Effector secretion is crucial for root endophytes to establish and protect their ecological niche. We used time-resolved transcriptomics to monitor effector gene expression dynamics in two closely related Sebacinales, Serendipita indica and Serendipita vermifera, during symbiosis with three plant species, competition with the phytopathogenic fungus Bipolaris sorokiniana, and cooperation with root-associated bacteria. We observed increased effector gene expression in response to biotic interactions, particularly with plants, indicating their importance in host colonization. Some effectors responded to both plants and microbes, suggesting dual roles in intermicrobial competition and plant-microbe interactions. A subset of putative antimicrobial effectors, including a GH18-CBM5 chitinase, was induced exclusively by microbes. Functional analyses of this chitinase revealed its antimicrobial and plant-protective properties. We conclude that dynamic effector gene expression underpins the ability of Sebacinales to thrive in diverse ecological niches with a single fungal chitinase contributing substantially to niche defense.

Bacterial biofilm formation and attachment to hosts are mediated by carbohydrate-binding lectins, exopolysaccharides, and their interactions in the extracellular matrix (ECM). During tomato infection Ralstonia pseudosolanacearum (Rps) GMI1000 highly expresses three lectins: LecM, LecF, and LecX. The latter two are uncharacterized. We evaluated the roles in bacterial wilt disease of LecF, a fucose-binding lectin, LecX, a xylose-binding lectin, and the Rps exopolysaccharide EPS I. Interestingly, single and double lectin mutants attached to tomato roots better and formed more biofilm under static conditions in vitro. Consistent with this finding, static bacterial aggregation was suppressed by heterologous expression of lecFGMI1000 and lecXGMI1000 in other Ralstonia strains that naturally lack these lectins. Crude ECM from a ΔlecF/X double mutant was more adhesive than the wild-type ECM, and LecF and LecX increased Rps attachment to ECM. The enhanced adhesiveness of the ΔlecF/X ECM could explain the double mutants hyper-attachment in static conditions. Unexpectedly, mutating lectins decreased Rps attachment and biofilm viscosity under shear stress, which this pathogen experiences in plant xylem. LecF, LecX, and EPS I were all essential for biofilm development in xylem fluid flowing through cellulose-coated microfluidic channels. These results suggest that under shear stress, LecF and LecX increase Rps attachment by interacting with the ECM and plant cell wall components like cellulose. In static conditions such as on root surfaces and in clogged xylem vessels, the same lectins suppress attachment to facilitate pathogen dispersal. Thus, Rps lectins have a dual biological function that depends on the physical environment.

Plants produce reactive oxygen species (ROS) upon infection, which typically trigger defence mechanisms and impede pathogen proliferation. Root-knot nematodes (RKNs, Meloidogyne spp.) represent highly detrimental pathogens capable of parasitizing a broad spectrum of crops, resulting in substantial annual agricultural losses. The involvement of ROS in RKN parasitism is well acknowledged. In this study, we identified a novel effector from Meloidogyne incognita, named CATLe, that contains a conserved catalase domain, exhibiting potential functions in regulating host ROS levels. Phylogenetic analysis revealed that CATLe is conserved across RKNs. Temporal and spatial expression assays showed that the CATLe gene was specifically up-regulated at the early infection stages and accumulated in the subventral oesophageal gland cells of M. incognita. Immunolocalization demonstrated that CATLe was secreted into the giant cells of the host plant during M. incognita parasitism. Transient expression of CATLe significantly dampened the flg22-induced ROS production in Nicotiana benthamiana. In planta assays confirmed that M. incognita can exploit CATLe to manipulate host ROS levels by directly degrading H2O2. Additionally, interfering with expression of the CATLe gene through double-stranded RNA soaking and host-induced gene silencing significantly attenuated M. incognita parasitism, highlighting the important role of CATLe. Taken together, our results suggest that RKNs can directly degrade ROS products using a functional catalase, thereby manipulating host ROS levels and facilitating parasitism.

Plants release a wealth of metabolites into the rhizosphere that can shape the composition and activity of microbial communities in response to environmental stress. The connection between rhizodeposition and rhizosphere microbiome succession has been suggested, particularly under environmental stress conditions, yet definitive evidence is scarce. In this study, we investigated the relationship between rhizosphere chemistry, microbiome dynamics, and abiotic stress in the bioenergy crop switchgrass grown in a marginal soil under nutrient-limited, moisture-limited, and nitrogen (N)-replete, phosphorus (P)-replete, and NP-replete conditions. We combined 16S rRNA amplicon sequencing and LC-MS/MS-based metabolomics to link rhizosphere microbial communities and metabolites. We identified significant changes in rhizosphere metabolite profiles in response to abiotic stress and linked them to changes in microbial communities using network analysis. N-limitation amplified the abundance of aromatic acids, pentoses, and their derivatives in the rhizosphere, and their enhanced availability was linked to the abundance of bacterial lineages from Acidobacteria, Verrucomicrobia, Planctomycetes, and Alphaproteobacteria. Conversely, N-amended conditions increased the availability of N-rich rhizosphere compounds, which coincided with proliferation of Actinobacteria. Treatments with contrasting N availability differed greatly in the abundance of potential keystone metabolites; serotonin and ectoine were particularly abundant in N-replete soils, while chlorogenic, cinnamic, and glucuronic acids were enriched in N-limited soils. Serotonin, the keystone metabolite we identified with the largest number of links to microbial taxa, significantly affected root architecture and growth of rhizosphere microorganisms, highlighting its potential to shape microbial community and mediate rhizosphere plant-microbe interactions.

BACKGROUND: Sclerotinia spp. are generalist fungal pathogens, infecting over 700 plant hosts worldwide, including major crops. While host resistance is the most sustainable and cost-effective method for disease management, complete resistance to Sclerotinia diseases is rare. We recently identified soft basal stem as a potential susceptibility factor to Sclerotinia minor infection in lettuce (Lactuca sativa) under greenhouse conditions. RESULTS: Analysis of stem and root cell wall composition in five L. sativa and one L. serriola accessions with varying growth habits and S. minor resistance levels revealed strong association between hemicellulose constituents, lignin polymers, disease phenotypes, and basal stem mechanical strength. Accessions resistant to basal stem degradation consistently exhibited higher levels of syringyl, guaiacyl, and xylose, but lower levels of fucose in stems. These findings suggest that stem cell wall polymers recalcitrant to breakdown by lignocellulolytic enzymes may contribute to stem strength-mediated resistance against S. minor. CONCLUSIONS: The lignin content, particularly guaiacyl and syringyl, along with xylose could potentially serve as biomarkers for identifying more resistant lettuce accessions and breeding lines. Basal stem degradation by S. minor was influenced by localized microenvironment conditions around the stem base of the plants.

