Abstract

Aim: Marginal tree populations, either those located at the edges of the species' range or in suboptimal environments, are often a valuable genetic resource for biological conservation. However, there is a lack of knowledge about the genetic consequences of population marginality, estimated across entire species' ranges. Our study addresses this gap by providing information about several genetic indicators and their variability in marginal and core populations identified using quantitative marginality indices. Location: Southwestern Europe and North Africa. Methods: Using 10,185 SNPs across 82 populations of maritime pine (Pinus pinaster Ait.), a widespread conifer characterised by a fragmented range, we modelled the relationship of seven genetic indicators potentially related to population evolutionary resilience, namely genetic diversity (based on both all SNPs and outlier SNPs), inbreeding, genetic differentiation, recessive genetic load and genomic offset, with population geographical, demo‐historical and ecological marginality (as estimated by nine quantitative indices). Models were constructed for both regional (introducing gene pool as a random factor) and range‐wide spatial scales. Results: We showed a trend towards decreasing overall genetic diversity and increasing differentiation with geographic marginality, supporting the centre‐periphery hypothesis (CPH). However, we found no correlation between population inbreeding and marginality, while geographically marginal populations had a lower recessive genetic load (only models without the gene pool effect). Ecologically marginal populations had a higher genomic offset, suggesting higher maladaptation to future climate, albeit some of these populations also had high genetic diversity for climate outliers. Main Conclusions: Overall genetic diversity (but not outlier‐based estimates) and differentiation patterns support the CPH. Ecologically marginal populations and those at the southern edge could be more vulnerable to climate change due to higher climate maladaptation, as predicted by genomic offsets, and/or lower potentially adaptive genetic diversity. Abstract

Most tree species underwent cycles of contraction and expansion during the Quaternary. These cycles led to an ancient and complex genetic structure that has since been affected by extensive gene flow and by strong local adaptation. The extent to which hybridization played a role in this multi‐layered genetic structure is important to be investigated. To study the effect of hybridization on the joint population genetic structure of two dominant species of the Eurasian boreal forest, Picea abies and P. obovata, we used targeted resequencing and obtained around 480 K nuclear SNPs and 87 chloroplast SNPs in 542 individuals sampled across most of their distribution ranges. Despite extensive gene flow and a clear pattern of Isolation‐by‐Distance, distinct genetic clusters emerged, indicating the presence of barriers and corridors to migration. Two cryptic refugia located in the large hybrid zone between the two species played a critical role in shaping their current distributions. The two species repeatedly hybridized during the Pleistocene and the direction of introgression depended on latitude. Our study suggests that hybridization helped both species to overcome main shifts in their distribution ranges during glacial cycles and highlights the importance of considering whole species complex instead of separate entities to retrieve complex demographic histories. Abstract

Marginal and peripheral populations are important for biodiversity conservation. Their original situation in a species’ geographic and ecological space often confers them genetic diversity and traits of high adaptive value. Yet theoretical hypotheses related to marginality are difficult to test because of confounding factors that influence marginality, namely environment, geography, and history. There is an urgent need to develop metrics to disentangle these confounding factors. We designed nine quantitative indices of marginality and peripherality that define where margins lie within species distributions, from a geographical, an environmental and a historical perspective. Using the distribution maps of eight European forest tree species, we assessed whether these indices were idiosyncratic or whether they conveyed redundant information. Using a database on marginal and peripheral populations based on expert knowledge, we assessed the capacity of the indices to predict the marginality status of a population. There was no consistent pattern of correlation between indices across species, confirming that the indices conveyed different information related to the specific geometry of the species distributions. Contrasting with this heterogeneity of correlation patterns across species, the relative importance of the indices to predict the marginality status of populations was consistent across species. However, there was still a significant country effect in the marginality status, showing a variation in expert opinion of marginality vis-á-vis the species distribution. The marginality indices that we developed are entirely based on distribution maps and can be used for any species. They pave the way for testing hypotheses related to marginality and peripherality, with important implications in quantitative ecology, genetics, and biodiversity conservation.

