The modified release of active substances such as chlorzoxazone from matrix tablets, based on Kollidon ® SR and chitosan, depends both on the drug solubility in the dissolution medium and on the matrix composition. The aim of this study is to obtain some new oral matrix tablet formulations, based on Kollidon ® SR and chitosan, in order to optimize the low-dose oral bioavailability of chlorzoxazone, a non-steroidal anti-inflammatory drug of class II Biopharmaceutical Classification System. Nine types of chlorzoxazone matrix tablets were obtained using the direct compression method by varying the components ratio as 1:1, 1:2, and 1:3 chlorzoxazone/excipients, 20-40 w/w % Kollidon ® SR, 3-7 w/w % chitosan while the auxiliary substances: Aerosil ® 1 w/w %, magnesium stearate 0.5 w/w % and Avicel ® up to 100 w/w % were kept in constant concentrations. Pharmaco-technical characterization of the tablets included the analysis of flowability and compressibility properties (flow time, friction coefficient, angle of repose, Hausner ratio, and Carr index), and pharmaco-chemical characteristics (such as mass and dose uniformity, thickness, diameter, mechanical strength, friability, softening degree, and in vitro release profiles). Based on the obtained results, only three matrix tablet formulations (F1b, F2b, and F3b, containing 30 w/w % KOL and 5 w/w % CHT, were selected and further tested. These formulations were studied in detail by Fourier-transform infrared spectrometry, X-ray diffraction, thermogravimetry, and differential scanning calorimetry. The three formulations were comparatively studied regarding the release kinetics of active substances using in vitro release testing. The results were analyzed by fitting into four representative mathematical models for the modified-release oral formulations. In vitro kinetic study revealed a complex mechanism of release occurring in two steps of drug release, the first step (0-2 h) and the second (2-36 h). Two factors were calculated to assess the release profile of chlorzoxazone: f1-the similarity factor, and f2-the factor difference. The results have shown that both Kollidon ® SR and chitosan may be used as matrix-forming agents when combined with chlorzoxazone. The three formulations showed optima pharmaco-technical properties and in vitro kinetic behavior; therefore, they have tremendous potential to be used in oral pharmaceutical products for the controlled delivery of chlorzoxazone. In vitro dissolution tests revealed a faster drug release for the F2b sample.

Cobalt(II) chloride (CoCl 2 ) being in the vicinity of polyimide chains entails modifications in terms of the molecular dynamics, which are mainly governed by the possible presence of amic acid residual groups, by the transition-metal-type characteristics of cobalt and by the CoCl 2 content. Polyimide was synthesized using poly(amic acid) according to the reaction of 2,2'-bis(3,4-dicarboxylphenyl)hexafluoropropane dianhydride (6FDA) with 3,3'-dimethyl-4,4'-diaminodiphenylmethane (MMDA) in N,N-dimethylacetamide. CoCl 2 was added before the thermal imidization of the poly(amic acid). An experimental approach was designed to establish the interaction between the polyimide and CoCl 2 and whether the interaction depends on the quantity of the salt. Evidence for the existence of residual amic acid groups was obtained using second derivative Fourier Transform Infrared Spectroscopy (FTIR) and with the help of 2D correlation spectroscopy (2D-COS). Moreover, FTIR, along with X-ray photoelectron spectroscopy (XPS), revealed the interaction between the polymer and CoCl 2 , primarily in the form of Co(II)-N coordinated bonds. Nevertheless, the coordination of cobalt with suitable atoms from the amic acid groups is not precluded. The results of dynamic mechanical analysis (DMA) featured a specific relaxation assigned to the presence of CoCl 2 in the polymeric film and demonstrated that its (non)reinforcing effect depends on its content in the polyimide.

Background: The inovirus Pf4 is a lysogenic bacteriophage of Pseudomonas aeruginosa ( Pa ). People with Cystic Fibrosis (pwCF) experience chronic airway infection with Pa and a significant proportion have high numbers of Pf4 in their airway secretions. Given the known severe damage in the airways of Pa -infected pwCF, we hypothesized a high Pf4 burden can affect airway healing and inflammatory responses. In the airway, basal epithelial cells (BCs) are a multipotent stem cell population critical to epithelium homeostasis and repair. We sought to investigate the transcriptional responses of BCs under conditions that emulate infection with Pa and exposure to high Pf4 burden., Methods: Primary BCs isolated from pwCF and wild-type (WT) donors were cultured in vitro and exposed to Pf4 or bacterial Lipopolysaccharide (LPS) followed by transcriptomic and functional assays., Results: We found that BCs internalized Pf4 and this elicits a strong antiviral response as well as neutrophil chemokine production. Further, we found that BCs that take up Pf4 demonstrate defective migration and proliferation., Conclusions: Our findings are highly suggestive of Pf4 playing a role in the pathogenicity of Pa in the airways. These findings provide additional evidence for the ability of inoviruses to interact with mammalian cells and disrupt cell function.

Liquid biopsies-tests that detect circulating tumor cellular components in the bloodstream-have the potential to transform cancer by reducing health inequities in screening, diagnostics, and monitoring. Today, liquid biopsies are being used to guide treatment choices for patients and monitor for cancer recurrence, and promising work in multi-cancer early detection is ongoing. However, without awareness of the barriers to adoption of this new technology and a willingness to build mitigation efforts into the implementation of widespread liquid biopsy testing, the communities that could most benefit may be the last to access and use them. In this work, we review the challenges likely to affect the accessibility of liquid biopsies in both the general population and underserved populations, and recommend specific actions to facilitate equitable access for all patients.

