It is based on the selection of microorganisms by sludge age, pH and temperature in order to set up a so-called partial nitrification, or nitrate shunt. Indeed, it is an aerobic nitritation directly followed by anaerobic denitrification. This innovative process is implemented in France on the Seine Grésillons wastewater plant (Yvelines, 1.2 million Equivalents.Habitants) operated by SIAAP. Placed in addition to the sludge treatment system, its role is to treat dehydration centers and digestion condensates of digested sludge from the site. Its interest is to avoid a too important supply of nitrogen on the water treatment line through the returns to the head; contributions that can represent 20% of the design load of the station. With the exception of bibliographic data, little technical and scientific knowledge is available on the operation of this process on an industrial scale. As part of the Mocopée research program, a fine follow-up of the process for more than two years has been carried out. The main objective of the project was to acquire a precise knowledge of the influence of the exploitation parameters on the composition of the microbial populations and thus on the treatment performances. After a literature review and a first measurement campaign in 2014, a set of recommendations were developed and implemented to ensure a proper functioning of the system. Following this preliminary step, successive modifications of the operating parameters (age of sludge, aeration, orthophosphate supply) were made according to an experimental protocol always identical: modification of an operating instruction followed by 3 weeks of stabilization of the reactor before implementation of a complete analytical campaign over 2 days (performances, kinetics, quantification of active nitrifying prokaryotic microorganisms - nitrating archaea or AOA, nitritating bacteria or AOB and nitrating bacteria or NOB). Three measurement campaigns were carried out in 2017. In general, the measured average NH4 concentrations appeared stable at entry (of the order of 875 mg N-NH4 / L) and variable at the outlet (from 30 to 215 mg N-NH4 / L). This variability in performance has been related to the operating modes of the process and the activity of the microorganisms. Thus, in addition to optimization of the operation (sizing of the boosters, feeding mode, reliability of continuous measurements) and the improvement of the analytical monitoring (filtration required, conservation of samples), the study made it possible to quantify precisely the performance of the process. Under appropriate conditions (T ° C above 40 ° C and sludge age of 3 to 4 days), the partial nitrification is in place and allows to obtain an N-NO2 / NOx concentration ratio greater than 95%. The results provided by this study are of prime importance for a reliable and efficient operation of the process on an industrial scale, making it possible to guarantee the respect of the statutory thresholds at the rejection of the plant.; Le procédé SHARON (Single reactor system for High Ammonia Removal Over Nitrite) est un système biologique de traitement des effluents fortement chargés en azote ammoniacal. Il est basé sur la sélection de micro-organismes par l'âge des boues, le pH et la température afin de mettre en place une nitrification dite partielle, ou shunt des nitrates. En effet, il s'agit d'une nitritation aérobie (NH4+NO2-) directement suivie d'une dénitrification anaérobie (NO2- N2). Ce procédé innovant est implanté en France sur l'usine d'épuration de Seine Grésillons (Yvelines, 1,2 millions d'Equivalents.Habitants) exploitée par le SIAAP. Placé en complément de la filière de traitement des boues, son rôle est de traiter les centrats de déshydratation et les condensats de séchage des boues digérées du site. Son intérêt est d'éviter un apport trop important d'azote sur la file de traitement des eaux par l'intermédiaire des retours en tête ; apports pouvant représenter 20% de la charge de dimensionnement de la station. A l'exception de données bibliographiques, peu de connaissances techniques et scientifiques sont disponibles sur le fonctionnement de ce procédé à l'échelle industrielle. Dans le cadre du programme de recherche Mocopée (MOdélisation, Contrôle et Optimisation des Procédés d'Epuration des Eaux), un suivi fin du procédé pendant plus de deux ans a été réalisé. L'objectif principal du projet était d'acquérir une connaissance précise de l'influence des paramètres d'exploitation sur la composition des populations microbiennes et donc, sur les performances de traitement. Après une étude bibliographique et une première campagne de mesures en 2014, un ensemble de préconisations ont été élaborées et mise en oeuvre afin d'assurer un fonctionnement adapté du système. Suite à cette étape préalable, des modifications successives des paramètres de fonctionnement (âge de boues, aération, apport d'orthophosphates) ont été apportées suivant un protocole expérimental toujours identique : modification d'une consigne d'exploitation suivie de 3 semaines de stabilisation du réacteur avant mise en oeuvre d'une campagne analytique complète sur 2 jours (performances, cinétiques, quantification des micro-organismes procaryotes nitrifiants actifs - les archées nitritantes ou AOA, les bactéries nitritantes ou AOB et les bactéries nitratantes ou NOB). Trois campagnes de mesures ont ainsi été réalisées en 2017. De manière générale, les concentrations moyennes en NH4 mesurées sont apparues stables en entrée (de l'ordre de 875 mg N-NH4/L) et variables en sortie (de 30 à 215 mg N-NH4/L). Cette variabilité des performances a été mise en relation avec les modes d'exploitation du procédé et l'activité des micro-organismes. Ainsi, outre l'optimisation de l'exploitation (dimensionnement des surpresseurs, mode d'alimentation, fiabilité des mesures en continues) et l'amélioration du suivi analytique (filtration nécessaire, conservation d'échantillons), l'étude a permis de chiffrer précisément les performances du procédé. Dans des conditions adaptées (T°C supérieure à 40°C et âge de boue de 3 à 4 jours), la nitrification partielle est bien en place et permet d'obtenir un ratio de concentrations N-NO2/NOx supérieur à 95 %. Les résultats apportés par cette étude sont de première importance pour une exploitation fiable et performante du procédé à l'échelle industrielle, permettant de garantir le respect des seuils réglementaires au rejet de l'usine.

