Objectives N6-methyladenosine (m6A) is one of the most common epigenetic modifications of eukaryotic RNA. Under the jointly reversible regulation of related enzymes, m6A regulates many aspects of RNA, such as translation, stability and degradation. The aim of this study is to investigate the role of m6A in placenta-related diseases. Methods Data were compiled from 2018 to 2021 citations in PubMed and Google Scholar using the keywords: placenta AND N6-methyladenosine. Seven studies were included. Results In this study, we introduced some conventional methods to detect m6A modification at the whole RNA, region (peak) and single base levels. We also summarized the current studies of m6A modification in the placenta and briefly describe m6A in placental-related diseases, including recurrent miscarriage (RM), preeclampsia (PE) and gestational diabetes mellitus (GDM). Conclusions Although the relevant reports are still in the preliminary stage and some results are inconsistent, studies on methylation m6A modification have contributed new ideas for the research of reproductive diseases, providing a new basis for the diagnosis, treatment, prognosis and monitoring of related diseases.

Endometritis is the inflammatory response of the uterine lining which is linked to infertility. Administration of platelet-rich plasma (PRP) represents a well-recommended strategy for the treatment of endometrium-associated infertility. In this study, we set to characterize the role and molecular mechanism of PRP intrauterine infusion in mice with endometritis.A mouse model of endometritis was established using lipopolysaccharide (LPS). Mouse endometrial epithelial cells were obtained in primary culture. PRP-treated cells were assayed for proliferative and apoptotic activities. Moreover, iNOS expression and chemokine and inflammatory factor contents in cells were assessed using RT-qPCR and ELISA. The mice were subjected to PRP intrauterine infusion. The expression of genes related to uterine development was analyzed by qPCR and the ki-67 content and caspase-3 activation in endometrial tissues were examined by immunohistochemistry. Finally, the Nrf2/HO-1 pathway activity in tissues was examined by Western blot.LPS induced inflammatory cell recruitment and tissue damage in the endometrium of mice, along with significantly increased levels of inflammatory and chemokine factors. PRP significantly enhanced endometrial epithelial cell activity, decreased apoptosis, and reduced inflammatory factor secretion. In addition, PRP intrauterine infusion significantly increased the expression of genes related to uterine development, promoted tissue proliferation, decreased apoptosis, and diminished inflammatory response in endometrial tissues of mice. PRP intrauterine infusion significantly elevated Nrf2/HO-1 pathway activity in endometrial epithelial cells and tissues.PRP intrauterine infusion significantly inhibited endometrial cell injury and alleviated the inflammatory response through activating the Nrf2/HO-1 pathway.

Fiber loop ring down spectroscopy has been developed into a high-sensitivity technique for sensing many important characteristics of samples, such as refractive index. In contrast to the previous demonstrations by using mode-locked fiber laser, this work is demonstrated to use chaotic laser combined with fiber loop ring down sensor. The formation and performance of fiber tapers was monitored by combining chaotic fiber ring down technology on line and in real time. Two sets of refractive index sensing experiments are performed and further analyzed by using the proposed setup. The results are shown that the detection limit of the chaotic correlation fiber loop ring down system is 3.45 × 10−4 RIU and the sensitivity is 0.0682 ns−1RIU−1 in 6.7 m, respectively. The performance of our system can be drastically improved by increasing the length of the etched fiber tapers to be tens of centimeters using the chaotic correlation fiber loop ring down system with the smaller fiber ring lengths.

Online monitoring of the polishing process of a D-shaped optical fiber sensor is performed in this paper by means of a chaotic correlation fiber loop ring down (CCFLRD) system. The ring down time of the autocorrelation coefficient decreases with the increase in polishing loss caused by different mesh sizes. A comparison of the sensitivity and resolution of the CCFLRD system with different polishing losses in the length of a fiber loop cavity are carried out. Online polishing in the fiber loop cavity with a very short length is proposed and demonstrated using different polishing mesh numbers to increase the sensitivity and resolution of the system. A high sensitivity of 31.871ns−1RIU−1 and resolution of 10−4RIU for refractive index sensing in the range of 1.3347–1.3721 correspond to different concentrations of sodium chloride.

