Abstract

A new measure $c(e)$ of the centrality of an edge $e$ in an undirected graph $G$ is introduced. It is based on the variation of the Kemeny constant of the graph after removing the edge $e$. The new measure is designed in such a way that the Braess paradox is avoided. A numerical method for computing $c(e)$ is introduced and a regularization technique is designed in order to deal with cut-edges and disconnected graphs. Numerical experiments performed on synthetic tests and on real road networks show that this measure is particularly effective in revealing bottleneck roads whose removal would greatly reduce the connectivity of the network.

Abstract

Consider a finite set of identical computational entities that can move freely in the Euclidean plane operating in Look-Compute-Move cycles. Let p(t) denote the location of entity p at time t; entity p can see entity q at time t if at that time no other entity lies in the line segment p(t)q(t). We consider the basic problem called Mutual Visibility: starting from arbitrary distinct locations, within finite time the entities must reach, without collisions, a configuration where they all see each other. This problem must be solved by each entity autonomously executing the same algorithm. We study this problem in the "luminous robots" model; in this generalization of the standard model of oblivious robots, each entity, called "robot", has an externally visible persistent light which can assume colors from a fixed set. The case where the number of colors is c=1 corresponds to the classical model without lights. In this paper we investigate under what conditions luminous robots can solve Mutual Visibility without collisions and at what cost (i.e., with how many colors). We establish a spectrum of results, depending on the power of the adversary, on the number c of colors, and on the a-priori knowledge the robots have about the system. Among such results, we prove that Mutual Visibility can always be solved without collisions in SSynch with c=2 colors and in ASynch with c=3 colors. If an adversary can interrupt and stop a robot moving to its computed destination, Mutual Visibility is still always solvable without collisions in SSynch with c=3 colors, and, if the robots agree on the direction of one axis, also in ASynch. All the results are obtained constructively by means of novel protocols. As a byproduct of our solutions, we provide the first obstructed-visibility solutions to two classical problems for oblivious robots: Collision-less Convergence to a point and Circle Formation., Comment: 60 pages, 15 figures

7. Factoring block Fiedler Companion Matrices

Abstract

When Fiedler published his “A note on Companion matrices” in 2003 in Linear Algebra and its Applications, he could not have foreseen the significance of this elegant factorization of a companion matrix into essentially two-by-two Gaus- sian transformations, which we will name (scalar) elementary Fiedler factors. Since then, researchers extended these results and studied the various resulting lineariza- tions, the stability of Fiedler companion matrices, factorizations of block companion matrices, Fiedler pencils, and even looked at extensions to non-monomial bases. In this chapter, we introduce a new way to factor block Fiedler companion matrices into the product of scalar elementary Fiedler factors. We use this theory to prove that, e.g., a block (Fiedler) companion matrix can always be written as the product of several scalar (Fiedler) companion matrices. We demonstrate that this factoriza- tion in terms of elementary Fiedler factors can be used to construct new lineariza- tions. Some linearizations have notable properties, such as low band-width, or allow for factoring the coefficient matrices into unitary-plus-low-rank matrices. Moreover, we will provide bounds on the low-rank parts of the resulting unitary-plus-low-rank decomposition. 8. Iterative methods for the delay Lyapunov equation with T-Sylvester preconditioning

Abstract

The delay Lyapunov equation is an important matrix boundary-value problem which arises as an analogue of the Lyapunov equation in the study of time-delay systems x˙(t)=A0x(t)+A1x(t−τ)+B0u(t). We propose a new algorithm for the solution of the delay Lyapunov equation. Our method is based on the fact that the delay Lyapunov equation can be expressed as a linear system of equations, whose unknown is the value U(τ/2)∈Rn×n, i.e., the delay Lyapunov matrix at time τ/2. This linear matrix equation with n2 unknowns is solved by adapting a preconditioned iterative method such as GMRES. The action of the n2×n2 matrix associated to this linear system can be computed by solving a coupled matrix initial-value problem. A preconditioner for the iterative method is proposed based on solving a T-Sylvester equation MX+XTN=C, for which there are methods available in the literature. We prove that the preconditioner is effective under certain assumptions. The efficiency of the approach is illustrated by applying it to a time-delay system stemming from the discretization of a partial differential equation with delay. Abstract

