Negative chemical ionization (NCI) and electron-capture negative ionization (ECNI) are gas chromatography–mass spectrometry (GC–MS) techniques that generate negative ions in the gas phase for compounds containing electronegative atoms or functional groups. In ECNI, gas-phase thermal electrons can be transferred to electrophilic substances to produce M–• ions and scarce fragmentation. As a result of the electrophilicity requirements, ECNI is characterized by high-specificity and low background noise, generally lower than EI, offering lower detection limits. The aim of this work is to explore the possibility of extending typical advantages of ECNI to liquid chromatography–mass spectrometry (LC–MS). The LC is combined with the novel liquid-EI (LEI) LC–EIMS interface, the eluent is vaporized and transferred inside a CI source, where it is mixed with methane as a buffer gas. As proof of concept, dicamba and tefluthrin, agrochemicals with herbicidal and insecticidal activity, respectively, were chosen as model compounds and detected together in a commercial formulation. The pesticides have different chemical properties, but both are suitable analytes for ECNI due to the presence of electronegative atoms in the molecules. The influence of the mobile phase and other LC- and MS-operative parameters were methodically evaluated. Part-per-trillion (ppt) detection limits were obtained. Ion abundances were found to be stable with quantitative linear detection, reliable, and reproducible, with no influence from coeluting interfering compounds from the sample matrix.

We present a modified microfluidic open interface (MOI) for the direct coupling of Bio-SPME to a liquid electron ionization-tandem mass spectrometry (LEI-MS/MS) system as a sensitive technique that can directly analyze biological samples without the need for sample cleanup or chromatographic separations as well as without measurable matrix effects (ME). We selected fentanyl as test compound. The method uses a C18 Bio-SPME fiber by direct immersion (DI) in urine and plasma and the subsequent quick desorption (1 min) in a flow-isolated volume (2.5 μL) filled with an internal standard−acetonitrile solution. The sample is then transferred to an EI source of a triple-quadrupole mass spectrometer via a LEI interface at a nanoscale flow rate. The desorption and analysis procedure requires less than 10 min. Up to 150 samples can be analyzed without observing a performance decline, with fentanyl quantitation at microgram-per-liter levels. The method workflow is extremely dependable, relatively fast, sustainable, and leads to reproducible results that enable the high-throughput screening of various biological samples.

Solid-phase microextraction (SPME) directly coupled to MS is a widespread technique for determining small molecules in different matrices in many application fields. Here we present a modified microfluidic open interface (MOI) connected to a passive-flow-splitter device (PFS) for the direct coupling of SPME to a liquid-electron ionization (LEI) interface in a tandem mass spectrometer for the analysis of complex biological samples. No chromatographic separation is involved. The new MOI-PFS configuration was designed to speed up the sample transfer to MS, improving the signal-to-noise ratio and peak shape and leading to fast and sensitive results. MOI-PFS-LEI-MS/MS experiments were conducted using fentanyl as a model compound in water and blood serum. The method uses a C18 Bio-SPME fiber by direct immersion (3 min) in 300 µL of the sample followed by rapid desorption (1 min) in a flow isolated volume (MOI chamber, 2.5 µL) filled with 100% acetonitrile. The PFS permits the rapid transfer of a fraction of the sample into the MS via the LEI interface. The optimal conditions were obtained at a flow rate of 10 µL·min

This review article traces the history of the use of liquid chromatography coupled with mass spectrometry (LC-MS) using electron ionization (EI) from the first attempts up to the present day. At the time of the first efforts to couple LC to MS, 70 eV EI was the most common ionization technique, typically used in gas chromatography-mass spectrometry (GC-MS) and providing highly reproducible mass spectra that could be collated in libraries. Therefore, it was obvious to transport this dominant approach to the early LC-MS coupling attempts. The use of LC coupled to EI-MS is challenging mainly due to restrictions related to high-vacuum and high-temperature conditions required for the operation of EI and the need to remove the eluent carrying the analyte before entering the ion source. The authors will take readers through a journey of about 50 years, showing how through the succession of different attempts it has been possible to successfully couple LC with EI-MS, which in principle appear to be incompatible.

Herein we present an efficient, column-switching method that relies on a custom-made T-union passive diffusion micromixer to assist water dilution and promote trap solute focusing of a high sample volume dissolved in pure organic solvent using a 0.075 mm i.d. nano-LC column. This method allows injecting 20 μL (or higher) of sample volume, speeding up the analysis time, with a 400-fold increase of the limits of quantitation for selected compounds. Five pesticides in different media were used as model compounds, and the analyses were carried out with a triple quadrupole mass spectrometer equipped with a Liquid Electron Ionization (LEI) LC-MS interface working in multiple reaction monitoring (MRM) mode. The system microfluidics were investigated using COMSOL modeling software. Robustness of the entire system was evaluated using a post-extraction addition soil extracts with limits of detection values spanning from 0.10 to 0.45 µg/L. Reproducible results in terms of peak area, peak shape, and retention times were achieved in soil matrix. Repeatability test on peak area variations were lower than 10%.

