Polybrominated diphenyl ethers (PBDEs) and their alternatives (e.g., dechlorane plus (DPs) and decabromodiphenyl ethane (DBDPE)) are ubiquitous in various environmental media. However, limited data is available on these chemicals in edible fish species from the wide-open South China Sea (SCS). In the present study, ten legacy PBDEs and three substitutions (DBDPE and two DPs) were analyzed in 16 wild fish species sampled from the open SCS to investigate their spatial and species-specific variations. The results showed that the total concentrations of PBDEs, DBDPE, and DPs in fish samples were in the range of 1.69-47.6, not detected (nd) to 21.0, and nd to 3.80 ng/g lipid weight (lw), respectively. BDEs 47, 209 and 100 were the dominant target PBDE congeners, representing 49.2%, 17.2% and 9.93% of the total PBDE concentrations, respectively. Higher concentrations of PBDEs, DBDPE, and DPs were found in fish species from the Wanshan Archipelago compared to those from the Mischief Reef and the Yongxing Island, suggesting the significant influence of anthropogenic activities. Species-specific differences in levels of PBDEs were observed, with the order of bathydemersal > demersal > pelagic ≈ reef-associated > benthopelagic species. The average f anti value of all fish samples was 0.68, suggesting commercial DP products as a contamination source. The levels of PBDEs, DPs, and DBDPE in fish samples were relatively low compared with those from other locations around the globe. Schisandra chinensis (Turcz.) Baill. ( S. chinensis ) and Schisandra sphenanthera Rehd. et Wils ( S. sphenanthera ) are called "Wuweizi" in traditional Chinese medicine, and they have distinct clinical applications. To systematically compare the differential characteristics of S. chinensis and S. sphenanthera , this study employed ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-QTOF-MS) and gas chromatography-mass spectrometry (GC-MS) to construct chemical profiles of these two species from different regions. In total, 31 non-volatiles and 37 volatiles were identified in S. chinensis , whereas 40 non-volatiles and 34 volatiles were detected in S. sphenanthera . A multivariate statistical analysis showed that the non-volatiles tigloygomisin P, schisandrol A, schisantherin C, and 6-O-benzoylgomisin O and the volatiles ylangene, γ-muurolene, and β-pinene distinguish these species. Additionally, the metabolism of oxygen free radicals can contribute to the development of various diseases, including cardiovascular and neurodegenerative diseases. Therefore, antioxidant activities were evaluated using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid (ABTS) scavenging assays. The results showed that S. sphenanthera exhibited significantly higher antioxidant potential. A gray relational analysis indicated that the key contributors to the antioxidant activity of S. chinensis were schisandrol A, gomisin G, schisantherin C, pregomisin, gomisin J, and schisantherin B. For S. sphenanthera , the key contributors included gomisin K2, schisantherin B, gomisin J, pregomisin, schisantherin C, schisandrin, gomisin G, schisantherin A, schisanhenol, and α-pinene. The identification of the differential chemical markers and the evaluation of the antioxidant activities provide a foundation for further research into the therapeutic applications of these species. This innovative study provides a robust framework for the quality control and therapeutic application of S. chinensis and S. sphenanthera , offering new insights into their medicinal potential.

Background: As a traditional Chinese herbal medicine, Schisandra chinensis exhibits various effects such as liver protection, blood sugar regulation, blood lipid regulation, immune function regulation, antidepressant activity, etc. However, because of its intricate composition, diverse origins, and medicinal effects depending on complex compound groups, there are differences in the lignan composition of S. chinensis from different origins. Therefore, it is currently difficult to evaluate the quality of medicinal materials from plants of different origins using a single qualitative quality control index., Purpose: This paper aims to investigate the potential relationship between the lignan components of S. chinensis from different origins and to establish stable assessment indices for determining the lignan content of S. chinensis from multiple perspectives., Methods: In this study, we collected S. chinensis samples of seven major origins in China, and randomly sampled 6-9 batches of each origin for a total of 60 batches. The lignan content was determined by HPLC, and its distribution law of the ratio of each lignan component of S. chinensis to Schisandrol A content was analyzed. Combining network pharmacology and differential analysis between samples, the stable and effective substances used as quality markers were determined., Results: There were some correlations among the lignan contents of S. chinensis, some correlations between schisandrin A and other lignans of S. chinensis could be determined. The ratio of each component to the indicator component schisandrol A was evenly distributed and reflected the lignan content of S. chinensis to some extent. Four substances (schisandrol A, schisandrol B, schisantherin A, and schisandrin C) were determined by network pharmacology combined with the analysis results of HCA, PCA and PLS-DA to further optimize the model. They displayed a strong connection with the core target, a large contribution rate to the principal components, and a stable content in each batch of samples, suggesting that these components may be the main active substances of S. chinensis lignans. Therefore, they could be used as main indicators evaluating the advantages and disadvantages of S. chinensis by examining the consistency of component proportions., Conclusion: This method can intuitively evaluate the content of main lignans in S. chinensis. Some Polycyclic Aromatic Compounds (PACs) such as nitrated-PAHs (NPAHs), oxygenated-PAHs (OPAHs) and methyl-PAHs (MPAHs) have attracted significant concern due to derivatives have greater potential to be more toxic at low environmental concentrations compared to their PPAHs, particularly in petrochemical industrial region and its surrounding areas surface soils in China. Hence, this article provides an insight into the fate, sources, impacts, and relevance to the external environment of PAH-derivatives based on important emissions source. Moreover, prospective health risk due to their exposure has also been discussed. In this study, the concentration (10 -3  ng/g) of Ʃ18PPAHs, Ʃ11MPAHs, Ʃ12NPAHs, and Ʃ4OPAHs in the park were 9.67 ± 1.40, 3.24 ± 0.54, 0.03 ± 0.02 and 0.19 ± 0.65, respectively, which were 4.47, 3.89, 2.04 and 1.17 times than of them surrounding the region. A decreasing trend of the low molecular weight (2-4Rings) contribution to the total amount of PAHs, while the fraction of high molecular weight (5-6Rings) species showed the opposite trend. According to the principal component analysis (PCA) and diagnostic ratios indicated PAHs in the soil samples have mixed sources from industrial activities, solid fuel combustion, and heavy traffic. Despite the high concentrations of MPAHs and OPAHs, the toxicity equivalency quotients (TEQs) of them were not calculated due to the lack of toxic equivalent factors (TEF), thus current studies on PAH and derivatives could have underestimated their exposure risks. The quality and sustainable management of soils are crucial for human health and sustainable development, while there is lack of public awareness of the severe issue of soil pollution. Airborne polycyclic aromatic hydrocarbons (PAHs) and their derivatives are of particular concern for population health due to their abundance and toxicity via inhalation. Lung toxicity testing includes exposing lung epithelial cell lines to PAHs in a culture medium containing inorganic species, lipids, proteins, and other biochemicals where the cell response is influenced among others by the toxic chemical accessibility in the medium. While inhalation bioaccessibility of PAHs and other toxicants was previously studied in surrogate lung fluids, studies measuring bioaccessibility in cell culture media are rare. In this work, a method was developed to characterize PAH bioaccessibility in a culture medium used for mouse lung epithelial (FE1) cells. Further, the optimised method was tested using commercially available standard reference material of urban particulate matter (PM) as well as polyurethane foam passive air samplers (PUF-PAS). The method provided a high precision and recovery of analytes, indicating no losses during sample processing and analysis. PAHs had non-linear concentration-responses, with the culture medium approaching saturation with PM concentration of 500 μg mL -1 . The results showed that phenanthrene, a 3-ring PAH, was significantly more bioaccessible than ≥4-ring congeners in the culture medium (up to ∼2.5 folds; p < 0.05). Finally, using pre-deployed PUF-PAS from a residential and an industrial site, five PAHs were found in the culture medium, including naphthalene, phenanthrene, anthracene, fluoranthene, and pyrene. China, one of the two dechlorane plus (DP) producers, might have become a major area of DP pollution. The environmental contamination status of DP in sediments across the whole of China has not yet been studied. In the current study, the pollution levels, spatial distribution, and compositions of DP were investigated comprehensively in surface sediments from 173 black-odorous urban rivers across China for the first time. Total DP concentrations varied from not-detected to 39.71 ng/g dw, with an average concentration of 3.20 ± 4.74 ng/g dw, which was polluted by local emission sources and presented significant differences among different sampling cities. Among the seven administrative regions of China, DP concentrations were the highest in South China and showed