Soil sealing is the permanent covering of the land surface by buildings, infrastructures or any impermeable artificial material. Beside the loss of fertile soils with a direct impact on food security, soil sealing modifies the hydrological cycle. This can cause an increased flooding risk, due to urban development in potential risk areas and to the increased volumes of runoff. This work estimates the increase of runoff due to sealing following urbanization and land take in the plain of Emilia Romagna (Italy), using the Green and Ampt infiltration model for two rainfall return periods (20 and 200 years) in two different years, 1976 and 2008. To this goal a hydropedological approach was adopted in order to characterize soil hydraulic properties via locally calibrated pedotransfer functions (PTF). PTF inputs were estimated via sequential Gaussian simulations coupled with a simple kriging with varying local means, taking into account soil type and dominant land use. Results show that in the study area an average increment of 8.4% in sealed areas due to urbanization and sprawl induces an average increment in surface runoff equal to 3.5 and 2.7% respectively for 20 and 200-years return periods, with a maximum > 20% for highly sealed coast areas.

In this contribution, new data concerning algae, bryophytes, fungi and lichens of the Italian flora are presented. It includes new records and confirmations for the algal genera Chara and Nitella, the bryophyte genera Brachythecium, Didymodon, Fissidens, Physcomitrium, and Riccia, the fungal genera Biatoropsis, Cantharellus, Coprinellus, Dacrymyces, Inosperma, Nigropuncta, Urocystis, and Xanthoriicola, and the lichen genera Arthonia, Bellemerea, Circinaria, Lecania, Lecanora, Lecidella, Mycobilimbia, Naetrocymbe, Parmelia, Peltigera, Porpidia, Scytinium, and Usnea.

Abstract Deficiency of adenosine deaminase 2 (DADA2) is an inborn error of immunity caused by loss-of-function mutations in the adenosine deaminase 2 (ADA2) gene. Clinical manifestations of DADA2 include vasculopathy and immuno-hematological abnormalities, culminating in bone marrow failure. A major gap exists in our knowledge of the regulatory functions of ADA2 during inflammation and hematopoiesis, mainly due to the absence of an ADA2 orthologue in rodents. Exploring these mechanisms is essential for understanding disease pathology and developing new treatments. Zebrafish possess two ADA2 orthologues, cecr1a and cecr1b, with the latter showing functional conservation with human ADA2. We establish a cecr1b-loss-of-function zebrafish model that recapitulates the immuno-hematological and vascular manifestations observed in humans. Loss of Cecr1b disrupts hematopoietic stem cell specification, resulting in defective hematopoiesis. This defect is caused by induced inflammation in the vascular endothelium. Blocking inflammation, pharmacological modulation of the A2r pathway, or the administration of the recombinant human ADA2 corrects these defects, providing insights into the mechanistic link between ADA2 deficiency, inflammation and immuno-hematological abnormalities. Our findings open up potential therapeutic avenues for DADA2 patients.

Abstract We present a European Union (EU)-wide dataset of estimated quantities of active substances of plant protection product applied on crops (also called “emissions”). Our estimates are derived from data reported by eight EU countries and extrapolated to encompass all EU regions using regression models. These models consider both climate and agricultural land use data. This allows us to spatially represent pesticide use at NUTS Level 3 of the European statistical mapping units, and within various agricultural land cover classes in each region. We compare our estimates with aggregated data provided by EUROSTAT and with independent, detailed data for the United Kingdom, highlighting an error typically within one order of magnitude. Our estimates can provide insights into the distribution and patterns of pesticide use in the EU around the year 2015. The estimate is most reliable for Western and Southern Europe. Outside these regions, data scarcity makes extrapolation more uncertain, potentially limiting the ability to accurate depict regional variations in pesticide use.

