Reduced defense against large herbivores has been suggested to be part of the island syndrome in plants. However, empirical evidence for this pattern is mixed. In this paper, we present two studies that compare putative physical and chemical defense traits from plants on the California Channel Islands and nearby mainland based on sampling of both field and common garden plants. In the first study, we focus on five pairs of woody shrubs from three island and three mainland locations and find evidence for increased leaf area, decreased marginal leaf spines, and decreased concentrations of cyanogenic glycosides in island plants. We observed similar increases in leaf area and decreases in defense traits when comparing island and mainland genotypes grown together in botanic gardens, suggesting that trait differences are not solely driven by abiotic differences between island and mainland sites. In the second study, we conducted a common garden experiment with a perennial herb-Stachys bullata (Lamiaceae)-collected from two island and four mainland locations. Compared to their mainland relatives, island genotypes show highly reduced glandular trichomes and a nearly 100-fold reduction in mono- and sesquiterpene compounds from leaf surfaces. Island genotypes also had significantly higher specific leaf area, somewhat lower rates of gas exchange, and greater aboveground biomass than mainland genotypes across two years of study, potentially reflecting a broader shift in growth habit. Together, our results provide evidence for reduced expression of putative defense traits in island plants, though these results may reflect adaptation to both biotic (i.e., the historical absence of large herbivores) and climatic conditions on islands.

The rice receptor kinase XA21 confers broad-spectrum resistance to Xanthomonas oryzae pv. oryzae (Xoo), the causal agent of rice bacterial blight disease. To investigate the relationship between the expression level of XA21 and resulting resistance, we generated independent HA-XA21 transgenic rice lines accumulating the XA21 immune receptor fused with an HA epitope tag. Whole-genome sequence analysis identified the T-DNA insertion sites in sixteen independent T0 events. Through quantification of the HA-XA21 protein and assessment of the resistance to Xoo strain PXO99 in six independent transgenic lines, we observed that XA21-mediated resistance is dose dependent. In contrast, based on the four agronomic traits quantified in these experiments, yield is unlikely to be affected by the expression level of HA-XA21. These findings extend our knowledge of XA21-mediated defense and contribute to the growing number of well-defined genomic landing pads in the rice genome that can be targeted for gene insertion without compromising yield.

Multiple Salmonella enterica serovars and strains have been reported to be able to persist inside the foliar tissue of lettuce (Lactuca sativa L.), potentially resisting washing steps and reaching the consumer. Intraspecies variation of the bacterial pathogen and of the plant host can both significantly affect the outcome of foliar colonization. However, current understanding of the mechanisms underlying this phenomenon is still very limited. In this study, we evaluated the foliar fitness of 14 genetically barcoded S. enterica isolates from 10 different serovars, collected from plant and animal sources. The S. enterica isolates were vacuum-infiltrated individually or in pools into the leaves of three- to four-week-old lettuce plants. To estimate the survival capacity of individual isolates, we enumerated the bacterial populations at 0- and 10- days post-inoculation (DPI) and calculated their net growth. The competition of isolates in the lettuce apoplast was assessed through the determination of the relative abundance change of barcode counts of each isolate within pools during the 10 DPI experimental period. Isolates exhibiting varying apoplast fitness phenotypes were used to evaluate their capacity to grow in metabolites extracted from the lettuce apoplast and to elicit the reactive oxygen species burst immune response. Our study revealed that strains of S. enterica can substantially differ in their ability to survive and compete in a co-inhabited lettuce leaf apoplast. The differential foliar fitness observed among these S. enterica isolates might be explained, in part, by their ability to utilize nutrients available in the apoplast and to evade plant immune responses in this niche.

SUMMARY Phytosulfokine (PSK) is a plant growth‐promoting peptide hormone that is perceived by its cell surface receptors PSKR1 and PSKR2 in Arabidopsis. Plants lacking the PSK receptors show phenotypes consistent with PSK signaling repressing some plant defenses. To gain further insight into the PSK signaling mechanism, comprehensive transcriptional profiling of Arabidopsis treated with PSK was performed, and the effects of PSK treatment on plant defense readouts were monitored. Our study indicates that PSK's major effect is to downregulate defense‐related genes; it has a more modest effect on the induction of growth‐related genes. WRKY transcription factors (TFs) emerged as key regulators of PSK‐responsive genes, sharing commonality with a pathogen‐associated molecular pattern (PAMP) responses, flagellin 22 (flg22), but exhibiting opposite regulatory directions. These PSK‐induced transcriptional changes were accompanied by biochemical and physiological changes that reduced PAMP responses, notably mitogen‐activated protein kinase (MPK) phosphorylation (previously implicated in WRKY activation) and the cell wall modification of callose deposition. Comparison with previous studies using other growth stimuli (the sulfated plant peptide containing sulfated tyrosine [PSY] and Pseudomonas simiae strain WCS417) also reveals WRKY TFs' overrepresentations in these pathways, suggesting a possible shared mechanism involving WRKY TFs for plant growth–defense trade‐off. Aphids are insects that feed on phloem and introduce effector proteins into plant cells through saliva. These effector proteins are key in regulating host plant defense and enhancing aphid host adaptation. We identified these salivary proteins in the cotton aphids genome and named them AgoArmet and AgoC002. Multiple sequence alignment, protein structure analysis, and phylogenetic analysis of these proteins with related proteins from other insects showed that AgoArmet and Armet of Aphis craccivora have high sequence identity (97%) and belong to the same evolutionary branch and that AgoC002 shares the highest sequence identity (80%) and closest evolutionary relationship with C002 of Aphis glyceins. Expression profiling of AgoArmet and AgoC002 showed that they were most highly expressed in cotton aphids during the adult-3d period. Cotton aphids transferred to zucchini leaves resulted in a significant increase in the expression of AgoArmet and AgoC002 within 48h. To investigate the functions of AgoArmet and AgoC002 , we decreased the expression of these genes in cotton using virus-induced gene silencing (VIGS), which ultimately led to a 38% and 26% decrease in cotton aphids fecundity, respectively. Moreover, the reduction in AgoC002 expression resulted in a significant (24%) reduction in body weight. Plant pathogens cause diseases by suppressing plant immune response and interacting with plant cells. Investigating these interactions assists in decoding the molecular strategies the pathogen uses to overcome plant immunity. Among plant pathogens, the nematodes parasitizing various plants incur a profound impact on food production across the globe. To deal with these parasites, plants have developed a complicated defense system, including performed defenses like rigid cell walls and reinforcements acting as the first line of defense to combat any invader. Plants also have a wide diversity of constitutively released phytochemicals that are toxic to the invading microbes as their defense arsenals. Additionally, a substantial system of host responses is triggered in response to infection based on the abilities of the host plants to sense and recognize the invading pathogen. Nematodes have evolved the strategies to perceive and respond to host defense through their nervous system which help them escape, avoid, or neutralize the host plant defense systems. For developing an effective management strategy, it is crucial to understand the mechanism by which the nematode suppress the host defense. Previous reviews mainly discussed the interaction of plants with the nematodes for their immunity against nematodes. The present review will discuss the strategies employed by the plant parasitic nematodes for suppressing plant defense along with an overall insights into the basic nematode recognition mechanism and basal immunity response of the host plant. The mechanism of modulating host defense by nematodes including the role of their effectors were also discussed. The latest research progress about the release of metabolites by plants, and the mode of action of these defensive chemicals at the molecular level in combating the nematode invasion was also analyzed. Plant diseases diminish crop yields and put the world's food supply at risk. Plant elicitor peptides (Peps) are innate danger signals inducing defense responses both naturally and after external application onto plants. Pep-triggered defense networks are compatible with pattern-triggered immunity (PTI). Nevertheless, in complex regulatory pathways, there is crosstalk among different signaling pathways, involving noncoding RNAs in the natural response to pathogen attack. Here, we used Prunus persica, PpPep2 and a miRNA-Seq approach to show for the first time that Peps regulate, in parallel with a set of protein-coding genes, a set of