Sciatic nerve injury is often associated with neuropathic pain and neuroinflammation in the central and peripheral nervous systems. In our previous work, Potamogeton perfoliatus L. displayed anti-inflammatory, antipyretic and analgesic properties, predominantly via the inhibition of COX-2 enzyme and attenuation of oxidative stress. Herein, we extended our investigations to study the effects of the plant's extract on pain-related behaviors, oxidative stress, apoptosis markers, GFAP, CD68 and neuro-inflammation in sciatic nerve chronic constriction injury (CCI) rat model. The levels of the pro-inflammatory marker proteins in sciatic nerve and brainstem were measured with ELISA 14 days after CCI induction. Pretreatment with the extract significantly attenuated mechanical and cold allodynia and heat hyperalgesia with better potential than the reference drug, pregabalin. In addition, CCI lead to the overexpression of prostaglandin E2 (PGE2), inducible nitric oxide synthase (iNOS), tumor necrosis alpha (TNFα), nuclear factor κB (NF-κB), cyclooxygenase-2 (COX-2), 5-lipoxygenase (5-LOX), and NADPH oxidase-1 (NOX-1) and decreased the catalase level in sciatic nerve and brainstem. The observed neuro-inflammatory changes were accompanied with glial cells activation (increased GFAP and CD68 positive cells), apoptosis (increased Bax) and structural changes in both brainstem and sciatic nerve. The studied extract attenuated the CCI-induced neuro-inflammatory changes, oxidative stress, and apoptosis while it induced the expression of Bcl-2 and catalase in a dose dependent manner. It also decreased the brainstem expression of CD68 and GFAP indicating a possible neuroprotection effect. The composition and nature of changes in the low-molecular-weight metabolome (LMWM) of Potamogeton perfoliatus L. growing in six biotopes of Lake Ladoga with different types of anthropogenic load have been analyzed. According to the results, it is found that the total number of low-molecular-weight organic compounds (LMWOCs) in the P. perfoliatus LMWM composition is directly dependent on anthropogenic load, which is well marked by the development of cyanobacteria. The greater the intensity of pollution or eutrophication of waters, or the higher the number of cyanobacteria, the lower the total number of LMWOCs and their concentration. A strongly pronounced dependence of the total concentrations of groups of LMWM compounds on the anthropogenic disturbance of the biotope and the concentration of cyanobacteria is revealed. A decrease in the number, relative amount, total concentration of carboxylic acids, and number and content of unsaturated fatty acids and an increase in the composition and content of phenols and the total content of aldehydes and ketones depends on an increase in anthropogenic pressure. The specific composition of the LMWM of claspingleaf pondweed depends on its response to biotic and abiotic factors of the aquatic (including anthropogenic) environment. The features of the change in the composition of the P. perfoliatus LMWM make it possible to use it as an integral indicator of the anthropogenic impact on the littoral biotopes of water bodies and the deterioration of their ecological state. Abstract The influence of macrophytes on the optical environment of the littoral zone was assessed by studying the effect of monospecific Potamogeton perfoliatus on the quantitative and qualitative properties of light and the response of plants to this altered environment. P. perfoliatus was shown to alter the optical environment and consequently its own architecture: in high-density pondweed patches, 67 percent of incident light was absorbed in the top 10 cm, while spectral properties of light was significantly altered. Leaf morphology and photophysiology adapted to these changes, with photosynthetically active biomass concentrated in the upper water layer and stem biomass increasing in the basal parts due to self-shading. This study highlights the importance of submerged macrophytes in shaping the optical environment and ecological dynamics of littoral zones. Not only do pondweed plants from different sites show very similar vertical patterns of morphological and physiological parameters, but they also contribute to similar vertical spatial variability in water optics, thus increasing habitat complexity. This added optical heterogeneity not only increases the diversity of the littoral zone, but also enriches the entire aquatic ecosystem of shallow lakes by providing additional optical ecological niches.

