BackgroundWhole genome duplication, which results in polyploidy, is a common feature of plant populations and a recurring event in the evolution of flowering plants. Polyploidy can result in changes to gene expression and epigenetic instability. Several epigenetic phenomena, occurring at the transcriptional or post-transcriptional level, have been documented in allopolyploids (polyploids derived from species hybrids) of Arabidopsis thaliana, yet findings in autopolyploids (polyploids derived from the duplication of the genome of a single species) are limited. Here, we tested the hypothesis that an increase in ploidy enhances transgene-induced post-transcriptional gene silencing using autopolyploids of A. thaliana.Methodology/principal findingsDiploid and tetraploid individuals of four independent homozygous transgenic lines of A. thaliana transformed with chalcone synthase (CHS) inverted repeat (hairpin) constructs were generated. For each line diploids and tetraploids were compared for efficiency in post-transcriptional silencing of the endogenous CHS gene. The four lines differed substantially in their silencing efficiency. Yet, diploid and tetraploid plants derived from these plants and containing therefore identical transgene insertions showed no difference in the efficiency silencing CHS as assayed by visual scoring, anthocyanin assays and quantification of CHS mRNA.Conclusions/significanceOur results in A. thaliana indicated that there is no effect of ploidy level on transgene-induced post-transcriptional gene silencing. Our findings that post-transcriptional mechanisms were equally effective in diploids and tetraploids supports the use of transgene-driven post-transcriptional gene silencing as a useful mechanism to modify gene expression in polyploid species.

Introduction: In randomized controlled trials, add-on brivaracetam (BRV) reduced seizure frequency in patients with drug-resistant focal epilepsy. Most real-world research on BRV has focused on refractory epilepsy. The aim of this analysis was to assess the 12-month effectiveness and tolerability of adjunctive BRV when used as early or late adjunctive treatment in patients included in the BRIVAracetam add-on First Italian netwoRk Study (BRIVAFIRST). Methods: BRIVAFIRST was a 12-month retrospective, multicenter study including adult patients prescribed adjunctive BRV. Effectiveness outcomes included the rates of sustained seizure response, sustained seizure freedom, and treatment discontinuation. Safety and tolerability outcomes included the rate of treatment discontinuation due to adverse events (AEs) and the incidence of AEs. Data were compared for patients treated with add-on BRV after 1-2 (early add-on) and ≥ 3 (late add-on) prior antiseizure medications. Results: A total of 1029 patients with focal epilepsy were included in the study, of whom 176 (17.1%) received BRV as early add-on treatment. Background: In randomized controlled trials, add-on brivaracetam (BRV) reduced seizure frequency in patients with drug-resistant focal epilepsy. Studies performed in a naturalistic setting are a useful complement to characterize the drug profile. Objective: This multicentre study assessed the effectiveness and tolerability of adjunctive BRV in a large population of patients with focal epilepsy in the context of real-world clinical practice. Methods: The BRIVAFIRST (BRIVAracetam add-on First Italian netwoRk STudy) was a retrospective, multicentre study including adult patients prescribed adjunctive BRV. Patients with focal epilepsy and 12-month follow-up were considered. Main outcomes included the rates of seizure‐freedom, seizure response (≥50% reduction in baseline seizure frequency), and treatment discontinuation. The incidence of adverse events (AEs) was also considered. Analyses by levetiracetam (LEV) status and concomitant use of strong enzyme-inducing antiseizure medications (EiASMs) and sodium channel blockers (SCBs) were performed. Results: A total of 1029 patients with a median age of 45years (33–56) was included. At 12 months, 169 (16.4%) patients were seizure-free and 383 (37.2%) were seizure responders. The rate of seizure freedom was 22.3% in LEV-naive patients, 7.1% in patients with prior LEV use and discontinuation due to insufficient efficacy, and 31.2% in patients with prior LEV use and discontinuation due to AEs (p

