7. Sequence typing of Haemophilus ducreyi isolated from patients in the Namatanai region of Papua New Guinea: Infections by Class I and Class II strain types differ in ulcer duration and resurgence of infection after azithromycin treatment.

Abstract

Haemophilus ducreyi (HD) is an important cause of cutaneous ulcers in several endemic regions, including the Western Pacific Region, especially among children. An HD sequence typing on swab samples taken from 1,081 ulcers in the Namatanai district of Papua New Guinea, during the pilot study for treatment of yaws, has been performed using the Grant typing system. Of the 363 samples that tested positive for the 16S rDNA of HD, the dsrA sequences of 270 samples were determined. Altogether they revealed 8 HD strain types circulating in Namatanai, including seven strain types of Class I (I.3, I.4, I.5, I.9, I.10, I.11, I.12) and one strain of Class II (II.3); four Class I types (I.9, I.10, I.11, I.12) were novel. The southern region of Namatanai (Matalai Rural) was identified as the region with the lowest genotype diversity and with most infections caused by HD Class II. The middle and northern subdistricts were affected mainly by HD Class I. Analysis of patient characteristics revealed that Class II HD infections were more often represented by longer-lasting ulcers than Class I HD infections. An increase in the prevalence of the I.10 strain was found after azithromycin administration compared to the untreated population at baseline likely reflecting higher infectivity of HD Class I, and more specifically strain type I.10. Author summary: Haemophilus ducreyi (HD) is one of the main causative agents of cutaneous ulcers in children across substantial parts of Africa and the Pacific. A yaws eradication study conducted in the Namatanai district of Papua New Guinea revealed that nearly one-third of all patients were infected with HD. Single locus sequence typing (SLST) of the HD dsrA locus revealed presence of multiple HD strains circulating in the human population of Namatanai region. In contrast to Treponema pallidum subsp. pertenue, the pathogen responsible for yaws, the prevalence of HD strains did not significantly decrease following mass drug administration. In addition, HD strains from patients in Namatanai region revealed a higher genetic diversity compared to the yaws agent present in the same area and the HD strain composition changed following antibiotic treatment. Infections caused by at least some different HD strain types appear to differ in reported ulcer duration and also in resurgence of infection after antibiotic treatment. Abstract

The treponemes infecting lagomorphs include Treponema paraluisleporidarum ecovar Cuniculus (TPeC) and ecovar Lepus (TPeL), infecting rabbits and hares, respectively. In this study, we described the first complete genome sequence of TPeL, isolate V3603-13, from an infected mountain hare (Lepus timidus) in Sweden. In addition, we determined 99.0% of the genome sequence of isolate V246-08 (also from an infected mountain hare, Sweden) and 31.7% of the genome sequence of isolate Z27 A77/78 (from a European hare, Lepus europeaus, The Netherlands). The TPeL V3603-13 genome had considerable gene synteny with the TPeC Cuniculi A genome and with the human pathogen T. pallidum, which causes syphilis (ssp. pallidum, TPA), yaws (ssp. pertenue, TPE) and endemic syphilis (ssp. endemicum, TEN). Compared to the TPeC Cuniculi A genome, TPeL V3603-13 contained four insertions and 11 deletions longer than three nucleotides (ranging between 6 and2,932 nts). In addition, there were 25 additional indels, from one to three nucleotides long, altogether spanning 36 nts. The number of single nucleotide variants (SNVs) between TPeC Cuniculi A and TPeL V3603-13 were represented by 309 nucleotide differences. Major proteome coding differences between TPeL and TPeC were found in the tpr gene family, and (predicted) genes coding for outer membrane proteins, suggesting that these components are essential for host adaptation in lagomorph syphilis. The phylogeny revealed that the TPeL sample from the European brown hare was more distantly related to TPeC Cuniculi A than V3603-13 and V246-08. [ABSTRACT FROM AUTHOR]