Purpose Impaired decidualization contributes to the infertility in recurrent implantation failure (RIF). Herein, we focused on the function and probable mechanisms of miR-133b in endometrial stromal cells decidualization.Methods miR-133b and KLF12 protein levels in midsecretory endometrial tissues derived from women with and without RIF were measured by qRT-PCR and Western blot. Primary human endometrial stromal cells (HESCs) were isolated and cultured for in vitro decidualization assays. Luciferase reporter, qRT-PCR and Western blot assays were used to measure the relationship between miR-133b and KLF12.Results miR-133b was significantly downregulated, whereas KLF12 was upregulated in endometrial tissues from RIF. miR-133b effectively promoted HESCs in vitro decidualization through the modulation of KLF12 expression and the activation of LIF/STAT3 pathway. Conversely, inhibition of miR-133b expression reversed these effects. In addition, the luciferase reporter system demonstrated that miR-133b directly inhibited the expression of KLF12 by interacting with 3’ untranslated region of KLF12.Conclusion Our data suggest that miR-133b promotes HESCs decidualization by targeting KLF12 and reverses the impaired decidualization in RIF.

In this paper, we describe a novel temperature sensing system based on chaotic correlation fiber loop ring down technique. A fiber Bragg grating is introduced into the fiber loop cavity. The effect of temperature on the central wavelength of fiber Bragg grating is characterized by the ring down time of the autocorrelation coefficient of chaotic laser in the fiber loop cavity. The relationship between the autocorrelation coefficient ring down time of the chaotic laser and wavelength shifting of the fiber Bragg gratings induced by temperature is theoretically and experimentally analyzed. The sensitivity of 3.52 ns/°C is achieved in the proposed temperature sensing system with fiber cavity length of 6.05 m. We also study the relationship between temperature and central wavelength of fiber Bragg grating by chaotic correlation fiber loop ring down system and receive the temperature sensitivity of 0.01 nm/°C of the FBG. This sensing method is not only simple and low cost, but also has the great potential applications for various industry and agriculture.

To assess the associations between mode of delivery and pregnancy outcomes in patients with intrahepatic cholestasis of pregnancy (ICP) based on the severity of maternal hypercholanemia. Endometrial cancer is the most common malignant tumor of female genital system worldwide. Homeobox A11 (HOXA11) is an evolutionarily conserved Homeobox gene closely implicated in carcinogenesis. However, the mechanisms of HOXA11 in the progression and cisplatin resistance of endometrial cancer remain unclear. The expression of HOXA11 was analyzed based on 548 endometrial cancer and 35 control tissues from The Cancer Genome Atlas (TCGA) database. Transwell assay was performed to investigate the effect of HOXA11 on endometrial cell migration and invasion. TUNEL staining was carried out to assay the role of HOXA11 in endometrial cell apoptosis. Western blot was employed to detect the protein levels of B cell lymphoma-2 (Bcl-2), Bcl-2 associated X (Bax), cleaved caspase-3, matrix metalloproteinase-2/9 (MMP/9), phosphatase and tensin homolog (PTEN), protein kinase B (AKT) and p-AKT. TCGA data showed that HOXA11 expression was significantly down-regulated in endometrial cancer tissue samples. The overexpression of HOXA11 promoted the apoptosis, but inhibited the proliferation, migration and invasion of endometrial cancer cells. HOXA11 knockdown with small interfering RNA (siRNA) considerably repressed cell apoptosis, while promoted cell proliferation, migration, and invasion through PTEN/AKT signaling pathway. Interestingly, HOXA11 was lowly expressed in Ishikawa cells treated with cisplatin. In addition, HOXA11 knockdown increased the resistance of endometrial cancer to cisplatin through activating PTEN/AKT signaling pathway. Low HOXA11 expression may promote the proliferation, migration, invasion of endometrial cancer cells, and increase their resistance to cisplatin through activating PTEN/AKT pathway.