Most of Earths trees rely on critical soil nutrients that ectomycorrhizal fungi (EcMF) liberate and provide, and all of Earths land plants associate with bacteria that help them survive in nature. Yet, our understanding of how the presence of EcMF modifies soil bacterial communities, soil food webs, and root chemistry requires direct experimental evidence to comprehend the effects that EcMF may generate in the belowground plant microbiome. To this end, we grew Pinus muricata plants in soils that were either inoculated with EcMF and native forest bacterial communities or only native bacterial communities. We then profiled the soil bacterial communities, applied metabolomics and lipidomics, and linked omics data sets to understand how the presence of EcMF modifies belowground biogeochemistry, bacterial community structure, and their functional potential. We found that the presence of EcMF (i) enriches soil bacteria linked to enhanced plant growth in nature, (ii) alters the quantity and composition of lipid and non-lipid soil metabolites, and (iii) modifies plant root chemistry toward pathogen suppression, enzymatic conservation, and reactive oxygen species scavenging. Using this multi-omic approach, we therefore show that this widespread fungal symbiosis may be a common factor for structuring soil food webs.IMPORTANCEUnderstanding how soil microbes interact with one another and their host plant will help us combat the negative effects that climate change has on terrestrial ecosystems. Unfortunately, we lack a clear understanding of how the presence of ectomycorrhizal fungi (EcMF)-one of the most dominant soil microbial groups on Earth-shapes belowground organic resources and the composition of bacterial communities. To address this knowledge gap, we profiled lipid and non-lipid metabolites in soils and plant roots, characterized soil bacterial communities, and compared soils amended either with or without EcMF. Our results show that the presence of EcMF changes soil organic resource availability, impacts the proliferation of different bacterial communities (in terms of both type and potential function), and primes plant root chemistry for pathogen suppression and energy conservation. Our findings therefore provide much-needed insight into how two of the most dominant soil microbial groups interact with one another and with their host plant.

Flatbed scanners are commonly used for root analysis, but typical manual segmentation methods are time-consuming and prone to errors, especially in large-scale, multi-plant studies. Furthermore, the complex nature of root structures combined with noisy backgrounds in images complicates automated analysis. Addressing these challenges, this article introduces RhizoNet, a deep learning-based workflow to semantically segment plant root scans. Utilizing a sophisticated Residual U-Net architecture, RhizoNet enhances prediction accuracy and employs a convex hull operation for delineation of the primary root component. Its main objective is to accurately segment root biomass and monitor its growth over time. RhizoNet processes color scans of plants grown in a hydroponic system known as EcoFAB, subjected to specific nutritional treatments. The root detection model using RhizoNet demonstrates strong generalization in the validation tests of all experiments despite variable treatments. The main contributions are the standardization of root segmentation and phenotyping, systematic and accelerated analysis of thousands of images, significantly aiding in the precise assessment of root growth dynamics under varying plant conditions, and offering a path toward self-driving labs.

Arbuscular mycorrhizal fungi (AMF) transport substantial plant carbon (C) that serves as a substrate for soil organisms, a precursor of soil organic matter (SOM), and a driver of soil microbial dynamics. Using two-chamber microcosms where an air gap isolated AMF from roots, we 13CO2-labeled Avena barbata for 6 wk and measured the C Rhizophagus intraradices transferred to SOM and hyphosphere microorganisms. NanoSIMS imaging revealed hyphae and roots had similar 13C enrichment. SOM density fractionation, 13C NMR, and IRMS showed AMF transferred 0.77 mg C g-1 of soil (increasing total C by 2% relative to non-mycorrhizal controls); 33% was found in occluded or mineral-associated pools. In the AMF hyphosphere, there was no overall change in community diversity but 36 bacterial ASVs significantly changed in relative abundance. With stable isotope probing (SIP)-enabled shotgun sequencing, we found taxa from the Solibacterales, Sphingobacteriales, Myxococcales, and Nitrososphaerales (ammonium oxidizing archaea) were highly enriched in AMF-imported 13C (> 20 atom%). Mapping sequences from 13C-SIP metagenomes to total ASVs showed at least 92 bacteria and archaea were significantly 13C-enriched. Our results illustrate the quantitative and ecological impact of hyphal C transport on the formation of potentially protective SOM pools and microbial roles in the AMF hyphosphere soil food web.

Tremendous plant metabolic diversity arises from phylogenetically restricted specialized metabolic pathways. Specialized metabolites are synthesized in dedicated cells or tissues, with pathway genes sometimes colocalizing in biosynthetic gene clusters (BGCs). However, the mechanisms by which spatial expression patterns arise and the role of BGCs in pathway evolution remain underappreciated. In this study, we investigated the mechanisms driving acylsugar evolution in the Solanaceae. Previously thought to be restricted to glandular trichomes, acylsugars were recently found in cultivated tomato roots. We demonstrated that acylsugars in cultivated tomato roots and trichomes have different sugar cores, identified root-enriched paralogs of trichome acylsugar pathway genes, and characterized a key paralog required for root acylsugar biosynthesis, SlASAT1-LIKE (SlASAT1-L), which is nested within a previously reported trichome acylsugar BGC. Last, we provided evidence that ASAT1-L arose through duplication of its paralog, ASAT1, and was trichome-expressed before acquiring root-specific expression in the Solanum genus. Our results illuminate the genomic context and molecular mechanisms underpinning metabolic diversity in plants.

Sorghum bicolor is among the most important cereals globally and a staple crop for smallholder farmers in sub-Saharan Africa. Approximately 20% of sorghum yield is lost annually in Africa due to infestation with the root parasitic weed Striga hermonthica. Existing Striga management strategies are not singularly effective and integrated approaches are needed. Here, we demonstrate the functional potential of the soil microbiome to suppress Striga infection in sorghum. We associate this suppression with microbiome-mediated induction of root endodermal suberization and aerenchyma formation and with depletion of haustorium-inducing factors, compounds required for the initial stages of Striga infection. We further identify specific bacterial taxa that trigger the observed Striga-suppressive traits. Collectively, our study describes the importance of the soil microbiome in the early stages of root infection by Striga and pinpoints mechanisms of Striga suppression. These findings open avenues to broaden the effectiveness of integrated Striga management practices.

BACKGROUND: Sulfur (S) is a mineral nutrient essential for plant growth and development, which is incorporated into diverse molecules fundamental for primary and secondary metabolism, plant defense, signaling, and maintaining cellular homeostasis. Although, S starvation response is well documented in the dicot model Arabidopsis thaliana, it is not clear if the same transcriptional networks control the response also in the monocots. RESULTS: We performed series of physiological, expression, and metabolite analyses in two model monocot species, one representing the C3 plants, Oryza sativa cv. kitaake, and second representing the C4 plants, Setaria viridis. Our comprehensive transcriptomic analysis revealed twice as many differentially expressed genes (DEGs) in S. viridis than in O. sativa under S-deficiency, consistent with a greater loss of sulfur and S-containing metabolites under these conditions. Surprisingly, most of the DEGs and enriched gene ontology terms were species-specific, with an intersect of only 58 common DEGs. The transcriptional networks were different in roots and shoots of both species, in particular no genes were down-regulated by S-deficiency in the roots of both species. CONCLUSIONS: Our analysis shows that S-deficiency seems to have different physiological consequences in the two monocot species and their nutrient homeostasis might be under distinct control mechanisms.

Soil microbiomes are highly diverse, and to improve their representation in biogeochemical models, microbial genome data can be leveraged to infer key functional traits. By integrating genome-inferred traits into a theory-based hierarchical framework, emergent behaviour arising from interactions of individual traits can be predicted. Here we combine theory-driven predictions of substrate uptake kinetics with a genome-informed trait-based dynamic energy budget model to predict emergent life-history traits and trade-offs in soil bacteria. When applied to a plant microbiome system, the model accurately predicted distinct substrate-acquisition strategies that aligned with observations, uncovering resource-dependent trade-offs between microbial growth rate and efficiency. For instance, inherently slower-growing microorganisms, favoured by organic acid exudation at later plant growth stages, exhibited enhanced carbon use efficiency (yield) without sacrificing growth rate (power). This insight has implications for retaining plant root-derived carbon in soils and highlights the power of data-driven, trait-based approaches for improving microbial representation in biogeochemical models.