Pseudomonas aeruginosa is a major human pathogen, particularly effective at colonizing the airways of patients with cystic fibrosis. Bacteriophages are highly abundant at infection sites, but their impact on mammalian immunity remains unclear. We previously showed that Pf4, a temperate filamentous bacteriophage produced by P. aeruginosa , modifies the innate immune response to P. aeruginosa infections via TLR3 signaling, but the underlying mechanisms remained unclear. Notably, Pf4 is a single-stranded DNA and lysogenic phage, and its production does not typically result in lysis of its bacterial host. We identified previously that internalization of Pf4 by human or murine immune cells triggers maladaptive viral pattern recognition receptors and resulted in bacterial persistence based on the presence of phage RNA. We report now that Pf4 phage dampens inflammatory responses to bacterial endotoxin and that this is mediated in part via bacterial vesicles attached to phage particles. Outer membrane vesicles (OMVs) are produced by Gram-negative bacteria and play a key role in host pathogen interaction. Recently, evidence has emerged that OMVs differentially package small RNAs. In this study, we show that Pf4 are decorated with OMVs that remain affixed to Pf4 despite of purification steps. These phages are endocytosed by human cells and delivered to endosomal vesicles. We demonstrate that short RNAs within the OMVs form hairpin structures that trigger TLR3-dependent type I interferon production and antagonize production of antibacterial cytokines and chemokines. In particular, Pf4 phages inhibit CXCL5, preventing efficient neutrophil chemotaxis in response to endotoxin. Moreover, blocking IFNAR or TLR3 signaling abrogates the effect of Pf4 bound to OMVs on macrophage activation. In a murine acute pneumonia model, mice treated with Pf4 associated with OMVs show significantly less neutrophil infiltration in BAL fluid than mice treated with purified Pf4. These changes in macrophage phenotype are functionally relevant: conditioned media from cells exposed to Pf4 decorated with OMVs are significantly less effective at inducing neutrophil migration in vitro and in vivo . In various drug delivery systems, solid lipid nanoparticles are dominantly lipid-based nanocarriers. Amiodarone hydrochloride is an antiarrhythmic agent used to treat severe rhythm disturbances. It has variable and hard-to-predict absorption in the gastrointestinal tract because of its low solubility and high permeability. The aims of this study were to improve its solubility by encapsulating amiodarone into solid lipid nanoparticles using two excipients-Compritol ® 888 ATO (pellets) (C888) as a lipid matrix and Transcutol ® (T) as a surfactant. Six types of amiodarone-loaded solid lipid nanoparticles (AMD-SLNs) were obtained using a hot homogenization technique followed by ultrasonication with varying sonication parameters. AMD-SLNs were characterized by their size distribution, polydispersity index, zeta potential, entrapment efficiency, and drug loading. Based on the initial evaluation of the entrapment efficiency, only three solid lipid nanoparticle formulations (P1, P3, and P5) were further tested. They were evaluated through scanning electron microscopy, Fourier-transform infrared spectrometry, near-infrared spectrometry, thermogravimetry, differential scanning calorimetry, and in vitro dissolution tests. The P5 formulation showed optimum pharmaco-technical properties, and it had the greatest potential to be used in oral pharmaceutical products for the controlled delivery of amiodarone.

Chronic wounds infected by Pseudomonas aeruginosa (Pa) are characterized by disease progression and increased mortality. We reveal Pf, a bacteriophage produced by Pa that delays healing of chronically infected wounds in human subjects and animal models of disease. Interestingly, impairment of wound closure by Pf is independent of its effects on Pa pathogenesis. Rather, Pf impedes keratinocyte migration, which is essential for wound healing, through direct inhibition of CXCL1 signaling. In support of these findings, a prospective cohort study of 36 human patients with chronic Pa wound infections reveals that wounds infected with Pf-positive strains of Pa are more likely to progress in size compared with wounds infected with Pf-negative strains. Together, these data implicate Pf phage in the delayed wound healing associated with Pa infection through direct manipulation of mammalian cells. Increasing interest on sustainable agriculture has led to the development of new materials which can be used as seed coating agents. In this study, a new material was developed based on gelatin film reinforced with cellulose nanocrystals (CNC) which was further used as nanocomposite matrix for Trichoderma harzianum KUEN 1585 spores. The nanocomposite films were characterized by Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD) and scanning electron microscopy (SEM), showing the formation of new hydrogen bonds between the components with a good compatibility between them. Measurements of water contact angles and tests of water vapor sorption and swelling degree revealed an improvement in the water vapor absorption properties of the films as a result of their reinforcement with CNC. Furthermore, by adding the Trichoderma harzianum KUEN 1585 spp. in the seed coating material, the germination percentage, speed of germination and roots length of the corn seeds improved. The polymeric coating did not inhibit the growth of T. harzianum KUEN 1585, with this material being a good candidate in modern agriculture.