Background Protoapigenone, as a flavonoid compound with a specific nonaromatic B-ring, exhibits extraordinary antitumor activities against a broad spectrum of human cancer cells. Here we developed a novel protoapigenone analog RY10-4, which induces the apoptosis of various tumor cells, especially for breast cancer cells, but the underlying mechanism involved in the apoptotic process remains unclear. Methods MTT assay, colony-formation assay and flow cytometry were applied to evaluate the proliferation and apoptosis of breast cancer cells. Cytoplasmic calcium ([Ca2+]c) and mitochondrial calcium ([Ca2+]m) of the breast cancer cells were measured by the Fluo-2 and Rhod-2 probes, respectively. The mitochondrial reactive oxygen species (mtROS), membrane potential (ΔΨm) and permeability transition pore (mPTP) were analyzed by MitoSOX, JC-1 probes and Calcein/AM, respectively. Furthermore, Western bolt assay was adopted for the exploration of the mitochondrial apoptosis pathway. Besides, the xenograft assay was performed to investigate the role of RY10-4 in breast cancer cells in vivo. Results Obviously, RY10-4 could effectively suppress the proliferation and induce the apoptosis of breast cancer cells. Furthermore, the [Ca2+]c and [Ca2+]m of MDA-MB-231 cells were up-regulated after the treatment of RY10-4, resulting in the mtROS accumulation, ΔΨm depolarization and mPTP opening. And finally, the mitochondrial apoptosis was activated by the release of cytochrome C. Interestingly, the inhibition of mitochondrial calcium uniporter (MCU) with Ru360 attenuated the overload of [Ca2+]m and blocked the apoptosis of MDA-MB-231 cells induced by RY10-4, which was also consistent with the in vivo results. Conclusions From the results we concluded that RY10-4 could induce apoptosis of breast cancer cells by elevating [Ca2+]m through MCU. Our work contributed previously unknown insights into the mechanisms involving in the clinical efficacy of RY10-4 on breast cancer cells, which also advanced calcium homeostasis as a potential target for chemotherapeutic drugs.

In this paper, the adhesion between irregularly shaped Martian dust particles and the Martian probe in the dust storm is studied. First, the characteristics of irregularly shaped Martian dust particles are analyzed using fractal theory and the discrete element simulation model of irregularly shaped Martian dust particles is built. Second, the mechanical model of the adhesion of Martian dust onto the Martian probe is established, and the analytical solution of the critical wind speed is calculated. Finally, a series of simulations based on DEM-CFD are performed to analyze the creeping of irregularly shaped Martian dust particles in the Martian atmospheric environment. The research can provide a theoretical basis for the processes of dust protection and cleaning in a Martian dust storm.

Neuroactive steroids such as progesterone, testosterone, and their derivatives have been widely studied for their neuroprotective roles in the nervous system. Autologous nerve transplantation is considered as the gold standard repair technique when primary suture is impossible; nevertheless, this method is far from ideal. In this study, we aimed to explore the impact of dihydrotestosterone (DHT), a 5α-reduced derivative of testosterone, on the recovery of peripheral nerve injury treated with autologous nerve transplantation. Sprague-Dawley rats were subjected to a 10-mm right side sciatic nerve reversed autologous nerve transplantation and randomly divided into groups that received DHT or DHT + flutamide (an androgen receptor blocker) daily for 8 weeks after operation. Our results demonstrated that DHT could speed up the rate of axonal regeneration and increase the expression of myelin protein zero (P0) in autograft reversal sciatic nerves. Thus, our study provided new insights into improving the prognosis of patients with long gap peripheral nerve defects.

RY10-4, an anti-tumor agent, exerts cytotoxicity to various human cancer cell lines. However, few studies reported the effect of combined application of RY10-4 and chemotherapeutic drugs against cancer cells with multidrug resistance (MDR). In this study, P-glycoprotein (P-gp), which is reported to mediate MDR to anti-cancer drugs, was proved to be overexpressed in the adriamycin (ADR)-resistant human breast cancer cells, namely MCF-7/ADR cells. Furthermore, RY10-4 application resulted in a downregulation of P-gp in MCF-7/ADR cells, thus leading to higher chemosensitivity to ADR. Our study further demonstrated that the MDR phenomenon was under the control of the PI3K/Akt/NF-κB pathway, which was suppressed by RY10-4, leading to MDR reversal effects in MCF-7/ADR cells. In vivo, MCF-7/ADR cells were effectively suppressed by the combined ADR/RY10-4 treatment compared with the ADR-alone treatment. Taken together, these results demonstrated that RY10-4 reverses the MDR phenotype in MCF-7/ADR cells by suppressing the PI3K/Akt/NF-κB pathway.