Introduction Breast milk is a dynamic type of nourishment that changes based on the needs of the child. An increasing amount of data suggests that mental health may be an important factor in such modulation. In addition, breast milk contains extracellular vesicles (EVs), which are currently considered an important dynamic system of communication between cells, even of different individuals. Objectives Purpose of this article is to investigate whether changes in breast milk in terms of EVs concentrations are related to maternal mental health. Methods This is a case-control study for which we enrolled mothers of infants with bronchiolitis (N=33) and mothers of healthy infants (N=13). Breast milk samples were taken and EVs concentrations were quantified. Maternal mental health was assessed by administration of five different psychometric scales: Edinburgh Postnatal Depression Scale (EPDS), State Trait Anxiety Inventory (STAI-S, STAI-T), Barkin Index of Maternal Functioning (BIMF), The Connor-Davidson Resilience Scale 10 items (CD-RISC). Subsequently, scale scores were related to evs concentrations by negative binomial regressions adjusted for case-control. Results As maternal resilience increases, the EVs of neutrophilic origin (p=0.0447) and those of endothelial origin (p=0.0078) decrease¹. In contrast, an increased EPDS score is associated with higher levels of B-lymphocyte EVs (p=0.0376). Cystic fibrosis (CF) is the most common inherited, life‐limiting disorder in Caucasian populations. It is caused by mutations in the gene encoding the cystic fibrosis transmembrane conductance regulator (CFTR), which lead to an impairment of protein expression and/or function. CFTR is a chloride/bicarbonate channel expressed at the apical surface of epithelial cells of different organs. Nowadays, more than 2100 CFTR genetic variants have been described, but not all of them cause CF. However, around 80–85% of the patients worldwide are characterized by the presence, at least in one allele, of the mutation F508del. CFTR mutations cause aberrant hydration and secretion of mucus in hollow organs. In the lungs, this condition favors bacterial colonization, allowing the development of chronic infections that lead to the onset of the CF lung disease, which is the main cause of death in patients. In recent years, evidence has reported that CFTR loss of function is responsible for alterations in a particular class of bioactive lipids, called sphingolipids (SL). SL are ubiquitously present in eukaryotic cells and are mainly asymmetrically located within the external leaflet of the plasma membrane, where they organize specific platforms capable of segregating a selected number of proteins. CFTR is associated with these platforms that are fundamental for its functioning. Considering the importance of SL in CFTR homeostasis, we attempt here to provide a critical overview of the literature to determine the role of these lipids in channel stability and activity, and whether their modulation in CF could be a target for new therapeutic approaches.

ABSTRACT Phage therapy represents a promising strategy for curing bacterial infections refractory to antibiotics. However, the success rate of phage therapy may be lowered by the emergence of bacterial resistance to the phages used for therapy. In this work, we studied the resistance to the CK4 cocktail, which is a mixture composed of four phages able to cure Pseudomonas aeruginosa infections in animal models. CK4-resistant mutants were easily isolated from cultures grown in either a standard laboratory medium or an artificial sputum medium mimicking the composition of the airway fluid of cystic fibrosis (CF) patients, who are highly susceptible to P. aeruginosa chronic lung infections. In both cases, CK4-resistant mutants resulted in being defective in lipopolysaccharide (LPS) biosynthesis. Accordingly, all CK4 phages were unable to infect wzy mutants lacking the O-antigen polymerase. A survey of the other 15 P. aeruginosa phages isolated from different environmental sources showed that they all needed either wzy or the Type IV-pilus (T4P) biosynthetic gene pilQ for the infection. Overall, our data suggest that 16 out of the 19 analyzed Pseudomonas phages may use either the LPS or the T4P as a receptor. Interestingly, CK4-resistant mutants devoid of the O-antigen had strongly attenuated virulence in a zebrafish embryo infection model, and the lack of T4P also decreased virulence in zebrafish. With respect to isolates from patients with CF, phages not reproducing in the Δwzy mutant had a wider host range than those requiring pilQ, suggesting that phages dependent on PAO1-type T4P may have limited therapeutic value for treating CF-related infections. IMPORTANCE In this work, we identified the putative receptors of 16 Pseudomonas phages and evaluated how resistance to phages recognizing different bacterial receptors may affect the virulence. Our findings are relevant for the implementation of phage therapy of Pseudomonas aeruginosa infections, which are difficult to treat with antibiotics. Overall, our results highlight the need to modify natural phages to enlarge the repertoire of receptors exploited by therapeutic phages and suggest that phages using the PAO1-type T4P as receptor may have limited value for the therapy of the cystic fibrosis infection.

Phosphorus (P) availability affects the response of terrestrial ecosystems to environmental and climate change (e.g., elevated CO2), yet the magnitude of this effect remains uncertain. This uncertainty arises mainly from a lack of quantitative understanding of the soil biological and geochemical P cycling processes, particularly the P exchange with soil mineral surfaces, which is often described by a Langmuir sorption isotherm. We first conducted a literature review on P sorption experiments and terrestrial biosphere models (TBMs) using a Langmuir isotherm. We then developed a new algorithm to describe the inorganic P exchange between soil solution and soil matrix based on the double-surface Langmuir isotherm and extracted empirical equations to calculate the sorption capacity and Langmuir coefficient. We finally tested the conventional and new models of P sorption at five beech forest sites in Germany along a soil P stock gradient using the QUINCY (QUantifying Interactions between terrestrial Nutrient CYcles and the climate system) TBM. We found that the conventional (single-surface) Langmuir isotherm approach in most TBMs largely differed from P sorption experiments regarding the sorption capacities and Langmuir coefficients, and it simulated an overly low soil P-buffering capacity. Conversely, the double-surface Langmuir isotherm approach adequately reproduced the observed patterns of soil inorganic P pools. The better representation of inorganic P cycling using the double-surface Langmuir approach also improved simulated foliar N and P concentrations as well as the patterns of gross primary production and vegetation carbon across the soil P gradient. The novel model generally reduces the estimates of P limitation compared with the conventional model, particularly at the low-P site, as the model constraint of slow inorganic P exchange on plant productivity is reduced.