plant miRNAs (~15%). Some PpPep2-regulated miRNAs have been described to participate in the response to pathogens in various plant–pathogen systems. In addition, numerous predicted target mRNAs of PpPep2-regulated miRNAs are themselves regulated by PpPep2 in peach trees. As an example, peach miRNA156 and miRNA390 probably have a role in plant development regulation under stress conditions, while others, such as miRNA482 and miRNA395, would be involved in the regulation of resistance (R) genes and sulfate-mediated protection against oxygen free radicals, respectively. This adds to the established role of Peps in triggering plant defense systems by incorporating the miRNA regulatory network and to the possible use of Peps as sustainable phytosanitary products. Elaeagnus angustifolia L. can attract adult Asian longhorned beetle (ALB), Anoplophora glabripennis (Motschulsky), and kill their offspring by gum secretion in oviposition scars. This plant has the potential to be used as a dead-end trap tree for ALB management. However, there is a limited understanding of the attraction ability and biochemical defense response of E. angustifolia to ALB. In this study, we conducted host selection experiments with ALB and then performed physiological and biochemical assays on twigs from different tree species before and after ALB feeding. We analyzed the differential metabolites using the liquid chromatograph–mass spectrometer method. The results showed that ALB's feeding preference was E. angustifolia > P.× xiaohei var. gansuensis > P. alba var. pyramidalis. After ALB feeding, the content of soluble sugars, soluble proteins, flavonoids, and tannins decreased significantly in all species. In three comparison groups, a total of 492 differential metabolites were identified (E. angustifolia:195, P.× xiaohei var. gansuensis:255, P. alba var. pyramidalis:244). Differential metabolites were divided into overlapping and specific metabolites for analysis. The overlapping differential metabolites 7-isojasmonic acid, zerumbone, and salicin in the twigs of three tree species showed upregulation after ALB feeding. The specific metabolites silibinin, catechin, and geniposide, in E. angustifolia, significantly increased after being damaged. Differential metabolites enriched in KEGG pathways indicated that ALB feeding activated tyrosine metabolism and the biosynthesis of phenylpropanoids in three tree species, with a particularly high enrichment of differential metabolites in the flavonoid biosynthesis pathway in E. angustifolia. This study provides the metabolic defense strategies of different tree species against ALB feeding and proposes candidate metabolites that can serve as metabolic biomarkers, potentially offering valuable insights into using E. angustifolia as a control measure against ALB. The effects of preharvest or/and postharvest treatments with salicylic acid (SA) on the physicochemical quality of peach fruit, infection with the fungus Monilinia laxa and the phenolic response to infection in SA treatments in different peach peel tissues were evaluated. Preharvest SA treatments had an effect on the physical quality parameters only at harvest on three times SA-sprayed peaches, with which a lower hue angle and a higher percentage of red blush occurred, indicating better coloration of the fruit. It was found that the intensity of M. laxa infection was not affected by preharvest SA treatments, only by soaking peach fruit in SA solution after harvest. On SA soaked peach fruit, M. laxa spread between 1.8 and 3.9 mm/day slower than on non-soaked fruit, resulting in a 34% lower intensity of infection five days after infection. Peach fruit treated with SA after harvest also had a significantly lower percentage of sporulating fruit (7.8%) compared to non-soaked fruit (68.9%). The phenolic response differed between tissues, since the proportion of hydroxycinnamic acids and anthocyanins was higher in the healthy tissue of infected fruit, while the proportion of flavanols was higher in the boundary tissue of infected fruit. The strongest phenolic response to infection and treatments with SA was in higher levels of neochlorogenic acid, cyanidin-3-glucosides and cyanidin-3-rutinosides. [ABSTRACT FROM AUTHOR]

Citrus is commercially propagated via grafting, which ensures trees have consistent fruit traits combined with favorable traits from the rootstock such as soil adaptability, vigor, and resistance to soil pathogens. Graft incompatibility can occur when the scion and rootstock are not able to form a permanent, healthy union. Understanding and preventing graft incompatibility is of great importance in the breeding of new fruit cultivars and in the choice of scion and rootstock by growers. The rootstock US-1283, a citrandarin generated from a cross of "Ninkat" mandarin (Citrus reticulata) and "Gotha Road" #6 trifoliate orange (Poncirus trifoliata), was released after years of field evaluation because of its superior productivity and good fruit quality on "Hamlin" sweet orange (C. sinensis) under Florida's growing conditions. Subsequently, it was observed that trees of "Bearss" lemon (C. limon) and "Valencia" sweet orange (C. sinensis) grafted onto US-1283 exhibited unhealthy growth near the graft union. The incompatibility manifested as stem grooving and necrosis underneath the bark on the rootstock side of the graft. Another citrandarin rootstock, US-812 (C. reticulata "Sunki" × P. trifoliata "Benecke"), is fully graft compatible with the same scions. Transcriptome analysis was performed on the vascular tissues above and below the graft union of US-812 and US-1283 graft combinations with "Bearss" and "Valencia" to identify expression networks associated with incompatibility and help understand the processes and potential causes of incompatibility. Transcriptional reprogramming was stronger in the incompatible rootstock than in the grafted scions. Differentially expressed genes (DEGs) in US-1283, but not the scions, were associated with oxidative stress and plant defense, among others, similar to a pathogen-induced immune response localized to the rootstock; however, no pathogen infection was detected. Therefore, it is hypothesized that this response could have been triggered by signaling miscommunications between rootstock and scion either through (1) unknown molecules from the scion that were perceived as danger signals by the rootstock, (2) missing signals from the scion or missing receptors in the rootstock necessary for the formation of a healthy graft union, (3) the overall perception of the scion by the rootstock as non-self, or (4) a combination of the above. Seaweeds are a natural marine resource containing many bioactive compounds such as amino acids, lipids, carbohydrates, proteins, phytohormones, and antimicrobial compounds. Since ancient times, seaweeds have been used in various sectors, including medicine, food, and the cosmetic industry. Currently, seaweeds are a promising alternative to reduce the application of harmful chemicals in agriculture. Seaweed and its derived products have been utilized for plant growth promotion, immunity enhancement, and the reduction of biotic and abiotic stresses. In the current global scenario, synthetic fertilizers and chemical pesticides are commonly used to increase agricultural crop production to meet the growing demands of the world population. However, these chemicals pose significant threats to the health of people, livestock, plants, soil, and the entire environment. In contrast, seaweed-based products are emerging as a newer option for stress mitigation and reduction, offering an alternative to synthetic chemicals. This article explains the use of seaweed extracts to increase the tolerance of plants to biotic and abiotic stresses. We also address the functions of various bioactive compounds present in seaweed extracts and the mechanisms by which they promote plant growth and induce defense against different stresses. Fusarium oxysporum causes devastating vascular wilt diseases in numerous crop species, resulting in substantial yield losses. The Arabidopsis thaliana - F. oxysporum f.sp. conglutinans (FOC) model system enables the identification of meaningful genotype–phenotype correlations and was applied in this study to evaluate the effects of overexpressing an NLR gene (AsTIR19) from Arachis stenosperma against pathogen infection. AsTIR19 overexpression (OE) lines exhibited enhanced resistance to FOC without any discernible phenotype penalties. To elucidate the underlying resistance mechanisms mediated by AsTIR19 overexpression, we conducted whole transcriptome sequencing of an AsTIR19-OE line and non-transgenic wild-type (WT) plants inoculated and non-inoculated with FOC using Illumina HiSeq4000. Comparative analysis revealed 778 differentially expressed genes (DEGs) attributed to transgene overexpression, while fungal inoculation induced 434 DEGs in the OE line, with many falling into defense-related Gene Ontology (GO) categories. GO and KEGG enrichment analysis showed that DEGs were enriched in the phenylpropanoid and flavonoid pathways in the OE plants. This comprehensive transcriptomic analysis underscores how AsTIR19 overexpression reprograms transcriptional networks, modulating the expression of stress-responsive genes across diverse metabolic pathways. These findings provide valuable insights into the molecular mechanisms underlying the role of this NLR gene under stress conditions, highlighting its potential to enhance resistance to Fusarium oxysporum. Summary: Lignin, a complex heterogenous polymer present in virtually all plant cell walls, plays a critical role in protecting plants from various stresses. However, little is known about how lignin modifications in sorghum will impact plant defense against