Mass spectrometry has been the "gold standard" for drugs of abuse (DoA) analysis for many decades because of the selectivity and sensitivity it affords. Recent progress in all aspects of mass spectrometry has seen significant developments in the field of DoA analysis. Mass spectrometry is particularly well suited to address the rapidly proliferating number of very high potency, novel psychoactive substances that are causing an alarming number of fatalities worldwide. This review surveys advancements in the areas of sample preparation, gas and liquid chromatography-mass spectrometry, as well as the rapidly emerging field of ambient ionization mass spectrometry. Dispersive liquid-liquid microextraction with and without ultrasound assistance (DLLME, UA-DLLME) and microextraction with packed sorbent (MEPS) methods for the extraction and determination of eight different benzodiazepines (BDZ) (chlordiazepoxide, flurazepam, bromazepam, oxazepam, lorazepam, clobazam, clonazepam, and flunitrazepam) in three commercial non-alcoholic and light alcoholic beverages were optimized and compared. Benzodiazepines are frequently used for their extensive diffusion and strong numbing effect in drug-facilitated crimes (DFC). The tiny small amount of sample required for DLLME and MEPS extraction makes them very suitable for specimens collected at the crime scene of DFCs. Microextraction techniques are of increasing interest thanks to their accordance to green analytical chemistry (GAC) guidelines providing good recovery values. Ultrasound assistance (UA-DLLME) was used to investigate whether this type of energy can improve the recoveries of the analytes. Analyses of the extracts were performed with reverse-phase capillary high-performance liquid chromatography with UV detection (HPLC − UV), thanks to low environmental impact, robustness, diffusion, and affordability. Recovery percentages at three different concentrations in the three beverages were between 14.30% and 103.28% with intraday and interday RSD lower than ± 2.78%. The same samples were extracted using a MEPS protocol, and the results were compared with those obtained with DLLME. MEPS gave recoveries between 20.90% and 101.88% for all matrices showing a better performance than DLLME at higher concentrations, though lower recoveries were observed with diluted samples.

Extra virgin olive oil (EVOO) is one of the main ingredients of the Mediterranean diet. It is claimed as a functional food for its unique content of health-promoting compounds. Tyrosol (Tyr), Hydroxytyrosol (Htyr), and their phenolic derivatives present in EVOO show beneficial properties, and their identification and quantification, both in their free form and after the hydrolysis of more complex precursors, are important to certify its quality. An alternative method for quantifying free and total Tyr and Htyr in EVOO is presented using an LC–MS interface based on electron ionization (EI), called liquid electron ionization (LEI). This method requires neither sample preparation nor chromatography, the sample is diluted and injected. The selectivity and sensitivity were assessed in multiple reaction monitoring mode (MRM), obtaining confirmation and quantification in actual samples ranging from 5 to 11 mg/Kg for the free forms and from 32 to 80 mg/Kg for their total amount after hydrolysis. Two MS/MS transitions were acquired for both compounds using the Q/q ratios as confirmatory parameters. Standard addition calibration curves demonstrated optimal linearity and negligible matrix effects, allowing a correct quantification even without expensive and difficult to find labeled internal standards. After several weeks of operation, the system’s repeatability was excellent, with an intraday RSD (%) spanning from five to nine and an interday RSD (%) spanning from 9 to 11.

Liquid Electron Ionization (LEI), is an innovative liquid chromatography-mass spectrometry (LC-MS) interface that converts liquid HPLC eluent to the gas-phase in a mass spectrometer equipped with an electron ionization (EI) source. LEI extends the electronic spectra libraries access to liquid chromatography, providing a powerful tool in the untargeted approacssh. Negligible matrix effects allow accurate quantitative information. The purpose of this research was to evaluate the main aspects concerning the interfacing process. These fundamental studies were necessary to understand the mechanism of LEI in details, and improve the interfacing process, especially regarding robustness and sensitivity. Hardware components were installed to prevent analytes precipitation, reduce thermal decomposition of sensitive compounds, and to stabilize the nano-flow delivery with different mobile-phase compositions. Particular attention was devoted to insulating the heated vaporization area from the LC part of the system. Experiments were performed to optimize the interface inner capillary dimensions, and other operative parameters, including temperature, gas and liquid flow rates. Test compounds of environmental interest were selected based on molecular weight, thermal stability, volatility, and polarity. Robustness was evaluated with a set of replicated injections and calibration experiments using a soil matrix as a test sample. MRM detection limits in the low-picogram range were obtained for five pesticides belonging to different classes in a soil sample. High-quality electron ionization mass spectra of a mixture of pesticides were also obtained.

