Abstract

The wide range of applications of nanoparticles (NPs) has increased the probability of environmental and occupational exposure. In a previous comparative study on cell line A549, we observed that oxidative stress caused by Co3O4 NPs affects the energetic homeostasis and the detoxification capacity, preventing autophagy induced by TiO2 NPs. In this study, we have investigated the effects of NPs on the cell cycle.Cytofluorimetric analysis showed a slow-down of the cell cycle progression for both NPs, with increases in the percentage of resting cells in the G0/G1. These observations were confirmed by a reduced expression of all cyclins, especially of CCNE1 and CDK2, involved in the late stages of the G1 phase, coupled with a significant increase in the expression of p21 only for Co3O4 NP exposure. On the contrary, the effects of TiO2 NPs were modest. Cell cycle related miRNA-34a, miRNA-126 and miRNA-1290 resulted increased at different early time-points (4-8h) but were down-expressed at 24h-48h only after TiO2 NP exposure. Our results show that NPs have an antiproliferative effect and deregulate cell cycle on A549 cells. These effects should not be underestimated because regulation of cycle progression is crucial for cell survival and repair of genetic damage.

Abstract

The available biomonitoring studies on workers producing/handling nanomaterials (NMs) focused on potential effects on respiratory, immune and cardio-vascular system. Aim of this study was to identify a panel of sensitive biomarkers and suitable biological matrices to evaluate particularly genotoxic and oxidative effects induced on workers unintentionally exposed to graphene or silica nanoparticles during the production process. These nanomaterials have been chosen for 'NanoKey' project, integrating the workplace exposure assessment (reported in part I) with the biomonitoring of exposed workers reported in the present work. Simultaneously to workplace exposure characterization, we monitored the workers using: Buccal Micronucleus Cytome (BMCyt) assay, fpg-comet test (lymphocytes), oxidized DNA bases 8-oxoGua, 8-oxoGuo and 8-oxodGuo measurements (urine), analysis of oxidative stress biomarkers in exhaled breath condensate (EBC), FENO measurement and cytokines release detection (serum). Since buccal cells are among the main targets of NM occupational exposure, particular attention was posed to the BMCyt assay that represents a noninvasive assay. This pilot study, performed on 12 workers vs.11 controls, demonstrates that BMCyt and fpg-comet assays are the most sensitive biomarkers of early, still reparable, genotoxic and oxidative effects. The findings suggest that these biomarkers could represent useful tools for the biomonitoring of workers exposed to nanoparticles, but they need to be confirmed on a high number of subjects. However, such biomarkers don't discriminate the effects of NM from those due to other chemicals used in the NM production process. Abstract Background The effects of changes in cooling temperature on biomarker levels in exhaled breath condensate have been little investigated. The aim of the study was to test the effect of condensation temperature on the parameters of exhaled breath condensate and the levels of selected biomarkers. Methods Exhaled breath condensate was collected from 24 healthy subjects at temperatures of -10, -5, 0 and +5 C degrees. Selected parameters (condensed volume and conductivity) and biomarkers (hydrogen peroxide, malondialdehyde) were measured. Results There was a progressive increase in hydrogen peroxide and malondialdehyde concentrations, and condensate conductivity as the cooling temperature increased; total condensate volume increased as the cooling temperature decreased. Conclusion The cooling temperature of exhaled breath condensate collection influenced selected biomarkers and potential normalizing factors (particularly conductivity) in different ways ex vivo. The temperature of exhaled breath condensate collection should be controlled and reported.

Abstract

Abstract Background Non-invasive diagnostic strategies aimed at identifying biomarkers of lung cancer are of great interest for early cancer detection. The aim of this study was to set up a new method for identifying and quantifying volatile organic compounds (VOCs) in exhaled air of patients with non-small cells lung cancer (NSCLC), by comparing the levels with those obtained from healthy smokers and non-smokers, and patients with chronic obstructive pulmonary disease. The VOC collection and analyses were repeated three weeks after the NSCLC patients underwent lung surgery. Methods The subjects' breath was collected in a Teflon® bulb that traps the last portion of single slow vital capacity. The 13 VOCs selected for this study were concentrated using a solid phase microextraction technique and subsequently analysed by means of gas cromatography/mass spectrometry. Results The levels of the selected VOCs ranged from 10-12 M for styrene to 10-9 M for isoprene. None of VOCs alone discriminated the study groups, and so it was not possible to identify one single chemical compound as a specific lung cancer biomarker. However, multinomial logistic regression analysis showed that VOC profile can correctly classify about 80 % of cases. Only isoprene and decane levels significantly decreased after surgery. Conclusion As the combination of the 13 VOCs allowed the correct classification of the cases into groups, together with conventional diagnostic approaches, VOC analysis could be used as a complementary test for the early diagnosis of lung cancer. Its possible use in the follow-up of operated patients cannot be recommended on the basis of the results of our short-term nested study.

Abstract

Nell’ambito della diagnosi precoce di tumore al polmone, una tematica di grande interesse è rappresentata dalla ricerca di nuove strategie diagnostiche non invasive per l’identificazione di nuovi indicatori. Tali tecniche potrebbero contribuire alla conferma della malattia nei casi sospetti e all’esclusione dei soggetti con noduli polmonari benigni, frequentemente individuati dalla moderne tecniche di imaging. Criteri puramente morfologici non sembrano sufficienti per distinguere con attentibilità univoca il tumore al polmone ad uno stadio precoce dai noduli benigni, aumentando così l’incidenza di falsi positivi. Tra i test diagnostici non invasivi, l’analisi dell’aria espirata rappresenta una metodica promettente, grazie ai buoni valori di sensibilità e specificità dimostrati. Lo scopo del presente studio è stato quello di identificare e quantificare i composti organici volatili (VOCs) nell’aria esalata di pazienti con tumore non a piccole cellule (NSCLC) e confrontarne i livelli con quelli ottenuti in soggetti asintomatici fumatori e non, e in pazienti con broncopneumopatia cronica ostruttiva (BPCO). L’analisi dell’aria espirata è stata ripetuta a distanza di tempo dall’intervento chirurgico per valutare l’effetto dell’intervento stesso sul profilo dei VOCs. Ulteriori esperimenti sono stati fatti per meglio comprendere l’origine dei VOCs ed il loro significato in relazione allo sviluppo della neoplasia. Non-invasive diagnostic strategies aimed at identifying biomarkers of lung cancer are of great interest for early cancer detection. They could be helpful in picking out lung cancer in suspected cases, but also and especially to exclude its occurrence in cases of those pulmonary nodules more and more frequently detected by CT scans. In fact, purely morphological criteria seem to be insufficient to confidently distinguish lung cancer at early stages from benign nodules, and false positives seem to occur frequently. Among non-invasive tests, analysis of exhaled air seems to be very promising thanks to its sensitivity and specificity. The aim of this study was to set up a new method for identifying and quantifying volatile organic compounds (VOCs) in exhaled air of patients with non-small cell lung cancer (NSCLC), by comparing the levels with those obtained from asymptomatic smokers and non-smokers, and patients with chronic obstructive pulmonary disease (cross sectional study). The VOC collection and analyses were repeated after the NSCLC patients undergone lung surgery (longitudinal study). Further experiments were performed to better understand the origin of the VOCs and their meaning related to the development of lung cancer.