Biopolymer-coated phosphorus fertilizers have shown promising results in slowing the release of P. However, their dissolution behaviour has been studied mainly in water, and their impacts on soil properties have rarely been addressed. The aim of this study was to compare the dissolution of P from uncoated triple superphosphate (TSP) and biopolymer-coated TSP fertilizers at the surface of cropped Mediterranean soils and to evaluate their impacts on the soil pH and the distributions of several P pools. The experimental treatments were: no fertilizer, uncoated TSP, TSP coated with lignin and TSP coated with lignin and carrageenan. The spatial and temporal variations in the pH of the fertilized soils were investigated using optodes. The concentrations of different pools of P (available P, microbial P), and total P as well as P recovery at different distances from the granules were determined. The coated TSP acidified the soil to a radius of 14 mm around each fertilizer granule, while the acidification spread to 21 mm in the treatment with the uncoated TSP. Moreover, an increase in microbial P was observed in the soil treated with coated TSP, possibly due to the carbon input and the consequent increase in microbial biomass. However, slower P-release was observed from the coated TSP due to the effect of the coating barrier. The P recovery results showed P migration within a 21 mm radius from each granule, and the P recovery from the TSP coated with lignin alone was significantly greater than that from the uncoated TSP treatment.

For about 24 000 river basins across Europe, we provide a continuous representation of the streamflow regime in terms of empirical flow-duration curves (FDCs), which are key signatures of the hydrological behaviour of a catchment and are widely used for supporting decisions on water resource management as well as for assessing hydrologic change. In this study, FDCs are estimated by means of the geostatistical procedure termed total negative deviation top-kriging (TNDTK), starting from the empirical FDCs made available by the Joint Research Centre of the European Commission (DG-JRC) for about 3000 discharge measurement stations across Europe. Consistent with previous studies, TNDTK is shown to provide high accuracy for the entire study area, even with different degrees of reliability, which varies significantly over the study area. In order to provide this kind of information site by site, together with the estimated FDCs, for each catchment we provide indicators of the accuracy and reliability of the performed large-scale geostatistical prediction. Summary: In the last years, extracellular vesicles (EVs) from different plant matrices have been isolated and gained the interest of the scientific community for their intriguing biological properties. In this study, we isolated and characterized nanovesicles from lemon juice (LNVs) and evaluated their antioxidant effects. We tested LNV antioxidant activity using human dermal fibroblasts that were pre-treated with LNVs for 24 h and then stimulated with hydrogen peroxide (H2O2) and UVB irradiation. We found that LNV pre-treatment reduced ROS levels in fibroblasts stimulated with H2O2 and UVB. This reduction was associated with the activation of the AhR/Nrf2 signaling pathway, whose protein expression and nuclear localization was increased in fibroblasts treated with LNVs. By using zebrafish embryos as in vivo model, we confirmed the antioxidant effects of LNVs. We found that LNVs reduced ROS levels and neutrophil migration in zebrafish embryos stimulated with LPS.

Pesticides can be an important stressor to aquatic ecosystems, and their use is strictly regulated in the European Union (EU). However, data on the use of pesticides are rather limited and poorly available, and monitoring is often insufficient to characterize their actual exposure and impact. The aim of the work presented here is to harness the limited data available and assess, for the first time, the distribution of concentrations and toxicity of 148 pesticide active substances (AS) for the whole EU. Starting from available estimates of pesticide use in agriculture and a simple screening-level model of their fate and transport, we quantify pesticide concentrations in soil and water. A comparison with monitoring data shows that predicted water concentrations are in plausible orders of magnitude, hence the model can be regarded as a first-approximation representation of the distribution of pesticides in the environment. The toxicity of individual pesticide active substances (AS) is characterized by their concentrations divided by the respective no observed effect concentrations (NOEC) for aquatic organisms, which represents the “toxic units” (TU) of each AS. The cumulative toxicity of pesticides in soils and streams of the EU is obtained by summing the TU of individual AS. We estimate that the toxicity of individual AS is generally well below 0.1 TU, indicating relatively safe environmental exposure. However, the cumulative toxicity of a mixture of AS can exceed 0.1 toxic units (TU) for more than 27% of the length of the EU’s stream network, and 1 TU for more than 4%. The cumulative toxicity at a given location is driven by only a handful of AS, but these differ from site to site reflecting the variability of pesticide use. Still, we estimate that only about 20 AS out of 148 appear among the top contributors to cumulative toxicity in most cases. While our assessment suggests a relatively widespread risk due to pesticide pollution, it also points to the important limitations concerning knowledge of pesticide use and monitoring of pesticide occurrence in the environment. These limitations need to be addressed in order to evaluate more accurately the effectiveness of EU pesticide policies. The assessment represents a proof-of-concept of a method that can be applied in support of the monitoring of pesticide policies implementation in the EU and elsewhere, once pesticide use can be estimated.