Buprenorphine and its metabolites are routinely monitored to assess patient compliance with drug detoxification programs or as pain killers. A rapid method for the simultaneous analysis of buprenorphine, norbuprenorphine, and glucuronides in urine using ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) was developed. Urine samples were diluted in water containing formic acid 0.1% and directly injected into the UHPLC-MS/MS system without any sample pretreatment. Quality control (QC) samples, prepared using 20 different urine matrices, fortified at 3 concentration levels, were quantified using four deuterated internal standards. The accuracy values obtained spanned from 90 to 114% with repeatability lower than 10% also in the inter-day between batch experiments. Matrix effects (ME), evaluated before correction with internal standards using Matuszewski procedure, mainly affected the analysis of buprenorphine glucuronide. The use of deuterated internal standards (IS) for each analyte was necessary to compensate for ME and was essential in the determination of glucuronides. The method was applied to 30 real urine samples from patients under a detoxification therapy. Duplicate analyses were performed with the presented method and compared with another method which involves a standard hydrolysis procedure. Real sample results were compared showing a good performance agreement, with differences between the two methods lower than ±20% in the quantification results.

The determination and quantification of benzodiazepines in very small residues of beverages, collected at the scene of drug-facilitated crimes are mandatory in legal procedures. A specific and sensitive analytical instrumentation is needed, involving little or no sample preparation. Here, a direct flow injection analysis of alcoholic beverages spiked with commercially available drugs containing diazepam and flunitrazepam is presented. The method proposed is very fast and requires neither sample preparation nor chromatographic separation. Linearity (R(2) ) was between 0.9977 and 0.9992; LOD and LOQ spanned from 0.01 to 0.02 ng/μL and from 0.1 to 0.5 ng/μL, respectively; intra- and interday repeatabilities were between 1 and 8%. No matrix effects were observed from the comparison of the linear regression curves obtained in real fortified samples and in pure ethanol. Vodka, whisky, and white wine specimens were fortified with commercial drugs, Valium(®) and Rohypnol(®) , at two different concentrations (20 and 50 ng/μL) to simulate the typical amounts found in adulterated real samples and analyzed to demonstrate the method applicability to forensic analyses.

Polycyclic aromatic hydrocarbons (PAHs) are USEPA regulated priority pollutants. Their low aqueous solubility requires very sensitive analytical methods for their detection, typically involving preconcentration steps. Presented is the first demonstrated 'proof of concept' use of condensed phase membrane introduction mass spectrometry (CP-MIMS) coupled with direct liquid electron ionization (DEI) for the direct, on-line measurement of PAHs in aqueous samples. DEI is very well suited for the ionization of PAHs and other nonpolar compounds, and is not significantly influenced by the co-elution of matrix components. Linear calibration data for low ppb levels of aqueous naphthalene, anthracene, and pyrene is demonstrated, with measured detection limits of 4 ppb. Analytical response times (t10%-90% signal rise) ranged from 2.8 min for naphthalene to 4.7 min for pyrene. Both intra- and interday reproducibility has been assessed (

Sudden infant death syndrome (SIDS) and sudden intrauterine unexpected death syndrome (SIUDS) are an unresolved teaser in the social-medical and health setting of modern medicine and are the result of multifactorial interactions. Recently, prenatal exposure to environmental contaminants has been associated with negative pregnancy outcomes, and verification of their presence in fetal and newborn tissues is of crucial importance. A gas chromatography-tandem mass spectrometry (MS/MS) method, using a triple quadrupole analyzer, is proposed to assess the presence of 20 organochlorine pesticides, two organophosphate pesticides, one carbamate (boscalid), and a phenol (bisphenol A) in human brain tissues. Samples were collected during autopsies of infants and fetuses that died suddenly without any evident cause. The method involves a liquid-solid extraction using n-hexane as the extraction solvent. The extracts were purified with Florisil cartridges prior to the final determination. Recovery experiments using lamb brain spiked at three different concentrations in the range of 1-50 ng g-1 were performed, with recoveries ranging from 79 to 106 %. Intraday and interday repeatability were evaluated, and relative standard deviations lower than 10 % and 18 %, respectively, were obtained. The selectivity and sensitivity achieved in multiple reaction monitoring mode allowed us to achieve quantification and confirmation in a real matrix at levels as low as 0.2-0.6 ng g-1. Two MS/MS transitions were acquired for each analyte, using the Q/q ratio as the confirmatory parameter. This method was applied to the analysis of 14 cerebral cortex samples (ten SIUDS and four SIDS cases), and confirmed the presence of several selected compounds.

An LC-MS method for the analysis of personal care and household products without sample preparation is presented. The method takes advantage of the Direct-electron ionization (EI) LC-MS interface for the quantitation of principal components, as well as for the identification of unknown or undeclared ingredients. The technique has proven its inertness toward matrix effects and the electron ionization allows quantitation and library identification. Commercially available products (shower gel, perfume, and hand cream) were diluted with methanol and injected directly into a nano-LC column. Limonene, linalool, and citral were selected as target compounds because of their use as fragrances in toiletry and detergent products. These and all other fragrances are commonly determined with GC-MS analysis, prior to sample cleanup, a procedure that can lead to analytes loss. The selected compounds are not detected with ESI because of their poor or very low response. Figures of merit and validation studies were executed and special attention was devoted to matrix-effects evaluation, because a sample preparation procedure is not involved. No matrix effects were observed, and the repeatability was excellent even after several weeks of operation. Products composition was investigated in full scan mode to determine the presence of unknown or not listed ingredients.

