Abstract Background Fusarium oxysporum f. sp. melonis Snyd. & Hans. (FOM) causes Fusarium wilt, the most important infectious disease of melon (Cucumis melo L.). The four known races of this pathogen can be distinguished only by infection on appropriate cultivars. No molecular tools are available that can discriminate among the races, and the molecular basis of compatibility and disease progression are poorly understood. Resistance to races 1 and 2 is controlled by a single dominant gene, whereas only partial polygenic resistance to race 1,2 has been described. We carried out a large-scale cDNA-AFLP analysis to identify host genes potentially related to resistance and susceptibility as well as fungal genes associated with the infection process. At the same time, a systematic reisolation procedure on infected stems allowed us to monitor fungal colonization in compatible and incompatible host-pathogen combinations. Results Melon plants (cv. Charentais Fom-2), which are susceptible to race 1,2 and resistant to race 1, were artificially infected with a race 1 strain of FOM or one of two race 1,2 w strains. Host colonization of stems was assessed at 1, 2, 4, 8, 14, 16, 18 and 21 days post inoculation (dpi), and the fungus was reisolated from infected plants. Markedly different colonization patterns were observed in compatible and incompatible host-pathogen combinations. Five time points from the symptomless early stage (2 dpi) to obvious wilting symptoms (21 dpi) were considered for cDNA-AFLP analysis. After successful sequencing of 627 transcript-derived fragments (TDFs) differentially expressed in infected plants, homology searching retrieved 305 melon transcripts, 195 FOM transcripts expressed in planta and 127 orphan TDFs. RNA samples from FOM colonies of the three strains grown in vitro were also included in the analysis to facilitate the detection of in planta-specific transcripts and to identify TDFs differentially expressed among races/strains. Conclusion Our data suggest that resistance against FOM in melon involves only limited transcriptional changes, and that wilting symptoms could derive, at least partially, from an active plant response. We discuss the pathogen-derived transcripts expressed in planta during the infection process and potentially related to virulence functions, as well as transcripts that are differentially expressed between the two FOM races grown in vitro. These transcripts provide candidate sequences that can be further tested for their ability to distinguish between races. Abstract Background Downy mildew is a destructive grapevine disease caused by Plasmopara viticola (Berk. and Curt.) Berl. and de Toni, which can only be controlled by intensive fungicide treatments. Natural sources of resistance from wild grapevine (Vitis) species are used in conventional breeding approaches, but the signals and effectors involved in resistance in this important crop species are not well understood. Results Early transcriptional changes associated with P. viticola infection in susceptible V. vinifera and resistant V. riparia plants were analyzed using the Combimatrix microarray platform. Transcript levels were measured 12 and 24 h post-inoculation, reflecting the time points immediately preceding the onset of resistance in V. riparia, as determined by microscopic analysis. Our data indicate that resistance in V. riparia is induced after infection, and is not based on differences in basal gene expression between the two species. The strong and rapid transcriptional reprogramming involves the induction of pathogenesis-related proteins and enzymes required for the synthesis of phenylpropanoid-derived compounds, many of which are also induced, albeit to a lesser extent, in V. vinifera. More interestingly, resistance in V. riparia also involves the specific modulation of numerous transcripts encoding components of signal transduction cascades, hypersensitive reaction markers and genes involved in jasmonate biosynthesis. The limited transcriptional modulation in V. vinifera represents a weak attempted defense response rather than the activation of compatibility-specific pathways. Conclusions Several candidate resistance genes were identified that could be exploited in future biotechnological approaches to increase disease resistance in susceptible grapevine species. Measurements of jasmonic acid and methyl jasmonate in infected leaves suggest that this hormone may also be involved in V. riparia resistance to P. viticola.

