Aim: It is a known fact that diabetes mellitus is increasing frequently and triggering many different diseases. Therefore, early diagnosis of the disease is important. This study was trying to predict the early diagnosis of the disease, according to machine learning methods by measuring plasma glucose concentration, serum insulin resistance, and diastolic blood pressure. Material and Methods: In the study, the public dataset from a website consists of 768 samples and nine variables. Three different machine learning strategies were used in the early diagnosis of diabetes mellitus (Support Vector Machine, Multilayer Perceptron, and Stochastic Gradient Boosting). 3 repeats and 10 fold cross-validation method was used to optimize the hyperparameters. The model’s performance parameters were evaluated based on accuracy, specificity, sensitivity, confusion matrix, positive predictive value (precision), negative predictive value, and AUC (area under the ROC curve). Results: According to the experimental results (the criteria of accuracy (0.79), sensitivity (0.57), specificity (0.91), positive predictive value (0.79), negative predictive value (0.80), and AUC (0.74)) the Support Vector Machine was more successful than other methods. Conclusion: Plasma glucose concentration, serum insulin resistance, and diastolic blood pressure markers are important indicators in the early diagnosis of diabetes mellitus. In this study, it was seen that these markers make a significant contribution to the early diagnosis of diabetes mellitus. However, it has been observed that these indicators alone will not be sufficient in the early diagnosis of the disease, especially since age, body mass index and pregnancy contribute significantly. Amaç: Diyabetin sıklıkla arttığı ve bir çok farklı hastalığı tetiklediği bilinen bir gerçektir. Bu nedenle hastalığın erken teşhisi önemlidir. Bu çalışmada plazma glukoz konsantrasyonu, serum insülin direnci ve diyastolik kan basıncı göstergelerinden, makine öğrenmesi yöntemlerine göre hastalığın erken teşhisi öngörülmeye çalışılmıştır. Materyal ve Metot: Çalışmada, bir web sitesinden alınan halka açık veri seti 768 örnek ve dokuz değişkenden oluşmaktadır. Diyabetin erken teşhisinde üç farklı makine öğrenme stratejisi kullanıldı (Destek Vektör Makineleri, Çok Katmanlı Algılayıcılar ve Stokastik Gradyan Artırma). Hiper parametre optimizasyonu için 3 tekrarlı 10 kat tekrarlı çapraz doğrulama yöntemi kullanıldı. Modellerin performansı doğruluk, seçicilik, duyarlılık, karışıklık matrisi, pozitif tahmin değeri (kesinlik), negatif tahmin değeri ve AUC (ROC eğrisi altında kalan alan) temel alınarak değerlendirilmiştir. Bulgular: Deneysel sonuçlara göre (doğruluk (0.79), duyarlılık (0.57), özgüllük (0.91), pozitif tahmin değeri (0.79), negatif tahmin değeri (0.80) ve AUC (0.74) kriterleri), Destek Vektör Makineleri diğer yöntemlere göre daha başarılı çıkmıştır. Sonuç: Diyabet hastalığının erken tanısında plazma glukoz konsantrasyonu, serum insülin direnci ve diastolik kan basinci belirteçleri önemli göstergelerdir. Bu çalışmada da bu belirteçlerin diyabetin erken tanısında önemli katkı sağladığı görülmüştür. Ancak tek başlarına bu göstergelerin hastalığın erken tanısında yeterli olmayacağı özellikle yaş, beden kitle indeksi ve gebeliğin de önemli derecede katkı sağladığı görülmüştür.