Nano-LC columns of different lengths (14–35 cm), 75 μm i.d., were packed with solid-core C18 particles using a conventional HPLC system at low pressure (375 bar) and without expensive tools and fittings. Solid-core particles consist of a solid, non-porous core surrounded with a shell of a porous layer with a very narrow particle size distribution. This geometry allows a faster diffusion of the analytes compared to porous particles, reducing the C term of the Knox plot. Different slurries of packing material were evaluated and tested. The packing procedure was carried out at room temperature and at 70 °C to evaluate the influence of this parameter on the overall process. The synergic action of pressure, temperature and sonication contributed to columns of various lengths in the packing process. The columns were tested at room temperature taking into account the following parameters: Knox plots, specific permeability and peak capacity. Reduced heights of theoretical plates, h, ranged between 3.8 and 5.1 at ν between 2 and 6. An LC-MS test was carried out with a Direct-EI LC-MS instrument.

Detection of target and non-target substances and their characterization in complex samples is a challenging task. Here we demonstrate that coating the electron ionization (EI) ion source of an LC-MS system with a sol-gel ceramic film can drastically improve the detection of high-molecular weight and high-boiling analytes. A new ion source coated with a ceramic material was developed and tested with a mixture of polycyclic aromatic hydrocarbons (PAH) with an increasing number of rings. Comparison of the results obtained with those for an uncoated stainless steel (SS) ion source shows a dramatic improvement in the MS signals, with a nearly 40-fold increase of the signal-to-noise ratio. We also demonstrate the ability of the new system to produce excellent chromatographic profiles for hard-to-detect hormones.

This article describes a nano-scale method for the determination and quantification of five benzodiazepines (BDZ) in an alcoholic grappa drink (chlordiazepoxide; lorazepam; diazepam; oxazepam; medazepam). BDZ are typically used in drug-facilitated crimes (DFC) for their accessibility and synergistic effects with alcohol. Specimens collected on the crime scene must be rapidly analyzed to prove the crime, though, in most cases, a very small amount is available. Off-line MEPS extraction of diluted grappa samples proved to be an efficient and reliable method for the recovery of the selected compounds. Requiring a very small amount of extraction solvents, MEPS is an environment-friendly technique. LC separation with UV detection was used as the analytical technique because it is simple, robust, relatively economic and easy-to-find in most laboratories. The method was validated in terms of precision, accuracy and recovery. Limits of detection and quantitation were in the range of 0.5-2ng/μL. Linearity (R(2)) spanned from 0.9994 and 1.0000. Intra- and inter-day repeatabilities were lower than 12% at any concentration. Recovery percentages of an equivalent-to-real sample at three different concentrations were between 70.7 and 74.1%.

We investigated the effect of temperature on the packing procedure of nano-LC columns (up to 50 cm) and on their performance. Several slurries of stationary phase were prepared using different solvent mixtures. Their stability was evaluated at several temperatures: 70°C, 50°C, and room temperature. At the higher temperature (70°C) the suspensions resulted to be stable for a longer time. For each slurry, we compared nano-LC columns packed with ultrasounds at 70°C and at room temperature. All the columns were tested with a standard mixture at 70°C, to reduce the solvent viscosity and the backpressure. Main chromatographic parameters such as the asymmetry factor, As, the reduced plate high, h, pattern in a Van Deemter plot, the total porosity, e(t), and the permeability, k, were calculated and discussed. One of the nano-LC columns was used to separate a mixture of pesticides in a LC-MS system with an electron ionization LC-MS interface (Direct-EI). From our knowledge, this is the first study on the role of temperature in the efficiency of slurry-packing procedure.

We present the validation of a multiresidue method for the concurrent SPE of organochlorine (OCP) and phenoxy-acidic pesticides (PA) in river water. The two classes of compounds show very different chemical characteristics. The analyses were performed by gas chromatography–mass spectrometry and by liquid chromatography–electrospray ionization–mass spectrometry. The method was validated at three different concentrations: method limit of quantitation, 0.5 and 5 μg L−1. The structure of the octadecyl (C18) chain layer attached to a silica surface in the presence of binary solvents (acetonitrile/water; methanol/water) was investigated by electron paramagnetic resonance (EPR) and reverse-phase high-performance liquid chromatography (RP-HPLC), using 4-hydroxy-2,2,6,6 tetramethylpiperidine-N-oxyl (Tempol) to mimic the behavior of pollutants with medium-low polarity. The computer-aided analysis of the EPR spectra provided structural and dynamical information of the probe and its environments which clarified the modifications of the chain conformations that occur at different solvent compositions. Capacity factors, k', were calculated as a function of the percentage of water/organic solvent (mobile phase), and the retention behavior of the C18-functionalized silica surface (stationary phase) was compared with the results obtained with EPR analysis under static conditions. In particular, EPR analysis showed that, at percentages of ACN equal or higher than 50%, the chain layer assume a quite ordered structure, whereas at percentages lower than 50% the chains tend to collapse and fold on the silica surface. In this latter situation, the hydrophobic net of the C18 chains strongly limits Tempol mobility. In methanol/water mixtures, both EPR and RP-HPLC analysis showed that the probe was adsorbed into a poorly ordered interphase. If the residual silanols at the silica surface were bonded to a sililating agent (endcapping), both EPR and RP-HPLC analysis showed a decreased adsorption of the probe with respect to the non-endcapped silica at the same mobile phase composition.

