African yam bean (AYB; Sphenostylis stenocarpa) is an underutilized legume indigenous to Africa with great potential to enhance food security and offer nutritional and medicinal opportunities. However, low grain yield caused by fungal diseases, including pod blight and leaf tip dieback, deters farmers from large‐scale cultivation. To determine the prevalence of fungal diseases affecting leaves, pods and flowers of AYB, a survey was conducted in 2018 and 2019 in major AYB‐growing areas in Nigeria. Leaf tip dieback, flower bud rot and pod blight were the most common symptoms. Morphological and molecular assays were conducted to identify the causal agents of the observed diseases. In all the samples examined, fungi from eight genera were isolated from diseased leaves, buds and pods. Koch's postulates were fulfilled only for fungi belonging to the Colletotrichum genus. Fungi from the other seven genera did not produce disease symptoms in healthy AYB tissues. Several Colletotrichum isolates were characterized by sequencing the rDNA internal transcribed spacer (ITS), glyceraldehyde‐3‐phosphate dehydrogenase, calmodulin and ApMAT loci. A combined phylogenetic analysis revealed four Colletotrichum species: C. siamense, C. theobromicola and C. fructicola, which were recovered from diseased leaves, and C. truncatum, recovered from diseased pods and buds. Our results are useful to gear efforts to develop integrated management strategies to control diseases affecting AYB in Nigeria and elsewhere. Availability of such strategies may stimulate greater AYB cultivation, which can contribute to diet diversification, something repeatedly advocated by a range of stakeholders to increase food security and prosperity of smallholder farmers. \nHighlightsIn this paper, we used the polyphasic approach combining 16S rRNA gene phylogenies, identity (p-distance), 16S-23S ITS secondary structures, and morphological and habitat analyses to describe two new genera of Leptolyngbyaceae, Pseudoleptolynbgya G.S. Hentschke gen. nov. (based on LEGE 16533 and LEGE 16651) and Leptolyngbyopsis G.S. Hentschke gen. nov. (based on LEGE 16524). These strains were sampled from freshwater environments at Figueira da Foz, Monchique and Coimbra, Portugal. Both genera are morphologically and ecologically identical to Leptolyngbya and can only be distinguished from it by molecular analyses. Based on morphological and ITS secondary structure analyses we describe two species of Pseudoleptolyngbya, P. figueirensis and P. monchiquensis. P. figueirensis differs from P. monchiquensis by the denser arrangement of the trichomes, the presence of firm sheaths, the common formation of shorter trichomes (up to 30 cells), and the olive or yellowish-green cell content, which is darker in P. monchiquensis. Moreover, the macroscopic appearance of P. figueirensis is a dense brownish mat, while P. monchiquensis is less dense and reddish. 16S rRNA gene sequences of Arthronema africanum CCALA020 are > 99% identical to Leptolyngbya boryana PCC6306, so we transfer A. africanum to Leptolyngbya, as L. africanoidea. Culture collections such as the Blue Biotechnology and Ecotoxicology Culture Collection (LEGE‐CC) hold approximately 1200 cyanobacterial strains and are critical community resources. However, many isolates in this and other collections have not been described with a polyphasic approach, and this limits further study. Here, we employed a polyphasic methodology that integrates 16S rRNA gene phylogenetic analyses, similarity (p‐distance), 16S‐23S ITS rRNA region secondary structures, morphological analyses, and habitat assessments to describe four novel cyanobacterial genera from the LEGE‐CC, Portugal. Pseudolimnococcus planktonicus gen. et sp. nov. (Chroococcales) is phylogenetically and morphologically related to Limnococcus. The 16S rRNA gene similarity between the types of both genera is only 93.1%. Morphologically, Pseudolimnococcus cells do not reach the original spherical shape before the next division or have aerotopes and firm mucilage, while Limnococcus cells reach the original shape, lack aerotopes, and have diffluent mucilage. Eucapsopsis lusitanus gen. et sp. nov. (Chroococcales) is morphologically similar to Eucapsis but differs from it by having aerotopes and diffluent envelope. Eucapsis lacks aerotopes and has firm mucilaginous envelopes, rarely diffluent. Both genera are phylogenetically very distant from each other and have only 90.68% 16S rRNA gene similarity. Pseudoacaryochloris arrabidensis gen. et sp. nov. (Acaryochloridales) differs from Acaryochloris by the lack of mucilaginous envelope, which is present in Acaryochloris. Both genera are phylogenetically distant and have only 94.1% 16S rRNA gene similarity. Moreover, Acaryochloris is marine (sponge symbiont), while Pseudoacaryochloris is from freshwater. Vasconcelosia minhoensis gen. et sp. nov. (Nodosilineales) is phylogenetically related to Cymatolege but has only 94.3% similarity with this genus. Morphologically both genera are distinct. Vasconcelosia has a Romeria‐like structure, while Cymatolege has a Phormidium‐like structure. In all cases the 16S‐23S ITS rRNA region secondary structures are in agreement with the other analyses. This investigation reports the polyphasic characterization of six cyanobacterial strains that were isolated from Basantgarh village of district Udhampur in the union territory of Jammu and Kashmir, India. Morphological examination of the isolated strains indicated that the strains are members of the genus Nostoc or its morphotypes. Phylogenetic analyses using the 16S rRNA gene showed that five strains clustered in the Nostoc sensu stricto clade, whereas one strain clustered in the Desmonostoc clade. Further, comparative studies with their phylogenetically related taxa, based on morphology, folded secondary structures, phylogeny of the ITS rRNA region, and the percent genetic homology of 16S rRNA gene and ITS rRNA region clearly established the strains as novel taxa belonging to the genera Nostoc and Desmonostoc. Also, two strains 21A‐PS and 2JNA‐PS emerged as conspecific to each other, representing the same species of Nostoc. Hence, in accordance with the International code of Nomenclature for algae, fungi, and plants, this study describes Nostoc jammuense, Nostoc globosum, Nostoc breve, and Nostoc coriaceum, as novel species of the genus Nostoc, while Desmonostoc raii is described as a novel species of the genus Desmonostoc. Until mid-1970s, cyanobacteria have been interpreted as algae despite they differed from other members of this taxonomic group by the absence of cell nucleus (that is currently considered among the characters of prokaryotic organization). However, when bacteria were interpreted as prokaryotes, blue-green algae became reattributed as cyanobacteria, and bacteriologists began to study their cultured strains with microbiology methods. But since these objects did not obey the provisions of bacteriological code (ICNB), development of their taxonomy had certain problems, especially regarding nomenclature, that could not be solved until early-2010s. Current changes in taxonomy of cyanobacteria result from the general progress in taxonomy of prokaryotes due to explosive accumulation of 16S rRNA gene sequencing data, particularly for uncultured objects. Advances in description of new cyanobacteral taxa are as follows: (I) based on polyphasic approach, new taxa ranging from species to order were published; (II) "dark" cyanobacteria demonstrating several types of non-photosynthetic metabolism were described; I(II) strains producing red-shifted chlorophylls d and f were published. Advances in nomenclature of cyanobacteria are: (IV) taxa validly published according to botanical code (ICN) are also considered valid under prokaryotic code (ICNP); (V) the category of phylum was introduced into ICNP, and due to valid publication of the type genus Cyanobacterium, phylum name Cyanobacteriota became legitimate; (VI) names of uncultured Candidatus objects could get a standing