KvLQT1 and hERG are the α-subunits of the voltage-gated K + channels which carry the cardiac repolarizing currents IKs and IKr, respectively. These currents function in vivo with some redundancy to maintain appropriate action potential durations (APDs) in cardiomyocytes. As such, protein-protein interactions between hERG and KvLQT1 may be important in normal cardiac electrophysiology, as well as in arrhythmia and sudden cardiac death. Previous phenomenological observations of functional, mutual downregulation between these complementary repolarizing currents in transgenic rabbit models and human cell culture motivate our investigations into protein-protein interactions between hERG and KvLQT1. Previous data suggest that a dynamic, physical interaction between hERG and KvLQT1 modulates the respective currents. However, the mechanism by which hERG-KvLQT1 interactions are regulated is still poorly understood. Phosphorylation is proposed to play a role since modifying the phosphorylation state of each protein has been shown to alter channel kinetics, and both hERG and KvLQT1 are targets of the Ser/Thr protein kinase PKA, activated by elevated intracellular cAMP. In this work, quantitative apFRET analyses of phosphonull and phosphomimetic hERG and KvLQT1 mutants indicate that unphosphorylated hERG does not interact with KvLQT1, suggesting that hERG phosphorylation is important for wild-type proteins to interact. For proteins already potentially interacting, phosphorylation of KvLQT1 appears to be the driving factor abrogating hERG-KvLQT1 interaction. This work increases our knowledge about hERG-KvLQT1 interactions, which may contribute to the efforts to elucidate mechanisms that underlie many types of arrhythmias, and also further characterizes novel protein-protein interactions between two distinct potassium channel families., (Copyright © 2021 Elsevier B.V. Due to ecological reason, in the last period there is an increased interest for changing the synthetic based materials with biodegradable ones. In this study, we prepared bio-nanocomposite formulations based on κ-Carrageenan (κ) and bentonite nanoclay (BT) with different component concentrations (0% BT, 5% BT, 10% BT and 15% BT respectively) and investigated their structural features, with focus on the interactions, sorption properties, and how the combination between them influences these properties. By infrared spectroscopy was identified that in the blending process hydrogen bonds and/or electrostatic interactions may have occurred between BT and κ. The water uptake and water sorption ability decreased with the increasing of the BT content in the formulations, from about 12% for κ to 9% for κ-BT 85:15% and from about 128% for κ to 121% for κ-BT 85:15%, respectively. Water adsorption can significantly decrease if the reinforcement materials are homogenusly dispersed in the matrix. Biobased materials present a great interest due to their properties and biodegradability. Cellulose nanocrystals (CNC) nanofiller, in various amounts, was incorporated into a carboxymethyl cellulose (CMC)-glycerol (G) matrix in order to obtain nanocomposite systems with improved properties. The effect of the nanofiller on the structural features was investigated by Fourier transform infrared (FT-IR) spectroscopy, principal component analysis (PCA), two-dimensional correlation spectroscopy (2D-COS), and X-ray diffraction, while the sorption properties were evaluated by water vapor isotherms using the gravimetric method coupled with infrared spectroscopy. We observed the presence of the interactions taking place between the CMC-G and CNC involving the hydroxyl and carboxylate groups, which decreased the number of water sorption sites. Following this, the moisture content in the nanocomposite films decreased with the increase in the amount of CNC. There is an increased interest on changing the synthetic based materials to biodegradable ones, especially with natural polymers, polysaccharides or proteins. In this research we prepared bio-nanocomposite formulations with different component concentrations and investigated their structural features, with focus on the interactions, sorption properties, and how the combination between them influences these properties. By infrared spectroscopy, principal component analysis (PCA) and two-dimensional correlation spectroscopy (2DCOS) was identified that in the blending process are involved the -SO 4 and C(4)-O-S groups of β-d-galactose, CO groups, (O=S=O) of carrageenan and -OH and CO groups from CNCs. The water uptake and water sorption properties decrease with increasing the CNCs content in the formulations from about 15% for κ to 10% for κC15 and from about 128% for κ to 115% for κC15, respectively. The increase of the CNCs content induced an increase of the water contact angle from 47° for κ to 90° for κC15, indicating once again the involvement of the free hydroxyl groups in the hydrogen bonded interactions., (Copyright © 2019 Elsevier B.V. Nanocomposite films containing cellulose nanocrystals (CNC), as the reinforcing phase, and poly(vinyl alcohol) (PVA)/starch (S) blend, as the matrix, were prepared by solution casting technique. For the preparation, 5, 10, 15 wt% CNC suspension were incorporated in blends containing 90 wt% PVA/10 wt% S, 75 wt% PVA/25 wt% S and 50 wt% PVA/50 wt% S. Structural evaluation was done by Fourier transform infrared spectrometry and X-ray diffraction, and the morphological aspects by atomic force microscopy. Infrared spectroscopy evidenced the presence of H-bond interactions between the PVA and S and between PVA/S blend and CNC, and also variation in the conformational rearrangement, while the X-ray diffraction showed that the crystallite size and the degree of crystallinity were strongly affected by blending of PVA with S and also by the incorporation of CNC in the nanocomposite films. Novel microarray platform for single nucleotide polymorphisms (SNPs) detection has been developed using silicon nanowires (SiNWs) as support and two different surface modification methods for attaining the necessary functional groups. Accordingly, we compared the detection specificity and stability over time of the probes printed on SiNWs modified with (3-aminopropyl)triethoxysilane (APTES) and glutaraldehyde (GAD), or coated with a simpler procedure using epoxy-based SU-8 photoresist. Scanning electron microscopy (SEM) and energy dispersive X-ray spectroscopy (EDX) were used for comparative characterization of the unmodified and coated SiNWs. The hybridization efficiency was assessed by comprehensive statistical analysis of the acquired data from confocal fluorescence scanning of the manufactured biochips. The high detection specificity between the hybridized probes containing different mismatch types was demonstrated on SU-8 coating by one way ANOVA test (adjusted p value *** < .0001). The stability over time of the probes tethered on SiNWs coated with SU-8 was evaluated after 1, 4, 8 and 21 days of probe incubation, revealing values for coefficient of variation (CV) between 2.4% and 5.6%. The signal-to-both-standard-deviations ratio measured for SU-8 coated SiNWs platform was similar to the commercial support, while the APTES-GAD coated SiNWs exhibited the highest values., (Copyright © 2018 Elsevier B.V. Spruce wood samples were subjected to different conditions of thermal and hydro-thermal treatment by varying the temperature, relative humidity and period of exposure. The obtained treated samples were evaluated using near infrared spectroscopy (NIR), principal component analysis (PCA) and hierarchical cluster analysis (HCA) in order to evidence the structural changes which may occur during the applied treatment conditions. Following this, modification in all wood components were observed, modifications which were dependent on the temperature, amount of relative humidity and also the treatment time. Therefore, higher variations were evidenced for samples treated at higher temperatures and for longer periods. At the same time, the increase in the amount of water vapours in the medium induced a reduced rate of side chains and condensation reactions occurring in the wood structure. The aim of this study is to conduct an extended surface and cross-section characterization of a denture base acrylic resin subjected to 500, 650, and 750 W microwave irradiation for 2, 3, and 5 min to assess its morphological modifications. A commercial heat-cured powder was polymerized according to the manufacturer's specifications and distributed into 20 circular samples. A stainless-steel wire was partially embedded in half of the discs, in order to investigate the metal-polymer interface. High-resolution scanning electron microscopy (SEM) imaging, white light interferometry, roughness measurements and Fourier transform infrared spectrometry were employed for morphological and structural evaluation of the irradiated polymer. Superficial adaptation was discovered after 5 min exposure at 500 W, 650 W, and 750 W, revealing significant roughness correction for 750 W. SEM characterization revealed the inner alteration of the resin for the 750 W protocol and a metal-polymer gap developed regardless of the irradiation conditions. The considerable temperature fluctuations that the samples were subject to during the experiments did not essentially change the poly(methyl-methacrylate) bond structure.