sugarcane aphids (SCA), a key pest of sorghum.We utilized the sorghum brown midrib (bmr) mutants, which are impaired in monolignol synthesis, to understand sorghum defense mechanisms against SCA. We found that loss of Bmr12 function and overexpression (OE) of Bmr12 provided enhanced resistance and susceptibility to SCA, respectively, as compared with wild‐type (WT; RTx430) plants.Monitoring of the aphid feeding behavior indicated that SCA spent more time in reaching the first sieve element phase on bmr12 plants compared with RTx430 and Bmr12‐OE plants. A combination of transcriptomic and metabolomic analyses revealed that bmr12 plants displayed altered auxin metabolism upon SCA infestation and specifically, elevated levels of auxin conjugate indole‐3‐acetic acid–aspartic acid (IAA–Asp) were observed in bmr12 plants compared with RTx430 and Bmr12‐OE plants. Furthermore, exogenous application of IAA–Asp restored resistance in Bmr12‐OE plants, and artificial diet aphid feeding trial bioassays revealed that IAA–Asp is associated with enhanced resistance to SCA.Our findings highlight the molecular underpinnings that contribute to sorghum bmr12‐mediated resistance to SCA. Plant hormones play a central role in various physiological functions and mediate defense responses against (a)biotic stresses. Jasmonic acid (JA) has emerged as one of the key phytohormones involved in the response to necrotrophic pathogens. Under stressful conditions, plants can also produce small molecules, such as methylglyoxal (MG), a cytotoxic aldehyde. The enzymes glyoxalase I (GLYI) and glyoxalase II primarily detoxify MG. In Arabidopsis thaliana, GLYI4 has been recently characterized as having a crucial role in MG detoxification and emerging involvement in the JA pathway. Here, we investigated the impact of a GLYI4 loss-of-function on the Arabidopsis JA pathway and how MG affects it. The results showed that the glyI4 mutant plant had stunted growth, a smaller rosette diameter, reduced leaf size, and an altered pigment concentration. A gene expression analysis of the JA marker genes showed significant changes in the JA biosynthetic and signaling pathway genes in the glyI4 mutant. Disease resistance bioassays against the necrotroph Botrytis cinerea revealed altered patterns in the glyI4 mutant, likely due to increased oxidative stress. The MG effect has a further negative impact on plant performance. Collectively, these results contribute to clarifying the intricate interconnections between the GLYI4, MG, and JA pathways, opening up new avenues for further explorations of the intricate molecular mechanisms controlling plant stress responses. Phytoliths, the microscopic silica structures formed within plant tissues, are an emerging component of many sustainable plant protection attempts. They offer defense in multiple directions, physically strengthening plant tissues and biochemically engaging with the surroundings, and can diminish reliance on chemical pesticides and fertilizers. Physically, phytoliths enhance plant tissue rigidity and toughness, rendering them indigestible and less nutritious to herbivores and pathogens, thereby reducing feeding damage and disease incidence. Biochemically, phytoliths influence plant–microbe and plant–herbivore interactions by decreasing leaf palatability to herbivores, altering rhizosphere microbial communities including silica-specializing, plant-growth-promoting rhizobacteria, and diminishing pathogen proliferation. These effects enhance plant health by reducing pathogen spread and improving overall resilience. Furthermore, phytoliths accomplish crucial biogenic environmental roles such as facilitating biogeochemical silica and participating in essential nutrient cycles that uphold soil pH, fertility, and agricultural sustainability. Their enduring presence in soil enhances its structure, augments water retention, and improves nutrient availability, thereby fostering optimal conditions for plant growth. Additionally, phytoliths play a pivotal role in carbon sequestration and can immobilize heavy metals, mitigating soil contamination and advocating safer agricultural practices. This dual function in bolstering direct plant defense and indirectly enhancing soil health through carbon sequestration underscores the significant potential of phytoliths in sustainable agriculture. SUMMARY Hexokinases (HXKs), which sense and catalyze cellular sugar, play a critical role in the growth and development of various plants, including wheat, a primary source of human calories frequently attacked by fungal pathogens. However, the evolutionary dynamics and functional diversification of HXKs in wheat, particularly their roles in plant defense, remain unclear. Here, we discovered that the wheat hexokinase gene family originated through multiple ancient gene duplications across different plant lineages and has undergone comprehensive, multidimensional functional specialization in gene expression, subcellular localization, enzyme activity, and regulation of plant defense responses. Gene expression analysis suggests that two‐thirds of the TaHXK genes are responsive to fungal infection. Subcellular analysis reveals that while six TaHXKs are localized in mitochondria, three TaHXKs from different phylogenetic branches are sorted into other cellular compartments. Notably, biochemical analysis shows that TaHXKs in mitochondria differ in their glucose‐catalyzing activity, with TaHXK5 and TaHXK3 exhibiting the highest and lowest enzyme activity, respectively. Consistently, transient expression analysis suggests that TaHXK5 induces various plant defense responses, while TaHXK3 is defective in activating some plant defense responses. Furthermore, inactivation of the glucokinase activity of TaHXK5 compromised its function in defense activation, suggesting that mitochondrial TaHXKs display functional divergence in both enzyme activity and defense‐inducing activity that are intrinsically connected. Overall, our findings reveal that the multidimensional specialization events following the ancient duplication events may have shaped the functional diversity of HXKs in wheat, shedding light on their evolutionary dynamics and potentially contributing to the improvement of wheat defense. Most coexisting insect species exhibit stunted growth compared to individual species on plants. This phenomenon reflects an interspecific antagonism drawing extensive attention, while the underlying mechanisms remain largely uncharacterized. Mirids (Apolygus lucorum) and cotton bollworms (Helicoverpa armigera) are two common cotton pests. We identified a secretory protein, ASP1, from the oral secretion of mirids, found in the nucleus of mirid-infested cotton leaves. ASP1 specifically targets the transcriptional co-repressor TOPLESS (TPL) and inhibits NINJA-mediated recruitment of TPL, promoting plant defense response and gossypol accumulation in cotton glands. ASP1-enhanced defense inhibits the growth of cotton bollworms on cotton plants, while having limited impact on mirids. The mesophyll-feeding characteristic allows mirids to avoid most cotton glands, invalidating cotton defense. Our investigation reveals the molecular mechanism by which mirids employ cotton defense to selectively inhibit the feeding of cotton bollworms. The study revealed the underlying mechanism by which mirids manipulate cotton defense to confront the cotton bollworms co-existing on cotton plants. Plant domestication often alters plant traits, including chemical and physical defenses against herbivores. In squash, domestication leads to reduced levels of cucurbitacins and leaf trichomes, influencing interactions with insects. However, the impact of domestication on inducible defenses in squash remains poorly understood. Here, we investigated the chemical and physical defensive traits of wild and domesticated squash (Cucurbita argyrosperma), and compared their responses to belowground and aboveground infestation by the root-feeding larvae and the leaf-chewing adults of the banded cucumber beetle Diabrotica balteata (Coleoptera: Chrysomelidae). Wild populations contained cucurbitacins in roots and cotyledons but not in leaves, whereas domesticated varieties lacked cucurbitacins in all tissues. Belowground infestation by D. balteata larvae did not increase cucurbitacin levels in the roots but triggered the expression of cucurbitacin biosynthetic genes, irrespective of domestication status, although the response varied among different varieties. Conversely, whereas wild squash had more leaf trichomes than domesticated varieties, the induction of leaf trichomes in response to herbivory was greater in domesticated plants. Leaf herbivory varied among varieties but there was a trend of higher leaf damage on wild squash than domesticated varieties. Overall, squash plants responded to both belowground and aboveground herbivory by activating chemical defense-associated gene expression in roots and upregulating their physical defense in leaves, respectively. While domestication suppressed both chemical and physical defenses, our findings suggest that it may enhance inducible defense mechanisms by increasing trichome induction in response to herbivory. Plants produce a great number of phytochemicals serving a variety of different functions. Recently, the chemodiversity of these compounds (i.e., the diversity of compounds produced by a plant) has been suggested to be an important aspect of the plant phenotype that may shape interactions between plants, their environment, and other organisms. However, we lack an agreement on how to quantify chemodiversity, which complicates conclusions about the functional importance of it. Here, we discuss how chemodiversity (deconstructed into components of richness, evenness and disparity) may relate to