in nomenclature based on SeqCode in which type material is represented by genomic DNA sequences. Advances in classification of cyanobacteria are: (VII) evolutionary tree of oxygenic phototrophs and related phylotypes was construed; (VIII) phylogenomic system of orders and families was elaborated; (IX) ecogenomic system combining genomie analysis with genome distribution data was offered. TN-15, a unique and less explored cyanobacterial strain, holds significant promise as a novel candidate for the synthesis of nanoparticles. This noticeable research gap underscores the novelty and untapped potential of Desertifilum sp. TN-15 in the field of nanomedicine. The characterization of the biogenically synthesized ZnO–NPs involved the application of diverse analytical techniques. Ultraviolet–visible spectroscopy revealed a surface plasmon resonance peak at 298 nm. Fourier transform infrared spectral analysis was utilized to confirm the involvement of biomolecules in the biogenic synthesis and stability. Scanning electron microscopy was employed to probe the surface morphology of the biogenic ZnO–NPs unveiling their size of 94.80 nm and star-shaped. Furthermore, X-ray diffraction analysis substantiated the crystalline nature of ZnO–NPs, with a crystalline size measuring 46 nm. To assess the physical stability of ZnO–NPs, zeta potential and dynamic light scattering measurements were conducted, yielding values of + 31.6 mV, and 94.80 nm, respectively, indicative of favorable stability. The antibacterial capabilities of Desertifilum sp. TN-15 are attributed to its abundance of bioactive components, including proteins, liposomes, amino derivatives, and carotenoids. Through the synthesis of zinc oxide nanoparticles (ZnO–NPs) with this strain, we have effectively used these chemicals to generate nanoparticles that exhibit noteworthy antibacterial activity against Staphylococcus aureus (MIC: 30.05 ± 0.003 µg/ml). Additionally, the ZnO–NPs displayed potent antifungal activity and antioxidant properties, as well as significant antihemolytic effects on red blood cells (IC50: 4.8 µg/ml). Cytotoxicity assessment using brine shrimps revealed an IC50 value of 3.1 µg/ml. The multifaceted actions of the biogenically synthesized ZnO–NPs underscore their potential applications in pharmacological and therapeutic fields. An unknown filamentous cyanobacterium was isolated from the Yuqiao Reservoir, a eutrophic drinking water reservoir in Tianjin city, northern China. The cyanobacterium shares some morphological similarities with most Leptolyngbya-like species, having thin trichomes that can facultatively produce sheaths, with one trichome per sheath, and contain peripheral thylakoids. Based on a combination of morphological, molecular, ultrastructural and ecological evidence, we propose the description of a new cyanobacterial taxon, Cyanoleptonema yuqiaoensis gen. & sp. nov. which expands our understanding of cyanobacterial diversity. Phylogenetic analyses based on 16S rRNA gene sequences placed this freshwater strain into a well-supported clade closely related to the marine or saline genera Sodalinema and Baaleninema. Genetic identity between Cyanoleptonema and other cyanobacteria, including Sodalinema and Baaleninema, was found to be less than 95% in 16S rRNA. Studies performed in North America, Africa, and South America have led to the isolation of four new species of Kastovskya, a filamentous cyanobacterial genus that before this manuscript had only one species, Kastovskya adunca from Chile. Kastovskya nitens and K. viridissima were isolated from soils on San Nicolas Island, K. sahariensis was isolated from hypolithic habitats from the Sahara Desert in Algeria, and K. circularithylacoides was isolated from hypolithic habitats in Chile. The molecular analyses are corroborated by morphological data, morphometric analysis, and ecological and biogeographical considerations for robust polyphasic descriptions of all taxa. The peculiar transatlantic distribution of this genus bears similarity to other taxa in recently published studies and is in agreement with a hypothesis suggesting that cyanobacteria in Africa may disperse to the Americas on dust particles during windstorms. This work is unusual in that species in a single rare cyanobacterial genus with a disjunct distribution are described simultaneously from three continents. The 16S rRNA gene analyses performed for this study also revealed that another recent genus, Arizonema, is clearly a later synonym of Symplocastrum. Analysis of cyanobacterial diversity is an important step towards identifying new biological resources for production of bioactive compounds. In this study cyanobacterial surface coverage was assessed through satellite image analysis of Tehran drinking water reservoirs in late summer and early autumn of 2018. We also analysed freshwater cyanobacteria including seven local strains purified from surface water samples. Following a polyphasic taxonomic approach combining morphological and molecular methods the culturable strains were classified into Synechococcales and Oscillatoriales, representing at least three families and four genera. Similarity-based analysis of their partial 16S rRNA genes showed 93.4–97.0% sequence identity among the strains from different ecosystems. Considering 95–96% as the overall cut-off for discriminating bacterial species, strains UTMC6011 and UTMC6013 are likely to be new species. Physiological characterization of the new strains and nine others isolated previously by our group from another freshwater ecosystem led to identification of seven nitrogen-fixing, 11 sulphide-tolerant, three salt-tolerant and three heterotrophic strains. Further taxonomic analyses are needed to support investigation of their biotechnological potential. The biodiversity and physiology of cyanobacteria in the main drinking water reservoirs of the megacity, Tehran, were studied by remote sensing and sample analysis. Seven cyanobacterial strains were isolated from the two freshwater lakes. Similarity-based analysis of obtained 16S rRNA gene sequence showed 93.4–97.0% sequence similarity with the strains from other ecosystems. This article presents the results of an ongoing inventory of Ascomycota in Yunnan, China, carried out as part of the research project series "Exploring ascomycete diversity in Yunnan". From over 100 samples collected from diverse host substrates, microfungi have been isolated, identified and are currently being documented. The primary objective of this research is to promote the discovery of novel taxa and explore the ascomycete diversity in the region, utilising a morphology-phylogeny approach. This article represents the second series of species descriptions for the project and introduces three undocumented species found in the families Bambusicolaceae, Dictyosporiaceae and Periconiaceae, belonging to the suborder Massarineae (Pleosporales, Dothideomycetes). These novel taxa exhibit typical morphological characteristics of Bambusicola, Periconia and Trichobotrys, leading to their designation as Bambusicola hongheensis, Periconia kunmingensis and Trichobotrys sinensis. Comprehensive multigene phylogenetic analyses were conducted to validate the novelty of these species. The results revealed well-defined clades that are clearly distinct from other related species, providing robust support for their placement within their respective families. Notably, this study unveils the phylogenetic affinity of Trichobotrys within Dictyosporiaceae for the first time. Additionally, the synanamorphism for the genus Trichobotrys is also reported for the first time. Detailed descriptions, illustrations and updated phylogenies of the novel species are provided, and thus presenting a valuable resource for researchers and mycologists interested in the diversity of ascomycetes in Yunnan. By enhancing our understanding of the Ascomycota diversity in this region, this research contributes to the broader field of fungal taxonomy and their phylogenetic understanding. Five cyanobacterial