Aim: Klotho, a key component of the endocrine fibroblast growth factor receptor—fibroblast growth factor axis, is a multi‐functional protein that impacts renal electrolyte handling. The physiological significance of Klotho will be highlighted in the regulation of calcium, phosphate, and potassium metabolism. Methods: In this review, we compare several murine models with different renal targeted deletions of Klotho and the insights into the molecular and physiological function that these models offer. Results: In vivo, Klotho deficiency is associated with severely impaired mineral metabolism, with consequences on growth, longevity and disease development. Additionally, we explore the perspectives of Klotho in renal pathology and vascular events, as well as potential Klotho treatment options. Conclusion: This comprehensive review emphasizes the use of Klotho to shed light on deciphering the renal molecular in vivo mechanisms in electrolyte handling, as well as novel therapeutic interventions. Purpose: We investigated the effects of low- and high-volume speed endurance training (SET), with a reduced training volume, on sprint ability, short- and long-term exercise capacity, muscle mitochondrial properties, ion transport proteins, and maximal enzyme activity in highly trained athletes. Methods: Highly trained male cyclists (maximal oxygen consumption (...): 68.3 ± 5.0 mL⋅min-1⋅kg-1, n = 24) completed 6 wk of either low (SET-L; 6 x 30-s intervals, n = 8) or high (SET-H; 12 x 30-s intervals, n = 8) volume SET twice per week with a 30% reduction in training volume. A control group (CON; n = 8) maintained their training. Exercise performance was evaluated by i) 6-s sprinting, ii) a 4-min time trial, and iii) a 60-min preload at 60% ... followed by a 20-min time trial. A biopsy of m. vastus lateralis was collected before and after the training intervention. Results: In SET-L, 4-min time trial performance was improved (P < 0.05) by 3.8%, with no change in SET-H and CON. Sprint ability, prolonged endurance exercise capacity, ..., muscle mitochondrial respiratory capacity, maximal citrate synthase activity, fiber type--specific mitochondrial proteins (complexes I-V), and phosphofructokinase (PFK) content did not change in any of the groups. In SET-H, maximal activity of muscle PFK and abundance of Na+-K+ pump-subunit α1, α2, β1, and phospholemman (FXYD1) were 20%, 50%, 19%, 24%, and 42% higher (P < 0.05), respectively after compared with before the intervention, with no changes in SET-L or CON. Conclusions: Low SET volume combined with a reduced aerobic low- and moderate-intensity training volume does improve short-duration intense exercise performance and maintain sprinting ability, ..., endurance exercise performance, and muscle oxidative capacity, whereas, high volume of SET seems necessary to upregulate muscle ion transporter content and maximal PFK activity in highly trained cyclists. Soil potassium deficiency is a common issue limiting agricultural productivity. Potassium-solubilizing bacteria (KSB) show significant potential in mitigating soil potassium deficiency, improving soil quality, and enhancing plant growth. However, different KSB strains exhibit diverse solubilization mechanisms, environmental adaptability, and growth-promoting abilities. In this study, we isolated a multifunctional KSB strain ZHS-1, which also has phosphate-solubilizing and IAA-producing capabilities. 16S rDNA sequencing identified it as Pantoea vagans. Scanning electron microscopy (SEM) showed that strain ZHS-1 severely corroded the smooth, compact surface of potassium feldspar into a rough and loose state. The potassium solubilization reached 20.3 mg/L under conditions where maltose was the carbon source, sodium nitrate was the nitrogen source, and the pH was 7. Organic acid metabolism profiling revealed that strain ZHS-1 primarily utilized the EMP-TCA cycle, supplemented by pathways involving pantothenic acid, glyoxylic acid, and dicarboxylic acids, to produce large amounts of organic acids and energy. This solubilization was achieved through direct solubilization mechanisms. The strain also secreted IAA through a tryptophan-dependent metabolic pathway. When strain ZHS-1 was inoculated into the rhizosphere of rice, it demonstrated significant growth-promoting effects. The rice plants exhibited improved growth and root development, with increased accumulation of potassium and phosphorus. The levels of available phosphorus and potassium in the rhizosphere soil also increased significantly. Additionally, we observed a decrease in the relative abundance of Actinobacteria and Proteobacteria in the rice rhizosphere soil, while the relative abundance of genera associated with acid production and potassium solubilization, such as Gemmatimonadota, Acidobacteria, and Chloroflexi, as well as Cyanobacteria, which are beneficial to plant growth, increased. These findings contribute to a deeper understanding of the potassium solubilization mechanisms of strain ZHS-1 and highlight its potential as a plant growth-promoting rhizobacteria. • Topping induces redistribution of potassium ions among different tissues of tobacco. • Topping-induced potassium redistribution is closely related to plant hormones. • IAA and CBL-CIPK pathway play key roles in the regulation of potassium metabolism. Topping is typically used in tobacco production to reduce reproductive consumption. However, following topping, declines in potassium content in tobacco leaves often have adverse effects on leaf growth, development, and quality. In this study, the mechanism of the topping-induced redistribution of potassium in the tobacco was studied based on physiological and ecological theories and molecular biology techniques. Our findings showed that the changes in potassium content in different parts of the tobacco after topping were closely correlated to the content of endogenous hormones, and the gray relational coefficient between potassium content and indole-3-acetic acid (IAA) content was the highest. Transcriptome sequencing and qRT-PCR analysis verified that the redistribution of potassium in tobacco plants induced by topping was mediated by the IAA signaling pathway and the CBL-CIPK signaling pathway. In addition, the genes of the NtPIN, NtPOT, NtSKOR , and NtCIPK families were found to play a significant role in the regulation of endogenous hormones and potassium metabolism in tobacco plants. Our study provides an important theoretical basis and data for elucidating the mechanism by which potassium metabolism is regulated in plants. Continuous cropping has a negative effect on soybean yield. In this study, a positioning experiment was conducted starting in 2015, with three treatments: maize–soybean rotation (SMR), 2-year maize, 2-year soybean rotation cropping (SC2), and 8-year soybean continuous cropping (SC8). We determined soybean yields (2015–2022) and analyzed soil microbial communities, functions, and metabolites composition in the 0–20 cm tillage layer using metagenomics technology and GC–MS technology during soybean flowering in 2022. Results indicated that continuous cropping (SC8) significantly reduced soybean yield compared to crop rotation (SMR) during the experimental period, while SC8 showed higher yield than SC2 in 2022. Compared to SMR, SC8 significantly increased soil N content and significantly decreased pH and TP, AP, and AK content. However, the pH and AK contents of SC8 were significantly higher than those of SC2. LeFSe analysis showed that Friedmanniella, Microlunatus, Nitrososphaera, Rubrobacter, Geodermatophilus, Nitriliruptor were enriched in SC8. Gaiella, Sphaerobacter, Methyloceanibacter were enriched in SC2. Sphingomonas, Cryobacterium, Marmoricola, Haliangium, Arthrobacter, Ramlibacter, Rhizobacter, Pseudolabrys, Methylibium, Variovorax were enriched in SMR. And the relative abundance of Cryobacterium, Marmoricola, Haliangium, Arthrobacter, Ramlibacter, Rhizobacter, Methylibium, Variovorax was significantly positively correlated with yield, while the relative abundance of Gaiella and Sphaerobacter was significantly negatively correlated with yield. SC8 significantly increased the abundance of genes in nitrogen metabolism and significantly decreased the abundance of genes related to phosphorus and potassium metabolism compared with SMR. However, the abundance of genes in potassium metabolism was significantly higher in SC8 than in SC2. Metabolomic analysis showed that compared to SMR, SC8 decreased the abundance of carbohydrates, ketones, and lipid. However, the abundance of carbohydrates, ketones, and lipid was significantly higher in SC8 than in SC2. Mantel test showed that soil pH and AK significantly affected soil microbial community, function, and metabolite composition. Introduction: This study will test the hypothesis that primary aldosteronism (PA) involves alterations in Na+, K+, and water content in the skin that are corrected by adrenalectomy. Aim and Methods: In skin biopsies, we will measure the content of Na+, K+, water, by physical-chemical methods and the osmotic-stress-responsive transcription factor Tonicity-responsive Enhancer Binding Protein (TonEBP, NFAT5) mRNA copy number by droplet digital PCR, in sex-balanced cohorts of 18 -75-year-old consecutive consenting patients with unilateral and bilateral PA, primary (essential) hypertension, and normotension. Before surgery, the patients with unilateral PA will receive the mineralocorticoid receptor antagonist (MRA) canrenone at doses that correct hypokalemia and high blood pressure values. They will be reassessed in an identical way one month after surgical cure, while off MRA. PA patients not selected for adrenalectomy will similarly be assessed at diagnosis and follow-up while on stable MRA treatment. Since a pilot study showed a direct correlation of dry weight (DW) with skin