Aerenchyma formation plays an important role in the survival of Potamogeton perfoliatus in submerged environment. To understand the regulatory role of reactive oxygen species (ROS) and caspase 3-like protein signaling molecules in aerenchyma formation, we investigated the effects of exogenous NADPH oxidase inhibitor (diphenyleneiodonium chloride, DPI), catalase inhibitor (3-amino-1,2,4-triazole, AT), and caspase-3-like protein inhibitor (AC-DEVD-CHO, DEVD) on morphological and physiological characteristics and aerenchyma formation in P. perfoliatus. The results showed that after DPI treatment, caspase-3-like protein activity decreased, ROS-related enzyme activities increased, and H2O2 content decreased, thereby inhibiting aerenchyma formation. When the concentration of DPI was approximately 1 μmol/L, the inhibitory effect was the most obvious. On the contrary, after the AT treatment, caspase-3-like protein activity increased, ROS-related enzyme activities decreased, and the H2O2 content increased, ultimately promoting aerenchyma formation, and the promotion was the most obvious under treatment with approximately 500 μmol/L AT. After DEVD treatment, the inhibition of vegetative growth caused by DPI or AT treatment was alleviated, significantly reducing caspase-3-like activity and inhibiting aerenchyma development. The results of this study show that ROS has a positive regulatory effect on aerenchyma formation, and caspase-3-like protein is activated to promote ROS-mediated aerenchyma formation. This experiment provides a new theoretical basis for further exploration of the signal transduction effects of ROS and caspase-3-like protein in plant cells and their roles in plant development. Clasping-leaved Potamogeton L. species growing in Turkey are P. praelongus Wulfen and P. perfoliatus L. There exists no detailed study about distribution, habitat requirements, and anatomical properties of the Turkish populations of the two species. Potamogeton perfoliatus is widespread throughout the country but P. praelongus was recorded only from a single locality. Therefore, P. praelongus is rare and endangered in Turkey. In this study, we recorded presence of P. perfoliatus in 54 wetlands based on examination of 86 herbarium specimens. Physical and chemical parameters of the water bodies where the two species occur were measured from 24 sites for P. perfoliatus and from one site for P. praelongus. According to our findings, P. praelongus grows in an alpine lake with oligotrophic, calcareous and alkaline water. Potamogeton perfoliatus occupies diverse habitats but prefers deep lentic water bodies with high pH and low salinity levels. Stem anatomy of the species were studied based on three individuals for P. praelongus and 35 individuals for P. perfoliatus. Morphological features of the species were also investigated and descriptions based on Turkish material were prepared. We provided the distinguishing anatomical and morphological characters between the species. Our anatomical findings showed that P. praelongus specimens have eight vascular bundles in contrast to previous reports on the species. Our results can be used for future monitoring of the two submerged Potamogeton species as we provide detailed information about their current distribution pattern and habitat features.

Natural antioxidants, especially those of plant origins, have shown a plethora of biological activities with substantial economic value, as they can be extracted from agro-wastes and/or under exploited plant species. The perennial hydrophyte, Potamogeton perfoliatus, has been used traditionally to treat several health disorders; however, little is known about its biological and its medicinal effects. Here, we used an integrated in vitro and in vivo framework to examine the potential effect of P. perfoliatus on oxidative stress, nociception, inflammatory models, and brewer’s yeast-induced pyrexia in mice. Our results suggested a consistent in vitro inhibition of three enzymes, namely 5-lipoxygenase, cyclooxygenases 1 and 2 (COX-1 and COX-2), as well as a potent antioxidant effect. These results were confirmed in vivo where the studied extract attenuated carrageenan-induced paw edema, carrageenan-induced leukocyte migration into the peritoneal cavity by 25, 44 and 64% at 200, 400 and 600 mg/kg, p.o., respectively. Moreover, the extract decreased acetic acid-induced vascular permeability by 45% at 600 mg/kg, p.o., and chemical hyperalgesia in mice by 86% by 400 mg/kg, p.o., in acetic acid-induced writhing assay. The extract (400 mg/kg) showed a longer response latency at the 3 h time point (2.5 fold of the control) similar to the nalbuphine, the standard opioid analgesic. Additionally, pronounced antipyretic effects were observed at 600 mg/kg, comparable to paracetamol. Using LC-MS/MS, we identified 15 secondary metabolites that most likely contributed to the obtained biological activities. Altogether, our findings indicate that P. perfoliatus has anti-inflammatory, antioxidant, analgesic and antipyretic effects, thus supporting its traditional use and promoting its valorization as a potential candidate in treating oxidative stress-associated diseases.