different ecologically relevant aspects of the phenotype. Then, we systematically review the literature on chemodiversity to examine methodological practices, explore patterns of variability in diversity across different levels of biological organization, and investigate the functional role of this diversity in interactions between plants and other organisms. Overall, the reviewed literature suggests that high chemodiversity is often beneficial for plants, although a heterogeneity of methodological approaches partly limits what general conclusions can be drawn. Importantly, to support future research on this topic, we provide a framework with a decision tree facilitating choices on which measures of chemodiversity are best used in different contexts and outline key questions and avenues for future research. The pine wood nematode (Bursaphelenchus xylophilus), the pathogen of pine wilt disease (PWD), has caused enormous economic losses in Asian forests. Whether venom allergen proteins (VAPs) are involved in the accumulation of key defense substances in pine trees during the interaction between B. xylophilus and host trees, and their specific function as putative effectors secreted through stylets, has not been fully elucidated. In this study, the role of the BxVAP2 effector protein in the infection process was analyzed through bioinformatics and phylogenetic tree construction. The expression profile of BxVAP2 during infection was analyzed using qRT-PCR, and its expression under the stress of Pinus massoniana metabolites was examined. Toxicity assays were conducted through the Agrobacterium transient expression of BxVAP2 in Nicotiana benthamiana, and its subcellular localization was investigated. The results showed that BxVAP2 contains a CAP domain and shares close evolutionary relationships with venom allergen proteins from related species, such as Bursaphelenchus mucronatus, Aphelenchoides besseyi, Aphelenchoides fujianensis, and Meloidogyne graminicola. BxVAP2 was upregulated during the infection of P. massoniana, indicating that BxVAP2 is a key effector in the infection and colonization process of B. xylophilus and may play an important role during the rapid population growth phase. BxVAP2 responds to P. massoniana metabolites, where different concentrations of α-pinene suppressed its expression, while high concentrations of β-pinene promoted its expression. Subcellular localization revealed that BxVAP2 localizes to the cell membrane and nucleus. The transient expression of BxVAP2 in N. benthamiana induced programmed cell death and regulated pattern-triggered immunity marker genes. These findings suggest that BxVAP2 plays an important role in the interaction between B. xylophilus and its host, responding to terpene stress and triggering plant defense. [ABSTRACT FROM AUTHOR]

Background: α-Hairpinins are a family of antimicrobial peptides, promising antimicrobial agents, which includes only 12 currently revealed members with proven activity, although their real number is supposed to be much higher. α-Hairpinins are short peptides containing four cysteine residues arranged in a specific Cys-motif. These antimicrobial peptides (AMPs) have a characteristic helix−loop−helix structure with two disulfide bonds. Isolation of α-hairpinins by biochemical methods is cost- and labor-consuming, thus requiring reliable preliminary in silico prediction. Methods: In this study, we developed a special algorithm for the prediction of putative α-hairpinins on the basis of characteristic motifs with four (4C) and six (6C) cysteines deduced from translated plant transcriptome sequences. We integrated this algorithm into the Cysmotif searcher pipeline and then analyzed all transcriptomes available from the One Thousand Plant Transcriptomes project. Results: We predicted more than 2000 putative α-hairpinins belonging to various plant sources including algae, mosses, ferns, and true flowering plants. These data make α-hairpinins one of the ubiquitous antimicrobial peptides, being widespread among various plants. The largest numbers of α-hairpinins were revealed in the Papaveraceae family and in Papaver somniferum in particular. Conclusions: By analyzing the primary structure of α-hairpinins, we concluded that more predicted peptides with the 6C motif are likely to have potent antimicrobial activity in comparison to the ones possessing 4C motifs. In addition, we found 30 α-hairpinin precursors containing from two to eight Cys-rich modules. A striking similarity between some α-hairpinin modules belonging to diverse plants was revealed. These data allowed us to assume that the evolution of α-hairpinin precursors possibly involved changing the number of Cys-rich modules, leading to some missing middle and C-terminal modules, in particular. The use of plant growth-promoting rhizobacteria presents a promising addition to conventional mineral fertilizer use and an alternative strategy for sustainable agricultural crop production. However, genotypic variations in the plant host may result in variability of the beneficial effects from these plant–microbe interactions. This study examined growth promotion effects of commercial vegetable crop cultivars of tomato, cucumber and broccoli following application with five rhizosphere bacteria. Biochemical assays revealed that the bacterial strains used possess several nutrient acquisition traits that benefit plants, including nitrogen fixation, phosphate solubilization, biofilm formation, and indole-3-acetic acid (IAA) production. However, different host cultivars displayed genotype-specific responses from the inoculations, resulting in significant (p < 0.05) plant growth promotion in some cultivars but insignificant (p > 0.05) or no growth promotion in others. Gene expression profiling in tomato cultivars revealed that these cultivar-specific phenotypes are reflected in differential expressions of defense and nutrient acquisition genes, suggesting that plants can be categorized into "microbe-friendly" cultivars (with little or no defense responses against beneficial microbes) and "microbe-hostile" cultivars (with strong defense responses). These results validate the notion that "microbe-friendly" (positive interaction with rhizosphere microbes) should be considered an important trait in breeding programs when developing new cultivars which could result in improved crop yields. Introduction: Fusarium wilt, a devastating soil-borne fungal disease in flax (Linum usitatissimum), is caused by Fusarium oxysporum f. sp. lini, a hemibiotrophic plant pathogen that penetrates plant roots. There are several reports of the molecular response of L. usitatissimum to F. oxysporum f. sp. lini; however, comparisons of the effects of mutualistic and pathogenic fungi on plants are more limited. Methods: In this study, we have integrated phenotyping and RNA-Seq approaches to examine the response of flax to F. oxysporum f.sp. lini and to a mutualistic arbuscular mycorrhizal fungus (AMF) Rhizoglomus irregulare. R. irregulare is a common soil fungus and also widely used as a commercial inoculant to improve plant growth. We measured flax growth parameters after plant inoculation with each or both fungi, in comparison with non-inoculated control. We performed transcriptome analysis of root tissues collected at 9 and 14 days post-inoculation. Results: We identified several differentially expressed genes (DEGs) in response to pathogenic and mutualistic fungi. These included genes related to ethylene and salicylic acid biosynthesis, carbohydrate binding, oxidoreductases, and sugar transmembrane transporters. Genes related to calcium signaling, nutrient transport, lipid metabolism, cell wall, and polysaccharide-modifying were upregulated by R. irregulare; however, the same genes were down-regulated by F. oxysporum f. sp. lini when treated independently. In the combined treatment, genes related to cell wall modifications, hormone regulation and nutrient uptake were up-regulated. These results suggest that inoculation with R. irregulare reduced gene expression related to F. oxysporum f. sp. lini infection, leading to a reduced response to the pathogen. In response to AMF, flax prioritized mutualism-related gene expression over defense, reversing the growth inhibition caused by F. oxysporum f. sp.lini in the combined treatment. Discussion: This research provides insights into the protective effects of AMF, revealing the pre-symbiotic gene expression profile of flax in response to mutualism in comparison with pathogenicity. Background: Tomato (Solanum lycopersicum L.) production is severely threatened by bacterial wilt, caused by the phytopathogenic bacterium Ralstonia solanacearum. Recently, nano-enabled strategies have shown tremendous potential in crop disease management. Objectives: This study investigates the efficacy of biogenic nanoformulations (BNFs), comprising biogenic silica nanoparticles (SiNPs) and melatonin (MT), in controlling bacterial wilt in tomato. Methods: SiNPs were synthesized using Zizania latifolia leaves extract. Further, MT containing BNFs were synthesized through the one-pot approach. Nanomaterials were characterized using standard characterization techniques. Greenhouse disease assays were conducted to assess the impact of SiNPs and BNFs on tomato plant immunity and resistance to bacterial wilt. Results: The SiNPs and BNFs exhibited a spherical morphology, with particle sizes ranging from 13.02 nm to 22.33 nm for the SiNPs and 17.63 nm to 21.79 nm for the BNFs, indicating a relatively uniform size distribution and consistent shape across both materials. Greenhouse experiments revealed that soil application of BNFs outperformed SiNPs, significantly enhancing plant immunity and reducing bacterial wilt incidence by 78.29% in tomato plants by maintaining oxidative stress homeostasis via increasing the activities of antioxidant enzymes such as superoxide dismutase (31.81%), peroxidase (32.9%), catalase (32.65%), and ascorbate peroxidase (47.37%) compared to untreated infected plants. Additionally, BNFs induced disease resistance by enhancing the production of salicylic acid and activating defense-related genes (e.g., SlPAL1, SlICS1, SlNPR1, SlEDS, SlPD4, and SlSARD1) involved in phytohormones signaling in infected tomato plants. High-throughput 16 S rRNA sequencing revealed that BNFs promoted growth of beneficial rhizosphere bacteria (Gemmatimonadaceae, Ramlibacter, Microscillaceae, Anaerolineaceae, Chloroplast and Phormidium) in both healthy and diseased plants, while suppressing R. solanacearum abundance in infected plants. Conclusion: Overall, these findings suggest that BNFs offer a more promising and sustainable approach for managing bacterial wilt disease in tomato plants. [ABSTRACT FROM AUTHOR]