Bio-nanocomposite films based on cellulose nanocrystals reinforced poly(vinyl alcohol) were obtained by solvent casting method. To assess the structural features of the films, different spectral techniques (FTIR, 2D COS and XRD) have been used. Infrared and 2D correlation spectroscopy evidenced the presence of H-bond interactions between the PVA and CNC, and the variation in the conformational rearrangements, while XRD showed that the crystallite size and the crystallinity degree were affected by the incorporation of CNC. At low content of CNC in the PVA matrix, the crystallinity degree decreased to 29.9%, while at higher CNC content increased to 80.6%, comparing to PVA (35.4%). To evaluate the interaction with water, contact angle measurement, water sorption and NIR spectroscopy were used, respectively. The increase of the CNC content induced a reduction in water sorption ability from 93% for PVA to 75% for PVA/CNC films, indicating the involvement of the hydroxyl groups in new hydrogen bonded interactions. By analyzing the variation of the NIR bands from 1930, 1902 and 1985nm, was observed that the water molecules interact with the polymer matrix through moderate hydrogen bond before diffusing into the free volume of the matrix and form stronger hydrogen bonds., (Copyright © 2017 Elsevier B.V. The combination of various mobile, non-invasive techniques (IR reflectography technique, optical microscopy, XRF, Raman and NIR spectroscopies) and lab-based devices (FTIR and XPS spectroscopies, SEM-EDX microscopy) lead to the first exhaustive investigation of pigments and materials used by the famous Romanian painter Nicolae Grigorescu in three cultural heritage paintings. As selenium is an important part of the antioxidant enzymes and also because there are several studies suggesting a possible link between cancer and selenium deficiency, this paper presents a spectrophotometric method for the assay of Se(IV), using N,N-diethyl-p-phenylenediamine monohydrochloride as reagent. The proposed method is based on the reaction between the selenium and potassium iodide in low acidic medium, when iodine is released. This last product will further oxidise the new reagent. The final obtained product is strongly coloured in red and has an absorption maximum at 552 nm and molar extinction coefficient (ε) of 6.1 × 10(4) L mol(-1) cm(-1). The optimum working conditions were established, and the developed method was validated, being characterised by a good linearity (in the range of 0.5-3.0 μg/mL), a limit of detection (0.0573 μg/mL) and a limit of quantification (0.1737 μg/mL). At the same time, the repeatability, the precision of the method and the accuracy were established. The proposed and validated method was applied with good results for the determination of Se(IV) in spring and bottled water from Iasi and also in pharmaceutical and cosmetic products.

