Abstract

Several crustaceans including shrimps change the amount of specific free amino acids to regulate the osmotic pressure in their bodies. Kuruma shrimp Penaeus japonicus also increases the concentration of alanine (Ala) in the abdominal muscle following the increase of environmental salinity. In the present study, to elucidate the mechanisms of changes in Ala accumulation of kuruma shrimp depending on salinity, we cloned the gene encoding alanine aminotransferase (ALT), an enzyme involved in Ala biosynthesis, and examined its expression profile. It was found that the full‐length kuruma shrimp ALT1 cDNA consisted of 3,301 bp, encoding 514 amino acids, and that all amino acid residues important for ALT activity were conserved. Phylogenetic analysis also indicated that the ALT gene cloned in this study was classified as ALT1. Moreover, we examined the expression levels of the ALT1 gene in the abdominal muscle and the hepatopancreas of kuruma shrimp acclimated at 17‰, 34‰, and 40‰ salinities, resulting that the mRNA levels of the ALT1 genes in both tissues of the shrimp acclimated at 40‰ were significantly higher than those at 17‰ for 12 h (p < 0.05). The mRNA levels of the ALT1 gene in the abdominal muscle of the shrimp acclimated for more than 24 h tended to increase following the increase of environmental salinity. These results indicate that ALT1 is responsible for the increase of free Ala concentration in the abdominal muscle of kuruma shrimp to regulate osmotic pressure at high salinity. 4. Molecular ecology and evolution of an acorn barnacle, Balanus improvisus (Darwin)

Abstract

The reproductive biology and population dynamics of B. improvisus were investigated. This barnacle species prefers to settle close to its conspecifics (0.4-0.8 cm), but there were more isolated individuals of B. improvisus (up to 3 cm apart) found compared with E. modestus (not more than 1.6 cm apart). This could be the result of competition for space between these two barnacles or a consequence of the ability of B. improvisus to self-fertilise, which allows the species to reproduce without the requirement of neighbouring individuals. In southern England, the breeding season of B. improvisus was between March and October, and the putative period of planktonic larvae release was from July to August. Settlement of this barnacle occurred between July and September. The population genetic structure of this barnacle was investigated both at the local- (within the U.K. and Baltic) and broader- (between the U.K. and the Baltic) levels using mtDNA PCR-RFLP analysis of the COI-COIII genes. The results of the local-scale study suggest that the populations around the British Isles are isolated and that the level of gene flow between the estuaries is low. In contrast, the level of gene flow between the populations within the Baltic is high. This indicates that the planktonic larvae of this barnacle are capable of dispersing throughout the Baltic. In the broad-scale study, there was no significant genetic differentiation between the British and Baltic populations. This suggests that the level of gene flow between the two regions is high. Combination of the results from both local- and board-scale studies indicated that the populations of this barnacle species in the two regions could be derived from the same gene pool.

17. Phylogenetic Relationships among Hoplobatrachus rugulosus in Thailand as Inferred from Mitochondrial DNA Sequences of the Cytochrome-b Gene (Amphibia, Anura, Dicroglossidae).

Abstract

A fragment (564 bp) of the mitochondrial cytochrome-b (Cyt-b) gene was studied for 73 individual rice field frogs (Hoplobatrachus rugulosus) from 18 geographical locations (populations) within Thailand. Sequence analysis revealed the presence of 12 haplotypes, with five haplotypes being represented in two or more populations, and the other seven being population-distinct haplotypes. Phylogenetic analysis by maximum parsimony, maximum likelihood, and neighbor joining analyses all placed the 12 haplotypes into two distinct and well-separated clades with high bootstrap support, reflecting the high sequence divergences between the clades (25.3-32.3%). The mountain ranges and the Isthmus of Kra are likely to have played important roles in hindering gene flow among H. rugulosus populations in Thailand. From the sequence divergence values, the two clades of H. rugulosus can be classified into two distinct species, and therefore, the strains of H. rugulosus bred in farm stocks should be restricted to a population of one clade so as to avoid cross breeding between the two clades.