To verify whether it is possible, in people with diabetes mellitus (DM) considered at very high cardiovascular (CV) risk, stratify this risk better and identify significant modifiable risk factor (including lifestyle habits) to help patients and clinicians improve CV prevention. People with DM and microvascular diseases or one or more CV risk factors (hypertension, hyperlipidemia, smoking, poor dietary habits, overweight, physical inactivity) included in the Risk and Prevention study were selected. We considered the combined endpoint of non-fatal acute myocardial infarction and stroke and CV death. A multivariate Cox proportional analysis was carried out to identify relevant predictors. We also used the RECPAM method to identify subgroups of patients at higher risk. In our study, the rate of major CV events was lower than expected (5 % in 5 years). Predictors of CV events were age, male, sex, heart failure, previous atherosclerotic disease, atrial fibrillation, insulin treatment, high HbA1c, heart rate and other CV diseases while being physically active was protective. RECPAM analysis indicated that history of atherosclerotic diseases and a low BMI defined worse prognosis (HR 4.51 95 % CI 3.04–6.69). Among subjects with no previous atherosclerotic disease, men with HbA1c more than 8 % were at higher CV risk (HR 2.77; 95 % CI 1.86–4.14) with respect to women. In this population, the rate of major CV events was lower than expected. This prediction model could help clinicians identify people with DM at higher CV risk and support them in achieving goals of physical activity and HbA1c.

Abstract

We confirmed the occurrence of IgG antibodies reacting with ox cardiac conducting tissue in the serum of some human subjects. These antibodies failed to react with all ox cardiac conducting tissue cells; they reacted only with the cells defined as Purkinje cells. Having checked 352 sera, we found that the prevalence of antibodies to Purkinje cells was 11% in normal subjects (no correlation with sex and age), 14% in systemic lupus erythematosus, 21% in rheumatoid arthritis, 18% in progressive systemic sclerosis, and 23% in Sjogren syndrome. In 50 patients with permanent pacemakers for chronic non-postinfarction atrioventricular (AV) block the prevalence was 30% (P = 0.008). In a selected set of 29 patients with clinically idiopathic AV block located at or below the level of the His bundle the prevalence was 34.5% (P = 0.006). The possible role of anti-Purkinje cell antibodies in autoimmune damage of cardiac conduction tissue is discussed.

Abstract

The patient admission scheduling (PAS) problem is a class of scheduling problems that must be handled by the managers of the hospital admission systems. The problem arises when patients arriving at the hospital need to be allocated to beds in an optimal manner, subject to the availability of beds and the needs of patients. The PAS problem in a dynamic context, as analysed in Ceschia and Schaerf [2] and Lusby et al. [6], considers a scenario in which random arrivals and unknown departures of patients are gradually revealed over the planning horizon. The problem was formulated as an integer programming model, and various procedures for computing the optimal solution were proposed. Ceschia and Schaerf [2] developed a metaheuristic algorithm based on simulated annealing and neighborhood search. Lusby et al. [6] developed an adaptive large neighbourhood search procedure to solve the problem. Although the arrivals and departures of patients are in general random, the models in [2, 6] assumed deterministic inputs such as a fixed length of stay for each patient, and a fixed number of arrivals at the start of each day. Here, we build on the analysis in Lusby et al. [6], and develop a model for the PAS problem in a dynamic context, which captures the random dynamics of the flow of the patients. Our aim here is to develop an improved mathematical model to solve the PAS problem in a dynamic environment with random arrivals and departures. Centrality measures of Integration and Choice have performed a crucial role for Space Syntax in depicting complex relations among form, function, and movement within cities. However, while still relevant, those measures are unable to address certain innate network properties regarding the relative importance of certain road elements, essential for urban analyses focused on road-network resilience. The overreliance on Integration and Choice metrics to explain urban phenomena left several configurational patterns derived from connectivity rather unaddressed by Space Syntax and currently constitutes the methodology’s main limitation. With those points in consideration, this paper proposes an initial overview regarding the adaptation of Markov-based centrality measures to the Space Syntax framework and graph representation. These measures, often computable only in primal graphs, are adapted to the Road-Centre Line representation, in a first approach that aims to further integrate them into the Angular Analysis’ framework. We use the measures of Normalized PageRank Centrality and Normalized Kemeny-based centrality that estimate the connective relative importance of individual road-elements within the system. These measures are based on the notions of strong-ties and weak-ties, both well-known concepts in social networks; Weak-ties are important to establish bridges among interconnected communities of strong-tied individuals. In the urban configuration, weak-ties give information about crucial bridges among spaces characterized by strong-ties, areas that possess a high number of interconnected road elements – common pattern found in urban settlements. Results indicate that adapting Markov-based centralities to Space Syntax is feasible and maintains a configurational and spatial sense, hence it introduces new dimensions to be evaluated in urban-regional analysis.