Aquatic plants play an important role in maintaining the health of water environment in nature. Studies have shown that linear alkylbenzene sulfonate (LAS), a type of omnipresent pollutant, can cause toxic damage to aquatic plants. In the present research, we studied the physiological and growth response of submerged plant Potamogeton perfoliatus L. to different concentrations of LAS (0.1, 1.0, 10.0, 20.0, and 50.0 mg l−1). The results showed that LAS is toxic to P. perfoliatus, and the toxicity is dose-dependent. Only slightly reversible oxidative damages were observed in the physiological parameters of P. perfoliatus when P. perfoliatus was exposed to lower LAS doses (< 10 mg l−1): soluble sugar, soluble protein, H2O2, and malondialdehyde (MDA) content in P. perfoliatus increased significantly at 0.1 mg l−1 and then returned to normal levels at 1.0 mg l−1. Antioxidant enzymes were activated before the LAS concentration reached 10 mg l−1, and the activities of superoxide dismutase (SOD), catalase (CAT), and photosynthesis pigment content declined significantly when the concentration of LAS exceeded 10 mg l−1. In addition, at higher concentrations (20-50 mg l−1) of LAS, dry weight and fresh weight of P. perfoliatus showed significant declines. In the present study, Pb, Cu, Cr and Ni accumulation capabilities of aquatic macrophytes Potamogeton pectinatus, Potamogeton perfoliatus and Groenlandia densa (Potamogetonaceae) were determined separately after treated with both single and triple. The macrophytes were treated with Pb, Cu, Cr and Ni at 0, 5, 25 and 50 mg/L concentrations in a controlled chamber during 4-days. The tested heavy metals were differently accumulated by macrophytes. The maximum Pb, Cu, Cr and Ni accumulations were determined in G. densa tissues followed by P. perfoliatus and P. pectinatus in both macrophyte treatments. In both macrophyte applications, mean accumulation values of the heavy metals in the macrophyte tissues decreased according to this sequence: Pb>Cu>Ni>Cr. Bioconcentration factor (BCF), which indicates the efficiency of macrophyte to accumulate the metal, were decreased with increasing external metal concentrations. According to this, Potamogetonaceae species had a high bioaccumulation potantial at low metal concentrations. On the other hand, metal accumulations in individual macrophytes were generally found to be higher than combined macrophytes. Phosphate (P i ) uptake kinetics were determined in shoot and root tissues for four freshwater macrophyte species, Littorella uniflora , Potamogeton perfoliatus , Myriophyllum alterniflorum and Elodea canadensis , using a radioactive 33 P phosphate technique. Collection of plant material in the oligotrophic softwater lake, Lake Hampen, Denmark, where P i limits macrophyte growth, enabled us to characterize and compare the P i uptake kinetics and competitive characteristics of the four species in a low level P i environment. The maximum P i uptake rates ( V max ), the half saturation constants ( K m ) together with the affinity at low P i concentrations ( V max / K m ) were determined by fitting data to the Michaelis-Menten kinetics. L. uniflora showed the highest V max / K m in the root tissue and the lowest K m . M. alterniflorum showed the highest and E. canadensis and P. perfoliatus the lowest V max / K m in leaf tissue. M. alterniflorum had the highest V max and, as the only species, a higher V max in leaves than in roots. Surface area explained about half of V max in M. alterniflorum leaves. Roots were the dominant organ for P i assimilation for all species at the Lake Hampen P i concentrations. K m showed positive correlation to%P content in root tissue. The results indicate that at low lake water P i concentrations L. uniflora is able to survive on the P i pools in the sediment porewater. M. alterniflorum showed high affinity for P i at both low and high P i concentrations by both roots and shoots, and suggests that M. alterniflorum is a strong competitor at both low and high P i concentrations. The comparative analysis of the anatomical study, chloroplast ultrastructure and pigments content of submerged leaves of Potamogeton perfoliatus, P. pectinatus and Myriophyllum spіcatum at budding-flowering stage with light microscopy, transmission electron microscopic and biochemical methods were realized. The absent of stomata, the presence of undifferentiated