23. The oldest known clones of Salix herbacea growing in the Northern Apennines, Italy are at least 2000 years old

Abstract

Premise: Dominant in many ecosystems around the world, clonal plants can reach considerable ages and sizes. Due to their modular growth patterns, individual clonal plants (genets) can consist of many subunits (ramets). Since single ramets do not reflect the actual age of genets, the ratio between genet size (radius) and longitudinal annual growth rate (LAGR) of living ramets is often used to approximate the age of clonal plants. However, information on how the LAGR changes along ramets and how LAGR variability may affect age estimates of genets is still limited. Methods: We assessed the variability of LAGR based on wood‐section position along the ramets and on the duration of the growing season on three genetically distinct genets of Salix herbacea growing in the Northern Apennines (Italy). We compared genet ages estimated by dividing genet radius by the LAGRs of its ramets. Results: LAGR increased significantly from the stem apex to the root collar; indicating that ramet growth rate decreased with time. Furthermore, a difference of ca. 2 weeks in the onset of the growing period did not impact LAGR. Considering the high LAGR variability, we estimated that the three genets started to grow between ~2100 and ~7000 years ago, which makes them the oldest known clones of S. herbacea even considering the most conservative age estimate. Conclusions: Our findings indicate that analyzing ramets at the root collar provides an integrative measurement of their overall LAGR, which is crucial for estimating the age of genets.

Abstract

New mutations provide the raw material for evolution and adaptation. The distribution of fitness effects (DFE) describes the spectrum of effects of new mutations that can occur along a genome, and is therefore of vital interest in evolutionary biology. Recent work has uncovered striking similarities in the DFE between closely related species, prompting us to ask whether there is variation in the DFE among populations of the same species, or among species with different degrees of divergence, i.e., whether there is variation in the DFE at different levels of evolution. Using exome capture data from six tree species sampled across Europe we characterised the DFE for multiple species, and for each species, multiple populations, and investigated the factors potentially influencing the DFE, such as demography, population divergence and genetic background. We find statistical support for there being variation in the DFE at the species level, even among relatively closely related species. However, we find very little difference at the population level, suggesting that differences in the DFE are primarily driven by deep features of species biology, and that evolutionarily recent events, such as demographic changes and local adaptation, have little impact.

Abstract

Aim: According to the Latitudinal Biotic Interaction Hypothesis (LBIH), the general increase in biodiversity towards lower latitudes can be partially explained by an increase in the intensity of biotic interactions. While LBIH received some support for plant-herbivores interactions, much less is known about how higher trophic levels may contribute to shape biotic interactions across latitudinal gradients. We hypothesized that the diversity of insectivorous birds increases towards lower latitude, leading to higher predation rates on insect herbivores. Location: Europe. Taxon: Insectivorous birds and pedunculate oaks. Methods: We deployed plasticine caterpillars in 138 oak trees in 47 sites along a 19° latitudinal gradient in Europe to quantify bird insectivory through predation attempts. In addition, we used passive acoustic monitoring to (i) characterize the acoustic diversity of surrounding soundscapes; and (ii) infer both taxonomic and functional diversity of insectivorous birds from recordings. Results: The functional diversity of insectivorous birds increased towards lower latitude. Bird predation increased with latitude, forest cover and bird acoustic diversity but decreased with mean annual temperature and functional richness of insectivorous birds. Contrary to our predictions, latitudinal clines in bird predation attempts were not directly mediated by changes in insectivorous bird diversity or acoustic diversity, but latitude and habitat still had independent effects on predation attempts. Main conclusions: Our study does not fully support the predictions of the LBIH of more biotic interactions southwards and advocates for better accounting for activity and abundance of insectivorous birds when studying the large-scale variation in insect-tree interactions.