Matrix-dependent signal suppression or enhancement represents a major drawback in quantitative analysis with liquid chromatography coupled to atmospheric pressure ionization mass spectrometry (LC-API-MS). Because matrix effects (ME) might exert a detrimental impact on important method parameters (limit of detection, limit of quantification, linearity, accuracy, and precision), they have to be tested and evaluated during validation procedure. This review gives a detailed description on when these phenomena might be expected, and how they can be evaluated. The major sources of ME are discussed and illustrated with examples from bioanalytical, pharmaceutical, environmental, and food analysis. Because there is no universal solution for ME, the main strategies to overcome these phenomena are described in detail. Special emphasis is devoted to the sample-preparation procedures as well as to the recent improvements on chromatographic and mass spectrometric conditions. An overview of the main calibration techniques to compensate for ME is also presented. All these solutions can be used alone or in combination to retrieve the performance of the LC-MS for a particular matrix-analyte combination.

Despite their enormous utility and diffusion, atmospheric pressure ionization mass spectrometry techniques are subjected to relevant drawbacks called matrix effects (ME). These effects could be summarized in matrix-dependent signal suppression or enhancement that could lead to erroneous quantitative results. The most important method parameters as well as linearity, precision, and accuracy could be modified due to interfering compounds present in the matrix. No validation methods could be accepted without a thorough evaluation of ME and possible strategies to minimize or to correct their influence should be addressed. In this article, we investigate mechanisms that lead to ME and discuss calibration techniques and other strategies to limit these phenomena. Significant examples from different fields of application are discussed as well. Sample preparation procedures, together with instrumental improvements and alternative calibration techniques used by many authors, are reported.

Environmental contextMarine mining activities are potential sources of environmental pollution. Diethylene glycol used in offshore platforms has been suspected to facilitate the release of toxic substances into the sea. The results obtained elucidate that this release is not significant for the metals examined here, apart from iron, nor for polycyclic aromatic hydrocarbons, even at very high diethylene glycol concentrations. AbstractThe role of diethylene glycol (DEG) as a co-solvent for selected organic and inorganic pollutants adsorbed onto the particulate matter in produced formation water (PFW) from offshore gas platforms is thoroughly evaluated. Artificial seawater samples were spiked with certified sediments containing several polycyclic aromatic hydrocarbons (PAH) and metals. Aliquots (1 L) containing no DEG and DEG at 3500 and 5000 mg L−1 were kept in static and dynamic modes for 24 h before analysis to allow sufficient partitioning time between solid and liquid phases for the selected analytes. The Italian legislation on this matter sets 3500 mg L−1 as the highest concentration for DEG in PFW. In our experiments, concentrations equal to and above the set limit were chosen to enhance any possible co-solvent effect. Real PFW samples were also analysed, both with and without DEG. The analyses were conducted by using GC-MS for the PAH, and ICP-MS for the metals. A minor co-solvent effect was observed for low-molecular-weight PAH in the artificial seawater in static mode. Among metals, only iron showed an increase in solubility in the presence of DEG, demonstrating the co-solvent effect of DEG. The experiments in dynamic mode revealed no increase in the solubility of any other analytes in the liquid phase compared with static mode.

Benzodiazepines (BDZs) are widely used as tranquilizers and anti-depressive drugs in common clinical practice. However, their ready availability and their synergistic effects with alcohol make them attractive for criminal intentions. To prove criminal action for legal reasons, it is often necessary to analyze beverage residues from a crime scene. Milk-based alcoholic drinks (whiskey creams) are gaining popularity due to their lower alcohol content pleasant taste. However, the complexity of this sample, containing proteins and fatty acids, can mask the presence of drugs or other substances in standard analysis methods. These characteristics make whiskey creams highly suitable for illicit purposes. In this study, eight BDZs, including diazepam, chlordiazepoxide, clobazam, flunitrazepam, bromazepam, flurazepam, nitrazepam and clonazepam, were extracted from whiskey cream using the Quick, Easy, Cheap, Effective, Rugged and Safe (QuEChERS) method and analyzed using GC-MS. The QuEChERS protocol can efficiently separate most of the matrix from the target compounds while maintaining acceptable recoveries. The presented method is simple and rapid and has been validated in terms of precision, accuracy and recoveries. Limits of detection and limits of quantitation were in the range of 0.02-0.1 and 0.1-0.5 µg/mL, respectively. Whiskey cream beverages, fortified with commercial drugs at 20 µg/mL, were extracted and analyzed demonstrating the applicability of the method in forensic analysis.