mesophyll was shown with anatomical study of hydrophytes leaves. The common and distinctions signs of chloroplast ultrastructure in leaf photosynthesizing parenchyma were revealed in dependent from species. Light within the littoral zone affects the productivity and interaction between periphyton and its macrophyte substrate. The effect of periphyton on macrophyte photosynthesis, seasonal variation and vertical distribution of periphyton on artificial substrates (plastic strips), and the effect of periphyton on the light environment was studied in Lake Balaton. Data showed that an average of 4.1 ± 0.4 mg (dry weight) cm of periphyton had accumulated on the plastic strips after 8.8 ± 0.4 days. This biomass corresponded to 294 ± 30 μg m chl- a of epiphytic algae and blocked 92.3 ± 0.8 % of the depth specific radiation. Seasonal variation and specific vertical distribution of periphyton were observed. The most active time of periphyton accumulation corresponded to spring up until mid-June. Later in the year, the amount of periphyton significantly decreased. The optimal conditions for periphyton accumulation were at 30-40 cm depth. Most of the light reaching the adaxial leaf surface was attenuated by periphyton, decreasing the production of Potamogeton perfoliatus by 60-80 %. This increased the importance of backscattered light that corresponded to 10-15 % of the macrophyte production. A smaller part of the periphyton consisted of precipitated inorganic material, while epiphytic algae, making up the majority of the periphyton, were connected to both benthic (dominantly benthic penales) and pelagic (very close seasonal dynamics of pelagic and epiphytic biomass) algae. Periphyton affects macrophyte production especially in spring and in the upper water layers even in a mesotrophic water body. This increases the importance of the light absorbed through the abaxial side of the leaf and confirm the role of periphyton in transition from clear to turbid water states. [ABSTRACT FROM AUTHOR]

Abstract: The uptake of Se(IV) by Myriophyllum spicatum, Ceratophyllum demersum and Potamogeton perfoliatus, and the effects of Se(IV) on their physiological and biochemical characteristics were studied. Plants were cultivated outdoors under semi-controlled conditions in water solution containing Na selenite (20μgSeL−1 and 10mgSeL−1). The higher concentration of Se lowered the photochemical efficiency of PSII in all species studied, while the lower concentration had no effect on any species. The higher concentration of Se lowered respiratory potential in M. spicatum. The response of M. spicatum and C. demersum to Se(IV) regarding chlorophylls was variable, however in the majority of cases, there was a trend of increasing the amount of chlorophylls, while in P. perfoliatus the amount of chlorophyll a decreased. The concentration of Se in plants cultured in 10mgSe(IV)L−1 ranged from 436 to 839μgSeg−1 DM in M. spicatum, 319 to 988μgSeg−1 DM in C. demersum and 310 to 661μgSeg−1 DM in P. perfoliatus. The amount of soluble Se compounds in enzyme extracts of high Se treatment was 27% in M. spicatum, 41% in C. demersum and 35% in P. perfoliatus. Se compounds were determined using HPLC–ICP-MS. Comparative significance and synchronicity of morphological and photosynthetic adjustments of Potamogeton perfoliatus to shore-specific environments were examined on plants growing at the maximum depth of colonisation of the northern and southern shores of Lake Balaton. The shore-specific environments did not affect photophysiological parameters: the photosynthesis of plants on both shores was high, coupled with low respiration and compensation irradiances. In contrast, morphological and habitual differences between the shores were significant: plants of the shady, northern shore had lighter, but larger leaves, and longer internodes concentrated in the apex of the plants. Abiotic heterogeneity of the littoral zone of Lake Balaton influences both horizontal and vertical distribution of macrophytes, but biotic differences could shape the nature of a community. Vertical and temporal (small timescale) biomass distributions of Potamogeton perfoliatus and Myriophyllum spicatum were analysed in relation to their photosynthetic capacities to understand their coexistence, general presence over the northern shore of the lake seasonal shift of dominance within the community.Our results indicated