The ontogenesis of the gastroesophageal mucosa involves morphological alterations related to its structure and the function of each segment. The present study describes the histogenesis of the mucus-secreting epithelium and glands of the esophagus, gizzard, and proventriculus of the chicken (Gallus gallus), and identifies alterations in the secretion pattern of glycosaminoglycans (GAG's). We analyzed 38 chicken embryos, processed the material collected following the histological routine, and then stained it with hematoxylin-eosin for the analysis of tissue structure and with Gomori's trichrome for the identification of conjunctive tissue and collagen fibers. We used the PAS histochemical technique for the analysis of neutral GAG's and the AB pH 2.5 histochemical technique for the analysis of acid GAG's. The embryos at late stage of development had the esophagus wall composed of four layers: mucosa, submucosa, muscularis, and serosa, whereas the proventriculus and the gizzard were composed of three layers: mucosa, muscularis, and serosa. In all three segments, we identified the superficial epithelium as mucus-secreting; in the esophagus this epithelium was mucus-secreting only at the initial development stages. The proventricular glands began to form at the initial development stages, whereas the tubular glands began to form in the gizzard just after the 15th day. The differences in the production of GAG's in these regions of the digestive tract are related to development stages, functions, and physiological requirements of each segment, and to the gradual adjustment of the body to the post-hatching life. La morfogénesis de la mucosa gastroesofágica implica alteraciones morfológicas relacionadas con su estructura y la función de cada segmento. El presente estudio describe la histogénesis del epitelio secretor mucinoso y de las glándulas del esófago, molleja y proventrículo del pollo (G. gallus), ademas de identificar los cambios em el patrón de secreción de glicocosaminoglicanos (GAG's). Se utilizaron treinta y ocho embriones de pollo. El material recogido fue procesado de acuerdo a la rutina histológica y posteriormente las secciones se tiñieron con hematoxilina-eosina para su análisis histológico, con tricrómico de Gomori para identificar el tejido conectivo y las fibras de colágeno y con PAS y AB pH 2,5 para el análisis GAG's neutro y ácido. En una etapa avanzada de desarrollo de los embriones, se pudo obervar en la pared del esófago cuatro capas: mucosa, submucosa, muscular y serosa, mientras que el proventrículo y molleja se muestra constituida por tres capas: mucosa, muscular y serosa. En los tres segmentos de la superficie, el epitelio se identificó como mucinoso y en el esófago sólo en la etapa inicial de desarrollo. Las glándulas del proventrículo se empiezan a formar en las primeras etapas de desarrollo, mientras que en las glándulas tubulares de la molleja comienzan su sólo después del día 15. Las diferencias en la producción de GAG's en estas regiones del tracto digestivo están relacionadas con las etapas de desarrollo, las funciones y necesidades fisiológicas de cada segmento del cuerpo y se adapta gradualmente a la vida después de la eclosión.

Objetivou-se avaliar as características morfométricas de ovários recuperados de bovinos em diferentes estádios reprodutivos. Os ovários utilizados (N=200) foram obtidos em matadouro e transportados para o laboratório, onde foram realizadas as mensurações do tamanho do órgão, dos folículos e corpos lúteos. Os pares de ovários foram classificados em função do estádio reprodutivo, da fase do ciclo estral ou do período de gestação dos animais dos quais foram recuperados. Observou-se que o tamanho do ovário diferiu (P;0,05) no tamanho do órgão entre os animais apresentando AOLC ou gestantes, porém nestes o corpo lúteo foi maior e o diâmetro do maior folículo menor (P

Trichogenes longipinnis (Britski & Ortega, 1983) is a leather fish restricted to the rivers on the shore in the southeast of Brazil. In this work, the oesophagous structure is being showed. The oesophagous appears itself short, dorsally inclinated toward pericardium and ventrally covered by liver lobe. By electron microscopy we can observe a mucosa layer formed by stratified squamous epithelium and lamina propria with stratum compactum. The epithelium is composed by three types of cells: superficial squamous cells; mucous cells, similar to the globet cells and polyhedric cells. The stratification of this epithelium provides a complete basal layer of proliferation polyhedric cells. The cells in the intermediate region of the epithelium are also polyhedric. These cells are just bellow the superficial squamous cells and packed among mucous cells.

The observations made about the stomach of Trichogenes longipinnis (Britski & Ortega, 1983) showed a rounded organ which has in its dorsal side an accessory structure - a cul de sac - that comunicates with it. The distal part of this dorsal accessory presents characteristic of an organ that exercise respiratory functions: reduction in the thickness of the mucosa and arrangement of an intrincate continuous capilar net in close contact with the superficial cells of the epithelium. In this portion there are no glands in the lamina propria. The epithelium shows a superficial squamous cels; they are very slim, and sometimes, with the capillary endothelium form a single barrier between the organ lumen and the blood. Polyhedric cells are also present in this epithelium; they are located under the squamous cells, sheltered between capillaries and basal cells.

The herein studied specimens are deposited in the Herpetological Collection of Federal University of Juiz de Fora. The animals? ovaries, epididymis and testes were removed and sent to the Histology and Embryology Laboratory (UFRRJ), where they were fixed, processed and then subjected to histological, histochemical and immunohistochemical techniques. The ovaries of these leiosauridae are functionally-active paired organs filled with ovarian follicles at different follicular development stages. These organs are covered by a simple cubic epithelium, which presents an underlying connective tissue layer called tunica albuginea. The testes are covered by the tunica albuginea and hold septa forming lobes where the seminiferous tubules are located in. The epididymis of these lizards are convolute ducts internally coated by an epithelium, whose shape ranges from simple cylindrical to simple cubic with stereocilia. The immunohistochemical analysis applied to the ovaries of the herein studied lizards showed moderate to strong reaction to the alpha smooth muscle actin in the teak layer and in endothelial cells of blood vessels. With respect to the species? testes, this cytoskeleton protein showed strong reaction in the tunica albuginea, interstitial tissue, and in endothelial cells of blood vessels. As for the epididymis, the alpha-actin showed immunohistochemical reaction in the capsule and in the interstitial tissue of the lizards. The antibody ?desmin? showed strong immunostaining in the albuginea and ovarian stroma of E. perditus, only; however, the endothelial cells in the blood vessels of the three herein analyzed species showed positive reaction to this intermediate filament. Desmin showed immunoreactivity in the testicular albuginea and in the endothelial cells of the blood vessels in the herein analyzed leiosauridae testes; however, only E. bilineatus showed immunostaining in the interstitial region. Desmin showed positive immunoreaction in the epididymis of E. perditus and E. bilineatus, only. Vimentin did not show immunostaining in the ovaries, testes and epididymis of the three analyzed lizards. The consistent results found in the current study provide additional data on the reproductive biology of the investigated species. The present study aimed to describe the histological and histochemical characteristics of the intestine and pancreas of four species of bat the family Phyllostomidae (Dryadonycteris, Capixaba, Pygoderma, bilabiatum, Chiroderma, Villosum and Phyllostomus discolor), as well as to analyze the presence of endocrine cells in these organs of the species studied, seeking to relate interspecifically the characteristics found. The animals were analyzed transferred from the zoological collection of the Microbiology Laboratory of the Department of Animal Biology (DBA) of the Universidade Federal Rural do Rio de Janeiro (UFRRJ).The organs were fixed (10% formalin) and conditioned in 70% alcohol. Subsequently, it was forwarded to the Laboratory of Histology and Embryology of UFRRJ, in which they were submitted to routine histological processing and immunohistochemical tests. Histologically the intestine and the pancreas of the species studied were similar. As for the immunohistochemical tests, two types of immunoreactive cells (IR) were identified in the intestinal mucosa of the studied species, 5HT and CCK producing cells. In the pancreas, immunoreactive cells (IR) were seen on insulin, glucagon on somatostatin and cholecystokinin. Through this study, there were differences in the distribution and relative frequency of endocrine cells in the small intestine of the animals studied. While in the pancreas species similarity was observed when compared to the histology and distribution of endocrine cells studied. Assim, o presente estudo visou descrever as caracter?sticas histol?gicas e histoqu?micas do intestino e p?ncreas de quatro esp?cies de morcegos da fam?lia Phyllostomidae (Dryadonycteris. capixaba, Pygoderma. bilabiatum, Chiroderma. villosum e Phyllostomus. discolor), bem como analisar a presen?a de c?lulas end?crinas nesses ?rg?os das esp?cies estudadas, buscando relacionar interespecificamente as caracter?sticas encontradas. Quanto aos testes imuno-histoqu?mico, dois tipos de c?lulas imunorreativas (IR) foram identificadas na mucosa intestinal das esp?cies estudadas, c?lulas produtoras de 5HT e CCK. The pancreas is a mixed gland that in involved in digestive and homeostatic processes. Understanding the morphology and physiology of the pancreas in all classes of vertebrates is fundamental to clarify this gland's function in the digestion of foods, to help shed light on nutritional factors that can improve the management and conditioning of species in their ecosystems. In the case of Brazilian species, this attention is particularly important, since many are classified as vulnerable by the IUCN (2017) due to anthropic alterations of their natural habitats. This article describes the histology and immunohistochemistry of the pancreas of two species of the Chelidae family. For this purpose, the pancreas of four specimens of Phrynops geoffroanus and four of Hydromedusa maximiliani that had been fixed in 10% formol and deposited in the collection of Juiz de Fora Federal University were routinely processed for histology to obtain sections with thickness of 5 ?m. The histology of the pancreas of P. geoffroanus and H. maximiliani is characterized by an endocrine portion dispersed among the pancreatic acini, forming or not forming endocrine groups similar to pancreatic islets. No connective tissue capsule is observed separating the pancreatic islets from the surrounding exocrine portion. The immunohistochemical analysis revealed four types of endocrine cells in P. geoffroanus: cells producing insulin, glucagon, somatostatin and pancreatic polypeptide. In H. maximiliani, only three cell types were found, producing insulin, glucagon and somatostatin. The cellular organization of the endocrine groups similar to pancreatic islets is different in the two species. In P. geoffroanus, the insulin-producing cells are positioned in the center, while those producing glucagon occupy the periphery. In turn, in H. maximiliani, these to cell types are interspersed, generally around the blood vessels. These observations will help understand the digestive physiology of the two species and provide data for comparative analyses with other freshwater turtles A compreens?o dos processos fisiol?gicos nos vertebrados ? essencial para obten??o de dados sobre a biologia das esp?cies. In Brazil, according to the National Cancer Institute, there were estimated 518,510 new cases of cancer for the years 2012 and 2013. From this estimation, 52,680 will correspond to breast cancer (BC), with an estimated risk of 52 new cases per 100,000 women. BC is one of the most common cancers worldwide. Today it is known that the treatment of the BC may lead to the emergence of different late adverse effects, including osteoporosis. One of the main causes of osteoporosis is the early menopause, which occurs with decreasing the serum estrogen concentration. This study aimed to evaluate the effects on bone matrix induced by chemotherapy , simulating a treatment for BC in Wistar rats .Wistar rats, approximately 3 months old, were divided into a control group and the group receiving polichemotherapy with docetaxel + cyclophosphamide (TC). Chemotherapy was administered in 4 cycles, with an interval of 1 week between them. The rats were euthanized 5 months after the end of treatment, so that the late effects could be evaluated. Several studies were performed: dosage of serum estradiol levels, histological tests by immunohistochemistry, micro X-ray fluorescence, micro-computed tomography and also transmission and scanning electron microscopy.Analyzing the results together, it is suggested that the initial step in the development of osteoporosis caused by multidrug TC is the reduction in the ovarian function. This event leads to decreased serum concentration of estrogen, which causes uterine atrophy. Concomitant to these facts, the TC causes a reduction in the concentration of zinc in the bone tissue. These results associated cause an imbalance in the osteoblast/osteoclast ratio in the bone tissue. The reduction in estrogen leads to decreased apoptosis of osteoclasts, while the reduction of zinc inhibits osteoblast function. This imbalance affects the bone turnover in order to increase bone resorption. Thus, the percentage of bone is reduced, and the trabeculae become thinner and spaced. The endpoint of this process is osteoporosis.The administration of docetaxel and cyclophosphamide together leads to decreased bone mass in the trabecular, thinning and increasing the space between them. These observations suggest that the rats treated with TC developed osteoporosis. We conclude that both the estrogen and the zinc play a fundamental role in the development of this disease after chemotherapy with TC. A Organização Mundial de Saúde (OMS) estima para 2030, 27 milhões de casos incidentes de câncer. No Brasil, segundo o Instituto Nacional do Câncer, foram estimados 518.510 casos novos de câncer para os anos de 2012 e 2013. Dessa estimativa, 52.680 correspondem ao câncer de mama (CM), com um risco estimado de 52 novos casos a cada 100.000 mulheres. O câncer de mama é um dos tipos de câncer mais comuns no mundo todo. Hoje se sabe que o tratamento para o CM pode levar ao surgimento de diferentes efeitos adversos tardios, entre eles a osteoporose. Uma das principais causas de surgimento da osteoporose é a menopausa precoce, que ocorre através da diminuição da concentração de estrogênio sérico. Este trabalho teve como objetivo avaliar os efeitos na matriz óssea induzidos pela quimioterapia, simulando um tratamento para o CM, em ratas Wistar.Ratas Wistar, com aproximadamente 3 meses de idade, foram divididas em: grupo controle e grupo que recebeu quimioterapia com poliquimioterápico docetaxel + ciclofosfamida (TC). A quimioterapia foi administrada em 4 ciclos, com intervalo de 1 semana entre eles. Os ratos foram submetidos à eutanásia 5 meses após o término do tratamento, para que os efeitos tardios pudessem ser avaliados. Vários estudos foram conduzidos: dosagem sorológica de estradiol, ensaios histológicos através de imunohistoquímica, micro-fluorescência de Raios-X, micro-tomografia computadorizada. Além de microscopia eletrônica de transmissão e varredura.Analisando os resultados obtidos em conjunto, sugere-se que a etapa inicial para o desenvolvimento da osteoporose, causada pelo poliquimioterápico TC, seja a diminuição da função ovariana. Este evento leva à diminuição da concentração de estrogênio sérico, o que causa a atrofia uterina. Concomitante a estes fatos, o TC causa redução na concentração de zinco no tecido ósseo. Estes resultados associados causam um desequilíbrio na relação osteoblastos/osteoclastos no osso. A redução do estrogênio leva à diminuição da apoptose de osteoclastos, enquanto que a redução do zinco inibe a função dos osteoblastos. Este desequilíbrio interfere no turnover do osso, de forma a aumentar a reabsorção óssea. Deste modo, o percentual de osso fica reduzido, as trabéculas tornam-se mais finas e espaçadas. O endpoint deste processo é a osteoporose.A administração dos poliquimioterápicos docetaxel e ciclofosfamida em conjunto leva a diminuição da massa óssea, a adelgaçamento das trabéculas e o aumento do espaço entre elas. Estas observações sugerem que realmente as ratas tratadas com TC apresentam osteoporose. Concluímos que tanto o estrogênio quanto o zinco têm papel fundamental no desenvolvimento desta patologia após quimioterapia com TC.

