Although lifetime-based luminescent manometers exploiting the luminescence kinetics of Mn
4+ ions exhibit remarkably attractive manometric performance, the number of studies reported in the literature devoted to this area is relatively small. Given that this type of manometer can exhibit very high sensitivity over a limited pressure range, in-depth studies on the correlation of the structure with the manometric properties of such pressure gauges are required to enable the design of manometers with predefined sensing performance. In response to these requirements, the present work investigates the spectroscopic properties of a new promising inorganic phosphor, i.e. K2 Ge4 O9 :Mn4+ , measured as a function of pressure and temperature. As is shown, the spectral shift of the Mn4+ R-line, the ratio of its emission intensity to phonon-progression bands, and the luminescence kinetics of the2 E state can be used for remote pressure readout, with sensitivities of 0.59 nm GPa−1 , SR = 21.7%/Gpa and 12%/Gpa, respectively. Notably, the developed manometer shows the highest sensitivity in the lifetime-based mode reported so far for pressure values above 2 Gpa. Furthermore, the considerable thermal sensitivity of the narrow emission line of Mn4+ ions (R-line) in the K2 Ge4 O9 :0.1% Mn4+ material, combined with its minimal shift under pressure for pressures below 1 GPa allows for the utilization of this phosphor as a luminescent sensor capable of concurrently measuring both pressure and temperature. - Abstract
Importance There is a major need for effective, well-tolerated treatments for idiopathic pulmonary fibrosis (IPF). Objective To assess the efficacy and safety of the autotaxin inhibitor ziritaxestat in patients with IPF. Design, Setting, and Participants The 2 identically designed, phase 3, randomized clinical trials, ISABELA 1 and ISABELA 2, were conducted in Africa, Asia-Pacific region, Europe, Latin America, the Middle East, and North America (26 countries). A total of 1306 patients with IPF were randomized (525 patients at 106 sites in ISABELA 1 and 781 patients at 121 sites in ISABELA 2). Enrollment began in November 2018 in both trials and follow-up was completed early due to study termination on April 12, 2021, for ISABELA 1 and on March 30, 2021, for ISABELA 2. Interventions Patients were randomized 1:1:1 to receive 600 mg of oral ziritaxestat, 200 mg of ziritaxestat, or placebo once daily in addition to local standard of care (pirfenidone, nintedanib, or neither) for at least 52 weeks. Main Outcomes and Measures The primary outcome was the annual rate of decline for forced vital capacity (FVC) at week 52. The key secondary outcomes were disease progression, time to first respiratory-related hospitalization, and change from baseline in St George’s Respiratory Questionnaire total score (range, 0 to 100; higher scores indicate poorer health-related quality of life). Results At the time of study termination, 525 patients were randomized in ISABELA 1 and 781 patients in ISABELA 2 (mean age: 70.0 [SD, 7.2] years in ISABELA 1 and 69.8 [SD, 7.1] years in ISABELA 2; male: 82.4% and 81.2%, respectively). The trials were terminated early after an independent data and safety monitoring committee concluded that the benefit to risk profile of ziritaxestat no longer supported their continuation. Ziritaxestat did not improve the annual rate of FVC decline vs placebo in either study. In ISABELA 1, the least-squares mean annual rate of FVC decline was –124.6 mL (95% CI, −178.0 to −71.2 mL) with 600 mg of ziritaxestat vs –147.3 mL (95% CI, −199.8 to −94.7 mL) with placebo (between-group difference, 22.7 mL [95% CI, −52.3 to 97.6 mL]), and –173.9 mL (95% CI, −225.7 to −122.2 mL) with 200 mg of ziritaxestat (between-group difference vs placebo, −26.7 mL [95% CI, −100.5 to 47.1 mL]). In ISABELA 2, the least-squares mean annual rate of FVC decline was –173.8 mL (95% CI, −209.2 to −138.4 mL) with 600 mg of ziritaxestat vs –176.6 mL (95% CI, −211.4 to −141.8 mL) with placebo (between-group difference, 2.8 mL [95% CI, −46.9 to 52.4 mL]) and –174.9 mL (95% CI, −209.5 to −140.2 mL) with 200 mg of ziritaxestat (between-group difference vs placebo, 1.7 mL [95% CI, −47.4 to 50.8 mL]). There was no benefit with ziritaxestat vs placebo for the key secondary outcomes. In ISABELA 1, all-cause mortality was 8.0% with 600 mg of ziritaxestat, 4.6% with 200 mg of ziritaxestat, and 6.3% with placebo; in ISABELA 2, it was 9.3% with 600 mg of ziritaxestat, 8.5% with 200 mg of ziritaxestat, and 4.7% with placebo. Conclusions and Relevance Ziritaxestat did not improve clinical outcomes compared with placebo in patients with IPF receiving standard of care treatment with pirfenidone or nintedanib or in those not receiving standard of care treatment. - Abstract
