Einige der 500 bekannten Sacoglossa-Arten können in ihrem eigenen Cytosol Chloroplasten photosynthetisch aktiv halten. Dieses als funktionale Kleptoplastie bezeichnete Phänomen ist eine spezielle Form der Photosymbiose und kommt in Tieren nur in Sacoglossa vor. Viele Fragen in Hinblick auf die benötigten Adaptationen der Schnecken und der Algen, um funktionale Kleptoplastie zu ermöglichen, sind entweder nur unzureichend oder noch nicht beantwortet. Besonders kontrovers wird der Nutzen der „gestohlenen Plastiden“ (Kleptoplasten) für die Schnecken diskutiert und wie die Schnecken die photosynthetisch fxierten Nährstoffe erhalten können. Hier könnte intensivere Forschung an Elysia viridis helfen, die auch plastidenfrei untersucht werden kann. Neben vielfältigen Forschungsfragen kann das Phänomen der funktionalen Kleptoplastie aber auch in der Schule, z. B. im Rahmen der Endosymbiose behandelt werden, um so bereits Schüler/innen mit diesen faszinierenden Schnecken in Kontakt kommen zu lassen und die Faszination für die Biologie zu steigern., Some of the 500 known Sacoglossa species can keep chloroplasts photosynthetically active in their own cytosol. This phenomenon, called functional kleptoplasty, is a special form of photosymbiosis and is only found in Sacoglossa among animals. With regard to the necessary adaptations to enable functional kleptoplasty in these slugs many questions are either insuffciently answered or not at all. Especially the beneft of the “stolen plastids” (kleptoplasts) for the slugs is discussed controversially and how they can obtain the photosynthetically fxed nutrients. More intensive research on Elysia viridis, which can also be examined free of plastids, could help to answer some of these questions. In addition to various research questions, the phenomenon of functional kleptoplasty can also be implemented at school, e. g. in the context of endosymbiosis, in order to show these fascinating slugs to schoolchildren and increase their fascination for biology.

Phytoplankton is the group of photosynthetic microorganisms (microalgae and cyanobacteria) living in suspension in marine and fresh waters. Through photosynthesis, phytoplankton produce large amounts of the oxygen essential for marine and terrestrial life. Marine microalgae are also promising organisms for biotechnological applications (human and animal food, biofuels). Because of their ecological and economic importance, the study of the phytoplankton responses to environmental challenged (including the ones induced by human activity and global warming) is a developing field of research. Phytoplankton activity is influenced by changes in the vertical stratification of the water column, which modulate light energy availability as well as nutrient supply to phytoplankton cells in a temperature-dependent manner. Based on light and nutrient availability, phytoplankton cells have evolved different lifestyles: autotrophy (photosynthetic activity), mixotrophy (simultaneous use of photosynthesis and respiration of exogenous carbon sources for growth) and photosymbiosis (symbiotic interactions with animal cells).In this thesis, I have studied the physiological responses of phytoplankton cells to environmental changes at the cellular and subcellular levels. To achieve this goal, I have developed a complete imaging workflow to perform quantitative morphometric analyses of entire algal cells, representatives of ecologically-successful and laboratory-model microalgal species. The protocol starts with FIB-SEM (Focused Ion Beam - Scanning Electron Microscopy) or SBF-SEM (Serial Block Facing – Scanning Electron Microscopy), to acquire high-resolution images. By implementing the 3D image analysis protocol, it is possible to obtained high-resolution whole cells models in three dimensions, suitable to perform quantitative analyses. Thanks to these tools, I have been able to image the adaptation of phytoplankton to various environmental conditions: i. changes in the size and morphology of plastids and mitochondria during light acclimation in diatoms, ii. changes in organelles interaction during nutrient acclimation in Nannochloropsis, iii. morphological changes occurring during photosymbiosis in Phaeocystis.Overall, this work reveals several scenarios of phytoplankton acclimation at both the cellular and subcellular levels. I have also validated the use of this protocol in plants in a study on chloroplast biogenesis during de-etiolation in Arabidopsis. of plastids., Le phytoplancton est le groupe de micro-organismes photosynthétiques (microalgues et cyanobactéries) vivant en suspension dans les eaux marines et douces. Grâce à la photosynthèse, le phytoplancton produit de grandes quantités d'oxygène indispensable à la vie marine et terrestre. Les microalgues marines sont également des organismes prometteurs pour les applications biotechnologiques (alimentation humaine et animale, biocarburants). En raison de leur importance écologique et économique, l'étude des réponses du phytoplancton aux défis environnementaux (y compris ceux induits par l'activité humaine et le réchauffement climatique) est un domaine de recherche en plein développement. L'activité du phytoplancton est influencée par les changements dans la stratification verticale de la colonne d'eau qui module, en fonction de la température, la disponibilité de l'énergie lumineuse ainsi que l'apport de nutriments aux cellules du phytoplancton. En raison de la disponibilité de la lumière et des nutriments, les cellules du phytoplancton ont évolué vers différents modes de