A conspicuous fraction of the water soluble organic compounds (WSOC) in fog and fine aerosol samples is composed by monosaccharide anhydrides, such as levoglucosan and its stereoisomers, galactosan and mannosan. Levoglucosan is produced exclusively during wood combustion processes, making it a very useful tracer for plant combustion emissions in the atmosphere. This paper describes a new experimental approach, based on electrospray-tandem mass spectrometry (ESI-MS/MS), for the identification of levoglucosan in fog water samples. The analytical method proposed allows to identify the specific sugar anhydrides directly in the liquid phase without the need of any derivatization process.

With the advent of soft ionization methods such as MALDI and ESI, mass spectrometry has become the most important technique for the analysis of proteins and peptides. ESI-MS is often preceded by separation of the peptide sample by reversed-phase liquid chromatography (LC). Acetonitrile (ACN) is the most commonly employed organic solvent in LC-ESI-MS analysis of peptides. In this report, we demonstrate that the use of methanol (MeOH) as the organic modifier improves the detection limits for analysis of peptide mixtures such as those found in tryptic digests of proteins. A nanoLC-ESI-quadrupole ion trap instrument (LCQ Deca, ThermoFinnigan) was used to analyze peptide standards, protein digests of known concentrations, and tryptic digests of 2-DGE-separated proteins. MeOH displayed excellent chromatographic performance (separation and sensitivity), and shorter gradient times were possible for chromatographic separation with MeOH versus ACN. Sensitivity levels of a few hundred attomoles were achieved with MeOH; those levels could not be achieved with ACN. In addition, MeOH-based nanoLC-MS/MS yielded superior results for the analysis of digests of 2-DGE-separated proteins. For the 14 protein spots analyzed, the success rate of protein identification with MeOH-based nanoLC-ESI-MS/MS was 100%, with multiple proteins identified in several of the spots. In contrast, ACN-based procedure failed to identify any proteins in 21% of the spots and overall identified 33% fewer proteins than the MeOH-based procedure. In summary, higher sensitivity and shorter gradient times make MeOH an excellent organic modifier for the use in nanoLC-ESI-MS/MS analysis of peptides.

We describe a method, based on a new nano-HPLC gradient generator coupled to a modified direct electron ionization (EI) LC–MS interface, for the analysis of several compounds of environmental interest. The gradient generator was installed on a conventional HPLC system and the nano-flow rate was achieved using a double splitter. Reproducible gradients of different shapes (i.e. linear, convex, concave) can be generated at micro- and nano-flow rates. This attribute makes this device particularly appropriate for coupling with direct-EI LC–MS interface. The combination of the gradient generator with the direct-EI interface optimizes the analytical conditions for the generation of good quality EI spectra for trace level analytes. The performance of this new system was tested at a flow rate of 250 nl min−1, evaluating linearity, intra-day reproducibility and limits of detection of several compounds of environmental interest belonging to three different classes: nitro-polycyclic aromatic hydrocarbons, pesticides and human hormones. Excellent signal stability and low detection limits were obtained in most cases. Enhanced performance was observed for critical separations due to the possibility to run gradients of different shapes.

Water-soluble organic compounds (WSOC) often represent a large fraction of the total organic mass found in the atmospheric aerosol. They play a very important role in determining the ability of aerosol particles to act as cloud condensation nuclei (CCN), influencing cloud and fog formation and cloud albedo. Molecular characterization of WSOC in fogwater samples was achieved using a two-stage ion-trap mass spectrometer equipped with electrospray ionization (ESIMS/MS). Negative ionization conditions in the electrospray interface finalized our characterization of the acidic fraction of WSOC that comprises both mono- and di-carboxylic acids and polycarboxylic acids for which a similarity was suggested with naturally occurring humic (or fulvic) acids, and which are sometimes referred to in the literature as humic-like substances (HULIS). Two recent approaches for coupling capillary scale liquid chromatography and electron ionization mass spectrometry are reviewed and discussed. The first one, Cap-EI, is the latest evolution of the micro-scale particle beam interface, in which the nebulizer has been optimized to overcome the limitations of the former approach, in terms of sensitivity and linearity. It can be easily hosted in pre-existing instruments without major modifications and can use helium and the less-expensive nitrogen to generate library-matchable electron ionization spectra. The second one is a miniaturized interface for nano- and micro-HPLC, in which the interfacing process takes place into a suitably modified ion source. Because the eluate from the column is completely transferred into the ion source for ionization, superior sensitivity, linearity, and reproducibility are obtained. No signs of chemical ionization are observed at flow rates up to 1.5 μL/min. These two interfaces demonstrate that electron ionization can be successfully used for the analysis of small—medium molecules of various polarities, and also at the trace level. A new solid-phase extraction on-line device for micro-HPLC is presented. This device optimizes the injection of very dilute samples into a packed capillary column. It consists of two capillary, reversed-phase, HPLC columns of different length that can be linked together as a single chromatographic column. The first segment, only 2 cm long is connected to the HPLC injector. When disconnected from the longer column, several milliliters of an aqueous sample can be passed through at a high flow rate for fast trapping. On the basis of the retention mechanism, all suitable compounds are focused on the short column head in a sharp band. As soon as the chromatographic column is recomposed, the trapped analytes are eluted and separated at the optimal flow rate and gradient conditions. Due to the high preconcentration factor, trace-level analysis can be performed successfully. Different classes of analytes of various polarities and molecular weights can be determined, depending on the stationary phase and on the detector used. Some pesticides belonging to different classes were chosen to evaluate the performance of the device using an electron ionization mass spectrometer as HPLC detector. A fungicide in an irrigation canal water was determined at a concentration level of 4.5 microg x L(-1).