Near-infrared (NIR) luminescence thermometry has been brought to the fore as a reliable approach for remote thermal sensing and imaging. Lanthanide (Ln
3+ )-based nanophosphors are often proposed as NIR nanothermometers of choice. However, the combination of Ln3+ with transition metal (TM) ions has recently emerged as a strategy to introduce additional emission bands and/or TM ↔ Ln3+ energy transfer pathways whose temperature dependence can be harnessed to increase the sensitivity of the thermometric approach. Yet, the examples of the combination of luminescence nanothermometers working in the NIR and hosting simultaneously TM and Ln3+ are scarce, leaving plenty of space for the exploration of these systems. Herein, we report on the preparation and optimization of the thermometric performance of Ba3 (VO4 )2 :Mn5+ ,Nd3+ nanophosphors. The different temperature dependences of the emission intensity of the two doped luminescent centers allow using the ratio between Mn5+ and Nd3+ as a reliable thermometric parameter with a relative thermal sensitivity of 1% K−1 close to room temperature. We then showcase the suitability of this nanophosphor for employment in 2D NIR luminescence thermal imaging. Lastly, we critically evaluate the possibility of using this thermal imaging approach through opaque media with the help of phantoms with tissue-like optical properties. As expected, a loss of reliability of the thermometric method is observed due to tissue-induced photon scattering and absorption that differentially affect the emission of Mn5+ and Nd3+ . Overall, the reported results underscore the good performance of the newly developed nanothermometer, while consolidating the call for the use of luminescence nanothermometers working in the time-domain (rather than in the spectral domain) for deep-tissue thermal readout/imaging. Finding thermal history phosphors with high sensitivity and a consistent readout is required for reliable thermal history determination with high temperature resolution. This work presents a new thermal history phosphor based on the luminescence of Eu3+ ions in LaVO4 to meet these requirements. As demonstrated, raising the annealing temperature causes a structural phase transition from a low-temperature tetragonal phase to a high-temperature single-stranded phase. The associated change in the local point symmetry of the crystallographic site occupied by Eu3+ ions result in a significant decrease in the emission intensity ratio of the 5D0 → 7F2 band relative to the 5D0 → 7F1 band, which enables the development of the ratiometric thermal history phosphor with the relative sensitivity of 0.38% °C−1 at 800 °C. - Abstract
Thermal nonradiative depopulation processes of excited levels significantly influence the potential application of phosphors. The high efficiency of thermal quenching of luminescence can limit a phosphor's potential applications as well as reduce the temperature range in which such a material can be used. This work presents a new strategy of shifting of usable temperature range of luminescent thermometer based on Eu
3+ ion emission that the molar ratio of (AsO4 )3− in respect to (VO4 )3− increases in YVx As1−x O4 host material. Consequently, the thermometric performance of the luminescence thermometer based on the single band intensity ratio corresponding to the5 D0 →7 F1 of Eu3+ ions upon optical excitations matching to the1 A2 (1 T1 ) →1 B1 (1 T2 ) and1 E(1 T1 ) →1 B1 (1 T2 ) electronic transition of (VO4 )3− group can be optimized. As shown, the use of YV0.25 As0.75 O4 :Eu3+ has an extended usable temperature range in respect to YVO4 :Eu3+ . This improvement, coupled with its high sensitivity (>1% K−1 ) make YV0.25 As0.75 O4 :Eu3+ a more stable material for luminescence thermometer applications. The presented strategy for modulating the thermal properties of luminescent thermometers through the introduction of the (AsO4 )3− groups is a step towards designing thermometers with on-demand thermometric performance. - Abstract