Emmenopterys henryi Oliv. (Rubiaceae) is an endangered tree species that is native to China. The wild populations of E. henryi have declined rapidly because of its poor natural regeneration, but the actual regeneration processes are not yet understood. In field tracking surveys and experiments in the Wuyishan Nature Reserve in Southeast China to determine the most important stage that affects the regeneration process, seed bank characteristics, seed germination, and seedling growth dynamics were studied in the typical habitats of E. henryi, Phyllostachys pubescens and broad-leaved forests. Results showed that in both P. pubescens and broad-leaved forests, more than 70% of the E. henryi seeds were distributed in moss and litter layers, and few were found in the soil beneath them. However, seed germination in the soil layer was significantly higher than in the moss and litter layers. Seed density, overall seed quality, and germination rate in the broad-leaved forest were significantly higher than in P. pubescens forest. Seed germination was highest in the microsites around the edge of the crown projection area of E. henryi mother trees. The order of survival rate of the seedlings on different ground surfaces was soil > moss > litter. In both habitats, the average seed density was 24.9 seeds m−2, and the total germination rate was less than 3.5‰. However, seedlings developed from only 1% of the germinated seeds, indicating that the seed germination is the most important stage in the natural regeneration of E. henryi.

Rationale: Peripheral nerve injury is common in clinic, which leads to severe atrophy and dysfunction of the denervated muscles, but the underlying mechanism is not fully understood. Recent studies advanced the causative role of mitochondrial dysfunction in muscle atrophy, while the upstream triggers remained unclear. Methods: In the present study, Atrophy of gastrocnemius and tibialis anterior (TA) were evaluated in mice sciatic nerve transection model. Transmission electron microscopy (TEM) was then used to observe the microstructure of atrophic gastrocnemius and mitochondria. Subsequently, small RNA sequencing, luciferase reporter assay and Electrophoretic Mobility Shift (EMSA) were performed to explore the potential signaling pathway involved in skeletal muscle atrophy. The effects of the corresponding pathway on mitochondrial function, mitophagy, apoptosis and muscle atrophy were further determined in C2C12 cells and denervated gastrocnemius. Results: Gastrocnemius and TA atrophied rapidly after denervation. Obvious decrease of mitochondria number and activation of mitophagy was further observed in atrophic gastrocnemius. Further, miR-142a-5p/ mitofusin-1 (MFN1) axis was confirmed to be activated in denervated gastrocnemius, which disrupted the tubular mitochondrial network, and induced mitochondrial dysfunction, mitophagy and apoptosis. Furthermore, the atrophy of gastrocnemius induced by denervation was relieved through targeting miR-142a-5p/MFN1 axis. Conclusions: Collectively, our data revealed that miR-142a-5p was able to function as an important regulator of denervation-induced skeletal muscle atrophy by inducing mitochondrial dysfunction, mitophagy, and apoptosis via targeting MFN1. Our findings provide new insights into the mechanism of skeletal muscle atrophy following denervation and propose a viable target for therapeutic intervention in individuals suffering from muscle atrophy after peripheral nerve injury.