An investigation of the constitutional adaptive gelation process of chitosan/cinnamaldehyde (C/Cy) dynagels is reported. These gels generate timely variant macroscopic organization across extended scales. In the first stage, imine-bond formation takes place "in-water" and generates low-ordered hydrogels. The progressive formation of imine bonds further induces "out-of-water" increased reactivity within interdigitated hydrophobic self-assembled layers of Cy, with a protecting environmental effect against hydrolysis and that leads to the stabilization of the imine bonds. The hydrophobic swelling due to Cy layers at the interfaces reaches a critical step when lamellar self-organized hybrids are generated (24 hours). This induces an important restructuration of the hydrogels on the micrometric scale, thus resulting in the formation of highly ordered microporous xerogel morphologies of high potential interest for chemical separations, drug delivery, and sensors., (© 2014 WILEY-VCH Verlag GmbH & Co. The modifications and/or degradation of lime (Tillia cordata) wood components during wood heat treatment under low temperature at about 140°C and 10% percentage of relative humidity were evaluated. The aim of this study was to obtain results by simple NIR coupled with second derivative, principal component analysis and two dimensional correlation spectroscopy in order to better understand how these techniques are able to evaluate structural differences resulted under hydro-thermal treatment of the wood over a period of 504h. The NIR spectra of treated samples were compared with the reference one. Due to the broad bands in the NIR spectra, the assignment and modifications occurring during treatment is difficult, therefore the second derivative principal component analysis were applied. Principal component analysis by first two components was able to differentiate the samples series, PC1 being considered as the time axis, and PC2 as the axis representing the structural modification of wood components. 2D NIR correlation spectroscopy was able to estimate the sequential order of the groups variations under the hydro-thermal treatment time as external perturbation, indicating as first moment changes the OH and CO groups from carbohydrates and lignin, followed by CarH, CH and CH2 groups from lignin, cellulose and hemicelluloses., (Copyright © 2013 Elsevier B.V. The obtaining of bio-polymer dispersed liquid crystal (bio-PDLC) systems based on a chitosan polymer matrix is reported here for the first time. The new PDLC composites have been obtained by encapsulation of 4-cyano-4'-pentylbiphenyl (5CB) as low molecular weight liquid crystal into chitosan, and they have been characterized by polarized optical microscopy, differential scanning calorimetry, electron and transmission scanning microscopy, Raman and fluorescence spectroscopy. Submicrometric liquid crystalline droplets with uniform size distribution and density have been obtained for low liquid crystal content into the PDLCs. This paper reports the morphological and thermal characterization of xanthan/lignin hydrogels. It has been emphasized the effect of the lignin type on the hydrogel properties. The hydrogels described here were obtained by chemical crosslinking, in the presence of epichlorohydrine as a cross-linker agent. The obtained materials were analyzed by AFM, TG/DTG, DSC, and FT-IR spectroscopy. It has been established that hydrogels have a porous morphology. The lignin type influences the hydrogel morphology which is either fibrilar as in case of hydrogel containing aspen wood lignin (which has the highest content of COOH groups and lowest content of phenolic OH groups) or smooth surface for other hydrogels. The specific intermolecular interactions are stronger in the case of 70 xanthan (X)/30 aspen wood lignin (AWL) hydrogel. The FTIR spectra were measured for G1 allyl terminated carbosilane dendrimer over a temperature range of 25-200°C to explore the temperature-dependent changes. The dendrimer spectra exhibited complicated patterns and thus were analyzed by both the exploratory principal component analysis (PCA) and two-dimensional (2D) correlation spectroscopy. Boltzmann fitting and score plots of PCA evidenced the phase transition temperature to be about 172°C. Frequency variables, which strongly contributed to each principal component highlighted in their loadings plot, were linked to the frequencies assigned to vibrations of the CH(2) and CH(3) groups, as well as to vibrations of the SiCH groups. 2D correlation spectroscopy discerned all the sequences of group motion of dendrimer, indicating that during the heating process a reorganization of core structure takes place, followed by a conformational rearrangement of the terminal units., (Copyright © 2012 Elsevier B.V. Generalized two-dimensional infrared (2D IR) correlation spectroscopy has been applied to the study of the conformational changes and specific interactions in blends of polyethylene adipate (PEA) and cholesteryl palmitate (CP). IR spectra of CP, PEA, and their blends of different compositions: 10/90, 16/84, 32/68, 64/36, and 80/20 have been recorded. In order to apply 2D IR correlation analysis, the samples are divided into two sets: set A with high PEA content 0/100, 10/90, 16/84, 32/68 CP/PEA and set B with high CP content 64/36, 80/20 and 100/0 CP/PEA. The 2D IR synchronous correlation analysis separates the bands of PEA from those of CP. The cross-peaks in 2D IR asynchronous correlation analysis are indicative of the specific interaction or the conformational change in the blends. The bands of CO, OH and C-O vibrations of PEA, and CH(3) and C-O vibrations of CP that are very sensitive to the intermolecular hydrogen bonding effect and consequently the partial miscibility of components, have been assigned by 2D correlation analysis., (Copyright © 2011 Elsevier B.V. In this study the weathering behavior of lime wood (Tillia cordata Mill.) has been examined using FT-IR and 2D IR correlation spectroscopy, which evidenced chemical changes induced by exposure to weathering conditions. It was showed that lignin is most sensitive component to the photodegradation processes as indicated by considerable decreases in the intensities of the characteristic aromatic lignin band at 1505cm(-1) and other associated bands. By 2D correlation spectroscopy has been demonstrated that the moment of CO from carboxyl and acetyl groups in hemicelluloses is changing first, followed by the CC of aromatic skeletal, CO in non-conjugated ketones, carboxyl groups and lactones, absorbed O-H and conjugated C-O groups in quinones. The carbonyl formation corresponded well with lignin degradation, indicating a close relationship between them. Comparing the rate of carbonyl formation and lignin decay clearly showed that the former is remarkably higher than the latter, indicating the formation of carbonyl bands at 1738cm(-1) probably resulted from not only lignin oxidation but also from reactions occurring in other components of the wood. Molecular mechanisms involved in the replenishment of the fast neurotransmitters glutamate and GABA are poorly understood. Glutamine sustains their generation. However, glutamine formation from the recycled transmitters is confined to glial processes and requires facilitators for its translocation across the glial and neuronal membranes. Indeed, glial processes are enriched with the system N transporter SN1 (Slc38a3), which, by bidirectional transport, maintains steady extracellular glutamine levels and thereby furnishes neurons with the primary precursor for fast neurotransmitters. We now demonstrate that SN1 is phosphorylated by protein kinase Cα (PKCα) and PKCγ. Electrophysiological characterization shows that phosphorylation reduces V(max) dramatically, whereas no significant effects are seen on the K(m). Phosphorylation occurs specifically at a single serine residue (S52) in the N-terminal rat (Rattus norvegicus) SN1 and results in sequestration of the protein into intracellular reservoirs. Prolonged activation of PKC results in partial degradation of SN1. These results provide the first demonstration of phosphorylation of SN1 and regulation of its activity at the plasma membrane. Interestingly, membrane trafficking of SN1 resembles that of the glutamate transporter GLT and the glutamate-aspartate transporter GLAST: it involves the same PKC isoforms and occurs in the same glial processes. This suggests that the glutamate/GABA-glutamine cycle may be modified at two key points by similar signaling events and unmasks a prominent role for PKC-dependent phosphorylation. Our data suggest that extracellular glutamine levels may be fine-tuned by dynamic regulation of glial SN1 activity, which may impact on transmitter generation, contribute to defining quantal size, and have profound effects on synaptic plasticity.