a decreasing trend from southeastern coastal areas to northwest inland regions. Spearman's correlation analysis suggested that the gross industrial output, gross domestic product, and daily wastewater treatment capacity were not the principal factors controlling the spatial distribution of DP. The f anti values (the concentration ratios of anti-DP to the sum of anti-DP and syn-DP) varied from 0.19 to 0.88, with those in most sediments falling in the range of DP technical product (0.60 ~ 0.80), suggesting no apparent stereoselective enrichment occurred. Moreover, the anti-Cl 11 -DP was detected in sediments (n.d. ~ 0.40 ng/g dw), which showed significantly and insignificantly positive correlation with the anti-DP levels and f anti , respectively, implying it might mainly originate from the byproduct of DP technical product rather than the dechlorination of anti-DP., (© 2024. In the context of a rising global temperature, biomass burning represents an increasing risk to human health, due to emissions of highly toxic substances such as polycyclic aromatic hydrocarbon (PAHs). Size-segregated particulate matter (PM) was collected in a region within the sugarcane belt of São Paulo state (Brazil), where biomass burning is still frequent, despite the phasing out of manual harvesting preceded by fire. The median of the total concentration of the 15 PAHs determined was 2.3 ± 1.8 ng m -3 (n = 19), where 63% of this content was in PM 1.0 . Concentrations of OPAHs and NPAHs were about an order of magnitude lower. PM 2.5 collected in the dry season, when most of the fires occur, presented PAHs and OPAHs total concentrations three times higher than in the wet season, showing positive correlations with fire foci number and levoglucosan (a biomass burning marker). These results, added to the fact that biomass burning explained 65% of the data variance (PCA analysis), evidenced the importance of this practice as a source of PAHs and OPAHs to the regional atmosphere. Conversely, NPAHs appeared to be mainly derived from diesel-powered vehicles. The B[a]P equivalent concentration was estimated to be 4 times higher in the dry season than in the wet season, and was greatly increased during a local fire event. Cytotoxicity and genotoxicity of PM 1.0 organic extracts were assessed using in vitro tests with human liver HepG2 cells. For both types of tests, significant toxicity was only observed for samples collected during the dry season. Persistent DNA damage that may have impaired the DNA repair system was also observed. Decades of research have established the toxicity of soot particles resulting from incomplete combustion. However, the unique chemical compounds responsible for adverse health effects have remained uncertain. This study utilized mass spectrometry to analyze the chemical composition of extracted soot organics at three oxidation states, aiming to establish quantitative relationships between potentially toxic chemicals and their impact on human alveolar basal epithelial cells (A549) through metabolomics-based evaluations. Targeted analysis using MS/MS indicated that particles with a medium oxidation state contained the highest total abundance of compounds, particularly oxygen-containing polycyclic aromatic hydrocarbons (OPAHs) composed of fused benzene rings and unsaturated carbonyls, which may cause oxidative stress, characterized by the upregulation of three specific metabolites. Further investigation focused on three specific OPAH standards: 1,4-naphthoquinone, 9-fluorenone, and anthranone. Pathway analysis indicated that exposure to these compounds affected transcriptional functions, the tricarboxylic acid cycle, cell proliferation, and the oxidative stress response. Biodiesel combustion emissions had higher concentrations of PAHs, OPAHs, and nitrogen-containing PAHs (NPAHs) compared with other fuels. Quinones and 9,10-anthraquinone were identified as the dominant compounds within the OPAH category. This knowledge enhances our understanding of the compounds contributing to adverse health effects observed in epidemiological studies and highlights the role of aerosol composition in toxicity.

The limited information in existing mass spectral libraries hinders an accurate understanding of the composition, behavior, and toxicity of organic pollutants. In this study, a total of 350 polycyclic aromatic compounds (PACs) in 9 categories were successfully identified in fine particulate matter by gas chromatography high resolution mass spectrometry. Using mass spectra and retention indexes predicted by in silico tools as complementary information, the scope of chemical identification was efficiently expanded by 27%. In addition, quantitative structure-activity relationship models provided toxicity data for over 70% of PACs, facilitating a comprehensive health risk assessment. On the basis of extensive identification, the cumulative noncarcinogenic risk of PACs warranted attention. Meanwhile, the carcinogenic risk of 53 individual analogues was noteworthy. These findings suggest that there is a pressing need for an updated list of priority PACs for routine monitoring and toxicological research since legacy polycyclic aromatic hydrocarbons (PAHs) contributed modestly to the overall abundance (18%) and carcinogenic risk (8%). A toxicological priority index approach was applied for relative chemical ranking considering the environmental occurrence, fate, toxicity, and analytical availability. A list of 39 priority analogues was compiled, which predominantly consisted of high-molecular-weight PAHs and alkyl derivatives. These priority PACs further enhanced source interpretation, and the highest carcinogenic risk was attributed to coal combustion.

Air pollution is a complex mixture of gases and particulate matter (PM) with local and non-local emission sources, resulting in spatiotemporal variability in concentrations and composition, and thus associated health risks. To study this in the greater Stockholm area, a yearlong monitoring campaign with in situ measurements of PM 10 , PM 1 , black carbon, NO x , O 3 , and PM 10 -sampling was performed. The locations included an Urban and a Rural background site and a Highway site. Chemical analysis of PM 10 was performed to quantify monthly levels of polycyclic aromatic compounds (PACs), which together with other air pollution data were used for source apportionment and health risk assessment. Organic extracts from PM 10 were tested for oxidative potential in human bronchial epithelial cells. Strong seasonal patterns were found for most air pollutants including PACs, with higher levels during the winter months than summer e.g., highest levels of PM 10 were detected in March at the Highway site (33.2 μg/m 3 ) and lowest in May at the Rural site (3.6 μg/m 3 ). In general, air pollutant levels at the sites were in the order Highway > Urban > Rural. Multivariate analysis identified several polar PACs, including 6H-Benzo[cd]pyren-6-one, as possible discriminatory markers for these sites. The main sources of particulate pollution for all sites were vehicle exhaust and biomass burning emissions, although diesel exhaust was an important source at the Highway site. In vitro results agreed with air pollutant levels, with higher oxidative potential from the winter samples. Estimated lung cancer cases were in the order PM 10  > NO 2  > PACs for all sites, and with less evident seasonal differences than in vitro results. In conclusion, our study presents novel seasonal data for many PACs together with air pollutants more traditionally included in air quality monitoring. Polycyclic aromatic compounds (PACs) are important hazardous air pollutants in China due to the country's coal-dominant energy structure. In order to reveal the pollution characteristics, sources, toxicity, and pollution historical trends of PACs in the atmosphere of the middle reach of the Huaihe River (MRHR), a large-scale coal-fired power base of China, tree barks and tree cores were collected and employed as passive air samplers and historical trend recorders, and 76 PACs were identified for the first time. ΣPACs in tree barks ranged from 170 to 3800 ng g -1 (mean = 700 ± 720 ng g -1 ), with the high concentrations observed mainly in the coal-mining and coal-bearing area. 16 priority PAHs (PriPAHs) were the predominant substances and accounted for 59 ± 8.3 % of ΣPACs. The combustion of coal and fuel oil was the most significant source of PACs, accounting for 43 % of ΣPACs, followed by the combustion of biomass (30 %) and non-combustion sources (27 %). Based on a bark-air partitioning model, volumetric air concentrations for ΣPACs were calculated to be 450-11,000 ng m -3 (mean = 1600 ± 2000 ng m -3 ). The BaP-toxic equivalent concentrations (TEQ BaP ) of ΣPACs (mean = 9.7 ± 15 ng m -3 ) were significantly higher than the Chinese guideline (1 ng m -3 ) and were mostly caused by coal & fuel oil combustion (55 ± 13 %). High molecular weight PACs were detected in lower percentages in tree cores than in tree barks, indicating that PACs in the particle phase were difficult to enter the tree core. Alkyl polycyclic aromatic hydrocarbons (APAHs) are more toxic and persistent than their parent compounds. In this study, the concentrations of polycyclic aromatic compounds (PACs) in dust, topsoil and coal gangue from Huaibei Coal mine, China were analyzed by gas chromatography-mass spectrometry, confirming APAHs were the dominant pollutants. The mean concentrations of APAHs were substantially higher than those of 16 PAHs in both dust and topsoil. The mean concentration of APAHs in dust was 9197 µg kg -1 , accounting for 80% of the total mean concentration of PACs. The mean concentration of APAHs in topsoil was 2835 µg kg -1 , accounting for 77% of the mean concentration of PACs. Alkyl naphthalenes and alkyl phenanthrenes were the primary pollutants in APAHs. Their mean concentrations in dust and topsoil were 7782 µg kg -1 and 2333 µg kg -1 , respectively. This accounted