electrolytes and water content and significant differences of biopsy DW between surgery and follow-up, meaningful comparison of the skin cations and water content and TonEBP mRNA copy number, between specimen obtained at different time points, will require DW- and total mRNA-adjustment, respectively. Conclusion: This study will provide novel information on the skin Na+, K+ and water content in PA, the paradigm of salt-dependent hypertension, and novel knowledge on the effect of surgical cure of hyperaldosteronism. Anthocyanin biosynthesis in plants is influenced by a wide range of environmental factors, such as light, temperature and nutrient availability. In this study, we revealed that the potassium-repressed anthocyanin accumulation in radish hypocotyls was associated with altered sugar distribution and sugar signaling pathways rather than changes in oxidative stress status. Sugar-feeding experiments suggested a hexokinase-independent glucose signal acted as a major contributor in regulating anthocyanin biosynthesis, transport and regulatory genes at the transcriptional level. Several R2R3-MYBs were identified as anthocyanin-related MYBs. Phylogenetic and protein sequence analyses suggested that RsMYB75 met the criteria of subgroup 6 MYB activator, while RsMYB39 and RsMYB82 seemed to be a non-canonical MYB anthocyanin activator and repressor, respectively. Through yeast-one-hybrid, dual-luciferase and transient expression assays, we confirmed that RsMYB39 strongly induced the promoter activity of anthocyanin transport–related gene RsGSTF12, while RsMYB82 significantly reduced anthocyanin biosynthesis gene RsANS1 expression. Molecular models are proposed in the discussion, allowing speculation on how these novel RsMYBs may regulate the expression levels of anthocyanin-related structural genes. Together, our data evidenced the strong impacts of potassium on sugar metabolism and signaling and its regulation of anthocyanin accumulation through different sugar signals and R2R3-MYBs in a hierarchical regulatory system. Potassium (K+) is an endogenous substance that is an essential dietary component. However, the interaction between dietary arrangements and specific effects of dietary K+ intake in bioequivalence studies remains unclear. To investigate the influence of dietary arrangement on the bioequivalence of potassium chloride (KCl) sustained‐release tablets in healthy Chinese volunteers, the pharmacokinetics of KCl were compared in two open‐label, single‐center, randomized, two‐period crossover studies with different dietary conditions. All volunteers received an oral dose of 6 g of KCl sustained‐release tablets under fasting conditions, with different dietary arrangements. Urine samples were collected on baseline days and 48 hours after tablet consumption. Inductively coupled plasma–optical emission spectrometry was used to measure the concentration of K+ in the urine samples. Pharmacokinetic parameters were analyzed using Phoenix WinNonlin software in a noncompartmental model. In either clinical trial, no significant differences were observed in the maximal rate of urinary excretion and cumulative urinary excretion from 0 to 24 hours of K+ between the reference and test drugs. In a recent study, Looser et al. identified oligodendrocyte Kir4.1 as the activity-dependent driver of oligodendrocyte glycolysis that ensures that lactate is supplied to active neurons. Background and aims: Long-term monocultures seriously compromise crop productivity, which is known as replanting obstacle. Flowers are the most valuable tissue in annual plants and closely associated with pollen production, fertilization and seed set, however, their roles in replanting obstacle are unknown. We here explored how long-term monocropping impacts flowers, and revealed the potential acclimation mechanisms employing by flowers to monocropping. Methods: Field trial and pot experiments with peanut monocropped and rotated regimes were performed to study the response of peanut flowers to monocropping with physiological, transcriptomic and fungal community analyses. Results: Long-term monocropping reduced flower formation, pollen viability, pollen germination and tube growth, thereby reducing seed set in peanut. The transcriptome of flowers revealed that genes associated with the anther dehiscence, heat stress response, flavonoid biosynthesis, glutathione metabolism and potassium uptake were enriched by monocropping, which indicated that flowers experienced water deficit and oxidative stress. Inhibition of reactive oxygen species (ROS) synthesis or scavenging excess ROS rescued pollen activity decline under monocropping. By screening flower physiology and soil physicochemical properties, combined with drought and soil aggregate composition mimic experiments, we confirmed that flower water deficit and oxidative stress under monocropping were mediated by the soil structure alterations, especially large macro-aggregates (> 2 mm) reduction. Conclusion: Long-term monocropping alters soil structures, resulting in a water deficit and ROS accumulation in the flowers, which decreases pollen activity, and a multi-faceted actions including accelerated anther dehiscence, enhanced potassium absorption and better ROS scavenging capacity, are taken by flowers to acclimate to monocropping. Simple Summary: Cell fate is ultimately determined by the precisely coordinated action of the Ca2+-signaling machinery. During carcinogenesis, Ca2+ signaling is significantly remodeled due to mutations and/or ectopic expression. Here, we summarize current knowledge on how alterations in Ca2+ signaling contribute to the development of different cancer hallmarks. Emphasis is placed on the structure/function relationship of the well-studied store-operated Ca2+ channel, i.e., Orai1, and the Ca2+-activated K+ channel, i.e., SK3, alongside their individual and joint roles in cancer. This review lays out the current state of knowledge of Ca2+-signaling effectors and proteins as potential targets for the treatment of certain cancer types, with Orai1 and SK3 presented as emerging therapeutic targets. Cancer represents a major health burden worldwide. Several molecular targets have been discovered alongside treatments with positive clinical outcomes. However, the reoccurrence of cancer due to therapy resistance remains the primary cause of mortality. Endeavors in pinpointing new markers as molecular targets in cancer therapy are highly desired. The significance of the co-regulation of Ca2+-permeating and Ca2+-regulated ion channels in cancer cell development, proliferation, and migration make them promising molecular targets in cancer therapy. In particular, the co-regulation of the Orai1 and SK3 channels has been well-studied in breast and colon cancer cells, where it finally leads to an invasion-metastasis cascade. Nevertheless, many questions remain unanswered, such as which key molecular components determine and regulate their interplay. To provide a solid foundation for a better understanding of this ion channel co-regulation in cancer, we first shed light on the physiological role of Ca2+ and how this ion is linked to carcinogenesis. Then, we highlight the structure/function relationship of Orai1 and SK3, both individually and in concert, their role in the development of different types of cancer, and aspects that are not yet known in this context. Cultivation of sloping land is a main cause for soil erosion. Conservation practices, such as soil and stone terraces, may reduce the impacts of erosion but their impacts on soil microbial diversity and functioning related to carbon (C) and nutrient metabolisms remain unclear. This study was conducted to evaluate the effects of slope gradients (5°, 8°, 15°, 25°) and conservation practices (cultivated, uncultivated, soil terrace, and stone terrace) on bacterial and fungal diversities, metagenomic and metabolomic functioning associated with basic soil properties. Our results showed that steep slopes at 25° significantly decreased soil pH, silt percentage, and bacterial and fungal abundances, but that soil and stone terraces increased soil organic C (SOC), silt and clay contents, and fungal abundance compared to sloping cultivated lands. In addition, soil and stone terraces increased both bacterial and fungal alpha diversities, and relative abundances of Crenarchaeota, Nitrospirota, and Latescibacterota, but reduced the proportions of Actinobacteriota and Patescibacteria, thus shifting microbial beta diversities, which were significantly associated with increased SOC and silt content. For metagenomics, soil and stone terraces greatly increased the relative abundance of functional genes related to Respiration, Virulence, disease and defense, Stress response, and nitrogen and potassium metabolisms, such as Denitrification and Potassium homeostasis. For soil metabolomics, a total of 22 soil metabolites was enriched by soil and stone terraces, such as Lipids and lipid-like molecules (Arachidonic acid, Gamma-Linolenic acid, and Pentadecanoic acid), and Organoheterocyclic compounds (Adenine, Laudanosine, Methylpyrazine, and Nicotinic acid). To sum up, soil and stone terraces could reduce some of the negative impacts of steep slope cultivation on soil microbial diversity as well as their metagenomic and metabolomic functioning related to C and nutrient metabolism useful for soil health improvement, potentially bolstering the impact of sustainable practices in erosion hotspots around the world. Abnormal levels of potassium are linked to several health conditions, including high blood pressure, cardiac dysfunction, kidney damage, and osteoporosis. Given the limited availability of in vivo measurement techniques, there is a need for novel methods to measure potassium to enhance the diagnosis and management of potassium metabolism related diseases. This study