UNLABELLED: Diazotrophic bacteria can reduce atmospheric nitrogen into ammonia enabling bioavailability of the essential element. Many diazotrophs closely associate with plant roots increasing nitrogen availability, acting as plant growth promoters. These associations have the potential to reduce the need for costly synthetic fertilizers if they could be engineered for agricultural applications. However, despite the importance of diazotrophic bacteria, genetic tools are poorly developed in a limited number of species, in turn narrowing the crops and root microbiomes that can be targeted. Here, we report optimized protocols and plasmids to manipulate phylogenetically diverse diazotrophs with the goal of enabling synthetic biology and genetic engineering. Three broad-host-range plasmids can be used across multiple diazotrophs, with the identification of one specific plasmid (containing origin of replication RK2 and a kanamycin resistance marker) showing the highest degree of compatibility across bacteria tested. We then demonstrated modular expression by testing seven promoters and eleven ribosomal binding sites using proxy fluorescent proteins. Finally, we tested four small molecule inducible systems to report expression in three diazotrophs and demonstrated genome editing in Klebsiella michiganensis M5al. ONE-SENTENCE SUMMARY: In this study, broad-host plasmids and synthetic genetic parts were leveraged to enable expression tools in a library of diazotrophic bacteria.

Plant roots integrate environmental signals with development using exquisite spatiotemporal control. This is apparent in the deposition of suberin, an apoplastic diffusion barrier, which regulates flow of water, solutes and gases, and is environmentally plastic. Suberin is considered a hallmark of endodermal differentiation but is absent in the tomato endodermis. Instead, suberin is present in the exodermis, a cell type that is absent in the model organism Arabidopsis thaliana. Here we demonstrate that the suberin regulatory network has the same parts driving suberin production in the tomato exodermis and the Arabidopsis endodermis. Despite this co-option of network components, the network has undergone rewiring to drive distinct spatial expression and with distinct contributions of specific genes. Functional genetic analyses of the tomato MYB92 transcription factor and ASFT enzyme demonstrate the importance of exodermal suberin for a plant water-deficit response and that the exodermal barrier serves an equivalent function to that of the endodermis and can act in its place.

ABSTRACT Drought limits crop performance worldwide. Plant roots' ability to grow toward moisture, termed hydrotropism, is considered one strategy for optimizing water recruitment from the growth medium. Based on the sequence of the hydrotropism‐indispensable MIZ1 protein in Arabidopsis thaliana, we identify hydrotropism and drought‐responsive genes in tomato. We utilized CRISPR/Cas9 genome‐editing technology for targeted mutagenesis of three hydrotropism‐associated loci (MIZ1‐like) in tomato (Solanum lycopersicum). We show that the three tomato MIZ1‐like genes are drought‐responsive and two of them are hydrostimulation‐responsive. Examination of the root hydrotropic response of triple and double mutants indicated the gene SlMIZ1‐1 as indispensable for tomato root hydrotropism. Moreover, expression of the SlMIZ1‐1 gene in the Arabidopsis miz1 mutant effectively complemented the lost MIZ1 functionality, including root hydrotropic bending and generation of hydrotropic Ca2+ signals. Transcriptome analysis of hydrostimulated tomato root tips under control gravity and continuous clinorotation conditions was performed to identify gravitropism‐ and hydrotropism‐responsive genes. This analysis suggested the involvement of ethylene and ABA signalling in modulating the interplay between hydrotropism and gravitropism. Unveiling the molecular mechanisms that underlie hydrotropism and drought response holds great potential for improving crop performance under limiting water availability due to global climate changes. Despite decades of research, a comprehensive understanding of trait coordination at the whole‐plant level remains elusive. Furthermore, while the link between above‐ground growth rates and leaf traits related to nutrient use is well established, much less is known about the below‐ground compartment. Herbs and woody species exhibit distinct above‐ground growth and nutrient‐use strategies, but whether these differences extend to root traits below‐ground remains unclear. We carried out a common garden experiment with 23 perennials (7 herbs and 16 woody species) coexisting in a Mediterranean shrubland and measured 17 above‐ground and below‐ground traits related to growth, nutrient use and size. We analysed the links between growth rate and nutrient use focusing especially on roots and considering potential differences between herbs and woody species. We also combined plant size data from experimental juveniles and field‐sampled adults to determine the effects of life stage on whole‐plant phenotypic integration. We found a significant relationship between growth rates (both above‐ground and below‐ground) and root nutrient‐use strategies. Root diameter was negatively associated to growth rate only in herbs. Specific root area and root tissue density were positively and negatively correlated with growth rate, respectively, in both herbs and woody species. Moreover, we found significant differences in roots traits between herbs and woody species. Plant growth rate and root nutrient‐use strategies were both positively associated to plant size (height and above‐ground diameter) in juveniles, while a negative relationship was observed in adults. Our work provides insights on the links between whole‐plant growth rate and nutrient‐use strategies in species from a dry Mediterranan shrubland. Specifically, we show that whole‐plant growth rate is strongly associated to root traits, with more acquisitive root nutrient‐use strategies related to faster growth rates, which in turn translated to greater plant size in juveniles but smaller in adults. Our results also highlight root functional differences and contrasting patterns of whole‐plant phenotypic integration between herbs and woody species, offering deeper insights into species coexistence in species‐rich dry Mediterranean environments. Abstract\nNOVELTY STATEMENTA hydroponic experiment was conducted to investigate the variations in membrane permeabilities, chlorophyll contents, antioxidase activities, the ascorbic acid (AsA)-glutathione (GSH) cycle, and the glyoxalase system in the leaves of Pontederia cordata with 0 ∼ 15.0 mg L−1 lead ion (Pb2+) exposure. The concentrations of Pb2+ accumulated in the plant roots, stems, and leaves were also evaluated. After 7 days of exposure, the plants maintained normal growth, and there was a significant increase in ascorbate peroxidase and dehydroascorbate reductase activities. With 5.0 mg L−1 Pb2+ exposure for 28 days, nearly 66.36% of Pb2+ accumulated in the roots, while excess Pb2+ immobilized in the leaves was not observed. Exposure to 10.0 and 15.0 mg L−1 Pb2+ for 28 days significantly increased Pb2+ contents in the leaves. This led to decrease in chlorophyll a, b, and carotenoid contents, and to increase in the methylglyoxal content in the leaves. With 10 and 15 mg L−1 Pb2+ exposure, NPT and PCs contents in leaves increased. however, the glyoxalase system did not function well in the plant tolerant to Pb2+ at higher concentrations. The AsA-GSH cycle did not cooperate with the glyoxalase system in the plant defense against Pb2+ exposure in the present investigation.The role of the ascorbic acid (AsA)-glutathione (GSH) cycle and the glyoxalase system separately has been reported in numerous studies. however, the synergistic role of both in plants’ defense against heavy metal exposure was discussed in our present study. Purpose: It is a central goal in current ecology to investigate the effects of global changes on ecosystem productivity, which includes above- and below-ground plant productivity as well as soil microbial productivity, but most studies focus on aboveground plant productivity. As plant and microbial communities have different attributes, it is intuitive to hypothesize that belowground plant root productivity will respond to global changes more similarly as aboveground stem/leaf productivity than soil microbial productivity; however, this hypothesis remains largely unexplored. Methods: Here we conducted a long-term nitrogen deposition experiment in the Eurasian steppe, manipulating nine rates (0–50 g N m− 2 yr− 1) at two frequencies under two management strategies (fencing or mowing). Results: Belowground plant productivity was found to respond similarly as soil microbial productivity rather than aboveground plant productivity, contrary to the hypothesis. And this pattern was more obvious under mowing than fencing, because mowing decreased soil water content and caused another pressure beyond the decreased pH induced by N addition. Conclusions: Overall, our results demonstrated the importance of microhabitat (below- or above-ground) relative to community attribute (plants or microorganisms) in determining productivity response to nitrogen deposition, emphasizing the necessity to integratively study the response of both above- and below-ground productivities to global changes. Background In epiphytic bromeliads, the roots were previously considered to be poorly functional organs in the processes of absorption and metabolization of water and nutrients, while the leaves were considered to always act as protagonists in both functions. More recent discoveries have been changing this old view of the root system. Scope In this review, we address previous ideas regarding the function performed by the roots of epiphytic bromeliads (mere holdfast structures with low physiological activity) and the importance of a reduced or lack of a root system for the emergence of epiphytism. We present indirect and direct evidence that contradicts this older hypothesis. Furthermore, the importance of the root absorptive function mainly for juvenile tankless epiphytic bromeliads and the characteristics of the root absorption process of adult epiphytic tank bromeliads are discussed thoroughly from a physiological perspective. Finally, some factors (species, substrate, environmental conditions) that influence the absorptive capability of the roots of epiphytic tank bromeliads are also be considered, highlighting the importance that the absorptive role of the roots has for the plasticity of bromeliads that live on trees, which is an environment characterized by intermittent availability of water and nutrients. Conclusions The roots of tank-forming epiphytic bromeliads play important roles in the absorption and metabolization of nutrients and water. The importance of roots is greatest for juvenile tankless bromeliads since the root is the main absorptive organ. In larger plants with a tank, although the leaves become the protagonists in the resource acquisition process, the roots complement the absorptive function of the leaf trichomes, resulting in improved growth of these bromeliad. The physiological and biochemical properties of the processes of absorption and distribution of resources in the tissues appear to differ between absorption by trichomes and roots. Shrubs are usually adapted to stressful environments in which soil resources are limited, and thus, roots are fundamental for their biological success. However, root measures are challenging to collect, especially in field conditions and at the individual level. For this study, we collected data on the three‐dimensional distribution of fine root biomass of twenty‐three individuals belonging to three shrub species in a mediterranean shrubland in central Spain: gum rockrose Cistus ladanifer, rosemary Salvia rosmarinus, and hairy‐fruited broom Cytisus striatus. Our goal was to determine the soil‐foraging strategies adopted by the plants. We hypothesized that plants would show stabilizing niche differences explaining the high plant biodiversity characteristic of mediterranean shrublands and that they would follow the game theory model's prediction of exploitative segregation of roots behaving territorially but also over‐proliferating roots close to their stem and engaging in a root tragedy of the commons. We found that two‐thirds to three‐fourths of the biomass was belowground, and the system's productivity was roughly 500–600 g C m−2 year−1. Only rosemary plants competed with neighbors following the exploitative segregation predictions. Broom plants had the shallowest and most widespread root systems but significantly reduced their root range toward competing neighbors. Gum rockrose presented deep, narrow root systems avoiding extensive overlap with neighbors but did not appear to respond to competitive pressure levels. Exogenous nitrate ingestion can improve exercise performance. This study investigated whether an isotonic beetroot drink could improve jump and sprint performance in active individuals. Twenty-three physically active participants (17 males, 6 females) (mean ± SD; age: 26 ± 4 years; body mass index: 22.4 ± 1.9 kg/m2) completed a double-blind, randomized, cross-over study where they consumed 570mL of either beetroot juice drink (BR) or isotonic beetroot juice drink (ISO-BR) three hours before performing countermovement (CMJ) and standing broad jump (SBJ) tests and a 30-second all-out sprint on a cycle ergometer. Both drinks contained equal nitrate (12.9 mmol) and carbohydrate (6.1g per 100mL) content but differed in osmolality (BR: 420 mOsm/kg vs. ISO-BR: 315 mOsm/kg). Salivary total nitrate and nitrite concentrations (NOx) 3 hours post-ingestion were significantly higher after consuming ISO-BR than BR. ISO-BR significantly improved peak power output compared to BR by 3.9% (ISO-BR: 11.4 ± 2.5 W/kg vs. BR: 11.0 ± 2.3 W/kg, p =.04) but not time to peak power (ISO-BR: 2.8 ± 1.7 s vs. BR: 2.9 ± 1.6 s, p =.62) or mean power output (ISO-BR: 7.3 ± 1.5 W/kg vs. BR: 7.3 ± 1.5 W/kg, p =.37). There were no significant differences in CMJ or SBJ between trials (p >.05). Sensory evaluation indicated that ISO-BR was preferred by 91% (n = 21) of participants compared with BR (average score; ISO-BR: 5.52 vs. BR: 3.52, p <.05). Arabidopsis thaliana synthesizes various medicinal compounds, and serves as a model plant for medicinal plant research. Single-cell transcriptomics technologies are essential for understanding the developmental trajectory of plant roots, facilitating the analysis of synthesis and accumulation patterns of medicinal compounds in different cell subpopulations. Although methods for interpreting single-cell transcriptomics data are rapidly advancing in Arabidopsis, challenges remain in precisely annotating cell identity due to the lack of marker genes for certain cell types. In this work, we trained a machine learning system, AtML, using sequencing datasets from six cell subpopulations, comprising a total of 6000 cells, to predict Arabidopsis root cell stages and identify biomarkers through complete model interpretability. Performance testing using an external dataset revealed that AtML achieved 96.50% accuracy and 96.51% recall. Through the interpretability provided by AtML, our model identified 160 important marker genes, contributing to the understanding of cell type annotations. In conclusion, we trained AtML to efficiently identify Arabidopsis root cell stages, providing a new tool for elucidating the mechanisms of medicinal compound accumulation in Arabidopsis roots. Three new prenylated C6–C3 compounds (1–3), together with two known prenylated C6–C3 compounds (4–5) and one known C6–C3 derivative (6), were isolated from the roots of Illicium brevistylum A. C. Smith. The structures of 1–3 were elucidated by spectroscopic methods including 1D and 2D NMR, HRESIMS, CD experiments and ECD calculations. The structure of illibrefunone A (1) was confirmed by single-crystal X-ray diffraction analysis. All compounds were evaluated in terms of their anti-inflammatory potential on nitric oxide (NO) generation in lipopolysaccharide-stimulated murine RAW264.7 macrophages and murine BV2 microglial cells, antiviral activity against Coxsackievirus B3 (CVB3) and influenza virus A/Hanfang/359/95 (H3N2). Compounds 3 and 4 exhibited potent inhibitory effects on the production of NO in RAW 264.7 cells with IC50 values of 20.57 and 12.87 μM respectively, which were greater than those of dexamethasone (positive control). The demand for food is increasing and the use of soil organic amendments in agricultural management practices has been instructed to increase crop yield and reduce dependence on synthetic inorganic fertilizers at low cost to limited resource farmers. However, the effect of organic amendments on the quality and nutritional composition of edible plants has received little attention. Locally available organic amendments (sewage sludge SS, chicken manure CM, cow manure Cow, vermicompost Vermi, and biochar Bio) were chosen to test their impact on field-grown sweet potato, Ipomoea batatas L. yield, root quality, and root nutritional composition. The results indicated that utilizing Cow manure in growing sweet potatoes significantly promoted root yield and root nutritional composition. Cow treatment produced the greatest number of roots compared to Bio, CM, SS, and the control treatments. The results also revealed that the concentrations of vitamin C (260. 