for 85% and 82% of the concentration of APAHs, respectively. Additionally, low-molecular-weight APAHs dominated the PACs of the coal mine, exhibiting petrogenic characteristics; distribution of C1-C4 NAP and C1-C4 PHE exhibited "bell shape" pattern indicated as petrogenic source. Source identification indicated that the PACs were mainly derived from petrogenic sources and vehicle emissions, followed by biomass and coal burning. Fingerprinting information of dust and topsoil were consistent with coal gangue, indicating that PACs are most likely derived from coal gangue. South Pars Industrial Energy Zone, located in the southwest of Iran along the Persian Gulf coast, encompasses many industrial units in the vicinity of urban areas. This research study investigated the effects of polycyclic aromatic hydrocarbons (PAHs) and polychlorinated biphenyls (PCBs) on human health and the environment. Suspended particulate matters (SPM) in the air sampled, in summer and winter 2019, from ten stations next to industrial units and residential areas. The samples were analyzed by gas chromatography-mass spectrometry (GC-MS). Spatial distribution maps of pollutants in the region were prepared using GIS software. The highest carcinogenic risk due to PAHs and PCBs measured as ([Formula: see text]) and ([Formula: see text], respectively. According to the US Environmental Protection Agency limit ([Formula: see text]), the cancer risks from PAH compounds were significant and need further investigation. The PCB cancer risks were within acceptable ranges. The highest adsorption ratios for PAHs were obtained through skin and PCBs by ingestion. The maximum measured non-carcinogenic hazard indexes (HI) turned out to be 0.037 and 0.023 for PAH and PCB, respectively, and were reported as acceptable risks. The predominant source of PAH in industrial areas was liquid fossil combustion, and in urban areas replaced by coal-wood-sugarcane combustion. Petrochemical complexes, flares, power plants (69%), electric waste disposal sites, and commercial pigments (31%) were reported as PCB sources. Industries activities were the most effective factors in producing the highest level of carcinogenic compounds in the region, and it is necessary to include essential measures in the reform programs., (© 2022. Coal gangue is a by-product of coal, the output of which is as high as 30% of raw coal, whereas only 30% of it is recycled. The leftover remains in the environment from gangue backfilling areas and overlap with residential, agricultural, and industrial areas. Coal gangue accumulated in the environment is easily weathered and oxidized and becomes a source of various pollutants. In this paper, 30 coal gangue samples (fresh and weathered coal gangues) were collected from three mine areas in Huaibei, Anhui province, China. Gas chromatography-triple quadrupole mass spectrometry (GC-MS/MS) was used to qualitatively and quantitatively analyze thirty polycyclic aromatic compounds (PACs), including 16 polycyclic aromatic hydrocarbons (16PAHs), preferentially controlled by the United States Environmental Protection Agency (US EPA), and the corresponding alkylated polycyclic aromatic hydrocarbons (a-PAHs). The results showed that PACs existed objectively in coal gangue, and the content of a-PAHs was higher than that of 16PAHs (average values for 16PAHs ranged from 77.8 to 581 ng/g; average values for a-PAHs ranged from 97.4 to 3179 ng/g). Moreover, coal types not only affected the content and type of PACs but also affected the distribution pattern of a-PAHs at different substitution sites. With the increase of gangue weathering degree, the composition of a-PAHs kept changing; the low ring a-PAHs were more easily diffused to the environment, and the high ring a-PAHs remained enriched in the weathered coal gangue. The correlation analysis showed that the correlation between fluoranthene (FLU) and alkylated fluoranthene (a-FLU) was as high as 94%, and the calculated ratios were not more than 1.5. The basic conclusion is that not only 16PAHs and a-PAHs objectively existed in the coal gangue, but also the characteristic compound belonging to the pollution source of coal gangue oxidation have been discovered. To fully understand the ecological and cumulative effects of mining activities on the surrounding aquatic systems of the Canadian oil sands region, it is essential to understand the consequences of exposure to bitumen-containing soils/sediment from natural geomorphological processes. Both physical and chemical stress on aquatic biota can potentially result from exposure to natural bitumen, resulting from hillslope erosional processes and slumping of bankside soils into the rivers, affecting both riverbed habitat and water quality. The magnitude and duration of bitumen-containing soil's fluvial soils/erosional input into receiving watersheds depends on the interannual variability in the regional hydroclimatology and related seasonal and extreme flow events. The primary objective of this study was to evaluate the potential ecotoxicological effects associated with erosional input of riverbank bitumen soils using controlled exposures. A series of integrated, laboratory-based ecotoxicological bioassays were conducted using organisms with different ecological and functional traits (Daphnia magna (Cladocera), Physa acuta (Gastropoda), and Aliivibrio fischeri (Bacteria)). All model organisms were exposed to elutriates produced from natural bitumen from four different regional rivers: fresh bitumen from shoreline outcrops at the Steepbank River upper and lower reaches (STB-CF and STB-MF, respectively) and the lower-Ells River (EL-MF) and aged, fluvially processed/weathered bitumen from the shoreline of the Athabasca River (ATB-MF). All tested organisms responded negatively to STB-MF and EL-MF elutriates. Low toxicity was also observed in the STB-CF and ATB-MF samples. These results follow the chemical analysis of the parental material and elutriates, where higher levels of metals, polycyclic aromatic compounds and naphthenic acids were detected in the EL-MF sample. In summary, this study shows that eroded and transported bitumen-containing soils and sediments could be a natural source of contaminant exposure to aquatic biota. As part of the pan-Canadian Maternal-Infant Research on Environmental Chemicals (MIREC) study, human milk samples were collected between 2008 and 2011, and analyzed for mirex, an organochlorine insecticide and flame retardant, in addition to dechlorane plus (syn- and anti-DDC-CO), the flame retardant replacement for mirex. Mirex was analyzed separately, using a method for the analysis of existing organochlorine insecticides, while the presence of DDC-CO isomers was determined using a method developed for the detection of emerging flame retardants. Mirex was detected in all samples analyzed (n = 298), while syn- and anti-DDC-CO were present in 61.0% and 79.5% of the samples, respectively (n = 541). Mirex concentrations have declined in human milk since the 1990s. Since this is the first pan-Canadian dataset reporting DDC-CO concentrations in human milk, no temporal comparisons can be made. Maternal age was correlated with concentrations of both compounds although parity did not impact concentrations of either analyte. Given the presence of this relatively recently identified flame retardant (DDC-CO) in human milk from women across Canada, studies to identify dominant sources of this compound are critical. The tissue-specific bioaccumulation of Dechlorane Plus (DP) isomers was investigated in two predator fish species (redtail catfish, RF; and oscar fish, OF) that were feeding on tiger barb (TB), which was exposed to syn-DP and anti-DP isomers. The biotransformation potential of DP isomers was examined by in vitro metabolism using fish liver microsomes. No difference in accumulation behaviors of DP isomers was observed between RF and OF, and the accumulation of both syn- and anti-DP isomers exhibiting a linear increase trend with the exposure time in all fish tissues. The assimilation efficiencies and depuration rates for syn-DP and anti-DP were determined to be the highest in the liver. Biomagnification factors (BMFs) for both syn-DP and anti-DP were higher than one in the serum and gastrointestinal tract of fish, whereas were less than one in the other tissues. The wet-weight concentrations of DP isomers in tissues were significantly correlated with the lipid contents in both fish species, indicating that the tissue distribution of DP isomers occurred through passive diffusion to the lipid compartments in vivo. Tissue-specific compositions of DP isomers were observed, with anti-DP selectively accumulating in the liver, gonad, serum, and gills, whilst syn-DP in the carcass and GI tract. However, after being normalized of all tissues, the fish showed no selective accumulation of DP isomers during the exposure period, and selective accumulation of syn-DP was observed during the depuration period. No potential DP metabolites were detected in the fish tissues and in vitro metabolism systems. The Pacific coast of Canada has a rich marine fauna and a growing human population with increasing potential for pollution releases, but there is currently little overlap between marine wildlife hotspots and ongoing biomonitoring efforts for less bioaccumulative contaminants such as polycyclic aromatic compounds (PAC) and trace metals (metals). We surveyed PACs and metals at marine bird breeding colonies in coastal British Columbia in 2018 by analyzing chemical residues in the soft tissue of bivalve Mytilus sp. mussels collected from stations (n = 3) at seven sites. The concentration of sum PACs (∑ 43 PAC) and high molecular weight (HMW) PACs were highest at the Second Narrows colony in Vancouver Harbour, a highly urbanized and industrialized port within the Salish Sea. For conservation areas, two Salish Sea and three Pacific Ocean coast colonies, PACs were generally lower. However, ∑ 43 PAC, ∑ HMW