This paper describes and illustrates a new species of Primulaceae, Primula wolongensis sp. nov. from Wolong National Nature Reserve in Sichuan Province, China. It is very rare and currently only known from its type locality. The new species belongs to subsection Chartacea of the section Petiolares on account of lacking bud scales at flowering, being efarinose and having distinct petiolate leaves with more or less rounded lamina. The new species can be differentiated from other members of the subsection by leaf blade margin dentate, and leaf veins which are not raised, scape shorter than or equal to pedicels, yellow flowers and location of stamens of the corolla tube at thrum flower. Molecular phylogenetic analysis based on nuclear ribosome internal transcribed spacer (nrITS) demonstrated that P. wolongensis was sister to subgen. Auriculastrum. Primula wolongensis is currently known from a single location in Wolong Town, and its conservation status is assessed as Data Deficient (DD).

22. Microbial mats as shelter microhabitat for amphipods in an intermittent karstic spring

Microbial mats represent complex communities where cyanobacteria and diatoms as key organisms provide shelter for diverse assemblages of aquatic invertebrates, like the small stygophilous amphipod Synurella ambulans. Studies addressing such communities in the karst springs have rarely examined springheads, and have ignored intermittent springs. During high flow conditions the stygophilic crustaceans are flushed to the surface of a temporary stream Krčić where microbial mats prevent their drift and enables their successful retreat into underground in the periods of drought. The objective of this study was to characterize the microbial mat community of the Krčić Spring as a shelter for S. ambulans during strong current and high water level. Representative samples for diatom and cyanobacterial species identification and composition, as well as the fresh mat material for potential animal activity and cyanobacterial phylogenetic analysis were collected. The most dominant diatom was Achnanthidium minutissimum, whilst Fragilaria capucina, Meridion circulare, Navicula cryptocephala and Nitzschia palea had abundance greater than 0.5%. Morphological observations of cyanobacteria revealed that Phormidium favosum was the most dominant, with Hydrocoleum muscicola as a subdominant. Cyanobacterial phylogenetic relationship revealed two distinct clusters: (i) "Phormidium cluster", confirming morphological observations in both winter and spring samples, and (ii) "Wilmottia cluster", a first report for Croatia and found exclusively in the winter sample. Laboratory observations revealed a small stygophilic amphipod S. ambulans, hiding and feeding inside the pockets of fresh microbial mat. The intermittent Krčić Spring as a predator-free and competitor-free ecosystem provides a spatiotemporal conformity between microbial mat and stygophilous amphipod.

Insight into how environmental change determines the production and distribution of cyanobacterial toxins is necessary for risk assessment. Management guidelines currently focus on hepatotoxins (microcystins). Increasing attention is given to other classes, such as neurotoxins (e.g., anatoxin-a) and cytotoxins (e.g., cylindrospermopsin) due to their potency. Most studies examine the relationship between individual toxin variants and environmental factors, such as nutrients, temperature and light. In summer 2015, we collected samples across Europe to investigate the effect of nutrient and temperature gradients on the variability of toxin production at a continental scale. Direct and indirect effects of temperature were the main drivers of the spatial distribution in the toxins produced by the cyanobacterial community, the toxin concentrations and toxin quota. Generalized linear models showed that a Toxin Diversity Index (TDI) increased with latitude, while it decreased with water stability. Increases in TDI were explained through a significant increase in toxin variants such as MC-YR, anatoxin and cylindrospermopsin, accompanied by a decreasing presence of MC-LR. - Abstract
To determine the drivers of phytoplankton biomass, we collected standardized morphometric, physical, and biological data in 230 lakes across the Mediterranean, Continental, and Boreal climatic zones of the European continent. Multilinear regression models tested on this snapshot of mostly eutrophic lakes (median total phosphorus [TP] = 0.06 and total nitrogen [TN] = 0.7 mg L−1), and its subsets (2 depth types and 3 climatic zones), show that light climate and stratification strength were the most significant explanatory variables for chlorophyll a (Chl a) variance. TN was a significant predictor for phytoplankton biomass for shallow and continental lakes, while TP never appeared as an explanatory variable, suggesting that under high TP, light, which partially controls stratification strength, becomes limiting for phytoplankton development. Mediterranean lakes were the warmest yet most weakly stratified and had significantly less Chl a than Boreal lakes, where the temperature anomaly from the long-term average, during a summer heatwave was the highest (+4°C) and showed a significant, exponential relationship with stratification strength. This European survey represents a summer snapshot of phytoplankton biomass and its drivers, and lends support that light and stratification metrics, which are both affected by climate change, are better predictors for phytoplankton biomass in nutrient-rich lakes than nutrient concentrations and surface temperature.