Background: The INBUILD trial investigated the efficacy and safety of nintedanib versus placebo in patients with progressive fibrosing interstitial lung diseases (ILDs) other than idiopathic pulmonary fibrosis (IPF). We aimed to establish the effects of nintedanib in subgroups based on ILD diagnosis. Methods: The INBUILD trial was a randomised, double-blind, placebo-controlled, parallel group trial done at 153 sites in 15 countries. Participants had an investigator-diagnosed fibrosing ILD other than IPF, with chest imaging features of fibrosis of more than 10% extent on high resolution CT (HRCT), forced vital capacity (FVC) of 45% or more predicted, and diffusing capacity of the lung for carbon monoxide (DLco) of at least 30% and less than 80% predicted. Participants fulfilled protocol-defined criteria for ILD progression in the 24 months before screening, despite management considered appropriate in clinical practice for the individual ILD. Participants were randomly assigned 1:1 by means of a pseudo-random number generator to receive nintedanib 150 mg twice daily or placebo for at least 52 weeks. Participants, investigators, and other personnel involved in the trial and analysis were masked to treatment assignment until after database lock. In this subgroup analysis, we assessed the rate of decline in FVC (mL/year) over 52 weeks in patients who received at least one dose of nintedanib or placebo in five prespecified subgroups based on the ILD diagnoses documented by the investigators: hypersensitivity pneumonitis, autoimmune ILDs, idiopathic non-specific interstitial pneumonia, unclassifiable idiopathic interstitial pneumonia, and other ILDs. The trial has been completed and is registered with ClinicalTrials.gov, number NCT02999178. Findings: Participants were recruited between Feb 23, 2017, and April 27, 2018. We conducted an international study of idiopathic pulmonary fibrosis (IPF) diagnosis among a large group of physicians and compared their diagnostic performance to a panel of IPF experts. A total of 1141 respiratory physicians and 34 IPF experts participated. Participants evaluated 60 cases of interstitial lung disease (ILD) without interdisciplinary consultation. Diagnostic agreement was measured using the weighted kappa coefficient (κw). Prognostic discrimination between IPF and other ILDs was used to validate diagnostic accuracy for first-choice diagnoses of IPF and were compared using the C-index. A total of 404 physicians completed the study. Agreement for IPF diagnosis was higher among expert physicians (κw=0.65, IQR 0.53–0.72, p20 years of experience (C-index=0.72, IQR 0.0–0.73, p=0.229) and non-university hospital physicians with more than 20 years of experience, attending weekly MDT meetings (C-index=0.72, IQR 0.70–0.72, p=0.052), did not differ significantly (p=0.229 and p=0.052 respectively) from the expert panel (C-index=0.74 IQR 0.72–0.75). Experienced respiratory physicians at university-based institutions diagnose IPF with similar prognostic accuracy to IPF experts. - Abstract
The introduction of PDDL+ allowed more accurate representations of complex real-world problems of interest to the scientific community. However, PDDL+ problems are notoriously challenging to planners, requiring more advanced heuristics. We introduce the Temporal Pattern Database (TPDB), a new domain-independent heuristic technique designed for PDDL+ domains with mixed discrete/continuous behaviour, non-linear system dynamics, processes, and events. The pattern in the TPDB is obtained through an abstraction based on time and state discretisation. Our approach combines constraint relaxation and abstraction techniques, and uses solutions to the relaxed problem, as a guide to solving the concrete problem with a discretisation fine enough to satisfy the continuous model's constraints.