PAC, and several HMW congeners at the remote site of Triangle Island, a Marine National Wildlife Area, were not significantly different from Second Narrows. The dominant PAC sources at all sites are likely pyrogenic rather than petrogenic, as suggested by PAC profiles, proportion of parent PACs, and source-indicator congeners. For metals, site differences were found for seven out of eight priority metals, but principal component analysis indicated that site differences, such as high mercury and cadmium at offshore sites, are likely related to environmental and biological variables including salinity, condition index, water temperature, and shell length. Polycyclic aromatic compounds (PACs) are organic compounds commonly found in contaminated soil. Previous studies have shown the removal of polycyclic aromatic hydrocarbons (PAHs) in creosote-contaminated soils during steam enhanced extraction (SEE). However, less is known about the removal of alkyl-PAHs and heterocyclic compounds, such as azaarenes, and oxygen- and sulfur-heterocyclic PACs (OPACs and PASHs, respectively). Further, the impact of SEE on the freely dissolved concentration of PACs in soil as well as the soil bioactivity pre- and post-SEE have yet to be addressed. To fulfil these research gaps, chemical and bioanalytical analysis of a creosote-contaminated soil, collected from a U.S. Superfund site, pre- and post-SEE were performed. The decrease of 64 PACs (5-100%) and increase in the concentrations of nine oxygenated-PAHs (OPAHs) (150%) during SEE, some of which are known to be toxic and can potentially contaminate ground water, were observed. The freely dissolved concentrations of PACs in soil were assessed using polyoxymethylene (POM) strips and the concentrations of 66 PACs decreased post-SEE (1-100%). Three in vitro reporter gene bioassays (DR-CALUX®, ERα-CALUX® and anti-AR CALUX®) were used to measure soil bioactivities pre- and post-SEE and all reporter gene bioassays measured soil bioactivity decreases post-SEE. Mass defect suspect screening tentatively identified 27 unique isomers of azaarenes and OPAC in the soil. Polycyclic aromatic compound (PAC) contamination in soil as a result of oil spills is a serious issue because of the huge global demand for fossil energy. This study assessed the vertical variation in polycyclic aromatic hydrocarbons (PAHs), derivatives of PAHs (dPAHs) and bacterial community structure in deep soil with long-term contamination by oil spillage. Our results suggest that the content of total PACs ranged from 1196.6 μg/kg to 14980.9 μg/kg and decreased with depth at all sites. PAHs were the most abundant PACs, with a mean concentration of 6640.7 μg/kg, followed by oxygenated PAHs (mean 156.3 μg/kg) and nitrated PAHs (mean 33.4 μg/kg). PAHs are mainly low molecular weight PACs such as naphthalene, fluorene and phenanthrene, while derivatives of PAHs are all low molecular weight PACs and mainly oxygenated PAHs. Low molecular weight PAHs were an important source of dPAHs under specific conditions. The bacterial community structure showed higher bacterial diversity and lower bacterial richness in shallow soil (2-6 m in depth) than in deep soil (8-10 m in depth). Spearman's analysis confirmed that dramatic bacterial community shifts are a response to contamination. At the genus level, the presence of PACs highly selected for Pseudomonas, belonging to Proteobacteria. Moreover, functional predictions based on Tax4Fun revealed that soil with long-term contamination had a strong potential for PAC degradation. In addition, statistical analysis showed that oxidation-reduction potential (Eh) was closely related to variations of bacterial community composition and function. Finally, Ramlibacter, Pseudomonas, Pseudonocardia, c&#95;MB-A2-108, f&#95;Amb-16S-1323, and Qipengyuania were identified by cooccurrence network analysis as keystone taxa contributing to the maintenance of bacterial ecological function. Dechlorane plus (DP) is a potential persistent organic pollutant and its distribution in various tissues and organs of terrestrial organisms is currently unknown. DP concentrations in sheep tissues were determined in this study. The DP concentrations in the tissues decreased in the following order: abdominal fat > liver > stomach > heart > outer tenderloin > lung > hind leg meat > kidney > small intestine > tail fat > spleen > brain. Apart from brain and fat, anti-DP is enriched more readily than syn-DP in sheep tissues, but syn-DP is more readily enriched in brain and abdominal fat. The factors influencing DP distributions in sheep tissues were assessed by determining the DP to sheep serum albumin binding forces, binding types, and binding sites by fluorescence spectroscopy. The results indicated that anti-DP more readily binds to sheep serum albumin than does syn-DP. Therefore, sheep serum albumin will more readily transport anti-DP than syn-DP to sheep tissues, and anti-DP will be enriched more than syn-DP in the tissues. The molecular diameter of DP is the main factor affecting DP concentrations in sheep brain and fat because of the blood−brain barrier and because the main source of DP to abdominal fat is dermal contact.

A magnetic hybrid material based on the use of the mixed-ligand Metal-Organic Framework (MOF) PUM198 is proposed for the magnetic dispersive micro solid-phase extraction (MD-μSPE) of the 16 polycyclic aromatic hydrocarbons (PAHs) included in the US-EPA priority pollutants list. PUM198 is a thermally robust MOF characterized by a doubly interpenetrated microporous framework in which Zn 2+ ions and carboxylate groups define 2D planes that are pillared by a bis-pyridine-bis amide ligand containing a biphenyl scaffold. PUM198 revealed to be ideal to adsorb PAHs efficiently through non-covalent interactions. A Plackett-Burman Design followed by a Central Composite Design and the multicriteria method of the desirability functions were applied to find the optimal conditions for the extraction of the investigated PAHs, resulting in a reduced solvent consumption, i.e., 50 μL of solvent per extraction for 5 mL of sample, approximatively 3-20 times lower than those reported in previous studies, thus satisfying the principles of green analytical chemistry. Method validation proved the reliability of the method for the determination of PAHs at trace level, obtaining detection limits in the 6.7-27 ng/L range, good precision with RSDs % lower than 19% and recovery rates in the 99 (±13)-126 (±8)% range near the quantitation limit. Mining activities in the Athabasca oil sands region (AOSR) have contributed to an increase of polycyclic aromatic compounds (PACs) locally. However, many PACs found in the AOSR, and the combined effects of PAC mixtures have not been evaluated for genotoxicity in wildlife. Here, we examine whether mutation frequencies in AOSR river otters are correlated to PAC tissue burdens. We used single-molecule polymerase chain reaction (SM-PCR) to measure the mutant frequency of unstable DNA microsatellite loci in the bone marrow of wild river otters (n = 11) from the AOSR. Microsatellite mutation frequencies were regressed against liver PAC burden (total, low/high molecular weight [LMW/HMW], and parent/alkylated PACs), and to the distances from where the samples were collected to nearby bitumen upgraders. We found that microsatellite mutation frequency was positively correlated with total liver PAC burden. LMW and alkylated PACs were detected at higher levels and had a stronger positive relationship with mutation frequency than HMW (alkylated and parent) PACs. There were no significant relationships detected between mutation frequency and LMW parent PACs or the distance from bitumen upgraders. Furthermore, pyrogenic and petrogenic signatures suggest PACs in animals with high mutation frequencies were associated with combustion processes; although further investigation is warranted, due to limitations of diagnostic ratio determination with biotic models. Our findings support the hypothesis that PACs found in the AOSR increase mutation frequency in wildlife. Despite several studies having addressed the bioaccumulation of Dechlorane Plus (DP) flame retardant in wildlife, there is still a dearth of information for reptiles in general and for snakes in particular. Here, we report the residue levels and trophic transfer of syn-, anti-, and anti-Cl 11 -DP in a frog-eating snake-namely, the striped keelback snake Amphiesma stolata-from a DP hotspot in South China. The concentrations of syn-, anti-, and anti-Cl 11 -DP in A. stolata ranged from 1.06-21.2, 2.13-21.5, and 0.16-10.6 ng/g lipid weight, respectively, with significantly higher levels in males compared with females. Statistical analysis showed that the concentrations of these chemicals were negatively correlated with body sizes (length and mass) of the snake. The fractional abundance of anti-DP (f anti ) did not significantly differ either between the sexes or between A. stolata and its diet (i.e., frogs). However, f anti showed positive correlations with the snake's body size and negative correlations with ∑DP concentrations (summed concentrations of syn- and anti-DP), indicating that body size and DP residue levels are important factors influencing DP isomeric profiles in these snakes. Biomagnification factors estimated based on the relationship between A. stolata and frogs were 0.49 ± 0.01 (mean ± SE), 0.44 ± 0.09, and 1.79 ± 0.54 for syn-, anti-, and anti-Cl 11 -DP, respectively, suggesting trophic dilution of syn- and anti-DP and a mild biomagnification of anti-Cl 11 -DP from frogs to snakes., (Copyright © 2022 Elsevier Ltd. Much attention is drawn to polycyclic aromatic hydrocarbons (PAHs) as an air pollutant due to their toxic, mutagenic and carcinogenic properties. Therefore, to understand the levels, seasonality, sources and potential health risk of PAHs in two distinct geographical locations at Karachi and Mardan in Pakistan, total suspended particle (TSP) samples were