The vestibular system in the inner ear plays a central role in sensorimotor control by informing the brain about the orientation and acceleration of the head. However, most experiments in neurophysiology are performed using head-fixed configurations, depriving animals of vestibular inputs. To overcome this limitation, we decorated the utricular otolith of the vestibular system in larval zebrafish with paramagnetic nanoparticles. This procedure effectively endowed the animal with magneto-sensitive capacities: applied magnetic field gradients induced forces on the otoliths resulting in robust behavioral responses comparable to that evoked by rotating the animal by up to 25°. We recorded the whole-brain neuronal response to this fictive motion stimulation using light-sheet functional imaging. Experiments performed in unilaterally injected fish revealed the activation of a commissural inhibition between the brain hemispheres. This magnetic-based stimulation technique for larval zebrafish opens new perspectives to functionally dissect the neural circuits underlying vestibular processing and to develop multisensory virtual environments, including vestibular feedback.HighlightsAfter injecting a ferrofluid into the inner ear of a larval zebrafish, the ear-stones can be actuated via magnetic forces.This method allows one to elicit vestibular-like behavioral responses without impairing physiological inner ear functions.It is compatible with brain-scale functional imaging and thus offers a promising avenue to investigate the neural underpinnings of vestibular-driven behaviors.eTOCFerrofluid injection into zebrafish inner ear allows magnetic manipulation of ear-stones to evoke vestibular responses in static animals. We present a simple method to produce giant lipid pseudo-vesicles (vesicles with an oily cap on the top), trapped in an agarose gel. The method can be implemented using only a regular micropipette and relies on the formation of a water/oil/water double droplet in liquid agarose. We characterize the produced vesicle with fluorescence imaging and establish the presence and integrity of the lipid bilayer by the successful insertion of α-Hemolysin transmembrane proteins. Finally, we show that the vesicle can be easily mechanically deformed, non-intrusively, by indenting the surface of the gel.

The Faint Intergalactic Medium Redshifted Emission Balloon (FIREBall) is a mission designed to observe faint emission from the circumgalactic medium of moderate redshift (z~0.7) galaxies for the first time. FIREBall observes a component of galaxies that plays a key role in how galaxies form and evolve, likely contains a significant amount of baryons, and has only recently been observed at higher redshifts in the visible. Here we report on the 2018 flight of the FIREBall-2 Balloon telescope, which occurred on September 22nd, 2018 from Fort Sumner, New Mexico. The flight was the culmination of a complete redesign of the spectrograph from the original FIREBall fiber-fed IFU to a wide-field multi-object spectrograph. The flight was terminated early due to a hole in the balloon, and our original science objectives were not achieved. The overall sensitivity of the instrument and telescope was 90,000 LU, due primarily to increased noise from stray light. We discuss the design of the FIREBall-2 spectrograph, modifications from the original FIREBall payload, and provide an overview of the performance of all systems. We were able to successfully flight test a new pointing control system, a UV-optimized, delta-doped and coated EMCCD, and an aspheric grating. Here we discuss advances in UV technology over the last decade, with an emphasis on photon counting, low noise, high efficiency detectors in sub-orbital programs. We focus on the use of innovative UV detectors in a NASA astrophysics balloon telescope, FIREBall-2, which successfully flew in the Fall of 2018. The FIREBall-2 telescope is designed to make observations of distant galaxies to understand more about how they evolve by looking for diffuse hydrogen in the galactic halo. The payload utilizes a 1.0-meter class telescope with an ultraviolet multi-object spectrograph and is a joint collaboration between Caltech, JPL, LAM, CNES, Columbia, the University of Arizona, and NASA. The improved detector technology that was tested on FIREBall-2 can be applied to any UV mission. We discuss the results of the flight and detector performance. We will also discuss the utility of sub-orbital platforms (both balloon payloads and rockets) for testing new technologies and proof-of-concept scientific ideas, Submitted to the Proceedings of SPIE, Defense + Commercial Sensing (SI19)

