Abstract

Assessing HER2 expression in breast cancer cells holds significant diagnostic and prognostic importance. Traditional methods like immunohistochemistry and in situ hybridization suffer from low sensitivity and misclassification rates. In this frame, techniques such as vibrational microscopies can ensure, together with low costs and analytical speed, both high accuracy and precision. Herein, we propose a combined Raman and SERS approach for characterizing 4 breast cancer cell lines and normal cells with varying HER2 expression levels. We show that Raman spectroscopy offers a promising alternative, providing unique molecular fingerprints for cell types based on their biochemical signatures. Its non-invasive nature and ability to detect subtle changes in cellular metabolism make it ideal for cancer cell analysis. Coupled with machine learning techniques like PCA and LDA, Raman spectroscopy can classify different breast cancer subcategories accurately. Surface Enhanced Raman Scattering (SERS) further enhances sensitivity, allowing the detection of single molecules like HER2 receptors. Overall, our results enable fast screening of cancer subpopulation in terms of HER2 concentration and macromolecule cell content. Integration of Raman spectroscopy with SERS offers precise identification and opens avenues for personalized therapies.

Abstract

Hepatocellular carcinoma (HCC), the most common form of primary liver cancer, represents a global health challenge due to its complexity and the limitations of current diagnostic techniques. By combining Raman spectroscopy and Artificial Intelligence (AI), we have succeeded in classifying tumor cells. In fact, we have performed a first Raman spectral analysis based on the characterization and differentiation between uncultured primary human liver cells derived from resected HCC tumor tissue and the adjacent non-tumor counterpart. Biochemical analysis of the collected Raman spectra revealed that there is more DNA in the nuclei of the tumor cells than in non-tumor cells. We then develop three machine learning approaches, including multivariate models and neural networks, to rapidly automate the recognition and classification of the Raman spectra of both cells. To evaluate the performance of the developed AI models, we prepared and analyzed two additional cell samples with a ratio of 4:1 and 3:1 between tumor and non-tumor cells and compared the obtained results with the nominal percentages (accuracy of 80 and 60%, respectively). These results confirm that the models are able to make classifications at the level of a single spectrum, indicating the possibility of rapidly analysing and classifying a primary HCC cell.

Abstract

Here we have developed an advanced surface-enhanced Raman scattering (SERS) platform that enables the ultrasensitive, rapid and highly specific identification of tumour biomarkers in liquid biopsies. Our focus is on the detection of Thyroglobulin (Tg), the most important tumour biomarker for the diagnosis and prognosis of thyroid cancer. Specifically, SERS-active substrates fabricated by nanosphere lithography on chip or on tips of optical fiber (OF) were functionalised with Tg Capture antibodies. Gold nanoparticles were functionalized with Detection antibodies and conjugated with a Raman reporter. The sandwich assay platform was validated in the planar configuration and a detection limit of only 7 pg/ml was successfully achieved. A careful morphological characterisation of the SERS substrates allowed us to strictly correlate the coverage area with the Tg concentration. The same approach was successfully demonstrated in the washout of fine needle aspiration biopsies from cancer patients. The strategy was transferred to the Lab On Fiber (LOF) SERS platform and successfully used to detect Tg concentration. The proposed SERS-enhanced immunoassay platform has proven to be highly versatile and can be used with both microfluidic chip POC devices and SERS-OF-based optrodes to perform sensitive, specific and rapid ex vivo assays for Tg detection in liquid intraoperative biopsies.

Abstract

The creation of SERS‐active substrates is investigated by self‐assembling hierarchical structures of plasmonic‐assisted nanospheres (HSNs). The hierarchical architecture exploits the advantages of the well‐known ordered configuration of a hexagonally closed‐packed array (CPA) of nanospheres. A further layer of upper nanospheres is used to provide regular and intense surface hotspots, located at the nanogaps between neighboring nanospheres. Numerical analysis is carried out to predict the SERS performances and to identify the more promising configurations, by offering design criteria and a physical insight on the conditions affecting the SERS response of the self‐assembled substrates. Two alternative self‐assembly fabrication methods have been pursued to realize HSNs, namely co‐deposition and sequential deposition. Morphological analysis revealed the formation of well‐ordered hierarchical structures with different ratios between the diameters of the bottom and upper nanospheres. Experimental analysis of the SERS response demonstrates that HSNs can work as cost‐effective SERS substrates with superior performances with respect to the simpler single‐layer CPA configurations. Abstract