collected for over one year period. The average total PAH concentrations were 31.5 ± 24.4 and 199 ± 229 ng/m 3 in Karachi and Mardan, respectively. The significantly lower concentration in Karachi was attributed to diffusion and dilution of the PAHs by the influence of clean air mass from the Arabian sea and high temperature, enhancing the volatilization of the particle phase PAHs to the gas phase. Conversely, the higher concentration (~6 times) in Mardan was due to large influence from local and regional emission sources. A clear seasonality was observed at both the sites, with the higher values in winter and post-monsoon due to higher emissions and less scavenging, and lower values during monsoon season due to the dilution effect. Diagnostic ratios and principal component analysis indicated that PAHs in both sites originated from traffic and mixed combustion sources (fossil fuels and biomass). The average total BaP equivalent concentrations (BaP eq ) in Karachi and Mardan were 3.26 and 34 ng/m 3 , respectively, which were much higher than the WHO guideline of 1 ng/m 3 . The average estimates of incremental lifetime cancer risk from exposure to airborne BaP eq via inhalation indicated a risk to human health from atmospheric PAHs at both sites., (Copyright © 2021. Dechlorane series flame retardants (DECs), e.g. Dechlorane plus (DP), have reportedly showed an increase in consumption since the phase-out of traditional brominated flame retardants (BFRs). Here we investigated DP and 7 structural analogues, as well as its 2 dechlorinated products in 76 surficial sediments from the Pearl River Delta (PRD), one of the three important manufacturing bases of China. The concentration of Σ 8 DECs varied from 28.1 to 38,000 pg g -1 dw in the PRD sediments, dominated by DP and Mirex. Spatially, sedimental DP concentrations were significantly and positively correlated with the municipal gross domestic product (GDP), population and sewage discharge of the PRD cities, but were insignificantly related to their industrial outputs. This indicates that DP in the PRD sediments mainly originated from urban activities instead of industrial ones. Although Mirex has been restricted for several decades, it demonstrated ubiquity in the PRD and considerably high levels in several termite control hot-spots (up to 34,200 pg g -1 ), implying its massive historical use in this subtropical region. Other DECs, however, exhibited quite low abundances, implying their limited applications in this region. In comparison to the historical data, sedimental DP concentrations presented an increasing trend in most rivers in the PRD except the West River. The fractions of anti-DP (f anti ) showed insignificant deviations from its technical value, suggesting that no obvious anti-DP transformation occurred in most PRD sediments. However, anti-Cl 11 -DP, an important dechlorination product of anti-DP, was ubiquitously found in the PRD sediments, and its concentrations were significantly and positively associated with those of anti-DP. Therefore, anti-Cl 11 -DP in the PRD sediments was deemed as the impurity co-emitted with anti-DP rather than its dechlorination byproduct. Ethnopharmacological Relevance: Shengmai San (SMS) has been commonly used as a traditional Chinese medicine for the treatment of cardiovascular disorders, of which drug interactions need to be assessed for the safety concern. There is little evidence for the alterations of hepatic and intestinal drug-metabolizing enzymes after repeated SMS treatments to assess drug interactions., Aim of the Study: The studies aim to illustrate the effects of repeated treatments with SMS on cytochrome P450s (CYPs), reduced nicotinamide adenine dinucleotide (phosphate)-quinone oxidoreductase (NQO), uridine diphosphate-glucuronosyltransferase (UGT), and glutathione S-transferase (GST) using in vivo rat model., Materials and Methods: The SMS was prepared using Schisandrae Fructus, Ginseng Radix, and Ophiopogonis Radix (OR) (1:2:2). Chromatographic analyses of decoctions were performed using ultra-performance liquid chromatography (UPLC) and LC-mass spectrometry. Sprague-Dawley rats were orally treated with the SMS and its component herbal decoctions for 2 or 3 weeks. Hepatic and intestinal enzyme activities were determined. CYP3A expression and the kinetics of intestinal nifedipine oxidation (NFO, a CYP3A marker reaction) were determined., Results: Schisandrol A, schisandrin B, ginsenoside Rb1 and ophiopogonin D were identified in SMS. SMS selectively suppressed intestinal, but not hepatic, NFO activity in a dose- and time-dependent manner. Hepatic and intestinal UGT, NQO and GST activities were not affected. A 3-week SMS treatment decreased the maximal velocity of intestinal NFO by 50%, while the CYP3A protein level remained unchanged. Among SMS component herbs, the decoction of OR decreased intestinal NFO activity., Conclusions: These findings demonstrate that 3-week treatment with SMS and OR suppress intestinal, but not hepatic CYP3A function. Here, a new gas chromatography-atmospheric pressure photoionization-high-resolution mass spectrometry (GC-APPI-HRMS) method combined with selective pressurized liquid extraction (sPLE) has been developed for the selective determination of Dechlorane Plus (DP) and its related compounds in gull egg samples used as a bioindicator of contamination. To the best of our knowledge, this is the first time these compounds have been analyzed by GC-MS using atmospheric pressure photoionization (APPI). Negative ion dopant-assisted APPI using vapors of diethyl ether and a source temperature of 250 °C provided high ionization efficiencies and mass spectra characterized by intense in-source fragment ions as well as the presence of molecular ion and characteristic cluster ions containing oxygen atoms in their chemical structure. This made it possible to improve the selectivity in the determination of these compounds compared to that obtained with traditional GC-MS ion sources. Under optimized conditions, the sPLE GC-APPI-HRMS (Orbitrap) method provided high recoveries (> 91%), good precisions (RSD% < 12%), and low method limits of detection (0.1-3.5 pg g -1 wet weight). The developed methodology has been applied to the determination of DP and related compounds in eggs of two gull species (L. michahellis and L. audouinii) from several Spanish protected areas. The results obtained showed significant differences in the DP concentration profiles in eggs from different gull breeding locations and between gull species of the same protected area. These results demonstrated the good performance of the GC-APPI-HRMS system to achieve a selective and sensitive determination of DP and related compounds in complex environmental samples.

In this study, roasting was applied to enhance the volatile compound content and antioxidant activity of hot-air dried omija (Schisandra chinensis Baillon) fruit. The major volatile compounds were furfural, 1,8-cineole and terpinen-4-ol. Total volatile compound concentration in omija roasted at 150 ℃ for 15 min was approximately 4 times higher than that in hot-air dried omija. Contents of monoterpenes and sesquiterpenes in roasted omija were significantly increased, compared to that of the hot-air dried omija (P < 0.05). The contents of schizandrin in extracts of hot-air dried omija and omija roasted at 150 ℃ for 10 min were determined to be 28.9 and 106.5 mg/100 g extract, respectively. The content of gomisin A from roasted omija was about 5 times higher than that of hot-air dried omija. Dechlorane Plus (DP) is found widely in the environment. It is important to study DP enrichment and biomagnification in terrestrial ecosystems to improve our understanding of the possible effects of DP on the environment and human health. A total of 90 samples, including plant and animal tissues, were collected from Xilingol Prairie in Inner Mongolia, China. The DP concentrations in different species were assessed, and transmission of DP through food webs containing ectotherms and endotherms was assessed. The compound was detected in the biotic samples (plant; range 0.133-0.422 ng/g dry wt), in animal muscle (range: not dected-5.70 ng/g lipid wt), and in animal hair (range: not dected-2.03 ng/g dry wt), indicating that DP is present in remote environments such as Xilingol Prairie. These findings suggest that DP can undergo long-distance transport in the environment. Biomagnification factors (ectotherms: range 0.146-88.0, endotherms: range 0.866-17.2) and anti-DP/total DP concentration ratios (f anti values of 0.412-0.787) for the prairie animals were calculated. Ectotherms were found to selectively enrich syn-DP, and stereoselective enrichment increased moving up the food web. Dechlorane Plus is a polychlorinated compound which has exclusively anthropic origin. This compound has been manufactured for close to 60 years for various applications, but mainly as flame retardant. Dechlorane Plus and other Dechlorane-related compounds (DRCs) are currently marketed as a replacement for Dechlorane, also known as Mirex, banned in 1978. These compounds share comparable properties to persistent organic pollutants (POPs), such as persistence in the environment, high lipophilicity, bioaccumulation through the food web and adverse effects on the environment and human health. Despite their long production history, they have been only recently reported in various environmental compartments, such as air, soil, and foodstuff. The aim of this review is to provide a picture of the current state of knowledge on worldwide DRC levels in food, in order to highlight gaps and research needs. The review compares the data on DRC contamination available in literature, considering different food categories and sampling country. In addition, it is specified whether the data were obtained from studies on foodstuff to estimate dietary intake, to evaluate the contamination near the e-waste treatment area or for environmental monitoring purposes.