strains exhibiting Nostoc‐like morphology were sampled from the biodiversity hotspots of the northeast region of India and characterized using a polyphasic approach. Molecular and phylogenetic analysis using the 16S rRNA gene indicated that the strains belonged to the genera Amazonocrinis and Dendronalium. In the present investigation, the 16S rRNA gene phylogeny clearly demarcated two separate clades of Amazonocrinis. The strain MEG8‐PS clustered along with Amazonocrinis nigriterrae CENA67, which is the type strain of the genus. The other three strains ASM11‐PS, RAN‐4C‐PS, and NP‐KLS‐5A‐PS clustered in a different clade that was phylogenetically distinct from the Amazonocrinis sensu stricto clade. Interestingly, while the 16S rRNA gene phylogeny exhibited two separate clusters, the 16S–23S ITS region analysis did not provide strong support for the phylogenetic observation. Subsequent analyses raised questions regarding the resolving power of the 16S–23S ITS region at the genera level and the associated complexities in cyanobacterial taxonomy. Through this study, we describe a novel genus Ahomia to accommodate the members clustering outside the Amazonocrinis sensu stricto clade. In addition, we describe five novel species, Ahomia kamrupensis, Ahomia purpurea, Ahomia soli, Amazonocrinis meghalayensis, and Dendronalium spirale, in accordance with the International Code of Nomenclature for algae, fungi, and plants (ICN). Apart from further enriching the genera Amazonocrinis and Dendronalium, the current study helps to resolve the taxonomic complexities revolving around the genus Amazonocrinis and aims to attract researchers to the continued exploration of the tropical and subtropical cyanobacteria for interesting taxa and lineages. SUMMARY: Genus Klisinema (Desertifilaceae, Desertifilales) has only one species, Klisinema persicum, reported worldwide, and this species was found in soils around thermal springs (50°C) of high radiation regions. Simple filament cyanobacteria belonging to the genus Klisinema were collected from Korea, and cultured samples were studied through morphological, ecological and molecular analyses. Both Korean K. persicum and Klisinema koreana were found as epilithic species in freshwater gravel, which have different ecological characteristics from previously reported K. persicum. As a result of morphological analysis, keritomized cytoplasm was observed in K. koreana, and this feature was also newly observed in K. persicum that was discovered in Korea. In addition, K. koreana has a larger width/length ratio compared to K. persicum, and both species have an irregular thylakoid arrangement. Furthermore, as a result of phylogenetic analysis of 16S rRNA, the genus Klisinema formed a monophyletic lineage, and K. koreana strains were clearly separated from K. persicum strains to form a phylogenetic tree. Moreover, when comparing the secondary structure of the 16S–23S internal transcribed spacer region, K. koreana has a structural difference from K. persicum, especially in the structure of the Box‐B helix. Therefore, Klisinema koreana sp. nov. was proposed as a new species belonging to the genus Klisinema of family Desertifilaceae. [ABSTRACT FROM AUTHOR]

To increase the knowledge on diversity of freshwater cyanobacteria, we have characterized a novel filamentous, benthic cyanobacterium isolated from a pond in Golestan province, Iran. The isolate was studied using a polyphasic approach based on morphological and molecular characteristics, including phylogenetic and structural analysis of the 16S rRNA gene sequence and 16S to 23S internal transcribed spacer (ITS), respectively. The overall morphology of the isolate was similar to Nostocalean member's strains with non-tapering filaments and distributed heterocytes. However, the molecular analyses suggested that it was more closely related to the lithobiont and rock-dwelling Goleter apudmare Miscoe & J.R.Johansen, which has tapered filaments and basal heterocytes. The 16S rRNA gene phylogenetic analysis, based on bayesian inference and maximum likelihood methods, placed the isolate close to G. apudmare. The D1-D1', Box-B and V3 helixes of the isolate's ITS sequence were different when compared to other Goleter ITS sequences. Similar to G. apudmare HA4356-MV2 (JN385288.1) and Goleter sp. CHAB TP201702 (MT488126.1), tRNA genes were absent from the amplified ITS sequence in the studied isolate. These morphological and molecular characteristics differentiate this isolate from all other Goleter taxa described so far, which contributes to extend the knowledge on cyanobacterial diversity and occurrence of benthic cyanobacteria in Iranian freshwaters. Pour accroître les connaissances sur la diversité des cyanobactéries d'eau douce, nous avons caractérisé une nouvelle cyanobactérie benthique filamenteuse isolée d'un étang de la province du Golestan, en Iran. L'isolat a été étudié en utilisant une approche polyphasique basée sur les caractéristiques morphologiques et moléculaires, comprenant l'analyse phylogénétique et structurelle de la séquence du gène de l'ARNr 16S et de l'espaceur transcrit interne (ITS) 16S à 23S, respectivement. La morphologie globale de l'isolat était similaire à celle des souches membres des Nostocales avec des filaments non effilés et des hétérocytes distribués. Cependant, les analyses moléculaires suggèrent qu'il était plus étroitement lié au lithobionte et au Goleter apudmare Miscoe & J.R.Johansen vivant dans les roches, qui possède des filaments effilés et des hétérocytes basaux. L'analyse phylogénétique du gène de l'ARNr 16S, basée sur les méthodes d'inférence bayésienne et de vraisemblance maximale, a placé l'isolat à proximité de G. apudmare. Les hélices D1-D1', Box-B et V3 de la séquence ITS de l'isolat étaient différentes par rapport aux autres séquences Goleter ITS. De la même manière que G. apudmare HA4356-MV2 (JN385288.1) et Goleter sp. CHAB TP201702 (MT488126.1), les gènes d'ARNt étaient absents de la séquence ITS amplifiée dans l'isolat étudié. Ces caractéristiques morphologiques et moléculaires différencient cet isolat de tous les autres taxons de Goleter décrits jusqu'à présent, ce qui contribue à étendre les connaissances sur la diversité cyanobactérienne et la présence de cyanobactéries benthiques dans les eaux douces iraniennes. Cyanobacteria inhabiting lotic environments have been poorly studied and characterized in Mexico, despite their potential risks from cyanotoxin production. This article aims to fill this knowledge gap by assessing the importance of benthic cyanobacteria as potential cyanotoxin producers in central Mexican rivers through: (i) the taxonomic identification of cyanobacteria found in these rivers, (ii) the environmental characterization of their habitats, and (iii) testing for the presence of toxin producing genes in the encountered taxa. Additionally, we introduce and discuss the use of the term “CyanoHAMs” for lotic water environments. Populations of cyanobacteria were collected from ten mountain rivers and identified using molecular techniques. Subsequently, these taxa were evaluated for genes producing anatoxins and microcystins via PCR. Through RDA analyses, the collected cyanobacteria were grouped into one of three categories based on their environmental preferences for the following: (1) waters with high ionic concentrations, (2) cold-temperate waters, or (3) waters with high nutrient enrichment. Populations from six locations were identified to genus level: Ancylothrix sp., Cyanoplacoma sp., and Oxynema sp. The latter was found to contain the gene that produces anatoxins and microcystins in siliceous rivers, while Oxynema tested positive for the gene that produces microcystins in calcareous rivers. Our results suggest that eutrophic environments are not necessarily required for toxin-producing cyanobacteria. Our records of Compactonostoc, Oxynema, and Ancylothrix represent the first for Mexico. Four taxa were identified to species level: Wilmottia aff. murrayi, Nostoc tlalocii, Nostoc montejanii, and