- Abstract
Despite the numerous advantages of lanthanide based luminescent thermometers, a major limitation is the limited brightness associated with the low absorption cross section of the 4f–4f transitions. In this paper, co-doping with Cr
3+/4+ is proposed as a strategy to enhance the near-infrared luminescence emission of Nd3+ /Er3+ -based luminescent thermometers, as well as to modulate the temperature sensitivity of the ratio of Nd3+ to Er3+ in YAG microcrystals. It was found that an increase in the Cr3+ concentration leads to the enhancement of the YAG:Nd3+ ,Er3+ emission intensity via the Cr3+ → Ln3+ sensitization process. The quantitative analysis of the absolute brightness indicates the 30-fold enhancement of their near infrared emission upon broad white light excitation for 20% Cr3+ with respect to the non co-doped with the Cr3+ counterpart. In addition, the phonon-assisted nature of the simultaneously occurring Cr3+ → Ln3+ and Nd3+ → Cr4+ → Er3+ energy transfers leads to a 7-fold increase of the relative temperature sensitivity of the ratio of Nd3+ to Er3+ emission. In conclusion, co-doping with transition metals can enhance simultaneously the luminescence brightness and the temperature sensitivity of NIR luminescent thermometers, holding promise for sensitive and robust remote temperature sensing at physiological temperatures. The co-doping of SrTiO
3 :Mn4+ luminescent nanocrystals with trivalent lanthanide ions (Ln3+ = Lu3+ , Tm3+ , Er3+ , Ho3+ , Dy3+ , Eu3+ , and La3+ ) is demonstrated as a new strategy for significant sensitivity improvement of lifetime-based luminescent thermometers. SrTiO3 :Mn4+ :Ln3+ nanocrystals of about 25 nm diameter were prepared by the modified Pechini method and characterized using X-ray powder diffraction and electron microscopy techniques. The temperature dependence of Mn4+ emission in SrTiO3 was considerably altered by the co-doping of Ln3+ due to the cooperating effects of Mn4+ → Ti3+ and Mn4+ → Ln3+ energy transfers. A substantial enhancement of the relative sensitivity of lifetime-based thermometry as high as 145% with respect to the unco-doped nanocrystals reveals the success of the approach. The obtained values of maximal relative sensitivity for different Ln3+ co-dopants are 5.10% K−1 at 290 K for Er3+ , 5.00% K−1 at 301 K for Eu3+ , 4.84% K−1 at 303 K for Dy3+ , 4.71% K−1 at 289 K for Ho3+ , and 3.87% K−1 at 290.7 K for Lu3+ . - Abstract
Planning with hybrid domains modelled in PDDL+ has been gaining research interest in the Automated Planning community in recent years. Hybrid domain models capture a more accurate representation of real world problems that involve continuous processes than is possible using discrete systems. However, solving problems represented as PDDL+ domains is very challenging due to the construction of complex system dynamics, including non-linear processes and events. In this paper we introduce DiNo, a new planner capable of tackling complex problems with non-linear system dynamics governing the continuous evolution of states. DiNo is based on the discretise-and-validate approach and uses the novel Staged Relaxed Planning Graph+ (SRPG+) heuristic, which is introduced in this paper. Although several planners have been developed to work with subsets of PDDL+ features, or restricted forms of processes, DiNo is currently the only heuristic planner capable of handling non-linear system dynamics combined with the full PDDL+ feature set.