This study was designed to determine adequate levels for sodium butyrate inclusion in pre-hatching and pre-starter feed in order to minimize the negative effects of post-hatch delayed placement on broiler chicks. Newly-hatched chicks were allotted in a completely randomized design, with five treatments, each comprising five replicates of ten birds each. Five dietetic levels of sodium butyrate (control, 0.050, 0.075, 0.100 and 0.200%) were used in the pre-starter feed offered to the chicks in the transporting box and during the pre-starter phase. Performance, yolk sac retraction, plasma glucose concentration, weight and histomorphometry of the small intestine were evaluated after 24 hours of feed access and at 7 days of age. A metabolic trial was performed when the chicks were seven to ten days of age. Supplementation of 0.1% sodium butyrate increased the development of broiler chicks’ intestinal villi at seven days of age but was not able to improve performance in the pre-starter phase. Supplementation with sodium butyrate in a diet offered in the transportation box does not improve broiler performance in the pre-starter phase. Supplementation of up to 0.16% is recommended, in order to improve the metabolizability of the ether extract for broilers at seven days of age. RESUMO Objetivou-se, com este estudo, encontrar níveis adequados de inclusão de butirato de sódio em ração pós-eclosão e pré-inicial, buscando minimizar os efeitos negativos do jejum de pintos de corte. Os animais foram distribuídos em delineamento inteiramente ao acaso, com cinco tratamentos e cinco repetições de 10 aves cada. Foram utilizadas cinco suplementações de butirato de sódio (controle; 0,05; 0,075; 0,100 e 0,200%) na ração pré-inicial, fornecidas aos pintos na caixa de transporte e durante a fase pré-inicial. Após 24 horas de acesso ao alimento e aos sete dias de idade, foram avaliados desempenho, retração do saco vitelino, concentração de glicose plasmática, peso e histomorfometria do intestino delgado. De sete a 10 dias de idade, realizou-se ensaio de metabolizabilidade. A suplementação de 0,10% de butirato de sódio aumentou o desenvolvimento das vilosidades intestinais dos pintos de corte aos sete dias de idade, mas não foi capaz de melhorar o desempenho na fase pré-inicial. A suplementação com butirato de sódio em dieta oferecida na caixa de transporte não melhora o desempenho dos frangos na fase pré-inicial. Recomenda-se suplementação de até 0,16%, a fim de melhorar a metabolizabilidade do extrato etéreo para frangos de corte aos sete dias de idade.

Plasma glucose concentration (PGC) and plasma insulin concentration (PIC) are two essential metrics for diabetic regulation, but difficult to be measured directly. Often, PGC and PIC are estimated from continuous glucose monitoring and insulin delivery data. Nevertheless, the inter-individual variability and external disturbance (e.g. carbohydrate intake) bring challenges for accurate estimations. This study is to estimate PGC and PIC adaptively by identifying personalized parameters and external disturbances. An observable glucose-insulin (OGI) dynamic model is established to describe insulin absorption, glucose regulation, and glucose transport. The model parameters and disturbances can be extended to observable state variables and be identified dynamically by Bayesian filtering estimators. Two basic Gaussian noise based Bayesian filtering estimators, extended Kalman filtering (EKF) and unscented Kalman filtering (UKF), are implemented. Recognizing the prevalence of non-Gaussian noise, in this study, two new filtering estimators: particle filtering with Gaussian noise (PFG), and particle filtering with mixed non-Gaussian noise (PFM) are designed and implemented. The proposed OGI model in conjunction with the estimators is evaluated using the data from 30 in-silico subjects and 10 human participants. For in-silico subjects, the OGI with PFM estimator has the ability to estimate PIC and PGC adaptively, achieving RMSE of PIC $ 9.49\pm3.81 $ mU/L, and PGC $ 0.89\pm0.19 $ mmol/L. For human, the OGI with PFM has the promise to identify disturbances ($ 95.46\%\pm0.65\% $ accurate rate of meal identification). OGI model provides a way to fully personalize the parameters and external disturbances in real time, and has potential clinical utility for artificial pancreas.