This study presents a comprehensive morphological comparison along with molecular phylogeny of the genus Gloydius based on five mitochondrial genes (12S, 16S, COI, cytb, and ND4). The specimens collected from Jiuzhaigou National Nature Reserve are shown to be a new species, Gloydius lateralis sp. nov. Zhang, Shi, Jiang & Shi based on a combination of morphological and molecular accounts. G. lateralis sp. nov. differs from other congeneric species by a series of diagnostic morphological characteristics and forms a strongly supported monophyletic group. The new species is phylogenetically closely related to G. swild, another recently described species from Heishui, Aba, Sichuan. [ABSTRACT FROM AUTHOR]

Biodiversity protection and sustainable management of freshwater karstic ecosystems require a set of good governmental policies and measures based on competent decision-making, and establishment of relevant practices founded on the best available technologies and high quality knowledge. Traditional monitoring systems have several substantial drawbacks, like tediousness and expensiveness, difficult automation of processes and doubtful sensitivity, as well as inability to provide prompt, immediate information about the ecological status of a lake. To overcome these deficiencies new innovative methods and alternative rapid monitoring tools, such as environmental DNA metabarcoding (eDNA), pigment analyses and automatic monitoring systems for high-frequency measuring and real-time management based on development of the Information and Communications Technology (ICT), were developed in recent years. The current study, carried out during August 2018 on Lake Visovac (Croatia), provides results of taxonomic analyses, phytoplankton biomass, in situ measuring of physical and chemical parameters, chlorophyll a concentration and eDNA analyses, all in correlation with data collected from the in situ water sensor installed on the buoy. In contrast to single point measurements of the water sensor, phytoplankton composition and biomass were investigated in vertical profiles on 10 sampling points along the limnetic and littoral zone of the Lake. A strong separation between limnetic and littoral zone samples was recorded, together with statistically significant differences in the ecological status based on phytoplankton as biological quality element (BQE). Grouping of buoy sensor data with samples from the littoral zone revealed that buoy data were not comparable with data from the monitoring site in the limnetic zone. Moreover, analyses of chlorophyll a and eukaryotic eDNA have shown comparable results with traditionally obtained ecological status based on BQE phytoplankton and can be successfully used in biomonitoring.

Krka River is a unique freshwater ecosystem situated in Dinaric karstic area of Croatia. Knowledge concerning diatom diversity, ecology and community associations especially of karstic environments in Croatia is still very scarce. Diversity of benthic diatom species from various habitats, both lotic and lentic, from springhead to the Krka River estuary was studied in September 2017. A total of 44 samples were collected and examined for taxonomic analyses using light and scanning electron microscopy. Altogether, 276 taxa (species, varieties and forms) from 69 genera were identified by traditional morphology analyses. Flora dominated by genera Navicula Bory sensu stricto, Nitzschia Hasall and Gomphonema Ehrenberg was observed. The most frequent species were Navicula cryptotenella Lange-Bertalot, N. radiosa Kützing, Pantocsekiella ocellata (Pantocsek) K.T. Kiss & E. Ács, Encyonopsis minuta Krammer & E.Reichardt, and Achnanthidium minutissimum (Kützing) Kützing. Species with unclear taxonomic status (Reimeria sp. aff. uniseriata, Aneumastus aff. tusculus, Aneumastus aff. stroesei, Diploneis submarginestriata) were also recorded together with Gomphosphaenia plenkoviciae Gligora Udovič & Žutinić, a newly described diatom species from Crveno jezero, Croatia. For molecular analyses, 38 samples were exposed using the universal eukaryotic primer pair for the hypervariable V9-region. When comparing morphological and genetic results of represented Operational Taxonomic Units (OTUs), the most frequent species were Aulacoseira ambigua (Grunow) Simonsen, Cocconeis placentula Ehrenberg, Cymatopleura elliptica (Brébisson) W.Smith, Aulacoseira granulata var. angustissima (Otto Müller) Simonsen, Staurosirella pinnata (Ehrenberg) D.M.Williams & Round, and Achnanthidium minutissimum (Kützing) Kützing. These preliminary results of V9 region performed high number of OTUs for diatom species, but OTUs didn’t show high resolution of overlap with traditional morphological-based identification. In order to obtain a better resolution, further research will include primers for the highly conserved rbcl gene.