Trade-offs are a fundamental concept in evolutionary biology because they are thought to explain much of nature's biological diversity, from variation in life-histories to differences in metabolism. Despite the predicted importance of trade-offs, they are notoriously difficult to detect. Here we contribute to the existing rich theoretical literature on trade-offs by examining how the shape of the distribution of resources or metabolites acquired in an allocation pathway influences the strength of trade-offs between traits. We further explore how variation in resource distribution interacts with two aspects of pathway complexity (i.e., the number of branches and hierarchical structure) affects tradeoffs. We simulate variation in the shape of the distribution of a resource by sampling 106 individuals from a beta distribution with varying parameters to alter the resource shape. In a simple "Y-model" allocation of resources to two traits, any variation in a resource leads to slopes less than −1, with left skewed and symmetrical distributions leading to negative relationships between traits, and highly right skewed distributions associated with positive relationships between traits. Adding more branches further weakens negative and positive relationships between traits, and the hierarchical structure of pathways typically weakens relationships between traits, although in some contexts hierarchical complexity can strengthen positive relationships between traits. Our results further illuminate how variation in the acquisition and allocation of resources, and particularly the shape of a resource distribution and how it interacts with pathway complexity, makes its challenging to detect trade-offs. Maize originated in southern Mexico and various hybrid varieties have been bred during domestication. All maize tissues are rich in specialized plant metabolites (SPMs), which allow the plants to resist the stresses of herbivores and pathogens or environmental factors. To date, a total of 95 terpenoids, 91 phenolics, 31 alkaloids, and 6 other types of compounds have been identified from maize. Certain volatile sesquiterpenes released by maize plants attract the natural enemies of maize herbivores and provide an indirect defensive function. Kauralexins and dolabralexins are the most abundant diterpenoids in maize and are known to regulate and stabilize the maize rhizosphere microbial community. Benzoxazinoids and benzoxazolinones are the main alkaloids in maize and are found in maize plants at the highest concentrations at the seedling stage. These two kinds of alkaloids directly resist herbivory and pathogenic infection. Phenolics enhance the cross‐links between maize cell walls. Meanwhile, SPMs also regulate plant–plant relationships. In conclusion, SPMs in maize show a large diversity of chemical structures and broad‐spectrum biological activities. We use these to provide ideas and information to enable the improvement of maize resistances through breeding and to promote the rapid development of the maize industry. [ABSTRACT FROM AUTHOR]

The co‐evolutionary arms race between herbivores and plants forces plants to evolve protection strategies that reduce the palatability of the plant modules attacked by the herbivores. These characteristics of traits have consequences for both the survival of plant individuals and the composition of plant communities. Thus, correlating traits of for instance leaves with herbivory is an important step toward understanding the dynamics of plant populations and communities. Traits can either be measured using conventional lab methods or recently developed spectral sensing techniques. We examined whether leaf traits of trees are related to herbivory in a multispecies approach. Furthermore, we explored whether leaf traits characterized by spectral sensing provide similar relations to herbivory as lab‐based leaf traits. We established nine 1‐ha square plots evenly distributed over three different forest types in Ecuadorian tropical montane rainforests where we estimated herbivory as the leaf area loss (in square centimeters) of 20 (±5) leaves sampled from the canopies of 380 tree individuals belonging to 51 tree species (7 ± 1 individuals/species) using lab‐ and spectral‐sensing‐based methods. For each methodological approach, we ran 100 linear mixed‐effects models with all respective leaf traits as predictor and herbivory as response variables for data subsets containing one randomly selected tree individual of each species to estimate the range of the regression coefficients for each trait. Automated stepwise backward selections determined the frequency of each trait having an important influence on herbivory. We found no clear relations between leaf traits and herbivory for neither lab‐ nor spectral‐sensing‐based traits. A nested variance component analysis demonstrated that the observed variability was mainly due to the variation in trait concentrations between tree individuals of a species. Our results suggest that snapshot data lead to a mismatch between herbivory and the concentrations of traits during the peak of herbivory. Another explanation could be that environmental conditions or processes along the food web are more important in structuring herbivory than leaf traits. [ABSTRACT FROM AUTHOR]

Chitosan is a natural elicitor, used for stimulating plant growth and inducing plant defense. However, due to difficulty in monitoring root growth and activity, the effects of chitosan treatment on plant root systems have been less studied as compared to plant shoot parts that include leaves, seeds, and fruits. This results in an indefinite outcome of the benefits of chitosan on plant roots. Therefore, this review aims to evaluate the effects of chitosan treatment on root growth and defense responses based on current evidence. Interestingly, many studies have demonstrated that chitosan can induce plant root defense systems, yet conversely inhibiting root growth. The effects were most clearly observed from studies using liquid or solid media as substrates, while the results from the studies using soil were inconclusive and require additional investigation to observe the effects of environmental factors. In addition, root chitosan treatment showed variable effects on shoot growth, where low chitosan concentrations tend to promote shoot growth, but high chitosan concentrations may affect shoot development. Additionally, this review discusses the potential methods of chitosan application onto plant roots. Water insolubility of chitosan is likely a major issue for root treatment. Chitosan can be dissolved in acids, but this could induce acidity stress in plant roots. Modified versions of chitosan, such as chitosan nanoparticles, carboxylated chitosan, and graft chitosan copolymers have been developed to improve solubility and functionality. Chitosan nanoparticles can also be used to encapsulate other biocontrol agents to augment biological effects on plant defense. In conclusion, root chitosan treatment could help to promote plant defense and prevent root infections, abating the uses of chemical fungicides in agriculture. However, further research is required to monitor the impact of root chitosan treatment on long-term plant growth in order to gain multifaceted information to maximize the effectiveness of root chitosan application. Abstract The study focuses on the in silico analysis of cysteine-rich secretory proteins and PR1-like (CAP) genes in finger lime (Citrus australasica), a citrus species known for its tolerance to Huanglongbing (HLB). We identified several PR1-like genes, all belonging to the CRISP family within the CAP superfamily. Of them, CaCAP2 transcript levels increased by over 300-fold in the finger lime compared to ‘Valencia' sweet orange upon infection with ‘Candidatus Liberibacter asiaticus' (CaLas). Localization studies using an EGFP fusion showed that the CAP2 protein is predominantly located in the nucleus, extracellular and plasma membrane. The study also examined CAP2 transcript levels in response to cold, drought stress, and salicylic acid application. Despite environmental stress causing apparent damage, CAP genes seem to play a significant role in managing both biotic and abiotic stresses. Analysis of CAP2 gene promoters from finger lime and sweet orange revealed 95.33% sequence identity, with variations in transcription factor-binding sites and cis-acting elements such as Stress Response Element (STRE: AGGGG), which might influence the differential expression of CAP2 between the two species. Additionally, expressing the finger lime-derived CaCAP2 gene in transgenic Nicotiana tabacum induced a strong defense response against Pseudomonas syringae pv. Tabaci., underscoring the CAP gene's crucial role in plant defense mechanisms against bacterial pathogens.