Dichothrix aff. willei, with only the first testing positive using PCR for anatoxin and microcystin-producing genes in siliceous rivers. Due to the differences between benthic growths with respect to planktonic ones, we propose the adoption of the term Cyanobacterial Harmful Algal Mats (CyanoHAMs) as a more precise descriptor for future studies. During the study of diversity in filamentous cyanobacteria in China, two strains (WZU0719 and WZU0723) with the form of thin filaments were isolated from the surface of Qiandao Lake, a large freshwater lake in Zhejiang Province, China. A comprehensive analysis was conducted, incorporating morphological, ecological, and molecular data. The morphological examination provided an initial identification as a Leptolyngbya-like cyanobacterium. Genetic characterization was also performed by amplifying the 16S rRNA gene and the 16S-23S rRNA internal transcribed spacer (ITS) region. The phylogenetic grouping based on the 16S rRNA gene demonstrates that the examined strain is unequivocally assigned to the Pegethrix genus. However, it possesses distinct phylogenetic divergence from the six described Pegethrix species. Additionally, discrepancies in habitat further differentiate it from other members of this genus. Employing the polyphasic approach, we present a comprehensive account of the newly discovered taxa: Pegethrix qiandaoensis sp. nov. As the taxonomic knowledge of cyanobacteria from terrestrial environments increases, it remains important to analyze biodiversity in areas that have been understudied to fully understand global and endemic diversity. This study was completed as part of a larger algal biodiversity study of the soil biocrusts of San Nicholas Island, California, USA. Among the taxa isolated were several new species in three genera (Atlanticothrix, Pycnacronema, and Konicacronema) which were described from, and previously restricted to, Brazil. New taxa are described herein using a polyphasic approach to cyanobacterial taxonomy that considers morphological, molecular, ecological, and biogeographical factors. Morphological data corroborated by molecular analysis including sequencing of the 16S rRNA gene, and the associated 16S–23S ITS rRNA region was used to delineate three new species of Atlanticothrix, two species of Pycnacronema, and one species of Konicacronema. The overlap of genera from San Nicolas Island and Brazil suggests that cyanobacterial genera may be widely distributed across global hemispheres, whereas the presence of distinct lineages may indicate that this is not true at the species level. Our data suggest that based upon global wind patterns, cyanobacteria in both Northern and Southern hemispheres of the Americas may have a more recent common ancestor in Northern Africa, but this common ancestry is distant enough that speciation has occurred since transatlantic dispersal. A new filamentous cyanobacterial strain (CHAB 4127) was successfully isolated from the algal mat during the field investigation of the open channel from Luanhe River to Tianjin, China. The polyphasic approach combining morphological, ultrastructural, ecological, and molecular features was used to characterize this studied strain. The strain is morphologically similar to the Microcoleus-like cyanobacterial taxa under light microscopy, and the radial arrangement of thylakoids is also consistent with that of Microcoleus-like groups. The phylogenetic position of CHAB 4127 based on 16S rRNA gene sequences, is shown to be clearly clustered into an independent clade with the newly established genus Microcoleusiopsis. The maximum similarity of 16S rRNA gene of the studied strain with other existing related cyanobacterial taxa is 93.97%, and the ITS secondary structures is also obviously different from other members of Microcoleaceae. Three simple filamentous cyanobacteria (strains S1, S6 and S10) were isolated from the upper surface of intertidal soil of an Indian mangrove forest, the Sundarbans. The three cyanobacterial strains formed a new cluster within the phylogenetic tree calculated based on 16S rRNA gene sequences, which was distinctive from the clade of the ecologically and morphologically most similar genus, Euryhalinema. Moreover, the sequence similarity of 16S rRNA gene was less than 94.4% compared with the reference strain of Euryhalinema, which confirmed that the new strains belonged to a novel genus. The folded secondary structure of D1-D1', Box-B and V3 helices of the internal transcribed spacer (ITS) region were different from those of Euryhalinema and other members of family Nodosilineaceae. P-distance analysis of the ITS region of the test strains showed 22.2% dissimilarity with the reference genus Euryhalinema, which exceeded the cut off (7%) and strongly supported that it is a novel species within a novel genus. Differential morphological and ultrastructural characters, such as the presence of various cellular inclusions (prominent large cyanophycin granule, glycogen α granules, lipid β granules and mucilaginous sheath), conical-shaped apical cell as well as cell width, supported the description of a new taxon. Therefore, based on morphological, ultrastructural and molecular analysis we propose that cyanobacterial strains S1, S6 and S10 belong to a new monophyletic genus and species Almyronema epifaneia gen. & sp. nov., in the recently established family Nodosilineaceae. Studies performed in North America, Africa, and South America have led to the isolation of four new species of Kastovskya, a filamentous cyanobacterial genus that before this manuscript had only one species, Kastovskya adunca from Chile. Kastovskya nitens and K. viridissima were isolated from soils on San Nicolas Island, K. sahariensis was isolated from hypolithic habitats from the Sahara Desert in Algeria, and K. circularithylacoides was isolated from hypolithic habitats in Chile. The molecular analyses are corroborated by morphological data, morphometric analysis, and ecological and biogeographical considerations for robust polyphasic descriptions of all taxa. The peculiar transatlantic distribution of this genus bears similarity to other taxa in recently published studies and is in agreement with a hypothesis suggesting that cyanobacteria in Africa may disperse to the Americas on dust particles during windstorms. This work is unusual in that species in a single rare cyanobacterial genus with a disjunct distribution are described simultaneously from three continents. The 16S rRNA gene analyses performed for this study also revealed that another recent genus, Arizonema, is clearly a later synonym of Symplocastrum. This issue is resolved here with the collapsing of the type species Arizonema commune into Symplocastrum flechtnerae.

Simple filamentous cyanobacteria comprise a diverse and polyphyletic group of species, primarily in the orders Leptolyngbyales and Oscillatoriales, that need more sampling to improve their taxonomy. Oceanic islands, such as the Azores archipelago, present unique habitats and biogeographic conditions that harbor an unknown range of diversity of microorganisms. Filamentous cyanobacteria isolated from aquatic habitats in the Azores and maintained in the BACA culture collection were described using morphology, both light and transmission electron microscopy, ecology, and genetic data of the 16S rRNA gene sequences and 16S–23S Internal Transcribed Spacer (ITS) rRNA region secondary structure. Our analyses revealed two new monophyletic genera: Tumidithrix elongata gen. sp. nov. (Pseudanabaenaceae) and Radiculonema aquaticum gen. sp. nov. (Leptolyngbyaceae). In addition, two new species Leptodesmis lacustris sp. nov. (Leptolyngbyaceae) and Pycnacronema lacustrum sp. nov. (Wilmottiaceae) are reported as the first aquatic species for these genera. The description of these new taxa and the genetic study of an isolate of Leptodesmis alaskaensis from the Azores followed the polyphasic approach, identifying diacritical features. Knowledge of the tropical terrestrial cyanobacterial flora