We used 12 specimens of each reservoir, which are collected using gillnets and sacrificed by hypothermia. After dissection occurred to the immediate removal of GIT. Subsequently, fragments of the stomach and gut were fixed for 8 hours in Bouin liquid and sent to the Laboratory of Histology and Embryology of UFRRJ, where they were subjected to histological processing and histochemical routine with periodic acid + reactive Schiff (PAS) and Alcian Blue pH 2.5 (AB) to posterior the realization of immunohistochemical study. Histologically the stomach of A. bimaculatus showed two distinct regions (glandular and non glandular) and gut was divided into anterior and posterior segment, being the only regions of the stomach and gut PAS reactive to reactive to PAS and AB respectively. In immunohistochemical study of three types of cells immunoreactive (IR) IR studied, only cells to 5-HT were identified in the stomach region (epithelia and glands) and gut. GAS cells to IR and SOM were only detected in the stomach. GAS cells to IR was detected only in the gastric glands. The regional distribution and frequency of endocrine cells varied across regions of the GIT and also among the specimens of each reservoir. It was noted that there was no difference in histological GIT of A. bimaculatus in the comparison of the three reservoirs and that it is compatible with your omnivorous. By immunohistochemistry we found that the regions of the stomach of A. bimaculatus occurred a frequency of almost exclusively three types of endocrine cells studied, which may indicate that this organ is the main site for digestion of food of this species, being also observed a similarity in the amount and distribution of these cells in the GIT of specimens coming from the different reservoirs. Knowledge of ecology and evolution is key to appropriate management, conservation and improvement of ecosystems. Therefore, clarification of the morphology of the digestive system is fundamental because this reflects to pressures exercised by the environment on the organism. The adaptation to different environmental niches leads to anatomic and physiological changes, mainly in the digestive system, among and within the different reptile orders. This work describes the histology histochemistry and immunohistochemical of the digestive tube of Phrynops geoffroanus, to relate the characteristics of the digestive organs to the species? eating habit. The esophagus, stomach and intestine of four specimens fixed in 10% formal and deposited in the reptile collection of Juiz de Fora Federal University were processed by routine histological techniques to obtain 5- ?m sections. These were stained with hematoxylin-eosin (HE), periodic acid-Schiff (PAS) and Alcian Blue (AB) pH 0.4 and 2.5. The tube is formed by the mucosa, submucosa, muscular and adventitia or serosa layers. The mucosa layer of the esophagus and stomach is lined with a simple cylindrical epithelium with mucus-secreting cells, with intraepithelial glands inserted in part of the esophagus and gastric fossettes emptying into tubular glands in the stomach. The stomach is divided into an anterior, middle and posterior region, according to the depth of the fossettes and the concentration of gastric glands. The intestine is lined by a simple cylindrical epithelium with striated boundaries and goblet cells and is divided into an anterior and posterior region, according to the pattern of folds of the mucosa layer and the number of goblet cells. Reactivity to PAS and AB was observed in the entire tube. The muscularis mucosa is present in all the segments. The muscular layer is formed by two sublayers of smooth muscle, except for the posterior part of the stomach. Immunoreactive (IR) cells to anti-serotonin (5-HT) are present among the epithelial cells and at the base of the intraepithelial glands in the esophagus and epithelium of the stomach and intestine, while at the base of gastric glands there are IR sells to anti-serotonin (5-HT), anti-somatostatin (SOM) and anti-insulin (INS). There was no reaction observed to anti-gastrin (GAS), anti-glucagon (GLUC) and anti-pancreatic polypeptide (PP). This study will help understanding of the digestive physiology of P. geoffroanus and provide data for comparative analyses with other turtles. The ontogenesis of the gastroesophageal mucosa involves morphological changes that very according to the structure and function of each organ and events such as cell proliferation, differentiation and apoptosis. The aim of this study was to describe the histogenesis of the gastroesophageal mucosa of G. gallus, identify the alterations in the secretion pattern of glycosaminoglycans (GAGs), analyze the expression of protein p53 and correlate it with the morphological changes during development. Thirty-eight chicken embryos were used, classified as in stages 37 to 45 according to Hamburger and Hamilton (1951). The material collected was processed according to routine histological techniques and stained with hematoxylin-eosin for analysis of the tissue structure, with Gomori?s trichrome for identification of the conjunctive tissue and mainly collagen fibers, as well as with the histochemical techniques PAS and AB pH 2.5 for analysis of neutral and acid GAGs, respectively. For the immunohistochemical technique, 26 specimens were used, 24 embryos and the thymus of 2 adult animals. The epithelium covering the gastroesophageal mucosa was found to secrete mucus, while in the esophagus, the mucosa only secreted mucus in the initial stages of development. The esophageal and proventricular glands start to form in the initial stages of development (stage 37), while in the gizzard the tubular glands start to form only after the 15th day of incubation (stage 41). The differences in production of GAGs in these regions of the digestive tract are related to the advancement of morphogenesis in the developmental stages, the functions and physiological needs of each segment and the progressive adaptation of the organism to life after hatching. Protein p53 was expressed at crucial moments of development, in the esophagus (during remodeling of the cells to form the esophageal glands, stage 42 to 45), in the proventriculus (during differentiation of the mucosal epithelium, stage 39 to 45) and in the gizzard (during formation of the tubular glands, stage 42 to 45). The expression of p53 in the gastroesophageal mucosa occurs mainly in the stages when tissue remodeling starts and accompanies the differentiation of these cells until formation of glandular structures with characteristics similar to those in the adult bird. The differences in the production of GAGs in these regions of the gastroesophageal tract are related to development stages, functions, and physiological requirements of each segment, and to the gradual adjustment of the body to the post-hatching life. These species are often found in domiciles in the state of Rio Grande do Sul, Brazil. Comparisons were made between the three species by morphology using the technique of optical microscopy and scanning electron microscopy. We illustrate the morphostructural differences adults in scutellar region and only one species showed heart-shaped median depression, T. circummaculata. T. carcavalloi and T. rubrovaria presented in the form of W. It was also observed concentration of sensilla on the surfaces of the scutellum of these three species. In analyzes of the cephalic region, the stridulatory sulcus is "U" shaped in T. circummaculata and "V" shaped in T. carcavalloi and T. rubrovaria, the buccula T. carcavalloi and T. rubrovaria presented in the form of "U" and T. circummaculata "V" shaped. The rostrum of T. carcavalloi, T. circummaculata and T. rubrovaria presented in apex two lateral rifts 1+1. Were analyzed and shown significant differences in the length, width and diameter of the body and operculum of the eggs of T. rubrovaria, T. carcavalloi and T. circummaculata. In the morphological analysis of antennas T. carcavalloi it was verified the occurence in both sexes more than 14 trichobotrium, in the thrird and fourth antennal articles. T. carcavalloi presented on head 1+1 post-ocelares spots. In the legs, a pair of subapical denticles in femures of male and female was found. Spongy fossette were found only in the males, in the t?bias of the anterior and medium legs. The results of morphological T. carcavalloi showed that some structures are important for the specific characterization, such: antennas, spongy fossette and subapical denticles. Morphological studies placed T. carcavalloi in "complex infestans" and "subcomplex rubrovaria" as well as T. rubrovaria, while T. circummaculata was placed in the "complex circummaculata". The phylogeny of the group consists of 16 species of triatomines was reassessed with the inclusion of T. carcavalloi by Bayesian analysis of mtDNA sequences of subunits 12S and 16S ribosomal RNA, and cytochrome oxidase I gene (COI). The phenotypic relationship between T. carcavalloi related triatomines was also inferred from morphometry. Phylogenetic results indicate that T. carcavalloi is a sister species to T. rubrovaria, and both were relocated in close relationship with T. circummaculata. Morphometrics studies confirmed the proximity of T. carcavalloi, T. rubrovaria and T. circummaculata, placing the latter species the "complex infestans" and "subcomplex rubrovaria". Regarding biological parameter, the average incubation period of the eggs in T. carcavalloi was 22.7 days. The highest number of eggs/female/week was observed during the hottest months. The first meal took place at 3.13 days after hatching. The intermoulting period N1-N2 was on average 18.52 days, N2-N3 62.77, N3-N4 86.93, N4-N5 119.05 and N5?adult 193.43. The average of feedings during the nymphal development was 13.4. Resistance to fasting in all nymph and adult (males and females) stages indicated that nymphs of 3rd, 4th and 5th stages presented higher resistance than adults, and in this case males were less resistant than females. The highest mortality rate was registered in N3 (22.2%). The average adult survival was 25.6 weeks. The total life cycle was highly long with average of 503.4 days. This is the first report on the biology of T. carcavalloi fed mice. This study enable expand the bionomic knowledge of this species, besides contribute to better maintenance colony and thus understand the possible participation in the transmission of T. cruzi. Nesta tese foram estudadas tr?s esp?cies do ?subcomplexo rubrovaria?: T. carcavalloi, T. circummaculata e T. rubrovaria que possuem h?bitos silvestres, mas com potencialidades na transmiss?o da doen?a de Chagas. Estas esp?cies s?o encontradas com frequ?ncia em domic?lios do estado do Rio Grande do Sul, Brasil. Foram feitas compara??es entre as tr?s esp?cies atrav?s da morfologia usando a t?cnica da microscopia ?tica e eletr?nica de varredura. Foram ilustradas as diferen?as morfoestruturais dos adultos na regi?o escutelar e apenas uma esp?cie apresentou a forma codiforme na depress?o mediana, T. circummaculata. J? T. carcavalloi e T. rubrovaria apresentaram a forma de W. Foi tamb?m observada a concentra??o de sensilas nas superf?cies do escutelo das tr?s esp?cies. Nas an?lises da regi?o cef?lica, o sulco estridulat?rio ? em forma de ?U? em T. circummaculata e em forma de "V" em T. carcavalloi e T. rubrovaria, as b?culas de T. carcavalloi e T. rubrovaria apresentaram a forma de ?U? e T. circummaculata de ?V?. J? o rostro de T. carcavalloi, T. circummaculata e T. rubrovaria apresentaram duas fendas laterais 1+1 no ?pice. Foram analisadas e evidenciadas diferen?as significativas no comprimento, largura e di?metro do corpo e do op?rculo dos ovos de T. rubrovaria, T. carcavalloi e T. circummaculata. Na an?lise morfol?gica das antenas de T. carcavalloi verificou a ocorr?ncia tanto em machos quanto em f?meas de mais de 14 tricob?trias, no terceiro e quarto art?culos antenais. T. carcavalloi apresentou na cabe?a 1+1 manchas p?s-ocelares. Nos f?mures das patas de ambos os sexos foi encontrado um par de dent?culos subapicais. As fossetas esponjosas foram encontradas somente nos machos, nas t?bias das patas anteriores e medianas. Os resultados morfol?gicos de T. carcavalloi evidenciaram que algumas estruturas s?o importantes para a caracteriza??o espec?fica, como: antenas, dent?culos subapicais e fossetas esponjosas. Estudos morfol?gicos colocaram T. carcavalloi no ?complexo infestans? e no ?subcomplexo rubrovaria? assim como T. rubrovaria, enquanto T. circummaculata foi colocado no ?complexo circummaculata". A filogenia do grupo composto de 16 esp?cies de triatom?neos foi reavaliado com a inclus?o de T. carcavalloi por an?lise Bayesiana utilizando sequ?ncias de mtDNA de subunidades 12S e 16S do RNA ribossomal, e genes do citocromo oxidase I (COI). A rela??o fenot?pica entre T. carcavalloi e triatom?neos afins tamb?m foi inferida a partir da morfometria. Resultados filogen?ticos indicaram que T. carcavalloi ? uma esp?cie irm? de T. rubrovaria, e ambos foram recolocados em estreita rela??o com T. circummaculata. Estudos morfom?tricos confirmaram a proximidade entre T. carcavalloi, T. rubrovaria e T. circummaculata, colocando a ?ltima esp?cie no ?complexo infestans? e no ?subcomplexo rubrovaria?. Em rela??o ao par?metro biol?gico, o per?odo m?dio de incuba??o dos ovos em T. carcavalloi foi de 22,7 dias. O maior n?mero de ovos / f?mea / semana foi observado durante os meses mais quentes e os ovos que n?o eclodiram eram f?rteis. O primeiro dia de repasto sangu?neo foi de 3,13 dias ap?s a eclos?o, em m?dia. O per?odo de intermuda N1-N2 foi em m?dia de 18,52 dias; N2-N3 foi 62,77 dias; N3-N4 foi 86,93 dias; N4-N5 foi 119,05 dias e N5-adulto foi 193,43 dias. A m?dia geral de alimenta??es durante todo o desenvolvimento ninfal foi de 13,4. Os resultados logrados para resist?ncia ao jejum indicaram que as ninfas de 3, 4 e 5 est?gios apresentam maior resist?ncia do que os adultos, e neste caso os machos foram menos resistente do que as f?meas. A taxa de mortalidade mais alta foi registrada em N3 (22,2%). A sobreviv?ncia m?dia foi de 25,6 semanas para os adultos. O ciclo de vida total foi longo, com m?dia de 503,4 dias. Este foi o primeiro relato sobre a biologia de T. carcavalloi alimentados com camundongos. Esse estudo possibilitou ampliar o conhecimento bion?mico desta esp?cie, al?m de contribuir para melhor mantuten??o de col?nias e dessa forma entender a poss?vel participa??o na transmiss?o do T. cruzi.