aimed to evaluate the feasibility of compact neutron generator based in vivo measurement system for quantification of potassium using rat carcasses. A cohort of thirty-nine rats (n = 20 males and 19 females, average weight 255 ± 15 and 163 ± 7 g) were sacrificed, and their carcasses were placed in polyethylene bottles. The rats were then positioned and irradiated in a carefully designed irradiation cave built alongside the neutron generator with an optimized thermal neutron flux and radiation dose ratio. The irradiation time was 10 min, followed by a 5-min decay and 2-h measurement using a high efficiency high purity germanium detector(HPGe). The average potassium concentration in male and female rats was found to be comparable (male 2874 ± 161 and female 2866 ± 144 μg/g). A marginally positive correlation between potassium concentration and weight was found in female rats only (male(20) = 0.07, P = 0.76 and female r(19) = 0.34, P = 0.15). We assessed the influence of manganese toxicity on potassium levels and observed no significant impact. These results were consistent with our previous study in mice. By controlling urinary potassium excretion, the kidneys play a key role in maintaining whole-body potassium homeostasis. Conversely, low urinary potassium excretion (as a proxy for insufficient dietary intake) is increasingly recognized as a risk factor for the progression of kidney disease. Thus, there is a reciprocal relationship between potassium and the kidney: the kidney regulates potassium balance but potassium also affects kidney function. This review explores this relationship by discussing new insights into kidney potassium handling derived from recently characterized tubulopathies and studies on sexual dimorphism. These insights reveal a central but non-exclusive role for the distal convoluted tubule in sensing potassium and subsequently modifying the activity of the sodium-chloride cotransporter. This is another example of reciprocity: activation of the sodium-chloride cotransporter not only reduces distal sodium delivery and therefore potassium secretion but also increases salt sensitivity. This mechanism helps explain the well-known relationship between dietary potassium and blood pressure. Remarkably, in children, blood pressure is related to dietary potassium but not sodium intake. To explore how potassium deficiency can cause kidney injury, we review the mechanisms of hypokalemic nephropathy and discuss if these mechanisms may explain the association between low dietary potassium intake and adverse kidney outcomes. We discuss if potassium should be repleted in patients with kidney disease and what role dietary potassium plays in the risk of hyperkalemia. Supported by data and physiology, we reach the conclusion that we should view potassium not only as a potentially dangerous cation but also as a companion in the battle against kidney disease. We investigated potassium (K) accumulation characteristics and expression of K metabolism related genes in one high-K variety (ND202) and a common variety (NC89) of tobacco (Nicotiana tabacum L.). Results showed that K accumulation and leaf K content in ND202 were higher than those in NC89. The distribution rate and K accumulation in the leaves of ND202 increased significantly, while the distribution rate in the roots and stems had lower values. In addition, the maximum K accumulation rate and high-speed K accumulation duration in ND202 were found to be better than those in NC89. The expression of NKT1 in the upper and middle leaves of ND202 had an advantage, and the relative expression of NtKC1 and NtTPK1 in both the upper and middle leaves, as well as the roots, was also significantly upregulated. Conversely, the expression of NTRK1 in the lower leaves and roots of ND202 was weaker. ND202 had significantly greater expression levels of NtHAK1 than NC89 in the upper and middle leaves and roots; moreover, the expression of NtKT12 in the upper leaves and roots of ND202 was also higher. In comparison with common varieties, high-K varieties had a stronger ability to absorb and accumulate K. They also possessed higher expression of K+ channel- and transporter-related genes and showed a superior K accumulation rate and longer duration of high-speed K accumulation. This article investigates the polynomial fuzzy impulsive control problem for positive nonlinear systems subject to parameter uncertainties. The positive nonlinear systems are represented as positive interval type-2 (IT2) polynomial fuzzy-model-based systems, while the parameter uncertainties are captured by the IT2 membership functions (MFs). Considering that the controller being designed needs to be implementable and practical, in addition to ensuring the positivity and stability of the system, there are still some potential requirements for the controller, such as lower implementation cost and control cost. To reduce the implementation cost, the IT2 polynomial fuzzy impulsive controller is designed under the imperfect premise matching concept that the premise MFs and the number of rules of fuzzy controller are different from those of fuzzy model. To reduce the control cost, the impulsive controller that can tolerate larger impulse control interval (ICI) is designed by the following two novel methods. The first one is the proposed impulse-time-dependent discretized polynomial copositive Lyapunov function whose Lyapunov variable is designed as the interpolation polynomial function of time. The second one is the advanced IT2 membership-function-dependent (IT2MFD) analysis method which is improved by a novel footprint of uncertainty partitioning method proposed by this paper, so that this advanced IT2MFD method can alleviate the negative impact of parameter uncertainties on the ICI. Finally, a simulation example using lipoprotein metabolism and potassium ion transfer model as the nonlinear system to be controlled is used to verify the effectiveness of the proposed impulsive control methods. Accumulation of coal gangue (CG) poses significant risks to both human health and ecological systems, underscoring the urgent need for sustainable methods to utilize this abundant waste material effectively. In this study, we successfully screened and isolated the Bacillus velezensis bacterial strain to enhance the solubilization of potassium (K) and phosphorus (P) nutrients from CG. The study employed XRD, FTIR, SEM, and HPLC techniques to investigate the underlying mechanisms of CG solubilization. Various parameters such as CG particle size, incubation time, initial inoculation ratio, concentration of CG, pH, and temperature were optimized to maximize solubilization efficiency. The Bacillus velezensis bacterium can dissolve minerals of CG by adhering to its surface and secreting various kinds of organic acids, particularly succinic acid. Pot experiments further demonstrated that Bacillus velezensis , in conjunction with CG, promotes alfalfa growth. These combined findings suggest that Bacillus velezensis and coal gangue hold promising potential as mineral soil conditioners, effectively enhancing plant growth. Cadmium (Cd) is a potentially harmful metal with significant biological toxicity that adversely affects plant growth and physiological metabolism. Excessive Cd exposure in plants leads to stunted plant growth owing to its negative impact on physiological functions such as photosynthesis, nutrient uptake, and water balance. Potassium-enriched biochar (KBC) and Bacillus altitudinis rhizobacteria (RB) can effectively overcome this problem. Potassium-enriched biochar (KBC) significantly enhances plant growth by improving the soil structure, encouraging water retention, and enhancing microbial activity as a slow-release nutrient. Rhizobacteria promote plant growth by improving root ion transport and nutrient availability while promoting soil health and water conservation through RB production. This study examined the effects of combining RB + KBC as an amendment to rice, both with and without Cd stress. Four treatments (control, KBC, RB, and RB + KBC) were applied using a completely randomized design (CRD) in four replications. The results showed that the combination of RB + KBC increased rice plant height (38.40%), shoot length (53.90%), and root length (12.49%) above the control under Cd stress. Additionally, there were notable improvements in chlorophyll a (15.31%), chlorophyll b (25.01%), and total chlorophyll (19.37%) compared to the control under Cd stress, which also showed the potential of RB + KBC treatment. Moreover, increased N, P, and K concentrations in the roots and shoots confirmed that RB + KBC could improve rice plant growth under Cd stress. Consequently, these findings suggest that RB + KBC is an effective amendment to alleviate Cd stress in rice. Shifts in cytosolic pH have been recognized as key signaling events and mounting evidence supports the interdependence between H + and Ca 2+ signaling in eukaryotic cells. Among the cellular pH-stats, K + /H + exchange at various membranes is paramount in plant cells. Vacuolar K + /H + exchangers of the NHX (Na + ,K + /H + exchanger) family control luminal pH and, together with K + and H + transporters at the plasma membrane, have been suggested to also regulate cytoplasmic pH. We show the regulation of vacuolar K + /H + exchange by cytoplasmic pH and the calmodulin-like protein CML18 in Arabidopsis. The crystal structure and physicochemical properties of CML18 indicate that this protein senses pH shifts. Interaction of CML18 with tonoplast exchangers NHX1 and NHX2 was favored at acidic pH, a physiological condition elicited by K + starvation in Arabidopsis roots, whereas excess K + produced cytoplasmic alkalinization and CML18 dissociation. These results imply that the pH-responsive NHX-CML18 module is an essential component of the cellular K + - and pH-stats.