Pour éviter cela, la station Seine Grésillons du SIAAP s'est dotée d'une étape de traitement des jus de digestion, avant retour en tête, par shunt des nitrates, le procédé SHARON®. Si le principe de shunt des nitrates est connu à l'échelle du laboratoire, sa mise en oeuvre à l'échelle industrielle reste très récente. C'est pourquoi les équipes d'IRSTEA, d'ECOBIO-Rennes et du SIAAP ont collaboré depuis 2014 dans le but de maîtriser l'exploitation du procédé et de proposer des voies d'optimisation de son fonctionnement. Ainsi, après un état des lieux de son fonctionnement, réalisé en 2014 et 2015, des paramètres d'exploitation ont été définis afin de favoriser le bon fonctionnement de l'unité de traitement. Après plus d'une année de fonctionnement et certaines améliorations de son exploitation (choix de matériel, suivi analytique), des voies d'optimisations ont été testées durant l'année 2017. Trois campagnes de mesures ont été réalisées sur le procédé, en période stabilisée, afin de tester des modifications de consignes d'exploitation, visant une optimisation de fonctionnement. Durant ces suivis, différentes approches (bilan 24H, prélèvements instantanés, vitesses en batch, microbiologique moléculaire) ont montré l'existence d'une nitrification partielle importante et constante sur le site mais variable d'une période à l'autre. Bien qu'aucune corrélation n'ait été observée entre les performances et les paramètres d'exploitation, il apparait que piloter le SHARON® avec une température supérieure à 35°C permettent d'assurer une nitrification partielle importante si la charge d'alimentation en N-NH4 reste stable et proche du nominal et si l'âge de boue total est maintenu de l'ordre de 3,35 jours.

Wolbachia are intracellular Gram-negative bacteria that are reproductive manipulators of many arthropods. In the isopod Armadillidium vulgare, the Wolbachia wVulC strain induces male feminization. Here, we characterized two vir operons which are expressed in all host tissues and which encode a type IV secretion system (T4SS) used to translocate bacterial effectors into host cytoplasm. Gene organization and sequence comparison in 37 Wolbachia strains highlighted the high conservation of both vir operons and their importance for the biology of the bacteria. We also identified in the on-going assembly of the wVulC genome, 66 ankyrin domain-encoding genes. Ankyrin motifs are known to mediate protein-protein interactions in eukaryotic organisms and thus are suggested to mediate in Wolbachia the interaction with host molecules. We showed that one of the three copies of the wVulC pk2 gene is only expressed in feminizing strains but not in the three strains inducing cytoplasmic incompatibility in terrestrial isopods. The associated Pk2 protein could be involved in male feminization. We thus tested the interaction between three T4SS proteins and five ankyrins (including Pk2) via the yeast two-hybrid and CRAfT (Cre-recombinase Reporter Assay for Translocation) methods. None of the five ankyrin proteins were revealed to be secreted by the wVulC strain. Nevertheless, this promising approach may enable us to identify Wolbachia effectors.; Wolbachia est une bactérie Gram(-) intracellulaire modifiant la reproduction de nombreux arthropodes. Background: In pathogens, certain genes encoding proteins that directly interact with host defences coevolve with their host and are subject to positive selection. In the lepidopteran host-wasp parasitoid system, one of the most original strategies developed by the wasps to defeat host defences is the injection of a symbiotic polydnavirus at the same time as the wasp eggs. The virus is essential for wasp parasitism success since viral gene expression alters the immune system and development of the host. As a wasp mutualist symbiont, the virus is expected to exhibit a reduction in genome complexity and evolve under wasp phyletic constraints. However, as a lepidopteran host pathogenic symbiont, the virus is likely undergoing strong selective pressures for the acquisition of new functions by gene acquisition or duplication. To understand the constraints imposed by this particular system on virus evolution, we studied a polydnavirus gene family encoding cyteine protease inhibitors of the cystatin superfamily. Results: We show that cystatins are the first bracovirus genes proven to be subject to strong positive selection within a host-parasitoid system. A generated three-dimensional model of Cotesia congregata bracovirus cystatin 1 provides a powerful framework to position positively selected residues and reveal that they are concentrated in the vicinity of actives sites which interact with cysteine proteases directly. In addition, phylogenetic analyses reveal two different cystatin forms which evolved under different selective constraints and are characterized by independent adaptive duplication events. Conclusion: Positive selection acts to maintain cystatin gene duplications and induces directional divergence presumably to ensure the presence of efficient and adapted cystatin forms. Directional selection has acted on key cystatin active sites, suggesting that cystatins coevolve with their host target. We can strongly suggest that cystatins constitute major virulence factors, as was already proposed in previous functional studies.