vie : autotrophie (activité photosynthétique), mixotrophie (utilisation simultanée de la photosynthèse et de l’utilisation ? de sources de carbone extérieures pour la croissance) et photosymbiose (interactions symbiotiques avec des cellules animales).Dans cette thèse, j'ai étudié les réponses physiologiques des cellules du phytoplancton aux changements environnementaux au niveau cellulaire et sous-cellulaire. Pour atteindre cet objectif, j'ai mis au point un processus d'imagerie complet permettant d'effectuer des analyses morphométriques quantitatives de cellules entières d'algues représentatives à la fois d'espèces à succès écologique et de modèles de laboratoire. Le protocole commence avec l’acquisition de séries d’images hautes résolutions soit par FIB-SEM (Focused Ion Beam - Scanning Electron Microscopy) ou SBF-SEM (Serial Block Facing – Scanning Electron Microscopy). Le protocole d'analyse d'images 3D développé dans ce travail permet d'obtenir des modèles tridimensionnels à haute résolution de cellules entières permettant la réalisation d'analyses quantitatives. Grâce à ces outils, j'ai pu imager l'adaptation du phytoplancton à diverses conditions environnementales révélant ainsi : 1) le changement de taille et de morphologie des plastes et des mitochondries lors de l'acclimatation à la lumière dans les diatomées, 2) le changement dans l'interaction des organites chez Nannochloropsis lors de l'acclimatation aux nutriments, 3) les changements morphologiques qui surviennent lors de la photosymbiose dans l’algue Phaeocystis.Ces travaux révèlent plusieurs scénarios d'acclimatation du phytoplancton au niveau cellulaire et subcellulaire. J'ai également pu valider l'utilisation de ce protocole chez les plantes dans une étude sur la biogenèse des chloroplastes lors de la dé-étiolation des cotylédons d’Arabidopsis.

Whereas most jellyfishes are strictly heterotrophic organisms, some of them undergo a photosymbiosis with autotrophic Dinophyceae (“zooxanthellae”). These zooxanthellate jellyfishes, as holobionts, are mixotrophic deriving nutrition from both predation and photosynthesis. However, the relative importance of autotrophic and heterotrophic nutrition can vary as a function of ontogeny, phylogeny and ecology. Such variations of nutrition have important consequences for the population dynamics of these organisms. It is therefore central to characterize the variability and the plasticity of the nutrition of zooxanthellate jellyfishes to understand their ecology. In this thesis, the nutrition of zooxanthellate jellyfishes was investigated using laboratory experimental systems and field studies. A first experiment allowed to confirm previous findings that autotrophic nutrition is of small importance for the polyp of zooxanthellate jellyfishes. A second experiment assessed how elemental and isotopic compositions of zooxanthellate jellyfishes could be used to study their nutrition. The findings of this experiment are then confronted with results from the field: The nutrition of zooxanthellate Mastigias papua medusae was studied in their natural environments (Palau) through the use of isotopic, elemental but also fatty acids compositions. These field results demonstrate the wide plasticity of the nutrition of Mastigias papua ranging from pure heterotrophy to dominant autotrophy.The existence of such a wide plasticity in the nutrition of zooxanthellate jellyfishes helps to understand some crucial aspect of their ecology such as their generally low ability to bloom relative to non-zooxanthellate jellyfishes, or their reactions to temperature-induced bleaching., Alors que la majorité des méduses sont des hétérotrophes strictes, certaines vivent en photosymbiose avec des Dinophyceae autotrophes (« zooxanthelles »). Ces méduses à zooxanthelles, en tant qu’holobiontes, sont mixotrophes, dérivant leur nutrition à la fois de la prédation et de la photosynthèse. Toutefois, l’importance relative de l’autotrophie et de l’hétérotrophie dans la nutrition peuvent varier en fonction de l’ontogénie, de la phylogénie, ou de l’écologie. De telles variations ont d’importantes conséquences pour la dynamique des populations de ces organismes. Il est donc important de pouvoir caractériser la variabilité et la plasticité de la nutrition des méduses à zooxanthelles pour comprendre leur écologie. Au cours de cette thèse, la nutrition des méduses à zooxanthelles a été étudiée par le biais d’expériences de laboratoire et d’observations de terrain. Une première expérience a permis de confirmer des résultats précédents suggérant que la nutrition autotrophe est de faible importance pour les polypes des méduses à zooxanthelles. Une seconde expérience a mis en évidence comment les compositions isotopiques et élémentaires des méduses à zooxanthelles peuvent être utilisées pour étudier leur nutrition. Ces résultats sont ensuite confrontés aux observations de terrain : la nutrition de la méduse à zooxanthelles Mastigias papua a été étudiée dans son environnement naturel (Palaos) via l’étude de leur compositions isotopiques, élémentaires, mais aussi en acides gras. Ces résultats de terrain démontrent l’importante plasticité de la nutrition de Mastigias papua, pouvant aller de la pure hétérotrophie, une autotrophie dominante. L’existence d’une telle plasticité dans la nutrition des méduses à zooxanthelles aide à comprendre certains aspects centraux de leur écologie, tels que leur tendance à former moins de blooms que les méduses sans zooxanthelles, ou leurs réactions aux évènements de blanchissement induit par la température.
