Abstract

Challenging neutron-capture cross-section measurements of small cross sections and samples with a very limited number of atoms require high-flux time-of-flight facilities. In turn, such facilities need innovative detection setups that are fast, have low sensitivity to neutrons, can quickly recover from the so-called $\gamma$-flash, and offer the highest possible detection sensitivity. In this paper, we present several steps toward such advanced systems. Specifically, we describe the performance of a high-sensitivity experimental setup at CERN n\_TOF EAR2. It consists of nine sTED detector modules in a compact cylindrical configuration, two conventional used large-volume C$_{6}$D$_{6}$ detectors, and one LaCl$_{3}$(Ce) detector. The performance of these detection systems is compared using $^{93}$Nb($n$,$\gamma$) data. We also developed a detailed \textsc{Geant4} Monte Carlo model of the experimental EAR2 setup, which allows for a better understanding of the detector features, including their efficiency determination. This Monte Carlo model has been used for further optimization, thus leading to a new conceptual design of a $\gamma$ detector array, STAR, based on a deuterated-stilbene crystal array. Finally, the suitability of deuterated-stilbene crystals for the future STAR array is investigaged experimentally utilizing a small stilbene-d12 prototype. The results suggest a similar or superior performance of STAR with respect to other setups based on liquid-scintillators, and allow for additional features such as neutron-gamma discrimination and a higher level of customization capability.

3. Estimating angular momenta of fission fragments from isomeric yield ratios

Abstract

Purpose: To deduce the angular momenta of fission fragments based on the observed isomeric yield ratios (IYR) in 25-MeV proton-induced fission of 238U and to compare these using Wilson's model. Method: A surrogate model of GEF has been developed to generate properties of primary fission fragments. Based on the excitation energy and angular momentum of fission fragments from GEF, an energy versus angular momentum matrix is reconstructed using a set of parameters. With such matrices as input, TALYS is used to calculate the de-excitation of the fission fragments, from which the IYRs are obtained. By varying one of the parameters, the root-mean-square angular momentum (Jrms), which determines the angular momentum distribution of the matrix, Jrms-dependent IYRs are obtained. Considering all primary fission fragments contributing to the IYR for a given fission product, the average angular momentum of those fragments is estimated. Results: Data of 31 IYRs in proton-induced fission of 238U were analysed. As a result, the average Jrms, equivalent to average angular momentum Jav, with uncertainties of 24 fission products, are presented. Considering the neutron emissions of the primary fission fragments, the Jav as a function of the primary fission fragment is presented. A mass dependency of Jav is observed in the proton-induced fission of 238U. Moreover, the Jav for A larger than 131 could be described by the parameterisation proposed by J. Wilson. In general, higher Jav are observed in the present work compared to those from Wilson et al. This is likely due to the higher excitation energy of the fissioning nuclei in this work compared to Wilson's. Furthermore, systematic measurements of the Jav of fission products in the symmetric mass region are presented for the first time. A decreasing trend with mass numbers is observed, which can not be explained by the proposal in Wilson's paper.

4. Pretreatment with IL-15 and IL-18 rescues natural killer cells from granzyme B-mediated apoptosis after cryopreservation.

Abstract

Human natural killer (NK) cell-based therapies are under assessment for treating various cancers, but cryopreservation reduces both the recovery and function of NK cells, thereby limiting their therapeutic feasibility. Using cryopreservation protocols optimized for T cells, here we find that ~75% of NK cells die within 24 h post-thaw, with the remaining cells displaying reduced cytotoxicity. Using CRISPR-Cas9 gene editing and confocal microscopy, we find that cryopreserved NK cells largely die via apoptosis initiated by leakage of granzyme B from cytotoxic vesicles. Pretreatment of NK cells with a combination of Interleukins-15 (IL-15) and IL-18 prior to cryopreservation improves NK cell recovery to ~90-100% and enables equal tumour control in a xenograft model of disseminated Raji cell lymphoma compared to non-cryopreserved NK cells. The mechanism of IL-15 and IL-18-induced protection incorporates two mechanisms: a transient reduction in intracellular granzyme B levels via degranulation, and the induction of antiapoptotic genes.