International audience; The Polarized Instrument for Long-wavelength Observation of the Tenuous interstellar medium (PILOT) is a balloon-borne experiment that aims to measure the polarized emission of thermal dust at a wavelength of 240 µm (1.2 THz). A first PILOT flight of the experiment took place from Timmins, Ontario, Canada, in September 2015 and a second flight took place from Alice Springs, Australia in April 2017. In this paper, we present the inflight performance of the instrument. Here we concentrate on the instrument performance as measured during the second flight, but refer to the performance observed during the first flight, if it was significantly different. We present a short description of the instrument and the flights. We measure the time constants of the detectors using the decay of the observed signal during flight following high energy particle impacts (glitches) and switching off the instrument's internal calibration source. We use these time constants to deconvolve the timelines and analyze the optical quality of the instrument as measured on planets. We then analyze the structure and polarization of the instrumental background. We measure the detector response flat field and its time variations using the signal from the residual atmosphere and from the internal calibration source. Finally, we analyze the spectral and temporal properties of the detector noise. The inflight performance is found to be satisfactory and globally in line with expectations from ground calibrations. We conclude by assessing the expected inflight sensitivity of the instrument in light of the measured inflight performance.

PILOT (Polarized Instrument for Long wavelength Observations of the Tenuous interstellar medium) is a balloonborne astronomy experiment designed to study the polarization of dust emission in the diffuse interstellar medium in our Galaxy. The PILOT instrument allows observations at wavelengths 240 μm (1.2THz) with an angular resolution about two arc-minutes. The observations performed during the first flight in September 2015 at Timmins, Ontario Canada, have demonstrated the optical performances of the instrument.

PILOT is a balloon-borne astronomy experiment designed to study the polarization of dust emission in the diffuse interstellar medium in our Galaxy at wavelengths 240 and 550 µm with an angular resolution of about two arc-min. PILOT optics is composed of an off-axis Gregorian telescope and a refractive re-imager system. All these optical elements, except the primary mirror, are in a cryostat cooled to 3K. We used optical and 3D measurements combined with thermo-elastic modeling to perform the optical alignment. FIREBALL (the Faint Intergalactic Redshifted Emission Balloon, funded by CNES-NASA, PI C.Martin, Caltech) is a balloon-borne 1m telescope coupled to an ultraviolet Multi Object Spectrometer (MOS), designed to study the faint and diffuse emission of the circumgalactic medium. The third flight of the experiment is planned in summer 2017. The goal of this paper is to describe the accurate pointing system of the 5-metres high / 1500kg gondola - that has been designed to fulfill stringent pointing requirements: less than 1 arcsec in elevation and cross elevation, and about 1 arcmin in field rotation (around the line of sight axis), over long integration time (a few hours). The pointing system is based on a multi stage closed loop scheme (4 Degrees Of Freedom), relying on a 1DOF gondola azimuth controller, a 2DOF gimbal frame supporting a 1.2-meter plano siderostat, and a 1DOF field rotation control system. The attitude determination is based on the hybridization of two accurate sensors: a Fiber Optic Gyrometer measurement unit and a star sensor integrated inside the instrument. The manuscript presents the design of the ACS. We also focus on flight train stability issues - due to the pendulum and torsion modes -, on the geometric equations specific to a siderostat pointing system, and on the description of the tests facilities.

PILOT is a stratospheric experiment designed to measure the polarization of dust FIR emission, towards the diffuse interstellar medium. The first PILOT flight was carried out from Timmins in Ontario-Canada on September 20th 2015. The flight has been part of a launch campaign operated by the CNES, which has allowed to launch 4 experiments, including PILOT. The purpose of this paper is to describe the performance of the instrument in flight and to perform a first comparison with those achieved during ground tests. The analysis of the flight data is on-going, in particular the identification of instrumental systematic effects, the minimization of their impact and the quantification of their remaining effect on the polarization data. At the end of this paper, we shortly illustrate the quality of the scientific observations obtained during this first flight, at the current stage of systematic effect removal.

Ce texte resulte de la transcription d’une table ronde qui s’est organisee en deux temps. Tout d’abord, les participants ont presente leurs analyses de l’etat actuel des enseignements de sciences humaines et sociales, que ce soit au lycee, dans les classes preparatoires, les filieres de grandes ecoles, et les universites. Les intervenants ont en general insiste sur les difficultes de positionnement des filieres de sciences humaines et les difficultes afferentes de proposer aux etudiants des debouches nombreux et attractifs. A partir de ce constat, un second temps de la discussion a permis d’evoquer differentes perspectives de sortie de crise. Tous les participants insistent sur la necessite de reviser les cursus et les pratiques pedagogiques.