To do so it used histological techniques, immunohistochemistry and morphometric analysis as well as a standardized experimental model for the induction of injury to the tibia of rats as proposed by Yuehuei and Friedman7. It is important to standardize a model of fracture induction for further investigation of the involvement of cells and extracellular matrix components in the fracture repair process, whereas the healing time depends significantly on the nature and type of lesion produced. Forty (n = 40) Wistar rats were subjected to fracture. The animals were divided into eight (n = 8) groups of five (n = 5). Each subgroup was observed after 12, 24, 48, 72, 96, 144, 192 and 240 hours of fracture (12 to 10 days). Immediately afterwards, the fractures were classified according to the system of international classification of fracture by Muller100, AO (Association for Osteosynthesis). Simple fractures were found in 86% of the total, among them, 68% were transverse and 18% were oblique. Complex fractures were found in 14% of the cases, among them 8% were irregular and 6% were segmental. These results demonstrated that the device enables researchers to standardize the type of fracture by X-ray, marked by the line separating the bone fragments. The qualitative results of the cells and extracellular matrix components of TGF-β, VEGF, PCNA, collagen I and II, osteopontin, proteoglycans, elastic fibers system with resorcin funcsin of Weigert, as well as the morphometric results suggest that the repair process of the fracture is not homogeneous for all components. Expression of the extracellular matrix components and cell proliferation modulation significantly depends on the local stresses generated by the type of the fracture. Such type can be decisive in determining time duration for bone regeneration and quality of the biomechanical properties restoration. Our findings may contribute to better understanding of bone fracture repair and for new therapeutic approaches that consider the biomechanical properties of bone tissue in repair in its different stages O propósito do presente trabalho foi investigar a participação da proliferação celular e da expressão dos componentes da matriz extracelular na cascata de eventos do processo de reparo da fratura óssea, empregando as técnicas histológica, imunohistoquímica e morfométrica, em um modelo experimental padronizado para a indução da lesão na tíbia de ratos a partir do método empregado por Yuehuei e Friedman7. É importante padronizar um modelo de indução da fratura, para posterior investigação da participação das células e dos componentes da matriz extracelular no processo de reparo da fratura, considerando que o tempo de consolidação depende significantemente da natureza e do tipo da lesão produzida. Quarenta (n = 40) ratos Wistar foram submetidos a fratura . Os animais foram avaliados em oito (n = 8) grupos de cinco (n = 5) animais, cada grupo emperimental com 12, 24, 48, 72, 96, 144, 192 e 240 horas após a fratura (12h até 10 dias). As fraturas foram classificadas de acordo com o sistema de classificação internacional de fratura de Muller100, AO (Associação para Osteosíntese). Foram encontradas fraturas simples em 86% do total, sendo 68% de fraturas transversas e 18% de fraturas obliquas, 14% do total de fraturas foram complexas, sendo 8% de fraturas irregulares e 6% de fraturas segmentares. Esses resultados demonstram que o aparelho permite padronizar radiológicamente o tipo de fratura, caracterizado pela linha que separa os fragmentos ósseos. Os resultados qualitativos dos componentes da matriz extracelular para TGF-β, VEGF, colágeno I e II, osteopontina, proteoglicanos, fibras do sistema elástico com a coloração de resorcina funcsina de Weigert, e para proliferação celular pelo PCNA, assim como os resultados morfométricos, sugerem que a modulação da expressão dos componentes da matriz extracelular e a proliferação celular durante o processo de reparo da fratura não é homogênea para todos os componentes teciduais, dependendo significantemente das tensões locais geradas pelo tipo da linha de fratura que pode ser determinante no tempo de regeneração do osso e na qualidade da restauração das propriedades biomecânica. For the chosen species no study is available in literature. There is no doubt that Diffuse Neuroendocrine System has an important role in the gastrointestinal function, however their phenotipic regulation in not completely elucidated, which indicates the importance of carrying out this search, which investigate species with different food habits. Whereas the basic premise of biological studies should be the Comparative Biology integrating different areas of knowledge, we sought to describe histologically the gastrointestinal tract wall of Asio clamator and Caracara plancus to evidenciate histochemically the presence of acids and neutral glycosaminoglycans and locate by immunohistochemistry Diffuse Neuroendocrine System cells with their respective peptides regulators. We hope demonstrate that this cells and their peptides are found in Falconiformes and Strigiformes species. To achieve these purposes, it was conducted an histological and histochemical (using Schiff periodic acid technique and alcian blue pH 2,5) study involving the description of gastrointestinal tract organs of two individuals from each of the following species Caracara plancus (Falconidae) e Asia clamator (Strigidae). Moreover, we detected and identified by immunohistochemistry cells which secrete gastrin, glucagon, somatostatin and serotonin. The cells were found in greater quantities in the proventriculus (stomach chemical) of the species investigated. The difference in habit (night and day) did not affect the distribution of cells of the Diffuse Neuroendocrine System. The results demonstrate that gastrointestinal tract wall organization is typical, as regards the mucosa, submucosa, muscle and serosa, since it presents adventitia in the esophagus and the ventricle without submucosal. The esophagus shows a stratified squamous epithelium without keratin containing few celular layers in e many of them in southern carcara. The mucosal glands of striped owl are tubular and can show ramifications. The mucosal glands in southern carcara are ?tubuloacinosas?. With respect to proper lamina, muscularis and adventitia, they are typical. The mucosal glands of esophagus were stained by histochemical techniques. Immunoreactive cells to sera used were not detected in esophagus. Proventriculus histological description is typical. The histochemical methods showed an mucosecretor epithelium. It was observed immunoreactive cells to gastrin, glucagon, serotonin and somatostatin in submucosal glands of striped owl. Immunoreactive cells to glucagon and somatostatin were found in southern carcara. In ventricle the histological description is typical. The histochemical showed a secretion product synthesized by tubular glands of mucosal region. It was not detected immunoreactive cells to anti-sera in any bird. The histological description of intestine is typical. The histochemical revealed the globet cells. In striped owl it was observed immunoreactive cells to glucagon and serotonin within epithelium and Lieberk?hn glands. Immunoreactive cells to glucagon was found in Lieberk?hn glands. Based on these results, it was found that different segments in the gastrointestinal tract of striped owl and of southern caracara have the same general structure of other vertebrates, and more specifically for avian; which cells producing glycosaminoglycans neutral and acidic are also observed in these species; and the location of endocrine cells in the gastrointestinal tract were essentially similar to those of other birds studied. Although most studies have shown the benefits of exercise on tendon regeneration, controversy still exists concerning non-steroidal antinflammatory drug (NSAID) effects in this context. Several experimental models have been used for the study of Achilles tendon injury. In this study, we describe the surgical technique of right Achilles tenotomy in rats and subsequently evaluate the effects of NSAID and aerobic exercise, in an isolated fashion and combined, on cell proliferation and biomechanical aspects of the Achilles tendon after tenotomy. Experimental study with 156 male Wistar rats with an average age of 3 months and with average weight of 300g. Surgical procedures were performed under light microscopy, after anesthesia with thiopental. A five millimeters posterior longitudinal incision was created, proximally directed, starting five millimeters proximal to the posterior calcaneal tuberosity. A complete tenotomy of the Achilles tendon was performed, seven millimeters away from the calcaneal tuberosity. The plantaris tendon was preserved. We used Hematoxilin and Eosin, Picrosirius-red and Weigert s Resorcin-fucsin to observe general tendon healing, especially regarding collagen and elastic fibers. After tenotomy, half of the rats received an intramuscular injection of tenoxican for 7 days and exercise was initiated on the 8th day for half the animals of each group. Rats were randomly divided into four treatment groups: A) no NSAID and no exercise; B) NSAID plus exercise; C) no NSAID, with exercise; D) NSAID and no exercise. Animals were sacrificed at 1, 2, 4 and 8 weeks after the tenotomy and cell proliferation was evaluated by immunohistochemistry for PCNA, biomechanical evaluation was performed with ultimate load and gait cycle analysis was also carried out. We used the test of analysis of variance, the Kruskal-Wallis test and also, Bonferroni method, in the R Project program 2.11.1. The mean operative time was one minute and 24 seconds, without complications observed until the 8th postoperative week. Histological studies showed cellular proliferation and fibrilogenesis at two weeks, with decreased amounts of cellularity and elastic fibers at the 8th week, besides changes in structural organization of collagen fibers. The highest intensity of PCNA immunostaining was found at 2 weeks in all groups except for group A (control) that had the highest intensity at 1 week. Animals submitted to exercise had significantly higher (P = 0.02) ultimate loads at 8 weeks after injury. The animals that received NSAID presented with a more stable gait cycle. The surgical technique described for complete Achilles tenotomy, under light microscopy and sparing the plantaris tendon, is simple and quick, shows signs of normal healing process, and it is easily reproducible in rats. Aerobic exercise, initiated early after a complete Achilles tendon tenotomy, was beneficial to the biomechanical aspects of the tendon during regeneration and the combined use of NSAID improved the gait s characteristics, which could be protective against reruptures Ruptura do tendão calcâneo é uma das lesões tendíneas mais frequentes. Embora a maioria dos trabalhos sugira que o exercício seja benéfico na cicatrização tendínea, não há consenso sobre o efeito do antiinflamatório neste contexto. Trabalhos experimentais tentam reproduzir lesão aguda deste tendão, em diferentes espécies animais. Neste estudo, descrevemos uma técnica de tenotomia completa do tendão calcâneo direito em ratos e, em seguida, avaliamos os efeitos do uso do antiinflamatório e do exercício aeróbico, isoladamente e em combinação, sobre a proliferação celular e o perfil biomecânico do tendão calcâneo, durante o processo de cicatrização após tenotomia. Estudo experimental com 156 ratos machos adultos, da raça Wistar, com idade