the adaptation of these macrophytes to the rapidly changing and mostly low irradiance of the Lake Balaton originated from its high turbidity: both P. perfoliatus and M. spicatum had high photosynthetic activity (20 to 50 mg O2.g drw−1.h−1), low dark respiration (around 12–14% of maximal photosynthetic capacity) and high shade tolerance (Ic of plants were 29±18 and 26±18 μmol.m−2.s−1, respectively). The majority of photosynthetic parameters had no (or little) seasonal changes. On the other hand, in Lake Balaton P. perfoliatus and M. spicatum differed in vertical distribution of biomass: Myriophyllum concentrated its biomass in the upper, well-lit region of the water more strongly than the Potamogeton.Results suggest that the autogenic shift of dominance from Potamogeton to Myriophyllum in Lake Balaton can't be explained only by seasonal patterns of photophysiological changes, but supported by constitutive differences in plants architecture could led to the described phenomena. 1. The perfoliate pondweed Potamogeton perfoliatus L. constitutes large monospecific macrophyte patches in many Central European lakes. Correlative evidence from the field suggests that P. perfoliatus is under an increasing grazing pressure during its short vegetation period from May to September due to seasonal outbreaks of the aquatic moth Acentria ephemerella Denis & Schiffermüller. We used a mesocosm experiment to determine the influence of A. ephemerella herbivory on P. perfoliatus shoot development and resting bud production and to study the defence strategies of this macrophyte. 2. Herbivory resulted in a reduction of the P. perfoliatus vegetation period by more than 2 months thereby reducing the average resting bud size and the overall resting bud biomass sevenfold. This suggests that besides its severe immediate effects, herbivory affects P. perfoliatus growth and dynamics also during the subsequent season. 3. As a response to herbivory P. perfoliatus translocated nutrients (phosphorus (P) and nitrogen (N)) from leaves towards buds. Acentria ephemerella larvae had a high P content, implying P limitation of larval growth especially within the herbivory treatment. This suggests that at least the P translocation from leaves towards resting buds may be viewed as an anti-predator strategy rather than as a nutrient conservation strategy. 4. Acentria ephemerella herbivory changed the allocation strategy of P. perfoliatus in the size versus number of resting buds: only the number, but not the size of resting buds was reduced under shoot grazing by Acentria ephemerella. This change in the number versus size trade-off might allow the plant to produce a minimum resting bud size necessary for successful sprouting in the next spring. 5. Synthesis. Overall, our results suggest an escape syndrome (after Agrawal & Fishbein, Ecology, 87 (2006) S132) as a defence strategy against herbivory for P. perfoliatus, consisting of a shortening of the growth period, a translocation of nutrients and a change in allocation strategy. The increased plant senescence that was accompanied by the shortening of the growth period has further implications for the usage of macrophyte patches as a habitat for invertebrates and fishes and for the structure of littoral food webs. Restoration of submerged aquatic vegetation from seed has been hampered by a lack of information on the appropriate conditions for collecting, processing, and storing seeds prior to dispersal. Seeds must be processed and stored under conditions that maintain seed viability, meet dormancy requirements, and prevent premature germination. This study examined the effects of collection date, processing technique, aeration, storage and induction temperature and salinity, and storage period on seed germination of two mesohaline aquatic species, Potamogeton perfoliatus and Ruppia maritima. Collection date and processing technique were significant factors affecting seed yield from donor populations. Seeds of both species remained viable and germinated best when stored at 4°C, and then exposed to freshwater induction conditions. However, their responses to other factors differed. Aeration during storage was necessary in order to maintain viability of P. perfoliatus seeds, whereas it was unnecessary for R. maritima seeds. Storage in freshwater at 4°C prevented germination of P. perfoliatus seeds, while high salinity during cold