- Abstract
Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease with a lifetime risk of one in 350 people and an unmet need for disease-modifying therapies. We conducted a cross-ancestry genome-wide association study (GWAS) including 29,612 patients with ALS and 122,656 controls, which identified 15 risk loci. When combined with 8,953 individuals with whole-genome sequencing (6,538 patients, 2,415 controls) and a large cortex-derived expression quantitative trait locus (eQTL) dataset (MetaBrain), analyses revealed locus-specific genetic architectures in which we prioritized genes either through rare variants, short tandem repeats or regulatory effects. ALS-associated risk loci were shared with multiple traits within the neurodegenerative spectrum but with distinct enrichment patterns across brain regions and cell types. Of the environmental and lifestyle risk factors obtained from the literature, Mendelian randomization analyses indicated a causal role for high cholesterol levels. The combination of all ALS-associated signals reveals a role for perturbations in vesicle-mediated transport and autophagy and provides evidence for cell-autonomous disease initiation in glutamatergic neurons.

- Abstract
Fused filament fabrication (FFF) has seen an upsurge in its utilization towards development of tailored made materials of polymer base. The advancement and diversity in fabricating the polymer composite parts by using FFF has seen the embracement of this technology in wider aspects, ranging from automotive, aerospace, construction and has marched towards day to day requirements. This research article focuses on development of polymer composite; by using flyash (FA), an industrial waste produced during coal combustion, as reinforcement in Acrylonitrile butadiene styrene (ABS) matrix, to study the physical and mechanical properties. FA, which is primarily made up of metal oxides, plays an imperative role as reinforcement. Easily and abundantly available, FA is being used in several applications to reduce the landfills utilization and also helps the environment. In this study FA was added as reinforcement in 5 and 10 wt. % respectively to ABS matrix and was developed into filament of 1.75 mm diameter. The developed ABS + FA polymer composite using FFF, were analyzed for physical and mechanical properties as per American Society for Testing and Materials (ASTM) standards. Microstructure studies were carried out for the developed composite to understand their behavior in enhancing the dimensional accuracy and tensile strength with incremental addition of FA up to 10 wt%. Tensile strength was enhanced by 28.19% and 36.13% for ABS + 5wt. % FA and ABS + 10wt. % FA respectively. Dimensional stability was also enhanced. Similarly, surface roughness analysis was carried out and it was observed to reduce with addition of FA. The surface roughness measurements provided suitable results of decrement by 9.64% and 14.6% for ABS + 5wt. % FA and ABS + 10wt. % FA respectively. Overall, the usage of FA along with FFF, has paved a path in sustainable and green technology in manufacturing.

Background: To assess Guillain-Barré syndrome (GBS) incidence we relied on the Italian Network for the study of GBS (ITANG) established in 2010 in 7 Italian regions to analyse the association between influenza vaccination and GBS. Methods: All individuals aged ≥18 years, presenting with clinical manifestations that suggested GBS according to the universally accepted Asbury's diagnostic criteria (1990) were prospectively notified to a centralised database by ITANG neurologists over the period October 1, 2010-September 30, 2011. Through a telephone survey, 9 trained interviewers followed up the cases to diagnosis and then for 1 year since hospital discharge. Validation of case reporting was performed with the support of administrative data in 5 regions. Results: We found 365 cases fulfilling the definition for GBS or one of its variants over 19,846,068 population ≥18 years of age, yielding an annual incidence rate of 1.84 per 100,000 (95% CI 1.65-2.03), 2.30 (95% CI 1.99-2.60) in men and 1.41 (95% CI 1.18-1.64) in women. A highly significant peak of incidence was observed in February 2011 as compared to reference month (September 2011, rate ratio 3.3:1, p < 0.01). Conclusions: In Italy, GBS incidence was among the highest reported in Europe and higher than previously observed in Italian studies.