média de 3 meses e peso médio de 300g. Após anestesia com tiopental e com auxílio da microscopia de luz, foi realizada incisão longitudinal posterior de cinco milímetros, em direção proximal, a partir da tuberosidade posterior do calcâneo da pata direita do rato. Foi feito corte transversal do tendão calcâneo, a sete milímetros da tuberosidade do calcâneo, com preservação do tendão plantar. Utilizamos as técnicas de Hematoxilina e Eosina, Picrosirius-red e Resorcina-fucsina de Weigert para avaliação da cicatrização tendínea e das fibras dos sistemas colágeno e elástico. Após a tenotomia, metade dos animais receberam tenoxicam intramuscular por 7 dias e no 8o dia iniciou-se protocolo de exercício em esteira na metade de cada grupo. Os ratos foram divididos aleatoriamente em 4 grupos de tratamento: A sem antiinflamatório E sem exercício (controle); B com antiinflamatório E com exercício; C sem antiinflamatório E com exercício; D com antiinflamatório E sem exercício. Os animais foram eutanasiados com 1, 2, 4 e 8 semanas após a tenotomia, para avaliação histológica pelo PCNA, e biomecânica através do teste de resistência à tração e da medida do ciclo locomotor. Foram realizados análise de variância, teste de Kruskal-Wallis e o método de Bonferroni, no programa R Project, versão 2.11.1. O tempo cirúrgico médio foi de 1 minuto e 24 segundos, sem complicações observadas até a 8a semana pós-operatória. Observamos proliferação celular e fibrilogênese com duas semanas, e diminuição da celularidade e das fibras elásticas na 8a semana, além de mudanças na organização estrutural do sistema colágeno. Encontramos pico da imunomarcação com PCNA na 2a semana em todos os grupos, exceto no grupo A, cujo pico aconteceu com 1 semana da tenotomia. Evidenciamos resistência à tração significativamente maior (p=0,02) nos ratos submetidos ao exercício, 8 semanas após ruptura. Nos grupos com antiinflamatório, observamos um ciclo locomotor mais estável durante todo o tempo avaliado. Consideramos a técnica cirúrgica experimental de tenotomia completa do tendão calcâneo, realizada com auxílio da microscopia de luz e preservação do tendão plantar, simples, rápida, com sinais de cicatrização tendínea normal e de fácil reprodução em ratos. O exercício aeróbico, iniciado precocemente após tenotomia completa do tendão calcâneo, é significativamente benéfico na sua recuperação biomecânica e o uso combinado com antiinflamatório confere maior estabilidade na marcha, o que pode proteger contra rerruptura tendínea em ratos

The aim of this work was study endocrine pancreas of bats with different feeding habits (hemataphagous D. ecaudata, nectarivorous A. Geoffroyi and insetivorous P. personatus) by histological, morphometric and stereological methods. Six animals was used in each group and they were collected in the caverna casa de Pedra (Sergipe). The pancreas was processed through routine histological techniques for paraffin embedding. Histologically, components of the exocrine pancreas (acini and their secretory ducts associated) and endocrine pancreas (pancreatic islet) were identified. Immunoreactive cells (IR) to insulin, glucagon and somatostatin were found isolated scattered in the exocrine pancreas and, in the tissue near the pancreatic ducts. It was also identified IR cells to insulin, glucagon and somatostatin. In the pancreatic islets, it was to indentifided cells with cytoplasmic processes. The total mass of pancreatic tissue in body weight does not exceed 0.6% and this relationship was higher in the insectivorous bats (0.6%) and lowest in nectarivorous bats (0.3%). The pancreatic islets diameter varied between species, being 88.16 ?m in the nectararivorous bat, in the vampire bat at 56.94 ?m and 40.47 ?m in the insectivorous bat. The stereological analysis showed a volume density (Vv) of the endocrine pancreas of 3.4% in the insectivorous bat, 6.1% in the nectarivorous bat and 4.1%. in the vampire bat. The islet mass was 0.15 in the insectivorous bat, 0.24 in the nectarivorous bat and 0.48 in the vampire bat. The volume density of ? cells was (25.8%, 14.7, 16.2%), ? cells (35.5%, 8.7%, 54.8%) and ? cells (3.9 %, 6.2%, 14.3%) in the P. personatus, A. geoffroyi and D. ecaudata bats, respectively. The differences observed between species in the various parameters analyzed may reflect inter-specific differences, as well as morphological adaptations of species to their eating habits. Este trabalho visou estudar o p?ncreas end?crino de morcegos com diferentes h?bitos alimentares (hemat?fago D. ecaudata, nectar?voro A. geoffroyi e inset?voro P. personatus), atrav?s de an?lise histol?gica, morfom?trica e estereol?gica. Foram utilizados ao todo seis animais de cada grupo, coletados na caverna casa de Pedra (Sergipe). O p?ncreas foi processado atrav?s das t?cnicas histol?gicas de rotina para inclus?o em parafina. Histologicamente, foram identificado os componentes ex?crinos (?cinos secretores e seus ductos associados) e end?crinos (ilhotas pancre?ticas) do p?ncreas. Foram observadas c?lulas imunoreativas (IR) ? insulina, glucagon e somatostatina espalhadas isoladamente no p?ncreas ex?crino e, na regi?o de tecido conjuntivo pr?ximo aos ductos pancre?ticos, foram identificadas c?lulas IR ? insulina, glucagon e somatostatina. C?lulas com a presen?a de processos citoplasm?ticos foram visualizadas nas ilhotas. A massa de tecido pancre?tico total sobre a massa corporal n?o ultrapassa a rela??o de 0,6%, sendo esta rela??o maior nos morcegos inset?voros (0,6%) e menor nos morcegos nectar?voros (0,3%). The chemical communication sign may have a highest importance in the space orientation and sexual behavior displayed by the chelonian during the reproductive period. There are not histological and morphological studies of the Brazilian chelonian species tegumental appendices, like Hydromedusa maximiliani and Phrynops geoffroanus. So, the actual study had the goal to describe and compare morphologically and histologically the tegumental appendices from two Brazilian tortoise species which belong to the Chelidae family. For this study, it was used adult specimens from the Cole??o Herpetol?gica do Departamento de Zoologia da Universidade Federal de Juiz de Fora of H. maximiliani (2 males and 2 females) and P. geoffroanus (4 males and 4 females), totalizing 12 specimens. The analysis of the external surface neck, head and chin region was done. The animals were dissected using stereoscopic microscope to take samples out of the epidermis and dermis from the neck and head region. To the microscopic analysis, it was done the routine techniques that are used in the histological studies. In P. geoffroanus, the keratinized epidermis layer is thicker when compared to the same epidermis layer of H. maximiliani. It was observed that in P. geoffroanus and H. maximiliani these regions may present epidermis invaginations with the detachments of the most external keratin layers. The histological and histochemical study done in the epidermis cuts of the specimens H. maximiliani and P. geoffroanus haven?t evidenced the presence of submandibular gland. The PAS method hasn?t shown the presence of neutral mucopolysaccharides in this submandibular gland, which are products part of the secretions produced by the exocrine glands. In this study, it was observed that the dewlap presented in P. geoffroanus are cylindrical tegumental appendices presented in the mandibular symphysis posterior region, which occurs in pairs, constituting of epidermis and dermis, and with a great concentration of nervous terminations. In the actual study, the P. geoffroanus specimens macroscopically presented conical shape tubers, rarely pointed and also with verrucous shape, visible in the dorsal and lateral region, from the anterior part up to the posterior part of the neck, not presented in the ventral region of the neck, which presents slots, grooves and depressions in the epidermis. In the actual study, it wasn?t observed morphological and histological differences regarding the tubers when compared by gender. In H. maximiliani the tubers are irregularly distributed in the lateral and dorsal regions of the neck, being absent in the region of the head and in the ventral region of the neck. They present pointed and tray shape, different from what was observed in the P. geoffroanus specimens. The tubers presented in the two studied species are different in macroscopic terms, but all of them present a random disposition and are also absent in the ventral region of the neck. For the two types of tegumental annexes (tubers, dewlap) analyzed in this study, it was verified that there are not differences between the genders. S?o inexistentes os estudos histol?gicos e morfol?gicos dos ap?ndices tegument?rios das esp?cies de quel?nios brasileiros, como Hydromedusa maximiliani e Phrynops geoffroanus. Dessa forma, o presente estudo teve como objetivos descrever e comparar morfol?gica e histologicamente os anexos tegumentares de duas esp?cies brasileiras de c?gados pertencentes ? Fam?lia Chelidae. Para esse estudo foram utilizados esp?cimes adultos de H. maximiliani (dois machos e duas f?meas) e P. geoffroanus (quatro machos e quatro f?meas), totalizando 12 esp?cimes, provenientes da Cole??o Herpetol?gica do Departamento de Zoologia da Universidade Federal de Juiz de Fora Foi realizada a an?lise da superf?cie externa da regi?o do pesco?o, cabe?a e da regi?o mentoniana. Os animais foram dissecados sob microsc?pio estereosc?pico para retirada de amostras da epiderme e derme das regi?es da cabe?a e pesco?o. Para an?lise microsc?pica foram realizados as t?cnicas de rotina utilizadas nos estudos de histologia. Em P. geoffroanus a camada queratinizada da epiderme ? mais espessa quando comparada com a mesma camada na epiderme de H. maximiliani. Foi observado em P. geoffroanus e H. maximiliani que essas regi?es podem apresentar invagina??es da epiderme com o desprendimento das camadas mais externas de queratina. O estudo histol?gico e histoqu?mico realizado nos cortes da epiderme de ambas esp?cies de, n?o evidenciou a presen?a da gl?ndula submandibular. O m?todo de PAS n?o mostrou a presen?a de mucopolissacar?deos neutros nessa regi?o submandibular, que s?o produtos que fazem parte das secre??es produzidas por gl?ndulas ex?crinas. Nesse estudo observou-se que as barbelas presentes em P. geoffroanus s?o ap?ndices tegumentares cil?ndricos presentes na regi?o posterior da s?nfise mandibular, que ocorrem aos pares, constituindo-se de epiderme e derme, e com grande concentra??o de termina??es nervosas. No presente estudo, os esp?cimes de P. geoffroanus macroscopicamente apresentaram tub?rculos de forma c?nica, raramente pontiagudos, e tamb?m de formato verrucoso, vis?veis na regi?o dorsal e lateral desde a parte anterior at? a parte posterior do pesco?o, n?o estando presentes na regi?o ventral do pesco?o, que apresenta ranhuras, estrias e depress?es da epiderme. No presente estudo n?o foram observadas diferen?as morfol?gicas e histol?gicas em rela??o aos tub?rculos quando comparados por sexo. Em H. maximiliani os tub?rculos est?o distribu?dos de forma irregular nas regi?es laterais e dorsais do pesco?o, estando ausentes na regi?o da cabe?a e na regi?o ventral do pesco?o. Apresentam formato pontiagudo e de tabuleiro, diferente do que foi observado nos esp?cimes de P. geoffroanus. Recent evidence suggests the involvement of nitric oxide (NO), a potent vasodilator and anti-aggregating effect, in the pathogenesis of cardiovascular and psychiatric diseases. Nitric oxide (NO) is synthesized