Purebred Holstein-Friesian cows are the main exotic breed used for milk production on large, medium, and small farms in Kenya. A study was undertaken on seven large-scale farms to investigate the genetic trends for milk production and fertility traits between 1986 and 1997 and the genetic relationships between the traits. This involved 3,185 records from 1,614 cows, the daughters of 253 sires. There was a positive trend in breeding value for 305-d milk yield of 12.9 kg/ yr and a drop in calving interval of 0.9 d/yr over the 11-yr period. Bulls from the United States (U.S.) had an average total milk yield breeding value 230 kg higher than the mean of all bulls used; Canada (+121 kg), Holland (+15 kg), the United Kingdom (U.K., 0 kg), and Kenya (-71 kg) were the other major suppliers of bulls. Average breeding values of bulls for calving interval by country of origin were -1.31 (Canada), -1.27 (Holland), -0.83 (U.S.), -0.63 (Kenya), and 0.68 d (U.K.). The genetic parameters for 305-d milk yield were 0.29 (heritability), 0.05 (permanent environment effect as proportion of phenotypic variance) resulting in an estimated repeatability of 0.34. Using complete lactation data rather than 305-d milk yield resulted in similar estimates of the genetic parameters. However, when lactation length was used as a covariate heritability was reduced to 0.25 and the permanent environment effect proportion increased to 0.09. There was little genetic control of either lactation length (heritability, 0.09) or calving interval (heritability, 0.05); however, there were strong genetic correlations between first lactation milk yield, calving interval, and age at first calving. In animals, new autosomal recessive genetic diseases (ARGD) arise all the time due to the regular, random mutations that occur during meiosis. In order to reduce the effect of any damaging new variant, it is necessary to find its cause. To evaluate the best way of doing this, 34 papers which found the exact location of a new genetic disease in livestock were reviewed and found to require at least two stages. In the initial stage the commonly used χ 2 method, applied in a case-control association analysis with single nucleotide polymorphism (SNP)-chip data, was found to have limitations and was almost always used in conjunction with a second method to locate the target region on the genome containing the variant. The commonly used methods had their drawbacks; so a new method was devised based on long runs of homozygosity, a common feature of new ARGD. This 'autozygosity by difference' method was found to be as good as, or better than, all the reviewed methods tested based on its ability to unambiguously find the shortest known target region in an already analysed data set. Mean target region length was found to be 4.6 megabases in the published reports. Success did not depend on the size of commercial SNP-chip used, and studies with as few as three cases and four controls were large enough to find the target region. The final stage relied on either sequencing the candidate genes found in the target region or using whole genome sequencing (WGS) on a small number of cases. Sometimes this latter method was used in conjunction with WGS on a number of control animals or resources such as the 1000 bull genomes data. Calculations showed that, in cattle, less than 15 animals would be needed in order to locate the new variant when using WGS data. This could be any combination of cases plus parents or other unrelated animals in the breed. Using WGS data, it would be necessary to search the three billion bases of the cattle genome for base positions which were homozygous for the same allele in all cases and heterozygous for that allele in parents, or not containing that homozygote in unrelated controls. This site could be confirmed on other healthy animals using much cheaper methods, and then a genetic test could be devised for that variant in order to screen the whole population and to devise a breeding programme to eliminate the disorder from the population. Milk production from dairy animals has been described in terms of 3 processes: the increase in secretory cell numbers in late pregnancy and early lactation, secretion rate of milk per cell, and the decline in cell numbers as lactation progresses. This latter process is thought to be determined by the level of programmed cell death (apoptosis) found in the animal. Until now, apoptosis has been measured by taking udder biopsies, using magnetic resonance imaging scans, or using animals postmortem. This paper describes an alternative, noninvasive method for estimating apoptosis by measuring microparticles in milk samples. Microparticles are the product of several processes in dairy animals, including apoptosis. Milk samples from 12 Holstein cows, at or past peak lactation, were collected at 5 monthly samplings. The samples (n = 57) were used to measure the number of microparticles and calculate microparticle density for 4 metrics: annexin V positive and merocyanine 540 dye positive, for both and total particles, in both whole milk (WM) and spun milk. Various measures of milk production were also recorded for the 12 cows, including daily milk yield, fat and protein percentage in the milk, somatic cell count, and the days in milk when the samples were taken. A high correlation was found between the 4 WM microparticle densities and days in milk (0.46 to 0.64), and a moderate correlation between WM microparticle densities and daily milk yield (-0.33 to -0.44). No significant relationships were found involving spun milk samples, somatic cell count, or fat and protein percentage. General linear model analyses revealed differences between cows for both level of microparticle density and its rate of change in late lactation. Persistency of lactation was also found to be correlated with the WM microparticle traits (-0.65 to -0.32). As apoptosis is likely to be the major contributor to microparticle numbers in late lactation, this work found a noninvasive method for estimating apoptosis that gave promising results. The rearing period has a key influence on the later performance of cattle, affecting future fertility and longevity. Producers usually aim to breed replacement heifers by 15 months to calve at 24 months. An age at first calving (AFC) close to 2 years (23 to 25 