Glyphosate, the active ingredient of glyphosate-based herbicides (GBHs), controls the growth of weeds by inhibiting the shikimate pathway, thereby interrupting amino acid biosynthesis in plants. However, several microbes have the shikimate pathway, and the effect of glyphosate on these non-target organisms is ignored. The action of GBHs is further complicated when used with other agrochemicals, such as phosphate fertilizers, often varying their mode of action depending on soil type or plant species. To address the impact of GBHs and phosphate fertilizers, we simulated agricultural application of GBHs and phosphate fertilizers in a field study, investigating the composition of endophytic microbial communities and correlation of phytohormone concentrations with the microbial diversity of potato (Solanum tuberosum). In leaves, glyphosate residues in soil from GBH treatment alone and in combination with phosphate significantly shifted the bacterial community, whereas phosphate alone and in combination with glyphosate significantly altered the composition of the fungal community. There were no significant changes in microbial communities in roots and tubers. Plants treated with GBHs showed higher ratios of potentially glyphosate-resistant bacteria, with Xanthomonadaceae and Moraxellaceae being more abundant. Additionally, phytohormone concentrations showed various correlations with bacterial and fungal diversity in different treatments. The study highlights the impact of GBH residues in soil, particularly in combination with phosphate fertilizers, on the composition of plant-associated microbial communities. Together with changes in phytohormone concentrations, plant health may be affected. Future studies could provide insights into whether these agrochemicals influence the plant microbiome, leading to changes in phytohormones or vice versa. The purpose of this article is to give a comprehensive overview of the fascinating world of food webs, where plants play a key role. To highlight the importance of studying this area, understanding the issues involved, and applying the knowledge gained with respect for nature to agroecosystems including important permanent crops including orchards. Plants produce chemicals called secondary metabolites that play an indispensable role in their interactions with their environment. Plants use these substances to attract pollinators, defend themselves against herbivorous arthropods, attract predators or parasitoids of herbivores and communicate with each other. Recognizing these "SMALL" interactions can have BIG implications useful in the fields of ecology, medicine, and agriculture.

Plants have evolved direct and indirect defences against herbivores, which may come at a cost to other plant functions. Many plants can uptake and deposit large amounts of silicon within plant tissue, creating structures which can reduce herbivore performance. Silicon uptake can increase plant defenses against herbivores, but it has also been shown to trade-off with defensive phenolic compounds due to interference with jasmonic acid (JA) signaling. Additionally, plants can recruit predacious insects with extrafloral nectar (EFN), a sugar secretion not involved in pollination. It is currently unclear whether silicon uptake reduces other putative defences associated with the JA pathway, like EFN production. We used faba bean (Vicia faba) to identify potential trade-offs between silicon accumulation, phenolic content, and EFN production. We grew four genotypes of faba bean that varied in tannin content in control soil, or soil supplemented with silicon. After five weeks of growth, we exposed plants to either a buffer or methyl jasmonate (MeJA) solution to simulate an herbivory response. We measured EFN production at 24 and 48 hours after treatment, and harvested leaves to quantify silicon and phenolic content. We found silicon supplementation, but not MeJA treatment, increased foliar silicon concentration. Silicon supplementation did not affect foliar phenolic content or EFN sugar content. Silicon concentration (ppm) and MeJA treatment did not decrease foliar phenolic content or EFN sugar content. However, we found an interaction between silicon concentration (ppm) and MeJA treatment with EFN sugar content: across MeJA-treated plants, we detected a positive association between foliar silicon concentration and the amount of sugar (mg) in EFN. This study is the first to show MeJA can interact with leaf silicon concentration to alter EFN response, with the potential for cascading effects on other trophic levels.

Serine protease inhibitors (SPIs) are ubiquitously distributed and structurally well-conserved proteins that interfere with serine protease catalysis and function as intrinsic defense molecules for plants. Potato inhibitor type II (Pin-II) SPIs found mainly in Solanaceous species are important in minimizing injury caused by pathogen infection and insect herbivory. Eggplant (Solanum melongena L.), an important vegetable crop, is severely damaged by insect pests and diseases. Multiple Pin-II SPI proteins had been identified in tomato, potato, tobacco, and pepper, with only one in eggplant. This study conducted a bioinformatics analysis and identified 15 Pin-2SPI genes in cultivated eggplant, herein named as SmePin-2SPI.1 to SmePin-2SPI.15. Phylogenetic and syntenic analyses confirmed the homology of these genes with Pin-II SPIs from other Solanaceae and non-Solanaceae plant species based on the conserved protease inhibitor 2 (PF02428; Prot-Inhib 2) domains and coding regions, indicating conservation within the Solanaceae. Functional annotation indicates that these genes are involved in the negative regulation of serine peptidase activity and plant defense. Major cis-acting regulatory elements are associated with plant hormones and stress responses. An in-silico analysis of transcriptomic profiles for constitutive and induced expression indicates that SmePin-2SPI genes are highly expressed in reproductive organs and exhibit strong reactions to biotic stress, particularly bacterial wilt infection. SmePin-2SPI.1 is highly expressed in most plant organs and shows differential expression in response to various stress treatments. This study also demonstrated sequence homology of Pin-2SPI genes using PCR amplification for three species of eggplant within the Solanum clade, the cultivated species S. melongena, S. aethiopicum, and the wild species S. mammosum. Abstract Background Rivea ornata, a rare species from the morning glory family, exhibits uncommon characteristics compared to other typical morning glories, including nocturnal flowers that fit the classic moth pollination syndrome. However, the accuracy of its predicted pollination syndrome and its mating system have never been assessed. Additionally, R. ornata flowers attract not only pollinators but also florivores, potentially reducing plant reproductive success. Therefore, this study examined two populations of R. ornata in Thailand and assessed traits related to pollinator attraction and reward, determined its mating system, identified floral visitors and effective pollinators, and investigated the effect of florivory on reproductive success. Results Rivea ornata is highly fertile but self-incompatible and an obligate outcrosser, rendering it highly dependent on pollinators. Lepidopterans, particularly nocturnal hawk moths, were found to account for a significant proportion of all visits and were the sole effective pollinators of this plant species, in correspondence with its predicted pollination syndrome. Surprisingly, florivory did not significantly reduce reproductive success. This phenomenon may be explained by the strategies employed by R. ornata, which align with the optimal defense hypothesis and functional trade-offs. Specifically, R. ornata appears to invest resources in defending key floral structures while, simultaneously, guard ants are conspicuously absent from flowers, resulting in some florivore damage to non-vital floral organs but ensuring that pollinators are not deterred by ants and thus maintaining high pollinator visitation rates. Conclusions Our findings indicate that reproduction-related traits in R. ornata, including those involved in pollinator attraction and reward and florivore defense, are highly effective and work in concert to maximize plant reproductive success. Therefore, a main risk that R. ornata faces is the decline or disappearance of hawk moths and other lepidopterans given its extreme specialization and high dependence on pollinators, and conservation efforts should include habitat protection for both R. ornata and its pollinators.