Chemical elements support various plant functions, and their reutilization is important for plant ecological adaptation. However, there is a lack of studies comparing the elemental concentration and their reutilization in floral petals and leaves of the same plant. To address this research gap, we conducted a comparative study across 38 plant species with diverse life forms in a common garden. Our investigation focused on the nutrient concentration of 10 elements in both petals and leaves and functional traits, including flower lifespan, dry mass per unit area, water concentration, and vein density. We have found that the elements of nitrogen (N), calcium (Ca), magnesium (Mg), iron (Fe), and manganese (Mn) were more abundant in leaves. In contrast, petals contained higher concentrations of phosphorus (P) and potassium (K). N, P, K, Ca, Mg, and sodium (Na) concentrations of petals were positively related to leaves. In herbaceous plants, their petals showed significant resorption of P and K, while N, P, and K were detected with significant resorption in leaves from all life forms. A positive correlation was found between the resorption of P in leaves and petals. From the perspective of the carbon economic spectrum, N and P showed a negative correlation with dry mass per unit area in leaves. Meanwhile, petal dry mass per area and floral longevity were significantly negatively correlated with P. The NHX1 gene encodes a Na+ /H+ antiporter located in the tonoplast membrane, which plays critical role in regulating plant salt tolerance. It is also involved in the uptake and accumulation of K in plants; however, its precise mechanism is unknown. In this research, we elucidated the physiological basis underlying the increases in K content induced by NHX1 . We evaluated main agronomic traits, leaf K content, K+ uptake kinetics, and root morphological and physiological characteristics from field-planted and hydroponic plants. We included a wild-type tobacco (Nicotiana tabacum ) variety (K326) and three transgenic tobacco lines (NK7, NK9, NK10) that overexpress AtNHX1 from Arabidopsis thaliana . Results demonstrated that the agronomic performance of the AtNHX1 -overexpressing tobacco lines was similar to K326 in field and hydroponic settings. The three AtNHX1 -overexpressing tobacco lines had significantly higher leaf K contents than K326. Under hydroponic condition, enhanced K uptake capacity and a larger maximum K uptake rate were seen in AtNHX1 -overexpressing tobacco lines. AtNHX1 -overexpressing lines also exhibited significantly superior root morphological and physiological traits relative to K326, including root biomass, root volume, absorption area, root activity, cation exchange capacity, soluble protein content, and H+ -ATPase activity. Overexpression of AtNHX1 in tobacco significantly improves the K uptake and accumulation. Therefore, leaf K content greatly increased in these transgenic lines in the end. Our findings strongly suggest that AtNHX1 overexpression increased leaf K content by boosting the capacity of enriching K in tobacco roots, thereby advancing the understanding of the function of AtNHX1 .

Anthropogenically elevated inputs of nitrogen (N), phosphorus (P), and potassium (K) can affect the carbon (C) budget of nutrient-poor peatlands. Fungi are intimately tied to peatland C budgets due to their roles in organic matter decomposition and symbioses with primary producers; however, the influence of fertilization on peatland fungal composition and diversity remains unclear. Here, we examined the effect of fertilization over 10 years on fungal diversity, composition, and functional guilds along an acrotelm (10-20 cm), mesotelm (30-40 cm), and catotelm (60-70 cm) depth gradient at the Mer Bleue bog, Canada. Simultaneous N and PK additions decreased the relative abundance of ericoid mycorrhizal fungi and increased ectomycorrhizal fungi and lignocellulose-degrading fungi. Fertilization effects were not more pronounced in the acrotelm relative to the catotelm, nor was there a shift toward nitrophilic taxa after N addition. The direct effect of fertilization significantly decreased the abundance of Sphagnum-associated fungi, primarily owing to the overarching role of limiting nutrients rather than a decline in Sphagnum cover. Increased nutrient loading may threaten peatland C stocks if lignocellulose-degrading fungi become abundant and accelerate decomposition of recalcitrant organic matter. Rice growth and yield response to salinity can be influenced by the duration and the timing of salt stress. The present study tested the effects of saline water irrigation from vegetative growth to maturity on rice growth and yield and ion concentrations in the straw and root and related them to changes in soil salinity and soil solute potential. The treatments consisted of five levels of saline water irrigation (electrical conductivity ~0.25 (control), 4, 6, 8, and 10 dS m-1) with two rice cultivars (BRRI dhan67 and BRRI dhan99) grown in pots in a rain shelter. Grain weight per pot, dry straw weight, and root weight were significantly reduced with increasing water salinity, but BRRI dhan99 was less affected. With prolonged saline water irrigation, salt concentration increased in the soil and lowered the soil solute potential. Increased saline water induced higher concentrations of Na+ in the straw (527-1200 mmol kg-1 at 4-10 dS m-1) relative to the root. By contrast, higher Cl- concentrations accumulated in the root than in the straw. The decrease of K+ in the straw and root for increasing salinity was inconsistent, but the Na+/K+ ratio sharply increased in the straw with higher water salinity. The increased Na+/K+ explained most grain weight loss due to higher salinity (R2 = 0.93) followed by Na+ (R2 = 0.87) and Cl-1 (R2 = 0.53). K + accumulation in plant tissues is a crucial factor for plant growth and development. The tomato high-affinity K + transporter SlHAK5 is essential for root K + acquisition from low external concentrations. It is also involved in K + accumulation in pollen and plant fertility as slhak5 KO plants show a low rate of pollen germination, impaired pollen tube growth and parthenocarpic fruits. Here, we present a thorough analysis of slhak5 flowers, which showed relevant defects at the anatomic, ionomic, metabolomic and transcriptomic levels. First, slhak5 flowers exhibited shorter styles and enlarged ovaries that, together with a low number of seeds in fruits from slhak5 X WT crosses, indicated an effect of the slhak5 mutation on female fertility. Second, a lower accumulation of Ca 2+ , as well as of several metabolites such as amino acids, citric acid and sugars, was observed in mutant flowers, whereas indole-3-acetic acid content was increased when compared to the wild-type. Third, RNAseq conducted on pistils and stamens of wild-type and slhak5 plants revealed that transport and signalling pathways are significantly enriched in the gene expression analyses of stamens. Patients with myotonia congenita suffer from slowed muscle relaxation caused by hyperexcitability. The diaphragm is only mildly affected in myotonia congenita; discovery of the mechanism underlying its resistance to myotonia could identify novel therapeutic targets. Intracellular recordings from two mouse models of myotonia congenita revealed the diaphragm had less myotonia than either the extensor digitorum longus (EDL) or the soleus muscles. A mechanism contributing to resistance of the diaphragm to myotonia was reduced depolarization of the interspike membrane potential during repetitive firing of action potentials, a process driven by build-up of K + in small invaginations of muscle membrane known as t-tubules. We explored differences between diaphragm and EDL that might underlie reduction of K + build-up in diaphragm t-tubules. Smaller size of diaphragm fibres, which promotes diffusion of K + out of t-tubules, was identified as a contributor. Intracellular recording revealed slower repolarization of action potentials in diaphragm suggesting reduced Kv conductance. Higher resting membrane conductance was identified suggesting increased Kir conductance. Computer simulation found that a reduction of Kv conductance had little effect on K + build-up whereas increased Kir conductance lessened build-up, although the effect was modest. Our data and computer simulation suggest opening of K + channels during action potentials has little effect on K + build-up whereas opening of K + channels during the interspike interval slightly lessens K + build-up. We conclude that activation of K + channels may lessen myotonia by opposing depolarization to action potential threshold without worsening K + build-up in t-tubules. KEY POINTS: In mouse models of the muscle