Abstract

The chronosequence approach, that is, analyses of several stages differing in age since disturbance or restoration but more or less comparable in site conditions, may well demonstrate, despite some limitations, a long‐term ecosystem development. Earlier, we described spontaneous vegetation succession in 11 differently aged industrially harvested raised bogs and suggested that spontaneous recovery of plant cover gradually proceeds toward the reference state of natural raised bogs. We repeated the analyses after 10 years and asked: (1) Are the successional trajectories of spontaneous restoration really directing toward reference natural sites? and (2) How do typical peatland plant species participate in re‐vegetation? Artificially afforested extracted peatland sites were used for comparison. We concluded that spontaneous recovery of plants does not proceed toward the reference sites. Instead it directs more toward the artificially afforested sites. Despite this, the spontaneously developed vegetation harbored many more peatland species. To turn the development toward the natural reference sites would require a substantial rise of the water table after ceasing peat extraction. The study demonstrates the importance of repeated analyses, which may give more reliable signs of the re‐vegetation direction than one‐time analyses. [ABSTRACT FROM AUTHOR]

Abstract

The aim of the diploma thesis "The Shepherd as the Anthropomorphism of God" is to clarify the role of the shepherd in antiquity and today and to present this knowledge in the use of the shepherd's anthropomorphism in biblical texts by means of literary research and comparison of texts, exegesis and reflection of own experience. In the introductory chapters we focus on the person of the shepherd, his characteristics, where special attention is paid to the shepherd's staff, as it also has an abundant biblical metaphorical use. The role of the shepherd in antiquity, in biblical texts and today is also discussed. Attention is also paid to the metaphor of the shepherd in ancient mythology, the use of this metaphor in various religious cults and its content. From a similar point of view, grazing animals are viewed, especially sheep, and special attention is paid to the role of the lamb in biblical texts. But we also talk about pigs and goats as pastoral animals. The following chapters first deal with anthropomorphisms in general, followed by chapters from specific biblical books where anthropomorphism is used. The texts are divided according to their inclusion in the Bible - first, Tanach texts are discussed, where the most common occurrence of anthropomorphisms is in the prophetic books. The next...

Abstract

In the genus Treponema there are several human and animal pathogens that include the causative agent of syphilis (Treponema pallidum ssp. pallidum; TPA), the causative agent of yaws (T. p. ssp. pertenue; TPE), and the causative agent of endemic syphilis (T. p. ssp. endemicum; TEN). T. paraluisleporidarum causes venereal spirochetosis in rabbits. We used whole genome sequences of 11 treponemal strains together with additional 62 draft genomic data to identify genes evolving under positive selection. The identified genes evolving under positive selection partly overlapped with the genes previously reported as recombinant and were found to be different in treponemal subspecies. Since both genetic recombination and positive selection could allow a survival of pathogenic bacteria despite the human immune response, identification of such genes could predict the major antigens recognized by the human immune system and also identify the most suitable components for development of an anti-treponemal vaccine.

Abstract

Syphilis, caused by Treponema pallidum subsp. pallidum (TPA), remains an important public health problem with an increasing worldwide prevalence. Despite recent advances in in vitro cultivation, genetic variability of this pathogen during infection is poorly understood. Here, we present contemporary and geographically diverse complete treponemal genome sequences isolated directly from patients using a methyl-directed enrichment prior to sequencing. This approach reveals that approximately 50% of the genetic diversity found in TPA is driven by inter- and/or intra-strain recombination events, particularly in strains belonging to one of the defined genetic groups of syphilis treponemes: Nichols-like strains. Recombinant loci were found to encode putative outer-membrane proteins and the recombination variability was almost exclusively found in regions predicted to be at the host-pathogen interface. Genetic recombination has been considered to be a rare event in treponemes, yet our study unexpectedly showed that it occurs at a significant level and may have important impacts in the biology of this pathogen, especially as these events occur primarily in the outer membrane proteins. This study reveals the existence of strains with different repertoires of surface-exposed antigens circulating in the current human population, which should be taken into account during syphilis vaccine development.