from the African continent is still limited. Of 31 strains isolated from soil and subaerial samples collected in Lagos State, Nigeria, three were found to be in the Oculatellaceae, including two species in a new genus. Subsequently, isolates from microbial mats in White Sands National Park in New Mexico, United States, and from a rock near the ocean in Puerto Rico, United States, were found to belong to the new genus as well. Cyanobacterial isolates were characterized microscopically, sequenced for the 16S rRNA gene and associated ITS region, and phylogenetically analyzed. Egbenema gen. nov., with three new species, as well as two new species of Albertania were differentiated from all other Oculatellaceae. Both genera belong to a supported clade within the Oculatellaceae that includes Trichotorquatus and Komarkovaea. The two new species of Albertania, A. egbensis and A. latericola, were from the same sample, but were evolutionarily separate based on 16S rRNA gene phylogenies, percent identity below the 98.7% threshold, and ITS rRNA percent dissimilarity >7.0%. Egbenema aeruginosum gen. et sp. nov. was phylogenetically separated from Trichotorquatus and Albertania but was in a clade with other strains belonging to Egbenema. The two Egbenema strains from the United States are here named Egbenema epilithicum sp. nov. and Egbenema gypsiphilum sp. nov. Our results support the hypothesis that further species discoveries of novel cyanobacteria will likely be made in soils and subaerial habitats, as these habitats continue to be studied, both in tropical and temperate biomes. In this study, 18 strains of cyanobacteria were isolated from seven sites, including the Han River and Anseong Stream in Republic of Korea, and we propose these isolated strains as Limnonema gen. nov., belonging to the Aerosakkonemataceae family of the Oscillatoriales order, and also, as L. hangangris sp. nov. and L. anseonga sp. nov. These strains were identified based on morphological data using a light microscope and a transmission electron microscope, and molecular data using 16S rRNA and 16S–23S ITS gene sequences. The genus Limnonema was mainly collected as planktons, and some L. hangangris (ACKU-695–697) appeared as epilithic cyanobacteria. The genus Limnonema showed filamentous trichomes, intracellular gas vacuoles, and irregular thylakoids arrangement, which was distinct from genera Cephalothrix, Microseira, and Potamosiphon belonging to the family Aerosakkonemataceae. Moreover, the cell widths of genus Limnonema were narrower than those of genus Aerosakkonema, which is the type genus of the family Aerosakkonemataceae, and L. anseonga contained more cells with wider widths than those of L. hangangris. In the 16S rRNA gene sequence phylogeny, genus Limnonema belonged to the family Aerosakkonemataceae and was distinguished from its close relatives, genera Aerosakkonema and Cephalothrix, and L. hangangris and L. anseonga formed different branches. In 16S rRNA gene sequence similarity, genus Limnonema showed 95.4–95.9% and 93.6–94.4% similarity with genera Aerosakkonema and Cephalothrix, respectively, and L. hangangris and L. anseonga showed 97.6–97.7% similarity between each other. In the 16S–23S ITS secondary structure, the D1–D1′, Box-B, and V3 helices of genus Limnonema were distinguished from genera belonging to the family Aerosakkonemataceae, and the V3 helices of L. hangangris and L. anseonga were also different from each other. Until recently, the study of cyanobacteria and microalgae has been hampered by the need to cultivate these organisms to gain insight into their cytomorphology, life cycle and molecular biology. However, various microbial species characterized by thick sheaths of exopolymeric substances were difficult to isolate in culture due to their associated symbiotic bacteria. Other microbes evaded culture. Such challenges have now been overcome by the development of metagenomic techniques that allow direct DNA sequencing from environmental samples, as well as high resolution microscopy techniques that permit direct imaging of environmental samples. The sampling of understudied taxa from extreme environments and of toxic species has been facilitated by specialized robotic equipment. Single-cell sequencing has allowed for the proper characterization of microalgal species and their response to environmental changes. Various strains of cyanobacteria, microalgae and macroalgae have gained renewed interest for their high-value metabolites. This paper provides an overview of the emerging technologies and explains how they are being used to identify such strains and their products for industrial application. Advances in genetic engineering and CRISPR technology have facilitated the production of strains that are more amenable to culture, metabolite extraction, scale-up and application in biorefinery approaches. Heterocyte-forming cyanobacteria form symbiotic relationships with several lineages of plants. Here, twenty (20) strains of endosymbiotic cyanobacteria (cyanobionts) with Nostoc-like morphologies were isolated from the highly specialized coralloid roots of five host species in Cycadales—Cycas debaoensis, C. fairylakea, C. elongata, Ceratozamia robusta, and Macrozamia moorei. Molecular phylogeny based on the 16S rRNA gene placed these strains into seven different taxa within the Nostocaceae, specifically under the genera Desmonostoc and Dendronalium. The percent dissimilarity and unique patterns in the secondary structures of the D1-D1′, Box-B, V2, and V3 helices, which were based on the 16S–23S rRNA internal transcribed spacer (ITS) regions, supported three distinct species in Desmonostoc. These three morphologically distinct novel species are described in this report: Desmonostoc debaoense sp. nov., Desmonostoc meilinense sp. nov., and Desmonostoc xianhuense sp. nov. Other investigated strains were phylogenetically identified as members of the recently discovered genus Dendronalium and represent the first report of association of that genus with cycads. Our findings suggest that the order Cycadales hosts diverse species of cyanobionts in their coralloid roots and that many potential unreported or novel taxa are present in cycads occurring in their natural habitat and await discovery. Soil cyanobacterium, FBCC-A195 were isolated from the soil underneath the Jangnak Bridge, Republic of Korea. The FBCC-A195 was studied using light microscopy (LM), transmission electron microscopy (TEM), 16S rRNA, 16S–23S ITS, and ecological data. FBCC-A195 showed the circular cross-section of the trichome, broad mucilaginous envelopes, a swirl-like pattern of thylakoids inside the cell, and the lowest length/width ratio within the genus. In the 16S rRNA phylogeny, FBCC-A195 showed a sister relationship of Hormoscilla and belonged to the family Gomontiellaceae. The p-distance of FBCC-A195 ranged from 1.7% with H. pringsheimii to 12% with Komvophoron kgarii in the family. In the secondary structure of 16S–23S ITS, D1–D1', Box-B, and V3 helix of FBCC-A195 were distinguished from those of other taxa in the Gomontiellaceae. Cyanobacteria represent a cosmopolitan group of oxyphototrophic bacteria. Although free-living cyanobacteria thriving in aquatic habitats as well as cyanobacteria in terrestrial symbiotic systems (lichens) have been extensively studied in Finland, the diversity of terrestrial rock-inhabiting cyanobacteria is overlooked. As part of an ongoing effort to study terrestrial epilithic cyanobacteria from Finland, we isolated two Pseudanabaena-like cyanobacterial strains and characterized them using a polyphasic approach. Although the two strains were firmly placed within the Pseudanabaena clade in the 16S rRNA phylogenetic analyses, relationships among species were better resolved when phylogenetic analyses were based on a concatenate alignment of 16S rRNA gene and 16S–23S Internal Transcribed Spacer (ITS) region. In addition, 16S–23S ITS percent dissimilarity proved to be more useful for species discrimination in Pseudanabaena compared to