Cell membrane chromatography (CMC) is effective and widely used in drug screening, especially for the analysis of complex matrixes. However, it is time-consuming and costly given that cells or animals are employed for activity confirmation, which leads to a large amount of waste being produced if the result is negative. Stepwise frontal analysis is employed to saturate the affinity stationary phase, by using a series of low- to high-concentration solutions which resultantly form a staircase pattern. In doing so, the waste of samples, caused by the balancing process, can be avoided. In this study, stepwise frontal analysis coupled with a CMC system was performed for screening and characterizing the affinity of an active compound from wuweizi. Schizandrin A was screened and identified by α 1A AR /CMC coupled with UHPLC-MS/MS. By comparing the values obtained with those related to the equilibrium dissociation constant (K d ) calculated by zonal elution, the accuracy of the stepwise frontal analysis was verified. Subsequently, the type of affinity force between Schizandrin A and α 1A AR was studied by thermodynamic parameters. Moreover, schizandrin A showed an antagonistic effect on phenylephrine-induced contractions, which relax prostate muscle strips in a non-competitive antagonism manner. It has already suggested that the active compound, schizandrin A, could be used as a lead compound for the treatment of benign prostate hyperplasia (BPH) and should be further studied. Dechlorane Plus (DP) is an unregulated, highly chlorinated flame retardant. It has been manufactured from past 40 years but its presence in the environment was initially reported in 2006. Later, it has been found in various biotic and abiotic environmental matrices. However, little attention has been paid to monitor its presence in Asia. Many studies have reported the occurrence of DP in the environment of Asia, yet the data are scarce, and studies are limited to few regions. The objective of present review is to summarize the occurrence, distribution, and toxicity of this ubiquitous pollutant in various environmental matrices (biotic and abiotic). DP has also been reported in the areas with no emission sources, which proves its long-range transport. Moreover, urbanization and industrialization also affect the distribution of DP, i.e., high levels of DP have been found in urban areas relative to the rural. Tidal movement also incorporates in transport of DP across the aquatic system. Further, bioaccumulation trend of DP in various tissues is kidney > liver > muscle tissues, whereas, blood brain barrier resists its accumulation in brain tissues. Additionally, gender-based accumulation trends revealed high DP levels in females in comparison to males due to strong metabolism of males. Furthermore, methodological aspects and instrumental analysis used in previous studies have also been summarized here. However, data on biomagnification in aquatic ecosystem and bioaccumulation of DP in terrestrial food web are still scarce. Toxicity behavior of syn-DP and anti-DP is still unknown which might gain the interest for future studies.

Increasing our understanding of the bioavailable fractions of polycyclic aromatic compounds (PACs) in an aquatic environment is important for the assessment of the environmental and human health risks posed by PACs. More importantly, the behaviour of polar polycyclic aromatic hydrocarbons (polar PAHs), which are metabolites of legacy PAHs, are yet to be understood. We, therefore, carried out a study involving Sydney rock oysters (Saccostrea glomerata) sourced from two locations, that had been exposed to PAH contamination, within an Australian south-east estuary. Biomonitoring of these oysters, following relocation from the estuary to a relatively isolated waterway, was done at 24 and 72 h after deployment and subsequently at 7, 14, 28, 52 and 86 days. Control samples from Camden Haven River were sampled for PAC analyses just before deployment, after 28 days and at the end of the study (day 86). Lipid-normalised concentrations in oyster tissues across the 86-day sampling duration, elimination rate constants (k 2 ), biological half-lives (t 1/2 ) and time required to reach 95% of steady-state (t 95 ) were reported for parent PAHs and the less-monitored polar PAHs including nitrated/oxygenated/heterocyclic PAHs (NPAHs, oxyPAHs and HPAHs) for the three differently sourced oyster types. Most of the depurating PAHs and NPAHs, as well as 9-FLO (oxyPAH), had k 2 values significantly different from zero (p < 0.05). All other oxyPAHs and HPAHs showed no clear depuration, with their concentrations remaining similar. Improving risk assessment and remediation rests on better understanding of contaminant bioavailability. Despite their strong toxicological attributes, little is known about the partitioning behaviour and bioavailability of polar polycyclic aromatic hydrocarbons (PAHs) in aquatic environments. The present study provides an insight into the bioavailable fractions of polar PAHs and their parent analogues in the tissues of the Sydney rock oyster, Saccostrea glomerata, a model aquatic bio-indicator organism. The concentration and distribution patterns of parent and polar PAHs including oxygenated PAHs (oxyPAHs), nitrated PAHs (NPAHs) and heterocyclic PAHs (HPAHs) were determined in water, sediment and oysters from an ecologically and economically important estuary of New South Wales, Australia. Total concentrations of PAHs, oxyPAHs, NPAHs and HPAHs were higher in sediments compared to oyster tissue and water. For most polar PAHs, total concentrations for water, sediment and oyster samples were <1 μg/g (μg/l for water) while parent PAH concentrations were several orders of magnitude higher. Computed biota-sediment accumulation factors (BSAFs) on lipid-normalized oyster concentrations revealed that while ∑oxyPAHs and ∑HPAHs exhibited low accumulation from sediment to oyster tissues (BSAF <1), ∑PAHs and ∑NPAH were found to be accumulated at high levels (BSAF >1). BSAF individual computation showed that bioaccumulation of nine investigated HPAHs in oyster tissues were relatively low and only 2-EAQ (oxyPAH) and 1N-NAP (NPAH) showed high levels of accumulation in oyster tissues, similar to parent PAHs. To the best of our knowledge, this is the first known study on the bioavailability of polar and non-polar PAHs in an Australian aquatic environment. A wide variety of plant raw materials thought to promote health are used as herbal medicines as well as foods. However, there is no legal maximum or minimum concentration limit on any herbal compound when these plant raw materials are used in processed foods. Legally, these processed foods are regulated only for harmful substances, and there is no other guarantee of their contents. Therefore, the objective of this study was to determine the concentrations of 12 herbal compounds (nodakenin, decursin, decursinol angelate, morroniside, loganin, glycyrrhizic acid, liquiritigenin, puerarin, daidzin, schisandrin, gomisin A, gomisin N) in commonly used plant raw materials, such as "Angelica Gigas root", "Cornus Fruit", "Liquorice Root", "Pueraria Root", and "Schisandra Fruit"; and also in 45 processed foods, using high-performance liquid chromatography-tandem mass spectrometry (LC-MS/MS). Method validation was performed successfully using the parameters of specificity, linearity, limit of detection (LOD), limit of quantification (LOQ), accuracy, precision, matrix effect, extraction recovery, and stability. The 12 herbal compounds were determined to be present in all the foods advertised as containing each ingredient, although in very low concentrations in some cases. Three solid samples labelled as 100% pure material from one herbal species also contained herbal compounds found in others, so that intentional or unintentional adulteration was suspected.

Pyrene-based cyclophanes have been synthesized with the aim to realize a bellows-type sensing mechanism for the ratiometric detection of nucleotide concentrations in a buffered aqueous solution. The sensing mechanism involves the encapsulation of a nucleobase between two pyrene rings, which affects the monomer-excimer equilibrium of the receptor in the excited state. The nature of the spacer and its connection pattern to pyrene rings have been varied to achieve high selectivity for ATP. The 1,8-substituted pyrene-based cyclophane with the 2,2'-diaminodiethylamine spacer demonstrates the best selectivity for ATP showing a 50-fold increase in the monomer-excimer emission ratio upon saturation with the nucleotide. The receptor can detect ATP within the biological concentrations range over a wide pH range. NMR and spectroscopic studies have revealed the importance of hydrogen bonding and stacking interactions for achieving a required receptor selectivity. Great ecological and human health risks may arise from the presence of polycyclic aromatic hydrocarbons (PAHs) in aquatic environments and particularly in sediments, where they often partition. In spite of the apparent risk, knowledge about PAHs and their polar derivatives in sediments is limited. We, therefore, carried out an assessment of the concentrations of parent PAHs and their derivatives (polar PAHs) in sediments of Lake Macquarie: the largest saltwater lake in the southern hemisphere. A total of 31 sediment samples along the pollution prone western shoreline of the estuary were analysed. Multiple source apportionment methods were used to investigate PAH sources contributing to parent and polar PAH concentrations in the estuarine sediments. Concentration levels were highest for high molecular weight (HMW) PAHs compared to low molecular weight (LMW) PAHs. The highest PAH concentrations were recorded for oxygenated PAHs (oxy-PAHs) compared to parent and other polar PAHs. Polycyclic aromatic hydrocarbon diagnostic ratios and compositional analysis showed that PAHs in Lake Macquarie were predominantly pyrogenic exhibiting strong positive correlation (R 2  = 0.972) with total PAH concentrations. Principal Component Analysis (PCA) identified three groupings of PAHs with oxy-PAHs and NPAHs dominating (40.2%). Carbazole, a heterocyclic PAH, was also a prominent contributor to sediment PAH concentrations. In situ chemical oxidations are known to remediate PAH contaminations in groundwater and soils. In this study, batch-scale oxidations aim to compare the PAC (polycyclic aromatic compound) degradation of three oxidation processes traditionally applied for soil treatment: permanganate, heat-activated persulfate (60 °C) and Fenton-like activated by magnetite, to results obtained with ferrates (Fe VI ). Widely studied for water treatments, ferrates are efficient on a wide range of pollutants with the advantage of producing nontoxic ferric sludge after reaction. However, fewer works focus on their action on soil, especially on semi-industrial grade ferrates (compatible with field application). Oxidations were carried out on sand spiked with dense non-aqueous phase liquid (DNAPL) sampled in the groundwater of a former coking plant. Conventional 16 US-EPA PAHs and polar PACs were monitored, especially potential oxygenated by-products that can be more harmful than parent-PAHs. After seven reaction days, only the Fenton-like showed limited degradation. Highest efficiencies were obtained for heat-activated persulfate with no O-PAC ketones formed. Permanganate gave important degradation, but ketones were generated in large amount. The tested ferrates not only gave slightly lower yields due to their auto-decomposition but also induced O-PAC ketone production, suggesting a reactional pathway dominated by oxidoreductive electron transfer, rather than a radical one.