L’article examine les problemes poses par la gouvernance des academiques dans leurs organisations. Apres avoir decrit ces personnels comme des professionnels et clarifie leurs missions, l’article montre comment une gouvernance coherente peut se traduire en choix et formes organisationnels mais aussi en modalites concretes de fonctionnement qui pourraient ameliorer le niveau d’excellence des institutions academiques.

We investigated the paraffinic hydrocarbon composition of cell suspensions of Euphorbia characias L. grown in different regimes of carbon and energy supply. Heterotropic (H), photomixotrophic (PM) and photoautotrophic (PA) cell suspensions were compared to leaves. Alkane compositions varied greatly both quantitatively and qualitatively according to the mode of culture. PM and H cells had indentical gaussian-like hydrocarbon distributions, with maxima around C 27 or C 28 compounds, while PA cells and leaf hydrocarbons were enriched in odd carbon chain-length compounds, especially C 29 , C 31 and C 33 In long-term cultivated suspensions, total hydrocarbon amounts of PM and PA cells were about 5% and 6% that of leaf, respectively, and PA suspensions presented a major enrichment of all odd carbon chain-length compounds. Minor differences were observed between the growth phases of PM as well as PA cells. On the other hand, major differences appeared between these two modes of culture and were amplified along subcultures, indicating that the paraffinic hydrocarbon composition was directly related to the mode of culture. We discuss the possible origins of these alkane patterns.

Photoautotrophic, photomixotrophic and heterotrophic cell suspension cultures of Euphorbia characias were analysed for fatty acid composition of their neutral lipids. Contents in exponential and stationary phases of growth were compared to those of seeds, leaves and calli from which cell suspensions were derived. Quantitatively, on a dry weight basis, neutral lipid amounts varied in the different cells from 15 to 40% of that in the leaves (5.14 mg per g dry wt) but they were very low compared to that in the seeds (330 mg per g dry wt). The highest values were observed during the exponential phase of growth with photosynthetic cell suspensions. Qualitatively, fatty acid distribution was similar in photoautotrophic cell suspensions and leaves with a high content of linolenic acid. The linolenic acid/linoleic acid ratio decreased from photoautotrophic to heterotrophic cell suspensions. In seeds, the maximal amount of linoleic acid is reached while linolenic acid almost disappears. The desaturation of C18 fatty acids is closely linked with the development of the photosynthetic apparatus and with a low growth rate.

The low-temperature linear dichroism spectrum of thylakoids oriented in polyacrylamide gel can be adequately described by a linear combination of the corresponding spectra of particles of light-harvesting complex, Photosystem I and Photosystem II, isolated by Triton X-100 extraction. The main conclusions which can be derived from this observation are: (1) The in vivo orientation of the pigments within each of the three complexes is not significantly affected by the extraction and purification procedures. (2) The various photosynthetic pigments are oriented roughly to the same extent in each of the three main biochemical constituents of the thylakoid. (3) All the complexes investigated behave like ellipsoids, the largest dimensions of which are lying in the plane of the photosynthetic membrane.

The nature and possible causes of polarized light-scattering artefacts in linear dichroism measurements are investigated. Using criteria described in this article, the available orientation techniques have been critically assessed in order to obtain the linear dichroism spectra of thylakoids and of pigment-protein complexes isolated from pea. It is demonstrated here that the polyacrylamide gel squeezing technique of Abdourakhmanov et al. (Abdourakhmanov, I.A., Ganago, A.O., Erokhim, Yu.E., Solov'ev, A.A. and Chugunov, V.A. (1979) Biochim. Biophys. Acta 546, 183–186) does not lead to pigment degradation and that the linear dichroism spectra obtained in these conditions are essentially free of scattering artefacts. The linear dichroism spectra of light-harvesting complex isolated in different states of aggregation or incorporated into phospholipid vesicles are compared to the spectra of thylakoids. This comparison indicates: (1) that the isolation procedure of Burke et al. (Burke, J.J., Ditto, C.L. and Arntzen, C.J. (1978) Arch. Biochem. Biophys. 187, 252–263) leads to light-harvesting complex in which the in vivo orientation of pigments is preserved; (2) that the antenna chlorophyll a molecules of this complex have a significant degree of orientation with respect to the plane of the thylakoid.