29. Markov and Monte Carlo simulation of Waste-to-Energy power plants considering variable fuel analysis and failure rates

- Abstract
In the present work, a method has been developed to estimate the annual capacity factor of waste to energy power plants. The case study is an under-construction power plant located in an area of high precipitation rate. Existing data of the waste analysis in the region shows that its content, including the mass fractions of the paper, food, and plastic, change daily and seasonally. The variation of the waste analysis in addition to components' failure and the off-design performance of the steam cycle make the behavior of the system highly stochastic. To deal with this stochastic nature, the probability distribution function of waste analysis is constructed to consider a range of possible inputs. Moreover, time-varying failure rates are used in Markov analysis to calculate the system's availability by considering infant failures, random failures, and aging. Also, the off-design performance of the power plant is simulated by THERMOFLEX to capture nonlinearities caused by steam flow reduction in the steam turbine. The probability distribution functions of the MSW physical analysis are estimated by sampling from a pit near the location. The Monte Carlo simulation has been used to couple the stochastic nature of the MSW content to the simulation and operation of the power plant, modeled by THERMOFLEX. The results show 2300 Kilowatt for power generation in full load condition in the first year, highlighting 30% power derate due to Municipal Solid Waste quality reduction.

30. Anticancer drug discovery using multicellular tumor spheroid models

- Abstract
ABSTRACTIntroduction: Despite the increasing financial outlay on cancer research and drug discovery, many advanced cancers remain incurable. One possible strategy for increasing the approval rate of new anticancer drugs for use in clinical practice could be represented by three-dimensional (3D) tumor models on which to perform in vitrodrug screening. There is a general consensus among the scientific community that 3D tumor models more closely recapitulate the complexity of tumor tissue architecture and biology than bi-dimensional cell cultures. In a 3D context, cells are connected to each other through tissue junctions and show proliferative and metabolic gradients that resemble the intricate milieu of organs and tumors.Areas covered: The present review focuses on available techniques for generating tumor spheroids and discusses current and future applications in the field of drug discovery. The article is based on literature obtained from PubMed.Expert opinion: Given the relative simplicity of spheroid models with respect to clinical tumors, we must be careful not to overestimate the reliability of their drug-response prediction capacity. The next challenge is to combine our knowledge of co-culture methodologies with high-content imaging and advanced microfluidic technologies to improve the readout and biomimetic potential of spheroid-based models.

This study presents the simulation of retrofitting measures in a residential apartment located in Sydney based on energy consumption, building characteristics, and climatic conditions through energy audits. From the results, an upgrade to a casement aluminum framed double glazing window with a thermal conductance (U-Value) of 2.3 W/m
2 /o C, having an initial cost of $6,570, will save 41.2 % and 21 % of the energy used for heating and cooling equipment with a simple payback of 6.3 years and fuel cost savings of $1,046. In addition, 787kWh of electricity saved from white goods by investing $1,050 has a payback of 2.2 years and fuel cost savings of $472. Finally, the building will save 2.7 tons of GHG emissions, equivalent to three people reducing energy use by 20 % in Australia. The research is vital because retrofitting multi-residential buildings in Australia has not been fully explored, leaving 7.8 million existing buildings that are not energy efficient. - Abstract
To assess Guillain-Barre syndrome (GBS) incidence we relied on the Italian Network for the study of GBS (ITANG) established in 2010 in 7 Italian regions to analyse the association between influenza vaccination and GBS. Methods: All individuals aged ≥18 years, presenting with clinical manifestations that suggested GBS according to the universally accepted Asbury's diagnostic criteria (1990) were prospectively notified to a centralised database by ITANG neurologists over the period October 1, 2010-September 30, 2011. Through a telephone survey, 9 trained interviewers followed up the cases to diagnosis and then for 1 year since hospital discharge. Validation of case reporting was performed with the support of administrative data in 5 regions. Results: We found 365 cases fulfilling the definition for GBS or one of its variants over 19,846,068 population ≥18 years of age, yielding an annual incidence rate of 1.84 per 100,000 (95% CI 1.65-2.03), 2.30 (95% CI 1.99-2.60) in men and 1.41 (95% CI 1.18-1.64) in women. A highly significant peak of incidence was observed in February 2011 as compared to reference month (September 2011, rate ratio 3.3:1, p < 0.01). Conclusions: In Italy, GBS incidence was among the highest reported in Europe and higher than previously observed in Italian studies.