storage was necessary to minimize premature germination of R. maritima. Mean germination time of P. perfoliatus was dependent on storage salinity; in contrast, mean germination time of R. maritima seeds was dependent on induction salinity. These differences indicate that the methods required to produce large quantities of underwater plant seed amenable to large-scale restoration efforts must be tailored to the specific requirements of individual species and must consider the range of processes from initial harvest through seed testing prior to field establishment. Protocols are now available for seed harvest, storage and germination of several mesohaline and polyhaline species; however, low seedling survival rates point to the need for an increased understanding of factors affecting seedling establishment. Depth of seed burial in sediments and initial seedling growth rates are shown to be limiting factors for photosynthetic competency of Ruppia maritima and Potamogeton perfoliatus. Seedling emergence is inversely proportional to planting depth on sediments ranging in grain size from coarse sands (850 μm) to silt (63 μm). Less than 6% of the seeds of either species emerged when buried to a depth of 3 cm in test sediments. Germination was greatest for seeds placed on the surface of sediments; however, these seedlings were subject to displacement because of the weak and fragile roots produced during early growth. Fine sediments may be more favorable for R. maritima seedling establishment, because seedling emergence and height decreased with increasing sediment grain size. Potamogeton perfoliatus seedlings seem to be more tolerant of a wider range of sediment grain sizes than R. maritima as indicated by the lack of an effect of sediment grain size on P. perfoliatus seed emergence, seedling height, and biomass. Increasing nutrients stimulated seedlings of both species; however, even at the highest concentrations tested, growth, as determined by shoot elongation and leaf and root formation, slowed within 7–10 days. The historic decline of submersed aquatic vegetation (SAV) in mesohaline regions of Chesapeake Bay, United States involved a diversity of plant species. The recent modest recovery is mostly, however, associated with a single, prolific but ephemeral species, Ruppia maritima. Two previously abundant and more stable species, Potamogeton perfoliatus and Stuckenia pectinata, have shown virtually no evidence of recovery. Based on previous studies that demonstrated the ability of R. maritima stands to enhance water clarity and nutrient conditions for SAV growth, we hypothesized that these beds would serve as effective “nursery” areas to incite transplant success for other SAV. We conducted experiments in a two-phase study at small and large spatial scales designed to explore this “nursery effect” as a restoration approach to increase plant species diversity. The first phase was conducted at small spatial scales to test effects of patch density by planting P. perfoliatus and S. pectinata into bare, sparse, and densely vegetated areas within three similar R. maritima beds in a tributary of Chesapeake Bay. Mean seasonal percent survivorship and shoot density were significantly higher in bare patches compared to vegetated patches. In the second phase of the study, P. perfoliatus was transplanted into separate R. maritima beds of different densities to test the effect of bed scale plant density on P. perfoliatus survival and growth. Epiphyton might have distinctive influence on the morphology of substrate macrophyte. In this article, we evaluate the influence of epiphyton on the morphological characteristics of their substrate submerged macrophyte, Potamogeton perfoliatus under two light intensities. The experiment was carried out for a period of 84 days in 12 glass aquaria under laboratory conditions. It was based on a 2 x 2 factorial design with epiphyton status (present or absent) and light intensity (200 or 80 µE m-2 s-1). Both epiphyton and light intensity had significant effects on the morphology and biomass allocation of the experimental plants. The average number of leaves, total length of newly recruited shoots and diameter of stems were greater in the epiphyton-free control plants than in the epiphyton-colonized plants under low light conditions. The plants with epiphyton allocated more biomass in their rhizomes and roots (% relative