Abstract Brain injury at birth is an important cause of neurological and behavioral disorders. Hypoxic-ischemic encephalopathy (HIE) is a critical cerebral event occurring acutely or chronically at birth with high mortality and morbidity in newborns. Therapeutic strategies for the prevention of brain damage are still unknown, and the only medical intervention for newborns with moderate-to-severe HIE is therapeutic hypothermia (TH). Although the neurological outcome depends on the severity of the initial insult, emerging evidence suggests that infants with mild HIE who are not treated with TH have an increased risk for neurodevelopmental impairment; in the current clinical setting, there are no specific or validated biomarkers that can be used to both correlate the severity of the hypoxic insult at birth and monitor the trend in the insult over time. The aim of this work was to examine the presence of autophagic and mitophagic proteins in bodily fluids, to increase knowledge of what, early at birth, can inform therapeutic strategies in the first hours of life. This is a prospective multicentric study carried out from April 2019 to April 2020 in eight third-level neonatal intensive care units. All participants have been subjected to the plasma levels quantification of both Parkin (a protein involved in mitophagy) and ATG5 (involved in autophagy). These findings show that Parkin and ATG5 levels are related to hypoxic-ischemic insult and are reliable also at birth. These observations suggest a great potential diagnostic value for Parkin evaluation in the first 6 h of life.

At the early stages of pedogenesis, the dynamics of phosphorus (P) in soils are controlled by microbial communities, the physicochemical properties of the soil and the environmental conditions. While various microorganisms involved in carrying out biogeochemical processes have been identified, little is known about the actual contribution of microbial processes, such as organic P hydrolysis and microbial P turnover, to P cycling. We thus focused on processes driven by microbes and how they affect the size and cycling of organic and inorganic soil P pools along a soil chronosequence in the Chamser Kangri glacier forefield (Western Himalayas). The rapid retreat of the glacier allowed us to study the early stages of soil formation under a cold arid climate. Biological P transformations were studied with the help of the isotopic composition of oxygen (O) in phosphate (δ18OP) coupled to sequential P fractionation performed on soil samples (0–5 cm depth) from four sites of different age spanning 0 to 100–150 years. The P bound to Ca, i.e., 1 M HCl-extractable P, still represented 95 % of the total P stock after approximately 100 years of soil development. Its isotopic composition was similar to the parent material at the most developed site. Primary phosphate minerals, possibly apatite, mostly comprised this pool. The δ18OP of the available P and the NaOH-extractable inorganic P instead differed from that of the parent material, suggesting that these pools underwent biological turnover. The δ18OP of the available P was mostly controlled by the microbial P, suggesting fast exchanges occurred between these two pools possibly fostered by repeated freezing–thawing and drying–rewetting cycles. The release of P from organic P becomes increasingly important with soil age, constituting one-third of the P flux to available P at the oldest site. Accordingly, the lighter isotopic composition of the P bound to Fe and Al oxides at the oldest site indicated that this pool contained phosphate released by organic P mineralization. Compared to previous studies on early pedogenesis under alpine or cold climate, our findings suggest a much slower decrease of the P-bearing primary minerals during the first 100 years of soil development under extreme conditions. However, they provide evidence that, by driving short-term P dynamics, microbes play an important role in controlling the redistribution of primary P into inorganic and organic soil P pools.

Current understanding of phosphorus (P) cycling in soils can be enhanced by integrating previously discrete findings concerning P speciation, exchange kinetics, and the underlying biological and geochemical processes. Here, we combine sequential extraction with P K-edge X-ray absorption spectroscopy and isotopic methods (33P and 18O in phosphate) to characterize P cycling on a climatic gradient in Hawaii. We link P pools to P species and estimate the turnover times for commonly considered P pools. Dissolved P turned over in seconds, resin-extractable P in minutes, NaOH-extractable inorganic P in weeks to months, and HCl-extractable P in years to millennia. Furthermore, we show that in arid-zone soils, some primary mineral P remains even after 150 ky of soil development, whereas in humid-zone soils of the same age, all P in all pools has been biologically cycled. The integrative information we provide makes possible a more dynamic, process-oriented conceptual model of P cycling in soils.