Abstract Background The oomycete Plasmopara viticola (Berk. and Curt.) Berl. and de Toni causes downy mildew in grapevine (Vitis vinifera L.). This pathogen is strictly biotrophic, thus completely dependent on living host cells for its survival. The molecular basis of compatibility and disease development in this system is poorly understood. We have carried out a large-scale cDNA-AFLP analysis to identify grapevine and P. viticola genes associated with the infection process. Results We carried out cDNA-AFLP analysis on artificially infected leaves of the susceptible cultivar Riesling at the oil spot stage, on water-treated leaves and on a sample of pure sporangia as controls. Selective amplifications with 128 primer combinations allowed the visualization of about 7000 transcript-derived fragments (TDFs) in infected leaves, 1196 of which (17%) were differentially expressed. We sequenced 984 fragments, 804 of which were identified as grapevine transcripts after homology searching, while 96 were homologous to sequences in Phytophthora spp. databases and were attributed to P. viticola. There were 82 orphan TDFs. Many grapevine genes spanning almost all functional categories were downregulated during infection, especially genes involved in photosynthesis. Grapevine genes homologous to known resistance genes also tended to be repressed, as were several resistance gene analogs and carbonic anhydrase (recently implicated in pathogen resistance). In contrast, genes encoding cytoskeletal components, enzymes of the phenylpropanoid and beta-oxidation pathways, and pathogenesis related proteins were primarily upregulated during infection. The majority of P. viticola transcripts expressed in planta showed homology to genes of unknown function or to genomic Phytophthora sequences, but genes related to metabolism, energy production, transport and signal transduction were also identified. Conclusion This study provides the first global catalogue of grapevine and P. viticola genes expressed during infection, together with their functional annotations. This will help to elucidate the molecular basis of the infection process and identify genes and chemicals that could help to inhibit the pathogen.

IntroductionThis is a prospective, single-center trial in pediatric patients with sarcoma aiming to evaluate [18F]FDG PET/CT as a tool for early response assessment to neoadjuvant chemotherapy (neo-CTX).MethodsBone or soft tissue sarcoma patients with (1) baseline [18F]FDG PET/CT within 4 weeks prior to the start of neo-CTX (PET1), (2) early interim [18F]FDG PET/CT (6 weeks after the start of neo-CTX (PET2), (3) evaluation of neo-CTX response by histology or MRI, and (4) definitive therapy after neo-CTX (surgery or radiation) were included. Semiquantitative PET parameters (SUVmax, SUVmean, SUVpeak, MTV and TLG) and their changes from PET1 to PET2 (ΔPET) were obtained. The primary endpoint was to evaluate the predictive value of PET1, PET2 and ΔPET parameters for overall survival (OS) and time to progression (TTP). The secondary outcome was to evaluate if [18F]FDG PET/CT can predict the response to neo-CTX assessed by histopathology or MRI. Primary and secondary outcomes were also evaluated in a subpopulation of patients with bone involvement only.ResultsThirty-four consecutive patients were enrolled (10 females; 24 males; median age 15.1 years). 17/34 patients (50%) had osteosarcoma, 13/34 (38%) Ewing's sarcoma, 2/34 (6%) synovial sarcoma and 2/34 (6%) embryonal liver sarcoma. Median follow-up was 39 months (range 16-84). Eight of 34 patients (24%) died, 9/34 (27%) were alive with disease, and 17/34 (50%) had no evidence of residual/recurrent disease. Abstract Background Homology-dependent selective degradation of RNA, or post-transcriptional gene silencing (PTGS), is involved in several biological phenomena, including adaptative defense mechanisms against plant viruses. Small interfering RNAs mediate the selective degradation of target RNA by guiding a multicomponent RNAse. Expression of self-complementary hairpin RNAs within two complementary regions separated by an intron elicits PTGS with high efficiency. Plum pox virus (PPV) is the etiological agent of sharka disease in Drupaceae, although it can also be transmitted to herbaceous species (e.g. Nicotiana benthamiana). Once inside the plant, PPV is transmitted via plasmodesmata from cell to cell, and at longer distances, via phloem. The rolC promoter drives expression in phloem cells. RolC expression is absent in both epidermal and mesophyll cells. The aim of the present study was to confer systemic disease resistance without preventing local viral infection. Results In the ihprolC-PP197 gene (intron hair pin rolC PPV 197), a 197 bp sequence homologous to the PPV RNA genome (from base 134 to 330) was placed as two inverted repeats separated by the DNA sequence of the rolA intron. This hairpin construct is under the control of the rolC promoter.N. benthamiana plants transgenic for the ihprolC-PP197 gene contain siRNAs homologous to the 197 bp sequence. The transgenic progeny of ihprolC-PP197 plants are resistant to PPV systemic infection. Local infection is unaffected. Most (80%) transgenic plants are virus free and symptomless. Some plants (20%) contain virus in uninoculated apical leaves; however they show only mild symptoms of leaf mottling. PPV systemic resistance cosegregates with the ihprolC-PP197 transgene and was observed in progeny plants of all independent transgenic lines analyzed. SiRNAs of 23–25 nt homologous to the PPV sequence used in the ihprolC-PP197 construct were detected in transgenic plants before and after inoculation. Transitivity of siRNAs was observed in transgenic plants 6 weeks after viral inoculation. Conclusions The ihprolC-PP197 transgene confers systemic resistance to PPV disease in N. benthamiana. Local infection is unaffected. This transgene and/or similar constructs could be used to confer PPV resistance to fruit trees where systemic disease causes economic damage.