- Abstract
Aim:HPV DNA has never been investigated in nipple discharges (ND) and serum–derived extracellular vesicles, although its presence has been reported in ductal lavage fluids and blood specimens. Materials & methods:We analyzed 50 ND, 22 serum-derived extracellular vesicles as well as 51 pathologic breast tissues for the presence of 16 HPV DNA types. Results:We show that the presence of HPV DNA in the ND is predictive of HPV DNA-positive breast lesions and that HPV DNA is more represented in intraductal papillomas. We also show the presence of HPV DNA in the serum-derived extracellular vesicles. Conclusion:Our data supports the use of liquid biopsy to detect HPV DNA in breast pathology.

38. Thermal-energy analysis of natural “cool” stone aggregates as passive cooling and global warming mitigation technique.

- Abstract
Recently, research focused on the development of roofing and paving systems aimed at (i) enhancing the passive cooling at single-building level and (ii) mitigating urban heat island at inter-building scale. In this work, the optic-thermal behavior of a naturally cool gravel for roof and paving application is studied, through lab and field experimental analyses. Specific evaluation about the cooling potential is carried out with varying grain size of the same gravel and stone typology. Additionally, its cool roof effect is investigated through dynamic simulation in a case study university campus in Italy. Finally, the analysis of the global warming contribution of the gravel was performed, with reference to the most performing gravel size. The results show differences up to 24% in terms of albedo with varying the only grain size. Additionally, the gravel with the smallest grain size resulted the most performing in terms of summer energy saving and indoor thermal comfort. By shifting from single building scale models to global climate energy balance models, an equivalent carbon emission offset of 4400 tCO 2eq was found, imputable to the (i) energy saving contribution, and (ii) global warming mitigation effect produced by the enhanced albedo of the case study area. Recently, huge efforts were made to develop new passive solutions for optimizing building summer thermal-energy behavior. While cool roofs are well investigated, a lack of knowledge is detected about the benefits deriving from the combination of cool roofs and cool façades. This work aims at determining the thermal performance of innovative cool roofing membrane and cool façade painting when applied on a prototype building, through continuous monitoring. Additionally, sensitivity analysis is performed to investigate the thermal benefits of the coupled solutions. The present study deals with the preparation of albumin nanocapsules containing fenretinide and their evaluation in experimental models of human non-small cell lung cancer. These nanocapsules showed enhanced antitumor activity with respect to free fenretinide due to the solubilization effect of albumin on the hydrophobic drug, known to improve bioavailability. The high expression of caveolin-1 on the A549 cell surface further enhanced the antitumor activity of the nanoencapsulated fenretinide. Caveolin-1 favored albumin uptake and improved the efficacy of the fenretinide-loaded albumin nanocapsules, especially in 3-D cultures where the densely packed 3-D structures impaired drug diffusibility and severely reduced the activity of the free drug. The efficacy of the fenretinide albumin nanocapsules was further confirmed in tumor xenograft models of A549 by the significant delay in tumor progression observed with respect to control after intravenous administration of the novel formulation. The world-wide effort to reduce the environmental impact associated to the industrial sector is quickly producing an increasing feedback on national and international decision makers. In this context, the analysis of life-cycle based assessments on the main impact categories associated to the pre-production, production, assembly, use, and end-of- life phases represents a powerful tool towards a holistic interpretation of the footprint from industrial buildings. The Italian prefabricated building sector, characterized, on average, by local enterprises with regional coverage, has been investigated in order to study the Carbon and Energy Footprints. Data from a large company, running several facilities spread on the national territory, have been collected and analyzed in order to provide a parameterized evaluation of the GHG emission and the energy consumption associated to the single phases of the building life cycle as a function of the sensible design requirements. The quantification of the Carbon and the Energy footprint, associated to the prefabricated industrial building sector, is presented. The assessment procedure is performed through a parametric modeling of the building properties bases on the analysis of different sizes and designs. Introduction: Preserving the dental enamel structure during removal of orthodontic accessories is a clinician's