through conversion of the amino acid L-arginine into L-citrulline and NO by the enzyme NO synthase (NOS). This thesis addresses the role of the L-arginine-NO pathway in platelets from patients with MDD and its association with platelet function and oxidative stress. For a behavioural analysis of depression in an animal model, we used the single maternal separation (SMS) model, where the offspring was separated from its mother for eight minutes. The animals were divided into four groups for the study: Sedentary Control Group (SCG), Exercise Control Group (ECG), SMS Sedentary Group (SSMS) and SMS Exercise Group (ESMS). Physical training (PT) of the animals took 8 weeks, with a duration of 30 minutes and a training speed established by the maximum test (ET) of 14m/min. For the human study, there were ten MDD patients with Hamilton score: 20 ± 1 (mean age: 38 ± 4 years) and 10 healthy subjects (mean age: 38 ± 3 years). The human and animal studies were approved by the Ethics Committee: 1436 - CEP/HUPE and CEUA/047/2010 respectively. The following were measured in human and animal models: L-arginine transport, NOS and superoxide dismutase (SOD) activities, cGMP concentration and analysis of systemic cortisol. Experiments performed only in humans: NOS, arginase and guanylate cyclase expression evaluated by Western Blotting. Platelet aggregation was induced by collagen, and C-reactive protein, fibrinogen, and L-arginine were systemically analyzed. For statistical treatment, three statistical tests were used to evaluate the differences in survival curves: Kaplan-Meier, and the Tarone-Ware and Peto-Prentice tests. In humans, there was a reduction in L-arginine transport, NOS and SOD activities, cGMP concentration and plasma concentrations of L-arginine in MDD group compared with the control group. On the other hand, an increase in plasmatic levels of fibrinogen was observed in TD. These results show an impairment of L-arginine-NO-cGMP and the anti-oxidant enzyme SOD in patients with MDD without affecting platelet function. Regarding the PT, for the animal model were found initials changes on the running distance and duration of execution of ET between control groups and groups SMSs, the latter displaying lower values for the ET. After 8 weeks of PT, there was an enhancement of L-arginine transport in ESMS which was activated by exercise. The differences observed for time and distance in the initial ET between control groups and SMU groups was reversed after 8 weeks of PT, demonstrating the beneficial effect of exercise training on cardiorespiratory capacity in models of depression. O transtorno depressivo (TD) é um fator de risco cardiovascular independente que apresenta elevada morbi-mortalidade. Recentes evidências sugerem a participação do óxido nítrico (NO), potente vasodilatador e anti-agregante plaquetário, na patogênese de doenças cardiovasculares e psiquiátricas. A síntese do NO ocorre através da conversão do aminoácido L-arginina em L-citrulina e NO, pela ação da enzima NO sintase (NOS). Esta tese aborda o papel da via L-arginina-NO em plaquetas de pacientes com TD e sua associação com a função plaquetária e estresse oxidativo. Para análise comportamental da depressão em modelo animal, foi utilizado o modelo de estresse pós-natal de separação única (SMU). Os animais foram divididos em quatro grupos para a realização do estudo: Grupo Controle Sedentário (GCS), Grupo Controle Exercício (GCE), Grupo SMU Sedentário (SMUS) e Grupo SMU Exercício (SMUE). O treinamento físico (TF) dos animais englobou 8 semanas, com duração de 30 minutos e uma velocidade de treinamento estabelecida pelo teste máximo (TE). Para o estudo em humanos, 10 pacientes com TD com score Hamilton: 20±1, (média de idade: 38±4anos), foram pareados com 10 indivíduos saudáveis (média de idade: 38±3anos). Os estudos em humanos e animais foram aprovados pelos Comitês de Ética: 1436 - CEP/HUPE e CEUA/047/2010, respectivamente. Foi mensurado em humanos e em animais: transporte de L-arginina, concentração GMPc, atividade das enzimas NOS e superóxido dismutase (SOD) em plaquetas e cortisol sistêmico. Experimentos realizados somente em humanos: expressão das enzimas NOS, arginase e guanilato ciclase através de Western Blotting. A agregação plaquetária foi induzida por colágeno e foi realizada análise sistêmica de proteína C-reativa, fibrinogênio e L-arginina. Para o tratamento estatístico utilizou-se três testes estatísticos para avaliar as diferenças das curvas de sobrevida: Kaplan-Meier, e os testes de Tarone-Ware e Peto-Prentice. Em humanos, houve uma redução do transporte de L-arginina, da atividade das enzimas NOS e SOD, e da concentração de GMPc em plaquetas, e nas concentrações plasmáticas de L-arginina no grupo com TD em relação ao grupo controle. Foi observado um aumento dos níveis plasmáticos de fibrinogênio no TD. Esses resultados demonstram uma inibição da via L-arginina-NO-GMPc e da enzima anti-oxidante SOD em pacientes com TD sem afetar a função plaquetária. Em relação ao TF, para o modelo animal, foram encontradas alterações iniciais quanto à distância percorrida e tempo de execução do TE entre os grupos controles e o grupos SMUs, apresentando estes últimos menores valores para o TE. Após 8 semanas de TF, verificou-se um maior influxo no transporte de L-arginina para o SMUE em comparação ao grupo SMUS. As diferenças observadas para o tempo e a distância percorrida no TE inicial entre os grupos controle e no modelo de estresse foram revertidas após as 8 semanas de TF, demonstrando o efeito benéfico do exercício físico na capacidade cardiorespiratória em modelos de depressão.

The endothelial dysfunction existent in both diseases, CRF and EH, contributes to the maintenance of the high peripheral resistance, and contribute to circulatory complications such as atherosclerosis. This endothelial dysfunction is part of a pro-thrombotic state, leading to cardiovascular events, which are the major cause of death in these disorders. Nitric oxide (NO) plays an important role in the modulation of platelet function. Abnormalities of NO synthesis or inactivation are described in CRF and EH. It was previously reported an inhibition of L-arginine transport in erythrocytes of hypertensive patients and in an animal model of hypertension. Moreover, we have also demonstrated an activation of L-arginine-NO pathway in platelets taken from uraemic patients. The aim of the present thesis is to investigate L-arginine-NO pathway in arterial hypertension and in different stages of chronic renal failure. It will also be evaluated urea cycle and the presence of oxidative stress markers in these patients. According to the present study it was not detected any alteration in erythrocytres NO synthesis in hypertension, however, there was an activation of urea cycle, which could be explained by an increase in L-arginine influx. The present study has not demonstrated significative difference in markers of lipid peroxidation in the serum, platelets or erythrocytes in hypertension. In platelets however, there was an inhibition of NO synthase (NOS) activity without any alterations of NOS isoforms, arginase, phosphodiesterase 5 (PDE5) or soluble guanylyl cyclase (sGC) expression. This reduction of NO synthesis may be explained by a lower influx of L-arginine that is present on hypertension. Erythrocytes from chronic renal failure patients under haemodyalysis have shown an increased influx of L-arginine associated with a higher expression and activity of arginase with no difference in NOS activity. Therefore, although it was not shown abnormalities of lipid per oxidation markers in erythrocytes and platelets, it was detected increased levels of these markers in the serum of chronic renal failure patients under hemodyalysis. On the other hand, platelets from the same patients exhibited increased expression of eNOS, iNOS and soluble guanylyl cyclase associated with reduced arginase activity, which can explain the platelet dysfunction observed in these patients. Platelets taken from patients with chronic renal failure under conservative treatment have shown increased NOS activity associated with higher expression of both iNOS and eNOS. Curiously, it was been detected a lower concentration of cyclic guanosine monophosphate (cGMP), although there was no difference in the patterns of platelet aggregation induced by collagen or adenosine diphosphate (ADP). The findings reported in this study may contribute to a better understanding of EH and CRF physiopathology. A insuficiência renal crônica (IRC) e a hipertensão arterial sistêmica (HAS) são patologias com alta morbidade e mortalidade, consumindo grandes verbas de saúde pública. A disfunção endotelial presente tanto na IRC, como na hipertensão, contribui para a manutenção de elevada resistência periférica, favorecendo complicações como a aterosclerose. Esta disfunção endotelial é parte de um estado pró-trombótico, levando à ocorrência de eventos cardiovasculares, principal causa de morte nestas patologias. O óxido nítrico (NO) tem um papel importante na modulação da atividade plaquetária. Anormalidades na síntese e/ou inativação do NO são descritas tanto na insuficiência renal crônica como na hipertensão. Estudos prévios demonstraram uma redução do influxo de L-arginina em eritrócitos e plaquetas de pacientes hipertensos e em um modelo animal de hipertensão. Além disso, em IRC, nosso grupo mostrou uma ativação da via L-arginina-NO em plaquetas. O objetivo dessa tese é avaliar a via L-arginina-NO na HAS e em diferentes estágios da IRC, bem como investigar o ciclo da uréia, e a presença de marcadores de estresse nesses pacientes. De acordo com o presente estudo pôde-se verificar que não houve alteração na síntese de NO em eritrócitos na hipertensão, todavia ocorre uma ativação do ciclo da uréia, que pode ser dada pelo aumento do influxo de L-arginina eritrocitário previamente demonstrado. Não foi demonstrada diferença significativa na peroxidação lipídica sistêmica, em plaquetas ou eritrócitos na HAS. Em plaquetas, no entanto, houve uma redução da atividade da NO sintase (NOS), que não foi acompanhada por alteração da expressão das isoformas da NOS, da arginase, da fosfodiesterase 5 (PDE5) ou da guanilato ciclase (GC) solúvel. Essa redução na síntese de NO em plaquetas pode ser explicada por um menor influxo de L-arginina que está presente na hipertensão. Os eritrócitos de pacientes renais crônicos em hemodiálise mostraram um maior influxo de L-arginina associado a um aumento da expressão e da atividade da arginase, não havendo diferença na atividade da NOS. Além disso, apesar e não ter sido mostrado alteração nos marcadores de peroxidação lipídica em eritrócitos e plaquetas, foi detectado um aumento dos mesmos no soro de pacientes com IRC em hemodiálise. Por outro lado, as plaquetas dos mesmos pacientes apresentaram uma maior expressão da eNOS, da iNOS e da GC solúvel, acompanhada de uma redução da atividade da arginase, o que pode justificar a disfunção plaquetária que existe nesses pacientes. Plaquetas de pacientes portadores de IRC em tratamento conservador mostraram um aumento da atividade da NOS associado com maior expressão tanto da iNOS como da eNOS. Curiosamente foram detectados menores concentrações de 3 5 -monofosfato de guanosina cíclica (GMPc), não havendo no entanto, diferença nos padrões de agregação plaquetária induzida por colágeno ou adenosina difosfato (ADP). As descobertas aqui apresentadas certamente contribuirão para uma melhor compreensão da fisiopatologia da HAS e da IRC.