A novel neutral exopolysaccharide (EPS-III) was isolated from culture broth of Cordyceps militaris (C. militaris). The EPS-III was a homogeneous polysaccharide with Mw of 1.56 × 103 kDa. The yield of EPS-III from culture broth was 123.2 ± 3.1 mg/L and the sugar content was 93.32 ± 0.87%. The backbone of EPS-III was mainly consisted of →4)-α-D-Galp-(1→, while →3, 6)-α-D-Manp-(1→, →4)-α-D-Manp-(1→, →3)-β-D-Galp-(1→ and →3)-α-D-Glcp-(1→ were distributed in the backbone or in the branch chains. The EPS-III had helix structure when dissolved in weak alkaline solution. It also had branched and intertwined form on the surface. The inhibition of α-glucosidase significantly increased as the increase of purity of exopolysaccharides. The EPS-III had effective inhibition on the α-glucosidase with dose-effect relationship. Besides, the results of hypoglycemic activity analysis in vivo indicated that EPS-III can alleviate weight loss, reduce plasma glucose concentration, improve glucose tolerance, protect immune organs and repair dyslipidemia to relieve diabetes in STZ-induced diabetic mice. The manuscript first studied the hypoglycemic activity of exopolysaccharide of by C. militaris, proving and promoting the application value of culture broth. The structure characterization of EPS-III laid experimental foundations on the exploration of structure-activity relationship.

Background: Each measurement is subject to measurement uncertainty (MU). Consequently, each measurement of plasma glucose concentration used for diagnosis and monitoring of diabetes mellitus (DM) is affected. Although concepts and methods of MU are well established in many fields of science and technology, they are presently only incompletely implemented by medical laboratories, neglecting MU of target values of internal quality control (IQC) materials. Methods: An empirical and practical approach for the estimation of MU based on the analysis of routine IQC using control samples with assigned target values is presented. Its feasibility is demonstrated exemplarily by analyzing IQC data from one year obtained for glucose employing the hexokinase method with IQC of two different concentrations. Results: Combined relative extended ( k = 2) MU comprising bias, coefficient of variation (CV), and MU of the target values assigned to control materials were about 9% with a lower (~ 56 mg/dL; ~3.1 mmol/L) and 8% with a higher (~ 346 mg/dL; ~19.2 mmol/L) concentration sample, analyzing IQC of one year from three different devices. Conclusions: Estimation of MU in this study is quite reliable due to the large number of IQC data from one year. The MU of the target values of the commercial control material in this study was considerably larger than other MU contributions, ie, standard deviation and bias. In the future, the contribution of MU of commercial IQC should be addressed more carefully and technologies to measure glucose should be geared toward smaller MU possible, as needed, especially for glucose concentration measurements in diagnosis and management of DM.

Transportation is one of the most stressful procedures that the farm animals encounter and numerous factors can affect an animal’s responses to stress caused by transportation. The aim of the study was to investigate the effects of weaning status (WS: suckling vs weaned) and transport duration (TD: 45-min vs 120-min) on some physiological responses to transportation, and on some behaviours expressed during the 2-h resting period following transportation in Kivircik lambs. All lambs suckled their mothers until 45 days of age. On day 45, a total of 40 Kivircik male lambs were randomly divided into two groups: half of the lambs continued to stay with their dams (Suckling, n = 20), the remaining half (Weaned, n = 20) was separated from their dams, and housed in a different pen. When mean lamb age reached 75 days, lambs from suckling and weaned groups were divided randomly into two subgroups according to TD: 45-min (T45) and 120-min (T120) transport. At the end of transportation, behavioural activities of the lambs were recorded during the 2-h resting period. WS had no significant influence on investigated biochemical and haematological parameters at any sampling time except plasma glucose concentration measured after resting. Glucose is utilized by fish as one of the sources of energy. It is a physiological parameter useful in the determination of the general health condition of cultured fish. The aim of this study was to determine the stability of plasma and blood glucose concentration, stored at different temperatures (4 °C and 30 °C) for 72 h. Two (2) milliliters of blood was collected via the caudal abdominal venipuncture of 10 female catfishes purchased. The fish were 8 months of age and were kept in plastic ponds at the Faculty of Veterinary Medicine, University of Nigeria. Blood was collected in blood tubes containing K2EDTA. Plasma was harvested from 1 mL of K2EDTA blood sample following centrifugation. Glucose concentration was determined using the Accu-Chek glucometer. Whole blood and plasma were analyzed immediately (0 h), and then after storage at 4 °C and 30 °C at 24 h, 48 h, and 72 h. Comparisons to the whole blood samples (0 h) stored at 30 °C revealed a significant (p 0.05) differences in the 24-h, 48-h, and 72-h plasma glucose concentration were observed. Therefore, whole blood and plasma stored at 4 °C for 72 h and plasma stored at 30 °C for 24 h can be used for glucose determinations in the female African catfish.

