Abstract

Radiation reaction describes the effective force experienced by an accelerated charge due to radiation emission. Quantum effects dominate charge dynamics and radiation production[1][2] for charges accelerated by fields with strengths approaching the Schwinger field, $\mathbf{E_{sch}=}$\textbf{\SI[detect-weight]{1.3e18}{\volt\per\metre}[3]. Such fields exist in extreme astrophysical environments such as pulsar magnetospheres[4], may be accessed by high-power laser systems[5-7], dense particle beams interacting with plasma[8], crystals[9], and at the interaction point of next generation particle colliders[10]. Classical radiation reaction theories do not limit the frequency of radiation emitted by accelerating charges and omit stochastic effects inherent in photon emission[11], thus demanding a quantum treatment. Two quantum radiation reaction models, the quantum-continuous[12] and quantum-stochastic[13] models, correct the former issue, while only the quantum-stochastic model incorporates stochasticity[12]. Such models are of fundamental importance, providing insight into the effect of the electron self-force on its dynamics in electromagnetic fields. The difficulty of accessing conditions where quantum effects dominate inhibited previous efforts to observe quantum radiation reaction in charged particle dynamics with high significance. We report the first direct, high significance $(>5{\sigma})$ observation of strong-field radiation reaction on charged particles. Furthermore, we obtain strong evidence favouring the quantum radiation reaction models, which perform equivalently, over the classical model. Robust model comparison was facilitated by a novel Bayesian framework which inferred collision parameters. This framework has widespread utility for experiments where parameters governing lepton-laser collisions cannot be directly measured, including those using conventional accelerators., Comment: 31 pages, 14 figures

However, poor bioavailability and selectivity might limit their clinical application. Therefore, bioavailability is considered a notable challenge to improve bio-efficacy in transporting dietary phytochemicals. Different methods have been proposed for generating effective carrier systems to enhance the bioavailability of phytochemicals. Among them, nano-vesicles have been introduced as promising candidates for the delivery of insoluble phytochemicals. Due to the easy preparation of the bilayer vesicles and their adaptability, they have been widely used and approved by the scientific literature. The first part of the review is focused on introducing phytosome technology as well as its applications, with emphasis on principles of formulations and characterization. The second part provides a wide overview of biological activities of commercial and non-commercial phytosomes, divided by systems and related pathologies. These results confirm the greater effectiveness of phytosomes, both in terms of biological activity or reduced dosage, highlighting curcumin and silymarin as the most formulated compounds. Finally, we describe the promising clinical and experimental findings regarding the applications of phytosomes. The conclusion of this study encourages the researchers to transfer their knowledge from laboratories to market, for a further development of these products.Keywords: phytochemical, nanomedicine, phytosome, delivery, vesicle, disease

Abstract

The overexpression of the ecotropic viral integration site-1 gene (EVI1/MECOM) marks the most lethal acute myeloid leukemia (AML) subgroup carrying chromosome 3q26 abnormalities. By taking advantage of the intersectionality of high-throughput cell-based and gene expression screens selective and pan-histone deacetylase inhibitors (HDACis) emerge as potent repressors of EVI1. To understand the mechanism driving on-target anti-leukemia activity of this compound class, here we dissect the expression dynamics of the bone marrow leukemia cells of patients treated with HDACi and reconstitute the EVI1 chromatin-associated co-transcriptional complex merging on the role of proliferation-associated 2G4 (PA2G4) protein. PA2G4 overexpression rescues AML cells from the inhibitory effects of HDACis, while genetic and small molecule inhibition of PA2G4 abrogates EVI1 in 3q26 AML cells, including in patient-derived leukemia xenografts. This study positions PA2G4 at the crosstalk of the EVI1 leukemogenic signal for developing new therapeutics and urges the use of HDACis-based combination therapies in patients with 3q26 AML.

La questione che il volume di Giancarlo Gaeta (In attesa del Regno. Il Cristianesimo alla svolta dei tempi, Quodlibet, 2022) pone esplicitamente nella postilla finale, ma che in realtà attraversa – seppur in modi diversi – tutti i saggi in esso raccolti, è al tempo stesso provocatoria e cruciale: cosa resta oggi, a duemila anni dalla sua comparsa, del cristianesimo? Nelle pagine che seguono, proveremo a rispondere indirettamente a questa domanda, partendo da alcune brevi riflessioni che riguarderanno un’altra questione – quella riguardante la crisi del cristianesimo – che in un certo senso precede quella del “cosa resta” e che ad essa inevitabilmen te si connette. Più precisamente, cercheremo di incrociare il tema della crisi con il tema della natura del cristianesimo, perché mi pare essi siano in qualche modo indissolubilmente legati, come proverò a mostrare.