A limiting factor is the radiation damage in healthy tissues neighboring the tumor. The irradiation of the skin, accidentally or for therapeutic purposes, can trigger a series of injuries culminating in fibrosis, causing functional alterations in this organ. The morphological evaluation of the effects associated with skin irradiation becomes essential to establish more effective strategies for irradiation and decreased morbidity, and in case of accidents, proper handling of the victim. The aim of this study was to evaluate the radiation-induced dermal changes, using a rats model. Male Wistar rats, three months old, had their skin irradiated, in a 3cm2 field, with single doses of 10, 40 and 60 Gy of electrons with nominal energy of 4MeV. After irradiation, the animals were kept under constant observation, lesions were recorded photographically. The animals were divided into groups and euthanized: on the day of irradiation, 5, 10, 15, 25 and 100 days after irradiation. Part of the skin was fixed in formaldehyde, embedded in paraffin and subjected to microtomy. Sections were stained with hematoxylin-eosin, picrosirius red and immunostained with anti-TGF-beta1. Another part of the tissue was fixed in glutaraldehyde and processed for scanning electron microscopy. It was observed macroscopic skin lesions similar to burns throughout the irradiated area. It was verified, by optical microscopy, the early development of lesions 5 days after irradiation. After 10 days of irradiation there was evidence of epidermal wound healing under the crust formed by the injury. At the day 15 days the tissue below the lesion was reconstructed and had characteristics of healing, displaying a significant polymorphonuclear cells infiltration. After 25 days, at the higher doses, the lesions persisted, which did not occur at the lowest dose, which the irradiated area was already completely healed. After 100 days of irradiation with 40 Gy the wound was healed. With 60 Gy, in some animals, the lesion persisted. In the animals which the healing took place there was a hypertrophy of the epidermis (acanthosis). It was observed a tissue totally morphological mischaracterized, and necrotic. The results obtained by scanning electron microscopy analysis corroborate with the optical microscopy findings, where the higher doses collagen fibers were mischaracterized. The results indicate that the doses used induced a significant inflammation in the skin, activating the immune system. This fact promoted an increased expression of TGFbeta1, which is responsible for an increased production of extracellular matrix (ECM) by various cell types, mainly fibroblasts in injured tissues. Besides the increased expression of ECM, the TGFbeta1 also promotes the inhibition of the degradation processes of the same. The intense expression of this cytokines in irradiated skin can trigger the process of fibrosis and, consequently, affect the homeostasis of the body due to accumulation of ECM. A radioterapia é uma das modalidades terapêuticas mais utilizadas no tratamento do câncer, visando à destruição das células neoplásicas, a partir da utilização de radiação ionizante. Um dos fatores limitantes da radioterapia é o dano em tecidos sadios vizinhos ao tumor. A irradiação da pele, acidental ou para fins terapêuticos, pode desencadear uma série de lesões culminando na fibrose, o que implica na alteração funcional deste órgão. A avaliação dos efeitos morfológicos associados à irradiação da pele torna-se fundamental para estabelecer estratégias de irradiação mais eficazes e diminuição da morbidade; e em caso de acidentes, adequado manuseio da vítima. O objetivo deste estudo foi avaliar as alterações dérmicas radioinduzidas, utilizando um modelo em ratos. Ratos Wistar, machos, com três meses de idade, tiveram sua pele irradiada, em um campo de 3cm2, com doses únicas de 10, 40 e 60 Gy de elétrons com energia nominal de 4MeV. Após a irradiação, os animais permaneceram sob avaliação constante, sendo as lesões registradas fotograficamente. Os animais foram divididos em grupos e eutanasiados: no dia da irradiação, 5, 10, 15, 25 e 100 dias após a irradiação. Parte da pele foi fixada em formaldeído, incluída em parafina e submetida à microtomia. Os cortes foram corados com hematoxilina-eosina, picrosirius red e imunomarcados com anticorpo anti-TGF-beta1. Outra parte do tecido foi fixada em glutaraldeido e processada para microscopia eletrônica de varredura. Foi observado macroscopicamente o surgimento de lesões cutâneas semelhantes a queimaduras em toda área irradiada. Ao microscópio óptico foi verificado o inicio de desenvolvimento de lesão 5 dias após irradiação. Decorridos 10 dias da irradiação observou-se indícios de cicatrização epidérmica abaixo da crosta formada pela lesão. Aos 15 dias após a irradiação o tecido abaixo da lesão apresentava epiderme reconstruída e características de cicatrização tecidual. Foi visualizado também um infiltrado de polimorfonucleares significativo. Após 25 dias nas doses mais elevadas as lesões persistiam, o que não ocorreu na menor dose, na qual a área irradiada dos animais já se encontrava completamente cicatrizada. Após 100 dias da irradiação na dose de 40 Gy ocorreu a cicatrização da ferida. Na dose de 60 Gy em alguns animais a lesão persistia. Nos animais em que ocorreu a cicatrização houve uma hipertrofia da epiderme (acantose). Foi visualizado um tecido com aspecto morfológico totalmente descaracterizado, e necrosado. Os resultados encontrados na analise através de microscopia eletrônica de varredura corroboram os dados encontrados na microscopia de luz, onde observou-se a descaracterização das fibras de colágeno nas doses mais elevadas. Os resultados indicam que as doses utilizadas induziram um processo inflamatório importante na pele, ativando o sistema imunológico. Este fato promoveu um aumento na expressão do TGFbeta1, um dos responsáveis pelo aumento da produção da matriz extracelular por vários tipos celulares, principalmente por fibroblastos em tecidos lesionados. Alem do aumento de expressão da MEC, o TGFbeta1 também promove a inibição dos processos de degradação da mesma. A intensa expressão desta citocina na pele irradiada pode desencadear o processo de fibrose e, conseqüentemente, afetar a homeostase deste órgão devido ao acúmulo da MEC.

To assess the volumetric density of collagen, elastic system fibers and smooth muscles cells in the corpora cavernosa (CC), corpus spongiosum (CE) and tunica albuginea (TA) in the penis of diabetic rabbits. Twenty-six New Zealand white rabbits were used. Diabetes was induced at 8 weeks of age in 13 rabbits by intravenous injection of 100 mg/kg of alloxan. The remaining 13 rabbits served as a control group. After 10 weeks, the rabbits were killed using sodium thiopenthal. Mid-shaft fragments of the penis were obtained and processed by routine histological techniques. Stereological analysis of collagen, elastic system fibers and smooth muscle was performed in 5-m sections by using a M42 System Test grid. Data were expressed as volumetric density (Vv - %). Collagen organization was evaluated by Picrosirius red staining under polarization. In the TA of diabetic rabbits, thickness increased by 88% (p < 0.0003) with an enhanced collagen turnover. Moreover, the elastic fiber content was 34% higher (p < 0.0001). In the CC of diabetics, collagen was diminished by 45% (p < 0.0001) with a more organized collagen. The elastic fibers were decreased by 46% (p < 0.0001). Diabetes induced a 11% increase in CS collagen (p < 0.0235) with an enhanced collagen turnover. Smooth muscle in the CC of diabetic rabbits was increased by 40% (p < 0.0001), whereas in the CS it was decreased by a similar amount (p < 0.0001). Penile tissues were affected differently by diabetes, possibly due to cellular heterogeneity. These changes could have an impact on blood flow and tissue resistance, and therefore, might adversely affect erection. Embora a disfunção erétil seja um problema frequente em pacientes com diabete mellitus, poucos estudos avaliaram as alterações da estrutura peniana nesta condição. O objetivo foi avaliar a densidade volumétrica de colágeno, fibras do sistema elástico e células musculares lisas no corpo cavernoso (CC), corpo esponjoso (CE) e túnica albugínea (TA) no pênis de coelhos diabéticos. Foram utilizados vinte seis coelhos Nova Zelândia. A diabete foi induzida em 13 coelhos com 8 semanas de idade com injeção intravenosa de 100 mg/ kg de aloxano. 13 coelhos normais serviram como controle. Após 10 semanas os animais foram mortos com overdose de thiopenthal sódico endovenoso. Os pênis foram dissecados retirando-se um fragmento da porção média sendo fixado em formol 10% tamponado (pH 7.3). Todo material foi processado para inclusão em parafina seguindo a técnica histológica de rotina. A análise estereológica do colágeno, das fibras do sistema elástico e do músculo liso foi realizada em cortes de 5m no sistema teste M42. Os dados foram expressos em densidade volumétrica (Vv %). A organização de colágeno foi avaliada pela coloração do Picrosirius red sob polarização. Vimos que na túnica albugínea de coelhos diabéticos, houve um aumento de 88% da espessura (p < 0.0003) com um acentuado turnover do colágeno. Nowadays, Diabetes Mellitus is considered as an important health concern due to its increasing prevalence and high morbimortality. Its clinical importance comes from the complications, especially microvascular. Chronic or transitory hyperglycemia has been identified as endothelial harm inductor factor, being this the first outcome of microvascular complications. Endothelial cells, under local hemodynamic strength, produce signal transduction (mechanotransduction), which can be responsible for the beginning of pathological events in vessels wall. In this regard, the objective of this study was to analyze hamster cheek pouch microcirculation under the influence of type 1 diabetes mellitus induced by streptozotocin, evaluating its morpho-functional aspects at 6 and 15 days of diseases evolution. Morphological characteristics of arterioles and venules were estimated by the measurement of lumen diameter and wall thickness; the volume density and area of these vessels from cheek pouch; immunohistochemistry of the expression of actin, talin, smooth muscle alpha-actin, vimentin, laminin and type IV collagen through light microscopy with the utilization of a semi-quantitative score system based on the intensity of the immunostaining; and transmission electron microscopy. It was also evaluated the endothelium dependent relaxation, measured by the variation of lumen diameter before and after acetylcholine administration and post-capillary venules permeability to histamine, determined by number of points of plasma extravasation. Our results reveal that arterioles and venules do not show differences between the groups concerning wall thickness, luminal diameter, density per area and volume density. Vascular permeability, after 2 minutes of histamine administration, was reduced significantly in diabetic groups. However, this finding was not observed after 5 minutes of administration, the same occurring with vascular reactivity. The expression of actin, talin, laminin and vimentin was higher in arterioles of diabetic group with 6 days of evolution, being this alteration persistent in diabetic group at 15 days of evolution for laminin and vimentin. In electron microscopy, colloidal gold particles conjugated with talin and laminin were distributed at cytoplasm and basal surface of endothelial cells. In basement membrane, the laminin was forming clusters. These evidences suggest that at 6 days of diabetes course the proteins related to extracellular matrix adhesion were altered possibly due to changes in local hemodynamic forces caused by the new physiologic condition induced by hyperglycemia. O Diabetes Mellitus é uma doença metabólica crônica com múltiplos fatores etiológicos (genético, viral e imunológico) que condiciona deficiência absoluta ou relativa de insulina, causando persistência de níveis elevados de glicose no sangue. Atualmente, o Diabetes Mellitus é considerado um importante problema de saúde devido a sua prevalência e alta morbimortalidade. Sua importância clínica resulta essencialmente de suas graves complicações, especialmente as microvasculares. A hiperglicemia crônica ou intermitente tem sido identificada como o fator indutor de lesão endotelial, sendo este, o agente desencadeante das complicações microvasculares. As células endoteliais, por serem influenciadas pela força hemodinâmica local, respondem com a transdução de sinais (mecanotransdução), as quais podem ser responsáveis pelo início de processos patológicos na parede dos vasos. Desta forma, o objetivo deste estudo foi analisar a microcirculação da bolsa da bochecha do hamster sob a influência do Diabetes Mellitus tipo 1 experimental induzido por estreptozotocina, avaliando seus aspectos morfofuncionais aos 6 e 15 dias de evolução da doença. As características morfológicas de arteríolas e vênulas foram estimadas por medidas do diâmetro do lúmen e da espessura da parede; pela densidade de volume e de área destes vasos na bolsa da bochecha; pela análise imunohistoquímica da expressão de actina, talina, alfa-actina de músculo liso, vimentina, laminina e colágeno IV através da microscopia de luz com a utilização um sistema semiquantitativo baseado em uma escala de intensidade de imunomarcação; e por microscopia eletrônica de transmissão. Também foi avaliado o relaxamento dependente do endotélio, medido pela variação do diâmetro do lúmen antes e após a aplicação de acetilcolina e a permeabilidade de vênulas pós-capilares à histamina, determinada pelo número de pontos de extravasamento plasmático. Nossos resultados mostraram que arteríolas e vênulas não apresentaram diferenças entre os grupos para o espessamento da parede, diâmetro luminal, densidade por área e de volume. A permeabilidade vascular, após 2 minutos da administração de histamina, foi diminuída significativamente nos grupos diabéticos, entretanto este achado não foi observado após 5 minutos da administração, o mesmo ocorrendo com a reatividade vascular. A expressão de actina, talina, laminina e vimentina esteve aumentada em arteríolas do grupo diabético com 6 dias de evolução da doença, sendo esta alteração persistente no grupo diabético de 15 dias de evolução para a laminina e a vimentina. Na microscopia eletrônica, partículas de ouro coloidal conjugadas a talina e a laminina se distribuíram no citoplasma e ao longo da superfície basal das células endoteliais. Na membrana basal, a laminina mostrou-se formando aglomerados. Essas evidências sugerem que aos 6 dias de evolução do diabetes as proteínas relacionadas a adesão com a matriz extracelular sofrem alteração possivelmente em resposta a mudanças na força hemodinâmica local promovida pela nova condição fisiológica induzida pela hiperglicemia.

