1. A Transition Edge Sensor Operated in Coincidence with a High Sensitivity Phonon Veto for Photon Coupled Rare Event Searches

Abstract

Experimental searches for axions or dark photons that couple to the standard model photon require photosensors with low noise, broadband sensitivity, and near zero backgrounds. Here, we introduce a Transition Edge Sensor (TES) based photon sensor with a high sensitivity athermal phonon sensor coupled to the substrate on which the TES is deposited. We show that single photons absorbed locally in the TES have $\sim$35% energy deposition in the electronic system of the TES, with $\sim$26% of the photon energy absorbed by the athermal photon sensor (due to athermal phonons leaking out of the TES during the downconversion process). Backgrounds are observed to be largely coupled to either the TES or phonon system. At high energies, these backgrounds can be efficiently discriminated from TES photon absorption events, while at low energies, their leakage into the passage band is well modeled. With significant sensitivity improvements to both the photon and phonon channel, this coincidence technique could be used to suppress backgrounds in bosonic dark matter searches down to energies near the superconducting bandgap of the sensor., Comment: 5 pages, 5 figures

Abstract

The performance of superconducting qubits is degraded by a poorly characterized set of energy sources breaking the Cooper pairs responsible for superconductivity, creating a condition often called quasiparticle poisoning. Both superconducting qubits and low threshold dark matter calorimeters have observed excess bursts of quasiparticles or phonons that decrease in rate with time. Here, we show that a silicon crystal glued to its holder exhibits a rate of low-energy phonon events that is more than two orders of magnitude larger than in a functionally identical crystal suspended from its holder in a low-stress state. The excess phonon event rate in the glued crystal decreases with time since cooldown, consistent with a source of phonon bursts which contributes to quasiparticle poisoning in quantum circuits and the low-energy events observed in cryogenic calorimeters. We argue that relaxation of thermally induced stress between the glue and crystal is the source of these events.

Abstract

The phonon-mediated hybrid detector is made out of a monolithic silicon crystal characterized by two interconnected regions linked through a narrow neck. Operating solely on phonon signal measurements, the hybrid design facilitates the differentiation between electron recoil and nuclear recoil events, effectively discerning two types of interaction down to low energy levels. With a newly implemented software triggering technique (SWT), low-energy nuclear recoil events of approximately $500~eV_{ee}$ have been measured. In addition to this, an electron recoil background reduction of up to $95\%$ has been successfully demonstrated.

Abstract

Superconducting sensors are a key enabling technology for many HEP experiments with advances in sensor capabilities leading directly to expanded science reach. The unique materials and processes required for the fabrication of these sensors makes commercial sourcing impractical in comparison with semiconducting devices. Subsequently, the development and fabrication of new sensors are often performed at academic cleanrooms supported through HEP basic detector research and/or project funds. While this operational model has been successful to date, we are at a turning point in the history of superconducting electronics, as evidenced by the rapid growth in the field of quantum computing, when scale and sophistication of these sensors can lead to significant progress. Abstract

We demonstrate single electron-hole pair resolution in a single-sided, contact-free 1 cm$^2$ by 1 mm thick Si crystal operated at 48 mK, with a baseline energy resolution of 3 eV. This crystal can be operated at voltages in excess of $\pm50$ V, resulting in a measured charge resolution of 0.06 electron-hole pairs. The high aluminum coverage ($\sim$70%) of this device allows for the discrimination of surface events and separation of events occurring near the center of the detector from those near the edge. Abstract

Keeping track of the ever-increasing body of scientific literature is an escalating challenge. We present PubTree a hierarchical search tool that efficiently searches the PubMed/MEDLINE dataset based upon a decision tree constructed using >26 million abstracts. The tool is implemented as a webpage, where users are asked a series of eighteen questions to locate pertinent articles. The implementation of this hierarchical search tool highlights issues endemic with document retrieval. However, the construction of this tree indicates that with future developments hierarchical search could become an effective tool (or adjunct) in the mining of biological literature., Comment: 7 pages, 2 figures

A dedicated facility has been commissioned for Cryogenic Dark Matter Search (CDMS) detector fabrication at Texas A&M University (TAMU). The fabrication process has been carefully tuned using this facility and its equipment. Production of successfully tested detectors has been demonstrated. Significant improvements in detector performance have been made using new fabrication methods/equipment and tuning of process parameters.