Aim: The aim of this study was to gain insight into the pathophysiological significance of elevated plasma glucose concentrations (mmol/L) 60 min post oral glucose load in apparently healthy individuals. Methods: Comparison of resistance to insulin action and associated cardio-metabolic risk factors in 490 apparently healthy persons, subdivided into those with a plasma glucose concentration 60 min following a 75-g oral glucose challenge of Results: Insulin resistance was significantly greater in persons with normal glucose tolerance whose 60-min glucose concentration was ⩾8.6, associated with higher blood pressure, plasma concentrations of glucose, insulin, triglyceride and lower high-density lipoprotein cholesterol concentrations. Insulin resistance (IR) is a cardinal feature of type 2 diabetes mellitus (T2DM). It also is associated with multiple metabolic abnormalities which are known cardiovascular disease (CVD) risk factors. Thus, IR not only contributes to the development of hyperglycemia in T2DM patients, but also to the elevated CVD risk. Improving insulin sensitivity is anticipated to both lower the plasma glucose concentration and decrease CVD risk in T2DM patients, independent of glucose control. We review the molecular mechanisms and metabolic consequences of IR in T2DM patients and discuss the importance of addressing IR in the management of T2DM.

The aim of this study was to examine the metabolic response to feed deprivation up to 48 h in low and high yielding lamb genotypes. It was hypothesised that Terminal sired lambs would have decreased plasma glucose and increased plasma non-esterified fatty acids (NEFA) and β-hydroxybutyrate (BHOB) concentrations in response to feed deprivation compared to Merino sired lambs. In addition, it was hypothesised that the metabolic changes due to feed deprivation would also be greater in progeny of sires with breeding values for greater growth, muscling and leanness. Eighty nine lambs (45 ewes, 44 wethers) from Merino dams with Merino or Terminal sires with a range in Australian Sheep Breeding Values (ASBVs) for post-weaning weight (PWT), post-weaning eye muscle depth and post-weaning fat depth (PFAT) were used in this experiment. Blood samples were collected via jugular cannulas every 6 h from time 0 to 48 h of feed deprivation for the determination of plasma glucose, NEFA, BHOB and lactate concentration. From 12 to 48 h of feed deprivation plasma glucose concentration decreased (Plt; 0.05) by 25% from 4.04 ± 0.032 mmol/l to 3.04 ± 0.032 mmol/l. From 6 h NEFA concentration increased (Plt; 0.05) from 0.15 ± 0.021 mmol/l by almost 10-fold to 1.34 ± 0.021 mmol/l at 48 h of feed deprivation. Feed deprivation also influenced BHOB concentrations and from 12 to 48 h it increased (Plt; 0.05) from 0.15 ± 0.010 mmol/l to 0.52 ± 0.010 mmol/l. Merino sired lambs had a 8% greater reduction in glucose and 29% and 10% higher NEFA and BHOB response, respectively, compared to Terminal sired lambs (Plt; 0.05). In Merino sired lambs, increasing PWT was also associated with an increase in glucose and decline in NEFA and BHOB concentration (Plt; 0.05). In Terminal sired lambs, increasing PFAT was associated with an increase in glucose and decline in NEFA concentration (Plt; 0.05). Contrary to the hypothesis, Merino sired lambs showed the greatest metabolic response to fasting especially in regards to fat metabolism.