Abstract

One of the critical aspects for the accurate determination of neutron capture cross sections when combining time-of-flight and total energy detector techniques is the characterization and control of systematic uncertainties associated to the measuring devices. In this work we explore the most conspicuous effects associated to harsh count rate conditions: dead-time and pile-up effects. Both effects, when not properly treated, can lead to large systematic uncertainties and bias in the determination of neutron cross sections. In the majority of neutron capture measurements carried out at the CERN n\_TOF facility, the detectors of choice are the C$_{6}$D$_{6}$ liquid-based either in form of large-volume cells or recently commissioned sTED detector array, consisting of much smaller-volume modules. To account for the aforementioned effects, we introduce a Monte Carlo model for these detectors mimicking harsh count rate conditions similar to those happening at the CERN n\_TOF 20~m fligth path vertical measuring station. The model parameters are extracted by comparison with the experimental data taken at the same facility during 2022 experimental campaign. We propose a novel methodology to consider both, dead-time and pile-up effects simultaneously for these fast detectors and check the applicability to experimental data from $^{197}$Au($n$,$\gamma$), including the saturated 4.9~eV resonance which is an important component of normalization for neutron cross section measurements.

6. Large changes in detected selection signatures after a selection limit in mice bred for voluntary wheel-running behavior.

Abstract

In various organisms, sequencing of selectively bred lines at apparent selection limits has demonstrated that genetic variation can remain at many loci, implying that evolution at the genetic level may continue even if the population mean phenotype remains constant. We compared selection signatures at generations 22 and 61 of the High Runner mouse experiment, which includes 4 replicate lines bred for voluntary wheel-running behavior (HR) and 4 non-selected control (C) lines. Previously, we reported multiple regions of differentiation between the HR and C lines, based on whole-genome sequence data for 10 mice from each line at generation 61, which was >31 generations after selection limits had been reached in all HR lines. Here, we analyzed pooled sequencing data from ~20 mice for each of the 8 lines at generation 22, around when HR lines were reaching limits. Differentiation analyses of allele frequencies at ~4.4 million SNP loci used the regularized T-test and detected 258 differentiated regions with FDR = 0.01. Comparable analyses involving pooling generation 61 individual mouse genotypes into allele frequencies by line produced only 11 such regions, with almost no overlap among the largest and most statistically significant peaks between the two generations. These results implicate a sort of genetic churn that continues at loci relevant for running. Simulations indicate that loss of statistical power due to random genetic drift and sampling error are insufficient to explain the differences in selection signatures. The 13 differentiated regions at generation 22 with strict culling measures include 79 genes related to a wide variety of functions. Gene ontology identified pathways related to olfaction and vomeronasal pathways as being overrepresented, consistent with generation 61 analyses, despite those specific regions differing between generations. Genes Dspp and Rbm24 are also identified as potentially explaining known bone and skeletal muscle differences, respectively, between the linetypes.

Abstract

One of the crucial ingredients for the improvement of stellar models is the accurate knowledge of neutron capture cross-sections for the different isotopes involved in the $s$-,$r$- and $i$- processes. These measurements can shed light on existing discrepancies between observed and predicted isotopic abundances and help to constrain the physical conditions where these reactions take place along different stages of stellar evolution.In the particular case of the radioactive $^{94}$Nb, the $^{94}$Nb($n$,$\gamma$) cross-section could play a role in the determination of the $s$-process production of $^{94}$Mo in AGB stars, which presently cannot be reproduced by state-of-the-art stellar models. There are no previous $^{94}$Nb($n$,$\gamma$) experimental data for the resolved and unresolved resonance regions mainly due to the difficulties in producing high-quality samples and also due to limitations in conventional detection systems commonly used in time-of-flight experiments.Motivated by this situation, a first measurement of the $^{94}$Nb($n$,$\gamma$) reaction was carried out at CERN n\_TOF, thereby exploiting the high luminosity of the EAR2 area in combination with a new detection system of small-volume C6D6-detectors and a high quality $^{94}$Nb-sample. The latter was based on hyper-pure $^{93}$Nb material activated at the high-flux reactor of ILL-Grenoble. An innovative ring-configuration detection system in close geometry around the capture sample allowed us to significantly enhance the signal-to-background ratio. This set-up was supplemented with two conventional C$_{6}$D$_{6}$ detectors and a high-resolution LaCl$_{3}$(Ce)-detector, which will be employed for addressing reliably systematic effects and uncertainties.At the current status of the data analysis, 18 resonance in $^{94}$Nb+$n$ have been observed for the first time in the neutron energy range from thermal up to 10 keV., Comment: NPA-X conference proceeding