months) is optimum for economic performance as it minimises the non-productive period and maintains a seasonal calving pattern. This is rarely achieved in either dairy or beef herds, with average AFC for dairy herds usually between 26 and 30 months. Maintaining a low AFC requires good heifer management with adequate growth to ensure an appropriate BW and frame size at calving. Puberty should occur at least 6 weeks before the target breeding age to enable animals to undergo oestrous cycles before mating. Cattle reach puberty at a fairly consistent, but breed-dependent, proportion of mature BW. Heifer fertility is a critical component of AFC. In US Holsteins the conception rate peaked at 57% at 15 to 16 months, declining in older heifers. Wide variations in growth rates on the same farm often lead to some animals having delayed first breeding and/or conception. Oestrous synchronisation regimes and sexed semen can both be used but unless heifers have been previously well-managed the success rates may be unacceptably low. Altering the nutritional input above or below those needed for maintenance at any stage from birth to first calving clearly alters the average daily gain (ADG) in weight. In general an ADG of around 0.75 kg/day seems optimal for dairy heifers, with lower rates delaying puberty and AFC. There is some scope to vary ADG at different ages providing animals reach an adequate size by calving. Major periods of nutritional deficiency and/or severe calfhood disease will, however, compromise development with long-term adverse consequences. Infectious disease can also cause pregnancy loss/abortion. First lactation milk yield may be slightly lower in younger calving cows but lifetime production is higher as such animals usually have good fertility and survive longer. There is now extensive evidence that as long as the AFC is >23 months then future performance is not adversely influenced. On the other hand, delayed first calving >30 months is associated with poor survival. Underfeeding of young heifers reduces their milk production potential and is a greater problem than overfeeding. Farmers are more likely to meet the optimum AFC target of 23 to 25 months if they monitor growth rates and adjust feed accordingly. [ABSTRACT FROM PUBLISHER]

The use of molecular genetic information in the evaluation of livestock has become more common. This study looks at the efficacy of using such information to improve the genetic evaluation of a rare breed of dual-purpose cattle. Data were available in the form of pedigree information on the Gloucester cattle breed in the United Kingdom and recorded milk and beef performance on a small number of animals. In addition, molecular genetic information in the form of multi-marker, multiple regression results converted to a 1 to 10 score (Igenity scores) and 123 single nucleotide polymorphism (SNP) genotypes for 199 non-recorded animals were available. Appropriate mixed-animal models were explored for the recorded traits and these were used to calculate estimated breeding values (EBV), and their accuracies, for 6527 animals in the breed’s pedigree file. Various ways to improve the accuracy of these EBV were explored. This involved using multivariate BLUP analyses, genomic estimated breeding values (GEBV) and combining Igenity scores with recorded traits in a series of bivariate genetic analyses. Using the milk recording traits as an example, the accuracy of a number of traits could be improved using multivariate analyses by up to 14%, depending on the combination of traits used. The level of increase in accuracy largely corresponded to the absolute difference between the genetic and residual correlations between two traits, but this was not always symmetrical. The use of GEBV did not increase the accuracy of milk trait EBV owing to the low proportion of variance explained by the 101 SNPs used. Using Igenity scores in bivariate analyses with the recorded data was more successful in increasing EBV accuracy. The largest increases were found in genotyped animals with no recorded performance (e.g. a 58% increase in fat weight in milk); however, the size of the increase depended on the level of the genetic correlation between the recorded trait and the Igenity score for that trait. Lower levels of improvements in accuracy were seen in animals that were recoded but not genotyped, and ancestors which were neither genotyped nor recorded. This study demonstrated that it was possible to improve the accuracy of EBV estimation by including Igenity score information in genetic analyses but it also concluded that increasing the level of performance recording in the breed would be beneficial. Higher milk production is commonly associated with poorer fertility in dairy cows. This study used a biological model of lactation to define more closely which characteristics of lactation were linked to increased calving interval, an easily recorded measure of fertility in commercial dairy herds. Large data sets from a national milk recording scheme in the United Kingdom, collected over a 10-yr period, were used to calculate the genetic and phenotypic correlations between calving interval and a range of lactation traits within the Holstein breed. A lactation curve was fitted to each lactation, and several characteristics of each lactation were calculated. These were used in a series of mixed-model bivariate analyses with calving interval to derive the genetic parameters. When heifer lactation curve trait data were used, the highest genetic correlations were found with peak yield, maximum secretion potential, and total lactation milk yield (0.59 ± 0.06 to 0.63 ± 0.05), reflecting the observed phenomenon of poorer fertility and higher milk production. Genetic correlations for calving interval were also calculated with the rate of increase in milk yield in early lactation (0.46 ± 0.08), persistency (0.36 ± 0.09), day of peak yield (0.20 ± 0.02), and relative cell death rate (-0.12 ± 0.09). The daily production of milk components was highly genetically correlated with calving interval, with values for fat, protein, lactose, and water being 0.73 ± 0.15, 0.48 ± 0.13, 0.57 ± 0.13, and 0.50 ± 0.13. With these results and breeding values derived from these analyses, 2 possible strategies were suggested for improving the relationship between milk yield and