disease myotonia congenita, the diaphragm has much less myotonia (muscle stiffness) than the extensor digitorum longus or soleus muscles. Identifying why the diaphragm is resistant to myotonia may help in developing novel therapy. We found the reason the diaphragm has less myotonia is that it is less prone to depolarization caused by K + build-up in t-tubules during repetitive firing of action potentials. Previous studies reported that there is an association between abnormal body fluid balance and prognosis in colitis patients. However, it remains to be clarified the effects of colitis on characteristics of body electrolytes or water content, including alternation in blood pressure. In this study, we examined the effects of colon injury on body water balance and blood pressure in the dextran sodium sulfate (DSS)-induced colitis mouse model. We evaluated body electrolytes and water content, blood pressure, and urea-associated water conservation in DSS mice. By 5 days after the treatment, DSS mice exhibited diarrhea but relatively maintained body weight and total body sodium, potassium, and water content by increases in water intake and hepatic ureagenesis. On 7 days after DSS treatment, when colitis becomes severe, DSS mice significantly decreased food and water intake, and body weight but significantly increased relative total body sodium, potassium, and water content per dry mass. Notably, DSS induced more total body dry mass loss relative to water loss. These body electrolytes and water accumulation on day 7 were associated with a reduction in urinary osmole excretion and urine volume accompanied by renal urea accumulation. DSS mice significantly increased blood pressure by day 5 and then decreased on day 7. These findings suggest that body electrolyte and fluid imbalance and alternations in blood pressure in colitis vary with the stage and severity of the condition. Previous studies on BSC2 have shown that it enhances yeast cell resistance to AmB via antioxidation and induces multidrug resistance by contributing to biofilm formation. Herein, we found that BSC2 overexpression could reverse the sensitivity of pmp3Δ to AmB and help the tested strains restore the intracellular sodium/potassium balance under exposure to AmB. Meanwhile, overexpression of the chitin gene CHS2 could simulate BSC2 to reverse the sensitivity of pmp3Δ and nha1Δ to high salt or AmB. However, BSC2 overexpression in flo11Δ failed to induce AmB resistance, form biofilms, and affect cell wall biogenesis, while CHS2 overexpression compensated the resistance of flo11Δ to AmB. Additionally, BSC2 levels were positively correlated with maintaining cell membrane integrity under exposure to AmB, CAS, or a combination of both. BSC2 overexpression in nha1Δ exhibited a similar function of CHS2, which can compensate for the sensitivity of the mutant to high salt. Altogether, the results demonstrate for the first time that BSC2 may promote ion equilibrium by strengthening cell walls and inhibiting membrane damage in a FLO path-dependent manner, thus enhancing the resistance of yeast cells to AmB. Established as a plant macronutrient, potassium (K) substantially bestows plant growth and thus, global food production. It is absorbed by plants as potassium cation (K + ) from soil solution, which is enriched through slow-release from soil minerals or addition of soluble fertilizers. Contribution of bioavailable K + from soil is usually insignificant (< 2 %), although the earth's crust is rich in K-bearing minerals. However, K is fixed largely in interlayer spaces of K-bearing minerals, which can be released by K-solubilizing bacteria (KSB) such as Bacillus, Pseudomonas, Enterobacter, and Acidithiobacillus. The underlying mechanisms of K dissolution by KSB include acidolysis, ion exchange reactions, chelation, complexolysis, and release of various organic and inorganic acids such as citric, oxalic, acetic, gluconic, and tartaric acids. These acids cause disintegration of K-bearing minerals and bring K + into soil solution that becomes available to the plants. Current literature review updates the scientific information about microbial species, factors, and mechanisms governing the bio-intrusion of K-bearing minerals. Moreover, it explores the potential of KSB not only for K-solubilization but also to enhance bioavailability of phosphorus, nitrogen, and micronutrients, as well as its other beneficial impact on plant growth. Agriculture is vital for global food security, and irrigation is essential for improving crop yields. However, irrigation can pose challenges such as mineral scarcity and salt accumulation in the soil, which negatively impact plant growth and crop productivity. While numerous studies have focused on enhancing plant tolerance to high salinity, research targeting various ecotypes of Arabidopsis thaliana has been relatively limited. In this study, we aimed to identify salt-tolerant ecotypes among the diverse wild types of Arabidopsis thaliana and elucidate their characteristics at the molecular level. As a result, we found that Catania-1 (Ct-1), one of the ecotypes of Arabidopsis, exhibits greater salt tolerance compared to Col-0. Specifically, Ct-1 exhibited less damage from reactive oxygen species (ROS) than Col-0, despite not accumulating antioxidants like anthocyanins. Additionally, Ct-1 accumulated more potassium ions (K + ) in its shoots and roots than Col-0 under high salinity, which is crucial for water balance and preventing dehydration. In contrast, Ct-1 plants were observed to accumulate slightly lower levels of Na + than Col-0 in both root and shoot tissues, regardless of salt treatment. These findings suggest that Ct-1 plants achieve high salinity resistance not by extruding more Na + than Col-0, but rather by absorbing more K + or releasing less K + . Ct-1 exhibited higher nitrate (NO 3 - ) levels than Col-0 under high salinity conditions, which is associated with enhanced retention of K + ions. Additionally, genes involved in NO 3 - transport and uptake, such as NRT1.5 and NPF2.3, showed higher transcript levels in Ct-1 compared to Col-0 when exposed to high salinity. However, Ct-1 did not demonstrate significantly greater resistance to osmotic stress compared to Col-0. These findings suggest that enhancing plant tolerance to salt stress could involve targeting the cellular processes responsible for regulating the transport of NO 3 - and K + . The integrated application of inorganic fertilizers, organic fertilizers, and biofertilizers helps sustain the nutrient pool and benefits the soil quality, thereby boosting plant health. The effect of different combinations of biofertilizers (consortium biofertilizer [CBF]-non-rhizobial PGPR), inorganic fertilizers, and organic fertilizers on soil health, growth, and yield of cowpea was evaluated by conducting a field experiment. The application of N 100  FYM + CBF resulted in significantly higher populations of bacteria, fungi, PSB, and diazotroph, as well as soil dehydrogenase and alkaline phosphatase enzyme activities. However, the application of N 100 FYM recorded a significantly higher actinomycetes population. The application of N 100 FYM + CBF resulted in significantly higher soil OC, available nitrogen, phosphorus, and potassium. The soil pH was recorded to be highest in control, and soil EC was recorded to be lowest in control. The plant uptake of nitrogen, phosphorus, and potassium was significantly higher with N 50 FYM + NP 50  + CBF. The root-shoot biomass, number of leaves, nodules/plant, number of pods/plants, pod biomass, pod length, and pod width were significantly higher in treatment having N 50 FYM + NP 50  + CBF. However, the height of the plant, number of branches, and biomass of leaves were highest in treatment with N 25 FYM + NP 75  + CBF. The pod and stover yield were significantly higher in treatment with N 50 FYM + NP 50  + CBF. Background: The integration of inorganic and organic fertilizers is increasingly being recommended to address the demand for sustainable cotton cultivation and to mitigate the ecological impacts of reliance on inorganic fertilizers. However, the nuanced effects of this combined fertilization approach on soil quality, cotton growth, yield, and their interaction mechanisms, remain unclear., Method: To elucidate this, a 2-year field trial (2022-2023) was conducted, incorporating five fertilization treatments: low inorganic fertilizer (BI1), high inorganic fertilizer (BI2), organic fertilizer (BO), combined low inorganic and organic fertilizer (BIO1), and combined high inorganic and organic fertilizer (BIO2). This study aimed to evaluate the influence of these treatments on soil quality, cotton growth, and yield., Results: The results indicate that the BO treatment