3 µg g−1), β-carotene (45.4 µg g−1), soluble sugars (16.7 mg g−1), and total phenols (196.3 3 µg g−1 fresh roots) were greater in the roots of plants grown in Cow compared to the roots of the control treatment. Phenolic allelochemicals from root exudates dominate rhizosphere formation, lead to autotoxicity in plants subjected to continuous monoculture (CM) stress and induce the emergence of replant disease. However, the regulatory mechanisms governing the transport of phenolics from plant roots to the rhizosphere remain poorly understood. A potential phenolic efflux transporter from Rehmannia glutinosa, designated RgMATE35, has been preliminarily characterized. The objective of this study was to elucidate the molecular function of RgMATE35 in the secretion of phenolics and to investigate its role in the development of plant replant disease using quantitative real-time PCR (qRT-PCR), genetic transformation, HPLC-Q-TOF-MS and other analytical techniques. A tissue expression pattern analysis of RgMATE35 revealed that it is highly expressed in plant roots. Transient expression analysis confirmed the localization of the protein in plasma membranes. An assessment of the transport activity of RgMATE35 in Xenopus oocytes indicated that it plays a role in facilitating the efflux of labeled ferulic acid ([2H3]-FA) and trans-p-coumaric acid [2H6]-pCA. The results of functional studies in R. glutinosa demonstrated that RgMATE35 positively mediates the secretion of FA and pCA from plant roots into the rhizosphere. A molecular and physiological analysis of RgMATE35 transgenic plants subjected to CM stress revealed that the overexpression or repression of RgMATE35 resulted in notable changes in the degree of autotoxic injury in plants. These findings demonstrate that RgMATE35 plays a positive role in the development of replant disease through the secretion of phenolic acids from plant roots. They also provide a fundamental framework for elucidating the molecular regulatory mechanism through which MATEs regulate replant disease through the root secretion of allelochemicals. ABSTRACT Most terrestrial plants can establish a reciprocal symbiosis with arbuscular mycorrhizal (AM) fungi to cope with adverse environmental stresses. The development of AM symbiosis is energetically costly and needs to be dynamically controlled by plants to maintain the association at mutual beneficial levels. Multiple components involved in the autoregulation of mycorrhiza (AOM) have been recently identified from several plant species; however, the mechanisms underlying the feedback regulation of AM symbiosis remain largely unknown. Here, we report that AM colonization promotes the flavonol biosynthesis pathway in tomato (Solanum lycopersicum), and an AM‐specific UDP‐glucosyltransferase SlUGT132, which probably has the flavonol glycosylation activity, negatively regulates AM development. SlUGT132 was predominantly expressed in the arbuscule‐containing cells, and its knockout or knockdown mutants showed increased soluble sugar content, root colonization level and arbuscule formation. Conversely, overexpression of SlUGT132 resulted in declined soluble sugar content and mycorrhization degree. Metabolomic assay revealed decreased contents of astragalin, tiliroside and cynaroside in slugt132 mycorrhizal roots, but increased accumulation of these flavonoid glycosides in SlUGT132‐overexpressing plant roots. Our results highlight the presence of a novel, SlUGT132‐mediated AOM mechanism, which enable plants to flexibly control the accumulation of soluble sugars and flavonoid glycosides in mycorrhizal roots and modulate colonization levels. This article extensively explores various bio-inspired techniques of interfacial strengthening. Bio-inspired composites fabricated by emulating five natural materials: nacre, byssus thread, plant roots, bone, and spider silk have been discussed. A detailed account of the strengthening mechanisms active in these natural materials responsible for improving fiber/matrix adhesion in artificial composites is presented. Dendritic growth on fiber surfaces is crucial in enhancing the mechanical properties via interlocking in root-inspired composites. Micro-waviness, nano-asperity interlocking, shear deformation, and mineral bridge fracture are the main strengthening mechanisms active in the nacre-inspired composites. Byssal threads have exceptional adhesive properties due to the sacrificial bonds, hidden length, and strong metal coordination cross-links. The typical ‘Bouligand’ microstructure of bone has inspired different composite materials with excellent crack deflection and arresting. Spider silk fibers have a highly oriented proteinous structure surrounded by a semi-amorphous matrix. Their skin-core structure provides tensile strength and toughness to these fibers, inspiring various artificial composites. This article explores additive manufacturing and chemical modification as the implementation techniques for bio-inspired strengthening. Applications of bio-inspired composites include biomedical, marine, aerospace, and tissue engineering. Challenges, such as optimal mechanical properties, sustainability, simulation, and integration with Industry 5.0, must be addressed systematically to augment their applicability. Summary: Climate and edaphic properties drive the biogeographic distribution of dominant soil microbial phylotypes in terrestrial ecosystems. However, the impact of plant species and their root nutritional traits on microbial distribution in coastal wetlands remains unclear.Here, we investigated the nutritional traits of 100 halophyte root samples and the bacterial communities in the corresponding soil samples from coastal wetlands across eastern China. This study spans 22° of latitude, covering over 2500 km from north to south.We found that 1% of soil bacterial phylotypes accounted for nearly 30% of the soil bacterial community abundance, suggesting that a few bacterial phylotypes dominated the coastal wetlands. These dominated phylotypes could be grouped into three ecological clusters as per their preference over climatic (temperature and precipitation), edaphic (soil carbon and nitrogen), and plant factors (halophyte vegetation, root carbon, and nitrogen). We further provide novel evidence that plant root nutritional traits, especially root C and N, can strongly influence the distribution of these ecological clusters.Taken together, our study provides solid evidence of revealing the dominance of specific bacterial phylotypes and the complex interactions with their environment, highlighting the importance of plant root nutritional traits on biogeographic distribution of soil microbiome in coastal wetland ecosystems. Summary: The ALMT1 transporter aids malate secretion, chelating Al3+ ions to form nontoxic Al–malate complexes, believed to exclude Al from the roots. However, the extent to which malate secreted by ALMT1 is solely used for the exclusion of Al3+ or can be reutilized by plant roots for internal Al tolerance remains uncertain.In our investigation, we explored the impact of malate secretion on both external and internal Al resistance in Arabidopsis thaliana. Additionally, we delved into the mechanism by which the tonoplast‐localized bacterial‐type ATP‐binding cassette (ABC) transporter complex STAR1/ALS3 promotes the degradation of the Al resistance transcription factor STOP1 to regulate ALMT1 expression.Our study demonstrates that the level of secreted malate influences whether the Al–malate complex is excluded from the roots or transported into root cells. The nodulin 26‐like intrinsic protein (NIP) subfamily members NIP1;1 and NIP1;2, located in the plasma membrane, coordinate with STAR1/ALS3 to facilitate Al–malate transport from root apoplasm to the symplasm and eventually to the vacuoles for the internal Al detoxification. ALS3‐dependent STAR1 interacts with and promotes the degradation of STOP1, regulating malate exudation.Our findings demonstrate the dual roles of malate exudation in external Al exclusion and Al absorption for internal Al detoxification. Background: In addition to their role as endosymbionts for plant roots, arbuscular mycorrhizal fungi (AMF) engage in complex interactions with various soil microorganisms, the rhizosphere, and the root endosphere of host plants. They also host diverse prokaryotic groups within their mycelia, contributing to what is termed multipartite symbiosis. In this study, we examined the impact of three AMF species—Rhizophagus irregularis, R. clarus, and R. cerebriforme—combined with microbial