Background: Proper positioning of a total hip arthroplasty (THA) plays a crucial role in the success and long-term survivorship of the implant. Cup positioning within the Lewinnek Safe Zone (LSZ) does not, however, avoid implant dislocation. Thus, the concept of a functional cup position has been introduced. The purpose of this study was to assess the discrepancy between LSZ and the acetabular cup position suggested by the patient's specific functional planning. The hypothesis was that a mismatch does exist. Methods: One hundred consecutive patients with primary hip osteoarthritis undergoing primary THA with a personalized functional preoperative planning and patient-specific cup implantation system were enrolled. Anatomical and spino-pelvic functional parameters were recorded and, for each patient, a "safe cup orientation" was suggested. The suggested functional safe zone was compared to the LSZ. Results: The mean suggested inclination was 39° ± 3° (range 32°–45°). The mean suggested anteversion was 21° ± 3° (range 12°–28°). The patient's functional acetabular inclination (AI) corresponded to the LSZ in one of the 100 patients, whereas the acetabular anteversion (AV) was outside the LSZ in 8 of the 100 patients. The mean pelvic tilt while standing and sitting were 0.5° ± 7° (range 21°–45°) and −6° ± 16.7° (range −63°–33°), respectively. The mean pelvic incidence was 52° ± 9.7° (range 33°–83°). Conclusion: When a functional patient's specific preoperative planning is performed, the LZS does not correspond to the patient's functional safe zone in about 8% of patients. The concept of a universal safe zone should be revisited, and a functional personalized safe zone may have to be more widely considered. [ABSTRACT FROM AUTHOR]

Ocean wind plays a fundamental role in the Earth's climate system by transporting heat, moisture, and gases globally while mediating their exchange with the ocean. How, and at what scales these processes occur is an active area of research, and modeling studies suggest that the strongest vertical transport and air‐sea exchanges occur at small scales. This paper introduces results from the S‐MODE campaign, during which a Doppler Scatterometer mapped 2D ocean vector wind on scales between 200 m and 100 km. While in an average sense, DopplerScatt wind data match classical k−5/3 ${k}^{-5/3}$ spectra, atmospheric rolls, convection, and underlying ocean currents all drive unique wind spectra that are inconsistent with the average. Frequent observations of strong atmospheric divergence imply small‐scale vertical transport reaching > ${ >} $10 km/day. These results are not consistent with Kolmogorov and two‐dimensional turbulence theories due to anisotropy and divergence. Plain Language Summary: Using a recently developed measurement technique, we mapped two‐dimensional ocean vector winds at scales about 100 times smaller than in the past, simultaneously with ocean currents. Our observations show that the wind is highly variable spatially, and that interactions between the ocean and atmosphere are ubiquitous. We find that small scale motions drive strong spreading and turning in the atmosphere that imply strong vertical transport, which is important for the transfer of heat and gases between the ocean and atmosphere. These findings do not support some of the most commonly applied theories of atmospheric dynamics. Understanding these dynamics, and properly incorporating them into climate and forecast models, will improve the accuracy of projections. The noncarious cervical lesions (NCCLs) recognize an etiological framework of onset very different from the carious processes with etiology whose bacteria aggregated in a biofilm play a predominant role, leading in this way to the loss of the mineralized structure of the tooth. The pathological picture of the NCCLs, which manifests itself with a clinical picture of dental wear, differs from caries because it mainly recognizes a series of pathological processes, such as erosion, through the action of generally acidic chemical agents and abrasion, which is basically expressed through repeated mechanical trauma characteristic of tooth brushing. However, in the literature, there is no unanimous agreement in identifying only these two mechanisms, but there are some who propose a more marked role of anomalous occlusal loads, which would be unloaded on some teeth which, in addition to both erosive and abrasive mechanisms, would give rise to abfraction. Therefore, the aim of this review was to collect literature etio-pathological information and discuss the mechanisms underlying NCCLs.