Cold plasma treatment is used to modify the cellulosic fibers for a variety of applications. The grafting of softwood unbleached (UBP) and bleached (BP) kraft pulp fibers has been performed under the action of cold plasma discharges, using different kinds of fatty acids. The grafted samples are characterized by FTIR spectroscopy, X-ray photoelectron spectroscopy (XPS), scanning electron microscopy (SEM), differential scanning calorimetry (DSC), termogravimetry (TG-DTG) and X-ray diffraction (XRD). All these methods confirm the morphological and structural changes after plasma treatment which determines the modification in cellulosic fiber properties. The active centers created within the cellulose chains by plasma treatment were used to initiate grafting reactions with fatty acids. In this paper biomaterials based on various polyurethane formulations have been physically characterized by FT-IR spectroscopy, contact angle measurements, DSC, TG/DTG and SEM methods. It has been established that the transition temperatures of soft and hard segments of polyurethane (glass transition or melting) depend on the blend composition. The melting temperature varies from 54.2 to 81.9°C for soft segments, and from 220 to 235°C for hard segments. FT-IR spectrometry allows identifying the functional groups involved in interactions and consequently the changes in polymer chain mobility. From SEM images, is evident that polyurethanic film is porous and spongious. By adding of the others components such as hydrolyzed collagen, elastin, chondroitin sulfate or hyaluronic acid, a reduction of porosity of films was obtained., (Copyright © 2010 Elsevier B.V. Fourier transform infrared (FT-IR) spectroscopy combined with 2D correlation spectroscopy has been used to offer some information about stability and structure of some soluble elastins. Temperature has been chosen as the perturbation to monitor the infrared behavior of various soluble elastins, namely, α-elastin p, α-elastin, and k-elastin. In the 3800-2700 cm⁻¹ region, the H-containing groups were analyzed. The bonded hydroxyls are found to decrease prior to the NH-related hydrogen bonds and also to the conformational reorganization of hydrocarbon chains. The transition temperatures were evaluated and they were found to agree with those obtained from DSC data. The FTIR spectra and their 2nd derivatives denote that α- elastins exhibited amide-I, -II and -III bands at 1656, 1539 and 1236 cm⁻¹, respectively, while in k-elastin these bands were found at 1652 cm⁻¹ for amide I, 1540 cm⁻¹ for amide II and 1248 cm⁻¹ for amide III. The macroscopic IR finger-print method, which combines: general IR spectra, secondary derivative spectra, and 2D-IR correlation spectra, is useful to discriminate different elastins. Thus using the differences of the position and intensity of the bands from "fingerprint region" of studied elastins, which include the peaks assigned to C==O, C--C groups from α-helix, β-turn, and the peaks assigned to the amide groups, it is possible to identify and discriminate elastins from each others. The main difficulties in wood and pulp analyses arise principally from their numerous components with different chemical structures. Therefore, the basic problem in a specific analytical procedure may be the selective separation of the main carbohydrate-derived components from lignin due to their chemical association and structural coexistence. The processing of the wood determines some structural modification in its components depending on the type of wood and the applied procedure. Fourier transform infrared (FT-IR) spectrometry and X-ray diffraction have been applied to analyze Eucalyptus g. wood chips and unbleached and chlorite-bleached pulp. The differences between samples have been established by examination of the spectra of the fractions obtained by successive extraction (acetone extractives, acetone free extractive samples, hemicelluloses, and lignins) by evaluating the derivative spectra, band deconvolution, etc. The energy and the hydrogen bonding distance have been evaluated. The relationship between spectral characteristics and sample composition has been established, as well as the variation of the degree of crystallinity after pulping and bleaching. The integral absorption and lignin/carbohydrate ratios calculated from FT-IR spectra of the IR bands assigned to different bending or stretching in lignin groups are stronger in the spectrum of eucalyptus chips than those from brown stock (BS) pulp spectra because of the smaller total amount of lignin in the latter. FT-IR spectra clearly show that after chlorite bleaching the structure of the wood components is partially modified or removed. Along with FT-IR data, the X-ray results confirmed the low content of lignin in the pulp samples by increasing the calculated values of the crystalline parameters. It was concluded that FT-IR spectroscopy can be used as a quick method to differentiate Eucalyptus globulus samples.

Clinical studies suggest that treatment with vaccines comprised of idiotype protein may be associated with improved clinical outcome in follicular lymphoma patients. The time-consuming process required to generate patient-specific vaccines is a major limitation, however. Here we report results of a pilot clinical trial with a novel autologous, tumor-derived proteoliposome vaccine formulation that could be rapidly produced within a single day. Vaccination was safe, induced autologous tumor-specific type 1 cytokine responses in 5 out of 10 follicular lymphoma patients, and was associated with induction of a sustained complete response in one patient. Other patients had large tumor burdens and progressed after a median duration of 8 months. These results suggest that further testing of this vaccine formulation, particularly in the setting of minimal disease, is warranted. Furthermore, the proteoliposome formulation may provide a model for vaccine development for other human cancers, for which tumor-associated antigens need not be defined.

Therapeutic vaccination against idiotype is a promising strategy for immunotherapy of B-cell malignancies. Its feasibility, however, is limited by the requirement for a patient-specific product. Here we describe a novel vaccine formulation prepared by simply extracting cell-membrane proteins from lymphoma cells and incorporating them together with IL-2 into proteoliposomes. The vaccine was produced in 24 hours, compared with more labor-intensive and time-consuming hybridoma or recombinant DNA methods. The vaccine elicited T-cell immunity in vivo, as demonstrated by secretion of type 1 cytokines. It protected against tumor challenge at doses of tumor antigen 50 to 100 times lower than that previously observed using either liposomes formulated with IL-2 and secreted lymphoma immunoglobulin or a prototype vaccine consisting of lymphoma immunoglobulin conjugated to keyhole limpet hemocyanin. The increased potency justifies testing similar patient-specific human vaccines prepared using extracts from primary tumor samples.

FT-IR and 2D correlation spectroscopy were employed to study the microstructural changes occurring during phase transitions of a liquid crystal poly(amidoamine) codendrimer (PAMAM (L1)16(L2)16) generation 3, functionalized on the terminal groups by one-chain promesogenic calamitic units (4-(4'-decyloxybenzoyloxy)salicylaldehyde (L1)) and two-chain promesogenic calamitic units (4-(3',4'-didecyloxybenzoyloxy)salicylaldehyde (L2)). Spectral modifications associated with molecular conformation rearrangements allowing for molecular shape change on going from a liquid-crystalline organization to another were found. The transition temperatures were calculated, and they are in good agreement with the DSC data. Spectral analysis gives evidence of the LC phase transitions and to an additional transition associated with the existence of conformers. Various types of hydrogen bonding have been established.

Purpose: The idiotype (Id) of the immunoglobulin on a given B-cell malignancy is a clonal marker that can serve as a tumor-specific antigen. We developed a novel vaccine formulation by incorporating Id protein with liposomal lymphokine that was more potent than a prototype, carrier-conjugated Id protein vaccine in preclinical studies. In the present study, we evaluated the safety and immunogenicity of this vaccine in follicular lymphoma patients., Experimental Design: Ten patients with advanced-stage follicular lymphoma were treated with five doses of this second generation vaccine after chemotherapy-induced clinical remission. All patients were evaluated for cellular and humoral immune responses., Results: Autologous tumor and Id-specific type I cytokine responses were induced by vaccination in 10 and 9 patients, respectively. Antitumor immune responses were mediated by both CD4+ and CD8+ T cells, were human lymphocyte antigen class I and II associated, and persisted 18 months beyond the completion of vaccination. Specific anti-Id antibody responses were detected in four patients. After a median follow-up of 50 months, 6 of the 10 patients remain in continuous first complete remission., Conclusions: This first clinical report of a liposomal cancer vaccine demonstrates that liposomal delivery is safe, induces sustained tumor-specific CD4+ and CD8+ T-cell responses in lymphoma patients, and may serve as a model for vaccine development against other human cancers and infectious pathogens.