secondary structures of conserved 16S–23S ITS domains (D1–D1′, box B, V2 and V3 helices). Considering morphological, molecular and ecological information, we describe P. epilithica sp. nov. and P. suomiensis sp. nov. under the provisions of the International Code of Nomenclature for Algae, Fungi and Plants. Neither toxins nor antimicrobial metabolites were detected during LC-MS analysis or antimicrobial susceptibility testing, respectively. Lastly, our phylogenetic analyses revealed that many Pseudanabaena strains are misidentified and highlight the need for taxonomic revision in this poorly studied cyanobacterial genus. An endophytic fungus designated as EIT4T (MCC 9756T) was isolated from the asymptomatic stem tissue of Ephedra gerardiana collected from the Kargil district of Ladakh Union territory, India. Phylogenetic analysis based on concatenated nuclear ribosomal ITS (internal transcribed spacer) and LSU (large ribosomal subunit) sequence datasets revealed its placement within the genus Astragalicola. However, it formed a separate clade exhibiting strong bootstrap support value (80%). The highest nrITS sequence similarity between EIT4T and species of Astragalicola was 95.19% (A. vasilyevae) and 94.26% (A. amorpha), while nrLSU sequence similarity was 99.27% (A. amorpha). Morphologically, EIT4T differs from the other species of Astragalicola in having larger sub-globose to pyriform conidiomata, smaller and mostly unbranched conidiophores, and polymorphic translucent conidia with two terminal guttules. Based on combined cultural, micromorphological, molecular, and phylogenetic analyses, EIT4T represents a novel species in the genus Astragalicola proposed here as Astragalicola ephedrae sp. nov. Zea mays var. amylacea and Zea mays var. indurata are maize ecotypes from Paraguay. Aspergillus section Flavi is the main spoilage fungus of maize under storage conditions. Due to its large intraspecific genetic variability, the accurate identification of this fungal taxonomic group is difficult. In the present study, potential mycotoxigenic strains of Aspergillus section Flavi isolated from Z. mays var. indurata and Z. mays var. amylacea that are marketed in the metropolitan region of Asunción were identified by a polyphasic approach. Based on morphological characters, 211 isolates were confirmed to belong to Aspergillus section Flavi. A subset of 92 strains was identified as Aspergillus flavus by mass spectrometry MALDI-TOF and the strains were classified by MALDI-TOF MS into chemotypes based on their aflatoxins and cyclopiazonic acid production. According to the partial sequencing of ITS and CaM genes, a representative subset of 38 A. flavus strains was confirmed. Overall, 75 A. flavus strains (86%) were characterized as producers of aflatoxins. The co-occurrence of at least two mycotoxins (AF/ZEA, FUM/ZEA, and AF/ZEA/FUM) was detected for five of the Z. mays samples (63%). Considering the high mycological bioburden and mycotoxin contamination, maize marketed in the metropolitan region of Asunción constitutes a potential risk to food safety and public health and requires control measures. Two cyanobacterial strains KUT1-PS and 18C-PS were collected from the soil surface and vernal pool, respectively, from the Basantgarh village, Udhampur district of the union territory of Jammu and Kashmir, India and characterized by a polyphasic approach. The morphological characterization indicated that both the strains showed typical Cylindrospermum-like morphology and probably belonged to the genus Cylindrospermum. Further, phylogenetic interpretations at the genus level were made using the 16S rRNA gene while the 16S-23S ITS region phylogenetic analysis and secondary structure analysis were conducted to enhance the resolution at the species level. The results from the comparative morphological analysis, the 16S rRNA gene percent similarity and phylogenetic analyses, the 16S-23S ITS percent dissimilarity and the ITS secondary structure analyses provided enough evidence that the strain 18C-PS is a representative of Cylindrospermum badium, providing further insights into its ecological distribution and morphological attributes. Additionally, the strain KUT1-PS was a novel species of the genus Cylindrospermum and is referred to herein as Cylindrospermum solincola sp. nov., in accordance with the International Code of Nomenclature for algae, fungi and plants. Chlorophyll (Chl) f was recently identified in a few cyanobacteria as the fifth chlorophyll of oxygenic organisms. In this study, two Leptolyngbya‐like strains of CCNU0012 and CCNU0013 were isolated from a dry ditch in Chongqing city and a brick wall in Mount Emei Scenic Area in China, respectively. These two strains were described as new species: Elainella chongqingensis sp. nov. (Oculatellaceae, Synechococcales) and Pegethrix sichuanica sp. nov. (Oculatellaceae, Synechococcales) by the polyphasic approach based on morphological features, phylogenetic analysis of 16S rRNA gene and secondary structure comparison of 16S‐23S internal transcribed spacer domains. Both strains produced Chl a under white light (WL) but additionally induced Chl f synthesis under far‐red light (FRL). Unexpectedly, the content of Chl f in P. sichuanica was nearly half that in most Chl f‐producing cyanobacteria. Red‐shifted phycobiliproteins were also induced in both strains under FRL conditions. Subsequently, additional absorption peak beyond 700 nm in the FRL spectral region appeared in these two strains. This is the first report of Chl f production induced by FRL in the family Oculatellaceae. This study not only extended the diversity of Chl f‐producing cyanobacteria but also provided precious samples to elucidate the essential binding sites of Chl f within cyanobacterial photosystems. Rice has been the staple food of the Iranian population, and nitrogen availability is usually a limiting factor to high yields in paddy fields. Among the nitrogen-fixing microorganisms, cyanobacteria play a major role in the nitrogen fixation process. In the present work, two nitrogen-fixing cyanobacteria isolated from a paddy field in Golestan Province, Iran, were analyzed using a polyphasic approach including morphological and molecular analyses. Complete 16S rRNA gene sequences were analyzed phylogenetically coupled with the 16S-23S internal transcribed spacer (ITS) secondary structures. Sequences of the rbcL, tufA, psbA and rpoC1 genes were investigated as additional determinants. The 16S rRNA phylogenetic analysis grouped the new isolates into the cluster of the genus Aliinostoc, which was supported by rbcL, tufA, psbA and rpoC1 phylogenetic trees. The 16S-23S ITS-folded structures of the D1-D1′ and Box-B regions differentiated the isolates from published Aliinostoc. We studied nineteen populations of A. platensis (Microcoleaceae, Cyanobacteria), from Pantanal (Brazil) shallow, saline and alkaline lakes. The lakes are connected to the Paraná and Paraguay Rivers Basins, and during wet seasons, the waters flow towards La Plata River estuary. Morphology of natural populations and cultures were analyzed using optical microscope, and 16S rDNA sequences were used for the BI and ML phylogenetic analysis. The morphological analysis shows that our populations fit in the original description of A. platensis, but with additional aerotopes. Also, it is evident that these structures are facultative and the species is planktonic, rather than benthic without aerotopes, as originally described. The phylogenetic analysis shows our strains in the monophyletic Limnospira clade. Considering that, in this paper we transfer the species A. platensis to the genus Limnospira, based on phylogenetic and morphological data. This new taxonomical combination is supported also by ecological data, and indicates that the species is more related and abundant in Pantanal, than in La Plata Basin, region from where it was originally described. According to our results, L. platensis is planktonic or benthic and typical from saline, alkaline and warm waters.