fertility in dairy cows. First, animals that break the correlations described above could be selected (e.g., bulls with high peak yield and low calving intervals). Many governments have signed up to greenhouse gas emission (GHGE) reduction programmes under their national climate change obligations. Recently, it has been suggested that the use of extended lactations in dairy herds could result in reduced GHGE. Dairy GHGE were modelled on a national basis and the model was used to compare emissions from lactations of three different lengths (305, 370 and 440 days), and a current ‘base’ scenario on the basis of maintaining current milk production levels. In addition to comparing GHGE from the average ‘National Herd’ under these scenarios, results were used to investigate how accounting for lactations of different lengths might alter the estimation of emissions calculated from the National Inventory methodology currently recommended by Intergovernmental Panel on Climate Change. Data for the three lactation length scenarios were derived from nationally recorded dairy performance information and used in the GHGE model. Long lactations required fewer milking cows and replacements to maintain current milk yield levels than short ones, but GHGEs were found to rise from 1214 t of CO2 equivalent (CE)/farm per year for lactations of 305 days to 1371 t CE/farm per year for 440-day lactations. This apparent anomaly can be explained by the less efficient milk production (kg milk produced per kg cow weight) found in later lactation, a more pronounced effect in longer lactations. The sensitivity of the model to changes in replacement rate, persistency and level of milk yield was investigated. Changes in the replacement rate from 25% to 20% and in persistency by −10% to +20% resulted in very small changes in GHGE. Differences in GHGE due to the level of milk yield were much more dramatic with animals in the top 10% for yield, producing about 25% less GHGE/year than the average animal. National Inventory results were investigated using a more realistic spread of lactation lengths than recommended for such calculations using emissions calculated in the first part of the study. Current UK emission calculations based on the National Inventory were 329 Gg of methane per year from the dairy herd. Using the national distribution of lactation lengths, this was found to be an underestimate by about 10%. This work showed that the current rise in lactation length or a move towards calving every 18 months would increase GHGE by 7% to 14% compared with the current scenario, assuming the same milk yield in all models. Increased milk yield would have a much greater effect on reducing GHGE than changes to lactation length, replacement rate or persistency. National Inventory methodology appears to underestimate GHGE when the distribution of lactation lengths is considered and may need revising to provide more realistic figures. The calving intervals of Holstein cows in many countries have been increasing in length over recent years. Many reports of lactation characteristics refer only to a standard 305-d lactation. This paper attempts to describe the characteristics of lactations of different lengths using a large database of nationally recorded data. Three different lactation lengths were studied: 305 d (the traditional annual calving interval), 370 d (the current UK national average), and 440 d (equivalent to an 18-mo calving interval). Test-day milk yield; fat, protein, and lactose composition; and somatic cell counts were analyzed. Characteristics of each of 29,838 lactations were calculated using a biological model of lactation and were analyzed to identify which factors affected them. Maximum secretion potential was the only trait not affected by lactation length. Day of peak yield, peak yield, persistency, and total milk yield all increased with lactation length, whereas relative cell death rate and the rate of milk increase in early lactation both decreased as lactation length increased. For most weeks of lactation, the 3 lactation-length groups differed from each other in most traits, following the order of their lengths; for example, milk yield was always higher for lactations of 440 d and lower for those of 305 d. The post-peak trends in all traits were found to continue in longer lactations. Thus, daily milk production and lactose percentage continued to decrease as lactations became longer, whereas fat percentage, protein percentage, and somatic cell count continued to increase. Pregnancy was found to affect all traits, leading to an accentuation of these trends in late lactation. However, the effect of pregnancy depended on the yield at about the fourth month of pregnancy. The autosomal genes mitochondrial transcription factor A (TFAM) and uncoupling protein 2 (UCP2) are both involved in moderating mitochondrial energy production. This study investigated single nucleotide polymorphisms (SNPs) in TFAM and UCP2 and their associations with growth, fertility, milk production and survival in UK dairy cows. Holstein–Friesian heifers (n = 509) were recruited at birth and monitored until they either reached the end of their second lactation or were culled. Weight, height, length and girth were measured at 1, 6 and 15 months of age to assess growth. Fertility records were obtained for both heifers and cows and production traits (milk per day, peak yield and 305-day yield) were recorded in the first and second lactation. Mixed model and survival analyses revealed that TFAM3 GG homozygotes, representing 49% of the population, were larger than the AG heterozygotes throughout the growing period, had worse fertility, produced less milk in both lactations and were more likely to be culled. TFAM3 AA homozygotes (7% of population) were generally smaller as heifers, had slightly worse fertility and also produced less milk. This suggests a clear advantage for maximizing the heterozygotic population for this SNP. Furthermore, the results for milk per day suggest an overdominant effect, as both homozygotes of TFAM3 had significantly lower yields than the heterozygote. For UCP2, the minority CG heterozygotes (6% of population) were larger as heifers, had a reduced age at first conception and a delayed return to cyclicity after calving, when compared against the majority (94%) GG homozygotes.