to total biomass basis) when compared to the control plants in both light intensities. There were also significant epiphyton-light interactions. The control plants under low light intensity showed higher internodal elongation in their main shoots when compared to the plants under high light intensity as an adaptation mechanism. Whereas the plants with epiphyton did not show such an adaptation. The new shoots of the control plants under low light intensity did not show any internodal elongation as observed in the main shoots. Furthermore, the length of the leaves of main shoots was larger in control plants with epiphyton and high light intensity than in plants with epiphyton and low light intensity, but such a variation was absent in the new shoots. Abstract: Macrophytes play a crucial role in the functioning of lake ecosystems. Until now most macrophyte models neglected the fact that the majority of macrophyte species expand clonally during the growing season. Inclusion of a detailed description of clonal growth in models can facilitate our understanding of space occupation and patch expansion and predict future macrophyte development. “CLOMO” is an individual-based model which includes a detailed, spatially explicit description of rhizome formation and clone expansion as well as a realistic description of photosynthesis including light limitation and temperature. The model also accounts for transfers of energy or resources between different parts of the clone (“clonal integration”). Although the clonal growth of macrophytes is complex and poorly known, the first model results for the macrophyte species Potamogeton perfoliatus were promising and compared well with the field data. The model can produce growth networks very similar to those found in the field. A Monte Carlo sensitivity analysis showed systematically which parameters have the largest effect on the architecture and expansion of the clones. The application of the model provided new insights into growth dynamics and patch development: (1) the model showed that a lack of branching will lead to the extinction of the clone after a certain number of years. This is due to the fact that the reproductive organs (turions) are formed at the end of a branch and even a small turion mortality will cause a reduction in surviving plant numbers; (2) the growth of rhizome axes relative to those in the previous year determines the patch density and patch expansion rate. Abstract: The littoral plankton of Lake Peipsi (3555 km2, mean depth 7.1 m) was studied in summer 1980 and 2000–2002, and the epiphyton was studied on two dominating macrophytes, Phragmites australis (Cav.) Trin. ex Steud. and Potamogeton perfoliatus L., in 2000. The purpose of the study was to estimate to what extent the littoral phytoplankton and epiphytic algae (their biomass, chlorophyll a content and dominant species) can be used as a criterion for the trophic state of the lake. In wind-open areas, phytoplankton biomass in the littoral is commonly more than ten times higher than in open water due to the presence of the macroscopic cyanobacterium Gloeotrichia echinulata (J.S. Smith) P. Richter. This alga is abundant in the moderately eutrophic northern lake''s part and rare in the strongly eutrophic southern part; hence also biomass in the southern part is considerably lower. In open water, phytoplankton biomass increases southward with increasing trophy. Algal biomass in the littoral depends on wind direction and can change completely in a few days. Epiphyton biomass and Chla content increased southward with increasing trophic state. They both revealed significant Spearman correlations (P < 0.05) with wind index and transparency (negative), and with abundance of the host plant, both reed and pondweed (positive). The phytoplankton biomass of the littoral of the large and shallow Lake Peipsi can not be used as a criterion of trophic state, however, the species composition of the dominants, particularly cyanobacteria, is well applicable for this purpose. The shift from a turbid-water state to a clear macrophyte-dominatedstate in the shallow lake Veluwemeer (The Netherlands) has led to nuisance forrecreational navigation. The nuisance concerns the dense beds of Potamogeton perfoliatus in particular, whereas thelow-canopy forming charophytes cause much less harm. On the other hand, theimportance of macrophyte cover for the