Cell suspension cultures of four plant species have been maintained under different carbon and energy supply regimes, i.e. photoautotrophy, photomixotrophy and heterotrophy, and have been assayed for their capacity to biotransform geraniol. The reactions of interconversion of geraniol into nerol, or oxidation of alcohols into their respective aldehydes, were mainly determined by the plant species, whatever the modes of culture were. A rapid metabolism of monoterpenic alcohols to unidentified compounds was observed. In one cell suspension ( Glycine max ), a high biotransformation activity (40–60%) into neral and geranial was detected.

The absorption and linear dichroism (LD) spectra (380–780 nm) of isolated light-harvesting complex (LHC), Photosystem I (PS I), Photosystem II (PS II), as well as intact thylakoids have been determined at 300 and 100 K. The samples were oriented in squeezed polyacrylamide gel. The low-temperature spectra of LHC and PS I present LD signals which are characteristic enough to be recognized in the LD spectrum of thylakoids. Tentative assignments of the various features of the LD spectra to the major photosynthetic pigments are discussed. A shoulder in the low-temperature absorption spectra is observed at about 673 nm in all the systems under investigation. The absence of an associated LD signal suggests that this ubiquitous chlorophyll (Chl) a form is non-dichroic. Furthermore, in the three isolated chlorophyll-protein complexes described in this study the sign of the LD signal indicates that both the Qy transition of the Chl a and the carotenoid molecules are preferentially oriented parallel to the largest dimension(s) of the particles.

Using a polyacrylamide gel squeezing technique, linear dichroism spectra of thylakoids from wild-type and chlorophyll- b less barley have been obtained at 100 K. The calculated difference linear dichroism spectra, based on normalization at 690–695 nm, are identical to those of the light-harvesting complex (LHC) isolated by Triton solubilization. This observation is in agreement with previous conclusions (Tapie, P., Haworth, P., Hervo, G. and Breton, J. (1982) Biochim. Biophys. Acta 682, 339–344) regarding: (i) scattering artifacts are absent in linear dichroism spectra determined using polyacrylamide gels, (ii) the in vivo orientation of LHC pigments is maintained in the isolated complex and (iii) the largest dimension(s) of the isolated LHC is (are), in vivo, parallel to the plane of the photosynthetic membrane.

The chlorophylle_protein complex LHC has been extensively studied in-vivo and in-vitro during the last years. Low temperature fluorescence measurements have shown that the fluorescence emission spectrum exhibits a more or less pronounced shoulder near 693 nm, acompaniing the main emission peak located at 682–685 nm, following the prepa rations. In order to analyse the elementary components constituing the emission spectra, we have carried out measurements under high frequency modulated ligth and the effect of agregation has been studied at two different temperatures: 300 and 77 K.

Using a mass-spectrometric (16)O(2)/(18)O(2)-isotope technique, we compared the nature and the relative importance of oxygen exchange in photomixotrophic (PM) and photoautotrophic (PA) suspensions of Euphorbia characias L. with those in intact leaves of the same species. Young and mature leaves, dividing and nondividing cell suspensions were characterized in short-term experiments. On chlorophyll basis, the gross photosynthetic activities at CO(2) saturating concentration of PA and PM suspensions varied little from those of leaves. On dry weight basis, gross photosynthesis of PA suspensions was equal to that of leaves because of their similar chlorophyll content. This was not the case in PM suspensions where gross photosynthesis was lower and largely varied during the growth cycle. The CO(2) compensation point of PA cells (155-265 parts per million) was much higher than that of leaves (50-80 ppm). Oxygen uptakes were analyzed in terms of mitochondrial respiration, photorespiration and light stimulation of oxygen uptake (LSOU), often identified to Mehlertype reactions. In PA and PM suspensions, mitochondrial respiration rates were higher than in leaves by a factor of 1.5 to 4.5. In PM suspensions, photorespiration and LSOU were observed only in nondividing cells. Photorespiration and LSOU rates were comparable in PA suspensions and leaves. Our results demonstrate that photorespiration of PA suspensions has not been affected by the 2% CO(2) concentration imposed during 2 years of culture.