Background Transforming growth factor beta 1 (TGF-β1) is a classical modulator of skeletal muscle and regulates several processes, such as myogenesis, regeneration and muscle function in skeletal muscle diseases. Skeletal muscle atrophy, characterized by the loss of muscle strength and mass, is one of the pathological conditions regulated by TGF-β1, but the underlying mechanism involved in the atrophic effects of TGF-β1 is not fully understood. Methods Mice sciatic nerve transection model was created and gastrocnemius were analysed by western blot, immunofluorescence staining and fibre diameter quantification after 2 weeks. Exogenous TGF-β1 was administrated and high-mobility group box-1 (HMGB1), autophagy were blocked by siRNA and chloroquine (CQ) respectively to explore the mechanism of the atrophic effect of TGF-β1 in denervated muscle. Similar methods were performed in C2C12 cells. Results We found that TGF-β1 was induced in denervated muscle and it could promote atrophy of skeletal muscle both in vivo and in vitro, up-regulated HMGB1 and increased autophagy activity were also detected in denervated muscle and were further promoted by exogenous TGF-β1. The atrophic effect of TGF-β1 could be inhibited when HMGB1/autophagy pathway was blocked. Conclusions Thus, our data revealed that TGF-β1 is a vital regulatory factor in denervated skeletal muscle in which HMGB1/ autophagy pathway mediates the atrophic effect of TGF-β1. Our findings confirmed a new pathway in denervation-induced skeletal muscle atrophy and it may be a novel therapeutic target for patients with muscle atrophy after peripheral nerve injury. Introduction Abnormal inflammation mediated by Toll-like receptor 4 (TLR4) signaling pathway contributes to preeclampsia (PE). Because curcumin can inhibit TLR4 signaling pathway, we investigated its effects on a PE rat model. Methods Twenty-one pregnant rats were randomly divided into three groups: 1) seven rats were injected 0.5 μg/kg lipopolysaccharide (LPS) on gestational day (GD) 5 to create a PE model (LPS-treated group), 2) seven rats were injected with a similar dosage of LPS and further treated with curcumin (0.36 mg/kg) (LPS-curcumin-treated group), 3) seven rats received saline (control group). Blood pressure and urinary protein level were observed. Immunostaining and periodic acid-Schiff staining of placenta were conducted. TLR4 and downstream Nuclear Factor-κB (NF-κB) expressions of placenta were analyzed by Western blot and immunohistochemistry. IL-6 and MCP-1 in rat serum and placenta were determined by ELISA and qRT-PCR. Results Compared to LPS-treated group, LPS-curcumin-treated group had decreased blood pressure and urinary protein level, similar to control group. Furthermore, deficient trophoblast invasion and spiral artery remodeling induced by LPS were improved by curcumin. Increased TLR4, NF-κB and IL-6, MCP-1 protein expressions in LPS-treated group were significantly decreased after curcumin administration. Discussion Curcumin improves the PE-like phenotype in rat model by reducing abnormal inflammation related to TLR4 signaling pathway.

Mars dust accumulated on the solar array can affect the long-term performance of the solar array. Due to the thin gas flow on Mars, it is difficult to carry out experimental research and this paper study the wind removal process of Martian dust through theory and simulation. Firstly, this paper analyzed the mechanical properties of Martian dust and simulated Martian dust and established the discrete element model of Martian dust. Then the effect of Martian dust on spacecraft under different turbulence models in DEM-CFD model was analyzed. Finally, the simulation was performed and the influence of various parameters on the critical start-up speed of Mars dust was analyzed.

In this paper, the performance of wavelet transform on the detection and location of pipeline leak are explored. The effects of three wavelets on different thresholds and different decomposition layers are studied. Firstly, the UWB(Ultra Wideband) chaotic signal generated by Colpittscircuit is used as the signal detection signal, and the leak point of the pipeline is detected and located by analyzing the cross-correlation coefficient between the detection signal and the echo signal after the pipeline reflection. Because there are some questions such as how to choose the optimal threshold function, wavelets and optimal number of decomposed layers, three wavelet thresholds (hard threshold, soft threshold and given threshold)which has different signal-to-noise ratio (5dB, 10dB, 30dB) are used in signal threshold denoising. The paper explores the denoising effects of three wavelets(sym4, db4, Haar) after three, four and five wavelet decomposition, and finally gets the optimal threshold method, wavelet function and decomposition layer. This is important to achieve high-precision detection and location of water pipeline leak.

We numerically investigate the complex and the broadband characteristics of semiconductor ring laser (SRL) with optical feedback. The results show that the SRL with optical feedback is brought into chaotic behaviors with proper parameters and the dynamic behavior of which is complex. The bifurcation diagrams of chaos generation processes and detailed states with different parameters are demonstrated. The bandwidth of the chaos is also studied in this paper and the widest bandwidth is about 18 GHz when the feedback coefficient is 0.5 which is wider than the traditional edge-emitting lasers with optical feedback.