Abstract

Complementary gene-resequencing and transcriptomic approaches reveal contrasted evolutionary histories in a species complex. Pinus halepensis and Pinus brutia are closely related species that can intercross, but occupy different geographical ranges and bioclimates. To study the evolution of this species complex and to provide genomic resources for further research, we produce and analyze two new complementary sets of genetic resources: (i) a set of 172 re-sequenced genomic target loci analyzed in 45 individuals, and (ii) a set of 11 transcriptome assemblies. These two datasets provide insights congruent with previous studies: P. brutia displays high level of genetic diversity and no genetic sub-structure, while P. halepensis shows three main genetic clusters, the western Mediterranean and North African clusters displaying much lower genetic diversity than the eastern Mediterranean cluster, the latter cluster having similar genetic diversity to P. brutia. In addition, these datasets provide new insights on the timing of the species-complex history: the two species would have split at the end of the tertiary, and the changing climatic conditions of the Mediterranean region at the end of the Tertiary-beginning of the Quaternary, together with the distinct species tolerance to harsh climatic conditions would have resulted in different geographic distributions, demographic histories and genetic patterns of the two pines. The multiple glacial-interglacial cycles during the Quaternary would have led to the expansion of P. brutia in the Middle East, while P. halepensis would have been through bottlenecks. The last glaciations, from 0.6 Mya on, would have affected further the Western genetic pool of P. halepensis.

Abstract

The dataset presented here was collected by the GenTree project (EU-Horizon 2020), which aims to improve the use of forest genetic resources across Europe by better understanding how trees adapt to their local environment. This dataset of individual tree-core characteristics including ring-width series and whole-core wood density was collected for seven ecologically and economically important European tree species: silver birch (Betula pendula), European beech (Fagus sylvatica), Norway spruce (Picea abies), European black poplar (Populus nigra), maritime pine (Pinus pinaster), Scots pine (Pinus sylvestris), and sessile oak (Quercus petraea). Tree-ring width measurements were obtained from 3600 trees in 142 populations and whole-core wood density was measured for 3098 trees in 125 populations. This dataset covers most of the geographical and climatic range occupied by the selected species. The potential use of it will be highly valuable for assessing ecological and evolutionary responses to environmental conditions as well as for model development and parameterization, to predict adaptability under climate change scenarios.

Abstract

Studying how genetic diversity is transmitted between generations is critical for assessing the survival prospects of fragmented forest tree populations. This is especially urgent in the case of rare, endangered species that are at high risk of extinction and/or exceptionally susceptible to climate change, and are thus to be prioritised for conservation initiatives. We performed the first assessment of gene flow patterns by parentage analysis in the IUCN red-listed Serbian spruce [Picea omorika (Pan.c.) Purk.], within a Genetic Conservation Unit (GCU) which was exposed to fire disturbance 30 years ago. Parentage analysis was based on assigning the parentage of 104 juveniles to all the adult trees (48) present in the 2.8 ha population. All individuals were genotyped at 11 highly informative nuclear microsatellite markers. We found that fire disturbance triggered natural regeneration, usually poor to absent in Serbian spruce, and boosted pollen immigration (66%), which is reported as rather modest among undisturbed populations of this species. Seed immigration was absent, coherently with what was previously hypothesised for natural, undisturbed populations. Thus, the immigration of new gametes into the studied Serbian spruce population was accomplished exclusively through pollen gene flow, at a rate sufficient to counteract, at least in the short term, genetic drift. Furthermore, the abundant post-disturbance pollen immigration most likely accounts for the lack of fine-scale spatial genetic structure (SGS) in the juvenile cohort, contrary to the SGS observed in the adult cohort which is amongst the strongest ever recorded in anemophilous forest tree species. Finally, as generally found in forest trees, individual reproductive success was uneven among individuals, with 25% of local adults siring 70% of the sampled juveniles. Abstract