Endophytic fungi are microorganisms known as plant-associated fungi and are typically found asymptomatically inside plant tissue. By directly generating secondary metabolites, endophytic fungi help their host plants grow and become more resistant to plant diseases. Furthermore, they can also biosynthesize bioactive compounds used for antimicrobials, which were previously believed to be produced only by the host plant. Medicinal plants as host plants for endophytic fungi which produce secondary metabolites and their ability as biological agents against pathogens. Calabash tree is a medicinal plant that contains active compounds that function as antifungal and antibacterial. This study aims to determine the antagonistic activity of endophytic fungi from calabash tree leaves against F. oxysporum Fos, the pathogen that causes fusarium wilt in shallot plants. Twelve endophytic fungi were obtained from isolation from the leaves of the calabash tree, namely EnM6C22, EnM11P2, EnM6A2, EnM5H21, EnM10H22, EnM12P, EnM11P, EnM111, EnM5H22, EnM9Pt22, EnM5H1 and EnM5K. Five of the twelve endophytic fungi of calabash tree leaves have been identified, as Fusarium sp., Rhizoctonia sp., Pestalotia sp., Colletotrichum sp., and Bipolaris sp. with inhibition percentages of 36.43%, 32.19%, 25.41%, 32.19%, and 27.53% respectively at 7 days after inoculation.

MicroRNAs (miRNAs) are a class of small non-coding RNAs, typically 20-25 nucleotides in length, that play a crucial role in regulating gene expression post-transcriptionally. They are involved in various biological processes such as plant growth, development, stress response, and hormone signaling pathways. Plants interact with microbes through multiple mechanisms, including mutually beneficial symbiotic relationships and complex defense strategies against pathogen invasions. These defense strategies encompass physical barriers, biochemical defenses, signal recognition and transduction, as well as systemic acquired resistance. MiRNAs play a central role in regulating the plant's innate immune response, activating or suppressing the transcription of specific genes that are directly involved in the plant's defense mechanisms against pathogens. Notably, miRNAs respond to pathogen attacks by modulating the balance of plant hormones such as salicylic acid, jasmonic acid, and ethylene, which are key in activating plant defense mechanisms. Moreover, miRNAs can cross boundaries into fungal and bacterial cells, performing cross-kingdom RNA silencing that enhances the plant's disease resistance. Despite the complex and diverse roles of miRNAs in plant defense, further research into their function in plant-pathogen interactions is essential. Introduction: Salicylic acid (SA) is a phenolic compound widely found in plants. It plays a key role in exerting plant disease resistance. Panax vietnamensis Ha & Grushv., a valuable medicinal plant, contains high levels of phenolic compounds, which contribute significantly to the resilience of the plant against stress. However, the precise role of SA in regulating the synthesis of secondary metabolites in P.vietnamensis remains elusive. Methods: Two-year-old P. vietnamensis seedlings were treated with exogenous SA. We systematically assessed the changes in the physiological parameters of SA-treated P. vietnamensis leaves, employing transcriptome and metabolome analyses to elucidate the underlying mechanisms. Results: Our results revealed a significant improvement of the plant's antioxidant capacity at 6 h post-treatment. Furthermore, exogenous SA treatment promoted the biosynthesis of lignin and flavonoids such as rutin, coumarin, and cyanidin. In addition, it increased the levels of endogenous SA and jasmonic acid (JA), promoting the disease resistance of the plants. Thus, SA pretreatment enhanced the defense of P. vietnamensis against pathogens. Conclusions: Our study provided novel insights into the potential molecular mechanisms underlying SA-mediated biosynthesis of secondary metabolites. Pathogenesis‐related proteins (PR), including osmotins, play a vital role in plant defense, being activated in response to diverse biotic and abiotic stresses. Despite their significance, the mechanistic insights into the role of osmotins in plant defense have not been extensively explored. The present study explores the cloning and characterization of the osmotin gene (WsOsm) from Withania somnifera, aiming to illuminate its role in plant defense mechanisms. Quantitative real‐time PCR analysis revealed significant induction of WsOsm in response to various phytohormones e.g. abscisic acid, salicylic acid, methyl jasmonate, brassinosteroids, and ethrel, as well as biotic and abiotic stresses like heat, cold, salt, and drought. To further elucidate WsOsm's functional role, we overexpressed the gene in Nicotiana tabacum, resulting in heightened resistance against the Alternaria solani pathogen. Additionally, we observed enhancements in shoot length, root length, and root biomass in the transgenic tobacco plants compared to wild plants. Notably, the WsOsm‐ overexpressing seedlings demonstrated improved salt and drought stress tolerance, particularly at the seedling stage. Confocal histological analysis of H2O2 and biochemical studies of antioxidant enzyme activities revealed higher levels in the WsOsm overexpressing lines, indicating enhanced antioxidant defense. Furthermore, a pull‐down assay and mass spectrometry analysis revealed a potential interaction between WsOsm and defensin, a known antifungal PR protein (WsDF). This suggests a novel role of WsOsm in mediating plant defense responses by interacting with other PR proteins. Phenolic compounds (phenolics) are secondary metabolites ubiquitous across plants. The earliest phenolics are linked to plants' successful transition from an aquatic to a terrestrial environment, serving as protection against damaging ultraviolet (UV) radiation, and as antioxidants to reduce oxidative stress in an atmosphere with an increasingly high O2:CO2 ratio. In modern plants, phenolics are best known for the defense against fungal and bacterial pathogens and as antifeedants that deter herbivory. Phenolics also play a role in seed dormancy, delaying germination, and lengthening viability in the seed bank. Many plants' seeds are endozoochorous – dispersed by animals, like birds, who eat and later excrete the seeds. Plants send visual signals to attract birds with UV‐sensitive (UVS) vision for pollination and seed dispersal. As fruits ripen, antioxidant activity and phenolic content decrease. The waxy cuticle of fruits increases in UV reflection as phenolic rings, which absorb UV light, degrade. The UV contrast that birds detect may act as an honest signal, indicating nutritional changes in the fruit. However, there is little evidence to support the evolution of UV coloration during ripening being driven by frugivore selection. Plants resist the invasion of plant-parasitic nematodes (PPNs) by releasing root chemical substances, activating immune responses, and utilizing plant resistance (R) genes. A balance of interaction is formed between plants and PPNs, including multilayer communications and signal exchanges. The signal network involved in plant–nematode competition includes attraction and repellency of root exudates, perception of nematode-associated signals, activation of plant signals, interference of PPNs with plant defense, and interplay of plant systemic signals. Plant-parasitic nematodes (PPNs) are widely distributed and highly adaptable. To evade the invasion and infection of PPNs, plants initiate a series of defense responses. In turn, PPNs secrete effectors into the host tissues to suppress plant defense. In this ongoing battle between PPNs and plants, complex signal transduction processes are typically involved. This article aims to review the plant signaling network involved in host perception by the nematode, nematode perception, and downstream activation of plant defense signaling and how nematodes attempt to interfere with this network. Our goal is to establish a foundation for elucidating the signaling and regulatory mechanisms of plant–nematode interactions, and to provide insights and tools for developing PPN-resistant crops and technologies. Bipolaris maydis is a prominent pathogen causing maydis leaf blight disease (MLB) in warm, humid regions worldwide. Structural components of plants, particularly stomata, play a crucial role in defending against pathogen invasion. In this study, we conducted a comprehensive investigation of stomatal interactions at the histopathological and molecular levels in various cultivated maize genotypes from Punjab state of India. Bipolaris maydis was isolated, purified, and inoculated onto six maize genotypes. Leaf samples were collected 48 h post-inoculation, representing the disease phase. Stomatal aperture bioassays and scanning electron microscopy (SEM) were performed on infected and non-infected samples. SEM analysis revealed distinct morphological variations, including stomatal aperture patterns and MLB pathogen invasion routes through stomata. Stomatal diameter varied among genotypes, and mycelium presence on the leaf surface was observed, with varying degrees of invasion through stomata. Genotypes exhibiting greater field symptom visibility displayed higher mycelium abundance and invasion. Additionally, high-performance liquid chromatography (HPLC) was employed to quantify abscisic acid (ABA) content in maize leaves during the Bipolaris maydis -maize interaction. This analysis aimed to elucidate the relationship between ABA and biotic stress, shedding light on its role in defense responses against the pathogen. Overall, our study enhances the