situ Insight into how environmental change determines the production and distribution of cyanobacterial toxins is necessary for risk assessment. Management guidelines currently focus on hepatotoxins (microcystins). Increasing attention is given to other classes, such as neurotoxins (e.g., anatoxin-a) and cytotoxins (e.g., cylindrospermopsin) due to their potency. Most studies examine the relationship between individual toxin variants and environmental factors, such as nutrients, temperature and light. In summer 2015, we collected samples across Europe to investigate the effect of nutrient and temperature gradients on the variability of toxin production at a continental scale. Direct and indirect effects of temperature were the main drivers of the spatial distribution in the toxins produced by the cyanobacterial community, the toxin concentrations and toxin quota. Generalized linear models showed that a Toxin Diversity Index (TDI) increased with latitude, while it decreased with water stability. Increases in TDI were explained through a significant increase in toxin variants such as MC-YR, anatoxin and cylindrospermopsin, accompanied by a decreasing presence of MC-LR. While global warming continues, the direct and indirect effects of increased lake temperatures will drive changes in the distribution of cyanobacterial toxins in Europe, potentially promoting selection of a few highly toxic species or strains.

Microbes show a recurrent structure in which a relatively small number of abundant bacteria coexist with the "rare" biosphere. Abundant taxa are dynamic over time and space and reactive to local conditions. Dispersal limitation could have a strong influence on the community composition. During a monitoring program in 2016 (April- September), we investigated the microbial community composition in four Croatian freshwater lakes with different origin and trophic status: Lake Visovac -mesotrophic karst lake located in the Krka National Park ; Lake Prošće - an oligo- to mesotrophic karst lake located in the Plitvice Lakes National Park ; Lake Jarun - a mesotrophic, anthropogenic lake formed 30 year ago in the city of Zagreb ; and Lake Sakadaš - an eutrophic floodplain lake in the Danubian valley of the Kopački Rit Nature Park. Lake Sakadaš showed the most even microbial community composition, with various Beta- and Gammaproteobacteria, Actinobacteria and Verrucomicrobia co- occurring. Microbial communities in Lakes Visovac and Jarun were strongly dominated by the actinobacterial genus Ca. Planktophila, a representative of the yet uncultivated acI lineage, frequently detected as the most numerous taxon among total freshwater bacterioplankton. Interestingly, Actinobacteria were not as dominant in Lake Prošće. Instead, various Comamonadaceae- and Burkolderiaceae- related Betaproteobacteria occurred in similar frequencies. Notably, stark shifts in the dominant community members were observed in Lake Prošće during our sampling, indicating a less stable microbial core community.