Abstract

Background/Purpose: Radiation-induced cardiac toxicity in lung cancer patients has received increased attention since RTOG 0617. However, large cohort studies with accurate cardiac substructure (CS) contours are lacking, limiting our understanding of the potential influence of individual CSs. Here, we analyse the correlation between CS dose and overall survival (OS) while accounting for deep learning (DL) contouring uncertainty, α/β uncertainty and different modelling approaches. Materials/Methods: This single institution, retrospective cohort study includes 730 patients (early-stage tumours (I or II). All treated: 2009–2019), who received stereotactic body radiotherapy (≥ 5 Gy per fraction). A DL model was trained on 70 manually contoured patients to create 12 cardio-vascular structures. Abstract

A new experimental area, the NEAR station, has recently been built at the CERN n TOF facility, at a short distance from the spallation target (1.5 m). The new area, characterized by a neutron beam of very high flux, has been designed with the purpose of performing activation measurements of interest for astrophysics and various applications. The beam is transported from the spallation target to the NEAR station through a hole in the shielding wall of the target, inside which a collimator is inserted. The new area is complemented with a {\gamma}-ray spectroscopy laboratory, the GEAR station, equipped with a high efficiency HPGe detector, for the measurement of the activity resulting from irradiation of a sample in the NEAR station. The use of a moderator/filter assembly is envisaged, in order to produce a neutron beam of Maxwellian shape at different thermal energies, necessary for the measurement of Maxwellian Averaged Cross Sections of astrophysical interest. A new fast-cycling activation technique is also being investigated, for measurements of reactions leading to isotopes of very short half life.

Abstract

This article presents a few selected developments and future ideas related to the measurement of $(n,\gamma)$ data of astrophysical interest at CERN n_TOF. The MC-aided analysis methodology for the use of low-efficiency radiation detectors in time-of-flight neutron-capture measurements is discussed, with particular emphasis on the systematic accuracy. Several recent instrumental advances are also presented, such as the development of total-energy detectors with $\gamma$-ray imaging capability for background suppression, and the development of an array of small-volume organic scintillators aimed at exploiting the high instantaneous neutron-flux of EAR2. Abstract

BackgroundDiabetic retinopathy (DR) affects about 25% of people with diabetes in Canada. Early detection of DR is essential for preventing vision loss. ObjectiveWe evaluated the real-world performance of an artificial intelligence (AI) system that analyzes fundus images for DR screening in a Quebec tertiary care center. MethodsWe prospectively recruited adult patients with diabetes at the Centre hospitalier de l’Université de Montréal (CHUM) in Montreal, Quebec, Canada. Patients underwent dual-pathway screening: first by the Computer Assisted Retinal Analysis (CARA) AI system (index test), then by standard ophthalmological examination (reference standard). We measured the AI system's sensitivity and specificity for detecting referable disease at the patient level, along with its performance for detecting any retinopathy and diabetic macular edema (DME) at the eye level, and potential cost savings. ResultsThis study included 115 patients. CARA demonstrated a sensitivity of 87.5% (95% CI 71.9-95.0) and specificity of 66.2% (95% CI 54.3-76.3) for detecting referable disease at the patient level. For any retinopathy detection at the eye level, CARA showed 88.2% sensitivity (95% CI 76.6-94.5) and 71.4% specificity (95% CI 63.7-78.1). For DME detection, CARA had 100% sensitivity (95% CI 64.6-100) and 81.9% specificity (95% CI 75.6-86.8). Potential yearly savings from implementing CARA at the CHUM were estimated at CAD $245,635 (US $177,643.23, as of July 26, 2024) considering 5000 patients with diabetes. ConclusionsOur study indicates that integrating a semiautomated AI system for DR screening demonstrates high sensitivity for detecting referable disease in a real-world setting. This system has the potential to improve screening efficiency and reduce costs at the CHUM, but more work is needed to validate it.