stability of the clear-water state hasbeen recognised. To assess the potential of mechanical removal of densemacrophyte beds, several cutting regimes were simulated in a mixed vegetation of P. perfoliatus and Chara aspera, usingthe individual-based model Charisma. These species occupy a wide range of waterdepths between 0.5 and 2.5 m, with C. asperadominating the shallower zone and P. perfoliatus dominatingthe deeper zone; intermediary is a zone where either species may dominate asalternative equilibria. Both the cutting height and timing affected the amountof biomass present in summer. The effect of cutting was more profound fortreatment later in the season. With a cutting level above the Chara-canopy, the simulations showed an increased biomassof C. aspera and reduction of P.perfoliatus. In the zone of alternative equilibria, it was possibleto provoke a sustainable shift from P. perfoliatusdominance to C. aspera dominance. To achieve this, annualrepetition of cutting for a number of years was necessary. A harvesting regimeaimed at shifting the vegetation dominance from P.perfoliatus towards C. aspera could be an optionfor management of the lake, since it holds the perspective of decreasedmanagement effort after a number of years. Summary •Investigations of seasonal temperature acclimation in gas exchange are few and only exist for terrestrial and marine plants. Here we report on results obtained for three freshwater macrophyte species (Callitriche obtusangula, Potamogeton pectinatus and Potamogeton perfoliatus). •We collected plants from the field at monthly intervals and measured photosynthetic and respiratory temperature-response curves. Fitted and calculated parameters were derived from the curves and a simple model was used to evaluate the acclimative capacity to seasonal variation in water temperature. •For all species, optimum temperatures for gross photosynthesis showed little temporal variation. In addition, the shape of the temperature-response curves at suboptimal temperature was not optimized to temporal differences in water temperature. The only consistent seasonal trend in gas exchange was a gradual decrease in photosynthetic and respiratory capacity over time. •Our measurements and model predictions did not point to an acclimative seasonal response in the thermal dependence of oxygen exchange. Hence, we conclude that either other processes constrain the plants’ response, or temporal variation in water temperature is less important than seasonal loss of photosynthetic capacity. Natural antioxidants, especially those of plant origins, have shown a plethora of biological activities with substantial economic value, as they can be extracted from agro-wastes and/or under exploited plant species. The perennial hydrophyte, Potamogeton perfoliatus, has been used traditionally to treat several health disorders; however, little is known about its biological and its medicinal effects. Here, we used an integrated in vitro and in vivo framework to examine the potential effect of P. perfoliatus on oxidative stress, nociception, inflammatory models, and brewer's yeast-induced pyrexia in mice. Our results suggested a consistent in vitro inhibition of three enzymes, namely 5-lipoxygenase, cyclooxygenases 1 and 2 (COX-1 and COX-2), as well as a potent antioxidant effect. These results were confirmed in vivo where the studied extract attenuated carrageenan-induced paw edema, carrageenan-induced leukocyte migration into the peritoneal cavity by 25, 44 and 64% at 200, 400 and 600 mg/kg, p.o., respectively. Moreover, the extract decreased acetic acid-induced vascular permeability by 45% at 600 mg/kg, p.o., and chemical hyperalgesia in mice by 86% by 400 mg/kg, p.o., in acetic acid-induced writhing assay. The extract (400 mg/kg) showed a longer response latency at the 3 h time point (2.5 fold of the control) similar to the nalbuphine, the standard opioid analgesic. Additionally, pronounced antipyretic effects were observed at 600 mg/kg, comparable to paracetamol. Using LC-MS/MS, we identified 15 secondary metabolites that most likely contributed to the obtained biological activities. Altogether, our findings indicate that P. perfoliatus has anti-inflammatory, antioxidant, analgesic and antipyretic effects, thus supporting its traditional use and promoting its valorization as a potential candidate in treating oxidative stress-associated diseases.