Abstract

Random walks on graphs are often used to analyse and predict epidemic spreads and to investigate possible control actions to mitigate them. In this study, we first show that models based on random walks with a single stochastic agent (such as Google's popular PageRank) may provide a poor description of certain features of epidemic spread: most notably, spreading times. Then, we discuss another Markov chain based method that does reflect the correct mean infection times for the disease to spread between individuals in a network, and we determine a procedure that allows one to compute them efficiently via a sampling strategy. Finally, we present a novel centrality measure based on infection times, and we compare its node ranking properties with other centrality measures based on random walks. Abstract

Azole resistance in Candida albicans may be due to several mechanisms. It has been demonstrated that C. albicans possesses sequences with a high degree of homology with the human MDR-1 gene coding for P-glycoprotein (P-gp), belonging to the ATP-binding cassette transporter (ABC) superfamily and responsible for the multidrug resistance (MDR) in tumor cells. On this basis, the expression and intracellular localization of human P-gp-like molecule in C. albicans strains showing different sensitivity to fluconazole were investigated by flow cytometry and immunoelectron microscopy. Post-embedding immunolabeling revealed that monoclonal antibody (mAb) MM4.17, which recognizes an external epitope of human P-gp, reacted with both fluconazole-sensitive (3153 and CO 23-1) and fluconazole-resistant (AIDS 68 and CO 23-2, isolated from AIDS patient and in vitro drug-selected, respectively) strains of C. albicans. However, the resistant strains displayed a number of MM4.17-reactive epitopes much higher than the drug-sensitive ones. The C. krusei ATCC 6458 strain, whose resistance is not mediated by the presence of ABC transporters, was not reactive at all with mAb MM4.17. The specificity of the immunolabeling was confirmed by a competitive inhibition assay performed by using phage clone particles capable of mimicking the MM4.17-reactive epitope. The flow cytometric analysis confirmed a higher level of intracytoplasmic P-gp expression in azole-resistant strains of C. albicans. Both cyclosporin A and verapamil, which are well-known MDR inhibitors, strongly reduced the MICs for fluconazole and itraconazole of the tested azole-resistant AIDS 68 strain, while they did not influence the MICs of either the sensitive 3153 strain of C. albicans or the ATCC 6458 strain of C. krusei. Overall, our data suggest the existence of a P-gp-like drug efflux pump in C. albicans that may participate in the mechanisms of azole-resistance of this fungus.

Abstract

We have identified a peptide region on CD18 molecule (the beta subunit of the LFA-1 molecule) involved in syncytia formation of HIV-1-infected lymphocytes. Several phage clones mimicking an epitope of the CD18 cell-surface determinant were isolated from two 9-mer random peptide phage-displayed libraries via their binding to the CD18-specific monoclonal antibody (mAb) MHM23, which in in vitro assay inhibits syncytia formation in HIV-1-infected cells. The peptide sequences displayed on phages that blocked immunolabeling of this mAb on LFA-1-expressing cells were used to identify the epitope recognized by mAb MHM23 by sequence comparison. On the basis of this analysis, two peptides which inhibited syncytia formation in HIV-1-infected cells in vitro were synthesized, thus confirming that they mimic a CD18 domain that is involved in this phenomenon. The results here presented highlight the potential of phage-display technology for the study of biological processes at the basis of virus infection, but also suggest new approaches for the therapy of AIDS.

Abstract

It is not rare that controversial indications about the presence or the expression level of multidrug-resistant (MDR) proteins come out from different laboratories upon examination of identical tumor specimens. Distinct aspects, including the use of weakly discriminating monoclonal antibodies (MAbs) and/or unsuitable techniques and procedures, contribute in generating differences in the MDR phenotype evaluation of cancer cells. In this regard we describe here an innovative immunohistochemical approach for the determination of P-glycoprotein expression in cells and tissues. The method is based on the ability of phage-displayed peptides to mimic antibody epitopes. For this purpose we utilized the phage clone #55, which was affinity-purified from a phage-displayed random-peptide library using the MAb MM4.17 (specific for MDR1-P-glycoprotein) as previously described. This clone has been chosen since it clearly and undoubtedly reacts with its cognate MAb, as was determined by ELISA and dot blot tests. Inhibition of the MAb MM4.17 binding to MDR1-P-glycoprotein-expressing cells could be performed by adding a calibrated concentration of phage clone #55 particles, which mimic MDR1-P-glycoprotein antigen. This methodology can eliminate misleading interpretations concerning the presence and expression level of MDR1-P-glycoprotein and might well contribute in routine clinical determinations of MDR in tumor specimens, thus contributing to our understanding of the basis of the mechanisms of tumor cell resistance to drugs.