Cryptophytes, chrysophytes and dinoflagellates are groups of phytoplankton largely consisting of species with well documented mixotrophy. These groups also comprise a major part of phytoplankton biomass in karstic lakes with good to high ecological status. The aim of the study was to establish the link between bacterial community and potential mixotrophs in the planktonic community of a karstic mesotrophic Visovac Lake (Krka National Park, Croatia), in order to provide a precise assessment of the ecological status of lake ecosystem. Integrated vertical samples for phytoplankton, samples for physical-chemical parameters, water chemistry analyses and bacteriology were taken simultaneously from the deepest point of the lake, once a month in the period between April and September 2016. Phytoplankton composition and biomass were determined by standard methods, while fluorescence in situ hybridization with signal amplification and catalyzed reporter deposition (CARD-FISH) was applied for bacterial microscopic quantification. Additionally, filtered water was used for DNA extraction and 16S rRNA gene sequencing for bacterial identification. Total phytoplankton biomass varied from 0.24 mgL-1 (June) to 0.75 mgL- 1 (July). Potentially mixotrophic species characterizing the community were: Dinobryon divergens, Dinobryon bavaricum, Dinobryon crenulatum, Chromulina sp., Ochromonas danica, Plagioselmis nannoplanctica, Cryptomonas sp., Parvodinium inconspicuum and Ceratium hirundinella. The lowest total bacterial abundance was determined in April (2.61 x 10x6 cell mL-1), whilst the highest was in July (11.39 x 10x6 cell mL-1). Sequencing of 6 integrated samples resulted in a total of 132 429 nucleotide sequences, with 5 bacterial groups identified: Betaproteobacteria, Actinobacteria, Bacterioidetes, Chloroflexi and Veruccomicrobia. Actinobacteria was the most abundant class in all investigated months (avg. 34%), while Betaproteobacteria was subdominant (avg. 22%). Multivariate analysis provided a statistically significant corelation between the selected phytoplankton species and determined bacterial groups. The results will be used to determine relation between mixotrophic species and bacterial groups in lakes with variable ecological status.

Under ongoing climate change and increasing anthropogenic activity, which continuously challenge ecosystem resilience, an in-depth understanding of ecological processes is urgently needed. Lakes, as providers of numerous ecosystem services, face multiple stressors that threaten their functioning. Harmful cyanobacterial blooms are a persistent problem resulting from nutrient pollution and climate-change induced stressors, like poor transparency, increased water temperature and enhanced stratification. Consistency in data collection and analysis methods is necessary to achieve fully comparable datasets and for statistical validity, avoiding issues linked to disparate data sources. The European Multi Lake Survey (EMLS) in summer 2015 was an initiative among scientists from 27 countries to collect and analyse lake physical, chemical and biological variables in a fully standardized manner. This database includes in-situ lake variables along with nutrient, pigment and cyanotoxin data of 369 lakes in Europe, which were centrally analysed in dedicated laboratories. Publishing the EMLS methods and dataset might inspire similar initiatives to study across large geographic areas that will contribute to better understanding lake responses in a changing environment. Iako su istraživanja fitoplanktona tradicionalno usmjerena prema određivanju sastava, brojnosti i biomase zajednice korištenjem metode mikroskopiranja, u novije se vrijeme razvijaju brze metode, poput analize pigmenata putem tekuće kromatografije visoke djelotvornosti (HPLC). Cilj istraživanja bio je opisati fitoplanktonsku zajednicu Visovačkog jezera (NP Krka, Hrvatska) pomoću metoda mikroskopije i analize pigmenata (HLPC) te usporediti rezultate metoda s ciljem procjene primjenjivosti i učinkovitosti analize pigmenata na slatkovodnim krškim sustavima. Uzorci vertikalnih profila prikupljeni su na svakih 5 metara dubine sa 7 postaja tijekom jednodnevnog terena u srpnju 2012. i svibnju 2013. godine na Visovačkom jezeru. Udio pigmenata u uzorcima određen je HPLC metodom, pri čemu su indikatorski pigmenti odabrani na temelju kemotaksonomskih značajki: fukoksantin kao indikator dijatomeja, peridinin kao indikator dinoflagelata, aloksantin kao indikator kriptofita, zeaksantin kao indikator cijanobakterija te klorofil b kao indikator zelenih algi. Udio pigmenata podudarao se s udjelom biomase pripadajućih skupina. Navedena se metoda pokazala usporedivom metodi mikroskopiranja u opisu fitoplanktonske zajednice jezera Visovac. Zbog svoje jednostavnosti i brzine uporaba HPLC analize u budućim bi istraživanjima krških slatkovodnih ekosustava, u kombinaciji s mikroskopiranjem, omogućila detaljniji i obuhvatniji uvid u prostornu i vremensku dinamiku fitoplanktonskih zajednica.