Marginal and peripheral populations are important for biodiversity conservation. Their original situation in a species’ geographic and ecological space often confers them genetic diversity and traits of high adaptive value. Yet theoretical hypotheses related to marginality are difficult to test because of confounding factors that influence marginality, namely environment, geography, and history. There is an urgent need to develop metrics to disentangle these confounding factors. We designed nine quantitative indices of marginality and peripherality that define where margins lie within species distributions, from a geographical, an environmental and a historical perspective. Using the distribution maps of eight European forest tree species, we assessed whether these indices were idiosyncratic or whether they conveyed redundant information. Using a database on marginal and peripheral populations based on expert knowledge, we assessed the capacity of the indices to predict the marginality status of a population. There was no consistent pattern of correlation between indices across species, confirming that the indices conveyed different information related to the specific geometry of the species distributions. Contrasting with this heterogeneity of correlation patterns across species, the relative importance of the indices to predict the marginality status of populations was consistent across species. However, there was still a significant country effect in the marginality status, showing a variation in expert opinion of marginality vis-á-vis the species distribution. The marginality indices that we developed are entirely based on distribution maps and can be used for any species. They pave the way for testing hypotheses related to marginality and peripherality, with important implications in quantitative ecology, genetics, and biodiversity conservation.

Abstract

Ecological modeling refers to the construction and analysis of mathematical models aimed at understanding the complexity of ecological processes and at predicting how real ecosystems might evolve. It is a quickly expanding approach boosted by impressive accelerations in the availability of computational resources and environmental databases. In the light of foreseeing the effect of climate change on forest ecosystems, the branch of ecological modeling focusing on species distribution models (SDMs) has become widely used to estimate indices of habitat suitability and to forecast future tree distributions. However, SDMs are usually informed based solely on environmental data without any reference to the genetic makeup underlying responses to the environment, the possibility of exchanging variants helping to persist in situ, or the capacity to chase suitable conditions elsewhere. Among the main evolutionary processes that may complement forecasts of range shifts are local adaptation and gene flow, i.e., the occurrence of genetic variants conferring a population the optimal fitness in its own habitat and the exchange of adaptive alleles between populations. Local adaptation and gene flow could be described by indices of genetic diversity and structure, genetic load, genomic offset, and an admixture of genetic lineages. Here, we advocate for the development of a new analytical approach integrating environmental and genomic information when projecting tree distributions across space and time. To this aim, we first provide a literature review on the use of genetics when modeling intraspecific responses to the environment, and we then discuss the potential improvements and drawbacks deriving from the inclusion of genomic data into the current SDM framework. Finally, we speculate about the potential impacts of genomic-informed predictions in the context of forest conservation and provide a synthetic framework for developing future forest management strategies. Abstract

Background: Progress in the field of evolutionary forest ecology has been hampered by the huge challenge of phenotyping trees across their ranges in their natural environments, and the limitation in high-resolution environmental information. Findings: The GenTree Platform contains phenotypic and environmental data from 4,959 trees from 12 ecologically and economically important European forest tree species: Abies alba Mill. (silver fir), Betula pendula Roth. (silver birch), Fagus sylvatica L. (European beech), Picea abies (L.) H. Karst (Norway spruce), Pinus cembra L. (Swiss stone pine), Pinus halepensis Mill. (Aleppo pine), Pinus nigra Arnold (European black pine), Pinus pinaster Aiton (maritime pine), Pinus sylvestris L. (Scots pine), Populus nigra L. (European black poplar), Taxus baccata L. (English yew), and Quercus petraea (Matt.) Liebl. (sessile oak). Phenotypic (height, diameter at breast height, crown size, bark thickness, biomass, straightness, forking, branch angle, fructification), regeneration, environmental in situ measurements (soil depth, vegetation cover, competition indices), and environmental modeling data extracted by using bilinear interpolation accounting for surrounding conditions of each tree (precipitation, temperature, insolation, drought indices) were obtained from trees in 194 sites covering the species’ geographic ranges and reflecting local environmental gradients. Conclusion: The GenTree Platform is a new resource for investigating ecological and evolutionary processes in forest trees. The coherent phenotyping and environmental characterization across 12 species in their European ranges allow for a wide range of analyses from forest ecologists, conservationists, and macro-ecologists. Also, the data here presented can be linked to the GenTree Dendroecological collection, the GenTree Leaf Trait collection, and the GenTree Genomic collection presented elsewhere, which together build the largest evolutionary forest ecology data collection available.