Abstract

Objectives: Extracellular vesicles (EVs) are heterogeneous membrane vesicles in diameter of 30-5000 nm, that transport proteins, non-coding RNAs (miRNAs), lipids and metabolites. Major populations include exosomes, ectosomes and apoptotic bodies. The purpose of this study was to compare the distribution of EVs obtained under different conditions of differential centrifugation, including ultracentrifugation, with the results developed based on a theoretical model. Methods: Immortalized endothelial cell line that expresses h-TERT (human telomerase) was used to release of EVs: microvascular TIME. EVs were isolated from the culture medium at different centrifugation parameters. The size distribution of the EVs was measured using TRPS technology on a qNano instrument. Surface markers were evaluated using flow cytometry. The isolated EV subpopulations were compared with the theoretical model developed by Livshits. Results: EVs isolated from endothelial cells show strong aggregating properties, which was confirmed by TEM, TRPS imaging and flow cytometry. Conclusions: Obtaining pure EV subpopulations is difficult because of the small differences in the diameter of ectosomes and exosomes, and the strong aggregating properties of EVs.

29. Metabolic changes in glioblastomas in response to choline kinase inhibition: In vivo MRS in rodent models.

Abstract

Changes in glioblastoma (GBM) metabolism was investigated in response to JAS239, a choline kinase inhibitor, using MRS. In addition to the inhibition of phosphocholine synthesis, we investigated changes in other key metabolic pathways associated with GBM progression and treatment response. Three syngeneic rodent models of GBM were used: F98 (N = 12) and 9L (N = 8) models in rats and GL261 (N = 10) in mice. Rodents were intracranially injected with GBM cells in the right cortex and tumor growth was monitored using T2‐weighted images. Animals were treated once daily with intraperitoneal injections of 4 mg/kg JAS239 (F98 rats, n = 6; 9L rats, n = 6; GL261 mice, n = 5) or saline (control group, F98 rats, n = 6; 9L rats, n = 2; GL261 mice, n = 5) for five consecutive days. Single voxel spectra were acquired on Days 0 (T0, baseline) and 6 (T6, end of treatment) from the tumor as well as the contralateral normal brain using a PRESS sequence. Changes in metabolite ratios (tCho/tCr, tCho/NAA, mI/tCr, Glx/tCr and (Lip + Lac)/Cr) were used to assess metabolic pathway alterations in response to JAS239. Tumor growth arrest was noted in all models in response to JAS239 treatment compared with saline‐treated animals, with a significant reduction (p < 0.05) in the F98 model. A reduction in tCho/tCr was observed with JAS239 treatment in all GBM models, indicating reduced phospholipid metabolism, with the highest reduction in 9L followed by GL261 and F98 tumors. A significant reduction (p < 0.05) in the tCho/NAA ratio was observed in the 9L model. A significant reduction in mI/tCr (p < 0.05) was found in JAS239‐treated F98 tumors compared with the saline‐treated animals. A non‐significant trend of reduction in Glx/tCr was observed only in F98 and 9L tumors. JAS239‐treated F98 tumors also showed a significant increase in Lip + Lac (p < 0.05), indicating increased cell death. This study demonstrated the utility of MRS in assessing metabolic changes in GBM in response to choline kinase inhibition. [ABSTRACT FROM AUTHOR]