- Abstract
To determine the drivers of phytoplankton biomass, we collected standardized morphometric, physical, and biological data in 230 lakes across the Mediterranean, Continental, and Boreal climatic zones of the European continent. Multilinear regression models tested on this snapshot of mostly eutrophic lakes (median total phosphorus [TP] = 0.06 and total nitrogen [TN] = 0.7 mg L−1), and its subsets (2 depth types and 3 climatic zones), show that light climate and stratification strength were the most significant explanatory variables for chlorophyll a(Chl a) variance. TN was a significant predictor for phytoplankton biomass for shallow and continental lakes, while TP never appeared as an explanatory variable, suggesting that under high TP, light, which partially controls stratification strength, becomes limiting for phytoplankton development. Mediterranean lakes were the warmest yet most weakly stratified and had significantly less Chl athan Boreal lakes, where the temperature anomaly from the long‐term average, during a summer heatwave was the highest (+4°C) and showed a significant, exponential relationship with stratification strength. This European survey represents a summer snapshot of phytoplankton biomass and its drivers, and lends support that light and stratification metrics, which are both affected by climate change, are better predictors for phytoplankton biomass in nutrient‐rich lakes than nutrient concentrations and surface temperature.

- Abstract
Insight into how environmental change determines the production and distribution of cyanobacterial toxins is necessary for risk assessment. Management guidelines currently focus on hepatotoxins (microcystins). Increasing attention is given to other classes, such as neurotoxins (e.g., anatoxin-a) and cytotoxins (e.g., cylindrospermopsin) due to their potency. Most studies examine the relationship between individual toxin variants and environmental factors, such as nutrients, temperature and light. In summer 2015, we collected samples across Europe to investigate the effect of nutrient and temperature gradients on the variability of toxin production at a continental scale. Direct and indirect effects of temperature were the main drivers of the spatial distribution in the toxins produced by the cyanobacterial community, the toxin concentrations and toxin quota. Generalized linear models showed that a Toxin Diversity Index (TDI) increased with latitude, while it decreased with water stability. Increases in TDI were explained through a significant increase in toxin variants such as MC-YR, anatoxin and cylindrospermopsin, accompanied by a decreasing presence of MC-LR. While global warming continues, the direct and indirect effects of increased lake temperatures will drive changes in the distribution of cyanobacterial toxins in Europe, potentially promoting selection of a few highly toxic species or strains.

- Abstract
Under ongoing climate change and increasing anthropogenic activity, which continuously challenge ecosystem resilience, an in-depth understanding of ecological processes is urgently needed. Lakes, as providers of numerous ecosystem services, face multiple stressors that threaten their functioning. Harmful cyanobacterial blooms are a persistent problem resulting from nutrient pollution and climate-change induced stressors, like poor transparency, increased water temperature and enhanced stratification. Consistency in data collection and analysis methods is necessary to achieve fully comparable datasets and for statistical validity, avoiding issues linked to disparate data sources. The European Multi Lake Survey (EMLS) in summer 2015 was an initiative among scientists from 27 countries to collect and analyse lake physical, chemical and biological variables in a fully standardized manner. This database includes in-situ lake variables along with nutrient, pigment and cyanotoxin data of 369 lakes in Europe, which were centrally analysed in dedicated laboratories. Publishing the EMLS methods and dataset might inspire similar initiatives to study across large geographic areas that will contribute to better understanding lake responses in a changing environment.

- Abstract
Under ongoing climate change and increasing anthropogenic activity, which continuously challenge ecosystem resilience, an in-depth understanding of ecological processes is urgently needed. Lakes, as providers of numerous ecosystem services, face multiple stressors that threaten their functioning. Harmful cyanobacterial blooms are a persistent problem resulting from nutrient pollution and climate-change induced stressors, like poor transparency, increased water temperature and enhanced stratification. Consistency in data collection and analysis methods is necessary to achieve fully comparable datasets and for statistical validity, avoiding issues linked to disparate data sources. The European Multi Lake Survey (EMLS) in summer 2015 was an initiative among scientists from 27 countries to collect and analyse lake physical, chemical and biological variables in a fully standardized manner. This database includes in-situ lake variables along with nutrient, pigment and cyanotoxin data of 369 lakes in Europe, which were centrally analysed in dedicated laboratories. Publishing the EMLS methods and dataset might inspire similar initiatives to study across large geographic areas that will contribute to better understanding lake responses in a changing environment.