significantly enhanced plant height growth rate, and BIO1 treatment increased leaf area index and dry matter accumulation growth rate. Critical soil parameters such as alkali-hydrolyzed nitrogen and available potassium emerged as pivotal determinants of soil quality over the trial period, corresponding to soil quality index (SQI) values of 0.482 and 0.478, and yields of 7506.19 kg ha -1 and 6788.02 kg ha -1 , respectively. Water productivity reached optimum levels at SQI values of 0.461 and 0.462, with corresponding efficiencies of 13.31 kg (ha mm) -1 and 12.16 kg (ha mm) -1 . Partial least squares path modeling revealed that integrating organic fertilizer with reduced inorganic fertilizer usage significantly boosts cotton yield by enhancing soil quality (path coefficient: 0.842)., Conclusion: In conclusion, this integrated fertilization strategy not only improves soil health but also increases agricultural productivity. Thallium (Tl) is a highly toxic heavy metal whose mechanism of toxicity is still not completely understood. The aim of this study was to test Tl cytotoxicity on several cell lines of different tissue origin in order to clarify specific Tl toxicity to a particular organ. In addition, possible interference of Tl with cell potassium (K) transport was examined. Human keratinocytes (HaCaT), human hepatocellular carcinoma (HepG2), porcine kidney epithelial cells (PK15), human neuroblastoma (SH-SY5Y) and Chinese hamster lung fibroblast cells (V79) were treated with thallium (I) acetate in a wide concentration range (3.9-500 µg/mL) for 24 h, 48 and 72 h. To assess competitive interaction between Tl and K, the cells were treated with four Tl concentrations close to IC 50 (15.63, 31.25, 62.50, 125 µg/mL) in combination with/or without potassium (I) acetate (500 µg/mL). The cells' morphology was monitored, and cytotoxic effect was assessed by 3-(4, 5-dimethylthiazole-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) test. The most sensitive to Tl exposure were SH-SY5Y cells, while HepG2 were the most resistant. The combined exposure to thallium (I) acetate and potassium (I) acetate for every cell line, except V79 cells, resulted in higher cell viability compared to thallium (I) acetate alone. Plants often face high salinity as a significant environmental challenge with roots being the first responders to this stress. Maintaining K + /Na + ratio within plant cells is crucial for survival, as the intracellular K + level decreases and the intracellular Na + level increases under saline conditions. However, knowledge about the molecular regulatory mechanisms of K + loss in response to salt stress through outward-rectifying K + channels in plants is largely unknown. In this study, we found that the Arabidopsis double mutant gorkskor, in which the GORK and SKOR genes are disrupted, showed an improved primary root growth under salt stress compared to wild-type (WT) and the gork and skor single-mutant plants. No significant differences in the sensitivity to mannitol stress between the WT and gorkskor mutant were observed. Accumulation of ROS induced by salt stress was reduced in the gorkskor roots. The gorkskor mutant seedlings had significantly higher K + content, lower Na + content, and a greater resultant K + /Na + ratio than the WT under salt stress. Moreover, salt-stress-induced elevation of cytosolic free Ca 2+ concentration was reduced in the gorkskor roots. The concentration of salt ions influences the availability and plant nutrients dynamics in the soil. Proper management of these ions can enhance food grain production, helping to feed the growing population. In this experiment, nine fertility combinations were followed to enhance the soil organic carbon and reduce the salt toxicity and monitor the plant nutrient availability. An incubation experiment was conducted for the period of one year with different organic soil amendments in combinations including biochar (BC), pressmud (PM), and farm yard manure (FYM) as follow: T1-control, T2-RDF, T3-FYM (10 t/ha), T4-PM (10 t/ha), T5-BC (10 t/ha), T6-FYM (5 t/ha) + PM (5 t/ha), T7-FYM (5 t/ha) + BC (5 t/ha), T8-PM (5 t/ha) + BC (5 t/ha), T9-FYM (5 t/ha) + BC (2.5 t/ha) + PM (2.5 t/ha). Results showed that addition of organic substance (10 t/ha) significantly (p < 0.05) affected soil pH and electric conductivity. Plant nutrient availability (N, K, and S) was also influenced by application of organic substance (10 t/ha). Organic C and available N were recorded the highest in the treatment T7 (FYM-5 t/ha + BC -5 t/ha); whereas, the highest available K and S were observed in treatment T5 (BC-10 t/ha). The microbial soil fertility indicators (alkaline phosphatases, arylsulphatase, dehydrogenase activity and microbial biomass carbon) were measured the highest in FYM (5 t/ha) + BC (5 t/ha) applied treatment. In conclusion, application of organic substance 10 t/ha (biochar alone or with FYM) improved the plant nutrient availability and soil microbial activities in saline soil. Potassium ion (K + ) is one of the most essential nutrients for the growth and development of tobacco (Nicotiana tabacum L.), however, the molecular regulation of K + concentration in tobacco remains unclear. In this study, a two-pore K (TPK) channel gene NtTPKa was cloned from tobacco, and NtTPKa protein contains the unique K + selection motif GYGD and its transmembrane region primarily locates in the tonoplast membrane. The expression of NtTPKa gene was significantly increased under low-potassium stress conditions. The concentrations of K + in tobacco were significantly increased in the NtTPKa RNA interference lines and CRISPR/Cas9 knockout mutants. In addition, the transport of K + by NtTPKa was validated using patch clamp technique, and the results showed that NtTPKa channel protein exclusively transported K + in a concentration-dependent manner. Arbuscular mycorrhizal (AM) fungi can enhance plant uptake of phosphorus (P) and potassium (K), but it is not yet clear whether rhizosphere bacteria can enhance the ability of AM fungi to acquire insoluble P and K from the soil. Here, pot experiments confirmed that AM fungus-promoted insoluble P and K uptake by plants requires rhizosphere bacteria. The changes of rhizosphere bacterial communities associated with AM fungi were explored by 16S rRNA amplicon sequencing and metagenomic sequencing. Five core bacteria genera identified were involved in P and K cycles. Synthetic community (SynCom) inoculation revealed that SynCom increased soil available P and K and its coinoculation with AM fungi increased P and K concentration in the plants. This study revealed that AM fungi interact with rhizosphere bacteria and promote insoluble P and K acquisition, which provided a foundation for the application of AM fungal-bacterial biofertilizers and was beneficial for the sustainable development of agriculture.

Potassium ions (K + ) are critical electrolytes that regulate multiple functions in immune cells. Recent studies have shown that the elevated concentration of extracellular potassium in the tumor interstitial fluid limits T cell effector function and suppresses the anti-tumor capacity of tumor-associated macrophages (TAMs). The effect of excess potassium on the biology of myeloid-derived suppressor cells (MDSCs), another important immune cell component of the tumor microenvironment (TME), is unknown. Here, we present data showing that increased concentrations of potassium chloride (KCl), as the source of K + ions, facilitate autophagy by increasing the expression of the autophagosome marker LC3β. Simultaneously, excess potassium ions significantly decrease the expression of arginase I (Arg I) and inducible nitric oxide synthase (iNOS) without reducing the ability of MDSCs to suppress T cell proliferation. Further investigation reveals that excess K + ions decrease the expression of the transcription factor C/EBP-β and alter the expression of phosphorylated kinases. While excess K + ions downregulated the expression levels of phospho-AMPKα (pAMPKα), it increased the levels of pAKT and pERK. Additionally, potassium increased mitochondrial respiration as measured by the oxygen consumption rate (OCR). Interestingly, all these alterations induced by K + ions were abolished by the autophagy inhibitor 3-methyladenine (3-MA). Our results suggest that hyperosmotic stress caused by excess K + ions regulate the mitochondrial respiration and signaling pathways in MDSCs to trigger the process of autophagy to support MDSCs' immunosuppressive function by mechanisms independent of Arg I and iNOS. Overall, our in vitro and ex vivo findings offer valuable insights into the adaptations of MDSCs within the K + ion-rich TME, which has important implications for MDSCs-targeted therapies.