Cartusia hunanesis sp. nov. was isolated from a stream in China, and two strains (ZJJ02 and ZJJ03) of which were inquired using morphological features, ecological evidence, and molecular data consisting of the 16S rRNA gene and 16S–23S rRNA intergenic transcribed spacer (ITS) region. Cartusia hunanesis varies from the type species Cartusia fontana by having only a single trichome in the sheath and large granules near the cross wall. The investigated strains of C. hunanesis were revealed to be a sister clade of C. fontana, according to the phylogenetic analysis based on the 16S rRNA gene. In addition, the Cartusia was clustered with the family Oculatellaceae members, genera Pegethrix and Elainella. These two strains displayed 97.6% similarity to C. fontana. The ITS region of C. hunanesis was found to be considerably distinct from that of C. fontana in terms of the secondary structure, which demonstrated that C. hunanesis is a novel species owing to the divergences in its morphological and genetic data compared with the related C. fontana. Cyanobacterial taxonomy is facing a period of rapid changes thanks to the ease of 16S rRNA gene sequencing and established workflows for description of new taxa. Since the last comprehensive review of the cyanobacterial system in 2014 until 2021, at least 273 species in 140 genera were newly described. These taxa were mainly placed into previously defined orders and families although several new families were proposed. However, the classification of most taxa still relied on hierarchical relationships inherited from the classical morphological taxonomy. Similarly, the obviously polyphyletic orders such as Synechococcales and Oscillatoriales were left unchanged. In this study, the rising number of genomic sequences of cyanobacteria and well‐described reference strains allowed us to reconstruct a robust phylogenomic tree for taxonomic purposes. A less robust but better sampled 16S rRNA gene phylogeny was mapped to the phylogenomic backbone. Based on both these phylogenies, a polyphasic classification throughout the whole phylum of Cyanobacteria was created, with ten new orders and fifteen new families. The proposed system of cyanobacterial orders and families relied on a phylogenomic tree but still employed phenotypic apomorphies where possible to make it useful for professionals in the field. It was, however, confirmed that morphological convergence of phylogenetically distant taxa was a frequent phenomenon in cyanobacteria. Moreover, the limited phylogenetic informativeness of the 16S rRNA gene, resulting in ambiguous phylogenies above the genus level, emphasized the integration of genomic data as a prerequisite for the conclusive taxonomic placement of a vast number of cyanobacterial genera in the future. Six strains of subaerial cyanobacteria were isolated from a Gomso saltern in the Republic of Korea, all of which were studied using morphological and molecular traits. Trichotorquatus salinus sp. nov. was studied using a light microscope (LM), transmission electron microscope (TEM), 16S rRNA, 16S–23S ITS region, and ecological data. T. salinus is a thin and simple filament with a false branch and a firm collar sheath. The phylogenetic analyses of 16S rRNA revealed that Trichotorquatus formed a monophyletic lineage and the strains of T. salinus formed a distinct clade among the species in the genus Trichotorquatus. In the statistical analysis, the inter-species genetic distance of the five species of Trichotorquatus, including T. salinus, is shown to be greater than the distance of the previously reported species of Trichotorquatus. Additionally, 16S–23S ITS gene sequences between T. salinus and four species of Trichotorquatus showed dissimilarities of 55.3–59.4%. In the secondary structure of 16S–23S ITS region (type 2 operon), D1–D1′, Box-B, and V3 helix of T. salinus were different from the other taxa in the genus Trichotorquatus. These results demonstrate that T. salinus sp. nov. has unique morphological, ecological, and molecular traits. Chili anthracnose caused by Colletotrichum spp. is a significant disease of chili cultivation in Indonesia. The current taxonomic status of Colletotrichum spp. has been rearranged due to a well-developed molecular study. Several species of Colletotrichum were re-identified after being analyzed by a polyphasic approach. The polyphasic method in this research combines morpho-cultural and molecular identification using ITS-rDNA and GADPH region. The pathogenicity test would be more reliable in identifying the species complexes of Colletotrichum that are difficult to differentiate. This study applies a polyphasic approach to identify Colletotrichum species causing chili anthracnose, especially in fruits collected from chili fields in Yogyakarta, Indonesia, and observe its pathogenicity in wounded and unwounded inoculation. The results of those combination methods showed that isolates collected from chili fields in Yogyakarta were C. scovillei. The pathogenicity test revealed that the fungus caused anthracnose disease in wounded and unwounded OR Twist 42 chili fruits. The unwounded infection was confirmed by amplification of the ChEC3 pathogenicity gene 24 h after inoculation, showing that appressoria production may possess an essential role in the unwounded inoculation. In this study, 18 strains of cyanobacteria were isolated from seven sites, including the Han River and Anseong Stream in Republic of Korea, and we propose these isolated strains as Limnonema gen. nov., belonging to the Aerosakkonemataceae family of the Oscillatoriales order, and also, as L. hangangris sp. nov. and L. anseonga sp. nov. These strains were identified based on morphological data using a light microscope and a transmission electron microscope, and molecular data using 16S rRNA and 16S–23S ITS gene sequences. The genus Limnonema was mainly collected as planktons, and some L. hangangris (ACKU-695–697) appeared as epilithic cyanobacteria. The genus Limnonema showed filamentous trichomes, intracellular gas vacuoles, and irregular thylakoids arrangement, which was distinct from genera Cephalothrix, Microseira, and Potamosiphon belonging to the family Aerosakkonemataceae. Moreover, the cell widths of genus Limnonema were narrower than those of genus Aerosakkonema, which is the type genus of the family Aerosakkonemataceae, and L. anseonga contained more cells with wider widths than those of L. hangangris. In the 16S rRNA gene sequence phylogeny, genus Limnonema belonged to the family Aerosakkonemataceae and was distinguished from its close relatives, genera Aerosakkonema and Cephalothrix, and L. hangangris and L. anseonga formed different branches. In 16S rRNA gene sequence similarity, genus Limnonema showed 95.4–95.9% and 93.6–94.4% similarity with genera Aerosakkonema and Cephalothrix, respectively, and L. hangangris and L. anseonga showed 97.6–97.7% similarity between each other. In the 16S–23S ITS secondary structure, the D1–D1′, Box-B, and V3 helices of genus Limnonema were distinguished from genera belonging to the family Aerosakkonemataceae, and the V3 helices of L. hangangris and L. anseonga were also different from each other.

Heterocyte-forming cyanobacteria form symbiotic relationships with several lineages of plants. Here, twenty (20) strains of endosymbiotic cyanobacteria (cyanobionts) with Nostoc-like morphologies were isolated from the highly specialized coralloid roots of five host species in Cycadales—Cycas debaoensis, C. fairylakea, C. elongata, Ceratozamia robusta, and Macrozamia moorei. Molecular phylogeny based on the 16S rRNA gene placed these strains into seven different taxa within the Nostocaceae, specifically under the genera Desmonostoc and Dendronalium. The percent dissimilarity and unique patterns in the secondary structures of the D1-D1′, Box-B, V2, and V3 helices, which were based on the 16S–23S rRNA internal transcribed spacer (ITS) regions, supported three distinct species in Desmonostoc. These three morphologically distinct novel species are described in this report: Desmonostoc debaoense sp. nov., Desmonostoc meilinense sp. nov., and Desmonostoc xianhuense sp. nov. Other investigated strains were phylogenetically identified as members of the recently discovered genus Dendronalium and represent the first report of association of that genus with cycads. Our findings suggest that the order Cycadales hosts diverse species of cyanobionts in their coralloid roots and that many potential unreported or novel taxa are present in cycads occurring in their natural habitat and await discovery.

In this study, we isolated and identified cyanobacterial strains from paddy soil in Hanoi (Vietnam) and investigated their cytotoxic activities. Five isolated cyanobacterial strains showed distinctive profiles of gene sequences (rRNA 16S and rbcL), phylogenetic placements, and morphological characteristics. Based on the polyphasic evaluation, they were classified as Scytonema bilaspurense NK13, Hapalosiphon welwitschii MD2411, Aulosira sp. XN1103, Desikacharya sp. NS2000, and Desmonostoc sp. NK1813. The cytotoxic screening revealed that the extract of strain Scytonema bilaspurense NK13 exhibited potent cytotoxic activities against four human cell lines of HeLa cells, OVCAR-8 cells, HaCaT cells, and HEK-293T cells, with IC50 values of 3.8, 34.2, 21.6, and 0.6μg/mL, respectively. Microcystis-dominated blooms cause environmental and ecological impacts worldwide. However, it is sometimes challenging to correctly identify Microcystis and its related genera. Radiocystis fernandoi Komárek et Komáková-Legnerová 1993, characterized by large cells and correspondingly large colonies with gas vesicles, has been frequently found in cyanobacterial blooms in Brazil. However, its molecular and phylogenetic identity is unknown because no valuable DNA sequences are available in GenBank. In this study, a strain with R. fernandoi morphology was successfully isolated from a cyanobacterial bloom sample in Lake Erhai, a subtropical plateau lake in China. Molecular characterization and phylogenetic analyses based on 16S rRNA and cpcBA genes revealed that the strain is different from the type species of the genus Radiocystis but formed a well-supported clade with the Microcystis species. The comparative results of the ITS region between 16S–23S rRNA genes also showed that the strain had very similar secondary structures to the species of Microcystis, supporting it as a new member of the genus Microcystis. Therefore, we propose the transfer of Radiocystis fernandoi into Microcystis fernandoi comb. nov. The correct identification and further taxonomic procedure for the bloom-forming cyanobacterial genera, such as Microcystis, Radiocystis, and Sphaerocavum, are also discussed in this study. Cyanobacteria represent a cosmopolitan group of oxyphototrophic bacteria. Although free-living cyanobacteria thriving in aquatic habitats as well as cyanobacteria in terrestrial symbiotic systems (lichens) have been extensively studied in Finland, the diversity of terrestrial rock-inhabiting cyanobacteria is overlooked. As part of an ongoing effort to study terrestrial epilithic cyanobacteria from Finland, we isolated two Pseudanabaena-like cyanobacterial strains and characterized them using a polyphasic approach. Although the two strains were firmly placed within the Pseudanabaena clade in the 16S rRNA phylogenetic analyses, relationships among species were better resolved when phylogenetic analyses were based on a concatenate alignment of 16S rRNA gene and 16S–23S Internal Transcribed Spacer (ITS) region. In addition, 16S–23S ITS percent dissimilarity proved to be more useful for species discrimination in Pseudanabaena compared to secondary structures of conserved 16S–23S ITS domains (D1–D1′, box B, V2 and V3 helices). Considering morphological, molecular and ecological information, we describe P. epilithica sp. nov. and P. suomiensis sp. nov. under the provisions of the International Code of Nomenclature for Algae, Fungi and Plants. Neither toxins nor antimicrobial metabolites were detected during LC-MS analysis or antimicrobial susceptibility testing, respectively. Lastly, our phylogenetic analyses revealed that many Pseudanabaena strains are misidentified and highlight the need for taxonomic revision in this poorly studied cyanobacterial genus.