Cancers appear to escape surveillance by the immune system at least in part because they fail to induce a protective immune response. Therapeutic vaccines based on specific tumour antigens and tumour cells modified ex vivo by genetic techniques are but two strategies being used to circumvent this problem. In this report, we describe a simple, yet effective alternative in which tumour-specific responses are induced by in situ administration of a well-characterized liposomal formulation of the cytokine interleukin 2 (IL-2). Using the non-immunogenic B16 melanoma model, intratumoural injections of liposomal IL-2 L(IL2), were shown to induce a long-lived immune response specific for the injected tumour. In conjunction with subsequent removal of the primary tumours by surgery, the injections increased mean survival to 57 days from a control value of 32 days and partially protected surviving mice against re-challenge with B16. L(IL2) induced an early infiltration of inflammatory cells within the tumours which was followed several days later by an influx of CD3+ T cells. The cellular influx and a coincident decrease in tumour growth were noted in both injected tumours and a second non-injected tumour on the same animal, thereby demonstrating the systemic nature of the immune response. Intratumoural injections of soluble IL-2 at the same dose failed to induce B16-specific cellular immunity or to prolong survival of the mice. A liposomal Muc1 mucin vaccine for treatment of adenocarcinomas was formulated by incorporating a synthetic Muc1 mucin-based lipopeptide and Lipid A into a DPPC/cholesterol bilayer. Vaccination of mice with the liposomal formulation produced a peptide-specific immune response dependent on the cholesterol content. The response occurred at a threshold of 20-23 mol% cholesterol, and was optimal at cholesterol levels of > or =30 mol%. To understand this cholesterol dependency, we studied the effect of cholesterol on the liposomal bilayer and surface properties. Freeze-fracture electron microscopy showed a unique surface texture that was codependent upon cholesterol (> or =20 mol%) and lipopeptide content. Fluorescence anisotropy measurements exhibited a significant decrease in the rotational motion of 1,6-diphenyl-1,3,5-hexatriene in formulations containing >20 mol% cholesterol and only in the presence of the lipopeptide. At 20 mol% cholesterol and with lipopeptide, DSC showed a significant increase in the main phase transition of the DPPC bilayers, while Raman spectroscopy indicated a more ordered arrangement of DPPC molecules compared to control liposomes containing DPPC/cholesterol alone. Taken together, the data suggest the presence of lipopeptide-rich microdomains at and above a threshold of 20 mol% cholesterol that may play a role in the induction of a peptide-specific immunological response.

Recombinant human interleukin-2 (rhIL-2) was incorporated in liposomes for potential therapeutic applications using a novel process. In this process, rhIL-2 caused the formation of large, unique multilamellar vesicles (MLVs) from small unilamellar vesicles (SUVs) of dimyristoylphosphatidylcholine (DMPC). Vesicle coalescence occurred most rapidly at 19 degrees C, between the pre- and main phase transition temperatures of DMPC, and showed a dependence upon pH (pH <5.5), ionic strength (>50 mM) and the initial size of the unilamellar vesicles (

Oncolipin is a multilamellar liposomal (dimyristoyl phosphatidylcholine) formulation of interleukin 2 (IL-2) and human serum albumin (HSA) with distinct surface characteristics which may influence its biological activities. IL-2 and HSA were detected on the surface of the liposomes using specific antibody staining. Surface expression of IL-2 was also demonstrated by the observation that Oncolipin bound to cells expressing IL-2 receptors (IL-2R) containing alphabetagamma or betagamma subunits. Binding and internalization of Oncolipin by cells expressing alphabetagamma or betagamma receptor subunits was blocked by excess free IL-2 or a neutralizing antibody against the beta chain. The display of surface IL-2 on Oncolipin's liposomes was maintained in vivo after intravenous injection into mice. IL-2 was also present between the lipid bilayers of the multilamellar liposomes based on the unique physical characteristics detected by freeze fracture electron microscopy. The bulk of the liposome-associated IL-2 was released from the liposomes upon incubation at 37 degrees C in medium containing serum, indicating that the IL-2 was not irreversibly entrapped on or in the liposome structure. Thus, Oncolipin is receptor-targeted to activated T and NK cells by virtue of its surface expression of IL-2 and has the potential to release IL-2 following deposition within lymphoid organs. A novel method was developed to determine the pharmacokinetics and biodistribution of cytokines and lymphokines based on time-resolved fluorometry (TRF) of europium (Eu). The comparison of two formulations of IL-2 was used to illustrate the sensitivity and applicability of this method as well as to extend the information on the pharmacokinetics of liposomal IL-2 and soluble IL-2. The blood kinetics and biodistribution of liposomal and soluble IL-2 in lymphoid organs and kidneys as measured by TRF were similar to those determined by the radioisotopic method. In both instances, the formulation of IL-2 into liposomes increased its serum half-life and accumulation in reticuloendothelial and lymphoid organs. The increased sensitivity of the Eu/TRF method permitted the extension of observational time points and the analysis of biodistribution in organs such as lymph nodes and bone marrow. These results suggest that Eu-labelled proteins in conjunction with TRF offer a suitable alternative to radiolabelled proteins for pharmacokinetics and tissue distribution studies in animals. This method offers distinct advantages over traditional techniques employing radioistopes since it has greater sensitivity, no half-life limitations and no radioactive or hazardous waste disposal., (Copyright 2000 Academic Press.)