Abstract

Even though actions we observe in everyday life seem to unfold in a continuous manner, they are automatically divided into meaningful chunks, that are single actions or segments, which provide information for the formation and updating of internal predictive models. Specifically, boundaries between actions constitute a hub for predictive processing since the prediction of the current action comes to an end and calls for updating of predictions for the next action. In the current study, we investigated neural processes which characterize such boundaries using a repertoire of complex action sequences with a predefined probabilistic structure. Action sequences consisted of actions that started with the hand touching an object (T) and ended with the hand releasing the object (U). These action boundaries were determined using an automatic computer vision algorithm. Participants trained all action sequences by imitating demo videos. Subsequently, they returned for an fMRI session during which the original action sequences were presented in addition to slightly modified versions thereof. Participants completed a post-fMRI memory test to assess the retention of original action sequences. The exchange of individual actions, and thus a violation of action prediction, resulted in increased activation of the action observation network and the anterior insula. At U events, marking the end of an action, increased brain activation in supplementary motor area, striatum, and lingual gyrus was indicative of the retrieval of the previously encoded action repertoire. As expected, brain activation at U events also reflected the predefined probabilistic branching structure of the action repertoire. At T events, marking the beginning of the next action, midline and hippocampal regions were recruited, reflecting the selected prediction of the unfolding action segment. In conclusion, our findings contribute to a better understanding of the various cerebral processes characterizing prediction during the observation of complex action repertoires.

Abstract

Splicing factor mutations are common in myelodysplastic syndrome (MDS) and acute myeloid leukemia (AML), but how they alter cellular functions is unclear. We show that the pathogenic [SRSF2.sup.P95H/+] mutation disrupts the splicing of mitochondrial mRNAs, impairs mitochondrial complex I function, and robustly increases mitophagy. We also identified a mitochondrial surveillance mechanism by which mitochondrial dysfunction modifies splicing of the mitophagy activator PINK1 to remove a poison intron, increasing the stability and abundance of PINK1 mRNA and protein. [SRSF2.sup.P95H]-induced mitochondrial dysfunction increased PINK1 expression through this mechanism, which is essential for survival of [SRSF2.sup.P95H/+] cells. Inhibition of splicing with a glycogen synthase kinase 3 inhibitor promoted retention of the poison intron, impairing mitophagy and activating apoptosis in [SRSF2.sup.P95H/+] cells. Abstract

Background: MRI-guidance may aid better discrimination between Organs at Risk (OARs) and target volumes in proximity of the mediastinum. We report the first clinical experiences with Stereotactic Body Radiotherapy (SBRT) of (ultra)central lung tumours on a 1.5 T MR-linac. Materials and Methods: Patients with an (ultra)central lung tumour were selected for MR-linac based SBRT treatment. A T2-weighted 3D sequence MRI acquired during free breathing was used for daily plan adaption. Prior to each fraction, contours of Internal Target Volume (ITV) and OARs were deformably propagated and amended by a radiation oncologist. Inter-fractional changes in volumes and coverage of target volumes as well as doses in OARs were evaluated in offline and online treatment plans. Results: Ten patients were treated and completed 60 Gy in 8 or 12 fractions. In total 104 fractions were delivered. The median time in the treatment room was 41 min with a median beam-on time of 8.9 min. No grade ≥3 acute toxicity was observed. In two patients, the ITV significantly decreased during treatment (58 % and 37 %, respectively) due to tumour shrinkage. In the other patients, 81 % of online ITVs were within ±15 % of the volume of fraction 1. Comparison with the pre-treatment plan showed that ITV coverage of the online plan was similar in 52 % and improved in 34 % of cases. Adaptation to meet OAR constraints, led to decreased ITV coverage in 14 %. Conclusions: We describe the workflow for MR-guided Radiotherapy and the feasibility of using 1.5 T MR-linac for SBRT of (ultra) central lung tumours.

