7. Designed analogues of the aplyronines for use in antibody-drug conjugates

Abstract

The aplyronines are a family of structurally complex marine macrolides, isolated from the sea hare Aplysia kurodai, that exhibit picomolar growth inhibitory activity in the NCI 60 cell line panel. An unprecedented heterotrimeric complex has been observed to form between aplyronine A and actin and tubulin together, resulting in disruption of cytoskeletal dynamics and subsequent cell death. This novel mode of action, coupled with their exquisite biological activity, renders the aplyronines compelling candidates for incorporation as payloads into antibody-drug conjugates provided the supply problem can be solved. Guided by structure-activity relationship (SAR) studies, a highly convergent route towards novel and simplified aplyronine structures has been designed. Routes to these function- oriented analogues should be more streamlined and efficient due to judicious structural editing, whilst functionality and potency are retained. Chapter 1 introduces marine natural products and illustrates their therapeutic potential alongside the challenges associated with these highly complex structures. The aplyronine family of natural products is introduced and a function-oriented approach towards analogue design is presented. The avenue of targeted therapies is discussed, with particular focus on the field of antibody-drug conjugates. Chapter 2 outlines the synthesis of our key simplified fragments. Significant optimisation efforts towards the synthesis of the side chain ketone are discussed, together with the synthesis of the northern fragment. Chapter 3 outlines two complementary fragment coupling strategies to access our designed analogues. Contrasting to the established sequence, an alternate approach is examined in which the southern and side chain fragments are first connected before the northern piece is appended. The strengths and shortcomings of each route are reviewed. Chapter 4 culminates in endgame manipulations to generate a range of simplified analogues and details their initial biological evaluation. Preliminary investigations into linker strategies using a quaternary amine approach are investigated. Chapter 5 summarises this work and presents suggestions for future directions of this research for the purpose of developing antibody-drug conjugates.

Analysis of the collected data uncovered common themes, including interpreting and framing the change, professional collaboration, impact of the change on teachers' professional and personal lives, and pacing, communication, and training.

24. Understanding Common Core Implementation: How Educators Intuit, Interpret, and Begin to Integrate Curriculum Reform

Abstract

The purpose of this comparative case study was to explore the ways educators at the school-level experience a federally directed curriculum policy change and examine the contextual factors that impacted the way it was initially implemented. Application of an organizational learning (OL) lens was incorporated to help illuminate the ways educators made sense of and began to implement the new Common Core State Standards. This study investigated the experiences and perspectives of teachers in two North Carolina elementary schools, as they initially implemented the new standards. Qualitative data was gathered through teacher surveys, faculty focus groups and interviews with each school principal and the two district Race to the Top coordinators. Analysis of the collected data uncovered common themes between the experiences at each school, as well as varied contextual factors that impacted implementation. Themes included interpretation of the policy, the role of professional collaboration, professional and personal impacts, time and pacing, and alignment to other initiatives. Comparison to organizational learning frameworks revealed numerous links between teacher experiences and elements of OL theory that helped explain the dynamics at hand. Findings from the study suggest several implications for policy and practice. Federal and state level decisions should consider ways to improve coordination and communication between and among the various levels and move to link federal efforts to the ground level in a more meaningful way. For district practitioners charged with enacting and supporting policy implementation, findings suggest a need for better communication and better designs for the management of training and resources. At the school level, principals and coaches, in the role of both supporting implementation and filtering information, must promote straightforward discussion focused on a clear vision and unambiguous expectations. For classroom teachers implementing Common Core, or any other large scale reform, the findings of this research reveal the importance of embracing change while remaining anchored to the goals and purposes of education at its core. 30. SOME POLITICS ARE STILL LOCAL: STRATEGIC POSITION TAKING IN CONGRESS & ELECTIONS

Abstract

In today's congressional elections, politicians are increasingly presumed to run on the same party-driven platforms, offering voters the same choices throughout the country. Many argue that "local" issues---policy priorities important to a specific constituency---barely register. My dissertation challenges this expectation. I demonstrate that---even though House elections attend more to national issues than before---candidates still often "go local." To measure the degree to which a candidate's campaign is locally-oriented, I employ text data on policy positions extracted from campaign websites for candidates who ran for the U.S. House of Representatives across the 2018 and 2020 elections. Pairing this original data collection with a variety of methods for quantitative text analysis, I show that our theories of strategic candidate behavior must be updated to better reflect what locally-oriented campaigning looks like in today's era of nationalized politics. I go on to demonstrate that politicians who employ locally-oriented rhetoric in their campaigns carry forward this same position taking behavior into the legislative arena. This finding underscores a critical, but underemphasize, continuity between an incumbent's electoral and legislative behavior. In sum, this dissertation aims to refocus the discipline's attention in an era of nationalized expectations back towards local considerations, reminding scholars that local politics are still relevant in modern campaigns.

31. No Experience Required: Early Donations and Amateur Candidate Success in Primary Elections.

Abstract

The electoral dominance of "quality" candidates—political insiders with a history of holding office—is well‐established. However, research on the recent rise in successful political neophytes is less studied. Despite longstanding trends in the predominance of experienced candidates in primary elections, nearly half of all quality candidates who ran in non‐incumbent races lost to a candidate without prior electoral experience in 2018. In this article, we investigate the success of political newcomers in elections for the U.S. House of Representatives by examining a topic often overlooked in the growing literature on primaries: campaign finance. We show that, from 2016 to 2020, political newcomers saw (1) greater success in future fundraising, and (2) an increased likelihood of primary election victory when they garnered more early contributions from outside their district. This contrasts with prior elections, where early money from inside a candidate's own congressional district served as the strongest predictor of future fundraising and electoral success.