Soil cyanobacterium, FBCC-A195 were isolated from the soil underneath the Jangnak Bridge, Republic of Korea. The FBCC-A195 was studied using light microscopy (LM), transmission electron microscopy (TEM), 16S rRNA, 16S–23S ITS, and ecological data. FBCC-A195 showed the circular cross-section of the trichome, broad mucilaginous envelopes, a swirl-like pattern of thylakoids inside the cell, and the lowest length/width ratio within the genus. In the 16S rRNA phylogeny, FBCC-A195 showed a sister relationship of Hormoscilla and belonged to the family Gomontiellaceae. The p-distance of FBCC-A195 ranged from 1.7% with H. pringsheimii to 12% with Komvophoron kgarii in the family. In the secondary structure of 16S–23S ITS, D1–D1’, Box-B, and V3 helix of FBCC-A195 were distinguished from those of other taxa in the Gomontiellaceae. Based upon morphological, ecological, and molecular traits, Katagnymene terrestris proved to be a unique and novel species of the Katagnymene.

Zea mays var. amylacea and Zea mays var. indurata are maize ecotypes from Paraguay. Aspergillus section Flavi is the main spoilage fungus of maize under storage conditions. Due to its large intraspecific genetic variability, the accurate identification of this fungal taxonomic group is difficult. In the present study, potential mycotoxigenic strains of Aspergillus section Flavi isolated from Z. mays var. indurata and Z. mays var. amylacea that are marketed in the metropolitan region of Asunción were identified by a polyphasic approach. Based on morphological characters, 211 isolates were confirmed to belong to Aspergillus section Flavi. A subset of 92 strains was identified as Aspergillus flavus by mass spectrometry MALDI-TOF and the strains were classified by MALDI-TOF MS into chemotypes based on their aflatoxins and cyclopiazonic acid production. According to the partial sequencing of ITS and CaM genes, a representative subset of 38 A. flavus strains was confirmed. Overall, 75 A. flavus strains (86%) were characterized as producers of aflatoxins. The co-occurrence of at least two mycotoxins (AF/ZEA, FUM/ZEA, and AF/ZEA/FUM) was detected for five of the Z. mays samples (63%). Considering the high mycological bioburden and mycotoxin contamination, maize marketed in the metropolitan region of Asunción constitutes a potential risk to food safety and public health and requires control measures.

Cyanobacteria are a group of oxygenic photosynthetic prokaryotes found in almost all habitats on earth including those characterized as extreme environments. It has been observed that the number of studies dealing with the biodiversity of extremophilic cyanobacteria is limited while studies exploring their bioactive potential are even scarcer. The taxonomy of three Nostoc-like cyanobacterial strains isolated from a shallow lake in Brazil was studied by applying a polyphasic approach. The bioactive potential of the strains was also evaluated using antimicrobial susceptibility testing. The metabolites present in the bioactive HPLC fractions were identified by UPLC/ESI/Q-TOF. Based on our phylogenetic inferences in combination with morphological and ecological information, we describe Aliinostoc alkaliphilum sp. nov., exhibiting antibacterial and antifungal activities. The main bioactive metabolite in all three strains was nocuolin A, which represents the first report of this metabolite in Aliinostoc. Our phylogenetic studies also revealed that many bioactive metabolite-producting strains that are currently assigned to Nostoc belong to other distinct evolutionary lineages. While nostocacean cyanobacteria are ubiquitous and play critical roles in terrestrial ecosystems, their taxonomy and biogeography still entail mysteries. We isolated two Nostoc‐like cyanobacteria from biological soil crusts of the Atacama (Chile) and Mojave (USA) Deserts. An initial 16S rRNA gene phylogeny placed both in monophyly with Mojavia pulchra. Here, we describe two new species of the previously monotypic Mojavia using a polyphasic approach including morphology, 16S rRNA phylogenies, secondary structure, and percent similarity of the 16S‐23S ITS region. Like M. pulchra, both new species produce compact microcolonies, arthrospore‐like akinetes, and monocytes, traits characteristic of the genus. Mojavia aguilerae sp. nov. is morphologically distinct from both other species in producing bluntly conical end cells, abundant enlarged akinetes in multiseriate filaments, and gold‐colored cells during senescence. Mojavia dolomitestris sp. nov. exhibited distinctly firm, light‐colored, compartmentalized mucilage. M. dolomitestris is somewhat cryptic with M. pulchra, but has more densely packed microcolonies, rarity and later onset of brownish sheath pigmentation, and an origin from soils derived from dolomite. The two new species strengthened the position of Mojavia as a robust genus sister to Nostoc. Although 16S rRNA gene data could not separate the Mojavia species from each other, the three species showed distinct dissimilarities in secondary ITS structure and differed greatly from Nostoc sensu stricto. The high dissimilarities between their 16S‐23S ITS regions suggest a long evolutionary history of the three species as separate lineages. Mojavia is an evolutionary and ecologically unique nostocacean genus, and its rarity and restricted habitat point to an urgent need for recognition and protection. [ABSTRACT FROM AUTHOR]

Aims: Explore the diversity of culturable actinobacteria isolated from the Pacific oyster Crassostrea gigas with special emphasis on their antimicrobial activity. Methods and Results: For the characterization of the isolated actinobacteria, a polyphasic approach was adopted and thereby phenotypic descriptions, phylogenetic analysis, evaluations of antimicrobial activities and chemical analyses of crude extracts through HPLC and LC‐HRESIMS were performed. Five strains were isolated from C. gigas. The 16S rRNA gene analysis revealed that three of them were taxonomically affiliated to the genus Streptomyces and the other two strains were related to Micromonospora. High inhibition was detected against different test microorganisms such as Candida albicans, Staphylococcus aureus, Bacillus subtilis and Mycobacterium smegmatis. On the basis of the chemical analysis, 11 compounds from the active fractions of the crude extracts were determined, and 8 were related putatively to previously reported compounds. Conclusions: Actinobacteria isolated from C. gigas represent an interesting reservoir of antimicrobial compounds, and further study to uncover the full capacity of this source is encouraged. Significance and Impact: At present, the study of actinobacteria and their antimicrobial potential from uncommon sources as C. gigas is vital to the development of new therapeutic agents to cope with the widespread resistance of human pathogens.