bioaugmentation on the diversity and composition of bacterial communities in the mycelia and hyphosphere. Using a microcosm design to separate the influence of host plant roots from AMF mycelia and Illumina MiSeq amplicon sequencing to analyze the bacterial communities. Results: Our results revealed that, while AMF identity and microbial bioaugmentation did not affect the structure of bacterial communities in the hyphosphere soil, they significantly altered the communities associated with their mycelia. Although all three AMF species belong to the same genus, with R. irregularis and R. clarus being closely related compared to R. cerebriforme, we observed variations in the bacterial communities associated with their mycelia. Interestingly, the mycelial bacterial community of R. cerebriforme contained 60 bacteriome core taxa exclusive to it, while R. clarus and R. irregularis had 25 and 9 exclusive taxa, respectively. Conclusion: This study suggests that organismal phylogeny influences the bacterial communities associated with AMF mycelia. These findings provide new insights into AMF and bacterial interactions, which are crucial for the successful deployment of AMF inoculants. The plant mycobiome plays a crucial role in the host life cycle, influencing both healthy and diseased states, and is essential for plant tolerance to drought. In this study, we used ITS metabarcoding to investigate the fungal community of the drought-resistant plant Malva sylvestris L. in Morocco along a gradient of precipitation, encompassing subhumid and semi-arid environments. We sampled three biotopes: rhizosphere, bulk soil, and root endosphere. Our findings revealed an absence of beta-diversity differences between bulk soil and rhizosphere, indicating that the plant does not selectively influence its rhizosphere mycobiome. Additionally, ASVs belonging to the genus Alternaria represented up to 30% of reads in the plant's roots and correlated with drought (p = 0.006), indicating a potential role for this fungal genus in mitigating drought, possibly as part of the dark septate endophyte group. Root staining and microscopic observation revealed extensive colonization by fungal hyphae and microsclerotia-like structures. Furthermore, ASVs identified as Fusarium equiseti were also correlated with low precipitation and recognized as a hub taxon in the roots. However, it remains uncertain whether this species is pathogenic or beneficial to the plant. These insights contribute to our understanding of the plant mycobiome's role in drought tolerance and highlight the importance of specific fungal taxa in supporting plant health under varying environmental conditions. Future research should focus on characterizing these taxa's functional roles and their interactions with the host plant to further elucidate their contributions to drought resistance. Introduction: Alzheimer's disease (AD) is the leading cause of dementia, and currently, no effective treatments are available to reverse or halt its progression in clinical practice. Although a plethora of studies have highlighted the benefits of physical exercise in combating AD, elder individuals often have limited exercise capacity. Therefore, mild physical exercise and nutritional interventions represent potential strategies for preventing and mitigating neurodegenerative diseases. Our research, along with other studies, have demonstrated that platycodin D (PD) or its metabolite, platycodigenin, derived from the medicinal plant Platycodon grandiflorus, exerts neuroprotective effects against amyloid b (Ab)-induced neuroinflammation. However, the combined effects of PD and physical exercise on alleviating AD have yet to be explored. The current study aimed to investigate whether combined therapy could synergistically ameliorate memory deficits and AD pathology in 5 FAD mice. Methods: Five-month-old 5 x FAD mice were randomly assigned to four groups, and received either PD (5 mg/kg/day, p.o.), voluntary running, or a combination of both for 47 days. Nest building test, locomotion test, and Morris water maze test were used to evaluate the cognitive function. Immunohistochemical and ELISA analysis was performed to determine Ab build-up, microglia and astrocytes hyperactivation, and survival neurons in the hippocampus and perirhinal cortex. Real-time quantitative PCR analysis was used to assess the polarization of microglia and astrocytes. HPLC analysis was performed to measure monoamine neurotransmitters in the hippocampus. Results and discussion: The combination of PD and voluntary running synergistically restored nest-building behavior, alleviated recognition and spatial memory deficits, and showed superior effects compared to monotherapy. In addition, the PD and voluntary running combination reduced Ab build-up, decreased hyperactivation of microglia and astrocytes in the hippocampus and perirhinal cortex, promoted the polarization of inflammatory M1 microglia and reactive astrocytes toward beneficial phenotypes, and lowered systemic circulating pro-inflammatory cytokines while increasing anti-inflammatory cytokines in 5 FAD mice. Furthermore, combined therapy effectively protected neurons and increased levels of 5-hydroxytryptamine (5-HT) and dopamine (DA) in the hippocampus of 5 FAD mice. Background: Clausena harmandiana (CH), commonly known as song fa dong, was a medicinal plant traditionally used to treat illnesses and as a health tonic. CH root extract (CHRE) exhibited various bioactivities, including neuroprotective, antioxidant, antimicrobial, antifungal, anti-inflammatory, and anti-cancer effects. However, CHRE data on neuroprotective in AD-like animal models were still scarce. Objectives: This study aimed to investigate the effects of CHRE on Aβ1-42-induced cognitive deficits, free radical damage, and neuronal death in rats. Methods: Forty-eight adult male Sprague-Dawley rats (250–300 g) were classified as sham control (SC), V+Aβ, Vit C+Aβ, CHRE125+Aβ, CHRE250+Aβ, and CHRE500+Aβ (n = 8 in each group). Animals were orally administered with 0.5% sodium carboxymethylcellulose, vitamin C (200 mg/kg BW), or CHRE (125, 250, and 500 mg/kg BW) and were untreated for 35 days. On day 21, all treated rats were injected with 1 µl of aggregated Aβ1-42 (1 µg/µl) into the lateral ventricles, bilaterally, whereas untreated rats were injected with sterilized normal saline (NS). The Morris water maze test estimated the rat's learning and memory one week later. At the end of the treatment, all rats were sacrificed, and their brains were removed and divided into two hemispheres. On the left, morphological changes and neuronal density were observed in hippocampal CA1 and CA3 regions. While, on the right, changes in free radical damage markers (SOD, CAT, GPx, MDA, and Nrf2) and protein expression of active caspase-3 were evaluated in the hippocampus. Results: Pretreatment with CHRE at all doses could alleviate spatial learning and memory defects. CHRE also improved morphological changes and a decrease in neuronal density in CA1 and CA3 regions. Additionally, CHRE significantly increased the activities of antioxidant enzymes (SOD, CAT, GPx) and Nrf2 expression. This was coupled with significantly decreased MDA levels and active caspase-3 expression in the hippocampus of Aβ1-42-induced rats, which was similar to vitamin C exposure. Conclusions: Our findings suggested that CHRE ameliorated cognitive deficits and exhibited neuroprotective effects by reducing free radical damage and mitigating neuronal abnormality and neuronal death. In mountainous areas, wild edible plants are an important part of the local diet. Climate change and anthropogenic activities have profound effects on wild edible plants in these areas. Ethnobotanical studies are important for understanding the use patterns and harvest impacts on these plants. In this regard, the Shahrood region, with its diverse historical/ethnic background, is an appropriate starting point to investigate exploitation patterns and impacts of harvest of wild edible plants. During 2021–2022, ethnobotanical surveys were conducted in 12 villages in the region using semi-structured interviews and participatory observations. Data collection included assessing wild edible plant species diversity, their habitats, collection time and quantities, plant parts used and methods of consumption and preparation. The data were analyzed using use report indicators, relative frequency of citations, and use value (UV). A total of 1086 use reports were documented from a total of 44 interviews. A total of 67 wild species (66 plants and 1 fungus) belonging to 54 genera and 24 families were used as edible in the study area. Rosaceae (9 species), Apiaceae (8), Lamiaceae (8), Asteraceae (7), and Amaryllidaceae (5) were the families with the most reported species. Allium (5 species) was the most diverse genus in terms of species diversity. Herbs were the most commonly used life form (79.1%), followed by shrubs (13.4%) and trees (6%). The most consumed plant parts were young leaves (25%) and young aerial parts (21.4%). There were 13 use categories identified. The majority of edible plants were collected in April–May, mainly from areas nearby villages (37.7%) and rangelands (33.7%). The most important edible plants in the study area were Allium iranicum (Wendelbo) Wendelbo (UV = 1.7), Mentha longifolia (L.) Huds. (1.5), Allium umbilicatum Boiss. (1.47), Tragopogon graminifolius DC. (1.38), Lepidium draba L. (1.27), Urtica dioica L. (1.18), Falcaria vulgaris Bernh. (1.13), Malva neglecta Wallr. (1.11) and Eremurus sp. (1.09). Potassium (K+) is essential to plant growth and development. The absorption and transport of K+ by plant roots occurs mainly through K+ channels or transporters. In this study, an AKT1-type channel named PbrAKT1 was successfully cloned and characterised by a series of detailed procedures, including protein sequence alignment, expression and subcellular localisation analysis, as well as functional verification by heterologous expression function validation in yeast mutant R5421, Arabidopsis akt1 mutant, and Xenopus oocytes. The results showed that PbrAKT1 is preferentially expressed in pear roots and exhibits plasma membrane localisation, but its expression is unaffected by low K+ treatment. PbrAKT1 exhibits the function of an inwardly-rectifying K+ channel, and its activity is autonomously regulated when expressed in Xenopus oocytes. In addition, a series of heterologous expression experiments confirmed that PbrAKT1 plays an indispensable role in the processes of K+ transport, as well as alleviating the low-K+ -sensitive phenotype of yeast mutant R5421 and Arabidopsis akt1 mutant. The effects of mechanical defoliation and pinching (1 cm tip cutting) on Buxus plant growth, nutrient mobilization, and root architecture were determined. When 100% defoliation was applied, the highest increase rates of 80.3% in shoots and 88% in leaves were observed compared to the control group. In contrast, the overall effects of defoliation and pinching were negative, with 100% defoliation having the most negative effects. The chlorophyll content of the newly formed young leaves was also 50% lower with 100% defoliation. Leaves and root nutrient mobilization changed significantly, depending on the effects of defoliation and pinching. Apart from a very small increase in root length and number of forks, the effects of the treatments were negative, with 100% defoliation having the greatest negative effect on root development. Most affected was the number of crossings, which was 78% lower than in the control. In addition, machine learning (ML) algorithms were used in the study, including multilayer perceptron, J48, PART, and logistic regression. The input variables were evaluated to model and predict the root features. The performance values of the ML algorithms were noted in the following order: Logistic Regression> PART> J48> MultilayerPerceptron. As the severity of defoliation increased, the losses of the plant also increased. However, boxwood has mechanisms to compensate for these losses even when it suffers complete defoliation. Leea indica (Burm.f.) Merr, commonly known as Memaye, is a shrub utilized by the Besemah tribe in Lahat Regency, South Sumatra, for treating infectious diseases such as hepatitis and warts, as well as degenerative diseases like liver disorders. This plant's medicinal properties are primarily derived from its roots and fruits, which contain secondary metabolites like alkaloids, flavonoids, and terpenoids. Anatomical studies are crucial to identify specific structures within the plant that may store these metabolite compounds. This research aims to analyze the anatomical structure and distribution of secretory structures in the vegetative organs (leaves, stems, and roots) of Leea indica. Using a descriptive method, the study employed Paraffin and Whole mount techniques to prepare samples for microscopic observation. Results revealed the presence of epidermal tissue, basic tissue, and vascular tissue. Additionally, internal secretory structures, including cavities and two types of idioblast cells, were found to function as storage sites for secondary metabolites. Plant roots represent about a quarter of global plant biomass and constitute a primary source of soil organic carbon (C). Yet, considerable uncertainty persists regarding root litter decomposition and their responses to global change factors (GCFs). Much of this uncertainty stems from a limited understanding of the multifactorial effects of GCFs and it remains unclear how these effects are mediated by litter quality, soil conditions and microbial functionality. Using complementary field decomposition and laboratory incubation approaches, we assessed the relative controls of GCF‐mediated changes in root litter traits and soil and microbial properties on fine‐root decomposition under warming, nitrogen (N) enrichment, and precipitation alteration. We found that warming and N enrichment accelerated fine‐root decomposition by over 10%, and their combination showed an additive effect, while precipitation reduction suppressed decomposition overall by 12%, with the suppressive effect being most significant under warming‐alone and N enrichment‐alone conditions. Significantly, changes in litter quality played a dominant role and accelerated fine‐root decomposition by 15% ~ 18% under warming and N enrichment, while changes in soil and microbial properties were predominant and reduced decomposition by 7% ~ 10% under precipitation reduction and the combined warming and N enrichment. Examining only the decomposition environment or litter properties in isolation can distort global change effects on root decomposition, underestimating precipitation reduction impacts by 38% and overstating warming and N effects by up to 73%. These findings highlight that the net impact of GCFs on root litter decomposition hinges on the interplay between GCF‐modulated root decomposability and decomposition environment, as well as on the synergistic or antagonistic relationships among GCFs themselves. Our study emphasizes that integrating the legacy effects of multiple GCFs on root traits, soil conditions and microbial functionality would improve our prediction of C and nutrient cycling under interactive global change scenarios. Cotton (Gossypium spp.) is the most important fibre crop worldwide. Black root rot and Fusarium wilt are two major diseases of cotton caused by soil-borne Berkeleyomyces rouxiae and Fusarium oxysporum f. sp. vasinfectum (Fov), respectively. Phenotyping plant symptoms caused by soil-borne pathogens has always been a challenge. To increase the uniformity of infection, we adapted a seedling screening method that directly uses liquid cultures to inoculate the plant roots and the soil. Four isolates, each of B. rouxiae and Fov, were collected from cotton fields in Australia and were characterised for virulence on cotton under controlled plant growth conditions. While the identities of all four B. rouxiae isolates were confirmed by multilocus sequencing, only two of them were found to be pathogenic on cotton, suggesting variability in the ability of isolates of this species to cause disease. The four Fov isolates were phylogenetically clustered together with the other Australian Fov isolates and displayed both external and internal symptoms characteristic of Fusarium wilt on cotton plants. Furthermore, the isolates appeared to induce varied levels of plant disease severity indicating differences in their virulence on cotton. To contrast the virulence of the Fov isolates, four putatively non-pathogenic Fusarium oxysporum (Fo) isolates collected from cotton seedlings exhibiting atypical wilt symptoms were assessed for their ability to colonise cotton host. Despite the absence of Secreted in Xylem genes (SIX6, SIX11, SIX13 and SIX14) characteristic of Fov, all four Fo isolates retained the ability to colonise cotton and induce wilt symptoms. This suggests that slightly virulent strains of Fo may contribute to the overall occurrence of Fusarium wilt in cotton fields. Roots play an important role in plant growth, including providing essential mechanical support, water uptake, and nutrient absorption. Nanomaterials play a positive role in improving plant root development, but there is limited knowledge of how nanomaterials affect lateral root (LR) formation. Poly (acrylic) acid coated nanoceria (cerium oxide nanoparticles, PNC) are commonly used to improve plant stress tolerance due to their ability to scavenge reactive oxygen species (ROS). However, its impact on LR formation remains unclear. In this study, we investigated the effects of PNC on LR formation in Arabidopsis thaliana by monitoring ROS levels and Ca2+ distribution in roots. Our results demonstrate that PNC significantly promote LR formation, increasing LR numbers by 26.2%. Compared to controls, PNC-treated Arabidopsis seedlings exhibited reduced H2O2 levels by 18.9% in primary roots (PRs) and 40.6% in LRs, as well as decreased O 2 · − levels by 47.7% in PRs and 88.5% in LRs. When compared with control plants, Ca2+ levels were reduced by 35.7% in PRs and 22.7% in LRs of PNC-treated plants. Overall, these results indicate that PNC could enhance LR development by modulating ROS and Ca2+ levels in roots. In this study, we showed that cerium oxide nanoparticles (PNC) promoted lateral root formation in Arabidopsis thaliana by modulating reactive oxygen species and Ca2+ distribution in both primary and lateral roots. Results indicated that applying PNC to plants have potential to improve the development of root system and thus overall plant growth performance. Effective colonization by microbe in the rhizosphere is critical for establishing a beneficial symbiotic relationship with the host plant. Bacillus subtilis, a soil-dwelling bacterium that is commonly found in association with plants and their rhizosphere, has garnered interest for its potential to enhance plant growth, suppress pathogens, and contribute to sustainable agricultural practices. However, research on the dynamic distribution of B. subtilis within the rhizosphere and its interaction mechanisms with plant roots remains insufficient due to limitations in existing in situ detection methodologies. To achieve dynamic in situ detection of the rhizosphere environment, we established iRhizo-Chip, a microfluidics-based platform. Using this device to investigate microbial behavior within the rhizosphere, we found obvious diurnal fluctuations in the growth of B. subtilis in the rhizosphere. Temporal dynamic analysis of rhizosphere dissolved oxygen (DO), pH, dissolved organic carbon, and reactive oxygen species showed that diurnal fluctuations in the growth of B. subtilis are potentially related to a variety of environmental factors. Spatial dynamic analysis also showed that the spatial distribution changes of B. subtilis and DO and pH were similar. Subsequently, through in vitro control experiments, we proved that rhizosphere DO and pH are the main driving forces for diurnal fluctuations in the growth of B. subtilis. Our results show that the growth of B. subtilis is driven by rhizosphere DO and pH, resulting in diurnal fluctuations, and iRhizo-Chip is a valuable tool for studying plant rhizosphere dynamics. Red bed composite polymers composed of weathered red bed soil, adhesive materials, and water-retaining materials have been applied as a new type of material for environmental restoration. However, the promotion and application of this material has been limited by a lack of understanding of its action mechanism in environmental restoration. The objective of this study is to innovatively propose a water‒soil-air‒plant mutual feedback mechanism based on this material. Therefore, water‒soil-air‒plant mutual feedback tests were conducted in this study under 3 initial water contents and 10 red bed composite polymers ratios. Key parameters, namely, water content, soil conductivity, pH, temperature, O2 and CO2 contents, pigeon pea (Cajanus cajan) germination number and plant height were monitored and analyzed. As the results, a mutual feedback mechanism driving water retention, soil consolidation, air retention, and plant rooting was revealed under the application of red bed composite polymers. And, suitable environments and optimal compositions for this material are proposed. The study results provide a theoretical basis for the large-scale application of red bed composite polymers. Root nitrogen (N) reallocation is a conservative N-use strategy involving root N remobilization for shoot growth, reproduction, and other metabolic processes, but a critical framework for how it varies among plants is missing. Community-level root N reallocation increases with richness of plant species and N-acquisition strategies because of complementarity in N uptake and storage. Root N reallocation is a conservative trait opposite to competitive root traits along the major axis of the fast–slow spectrum of the root economics space. Root N reallocation declines with more abundant soil microbial K -strategists, hyphosphere/rhizosphere microbial recruitment, and mycorrhizal colonization due to acceleration of N release from recalcitrant organic matter and facilitation of plant efficiency in N foraging. Root nitrogen (N) reallocation involves remobilization of root N-storage pools to support shoot growth. Representing a critical yet underexplored facet of plant function, we developed innovative frameworks to elucidate its connections with key ecosystem components. First, root N reallocation increases with plant species richness and N-acquisition strategies, driven by competitive stimulation of plant N demand and synergies in N uptake. Second, competitive root traits and mycorrhizal symbioses, which enhance N foraging and uptake, exhibit trade-offs with root N reallocation. Furthermore, root N reallocation is attenuated by N-supply attributes such as increasing litter quality, soil fungi-to-bacteria ratios, and microbial recruitment in the hyphosphere/rhizosphere. SUMMARY: Plants synthesize hundreds of small secretory peptides, which are perceived by the receptor‐like kinase (RLK) family at the cell surface. Various signaling peptide‐RLK pairs ensure plant adaptation to distinct environmental conditions. Here, we report that SERINE RICH ENDOGENOUS PEPTIDE (SCOOP) immune peptides modulate root growth and development by regulating PIN‐FORMED (PIN)‐regulated polar auxin transport in Arabidopsis. The SCOOP4 and SCOOP12 treatments impaired root gravitropic growth, auxin redistribution in response to gravistimulation, and PIN abundance in the PM. Furthermore, genetic and cell biological analyses revealed that these physiological and cellular effects of SCOOP4 and SCOOP12 peptides are mediated by the receptor MALE DISCOVERER1‐INTERACTING RECEPTOR LIKE KINASE2 (MIK2) and the downstream mitogen‐activated kinase MPK6. Biochemical evidence indicates that MPK6 directly phosphorylates the cytosolic loop of PIN proteins. Our work established a link between the immune signaling peptide SCOOPs and root growth pathways, providing insights into the molecular mechanisms underlying plant root adaptive growth in the defense response. An ethnobotanical survey was conducted to determine the plant species used against gout and gout-associated conditions (GACs) in Monduli, Arusha, Tanzania. The survey that involved 21 Maasai traditional food system (TFS) and traditional medicine (TM) practitioners revealed that the study area had 101 plant species distributed in 84 genera and 42 families. About 79% of the species were used as medicine; some were used as food or for processing. The medicinal plants (MPs) managed 69 health conditions including gout, "Olgila" and GACs. The MPs had potential nutritional and antioxidant agents against diseases including metabolic diseases (MDs), like gout. The root parts of the plants (54% species) were exploited the most; most plants (94%) were sourced from the wild. Medicinal plants and associated indigenous knowledge was surveyed in Gishe Rabel District, Ethiopia, from 60 informants using semi-structured interviews, focus group discussions, and guided field walks. Preference rankings, paired comparisons, direct matrix rankings, and informant consensus factors (ICFs) were calculated. Ninety-five medicinal plants from 82 genera and 47 families were identified, five of which were unique to Ethiopia. Sixty-nine species were being used in treating human diseases. The leaves were the most commonly used plant part, followed by the roots. One new canthinone glycoside (1), together with six known compounds (2–7) including three lignans (2–4), two coumarins (5–6) and one phenol (7) was isolated from the root barks of Ailanthus altissima. The structure of new compound 1 was established by the interpretation of UV, IR, MS and NMR data, while its absolute configuration was determined by acid hydrolysis and GIAO NMR calculations with DP4+ probability analysis. The inhibitory effects of all compounds on Nitric oxide (NO) production were investigated in lipopolysaccharide (LPS)-induced RAW 264.7 cells.