The crystal structures of the azamacrocycle 1,7–bis(2–hydroxy-5–nitrobenzyl)-4,10–dimethyl-1, 4,7,10–tetraazadodecane (L) (zwitterionic form) (L-0.5CH3OH-3.5H2O) and the monocharged derivative (HL+) (HL-Br-2C2H5OH) are reported. The crystals of L-0.5CH3 OH-3.5H2O are rhombohedral (hexagonal axes), space group R3, a = b = 42.472(7), c = 9.415(9) A, Z = 18, R = 0,034, whereas the crystals of HL-Br-2C2H5OH are monoclinic, space group C2/c, a = 20.418(5), b = 13.814 (4), c = 11.785(3) A, β = 99.00(2)°, Z = 4, R = 0.067. The differences in their conformation are discussed. Molecular mechanics calculations were carried out and indicate that the energy difference between the two species is very small. The stepwise enthalpies of protonation in solution were determined by direct microcalorimetry: ΔH° = −23.8(4) kJ mol−1 for H−1L− + H+ = L; ΔH°≅O for reactions L + H+ = HL+ and HL+ + H+ = H2L2+. The addition of the third proton is also athermic and fully entropy-controlled.

Endocrine disrupting compounds (EDCs) include organochlorine pesticides (OCPs), organophosphate pesticides (OPPs), carbamate pesticides, and plasticizers, such as bisphenol A (BPA). They persist in the environment because of their degradation resistance and bioaccumulate in the body tissues of humans and other mammals. Many studies are focused on the possible correlation between in utero exposure to EDCs and adverse health hazards in fetuses and newborns. In the last decade, environmental pollution has been considered a possible trigger for Sudden Infant Death Syndrome (SIDS) and Sudden Intrauterine Unexplained Death Syndrome (SIUDS), the most important death-causing syndromes in fetuses and newborns in developed countries. In this work, a rapid and sensitive analytical method was developed to determine the level of OCPs and OPPs, carbamates, and phenols in human fetal and newborn tissues (liver and brain) and to unveil the possible presence of non-targeted compounds. The target analytes where selected on the basis of their documented presence in the Trentino-Alto Adige region, an intensive agricultural area in northern Italy. A liquid-solid extraction procedure was applied on human and animal tissues and the extracts, after a solid phase extraction (SPE) clean-up procedure, were analyzed by gas chromatography coupled to a quadrupole mass spectrometric detector (GC-qMS). A GC-TOFMS (time-of-flight) instrument, because of its higher full-scan sensitivity, was used for a parallel detection of non-targeted compounds. Method validation included accuracy, precision, detection, and quantification limits (LODs; LOQs), and linearity response using swine liver and lamb brain spiked at different concentrations in the range of 0.4–8000.0 ng/g. The method gave good repeatability and extraction efficiency. Method LOQs ranged from 0.4–4.0 ng/g in the selected matrices. Good linearity was obtained over four orders of magnitude starting from LOQs. Isotopically labeled internal standards were used for quantitative calculations. The method was then successfully applied to the analysis of liver and brain tissues from SIUDS and SIDS victims coming from the above mentioned region.

We investigated the effect of temperature on the packing procedure of nano-LC columns (up to 50 cm) and on their performance. Several slurries of stationary phase were prepared using different solvent mixtures. Their stability was evaluated at several temperatures: 70°C, 50°C, and room temperature. At the higher temperature (70°C) the suspensions resulted to be stable for a longer time. For each slurry, we compared nano-LC columns packed with ultrasounds at 70°C and at room temperature. All the columns were tested with a standard mixture at 70°C, to reduce the solvent viscosity and the backpressure. Main chromatographic parameters such as the asymmetry factor, As, the reduced plate high, h, pattern in a Van Deemter plot, the total porosity, ε(t), and the permeability, k, were calculated and discussed. One of the nano-LC columns was used to separate a mixture of pesticides in a LC-MS system with an electron ionization LC-MS interface (Direct-EI). From our knowledge, this is the first study on the role of temperature in the efficiency of slurry-packing procedure.

An innovative approach for nano-liquid chromatography (LC) gradient generation is presented. This system represents an optimized and refined version of a prototype proposed by the authors a few years ago: the current version is characterized by a new configuration that guarantees complete automation and easier operation. The core of the system is an electronically controlled, multiposition valve that hosts six loops, filled with different mobile phase compositions of increasing strength. A conventional flow rate of water is reduced at nano-scale through a split device to push the content of the on-line loop into the column. No mixing occurs between solvents inside the loops, due to the low flow rate and the reduced loop diameter. Valve actuation allows the selection of the on-line loop to obtain the solvent gradient. The evaluation of the system performance takes into account gradient accuracy, precision, delay time, shape (linear, convex, or concave), and organic solvent consumption. Results highlight the reliability and the competitiveness of the system, especially in terms of accuracy and precision. A comparison between the described system and a conventional split-based one demonstrates that the new approach reduces the solvent consumption by about 40 times, improving green chromatography and cutting laboratory costs.