40. Large changes in detected selection signatures after a selection limit in mice bred for voluntary wheel-running behavior.

Abstract

In various organisms, sequencing of selectively bred lines at apparent selection limits has demonstrated that genetic variation can remain at many loci, implying that evolution at the genetic level may continue even if the population mean phenotype remains constant. We compared selection signatures at generations 22 and 61 of the "High Runner" mouse experiment, which includes 4 replicate lines bred for voluntary wheel-running behavior (HR) and 4 non-selected control (C) lines. Previously, we reported multiple regions of differentiation between the HR and C lines, based on whole-genome sequence data for 10 mice from each line at generation 61, which was >31 generations after selection limits had been reached in all HR lines. Here, we analyzed pooled sequencing data from ~20 mice for each of the 8 lines at generation 22, around when HR lines were reaching limits. Differentiation analyses of allele frequencies at ~4.4 million SNP loci used the regularized T-test and detected 258 differentiated regions with FDR = 0.01. Comparable analyses involving pooling generation 61 individual mouse genotypes into allele frequencies by line produced only 11 such regions, with almost no overlap among the largest and most statistically significant peaks between the two generations. These results implicate a sort of "genetic churn" that continues at loci relevant for running. Simulations indicate that loss of statistical power due to random genetic drift and sampling error are insufficient to explain the differences in selection signatures. The 13 differentiated regions at generation 22 with strict culling measures include 79 genes related to a wide variety of functions. Gene ontology identified pathways related to olfaction and vomeronasal pathways as being overrepresented, consistent with generation 61 analyses, despite those specific regions differing between generations. Genes Dspp and Rbm24 are also identified as potentially explaining known bone and skeletal muscle differences, respectively, between the linetypes.

Abstract

The heterogeneity of data poses a great challenge when data from different sources is to be merged for one application. Solutions for this are offered, for example, by ontology-based data management (OBDM). A challenge of OBDM is the automatic creation of semantic models from datasets. This process is typically performed either data- or label-driven and always involves manual human intervention. We identified textual descriptions of data, a form of metadata, quickly to be produced and consumed by humans, as third possible basis for automatic semantic modelling. In this paper, we present, how we plan to use textual descriptions to enhance semantic data management. We will use state of the art NLP technologies to identify concepts within textual descriptions and build semantic models from this in combination with an evolving ontology. We will use automatically identified models in combination with the human data provider to automatically extend the ontology so that it learns new verified concepts over time. Finally, we will use the created ontology and automatically identified semantic models to either rate descriptions for new data sources or even to automatically generate descriptive texts that are easier to understand by the human user than formal models. Abstract

Predicting other people's action is key to successful social interactions, enabling us to adjust our own behavior to the consequence of the others' future actions. Studies on action recognition have focused on the importance of individual visual features of objects involved in an action and its context. Humans, however, recognize actions on unknown objects or even when objects are imagined (pantomime). Other cues must thus compensate the lack of recognizable visual object features. Here, we focus on the role of inter-object relations that change during an action. We designed a virtual reality setup and tested recognition speed for 10 different manipulation actions on 50 subjects. All objects were abstracted by emulated cubes so the actions could not be inferred using object information. Instead, subjects had to rely only on the information that comes from the changes in the spatial relations that occur between those cubes. In spite of these constraints, our results show the subjects were able to predict actions in, on average, less than 64% of the action's duration. We employed a computational model -an enriched Semantic Event Chain (eSEC)- incorporating the information of spatial relations, specifically (a) objects' touching/untouching, (b) static spatial relations between objects and (c) dynamic spatial relations between objects. Trained on the same actions as those observed by subjects, the model successfully predicted actions even better than humans. Information theoretical analysis shows that eSECs optimally use individual cues, whereas humans presumably mostly rely on a mixed-cue strategy, which takes longer until recognition. Providing a better cognitive basis of action recognition may, on one hand improve our understanding of related human pathologies and, on the other hand, also help to build robots for conflict-free human-robot cooperation. Abstract