Abstract

In this paper we propose a reflection on the intertwining of mind, body, and words in ancient Greek thought, with specific reference to Gorgias and Aristotle. In particular, we believe that it was the reflection developed within rhetoric – though not the only one – that played a central role in the first articulation of the conceptual framework where the relationship between soul and body, in a very peculiar and specific way, takes place. There are at least two salient features of this rhetorical perspective, which will be investigated here with specific reference, as mentioned, to Gorgias and Aristotle: the insistence on the close interrelationship between body and soul, an interrelationship highlighted starting from the emotional sphere in which it shows itself in the most explicit manner and never resolved in terms of a pure and simple subordination of one to the other; the role that logos, by virtue of its nature and characteristics, plays as point of intersection of somatic and psychic dimension of human experience.

Abstract

Arrhythmogenic cardiomyopathy (ACM) is a genetic disease associated with sudden cardiac death and cardiac fibro-fatty replacement. Over the last years, several works have demonstrated that different epigenetic enzymes can affect not only gene expression changes in cardiac diseases but also cellular metabolism. Specifically, the histone acetyltransferase GCN5 is known to facilitate adipogenesis and modulate cardiac metabolism in heart failure. Our group previously demonstrated that human primary cardiac stromal cells (CStCs) contribute to adipogenesis in the ACM pathology. Thus, this study aims to evaluate the role of GCN5 in ACM intracellular lipid accumulation. To do so, CStCs were obtained from right ventricle biopsies of ACM patients and from samples of healthy cadaveric donors (CTR). GCN5 expression was increased both in ex vivo and in vitro ACM samples compared to CTR. When GCN5 expression was silenced or pharmacologically inhibited by the administration of MB-3, we observed a reduction in lipid accumulation and a mitigation of reactive oxygen species (ROS) production in ACM CStCs. In agreement, transcriptome analysis revealed that the presence of MB-3 modified the expression of pathways related to cellular redox balance. Altogether, our findings suggest that GCN5 inhibition reduces fat accumulation in ACM CStCs, partially by modulating intracellular redox balance pathways.

The question that lies at the heart of this short essay concerns the effects of laughter on the relationship between the right to freedom of expression, on the one hand, and the potential exercise of verbal violence on the other. In short, the question we ask is whether the presence of laughter increases the spectrum of freedom of expression by reducing that of verbal violence, or, on the contrary, whether the very element of laughter does not laughter does not, on the contrary, determine a multiplication of violence in certain discursive situations.

Abstract

A partire da confronto tra alcuni passi dei Vangeli e testi di Wittgenstein e Kuhn, il saggio ragiona sulla su alcune questioni teoriche che ruotano attorno al tema della credenza e del cambiamento di credenza, a partire da una nozione molto densa del lessico greco antico, quella di pistis, che tra i suoi significati ha anche quelli di fede e fiducia. In particolare, si cerca di fare emergere come la dicotomia classica tra due tipi di pistis (da intendere qui genericamente come “credenza”), una pistis filosofico-scientifica razionale e una pistis religiosa irrazionale – distinzione che rimanda all’ancor più classica dicotomia “razionale” vs “irrazionale” – sia non soltanto difficile da sostenere, ma anche difficile da identificare.

Abstract

Background There is mounting evidence on the existence of a Pediatric Inflammatory Multisystem Syndrome-temporally associated to SARS-CoV-2 infection (PIMS-TS), sharing similarities with Kawasaki Disease (KD). The main outcome of the study were to better characterize the clinical features and the treatment response of PIMS-TS and to explore its relationship with KD determining whether KD and PIMS are two distinct entities. Methods The Rheumatology Study Group of the Italian Pediatric Society launched a survey to enroll patients diagnosed with KD (Kawasaki Disease Group – KDG) or KD-like (Kawacovid Group - KCG) disease between February 1st 2020, and May 31st 2020. Demographic, clinical, laboratory data, treatment information, and patients’ outcome were collected in an online anonymized database (RedCAP®). Relationship between clinical presentation and SARS-CoV-2 infection was also taken into account. Moreover, clinical characteristics of KDG during SARS-CoV-2 epidemic (KDG-CoV2) were compared to Kawasaki Disease patients (KDG-Historical) seen in three different Italian tertiary pediatric hospitals (Institute for Maternal and Child Health, IRCCS “Burlo Garofolo”, Trieste; AOU Meyer, Florence; IRCCS Istituto Giannina Gaslini, Genoa) from January 1st 2000 to December 31st 2019. Chi square test or exact Fisher test and non-parametric Wilcoxon Mann-Whitney test were used to study differences between two groups. Results One-hundred-forty-nine cases were enrolled, (96 KDG and 53 KCG). KCG children were significantly older and presented more frequently from gastrointestinal and respiratory involvement. Cardiac involvement was more common in KCG, with 60,4% of patients with myocarditis. 37,8% of patients among KCG presented hypotension/non-cardiogenic shock. Coronary artery abnormalities (CAA) were more common in the KDG. The risk of ICU admission were higher in KCG. Lymphopenia, higher CRP levels, elevated ferritin and troponin-T characterized KCG. KDG received more frequently immunoglobulins (IVIG) and acetylsalicylic acid (ASA) (81,3% vs 66%; p = 0.04 and 71,9% vs 43,4%; p = 0.001 respectively) as KCG more often received glucocorticoids (56,6% vs 14,6%; p Conclusion Our study suggests that SARS-CoV-2 infection might determine two distinct inflammatory diseases in children: KD and PIMS-TS. Older age at onset and clinical peculiarities like the occurrence of myocarditis characterize this multi-inflammatory syndrome. Our patients had an optimal response to treatments and a good outcome, with few complications and no deaths.