The understanding of cellular Cd 2+ accumulation and toxicity is hampered by a lack of fluorescent indicators selective for intracellular free Cd 2+ ([Cd 2+ ] i ). In this study, we used depolarized MIN6 mouse pancreatic beta cells as a model for evaluating [Cd 2+ ] i detection with commercially available fluorescent probes, most of which have been traditionally used to visualize [Ca 2+ ] i and [Zn 2+ ] i . We trialed a panel of 12 probes including fura-2, FluoZin-3, Leadmium Green, Rhod-5N, indo-1, Fluo-5N, and others. We found that the [Zn 2+ ] i probe FluoZin-3 and the traditional [Ca 2+ ] i probe fura-2 responded most consistently and robustly to [Cd 2+ ] i accumulation mediated by voltage-gated calcium channels. While selective detection of [Cd 2+ ] i by fura-2 required the omission of Ca 2+ from extracellular buffers, FluoZin-3 responded to [Cd 2+ ] i similarly in the presence or absence of extracellular Ca 2+ . Furthermore, we showed that FluoZin-3 and fura-2 can be used together for simultaneous monitoring of [Ca 2+ ] i and [Cd 2+ ] i in the same cells. None of the other fluorophores tested were effective [Cd 2+ ] i detectors in this model.

Dechlorane Related Compounds (DRCs), including Dechlorane Plus (syn/anti-DP or syn/anti-DDC-CO) and related compounds (Dec-601 or DDC-ID, Dec-602 or DDC-DBF, Dec-603 or DDC-Ant and Chlordene Plus or DDC-PDD), are a group of polychlorinated flame retardants of concern since they were first reported in various environmental and biota matrices about one decade ago. In this work, we investigated the dietary intake of the Lebanese population to these lipophilic environmental contaminants upon the evaluation of selected foodstuff contamination. Collected food samples (n = 58) were selected to be representative of the lipid fraction of the whole diet of the Beiruti population. The samples were analysed using pressurized liquid extraction, silica multilayer column followed by gel permeation chromatography for purification and GC-EI-HRMS for separation and detection. Detection frequency of at least one compound among Dechlorane Plus (syn-DP and anti-DP), Dechlorane 602, 603 and Chlordene Plus) was 91%. The mean concentrations of ∑ 6 DRCs, by food group, ranged from 4.7 to 29.5 pg g -1 wet weight in lowerbound (LB) and from 6.7 to 76.9 pg g -1 wet weight in upperbound (UB). Based on food habits, the dietary intake of Beiruti adults was estimated to be between 3.71 (LB) and 5.61 (UB) ng day -1 . Dechlorane Plus and Dechlorane 602 were the dominant compounds, contributing to 70 and 24% of the total intake (LB value), respectively. With the phasing out of traditional polybrominated diphenyl ethers (PBDEs), significant volumes of alternative brominated flame retardants (aBFRs) are being used and released into the environment compartment, especially in coastal regions. The levels and distribution of PBDEs, aBFRs, and dechlorane plus (DPs) were investigated in the surface sediments of the Yellow Sea (YS) and East China Sea (ECS) to examine the distribution and sources of these hydrophobic contaminants. The level and distribution of pollutants in the sediments of YS and ECS show obvious regional differences. As a major replacement for decabromodiphenyl ether (BDE 209), decabromodiphenyl ethane (DBDPE) was the dominant compound observed in the surface sediments, with a concentration one order of magnitude higher than that of BDE 209. High concentrations were found in the depositional zones of the YS, indicating that these contaminants may originate from land-based pollution sources (likely from the Laizhou Bay manufacturing base) near the Bohai Sea. The pollutants can be carried by the coastal current together with the sediment from the Yellow River, transported through the Bohai Strait and deposited in the mud zone of Northern and Southern YS. Low levels of halogenated flame retardants (HFRs) were found in the estuary of the Yangtze River and ECS, indicating that Yangtze River contributes less HFRs to the region. Numerous studies have reported bioaccumulation of halogenated flame retardants (HFRs) and polychlorinated biphenyls (PCBs) in wildlife from electronic waste (e-waste) recycling sites. However, the concentrations and hazards of HFRs and PCBs in wildlife from non-e-waste sites which were not involved in any known e-waste recycling activities in the e-waste-impacted region are still unclear. Polybrominated diphenyl ethers (PBDEs), alternative HFRs (AHFRs; including dechlorane plus, decabromodiphenyl ethane, and 1,2-bis(2,4,6-tribromophenoxy) ethane), and PCBs were quantified in common kingfishers (Alcedo atthis) from a region affected by e-waste recycling in South China, and potential adverse effects were evaluated. Concentrations of ∑PBDEs and ∑PCBs in kingfishers ranged from 2.1 × 10 3 -1.3 × 10 5  ng/g lipid mass (lm) and 2.1 × 10 3 -1.5 × 10 6  ng/g lm, respectively. At e-waste recycling sites, these concentrations were 100- to 1000-fold greater than those in kingfishers from non-e-waste areas, where concentrations of ∑PBDEs and ∑PCBs were 16-1.2 × 10 3 and 39-3.0 × 10 3  ng/g lm, respectively. Concentrations of ∑AHFRs in kingfishers from e-waste sites and non-e-waste sites ranged from 8.5 to 3.6 × 10 2 and 0.8-2.9 × 10 2  ng/g lm, respectively. The greatest concentrations of PCBs in kingfishers were measured from the e-waste sites. Additionally, kingfishers from four non-e-waste sites in the vicinity of e-waste sites had greater PCB concentrations compared to the other six non-e-waste sites. Concentrations of AHFRs were negatively and significantly correlated with distance from an e-waste site, which indicated that AHFRs from non-e-waste sites might be influenced by point sources. Further, a significant (r 2  = 0.53, p = 0.02) positive correlation between human population density and concentrations of ∑PBDEs in kingfishers from non-e-waste sites was observed. Concentrations of either PBDEs or PCBs from e-waste sites might pose severe, adverse reproductive effects to kingfishers, while the potential for adverse effects of PBDEs and PCBs to kingfishers from most non-e-waste sites seemed minimal., (Copyright © 2019 The Authors. Dechlorane plus (DP) is found widely in environmental media. Determining DP concentrations in dust and hair and identifying relationships between the concentrations in dust and hair could improve our understanding of the effects of DP in humans. DP concentrations in 48 hair samples from male and female students and in 30 indoor dust samples from dormitories and classrooms used by the students were determined. The objective was to determine the difference of DP concentration between indoor dust and hair, and to explore the effect of DP in dust on DP in hair of male and female students. The mean DP concentration was significantly higher in male dormitory dust than female dormitory dust, and the estimated DP dose through exposure to dust was also significantly higher for males than females. However, the mean DP concentration was significantly higher for hair from females than from males. The median DP f anti was significantly lower for hair than dust, indicating DP may be stereoselectively metabolized by humans. The median DP f anti was significantly higher for hair from females than from males, indicating DP may be metabolized differently by males and females. Human serum albumin preferred combination with anti-DP, rather than syn-DP. DP in indoor dust was not a major direct source of the DP in the hair, meaning DP in hair mainly came from within the body. Traditional Chinese Medicines (TCMs) have been widely used in orient countries for thousands of years, while their inconsistent quality and therapy issues have become increasingly serious as a result of the absence of effective methods for quality control. Therefore, it is necessary to develop a novel and specific evaluation system for TCMs' quality involved with not only composition but also bioactivity. In this study, we used Schisandra chinensis (Turcz.) Baill as an example and developed a novel integrated approach involved with various chemical analysis and data processing methods to explore its quality marker (Q-marker) underlying its anti-depressive effects. First, six bioactive lignans were identified and semi-quantified in rat brain samples via high resolution mass spectrometry. Then, the bioinformation analysis showed that all the six bioactive components could modulate various diseases relative to noradrenergic, dopaminergic and serotonergic pathways. Thus, the monoaminergic metabolites contained in these three pathways were selected to screen potential biomarkers of depression treated by S. chinensis based on target metabolomics using a rapid HPLC-MS/MS method. Finally, the correlation analysis between the six components and potential biomarkers was employed to uncover the Q-markers of S. chinensis. It is suggested that schisandrol A, schisandrin A, schisandrin C and gomisin N could be determined as Q-markers for S. chinensis. Thus, the integrated approach describing here for discovering Q-markers was expected to offer an alternative quality assessment strategy of herbal medicines for the first time., (Copyright © 2019. A reverse phase (RP)-HPLC method for separation and determination of Schisandrin A and Schisandrin B was presented, using a C18 Bondclone column, with methanol-water (v/v = 68 : 32) as mobile phase at a flow-rate of 1.00 mL·min -1 , and UV detection at 220 nm. The tested parameters included mobile phase composition and UV detection wavelength. Good linearities were observed within concentration ranges of Schisandrin A 0.008-4.8 mg·L -1 (r = 0.9996), and Schisandrin B 0.005-3.1 mg·L -1 (r = 0.9994), respectively. The limit of detection (LOD) (S/N = 3) were 0.005 mg·L -1 Schisandrin A and 0.002 mg·L -1 Schisandrin B, respectively. The method was applied to determine the 2 compounds in a traditional Chinese medicine preparation for treatment of hepatic diseases, Huganpian tablet. To eliminate matrix effect, Oasis hydrophilic lipophilic balance (HLB) solid-phase extraction (SPE) was used to purify the ultra-sonicately extracted solution of the drug sample. Combined with the HLB SPE purification procedure, the HPLC method gave satisfactory results for quantitation of Schisandrin A and Schisandrin B in 3 types of Huganpian tablet samples, with spiking recoveries ca. 98% (relative standard deviation (R.S.D.) ≤ 3.5%) (n = 5).