44. La vivaistica forestale in Italia al bivio: sfide e strategie.

Abstract

The recognition of the key role of forests in contrasting the dramatic effects of climate change and biodiversity crisis is the pillar of many initiatives on a global, European, and national scale calling for afforestation campaigns. The Italian forest nursery sector is currently inadequate to meet the demand for tree seedlings for the national campaigns. Forest nursery production is characterized by regional or local companies that are remarkably different from each other in organization and efficiency. It is therefore urgent to develop a comprehensive restructuring of the entire sector to be able to respond to the specific needs of forestry projects in Italy and Europe. In this paper, we present a series of key principles and criteria aimed at guiding the renaissance of the sector. Strategic actions are proposed by integrating research, governance, public/private partnership, training, and communication. The strategic approach presented is based on a collaborative structure integrating various skills and responsibilities. The first step is an expert review of the Basic Materials (BM) included in the National Register thanks to the development of the genetic studies of forest stands and the inclusion of shrub and herbaceous species, essentials for ecosystem restoration projects counteracting the biodiversity crisis. A series of actions concerns the aspects of certification, voluntary or prescriptive, of the quality of BM, and the ultimate harmonization of national production to European standards. Particular importance needs to be devoted to the collection, evaluation, and conservation of seeds to develop innovative solutions both for the production of BM and for specific implementation phases of afforestation projects and ecosystem restoration. The governance phase might be implemented through the creation of interregional centers with the duty of collecting and conserving seeds, thus enhancing the existing experiences of outstanding regional and provincial tree nurseries with the support of the National Centers for the study and conservation of Forest Biodiversity. Furthermore, the promotion of partnerships between public and private companies and the creation of a shared and accessible national platform represent strategic actions of primary importance. A fundamental role is then assigned to the implementation of training programs and the construction of an open and incremental communication plan. Thanks to these actions it will be possible to place forest nurseries at the center of the relationships between tree planting and afforestation plans and programs, integrated forest design, implementation of new forests, and their adaptive management. Abstract

Motivation: Trait variation within species can reveal plastic and/or genetic responses to environmental gradients, and may indicate where local adaptation has occurred. Here, we present a dataset of rangewide variation in leaf traits from seven of the most ecologically and economically important tree species in Europe. Sample collection and trait assessment are embedded in the GenTree project (EU‐Horizon 2020), which aims at characterizing the genetic and phenotypic variability of forest tree species to optimize the management and sustainable use of forest genetic resources. Our dataset captures substantial intra‐ and interspecific leaf phenotypic variability, and provides valuable information for studying the relationship between ecosystem functioning and trait variability of individuals, and the response and resilience of species to environmental changes. Main types of variable contained: We chose morphological and chemical characters linked to trade‐offs between acquisition and conservation of resources and water use, namely specific leaf area, leaf size, carbon and nitrogen content and their ratio, and the isotopic signature of stable isotope 13C and 15N in leaves. Spatial location and grain: We surveyed between 18 and 22 populations per species, 141 in total, across Europe. Time period: Leaf sampling took place between 2016 and 2017. Major taxa and level of measurement: We sampled at least 25 individuals in each population, 3,569 trees in total, and measured traits in 35,755 leaves from seven European tree species, i.e. the conifers Picea abies, Pinus pinaster and Pinus sylvestris, and the broadleaves Betula pendula, Fagus sylvatica, Populus nigra and Quercus petraea.