Outer Space Activities & Environmental Protection Abstract This thesis aims at addressing various connecting factors between outer space activities and environmental protection. It examines the present legal regime of the outer space with particular regard to the environmental protection. The analysis of the most relevant space treaties is provided, reviewing the provisions concerning the protection of the environment, while addressing the emerging environmental issues. The significant role of international bodies is being emphasized, as well as the current challenges the regime of outer space is facing. Although the space-related activities offer an extensive tool to enhance environmental protection of Earth, the environment of space is nowadays being exposed to an imminent threat called space debris, which could prevent all possible future space activities if left unattended. The pollution of the space environment is at a critical level, and the international community is required to respond within a short time in the future. The introduction of mitigating efforts will no longer itself be effective without the addition of active employment of remediating measures, such as the active debris removal. On the other hand, space activities already widely contribute to preserving the environment of Earth...

Background: Whole-genome sequencing (WGS) of Treponema pallidum subspecies pallidum (TPA) has been constrained by the lack of in vitro cultivation methods for isolating spirochetes from patient samples.Methods: We built upon recently developed enrichment methods to sequence TPA directly from primary syphilis chancre swabs collected in Guangzhou, China.Results: By combining parallel, pooled whole-genome amplification with hybrid selection, we generated high-quality genomes from 4 of 8 chancre-swab samples and 2 of 2 rabbit-passaged isolates, all subjected to challenging storage conditions.Conclusions: This approach enabled the first WGS of Chinese samples without rabbit passage and provided insights into TPA genetic diversity in China. This thesis deals with evaluation of financial and economical situation in furniture company NADOP-výroba nábytku a.s., for the period 2011-2015. Goal of the work is an elaboration of financial analysis of company's current condition using methods of horizontal and vertical analysis, differential indicators and ratio analysis. Theoretical and methodological basis and other aspects are described in the theoretical part. Practical part contains developed financial analysis based on which are evaluated the current situation and further developement of the company, and a prediction of financial distress. Part of the thesis is also proposal of tools and procedures for improving financial management based on the found results.

Abstract

Bachelor thesis "The infuence of opinion leaders on crystallizing of attitudes in small group" deals with the subject of opinion leadership and its role in shaping the opinion of an individual in a group. The author of the study analyses whether aspects of opinion leadership, discussed in the theoretical part, could be found in two chosen groups, and if so, how these groups differ from each other. Two mixed design research methods are used, specifically methods of informants and self-identification S-GOLS (Generalized Opinion Leadership Scale) method. Opinion leaders were in the quantitative part of the research identified and the area of their influence was specified. In the qualitative part of the research were then made in- depth interviews with such influential people. That leads to description of common characteristics of these opinion leaders, determine their inclusion in existing typologies and pointed variances between them. The differences between the data obtained by using both quantitative methods were analysed and their strengths and weaknesses were reflected. The study is based on findings of classical authors in the area of opinion leadership on which the research hypotheses have been also established.

The aim of my bachelor thesis is to detach chemistry curriculum from its stereotypical image of dull subject and bring it close to students as an essential element of everyday life. The first part deals with the problem of popularization of chemistry and a variety of didactic means and principles of their creating, which are used in Czech schools. In the practical part of the thesis an interactive Web sites focused on carboxylic acids were made, which offer a depersonalized world of chemistry in a funny way for students and it provids teachers with modern teaching aid. A set of worksheets with the author's answers is a part of these websites as well as powerpoint presentation "Carboxylic acids around us" and suggested experiments that are designed to link theoretical knowledge of students with practical skills and bring chemical aspects of everyday life naturally to them.

Occupational diseases of the nervous and musculoskeletal system caused overloading is one of the most common diseases, which belongs to the group occupational diseases. They represent about one-sixth of the number of reported occupational diseases. The number of reported cases is increased, but that there were 7 number of preventive measures, which include the introduction of new technologies, reducing physical work load and other organizational measures. The importance of occupational diseases strain is given only to their proliferation. Although it is mostly a disease from a purely medical point of view rather light, the life of a propagating that the patient more of a nuisance than it was threatened, often lead to exclusion from the employee's current job. Compensation for loss of earnings of these workers for the company financially challenging, and also due to the loss of profits paid to them often until retirement age. It is these social and especially economic reasons are disease as significant for society and for individuals.