Recent advancements in sustainable agriculture have spurred interest in hydroponics as an alternative to conventional farming methods. However, the lack of data-driven approaches in hydroponic growth presents a significant challenge. This study addresses this gap by varying nitrogen, magnesium, and potassium concentrations in hydroponically grown soybeans and conducting essential nutrient profiling across the growth cycle. Statistical techniques like Linear Interpolation are employed to interpolate nutrient data and a feature selection pipeline consisting of chi-squared testing methods, Linear Regression with Recursive Feature Elimination (RFE) and ExtraTreesClassifier have been used to select important nutrients for predicting water uptake using non-parametric regression methods. For different nutrient growth media, i.e. for soybeans grown in Hoagland + Nitrogen and Hoagland + Magnesium media, the Random Forest regressor outperformed other methods in predicting water uptake, achieving testing Mean Squared Error (MSE) scores of 24.55 ( R 2 score 0.63) and 8.23 ( R 2 score 0.81), respectively. Similarly, for soybeans grown in Hoagland + Potassium media, Support Vector Regression demonstrated superior performance with a testing MSE of 4.37 and R 2 score of 0.85. SHapley Additive exPlanations (SHAP) values are examined in each case to elucidate the contributions of individual nutrients to water uptake predictions. Erythropalum scandens Bl. is a medicinal woody vegetable found in southern China and parts of Southeast Asia. Studies have shown improper substrate hindered E. scandens seedling growth, causing water accumulation and nutrient deficiency. In pursuit of an ideal growth medium for E. scandens seedlings during the early stages, this study conducted a pot experiment to identify a mixed substrate with optimal water permeability and fertility. In this study, pure Alfisols soil treatment as the control (CK), and two soilless substrates (peat soil and perlite) were combined with Alfisols soil into different volume ratios, in order to better use soil resources from understory space and balance the texture of mixed substrates. The growth, physiological characteristics and nutrient status of 24-month-old E. scandens seedlings were determined after planting in different mixed ratios. The results showed that as the proportion of peat soil increased in the mix, most indexes exhibited an initial increase followed by a decline, while soluble protein content decreased consistently. Conversely, an increasing perlite ratio resulted in a general decline in most growth and physiological indexes. Root growth, biomass accumulation and chlorophyll content, peaked in the 66.67% Alfisols soil + 33.33% perlite (T4) treatment. Notably, T3 (66.67% Alfisols soil + 33.33% peat soil) showcased the best above-ground growth, while T1 (50.00% Alfisols soil + 50.00% peat soil) excelled in element content accumulation. In conclusion, the cultivation substrate should primarily consist of Alfisols soil, constituting at least 50%. The addition of peat soil enhances above-ground growth and nutrients accumulation, while perlite contributes to robust root development. One third of peat soil and a small amount of perlite can be added to the substrate during E. scandens seedling cultivation, and proper fertilization should also be used in order to increase nutrient accumulation in aboveground and underground parts. Salt stress can limit crop productivity, and there are differences in salt tolerance among plant varieties; however, we lack a comprehensive understanding of how keystone species obtained from different plant varieties under salt stress change plant biomass by driving root exudate secretion and regulating the Na + :K + ratio. We conducted a pot experiment for three wheat varieties (JiMai32 (JM32), XiaoYan60 (XY60), and ShanRong3 (SR3)) under saline/nonsaline soil conditions. Salt stress tended to significantly reduce wheat biomass, and the biomass reduction rates of the different varieties decreased in the order JM32 < XY60 < SR3. The compositions of the bacterial and fungal communities in the root endosphere, rhizosphere and bulk soil were measured, and salt-induced microbial taxa were isolated to identify keystone species from the co-occurrence networks and to study their effects on physiological responses to salinity in wheat varieties. We observed that root exudates participated in the regulation of the Na + :K + ratio, thereby affecting wheat biomass, and this process was regulated by keystone species. JM32 was enriched in microorganisms that promote plant growth and resistance to salt stress, such as Burkholderiales, Sordariomycetes, Alteromonadaceae, Acremonium, and Dokdonella, and inhibited microorganisms that are sensitive to the environment (salt, nutrients) and plant pathogens, such as Nocardioidaceae, Nitrospira, Cytophagaceae, Syntrophobacteriaceae, and Striaticonidium. XY60 inhibited microorganisms with biological control and disease inhibition potential, such as Agromyces and Kaistobacter. SR3-enriched pathogens, such as Aurantimonadaceae and Pseudogymnoascus, as well as microorganisms with antagonistic pathogen potential and the ability to treat bacterial infections, such as RB41 and Saccharothrix, were inhibited. Background: Soil salinization is a serious environmental hazard, limiting plant growth and production in different agro-ecological zones worldwide. Diethyl aminoethyl hexanoate (DA-6) as an essential plant growth regulator (PGR) exhibits a beneficial role in improving crop growth and stress tolerance. However, the DA-6-regulated effect and mechanism of salt tolerance in plants are still not fully understood. The objective of current study was to disclose salt tolerance induced by DA-6 in relation to changes in water and redox balance, photosynthetic function, ionic homeostasis, and organic metabolites reprogramming in white clover (Trifolium repens)., Results: A prolonged duration of salt stress caused water loss, impaired photosynthetic function, and oxidative injury to plants. However, foliar application of DA-6 significantly improved osmotic adjustment (OA), photochemical efficiency, and cell membrane stability under salt stress. In addition, high salinity induced massive accumulation of sodium (Na), but decreased accumulation of potassium (K) in leaves and roots of all plants. DA-6-treated plants demonstrated significantly higher transcript levels of genes involved in uptake and transport of Na and K such as VP1, HKT8, SOS1, NHX2, NHX6, and SKOR in leaves as well as VP1, HKT1, HKT8, H + -ATPase, TPK5, SOS1, NHX2, and SKOR in roots. Metabolomics analysis further illustrated that DA-6 primarily induced the accumulation of glucuronic acid, hexanoic acid, linolenic acid, arachidonic acid, inosose, erythrulose, galactopyranose, talopyranose, urea, 1-monopalmitin, glycerol monostearate, campesterol, stigmasterol, and alanine., Conclusions: The DA-6 significantly up-regulated transcript levels of multiple genes associated with increased Na + compartmentalization in vacuoles and Na + sequestration in roots to reduce Na + transport to photosynthetic organs, thereby maintaining Na + homeostasis under salt stress. The accumulation of many organic metabolites induced by the DA-6 could be attributed to enhanced cell wall and membrane structural stability and functionality, OA, antioxidant defense, and downstream signal transduction in leaves under salt stress. Nutrient requirement for crop growth, defined as the amount of nutrient that crops take up from soil to produce a specific grain yield, is a key parameter in determining fertilizer application rate. However, existing studies primarily focus on identifying nitrogen (N), phosphorus (P), and potassium (K) requirements solely in relation to grain yield, neglecting grain protein content, a crucial index for wheat grain quality. Addressing this gap, we conducted multi-site, multi-cultivar, and multi-year field trials across three ecological regions of China from 2016 to 2020 to elucidate variations in nutrient requirements for grain yield and grain protein. The research findings revealed that wheat grain yield ranged from 4.1 to 9.3 Mg ha -1 (average 6.9 Mg ha -1 ) and grain protein content ranged from 98 to 157 g kg -1 (average 127 g kg -1 ) across the three regions. Notably, the N requirement exhibited a nonlinear correlation with the wheat grain yield but a linear increase with increasing grain protein, while the P and K requirements positively correlated with grain yield and protein content. Regression models were formulated to determine the nutrient requirements (M ENR ), enabling the prediction of N, P, and K requirements for leading cultivars with varying grain yields and protein contents. Implementing nutrient requirements based on M ENR projections resulted in substantial reductions in fertilizer rates: 22.0 kg ha -1  N (10.7 %), 9.9 kg ha -1 P (20.2 %), and 8.1 kg ha -1  K (16.3 %). This translated to potential savings of 0.4 Mt. N, 0.23 Mt. P, and 0.17 Mt. K, consequently mitigating 5.5 Mt. CO 2 greenhouse-gas emission and yielding an economic benefit of 0.8 billion US$ annually in China. Metal ions have important roles in supporting the catalytic activity of DNA-regulating enzymes such as topoisomerases (topos). Bacterial type II topos, gyrases and topo IV, are primary drug targets for fluoroquinolones, a class of