The valve disease is a disorder that can affect any of the heart valves, marked by any change in its structure that prevents its proper operation. Studies show that the mechanical stress caused by blood flow may interfere with the organizational structure of cells and extracellular matrix of the valve. This study aimed to investigate the possible structural and ultrastructural changes that hypertension, induced by L-NAME, can cause to the left atrioventricular valve (mitral) from hearts of rats. The animals received L-NAME (40mg/kg/dia), previously diluted in water, for five weeks. At the end of the fifth week euthanasia of animals was performed and the valve parietal fragments were collected and processed for light microscopy (histochemistry) and electron microscopy (ultrastructural histochemistry and ultrastructural immunohistochemistry). It was analyzed the morphological structure of the valves at both structural (light microscopy) and ultrastructural (electron microscopy), as well as the thickness of the valve and quantification (% / area) of the elastic and collagen fibers through the ImagePro-Plus 4.5. The animals treated with L-NAME developed hypertension from the second week of administration of the drug. Structurally, it was possible to identify a disorganization of the elastic and collagen fibers with regions of fragmentation. The hypertension animals showed thicker valve regions than control, plus a smaller amount of collagen fibers. Ultrastructurally, it was identified changes of the endothelium in animals treated with L-NAME, where it presented irregular contours and accumulations of vesicles inside. Furthermore, it was also observed fragmentation of basal lamina, disorientation of collagen fibers, fiber oxytalan accumulation near the endothelium and fragmentation of elastic fibers of the atrial layer. This study concluded that increased blood pressure, characterized by hypertension, causes changes in the morphological structure of the valve, which may be linked in with the synergistic inhibition of nitric oxide synthesis, and may thus undermine its perfect operation. As valvas cardíacas têm como função manter o fluxo unidirecional do sangue e para isso conta com uma arquitetura anisotrópica da matriz extracelular que assegura e sustenta uma adequada função sob condições de alta e baixa pressão. A valvopatia é uma desordem que pode acometer qualquer uma das valvas cardíacas por qualquer modificação em sua estrutura que impossibilita o seu adequado funcionamento. Estudos demonstram que o stress mecânico ocasionado pelo fluxo sanguíneo pode interferir na estrutura organizacional das células e matriz extracelular da valva. O presente estudo teve como objetivo investigar as possíveis alterações estruturais e ultraestruturais que a hipertensão arterial, induzida por L-NAME, pode ocasionar na valva atrioventricular esquerda (mitral) de corações de ratos. Os animais receberam L-NAME (40mg/kg/dia), previamente diluído na água de beber, durante um período de cinco semanas. Ao final da quinta semana foi realizada a eutanásia dos animais e fragmentos da válvula parietal foram coletados e processados para microscopia de luz e eletrônica. Foram analisadas as estruturas morfológicas das válvulas tanto para microscopia óptica (histoquímica) quanto microscopia eletrônica (histoquímica ultraestrutural e imunohistoquímica ultraestrutural), assim como, a espessura da válvula e a quantificação (%/area) das fibras elásticas e colágenas através do programa ImagePro-Plus 4.5. Os animais tratados com L-NAME desenvolveram hipertensão a partir da segunda semana de administração do fármaco. Estruturalmente foi possível identificar uma desorganização das fibras elásticas e colágenas com regiões de fragmentação. Os animais hipertensos apresentaram regiões da válvula mais espessas do que o controle, além de uma menor porcentagem de fibras colágenas. Ultraestruturalmente, identificamos modificações do endotélio nos animais tratados com L-NAME, que se apresentou com contorno irregular e com acúmulos de vesículas no seu interior. Além disso, foi observada também fragmentação da lâmina basal, desorientação das fibras colágenas, acúmulo de fibras oxitalânicas próximo ao endotélio e fragmentação das fibras elástica da região atrial. O presente trabalho concluiu que o aumento da pressão sanguínea, caracterizada pela hipertensão, ocasiona modificações na estrutura morfológica da válvula, que pode estar associado de forma sinérgica com a inibição da síntese óxido do nítrico, podendo assim, prejudicar o seu perfeito funcionamento.

These mediators may have cell origin as the CGRP, VEGF and TGFß. The aim of this research were to evaluate the expression and distribution of the CGRP, VEGF and TGFß in the gingiva of the lower left first molar of rats in the seven and fourteen days after the ligature for inflammation induction; to analyse the expression and distribution of the CGRP, VEGF and TGFß in the gingiva of the correspondent counter lateral tooth without ligature in the seven and fourteen days and if the induction of periodontitis causes gingival inflammation of the counter lateral tooth after seven and fourteen days after ligature. For the development of this work were selected fifteen Rattus Novergicus, Albinus,Wistar. The experimental group of 12 rats was divided into 2 groups consisting of 6 rats each distributed of the following manner: the subgroup A1 remained with ligature in the first molar and left for 7 days before the sacrificed and the subgroup A-2 remained with the ligature for 14 days before the sacrificed. Another 3 animals constituted the control group. After the sacrificed of the 12 experimental and 3 control animals were immersedin neutral ethylenidiaminetetracetic (EDTA) for the decalcification. Then were processed for paraffin embedding. The sections were stained with hematoxylin-eosin and submitted to immunohistochemical techniques to imunostaining for CGRP, VEGF and TGFß. At 7 days of ligature was observed in the gingival lamina propria, junctional epithelium and oral epithelium, strong staining for CGRP. The intensive expression of VEGF occur in the lamina propria and scarce or no staining in the oral and junctional epithelium. The TGFß shows scarce staining in the lamina propria and no staining in the oral and junctional epithelium. At the fourteen days of ligature we observed strong staining of CGRP in the lamina propria and oral and junctional epithelium. The VEGF and TGFß show strong staining in the lamina propria and scarce or no staining in the junctional and oral epithelium. At the gingival of the counterlateral teeth in the 7 and 14 days there was scarce staining for CGRP and TGFß in the lamina propria and strong staining to VEGF. In the gingival of couterlateral teeth there was strong staining to CGRP in the junctional and oral epithelium and lamina propria. The TGFß and VEGF show scarce or none staining. Because the strong staining of VEGF in the lamina propria of the counterlateral teeth remain inconclusive the adequacy of the use of the counterlateral teeth in the experimental studies of the periodontal diseases, though the expression of the TGFß and CGRP has been smaller in this teeth. The bigger staining of CGRP, VEGF and TGFß in the animals with 14 days of ligature than those 7 days shows the evolution of the chronic inflammation process. We don t observe the process of tissue healing. No momento em que há a agressão tecidual e a defesa inata é deflagrada, mediadores químicos são liberados no local afetado. Esses mediadores podem ser de origem celular tais como CGRP, VEGF e TGFß. Os objetivos desta pesquisa foram avaliar a expressão e distribuição de CGRP, TGFß e VEGF na gengiva do primeiro molar inferior esquerdo de rato no 7° e 14° dias após a indução por ligadura; a expressão e distribuição de CGRP, TGFß e VEGF na gengiva do dente contralateral correspondente sem ligadura, no 7° e 14° dias, e se a indução da periodontite por ligadura no dente experimental provoca uma inflamação na gengiva do dente contralateral correspondente no 7° e 14° dias após a ligadura. Para o desenvolvimento deste trabalho foram selecionados 15 ratos Rattus Novergicus, Albinus, Wistar. O grupo experimental de 12 ratos foi dividido em 2 subgrupos compostos por 6 ratos cada um deles distribuídos da seguinte maneira: os do subgrupo A1 permaneceram com a ligadura no primeiro molar inferior esquerdo por 7 dias e foram sacrificados; os do subgrupo A-2 -permaneceram com a ligadura no primeiro molar inferior esquerdo por 14 dias e foram sacrificados. Outros três animais constituíram o grupo controle. Após o sacrifício dos 12 animais dos grupos experimentais e controle suas mandíbulas foram colocadas em ácido etilenodiaminotetracético (EDTA) neutro para sofrerem descalcificação. Então foram processadas para inclusão em parafina e os cortes histológicos foram corados pela hematoxilina-eosina e submetidos à técnica imunohistoquímica para imunomarcação de CGRP, VEGF e TGFß. Aos 7 dias de ligadura observou-se na lâmina própria gengival, epitélio juncional e epitélio oral, expressiva marcação para CGRP. A expressão de VEGF foi intensa na lamina própria e com pouca ou nenhuma marcação no epitélio oral e juncional. O TGFß apresentou pouca marcação na lâmina própria ou nenhuma marcação no epitélio oral e juncional. Aos 14 dias de ligadura houve expressiva marcação de CGRP na lâmina própria, epitélios oral e juncional. O VEGF e o TGFß apresentaram muita marcação na lâmina própria e pouca ou nenhuma marcação no epitélio oral e juncional. Na gengiva dos dentes contralaterais nos 7 e 14 dias houve pouca marcação do CGRP do TGFß na lâmina própria e muita marcação do VEGF. Na gengiva dos dentes controle observou-se muita marcação do CGRP no epitélio juncional e oral e na lâmina própria. O TGFß e o VEGF se expressaram muito pouco ou não se expressaram. Devido à marcação expressiva do VEGF na lâmina própria dos dentes contralaterais, permanece inconclusiva a adequação do uso dos dentes contralaterais nos estudos experimentais das doenças periodontais, embora a expressão de TGFß e CGRP tenham sido menores nestes dentes. A maior marcação do CGRP, VEGF e TGFß nos animais com 14 dias de ligadura do que aos 7 dias demonstra a progressão do processo inflamatório crônico, não se observando processo de reparação cicatricial.

The aim of this study is to evaluate the parasite diversity in the three sites (two in the municipality of Santa Maria Madalena and one in Concei??o de Macabu) throught morphobiology, biometric, ultrastructural and histopatological parameters. Seven isolates identifield as T. cruzi were maintained in NNN+LIT+20% bovine fetal serum. Morphometric studies and cellular differentiation were based on epimastigotes and trypomastigotes forms obtained in the growth curves in the 4, 7, 10, 13, 17 and 20? days for SMM32, SMM53, SMM81, SMM98, SMM100 proceeding from the area A, SMM36 proceeding from area B, SMM 81 and SMM1 proceeding from area F and extended to 23, 27, 30, 33, 37 and 40? days to the SMM 36 and SMM 98 samples. The maximum growth was observed in the 4 th day for SMM100 isolate (45.000 parasites/?l); the 7 day for SMM53 (52.750 parasites/?l) the 10 th day for SMM1 (21.000 parasites/?l) and SMM32 (33.125 parasites/?l); the 13 th day for SMM81 (46.000 parasites/?l) and 20 th day for SMM36 (54.375 parasites/?l) and SMM98 (44.000 parasites/?l). The SMM32 and SMM100 isolates return to grow after a slight decline. The rate of metacyclogenese was lesser than 30% in SMM32, SMM53, SMM100 and SMM1 isolates and higher than 80% in SMM81 and SMM98 isolates. These results demonstrate diversity in the behavior of these isolates. Morphometrical analysis based on epimastigotes and trypomastigotes forms found in the10 th, 17 th and 20 th days of the growth curve, suggests heterogeneity. Althrough the majority ultrastructural data of T. cruzi has been obtained from epimastigote forms maintained in culture was media, this study carried out with isolated forms from triatomines showing the same tipical structures present as the kinetoplast, the sub-pellicular microtubules, the flagellar pocket and the axoneme.The SMM36 strain was the most lethal a high mortality. Histopathological results with 10 and 60 days after inoculation have shown an intense tropism for cardiac muscle and intense mononuclear infiltrate. These results give evidences of the diversity of parasites that infection specimens of T. vitticeps, emphasizing the wild habit of this species and the epidemiological complexity of studied region. A amostra SMM32 e SMM100 apresentaram crescimento depois de um leve decl?nio. A taxa de metaciclog?nese era menor que 30% nos isolados SMM32, SMM53, SMM100 e SMM1 e mais alto que 80% em SMM81 e SMM98. Com o aumento de dias de avalia??o da curva de crescimento, a amostra SMM36 apresentou crescimento m?ximo no 33? dia (90.125 c?lulas/?l) decl?nando ao final (40? dia). Estes resultados demonstram uma diversidade no comportamento destes isolados. A morfometria foi baseada nas formas epimastigotes e tripomastigotas encontradas no 10?, 17? e 20? dias da curva de crescimento, tamb?m sugere a heterogeneidade destes isolados. Embora a maioria dos dados sobre a ultraestrutura de T. cruzi tenham sido obtidos de formas epimastigotas mantidas em meio de cultura, o estudo realizado nas formas isoladas de triatom?neos mostrou as mesmas estruturas t?picas presentes tais como cinetoplasto, microt?bulos subpeliculares, bolsa flagelar e axonema. A amostra SMM36 produziu uma alta mortalidade em camundongos inoculados com tripomastigotas. Os resultados histopatol?gicos com 10 e 60 dias ap?s infec??o mostram um intenso tropismo para m?sculo card?aco e intenso infiltrado mononuclear. The results are expected to be used in eventual population control, if and when its growth is considered harmful. The animals under study were borrowed from the Centro de Primatologia do Rio de Janeiro (CPRJ/FEEMA), to be used as necessary. The study of the ovaries revealed collagenous albuginea , under a single layer of squamous epithelium, - with strong mark for type I collagen. Among the follicles were observed a weak mark for the same collagen. In the unilaminar primary follicles we observed neutral glycoproteins called the zona pellucida; in the secondary and mature follicles these glicoproteins were seen in the zona pellucida and in the follicular fluid. In the multilaminar primary follicle, before the formation of the antrum, we observed several spaces which stain positively with PAS reaction and are called Call-Exner bodies. These are, surely, the precursors of the follicular fluid. They are more evident than in the human ovary. .An ovarian follicle with a completely formed antrum and cumulus oophorus is considered a mature follicle. In the corpus luteum, in several levels of development, we saw neuter glycoprotein interspersed with granulosa lutein cells and theca lutein cells, and, in the remnants of the follicular fuid, while present. The walls of the oviducts are composed of three layers: mucosa, muscularis and serosa. In the more aged specimens we observed atrophy of the oviducts. The uterine wall of the body and the fundus is composed of an endometrium, myometrium, and either an adventitia or a serosa. The endometrium is composed of a simple columnar epithelium and a lamina propria with simple branched tubular glands, whose product reacted positively with PAS. The cervix did not present glands; the uterine glands did not extend until it. The vagina was a fibromuscular tube with the mucous membrane consisting of an stratified squamous epithelium , not strictly keratinized, resting in a fibrous lamina propria. The vulva was represented by its vestibule and majora labia. Each testis is surrounded by a capsule of dense, irregular collagenous tissue, with predominance of type I collagen, known as tunica albuginea. Incomplete septs part from this capsule. These septa are thin and do not totally compart the testis.The seminiferous tubule presents itself covered by the seminiferous epithelium, which is supported by a basal lamina reactive to PAS. Both the tubuli recti and the rete testis are covered by a simple cuboidal epithelium. Emerging from the rete testis we could devise ductuli efferentes which shows a pseudostratified epithelium The ductus epididymis is lined by a stereociliated pseudostratified columnar epithelium. The epithelium is separated by a basal lamina from the underlying loose connective tissue. A layer of circularly arranged smooth muscle cells surrounds the connective tissue layer which exhibits a gradual increase proximo-distal in the thickness. Between the folds of this duct colagen type I was detected. The epididymis epithelium and its secretion revealed strong reaction to PAS. The ductus deferens is the continuation of the epididymis, The seminal vesicle arise as outgrowths from the ductus deferens. After the emergency of the seminal vesicle, the ductus deferens receives the denomination of ejaculatory duct and terminates into the pelvic uretra, which is enveloped by the prostrate. The prostate is a conglomeration of compound tubuloalveolar glands and the glands are lined by a pseudostratified columnar epithelium. O estudo objetivou descrever histologicamente os sistemas reprodutores feminino e masculino de primatas neotropicais da esp?cie Callithrix jacchus, conhecidos na nomenclatura usual como sag?i-de-tufo-branco, visto que este estudo poder? ser usado para conhecimento da esp?cie, ou para que se possa controlar a popula??o de sag?is, quando a mesma for considerada praga. Os animais estudados fazem parte da cole??o do Centro de Primatologia do Rio de Janeiro (CPRJ / FEEMA) e foram emprestados para esta pesquisa. Ao estudarmos os ov?rios das f?meas, detectamos, abaixo do epit?lio de revestimento uma albug?nea colagenosa com marca??o forte para col?geno tipo I. Entre os fol?culos observamos marca??o fraca para este col?geno. Nos fol?culos prim?rios unilaminares observamos glicoprote?nas neutras na zona pel?cida; nos fol?culos secund?rios e maduros observamo-las na zona pel?cida e no l?quido antral. Nos corpos l?teos, em v?rios n?veis de desenvolvimento, observamos glicoprote?na neutra por entre as c?lulas granuloso-lute?nicas e teco-lute?nicas e no restante de l?quido folicular, enquanto este ainda estava presente. Nos fol?culos ovarianos multilaminares observamos, antes da forma??o do antro, v?rios espa?os com rea??o positiva ao PAS: os chamados corp?sculos de Call Exner. Estes s?o, certamente, os precursores do l?quido folicular. Eles s?o mais evidentes do que no ov?rio humano. A parede da tuba uterina foi vista com uma t?nica mucosa, uma muscular e externamente, uma serosa. Observamos, nos exemplares mais idosos, uma atrofia das trompas. Na parede uterina o endom?trio apresentou epit?lio simples cil?ndrico com gl?ndulas simples e ramificadas cujo produto reagiu positivamente ao PAS. A c?rvix n?o se mostrou glandular; as gl?ndulas uterinas n?o se estendiam at? ela. A vagina apresentou-se como um tubo fibro-muscular, a mucosa consistindo de um epit?lio estratificado pavimentoso pouco queratinizado, repousando numa l?mina pr?pria fibrosa. A vulva representou-se pelo vest?bulo e grandes l?bios. Os test?culos em Calithrix jacchus, mostravam-se envolvidos por uma c?psula de tecido conjuntivo, com predomin?ncia de fibras col?genas do tipo I. A partir desta c?psula, denominada albug?nea, partiam septos incompletos delgados, que n?o compartimentalizam totalmente o test?culo. O col?geno da albug?nea se continuava nos septos e com o tecido intertubular. O t?bulo semin?fero apresentava-se revestido pelo epit?lio semin?fero, que apoiava-se em uma l?mina basal reativa ao PAS. Tanto t?bulos retos quanto ? rede testicular apresentavam-se revestidos por epit?lio c?bico simples. Dessa rede emergiam dutos eferentes sendo que j? observ?vamos um epit?lio pseudoestratificado cil?ndrico. Esses dutos formavam o duto epididim?rio. O duto epididim?rio apresentava um epit?lio pseudoestratificado cil?ndrico com esteroc?lios. Externamente ao epit?lio e ? l?mina basal localiza-se uma musculatura lisa, que exibe gradual aumento pr?ximo-distal em espessura. Entre as dobras desse duto observamos col?geno do tipo I. O epit?lio do epid?dimo e sua secre??o demostraram rea??o forte ao PAS. O duto deferente se continuava ao duto epididim?rio, localizando-se dentro do cord?o esperm?tico, circundado pelas veias do plexo pampiniforme, art?rias, vasos linf?ticos, nervos e c?lulas musculares estriadas esquel?ticas do m?sculo cremaster. A parte terminal do duto ao ampliar-se, evagina-se, formando as ves?culas seminais. Ap?s a emerg?ncia da ves?cula seminal, o duto deferente recebe a denomina??o de dutoejaculat?rio e desemboca na uretra p?lvica, a qual ? envolvida pela pr?stata. Foram produzidas seções histológicas de 80 espécimes. Métodos de colorações histológicas e histoquímicas foram empregados. Os preparados histológicos foram examinados e fotomicrografias foram realizadas usando um vídeo-microscópio. Cinco macro-categorias do estômago são propostas a partir das variações macroscópicas identificadas: Hematófaga, Simples-sacular, Sacular, Intermediária-sacular, e Complexasacular. Quatro padrões regionais de pregas gástricas (longitudinal e paralelo, ondulado e paralelo, complexo e com interdigitações, e tortuoso e anastomosado) são reconhecidos, assim como dois padrões de esfincter pilórico. Os resultados histológicos mostram que os tipos celulares típicos das glândulas gástricas variam regionalmente dentro da mucosa e principalmente nas glândulas fúndicas. As glândulas fúndicas de Desmodontinae apresentaram a menor profundidade e número reduzido de células. Três padrões morfológicos das vilosidades intestinais (digitiforme, foliáceo e piramidal) são reconhecidos na junção do gastroduodenal. As glândulas de Brünner estão presentes na submucosa duodenal da junção do gastroduodenal em quase todos os Phyllostomidae, à exceção de Uroderma e Platyrrhinus. A histoquÍmica revela diferenças entre a distribuição das glicoproteínas e proteoglicanas na mucosa gástrica. Este é o primeiro estudo detalhado da distribuição e da caracterização das fibras colágenas e elásticas na parede do estômago dos Chiroptera. Avaliações cladísticas das informações contidas no trato digestório de filostomídeos indicam resultados promissores. Comparative morphological study of the stomach and the gastroduodenal junction is undertaken in 12 genera of bats representing five sub-families of the family Phyllostomidae. We dissected and prepared histological sections of the stomach and gastroduodenal junction of 80 microchiropteran specimens. Standard staining and histochemical procedures were employed. Sections were examined under light microscopy and photomicrographies were taken using a video microscope. Line drawings were made for recording gross morphological variations, which indicate that five macro categories are identifiable: Hematophaga, Simple-sacular, Sacular, Intermediate-sacular, and Com pie xsacular. We identified four regional gastric pattems of the rugae (longitudinal and parallel, undulated and parallel, complex and interdigitating, and finally tortuous and ana~tomosed) as well as two pyloric sphincter types. Histological results show that the typical cellular types vary regionally within the gastric mucosae and principally in the fundic glands among genera and between sub-families. The fundic glands of Desmodontinae showed the shallowest depth and the least number of cells. Three pattems of villi morphology (digitiform, foliaceous and pyramidal) are recognized at the gastroduodenal junction. The Brünner' s gland is present at the submucosa of the gastroduodenal j unction in almost all Phyllostomidae except Uroderma and Platyrrhinus. The histochemistry demonstrates differences between the distribution of glycoproteins and proteoglycans which can be possibly related to diet representing adaptations to it. This is the first detailed study of the distribution and characterization of elastic and collagenous fibers in the stomach wall. The distribution of the enteroendocrine cells is also described. Cladistic evaluations of the information content of phyllostomid digestive system indicate quite promising results.

Only the molluscs belonging to classes A and B acquired the infection. In order to obtain eggs of E. coelomaticum three physiological solutions were tested: Earle solution, Locke solution, and physiological saline 0.85%. The physiological solution which induced the best egg realese was the Locke solution. The route of migration of the larval stages of E. coelomaticum through the visceral mass of B. similaris was followed through histology. The emergente of the second generation sporocysts occurred through the pneumostome, always during the night. The rhythm of elimination of the second x x i i generation sporocysts showed a positve correlation to the increase in temperature. The minimal period for the intramolluscan development, ending a t the e x p e l l i n g o f the f i r s t s p o r o c y s t s , was 79 days for the November/1989 infection, and 107 days for the Match/t990 infection. During the intramolluscan development of E. coelomaticum data on the oxygen consumption and calcium deposition on shells were collected and statistically a n a l y s e d f o r i n f e c t e d and u n i n f e c t e d molluscs. Oxygen consumption was analysed t h r o u g h the Student "T" t e s t ( L = 5%) and simple linear regression, indicating that the infection did not alter the mollusc's oxygen consumption. Meanwhile, the simple linear regression showed a positive correlation (r = 0.105) between post-infection time and oxygen consumption by molluscs in the control group (uninfected) in the March/1990 infection. In relation to the calcium deposition on the shells, the simple linear regression analysis showed a negative correlation (r = -0.277) between the shell weight and the calcium concentration in the infected molluscs, when compared to the uninfected. Histochemistry study of infected and uninfected molluscs showed an increase in calcium content in the cells of the digestive gland of the infected molluscs, suggesting mobilization of calcium from the shell to the digestive gland. Histological study of infected and uninfected molluscs showed alteration in the ovotestis of the infected molluscs, suggesting the occurrence of indirect parasitic x x i i i castration in these hosts, while that of the expelled sporocysts showed that the cellular structure of the sporocyst wall of E. coelomaticum was desintegrated with degenerated nuclei. Acid and neutral glycoconjugates were detected surrounding the cercaria, suggesting the beginning of the metac e r c a r i a c y s t wall f o r m a t i o n . Glycogen was d e t e c t e d in the parenchyma of the sporocyst around the cercaria. A emerg?ncia dos esporocistos de segunda gera??o ocorreu via pneum?stoma do molusco, sempre durante a noite. O r?tmo de elimina??o dos esporocistos de segunda gera??o mostrou uma correla??o positiva com aumento da temperatura. A dura??o m?nima do desenvolvimento larval, intramolusco, do E. coelomaticum , culminando com a primeira elimina??o dos esporocistos de segunda gera??o, foi de 79 dias para a infec??o realizada em novembro/1989 e 107 dias para a infec??o realizada em mar?o/1990. Durante o desenvolvimento larval, intramolusco, do E. coelomaticum foram registrados e analisados estatisticamente, o consumo de oxig?nio por moluscos infectados e n?oinfectados e a deposi??o de c?lcio nas conchas dos moluscos infectados e n?o-infectados. Os resultados relativos ao consumo de oxig?nio foram analisados estatisticamente pelo teste "T" de Student (L = 5%) e pela regress?o linear simples, tendo ambos mostrado que a infec??o n?o alterou o consumo de oxig?nio pelos moluscos. Entretanto, a regress?o linear simples mostrou uma correla??o positiva (r = 0,105) entre o tempo p?s-infec??o e o consumo de oxig?nio pelos moluscos do grupo controle (n?o-infectados), na infec??o de mar?o/1990.