obligation. Hence the search for an evidence based debonding protocol. Objective: to evaluate and compare, by means of scanning electron microscopy (SEM), the effects of four different protocols of bracket debonding and subsequent polishing on enamel surface, and to propose a protocol that minimizes damage to enamel surface. Methods: Twelve bovine permanent incisors were divided into four groups according to the instrument used for debonding and removal of the adhesive remnant. In groups 1 and 2, brackets were debonded with a straight debonding plier (Ormco Corp., Glendora, California, USA), and in groups 3 and 4, debonding was performed with the instrument Lift-Off (3M Unitek, Monrovia, California, USA). In groups 1 and 3, the adhesive remnant was removed using a long adhesive removing plier (Ormco Corp., Glendora, California, USA) and in groups 2 and 4, residual adhesive was removed with a tungsten carbide bur (Beavers Dental) at high-speed. After each stage of debonding and polishing, enamel surfaces were replicated and electron micrographs were obtained with 50 and 200X magnification. Results: All four protocols of debonding and polishing caused enamel irregularities. Conclusion: Debonding brackets with straight debonding plier, removal of adhesive remnant with a tungsten carbide bur and polishing with pumice and rubber cup was found to be the protocol that caused less damage to enamel surface, therefore this protocol is suggested for debonding brackets. This study aimed to compare in vitro the shear bond strength between metallic brackets (Abzil) with conventional mesh bases and metallic brackets with bases industrially sandblasted with aluminum oxide using three adhesive systems, in order to assess the influence of sandblasting on adhesiveness and to compare 3 different bonding systems. Two hundred and forty bovine incisors were used and randomly divided into 6 groups (40 teeth in each group), according to the bracket base and to the bonding system. The brackets were direct-bonded in bovine teeth with 3 adhesive systems: System A - conventional Transbond™ XT (3M - Unitek); System B - Transbond™ Plus Self Etching Primer + Transbond™ XT (3M - Unitek) and System C - Fuji ORTHO LC resin-reinforced glass ionomer cement in capsules (GC Corp.). Shear bond strength tests were performed 24 hours after bonding, in a DL-3000 universal testing machine (EMIC), using a load cell of 200 kgf and a speed of 1 mm/min. The results were submitted to statistical analysis and showed no significant difference between conventional and sandblasted bracket bases. However, comparison between the bonding systems presented significantly different results. System A (14.92 MPa) and system C (13.24 MPa) presented statistically greater shear bond strength when compared to system B (10.66 MPa). This study investigates public participation in heat impact reduction by analysing adaptive behaviours, familiarity with urban heat island (UHI) and cooling strategies, the perceived urgency of heat impact actions and citizen's willingness to pay through a questionnaire survey in Chongqing, China. The results indicate that air-conditioning systems are the dominant cooling facility in both work and living environments. Respondents had a moderately familiar understanding of several cooling strategies such as urban vegetation, shading devices, water-based artificial facilities, urban design for shading and ventilation and water bodies. Familiarity with innovative materials and techniques for pavements, roofs and façades was less than moderate. Urban planning and design for heat resilient cities was thought to be the most urgent intervention, followed by the establishment of temporary cooling facilities. Most respondents indicated that cost-sharing mechanisms for urban heat prevention and control systems should at least include the government, whilst 50% of the respondents preferred collaborative payment among government, developers, and owners. Only 41.6% of the interviewees expressed their willingness to pay, with a share varying between <20 and >80 RMB. A conservative estimate indicated that there could be an average payment of 45.95 RMB and 19.10 RMB among the 234 respondents who were willing to pay and all 562 respondents regardless of willingness, respectively. Respondents' heat-related responses and actions towards urban heat challenges were dependent on a wide range of factors like gender, age, education, economic status, health, exposure, habit formation and behavior change, social acceptance, etc. Moreover, such factors could interact with each other affecting public behavior with different weights. Overall, this study increases our understanding of people's perceptions and proactiveness in reducing urban heat and provides guidance for decision-makers towards a novel user-aware approach to the implementation of urban heat prevention, adaptation, and mitigation strategies. 48. Influence of Ridge Inclination and Implant Localization on the Association of Mandibular Kennedy Class I Removable Partial Denture