clinically relevant antibacterials requiring metal ions for efficient drug binding. While the presence of metal ions in topos has been elucidated in biochemical studies, accurate location and assignment of metal ions in structural studies have historically posed significant challenges. Recent advances in X-ray crystallography address these limitations by extending the experimental capabilities into the long-wavelength range, exploiting the anomalous contrast from light elements of biological relevance. This breakthrough enables us to confirm experimentally the locations of Mg 2+ in the fluoroquinolone-stabilized Streptococcus pneumoniae topo IV complex. Moreover, we can unambiguously identify the presence of K + and Cl - ions in the complex with one pair of K + ions functioning as an additional intersubunit bridge. Plants can survive in soils of low micromolar potassium (K + ) concentrations. Root K + intake is accomplished by the K + channel AKT1 and KUP/HAK/KT type high-affinity K + transporters. Arabidopsis HAK5 mutants impaired in low K + acquisition have been identified already more than two decades ago, the molecular mechanism, however, is still a matter of debate also because of lack of direct measurements of HAK5-mediated K + currents. When we expressed AtHAK5 in Xenopus oocytes together with CBL1/CIPK23, no inward currents were elicited in sufficient K + media. Under low K + and inward-directed proton motive force (PMF), the inward K + current increased indicating that HAK5 energetically couples the uphill transport of K + to the downhill flux of H + . At extracellular K + concentrations above 25 μM, the initial rise in current was followed by a concentration-graded inactivation. When we replaced Tyr450 in AtHAK5 to Ala the K + affinity strongly decreased, indicating that AtHAK5 position Y450 holds a key for K + sensing and transport. When the soil K + concentration drops toward the range that thermodynamically cannot be covered by AKT1, the AtHAK5 K + /H + symporter progressively takes over K + nutrition. High-affinity potassium transporters (HKTs) play an important role in plants responding to salt stress, but the transport properties of the soybean HKT transporters at the molecular level are still unclear. Here, using Xenopus oocyte as a heterologous expression system and two-electrode voltage-clamp technique, we identified four HKT transporters, GmHKT1;1, GmHKT1;2, GmHKT1;3 and GmHKT1;4, all of which belong to type I subfamily, but have distinct ion transport properties. While GmHKT1;1, GmHKT1;2 and GmHKT1;3 function as Na+ transporters, GmHKT1;1 is less selective against K+ than the two other transporters. Astonishingly, GmHKT1;4, which lacks transmembrane segments and has no ion permeability, is significantly expressed, and its gene expression pattern is different from the other three GmHKTs under salt stress. Interestingly, GmHKT1;4 reduced the Na+/K+ currents mediated by GmHKT1;1. Further study showed that the transport ability of GmHKT1;1 regulated by GmHKT1;4 was related to the structural differences in the first intracellular domain and the fourth repeat domain. Overall, we have identified one unique GmHKT member, GmHKT1;4, which modulates the Na+ and K+ transport ability of GmHKT1;1 via direct interaction. Changes in intracellular concentrations of Na + and K + are shown to alter gene expression. Another monovalent cation, Li + , is well known as a medicine for treatment of psychiatric disorders, but mechanism of its action is obscure. Thus, it is important to evaluate the effect of Li + on gene expression in endothelial cells. Here we studied influence of the increased intracellular Na + or Li + concentrations on transcription of Na + i /K + i -sensitive genes. Treatment of the human endothelial cells (HUVEC) with LiCl for 1.5 h resulted in accumulation of Li + in the cells. This was followed by increase in the FOS and EGR1 mRNAs levels and decrease in the JUN and MYC mRNA levels. Treatment of HUVEC with the Na + -ionophore monensin led to accumulation of Na + and loss of K + ions. However, monensin had no significant effect on gene expression. Incubation of HUVEC with elevated extracellular NaCl concentration increased intracellular K + concentration and transcription of the ATF3 gene, while transcription of the JUN gene decreased. These results indicate that Na + and Li + ions have different effects on the gene expression profile in the cells that is likely associated with the fact that they affect differently the intracellular monovalent cations ratio.

K + secretion in the distal nephron has a critical role in K + homeostasis and is the primary route by which K + is lost from the body. Renal K + secretion is enhanced by increases in dietary K + intake and by increases in tubular flow rate in the distal nephron. This review addresses new and important insights regarding the mechanisms underlying flow-induced K + secretion (FIKS). While basal K + secretion in the distal nephron is mediated by renal outer medullary K + (ROMK) channels in principal cells (PCs), FIKS is mediated by large conductance, Ca 2+ /stretch activated K + (BK) channels in intercalated cells (ICs), a distinct cell type. BK channel activation requires an increase in intracellular Ca 2+ concentration ([Ca 2+ ] i ), and both PCs and ICs exhibit increases in [Ca 2+ ] i in response to increases in tubular fluid flow rate, associated with an increase in tubular diameter. PIEZO1, a mechanosensitive, nonselective cation channel, is expressed in the basolateral membranes of PCs and ICs, where it functions as a mechanosensor. The loss of flow-induced [Ca 2+ ] i transients in ICs and BK channel-mediated FIKS in microperfused collecting ducts isolated from mice with IC-specific deletion of Piezo1 in the CCD underscores the importance of PIEZO1 in the renal regulation of K + transport., (© 2024 The Author(s). Plants require nutrients for growth, which they obtain from the soil via the root system. Fertilizers offer the essential nutrients (nitrogen, phosphorus, and potassium, as well as critical secondary elements) required by plants. Soil productivity falls with each crop until nutrients are provided. A wide range of so-called fertilizer products, such as organic fertilizers, argon mineral fertilizers, and mineral fertilizers, can assist farmers in adjusting fertilization methods based on the environment and agricultural conditions (inhibitors, restricted materials, growth mediums, plant bio-stimulants, etc.). Agricultural land is reduced by erosion, pollution, careless irrigation, and fertilization. On the other hand, more agricultural production is needed to meet the demands of expanding industries and the nutritional needs of a growing population. Nano fertilizers have recently started to be manufactured to obtain the highest yield and its quality per unit area. Previous researchers found that nano fertilizers could improve plant nutrient uptake efficiency, lower soil toxicity, mitigate the potential negative effects of excessive chemical fertilizer use, and reduce the frequency of fertilization. To maximize crop yields and optimize nutrient use while reducing the overuse of chemical fertilizers, nano fertilizersNFs are crucial in agriculture. The key component of these fertilizers is that they contain one or more macro- and micronutrients that can be applied regularly in minute doses while not damaging the environment. However, they have a minimal effect on plant growth and agricultural yields when employed in high numbers, like synthetic fertilizers. Periodontitis is a chronic inflammatory disease involving plaque biofilm as a pathogenic factor. Potassium ion plays an important role in cellular homeostasis; a large outflow of potassium may lead to local inflammation progression. In this work, the multifunctional short peptide molecule BmKTX-33 was designed by modifying the BmKTX, a Kv1.3 potassium channel inhibitor. This was to explore its antibacterial properties, capability of maintaining cell ion homeostasis, and bone-forming capacity. The results showed that BmKTX-33 had inhibitory effects on S. gordonii, F. nucleatum, and P. gingivalis. Moreover, BmKTX-33 also inhibited excessive potassium outflow in inflammatory environments. Finally, BmKTX-33 promoted MC3T3-E1 early osteogenesis while suppressing the NLRP3 inflammasome's production. In E. coli K-12, the absence of unphosphorylated PtsN (unphospho-PtsN) has been proposed to cause an L-leucine-sensitive growth phenotype (Leu S ) by hyperactivated K + uptake mediated impairment of the expression of the ilvBN operon, encoding subunits of the L-valine (Val)-sensitive acetohydroxyacid synthase I (AHAS I) that renders residual AHAS activity susceptible to inhibition by Leu and K + . This leads to AHAS insufficiency and a requirement for L-isoleucine (Ile). Herein, we provide an alternate mechanism for the Leu S of the ∆ptsN mutant. Genetic and physiological studies with suppressors of the Leu S indicate that impaired expression of the ilvBN operon jointly caused by the absence of unphospho-PtsN and the presence of Leu coupled to Leu-mediated repression of expression of AHAS III leads to AHAS insufficiency rendering residual AHAS activity susceptible to chronic Val stress that may be generated by exogenous Leu. Hyperactivated K + uptake and an elevated α-ketobutyrate level mediate elevation of ilvBN expression and alleviate the Leu S .