The generation of a micro-gradient for micro HPLC capillary columns is described. The system used is a modified and improved version of a split flow gradient elution device. Gradient tests have been performed and have shown good reproducibility under all conditions. The system has been successfully coupled with a particle beam LC-MS interface and the analysis of a mixture of twenty seven basic or neutral pesticides is reported.

One of the crucial tasks of pharmaceutical industry is to quantify the potential genotoxic impurities (PGIs) coming from the process of drug production. The European Medicines Agency (EMEA) imposes analytical testing limits in the order of μg/g, depending on drug dosage and exposure period, that means the need of a sensitive and selective method of analysis. Liquid chromatography coupled to electrospray ionization mass spectrometry (LC-ESI-MS) has been demonstrated as the most versatile approach to detect PGIs in complex matrices. However, time consuming derivatization processes are needed to enhance sensitivity and selectivity, and to overcome matrix effects (ME) that may arise from active pharmaceutical ingredients (APIs) or excipients. We propose the use of the Direct-EI LC-MS as an alternative approach to detect and quantify PGIs in drug formulations. The Direct-EI LC-MS interface is based on electron ionization (EI) which is well suited for the detection of low molecular weight compounds of different polarity, without derivatization and with no sign of ME. The method has been successfully applied to the detection of PGIs belonging to the class of alkylation agents. Calibration experiments show satisfactory linearity and precision data. Recoveries in low enriched samples spanned from 55 to 82%, and were not affected by ME. The method limits of detection (LODs), varying from 0.13 to 1.5 μg/g, were satisfactory for the quantitation of the target PGIs at the level required by regulatory agencies.

The purpose of this article is to underline the possibility of efficiently using electron ionization (EI) in liquid chromatography (LC) and mass spectrometry (MS). From a historical perspective, EI accompanied the first attempts in LC-MS but, owing to several technical shortcomings, it was soon outshined by soft, atmospheric pressure ionization (API) techniques. Nowadays, two modern approaches, supersonic molecular beam LC-MS and direct-EI LC-MS, offer a valid alterative to API, and preserve the advantages of EI also in LC-MS applications. These advantages can be summarized in three crucial aspects: automated library identification; identification of unknown compounds, owing to EI extensive fragment information; inertness to coeluted matrix interferences owing to very unlikely ion–ion and ion–molecule interactions in the EI gas-phase environment. The direct-EI LC-MS interface is a simple and efficient solution able to produce high-quality, interpretable EI spectra from a wide range of low molecular weight molecules of different polarity. Because of the low operative flow rates, this interface relies on a nano-LC technology that helps in reducing the impact of the mobile phase on the gas-phase environment of EI. This review provides an extensive discussion on the role of EI in LC-MS interfacing, and presents in detail several performance aspects of the direct-EI LC-MS interface, especially in terms of response, mass-spectral quality, and matrix effects. In addition, several key applications are also reported.

Matrix-dependent signal suppression or enhancement represents a major drawback in quantitative analysis with liquid chromatography coupled to atmospheric pressure ionization mass spectrometry (LC-API-MS). Because matrix effects (ME) might exert a detrimental impact on important method parameters (limit of detection, limit of quantification, linearity, accuracy, and precision), they have to be tested and evaluated during validation procedure. This review gives a detailed description on when these phenomena might be expected, and how they can be evaluated. The major sources of ME are discussed and illustrated with examples from bioanalytical, pharmaceutical, environmental, and food analysis. Because there is no universal solution for ME, the main strategies to overcome these phenomena are described in detail. Special emphasis is devoted to the sample-preparation procedures as well as to the recent improvements on chromatographic and mass spectrometric conditions. An overview of the main calibration techniques to compensate for ME is also presented. All these solutions can be used alone or in combination to retrieve the performance of the LC-MS for a particular matrix-analyte combination.

A major limitation in quantitative analysis with electrospray ionization mass spectrometry (ESI-MS) is represented by the so-called matrix effects in which the matrix coextracted with the analytes can alter the signal response, causing either suppression or enhancement, resulting in poor analytical accuracy, linearity, and reproducibility. In the direct electron ionization liquid chromatography-mass spectrometry (direct-EI LC-MS) interface the ionization process is based on electron impact ionization, and it occurs in the gas phase and is not influenced by coeluted matrix compounds. In this work we quantitatively evaluated matrix effects on enriched environmental and biological samples, with different extraction procedures, using ESI and direct-EI LC-MS. As expected, the samples analyzed with direct-EI were not affected by matrix composition, whereas with ESI we observed either signal suppression or enhancement, depending on the sample nature.