- Abstract
The aim of this study was to evaluate the tendency of displacement of the supporting structures of the distal extension removable partial denture (DERPD) associated to the implant with different inclinations of alveolar ridge and implant localizations through a two-dimensional finite-element method. Sixteen mandibular models were fabricated, presenting horizontal, distally descending, distally ascending, or descending-ascending ridges. All models presented the left canine and were rehabilitated with conventional DERPD or implant-retained prosthesis with the ERA system. The models were obtained by the AutoCAD software and transferred to the finite-element software ANSYS 9.0 for analysis. A force of 50 N was applied on the cusp tips of the teeth, with 5 points of loading of 10 N. The results were visualized by displacement maps. For all ridge inclinations, the assembly of the DERPD with distal plate retained by an anterior implant exhibited the lowest requisition of the supporting structures. The highest tendency of displacement occurred in the model with distally ascending ridge with incisal rest. It was concluded that the association of the implant decreased the displacement of the DERPD, and the anterior positioning of the implant associated to the DERPD with the distal plate preserved the supporting structures for all ridges.

49. Case Reports: Follicular Carcinoma Presenting as Autonomous Functioning Thyroid Nodule and Containing an Activating Mutation of the TSH Receptor(T620I) and a Mutation of The Ki-RAS(G12C) Genes

- Abstract
Most autonomous functioning thyroid nodules (AFTN) are benign thyroid follicular neoplasms. There are rare reports of malignant hot nodules, in which activating mutations of the TSH receptor(TSHR) were found. We report a case of follicular carcinoma presenting as an AFTN causing subclinical hyperthyroidism in a 64-year-old woman who had a 6-cm hot nodule in the left thyroid lobe. Genomic DNA was extracted from paraffin-embedded tissues from the tumor and extratumoral thyroid tissue. Sequence analyses revealed point mutations in two different genes: the normal ACC sequence at codon 620 of the TSHRgene was replaced by ATC, changing the threonine by isoleucine (T620I); and the wild-type GGT at codon 12 of Ki-RASmutated to TGT, replacing glycine by cysteine (G12C). In transfection experiments the T620I mutant showed constitutive activity in terms of cyclic adenosine monophosphate (cAMP) production when permanently transfected in 3T3 cells. Here, we describe for the first time an activating mutation in 3codon 620 of the TSHR. In addition, the cancerous AFTN also contained a G12C Ki-RASmutation. We hypothesize that the combination of these two mutations might have played an important role in both the hyperfunction of the tumor and the carcinogenetic process.

50. Two Episodes of Ifosfamide-Related Neurotoxicity in the Same Patient following Different Schedules and Doses of the Drug. A Case Report

- Abstract
We report the first case of recurrent ifosfamide-related neurotoxicity in the same patient following two distinct administrations of the drug at different doses and schedules and with a long interval between the two episodes. Remarkably, the first event was characterized by confusion and hallucinations, while the second, 29 months later, was characterized by partial and generalized seizures. Between the two episodes the patient had received high-dose cyclophosphamide, an oxazophoshorine agent closely related to ifosfamide, without any neurological side effects. We briefly discuss the diagnosis and management of ifosfamide-related encephalopathy.