Abstract

The MDR1-P-glycoprotein binding sites of three different murine monoclonal antibodies (MM4.17, MM6.15 and MC57), directed towards living, intact human multidrug-resistant cells were investigated in order to study P-glycoprotein topology. By using synthetic peptide scanning, we demonstrated that well-defined regions localized on the predicted first, fourth and sixth extracellular loops are external. On the basis of the structure of MM6.15 epitope, which is distributed on the above three different extracellular loops (and thus is discontinuous), P-glycoprotein molecules result to be differently organized in the lipid bilayer. Moreover, the outcome of the MC57 and MM4.17 epitopes localization experiments, obtained through the use of phage-displayed peptide libraries, represent an additional challenge to the classical 12-transmembrane domain model of P-glycoprotein, since they agree with the novel topography of the molecule (10-transmembrane domain), which was recently proposed on the basis of biochemical and expression studies.

Abstract

To identify an MC57 epitope which is more efficiently expressed on inactivated forms of P-glycoprotein we utilized peptide libraries displayed on filamentous phage. Using this technology, we selected specific phage clones blocking the binding of the murine monoclonal (MAb) MC57 with live human multi-drug-resistant (MDR) cells, and sequenced and analyzed their DNA. The results we obtained indicate that MAb MC57 epitope could be formed by 2 regions localized on the predicted fourth and sixth extracellular loops of the current 12-transmembrane-domain model predicted for MDR1-P-glycoprotein. Surprisingly, a third region, defined by residues 800-807 of the MDR1-P-glycoprotein sequence and postulated to be intracellular, was also identified as a putative part of the MC57 epitope. This finding adds weight to the interesting hypothesis that a P-glycoprotein structure different from the current model may exist.

A new murine monoclonal antibody (MAb), MM6.15, to human MDR1 P-glycoprotein was found to be reactive in ELISA with synthetic peptides selected from the predicted sequences of the first, fourth and sixth extracellular loop of MDR1-P-glycoprotein. In order to precisely define the MM6.15-binding site, a peptide library of overlapping 5- to 9-mer residues covering the entire sixth extracellular loop of both human and rodent class-1 P-glycoproteins was synthesized on polyethylene pins and tested for MAb binding. The results of this ELISA demonstrated that the MAb MM6.15 reacts only with human synthetic peptides and that the critical component of the MAb recognition is made up of the amino-acid sequence LVAHKL (residues 963-968 of the MDR1-P-glycoprotein) with histidine (H), lysine (K) and possibly leucine (L), key residues of this immunogenic domain.

Abstract

To study the structural conformation of the MM4.17 monoclonal antibody (mAb) epitope, twenty-six mAb MM4.17-specific phage clones were affinity-isolated and their inserts characterized for amino acid composition and homology with MDR1 gene product (MDR1-P-glycoprotein). The resulting sequence alignment shows that a unique consensus sequence, which corresponds to the previously mapped TRIDDPET linear peptide identified through synthetic peptide scanning, could not be identified. However, similarities between the inserts of positive phage clones and P-glycoprotein primary structure, consisting in two or three amino acid-long sequences, were observed. An analysis of the over-represented amino acid residues in the inserts of positive clones, and their comparison with the sequence of the antigen was also performed. The two different procedures led to the identification of four regions in which these similarities are clustered, indicating that four different antigen regions, one of which includes the TRIDDPET linear amino acid sequence, might participate in forming the structure of monoclonal antibody MM4.17 epitope.

Peripheral venous blood lymphocytes were defined, in 21 subjects with essential hypertension and 20 controls, on the basis of reactivity with monoclonal antibodies OKT3, OKT4, OKT8, OK1Ia. Total lymphocyte count, OKT3+ and OKT4+ cells percentage were similar in normal and hypertensive. OKT8+ cells percentage was significantly lower only in hypertensive stage II and III patients (WHO classification).