Abstract

Using the Cap Analysis of Gene Expression (CAGE) technology, the FANTOM5 consortium provided one of the most comprehensive maps of transcription start sites (TSSs) in several species. Strikingly, ~72% of them could not be assigned to a specific gene and initiate at unconventional regions, outside promoters or enhancers. Here, we probe these unassigned TSSs and show that, in all species studied, a significant fraction of CAGE peaks initiate at microsatellites, also called short tandem repeats (STRs). To confirm this transcription, we develop Cap Trap RNA-seq, a technology which combines cap trapping and long read MinION sequencing. We train sequence-based deep learning models able to predict CAGE signal at STRs with high accuracy. These models unveil the importance of STR surrounding sequences not only to distinguish STR classes, but also to predict the level of transcription initiation. Importantly, genetic variants linked to human diseases are preferentially found at STRs with high transcription initiation level, supporting the biological and clinical relevance of transcription initiation at STRs. Abstract

It is well known that physical and surface properties of nanomaterials are promising to enhance efficiency of nanostructured devices for sensing and for sustainable energy production, conversion, and storage. However, the practical use of nanomaterials is often complicated by the lack of scalable and cost-efficient synthesis procedures and the challenge of integrating into devices 1D nanomaterials saving their structural features. In this field, one of the most severe challenges is to find suitable methods for fabricating nanomaterials. Over the years, numerous preparation methods were proposed in the literature, but not all of them are easily scalable and economically advantageous for industrial application. In this context, electrochemical deposition in template is a facile method for fabricating either two- or one-dimensional nanostructured materials because it allows to easily adjusting the fundamental parameters controlling their final features. Electrochemical processes are, usually, cheap and environmental friendly, and they can be easily scaled-up from lab to industrial level. In this chapter, we will describe different electrochemical methods, electrodeposition, galvanic deposition electroless deposition and electrogeneration of base, that permit to obtain different type of nanomaterials such as metals, oxide, and semiconductors. In addition, also the performances of different nanostructured materials are presented.

Un uso che ha a che fare con la questione dei principi primi, con il tema dell'adeguatezza epistemologica e con la relazione conflittuale e problematica con la figura del recalcitrante.

Abstract

Background: Ochratoxin A is a nephrotoxin which may occur in wines characterised by higher pH than the average. In the last decades the mechanisms responsible for ochratoxin A reduction by lactic acid bacteria have been investigated and identified as mainly cell walls adsorption and / or enzymatic conversion to ochratoxin-α, a non-toxic metabolite. Since lactic acid bacteria are involved in the malolactic fermentation during the wine-making process, selected starter cultures could be exploited to guarantee safe ochratoxin A level in wines also from contaminated grapes. A lactic acid bacteria strain (Lactobacillus plantarum V22) was previously selected for its ability of both degrading ochratoxin A and carrying out malolactic fermentation at high pH. Objective: This study was aimed at assessing if the selected L. plantarum strain, can reduce ochratoxin A because it can use it as a carbon source. Methods: L. plantarum V22 was grown in the presence of ochratoxin A in two different synthetic substrates, with or without malic acid, monitoring the reduction of ochratoxin A and the presence of ochratoxin α as an indicator for a toxin enzymatic hydrolysis. The presence of residual not hydrolysed ochratoxin A bound to the bacteria cell walls was also evaluated to quantify the ochratoxin A removal due to simple adsorption. Result: A significant reduction of 19.5 ± 2.0% in ochratoxin A concentration was observed only in the presence of malic acid. The quantified fraction of ochratoxin A adsorbed on cell walls was irrelevant and the metabolite ochratoxin α could not be detected. Conclusion: There is a possibility that L. plantarum V22 can degrade ochratoxin A through a not yet identified metabolic pathway.