understanding of stomatal interactions, surface morphology, and ABA dynamics in maize- Bipolaris maydis interactions. These findings have implications for breeding programs seeking to develop resistant maize varieties and contribute to the overall management of MLB disease. This study explores the impact of exogenous salicylic acid (SA) alongside conventional treatment by farmers providing positive (Mancozeb 80 % WP) and negative (water) controls on rice plants (Oryza sativa L.), focusing on antioxidant enzyme activities, phytohormone levels, disease resistance, and yield components under greenhouse and field conditions. In greenhouse assays, SA application significantly enhanced the activities of peroxidase (POX), polyphenol oxidase (PPO), catalase (CAT), and superoxide dismutase (SOD) within 12–24 h post-inoculation (hpi) with Magnaporthe oryzae. Additionally, SA-treated plants showed higher levels of endogenous SA and indole-3-acetic acid (IAA) within 24 hpi compared to the controls. In terms of disease resistance, SA-treated plants exhibited a reduced severity of rice blast under greenhouse conditions, with a significant decrease in disease symptoms compared to negative control treatment. The field study was extended over three consecutive crop seasons during 2021–2023, further examining the efficacy of SA in regular agricultural practice settings. The SA treatment consistently led to a reduction in rice blast disease severity across all three seasons. Yield-related parameters such as plant height, the number of tillers and panicles per hill, grains per panicle, and 1000-grain weight all showed improvements under SA treatment compared to both positive and negative control treatments. Specifically, SA-treated plants yielded higher grain outputs in all three crop seasons, underscoring the potential of SA as a growth enhancer and as a protective agent against rice blast disease under both controlled and field conditions. These findings state the broad-spectrum benefits of SA application in rice cultivation, highlighting its role not only in bolstering plant defense mechanisms and growth under greenhouse conditions but also in enhancing yield and disease resistance in field settings across multiple crop cycles. Tithonia diversifolia has the potential for use as a reservoir for entomophages in pest control, as it attracts and preserves natural enemies before migrating to the target crop, where a high incidence of pests occurs. For example, in tomato crop production one of the limiting factors, under field condition, is the incidence of phytophagous insects. We determined the effects of Tithonia diversifolia on species assemblage structure of natural enemies and pest insects (abundance and diversity), plant growth (leaf area, stem length and diameter, number of leaves, flowers and fruits, and fruits weight/per plant), plant defense (leaf thickness and hardness) and fruit colouration on tomato plants in open conditions. Through direct and indirect samplings, a total of 456 insects were collected, comprising 76 natural enemies, 269 pests and 111 belonging other guilds. Length, stem diameter, number of flowers, leaf thickness, and hardness increased, while the number of leaves decreased. T. diversifolia plants increased natural enemies but not pest abundance in tomato plants, demonstrating that this plant can alter the structure of the insect community associated with tomato. In the search for environmentally friendly options to manage plant pests and enhance productivity with fewer external inputs in agriculture, the screening of landraces has gained importance. The aims of this study were to characterize functional leaf traits in maize landraces, including morphological traits such as leaf toughness, thickness, specific leaf area (SLA), and number of trichomes, as well as physiological leaf traits such as photosynthesis, transpiration rate, and stomatal conductance. We aimed to determine the association of these traits with leaf damage caused by Spodoptera frugiperda across 24 maize landraces. Relationships of dependence among leaf traits and leaf damage were determined using principal component analysis, multiple regression analysis and sequential path analyses. All morphological leaf traits showed differences among maize landraces mainly at 20 and 30 days after emerge (DAE) whereas at 40 DAE the differences decreased. By the contrast, physiological leaf traits had high variation at 40 DAE among maize landraces. We found a negative dependency of principal component conformed by photosynthesis, transpiration rate, and stomatal conductance over leaf damage. Based on the level of leaf damage, we categorized maize landraces into two groups with low and high susceptibility. Surprisingly, we observed that in the former group, physiological leaf traits affected the number of trichomes, whereas in the latter group, they influenced leaf thickness. This finding highlights the necessity of detailed assessments of foliage properties to understand the impact of S. frugiperda infestations. Plants and insects have co-existed for almost 400 million years and their interactions can be beneficial or harmful, thus reflecting their intricate co-evolutionary dynamics. Many herbivorous arthropods cause tremendous crop loss, impacting the agro-economy worldwide. Plants possess an arsenal of chemical defenses that comprise diverse secondary metabolites that help protect against harmful herbivorous arthropods. In response, the strategies that herbivores use to cope with plant defenses can be behavioral, or molecular and/or biochemical of which salivary secretions are a key determinant. Insect salivary secretions/oral secretions (OSs) play a crucial role in plant immunity as they contain several biologically active elicitors and effector proteins that modulate plants' defense responses. Using this oral secretion cocktail, insects overcome plant natural defenses to allow successful feeding. However, a lack of knowledge of the nature of the signals present in oral secretion cocktails has resulted in reduced mechanistic knowledge of their cellular perception. In this review, we discuss the latest knowledge on herbivore oral secretion derived elicitors and effectors and various mechanisms involved in plant defense modulation. Identification of novel herbivore-released molecules and their plant targets should pave the way for understanding the intricate strategies employed by both herbivorous arthropods and plants in their interactions. Rodents can cause considerable changes in plant community composition. However, relationships between shifts in species dominance and plant functional traits caused by rodents have seldom been investigated, especially for belowground functional traits. In this study, a set of enclosures was constructed to analyze the effects of 10 years of Brandt's voles' activities on the defense strategies and dominant position changes of three gramineous plants (Leymus chinensis, Stipa krylovii, and Cleistogenes squarrosa) in Inner Mongolia. Here, we measured the dominance, biomass, and fourteen functional traits of three plants. The effects of Brandt's voles on dominance, biomass, and functional traits were analyzed, and then we explored the effect of functional traits on plant dominance by using the structural equation model. Results showed that long‐term feeding by Brandt's voles resulted in a significant decrease in the dominance of L. chinensis and S. krylovii, whereas C. squarrosa was positively affected. The belowground biomass of L. chinensis and S. krylovii was higher in the vole treatment, which showed that they were increasing their escape characteristics. The leaf thickness of L. chinensis and the leaf C:N ratio of S. krylovii significantly increased, while the specific leaf area of C. squarrosa significantly decreased. All three gramineous showed increased resistance traits in response to Brandt's voles, which positively affected their dominance. Tolerance‐related traits of S. krylovii significantly increased, with the increasing growth rate of root length contributing to enhancing its dominance. We highlight that selective feeding by rodents led to the selection of different defense strategies by three gramineous plants, and that changes in biomass allocation and functional traits in the different species affected plant dominance, driving changes in the plant communities. Dynamic climate changes pose a significant challenge for plants to cope with numerous abiotic and biotic stressors of increasing intensity. Plants have evolved a variety of biochemical and molecular defense mechanisms involved in overcoming stressful conditions. Under environmental stress, plants generate elevated amounts of reactive oxygen species (ROS) and, subsequently, modulate the activity of the antioxidative enzymes. In addition, an increase in the biosynthesis of important plant compounds such as anthocyanins, lignin, isoflavonoids, as well as a wide range of low molecular weight stress-related proteins (e.g., dehydrins, cyclotides, heat shock proteins and pathogenesis-related proteins), was evidenced. The induced expression of these proteins improves the survival rate of plants under unfavorable environmental stimuli and enhances their adaptation to sequentially interacting stressors. Importantly, the plant defense proteins may also have potential for use in medical applications and agriculture (e.g., biopesticides). Therefore, it is important to gain a more thorough understanding of the complex biological functions of the plant defense proteins. It will help to devise new cultivation strategies, including the development of genotypes characterized by better adaptations to adverse environmental conditions.