We developed a liposome carrier for a model nonimmunogenic, self Ag. This carrier reproducibly converted lymphoma Ig into a potent tumor rejection Ag in mice. A single immunization induced protection against challenges representing 20 to 100 times the minimum lethal dose of parental tumor. This protective effect required minimal amounts of incorporated Ag and IL-2 and elicited specific Abs (compared with free Ag or liposomal control Ig which did not elicit any specific Abs); depletion experiments demonstrated a requirement for effector CD4+ and CD8+ T cells. Head-to-head comparisons, indicating superior potency and induction of specific T cell activation, distinguished liposomal from prototype, carrier-conjugated Ag. These results provide a strategy for formulating weak tumor or other clinically important Ags into vaccines.

When interacting with phospholipid in an aqueous environment, amphotericin B forms unusual structures of markedly reduced toxicity (Janoff et al. (1988) Proc. Natl. Acad. Sci. USA 85, 6122-6126). These structures, which appear ribbon-like by freeze-fracture electron microscopy (EM), are found exclusively at amphotericin B to lipid mole ratios of 1:3 to 1:1. At lower mole ratios they occur in combination with liposomes. Circular dichroism (CD) spectra revealed two distinct modes of lipid-amphotericin B interaction, one for liposomes and one for the ribbon-like structures. In isolated liposomes, amphotericin B which comprised 3-4 mole percent of the bulk lipid was monomeric and exhibited a hemolytic activity comparable to amphotericin B suspended in deoxycholate. Above 3-4 mole percent amphotericin B, ribbon-like structures emerged and CD spectra indicated drug-lipid complexation. Minimal inhibitory concentrations for Candida albicans of liposomal and complexed amphotericin B were comparable and could be attributed to amphotericin a release as a result of lipid breakdown within the ribbon-like material by a heat labile extracellular yeast product (lipase). Negative stain EM of the ribbon-like structures indicated that the ribbon-like appearance seen by freeze-fracture EM arises as a consequence of the cross-fracturing of what are aggregated, collapsed single lamellar, presumably interdigitated, membranes. Studies examining complexation of amphotericin B with either DMPC or DMPG demonstrated that headgroup interactions played little role in the formation of the ribbon-like structures. With these results we propose that ribbon-like structures result from phase separation of amphotericin B-phospholipid complexes within the phospholipid matrix such that amphotericin B release, and thus acute toxicity, is curtailed. Formation of amphotericin B-lipid structures such as those described here indicates a possible new role for lipid as a stabilizing matrix for drug delivery of lipophilic substances, specifically where a highly ordered packing arrangement between lipid and compound can be achieved.

This report describes the properties of a novel sustained-release drug delivery system comprising liposomes sequestered in a collagen gel. Two peptide hormones, insulin and growth hormone encapsulated in vesicles sequestered within the matrix, are slowly released into the circulation from either an intramuscular or subcutaneous injection site. A maximum 3-5-d release for insulin or a 14-d growth hormone release was observed. Enhanced sequestration of liposomes with the collagen can be achieved by modifying the liposome surface with fibronectin. The liposome gel delivery system appears to offer several advantages over other liposome formulations or gel formulations constructed only with free drug.

Thirty cows naturally infected with Brucella abortus were treated by various routes, using free or liposomal streptomycin or a combination of liposomal streptomycin and a long-acting oxytetracycline preparation. Of 21 cows treated with liposomal streptomycin alone, 3 (14%) were culture negative and 3 had 10 or fewer bacterial colonies isolated from tissues obtained at necropsy. Thirteen (62%) cows continued to shed organisms in udder secretions and were considered treatment failures. Of 9 cows that were given a combination of liposomal streptomycin and long-acting oxytetracycline, 5 (56%) were cured, 3 had 10 or fewer colonies on culture plates of tissue after necropsy and only 1 continued to shed B abortus in udder secretions after treatment. Eleven cows were given streptomycin liposomes by intramammary infusion with or without IM administration of long-acting oxytetracycline. The most effective regimen consisted of 2 intramammary infusions of streptomycin liposomes and 2 doses of oxytetracycline administered IM. Of 5 cows treated thusly, 2 were cured and all others had fewer than 10 B abortus colonies isolated from tissues obtained at necropsy.

Increased synthesis of prostaglandins (PGs) may be an important promoting factor in the development of lymphosarcoma. Thus, inhibition of PG synthesis may represent a logical way of controlling the growth of lymphosarcomatous cells. In order to verify this hypothesis, two groups of rats with lymphosarcoma have been used: the test-group received acetyl salicylic acid (54 mg/animal/day) and the control-group received the same quantity of physiological saline solution. After 4 days of treatment, the lymph nodes stopped growing in the test-group. Microscopic and electron microscopic examinations of lymph node sections were carried out after 11 days of treatment. These showed for the test-group the disappearance of the characteristics of malignancy.

The potential application of liposomes to drug delivery has been apparent since 1965, when these phospholipid vesicles were first described by Bangham. Since then, experiments on animals have shown that liposome encapsulation can dramatically alter the distribution of drugs in the body and their rate of clearance. These pharmacokinetic differences, as well as other less well-understood effects, can result in reduced toxicity and enhanced efficacy of the encapsulated drug. The vast majority of studies on the therapeutic use of liposomes have involved the delivery of drugs used in cancer chemotherapy and metabolic storage diseases, but there is now more literature on the use of liposomes for the delivery of antimicrobial drugs and immunomodulating agents. This review briefly discusses the general properties of liposomes and the rationale for their use in antimicrobial drug delivery and immunomodulation, as well as the encapsulation of specific agents and the effect of encapsulation on the treatment of infectious diseases.

Ribbon-like structures result when amphotericin B interacts with lipid in an aqueous environment. At high ratios of amphotericin to lipid these structures, which are lipid-stabilized amphotericin aggregates, become prevalent resulting in a dramatic attenuation of amphotericin-mediated mammalian cell, but not fungal cell, toxicity. Studies utilizing freeze-etch electron microscopy, differential scanning calorimetry, 31P NMR, x-ray diffraction, and optical spectroscopy revealed that this toxicity attenuation is related to the macromolecular structure of the complexes in a definable fashion. It is likely that amphotericin in this specific form will have a much improved therapeutic utility.