Genetic approaches in model organisms have consistently demonstrated that molecular traits such as gene expression are under genetic regulation, similar to clinical traits. The resulting expression quantitative trait loci (eQTL) have revolutionized our understanding of genetic regulation and identified numerous candidate genes for clinically relevant traits. More recently, these analyses have been extended to other molecular traits such as protein abundance, metabolite levels, and miRNA expression. Here, we performed global hepatic eQTL and microRNA expression quantitative trait loci (mirQTL) analysis in a population of Diversity Outbred mice fed two different diets. We identified several key features of eQTL and mirQTL, namely differences in the mode of genetic regulation (cis or trans) between mRNA and miRNA. Approximately 50% of mirQTL are regulated by a trans-acting factor, compared to ∼25% of eQTL. We note differences in the heritability of mRNA and miRNA expression and variance explained by each eQTL or mirQTL. In general, cis-acting variants affecting mRNA or miRNA expression explain more phenotypic variance than trans-acting variants. Finally, we investigated the effect of diet on the genetic architecture of eQTL and mirQTL, highlighting the critical effects of environment on both eQTL and mirQTL. Overall, these data underscore the complex genetic regulation of two well-characterized RNA classes (mRNA and miRNA) that have critical roles in the regulation of clinical traits and disease susceptibility.

Abstract

Abstract During preimplantation development, contractile forces generated at the apical cortex segregate cells into inner and outer positions of the embryo, establishing the inner cell mass (ICM) and trophectoderm. To which extent these forces influence ICM-trophectoderm fate remains unresolved. Here, we found that the nuclear lamina is coupled to the cortex via an F-actin meshwork in mouse and human embryos. Actomyosin contractility increases during development, upregulating Lamin-A levels, but upon internalization cells lose their apical cortex and downregulate Lamin-A. Low Lamin-A shifts the localization of actin nucleators from nucleus to cytoplasm increasing cytoplasmic F-actin abundance. This results in stabilization of Amot, Yap phosphorylation and acquisition of ICM over trophectoderm fate. By contrast, in outer cells, Lamin-A levels increase with contractility. This prevents Yap phosphorylation enabling Cdx2 to specify the trophectoderm. Thus, forces transmitted to the nuclear lamina control actin organization to differentially regulate the factors specifying lineage identity.

Abstract

Abstract During the second long shutdown period of the CERN accelerator complex (LS2, 2019-2021), several upgrade activities took place at the n_TOF facility. The most important have been the replacement of the spallation target with a next generation nitrogen-cooled lead target. Additionally, a new experimental area, at a very short distance from the target assembly (the NEAR Station) was established. In this paper, the core commissioning actions of the new installations are described. The improvement in the n_TOF infrastructure was accompanied by several detector development projects. All these upgrade actions are discussed, focusing mostly on the future perspectives of the n_TOF facility. Furthermore, some indicative current and future measurements are briefly reported.

Abstract

Smart cities around the world have begun monitoring parking areas in order to estimate available parking spots and help drivers looking for parking. The current results are promising, indeed. However, existing approaches are limited by the high cost of sensors that need to be installed throughout the city in order to achieve an accurate estimation. This work investigates the extension of estimating parking information from areas equipped with sensors to areas where they are missing. To this end, the similarity between city neighborhoods is determined based on background data, i.e., from geographic information systems.