Abstract

Many low-threshold experiments observe sharply rising event rates of yet unknown origins below a few hundred eV, and larger than expected from known backgrounds. Due to the significant impact of this excess on the dark matter or neutrino sensitivity of these experiments, a collective effort has been started to share the knowledge about the individual observations. For this, the EXCESS Workshop was initiated. In its first iteration in June 2021, ten rare event search collaborations contributed to this initiative via talks and discussions. The contributing collaborations were CONNIE, CRESST, DAMIC, EDELWEISS, MINER, NEWS-G, NUCLEUS, RICOCHET, SENSEI and SuperCDMS. They presented data about their observed energy spectra and known backgrounds together with details about the respective measurements. In this paper, we summarize the presented information and give a comprehensive overview of the similarities and differences between the distinct measurements. Abstract

Patients suffering from cystic fibrosis (CF) commonly harbor the important pathogen Pseudomonas aeruginosa in their airways. During chronic late-stage CF, P. aeruginosa is known to grow under reduced oxygen tension and is even capable of respiring anaerobically within the thickened airway mucus, at a pH of ~6.5. Therefore, proteins involved in anaerobic metabolism represent potentially important targets for therapeutic intervention. In this study, the clinically relevant 'anaerobiome' or 'proteogenome' of P. aeruginosa was assessed. First, two different proteomic approaches were used to identify proteins differentially expressed under anaerobic versus aerobic conditions. Microarray studies were also performed, and in general, the anaerobic transcriptome was in agreement with the proteomic results. However, we found that a major portion of the most upregulated genes in the presence of N[O.sub.3.sup.-] and N[O.sub.2.sup.-] are those encoding Pfl bacteriophage. With anaerobic [O.sub.2.sup.-], the most downregulated genes are those involved postglycolytically and include many tricarboxylic acid cycle genes and those involved in the electron transport chain, especially those encoding the NADH dehydrogenase I complex. Finally, a signature-tagged mutagenesis library of P. aeruginosa was constructed to further screen genes required for both N[O.sub.3.sup.-] and N[O.sub.2.sup.-] respiration. In addition to genes anticipated to play important roles in the anaerobiome (anr, dnr, nar, nir, and nuo), the cysG and dksA genes were found to be required for both anaerobic N[O.sub.3.sup.-] and N[O.sub.2.sup.-] respiration. This study represents a major step in unraveling the molecular machinery involved in anaerobic N[O.sub.3.sup.-] and N[O.sub.3.sup.-] respiration and offers clues as to how we might disrupt such pathways in P. aeruginosa to limit the growth of this important CF pathogen when it is either limited or completely restricted in its oxygen supply.

37. Mass spectrometric identification of phosphorylation sites of rRNA transcription factor upstream binding factor

Abstract

rRNA transcription is a fundamental requirement for all cellular growth processes and is activated by the phosphorylation of the upstream binding factor (UBF) in response to growth stimulation. Even though it is well known that phosphorylation of UBF is required for its activation and is a key step in activation of rRNA transcription, as yet, there has been no direct mapping of the UBF phosphorylation sites. The results of the present studies employed sophisticated nano-flow HPLC-microelectrospray-ionization tandem mass spectrometry (nHPLC-[mu]ESI-MS/MS) coupled with immobilized metal affinity chromatography (IMAC) and computer database searching algorithms to identify 10 phosphorylation sites on UBF at serines 273, 336, 364, 389, 412, 433, 484, 546, 584, and 638. We then carried out functional analysis of two of these sites, serines 389 and 584. Serine-alanine substitution mutations of 389 (S389A) abrogated rRNA transcription in vitro and in vivo, whereas mutation of serine 584 (S584A) reduced transcription in vivo but not in vitro. In contrast, serine-glutamate mutation of 389 (S389E) restored transcriptional activity. Moreover, $389A abolished UBF-SL1 interaction in vitro, while S389E partially restored UBF-SL1 interaction. Taken together, the results of these studies suggest that growth factor stimulation induces an increase in rRNA transcriptional activity via phosphorylation of UBF at serine 389 in part by facilitating a rate-limiting step in the recruitment of RNA polymerase I: i.e., recruitment of SL1.