We report on the first experimental evidence of guided resonances (GRs) in photonic crystal slabs based on aperiodically-ordered supercells. Using the Ammann-Beenker (quasiperiodic, 8-fold symmetric) tiling geometry, we present our study on the fabrication, experimental characterization, and full-wave numerical simulation of two representative structures (with different filling parameters) operating at near-infrared wavelengths (1300-1600 nm). Our results show a fairly good agreement between measurements and numerical predictions, and pave the way for the development of new strategies (based, e.g., on the lattice symmetry breaking) for GR engineering., Comment: 10 pages, 5 figures (minor revisions)

Abstract

In this paper, we present a comparative study of the tuning efficiency and sensitivity of guided resonances (GRs) in photonic crystal (PC) holed slabs based on periodic and aperiodically-ordered unit cells, aimed at assessing the applicability of these important technology platforms to ultra-compact optical sensors and active devices. In particular, with specific reference to square-lattice periodic PCs and aperiodically-ordered Ammann-Beenker photonic quasi-crystals, we study the effects of the hole radius, slab thickness, and refractive index on the GR sensitivity and tunability with respect to variation in the hole refractive index. Finally, we carry out a theoretical and numerical analysis in order to correlate the GR shift with the field distribution of the unperturbed (air holes) structures. Our results indicate that the spatial arrangement of the holes may strongly influence the tuning and sensitivity efficiency, and may provide new degrees of freedom and tools for the design and optimization of novel photonic devices for both sensing and telecommunication applications., Comment: 14 pages, 11 figures, 4 tables; minor changes

Abstract

In this paper, we study the excitation of guided resonances (GRs) in photonic-crystal slabs based on point-defected aperiodically-ordered supercells. With specific reference to perforated-slab structures and the Ammann-Beenker octagonal lattice geometry, we carry out full-wave numerical studies of the plane-wave responses and of the underlying modal structures, which illustrate the representative effects induced by the introduction of symmetry-preserving and symmetry-breaking defects. Our results demonstrate that breaking the supercell mirror symmetries via the judicious introduction of point-defects enables for the excitation of otherwise uncoupled GRs, with control on the symmetry properties of their field distributions, thereby constituting an attractive alternative to those GR-engineering approaches based on the asymmetrization of the hole shape. Abstract

In this paper, we report on the first evidence of guided resonances (GRs) in aperiodically-ordered photonic crystals, tied to the concept of "quasicrystals" in solid-state physics. Via a full-wave numerical study of the transmittance response and the modal structure of a photonic quasicrystal (PQC) slab based on a representative aperiodic geometry (Ammann-Beenker octagonal tiling), we demonstrate the possibility of exciting GR modes, and highlight similarities and differences with the periodic case. In particular, we show that, as for the periodic case, GRs arise from the coupling of the incident plane-wave with degenerate modes of the PQC slab that exhibit a matching symmetry in the spatial distribution, and can still be parameterized via a Fano-like model. Abstract

This review presents an overview of "Lab on Fiber" technologies and devices with special focus on the design and development of advanced fiber optic nanoprobes for biological applications. Depending on the specific location where functional materials at micro and nanoscale are integrated, "Lab on Fiber Technology" is classified into three main paradigms: Lab on Tip (where functional materials are integrated onto the optical fiber tip), Lab around Fiber (where functional materials are integrated on the outer surface of optical fibers), and Lab in Fiber (where functional materials are integrated within the holey structure of specialty optical fibers). This work reviews the strategies, the main achievements and related devices developed in the "Lab on Fiber" roadmap, discussing perspectives and challenges that lie ahead, with special focus on biological sensing applications.

Abstract

Optical fibers are the technological innovation that has revolutionized the way we communicate. The development of optical fiber technology started from the earliest studies and pioneering experiments on total internal reflection, and passed through several technological advancements, ending with the achievement of modern optical fiber devices and systems for communication and sensing applications. Nowadays, the research and development of optical fiber technology is very wide and diversified. A multitude of aspects are singularly investigated by several researchers with different scientific backgrounds.