This study was conducted to evaluate the potential of low-cost hyperspectral sensors for the early detection of Flavescence dorée (FD) from asymptomatic samples prior to symptom development. In total, 180 leaf spectra from 60 randomly selected plants (three leaves per plant) were collected by using two portable mini-spectrometers (Hamamatsu: 340–850 nm and NIRScan: 900–1700 nm) at five vegetative growth stages in a vineyard with grape variety Garganega. High differences in the Hamamatsu spectra of the two groups were found in the VIS-NIR (visible–near infrared) spectral region while very small differences were observed in the NIRScan spectra. We analyzed the spectral data of two sensors by using all bands, features reduced by an ensemble method, and by genetic algorithms (GA) to discriminate the asymptomatic healthy (FD negative) and diseased (FD positive) leaves using five different classifiers. Overall, high classification accuracies were found in case of the Hamamatsu sensor compared to the NIRScan sensor. The feature selection techniques performed better compared to all bands, and the highest classification accuracy of 96% was achieved when GA features of the Hamamatsu sensor were used with the logistic regression (LR) classifier on test samples. A slightly low accuracy of 85% was achieved when the features (selected by the ensemble method) of the Hamamatsu sensor were used with the support vector machine (SVM) classifier by using leave-one-out (LOO) cross-validation on the whole dataset. Results demonstrated that employing a feature selection technique can provide a valid tool for determining the optimal bands that can be used to identify FD disease in the vineyard. However, further validation studies are required, as this study was conducted using a small dataset and from the single grapevine variety.

The past two decades have seen increasing interest in the quality of government as an explanatory factor for the performance of European Union (EU) Cohesion Policy. An under-researched strand of these studies is administrative capacity in Cohesion Policy. Against this background, this mini-special issue provides new perspectives on the theme of administrative capacity, providing new insights into the operation and effectiveness of administrative capacity-building measures implemented in the EU countries, particularly through the support provided by the Structural Funds. The study examines the implementation of urban development strategies in Scotland, UK, and the Veneto Region of Italy and fills an important gap in the knowledge about the capacity-building potential of such strategies. Following a principal-agent approach, the investigation shows that narrow delegation models incentivize compliance, are less conducive to capacity-building and reinforce an administrative capacity paradox. By contributing an original framework for the study of delegation models, a fine-grained understanding of administrative capacity that acknowledges the importance of agency for the success of capacity-building initiatives and policy recommendations for the period 2021–27, the research will be of interest to scholars and practitioners alike. Iterative reconstructions (IR) might alter radiomic features extraction. We aim to evaluate the influence of Adaptive Statistical Iterative Reconstruction-V (ASIR-V) on CT radiomic features. Patients who underwent unenhanced abdominal CT (Revolution Evo, GE Healthcare, USA) were retrospectively enrolled. Raw data of filtered-back projection (FBP) were reconstructed with 10 levels of ASIR-V (10–100%). CT texture analysis (CTTA) of liver, kidney, spleen and paravertebral muscle for all datasets was performed. Six radiomic features (mean intensity, standard deviation (SD), entropy, mean of positive pixel (MPP), skewness, kurtosis) were extracted and compared between FBP and all ASIR-V levels, with and without altering the spatial scale filter (SSF). CTTA of all organs revealed significant differences between FBP and all ASIR-V reconstructions for mean intensity, SD, entropy and MPP (all p < 0.0001), while no significant differences were observed for skewness and kurtosis between FBP and all ASIR-V reconstructions (all p > 0.05). A per-filter analysis was also performed comparing FBP with all ASIR-V reconstructions for all six SSF separately (SSF0-SSF6). Results showed significant differences between FBP and all ASIR-V reconstruction levels for mean intensity, SD, and MPP (all filters p < 0.0315). Skewness and kurtosis showed no differences for all comparisons performed (all p > 0.05). The application of incremental ASIR-V levels affects CTTA across various filters. Skewness and kurtosis are not affected by IR and may be reliable quantitative parameters for radiomic analysis.