Hydroxylated polybrominated diphenyl ethers (OH-PBDEs), naturally produced by algae and cyanobacteria in the Baltic Sea, are potent disrupters of energy metabolism as well as endocrine disruptors and neurotoxins. In this study, European perch (Perca fluviatilis) from the Baltic Sea were sampled from May until October. OH-PBDEs and ten biomarkers were measured in each individual (n = 84 over 18 sampling time points) to study potential correlations between exposure to OH-PBDEs and changes in biomarkers. Several biomarkers showed significant non-linear seasonal variation. In the perch, ethoxyresorufin-O-deethylase (EROD) activity, plasma lactate concentration, and plasma glucose concentration showed a significant positive log-linear correlation with OH-PBDEs, whereas lipid percentage and liver somatic index showed a significant negative log-linear correlation with OH-PBDEs. These results strengthen the concern that OH-PBDEs could cause negative health effects for fish in the Baltic Sea.

This study aimed to evaluate the performance and metabolic changes in dairy calves supplemented with lysine and methionine in milk replacer (MR) or starter concentrate (SC). Male Holstein calves (n = 45) were blocked and distributed in Control without supplementation (1) and, Lysine and Methionine supplementation to achieve an intake of 17 and 5.3 g/d in the SC (2) and to achieve of 17 and 5.3 g/d in the MR (3). MR was fed (6 L/d) until the 8th week of life when weaning occurred. Calves were followed until the 10th week of age. Feed intake was measured daily. Weight and body measurements were registered weekly. Blood samples were collected biweekly to evaluate the intermediate metabolism. The AA supplementation resulted in lower body weight at weaning and week 10. Calves fed SC Lys:Met had lower SC intake and lower total feed intake at weaning when compared to control. Calves fed control had higher heart girth, hip-width, and plasma glucose concentration. The supplementation with Lys and Met did not benefit dairy calves&rsquo, performance nor metabolism in this study. Supplementation through the MR was more efficient than SC to result in adequate daily intakes of AA. Further studies are needed to understand the negative effects of AA on calf starter intake.

The correct volume of sample and time of storing prior to the analysis are important considerations in the estimation of plasma glucose concentration of patients. The present study was to determine the effect of sample volume variation and time delay in the analysis of plasma glucose results in healthy adults. A total of 30 individuals aged between 20 and 30 years were selected for the study. Blood samples were collected into three fluoride-oxalate collection tubes separately. The results revealed that the sample volume variation from 2.0 mL fluoride-oxalate tube to 1.0 mL and 3.0 mL did not significantly affect the plasma glucose concentration ( p > 0.05). However, the plasma glucose concentration in the sample significantly decreased upon delaying the analysis. The mean fasting plasma glucose concentration of analysis after one hour of collection and analysis after three hours of collection was not significantly different ( p > 0.05). The mean fasting plasma glucose concentrations between one hour and five hours timepoints after collection ( p p = 0.014) were significantly different. In conclusion, overfilling and underfilling (2.0 ± 1.0 mL) of fluoride-oxalate tubes did not affect the plasma glucose results significantly. If the samples are analyzed within three hours of collection, the time dependent change too is not statistically significant.

Objective Post-transplantation diabetes mellitus (PTDM) is a frequent complication after heart transplantation. We investigated the specific predictors of PTDM in Chinese heart transplant recipients and the prognostic value of these predictors. Methods We retrospectively analyzed 122 adult patients who underwent heart transplantation. Comparisons were made between patients with PTDM (n = 44) and those without PTDM (n = 78). Results During the median follow-up of 44 months, the cumulative incidence of PTDM was 19.7% at 1 year after transplantation and 36.1% at the endpoint. PTDM was associated with a significantly higher preoperative body mass index (BMI) (odds ratio [OR] = 1.349), fasting plasma glucose (FPG) concentration (OR = 2.538), and serum uric acid concentration (OR = 1.005) after transplantation. The area under the receiver operating characteristic curve was 0.708 and 0.763 for the BMI and FPG concentration, respectively. The incidence of acute rejection and infection were higher and the all-cause mortality rate was considerably greater in patients with than without PTDM. Conclusions A higher preoperative BMI (>23 kg/m 2 ), FPG concentration (>5.2 mmol/L), and uric acid concentration could potentially predict PTDM in Chinese heart transplant recipients. PTDM influences long-term survival after heart transplantation.