- Abstract
Purpose: In order to gain an insight into determinants of reported variability in immune responses to respiratory viruses in human bronchial epithelial cells (HBECs) from asthmatics, the responses of HBEC to viral infections were evaluated in HBECs from phenotypically heterogeneous groups of asthmatics and in healthy controls. Methods: HBECs were obtained during bronchoscopy from 10 patients with asthma (6 atopic and 4 non-atopic) and from healthy controls (n=9) and grown as undifferentiated cultures. HBECs were infected with parainfluenza virus (PIV)-3 (MOI 0.1) and rhinovirus (RV)-1B (MOI 0.1), or treated with medium alone. The cell supernatants were harvested at 8, 24, and 48 hours. IFN-α, CXCL10 (IP-10), and RANTES (CCL5) were analyzed by using Cytometric Bead Array (CBA), and interferon (IFN)-β and IFN-γ1 by ELISA. Gene expression of IFNs, chemokines, and IFN-regulatory factors (IRF-3 and IRF-7) was determined by using quantitative PCR. Results: PIV3 and RV1B infections increased IFN-γ1 mRNA expression in HBECs from asthmatics and healthy controls to a similar extent, and virus-induced IFN-γ1 expression correlated positively with IRF-7 expression. Following PIV3 infection, IP-10 protein release and mRNA expression were significantly higher in asthmatics compared to healthy controls (median 36.03-fold). No differences in the release or expression of RANTES, IFN-γ1 protein and mRNA, or IFN-α and IFN-β mRNA between asthmatics and healthy controls were observed. However, when asthmatics were divided according to their atopic status, HBECs from atopic asthmatics (n=6) generated significantly more IFN-γ1 protein and demonstrated higher IFN-α, IFN-β, and IRF-7 mRNA expressions in response to PIV3 compared to non-atopic asthmatics (n=4) and healthy controls (n=9). In response to RV1B infection, IFN-β mRNA expression was lower (12.39-fold at 24 hours and 19.37-fold at 48 hours) in non-atopic asthmatics compared to atopic asthmatics. BACKGROUND: Patients with systolic heart failure and anemia have worse symptoms, functional capacity, and outcomes than those without anemia. We evaluated the effects of darbepoetin alfa on clinical outcomes in patients with systolic heart failure and anemia. METHODS: In this randomized, double-blind trial, we assigned 2278 patients with systolic heart failure and mild-to-moderate anemia (hemoglobin level, 9.0 to 12.0 g per deciliter) to receive either darbepoetin alfa (to achieve a hemoglobin target of 13 g per deciliter) or placebo. The primary outcome was a composite of death from any cause or hospitalization for worsening heart failure. RESULTS: The primary outcome occurred in 576 of 1136 patients (50.7%) in the darbepoetin alfa group and 565 of 1142 patients (49.5%) in the placebo group (hazard ratio in the darbepoetin alfa group, 1.01; 95% confidence interval, 0.90 to 1.13; P=0.87). There was no significant between-group difference in any of the secondary outcomes. The neutral effect of darbepoetin alfa was consistent across all prespecified subgroups. Fatal or nonfatal stroke occurred in 42 patients (3.7%) in the darbepoetin alfa group and 31 patients (2.7%) in the placebo group (P=0.23). Thromboembolic adverse events were reported in 153 patients (13.5%) in the darbepoetin alfa group and 114 patients (10.0%) in the placebo group (P=0.01). Cancer-related adverse events were similar in the two study groups. CONCLUSIONS: Treatment with darbepoetin alfa did not improve clinical outcomes in patients with systolic heart failure and mild-to-moderate anemia. - Abstract
Luminescence (nano)thermometry is an emerging and promising field for remote temperature sensing and thermal imaging of both the surface and interior of objects. While the field is dominated by trivalent lanthanide ions (Ln
3+ ) the transition metal ions have a so far underestimated role in that regard, although their luminescence properties are far more sensitive to changes in the embedding host matrix than in the case of the Ln3+ ions. In this work, we demonstrate the potential of the abundant Fe3+ ion as a candidate for sensitive luminescence thermometry in the aluminate hosts MAl4 O7 (M = Ca, Sr, Ba). Although this ion is usually considered an efficient luminescence quencher, it is shown that a targeted control of its luminescence properties makes this activator an interesting and only scarcely investigated alternative to the Ln3+ ions. We investigated the possibility for different modes of luminescence thermometry with Fe3+ (eventually co-doped with Tb3+ as internal thermally insensitive intensity reference), which include absolute intensity, ratiometric or lifetime thermometry. By selective changes in the Fe–O bond distances, it is possible to modulate the relative sensitivity towards values as high as around 2% K−1 at room temperature in CaAl4 O7 :Fe3+ ,Tb3+ and connected relative temperature uncertainties of around 0.3%.