Polybrominated diphenyl ethers (PBDEs), decabromodiphenyl ethane (DBDPE), 1,2-bis(2,4,6-tribromophenoxy)ethane (BTBPE), tetrabromobisphenol A (TBBPA), hexabromocyclododecane (HBCDD) and dechlorane plus (DP) were measured in sediments collected from three mangrove wetlands of the Pearl River Estuary (PRE) in South China. This study aims to investigate the distribution of these halogenated flame retardants (HFRs) and the correlations between HFRs and microbial community structure in mangrove sediments. Concentrations of PBDEs, DBDPE, BTBPE, TBBPA, HBCDD and DP in mangrove sediments ranged from 6.97 to 216.1, 3.70-26.0, 0.02-0.73, 0.02-37.5, 0.44-127.5 and 0.07-2.23 ng/g dry weight, respectively. Higher levels of PBDEs, BTBPE, HBCDD and DP were observed in sediments from Futian mangrove wetland of Shenzhen, the only nature reserve located in the downtown of China. The highest concentration of TBBPA found in mangrove sediments from Guangzhou was proximate to a ferry terminal and a dockyard where TBBPA is widely used in the coatings. PBDEs were the predominant HFRs in mangrove sediments, with an average contribution of 63.0%. Mangrove sediments from Guangzhou and Zhuhai showed an enrichment of (-)-α-HBCDD, (-)-β-HBCDD and (-)-γ-HBCDD. Concentrations of HFRs in mangrove sediments from Guangzhou increased significantly from 2012 to 2015, which was probably due to the establishment and rapid development of Nansha Free Trade Zone of Guangzhou. Although isomer-specific bioaccumulation of dechlorane plus (DP) has been addressed in many studies, it remains unclear which factors determine this process and whether biotransformation of DP occurs in organisms. Comparative experiments were conducted in both in vivo and in ovo incubation using hens and eggs to identify the dominant factors determining the bioaccumulation of DP. Hens and fertilized eggs were exposed to DP isomers (syn- and anti-DP) by feeding and spiking, respectively, to investigate absorption, elimination, and metabolism. No significant differences were found between absorption efficiencies of DP isomers in the adult hens. Following first-order kinetics, anti-DP exhibited a slightly longer half-life than syn-DP as well as an elevated anti-DP fraction in laid eggs, thereby suggesting selective enrichment of anti-DP in adult hens. However, chicken embryos metabolized approximately 12% and 28% of the absorbed syn- and anti-DP, respectively, thereby verifying that anti-DP was preferably metabolized. This result indicated that stereo-selective excretion of syn-DP, rather than preferred metabolism of anti-DP, played a more prominent role in isomer-specific bioaccumulation of DP in chickens. Arboreous fruit of Schisandra chinensis Baillon, Schisandra Fruit (SF), is a crude drug used in Japanese traditional Kampo medicine. The marker compounds of SF for quality control are lignans, such as schizandrin (Sz) and gomisin A (GmA). Kampo formulation containing SF is usually prepared as decoctions in the dosage form of whole crude drug (W), as its size is small enough to measure using a spoon. However, in some traditional books, it has been described that SF must be used in the dosage form of crushed or cut pieces (Cr). In this study, we evaluated the transferring ratio of lignans from SF to the decoction, and the stability and taste of the decoctions of shoseiryuto (SST) and ninjin'yoeito (NYT) using each dosage form, i.e., Cr and W, of SF. The transferring ratio of Sz and GmA was significantly higher in the decoction prepared with the Cr form than that prepared using the W form in both SST and NYT. The concentration of Sz and GmA in the decoctions was stable when maintained at 4 °C for 35 days. The taste of SST decoction prepared using the Cr form was more acidic, harsher, and bitterer than SST decoction prepared using the W form, and the taste of NYT decoction prepared using the Cr form was harsher than NYT decoction prepared using the W form. In conclusion, when SF is used in Kampo prescription, crushing the fruits and seeds can increase its effectiveness.

Under the Stockholm Convention, signatory countries are obliged to direct source inventories, find current sources, and provide ecological monitoring evidence to guarantee that the encompassing levels of persistent organic pollutants (POPs) are declining. However, such monitoring of different types of POPs are to a great degree constrained in most developing countries including Nepal and are primarily confined to suspected source area/ densely populated regions. In this study, 9 polybrominated diphenyl ethers (PBDEs), 2 dechlorane plus (DPs), 6 novel brominated flame retardants (NBFRs) and 8 organophosphate ester flame retardants (OPFRs) were investigated in indoor dust from a rural area (Kopawa) in Nepal in order to evaluate their occurrence/level, profile, spatial distribution and their sources. Additionally, health risk exposure was estimated to anticipate the possible health risk to the local population. The results showed that OPFRs was the most abundant FR measured in the dust. The concentration of ∑ 8 OPFRs was about 2, 3 and 4 orders of magnitude higher than the ∑ 6 NBFRs, ∑ 9 PBDEs, and ∑ 2 DPs, respectively. Tris (methylphenyl) phosphate (TMPP) and Tris (2-ethylhexyl) phosphate (TEHP) were the most abundant OPFRs analyzed in the dust; while decabromodiphenyl ethane (DBDPE) exceeded among NBFRs. Likewise, 2,2',3,3',4,4',5,5',6,6'-decabromodiphenylether (BDE-209) was the most identified chemical among PBDEs. The total organic carbon (TOC) content in dust was significantly and positively connected with octa-BDEs (Rho = 0.615, p < 0.01), BTBPE (Rho = 0.733, p < 0.01), TPHP (Rho = 0.621, p < 0.01), TEHP (Rho = 0.560, p < 0.01) and TMPPs (Rho = 0.550, p < 0.01), while black carbon (BC) was either weakly related or not related, suggesting little or no impact of BC in the distribution of FRs. Principal component analysis indicated the contribution from commercial penta-, octa- and deca-BDEs formulation, the adhesive substance, food packaging and paints, and degradation of BDE-209 as the essential sources of FRs. Health risk exposure estimates showed that dermal absorption via dust as the primary route of FRs intake. The estimated daily exposure of PBDEs, NBFRs and OPFRs were 2-10 orders of magnitude lower than their corresponding reference dose (RfD), suggesting insignificant risk.
