1. Impact of oil on bacterial community structure in bioturbated sediments.

Abstract: Oil spills threaten coastlines where biological processes supply essential ecosystem services. Therefore, it is crucial to understand how oil influences the microbial communities in sediments that play key roles in ecosystem functioning. Ecosystems such as sediments are characterized by intensive bioturbation due to burrowing macrofauna that may modify the microbial metabolisms. It is thus essential to consider the bioturbation when determining the impact of oil on microbial communities. In this study, an experimental laboratory device maintaining pristine collected mudflat sediments in microcosms closer to true environmental conditions--with tidal cycles and natural seawater--was used to simulate an oil spill under bioturbation conditions. Different conditions were applied to the microcosms including an addition of: standardized oil (Blend Arabian Light crude oil, 25.6 mg.g⁻¹ wet sediment), the common burrowing organism Hediste (Nereis) diversicolor and both the oil and H. diversicolor. The addition of H. diversicolor and its associated bioturbation did not affect the removal of petroleum hydrocarbons. After 270 days, 60% of hydrocarbons had been removed in all microcosms irrespective of the H. diversicolor addition. However, 16S-rRNA gene and 16S-cDNA T-RFLP and RT-PCR-amplicon libraries analysis showed an effect of the condition on the bacterial community structure, composition, and dynamics, supported by PerMANOVA analysis. The 16S-cDNA libraries from microcosms where H. diversicolor was added (oiled and un-oiled) showed a marked dominance of sequences related to Gammaproteobacteria. However, in the oiled-library sequences associated to Deltaproteobacteria and Bacteroidetes were also highly represented. The 16S-cDNA libraries from oiled-microcosms (with and without H. diversicolor addition) revealed two distinct microbial communities characterized by different phylotypes associated to known hydrocarbonoclastic bacteria and dominated by Gammaproteobacteria and Deltaproteobacteria. In the oiled-microcosms, the addition of H. diversicolor reduced the phylotype-richness, sequences associated to Actinobacteria, Firmicutes and Plantomycetes were not detected. These observations highlight the influence of the bioturbation on the bacterial community structure without affecting the biodegradation capacities.

2. Effects of hydrostatic pressure on growth and luminescence of a moderately-piezophilic luminous bacteria Photobacterium phosphoreum ANT-2200.

Abstract: Bacterial bioluminescence is commonly found in the deep sea and depends on environmental conditions. Photobacterium phosphoreum ANT-2200 has been isolated from the NW Mediterranean Sea at 2200-m depth (in situ temperature of 13°C) close to the ANTARES neutrino telescope. The effects of hydrostatic pressure on its growth and luminescence have been investigated under controlled laboratory conditions, using a specifically developed high-pressure bioluminescence system. The growth rate and the maximum population density of the strain were determined at different temperatures (from 4 to 37°C) and pressures (from 0.1 to 40 MPa), using the logistic model to define these two growth parameters. Indeed, using the growth rate only, no optimal temperature and pressure could be determined. However, when both growth rate and maximum population density were jointly taken into account, a cross coefficient was calculated. By this way, the optimum growth conditions for P. phosphoreum ANT-2200 were found to be 30°C and, 10 MPa defining this strain as mesophile and moderately piezophile. Moreover, the ratio of unsaturated vs. saturated cellular fatty acids was found higher at 22 MPa, in agreement with previously described piezophile strains. P. phosphoreum ANT-2200 also appeared to respond to high pressure by forming cell aggregates. Its maximum population density was 1.2 times higher, with a similar growth rate, than at 0.1 MPa. Strain ANT-2200 grown at 22 MPa produced 3 times more bioluminescence. The proposed approach, mimicking, as close as possible, the in situ conditions, could help studying deep-sea bacterial bioluminescence and validating hypotheses concerning its role into the carbon cycle in the deep ocean.

Abstract: International audience; Caleta Valdés (CV) is a coastal lagoon of the Patagonian Atlantic coast located in the Península de Valdés declared as Humanity Mundial Patrimony due to its remarkable biodiversity, ecosystemic richness, and pristine state. Marine mammal populations are well documented in this area but few studies have been carried out on the local macrobenthic communities. The goals of this study were (a) to evaluate for the first time the seasonal variation of the structure and activity (i.e., sediment reworking) of the intertidal macrobenthic communities of CV, and (b) to validate an ex situ experimental protocol for future ecotoxicological studies. To do so, sedimentary (granulometry, water content, and organic matter) and biological (macrobenthic assemblages, biodiffusive-like and advective sediment reworking components) parameters were analyzed using a combination of in situ and ex situ measurements. Overall, polychaete and crustacean dominated the macrobenthic community. The highest abundances were found in spring–summer along with a predominance of biodiffusors versus conveyors. Ex situ and in situ measurements demonstrated similar results, thus allowing validation of an ex situ experimental procedure for macrobenthic community and functioning studies. In addition, these results provide a first baseline of benthic information on CV that will be helpful to monitor the effects of potential pollution in Patagonian coastal systems.

Abstract: International audience; This article examines how the populations of French Guiana have, since the colonization period until today, inhabited the coast, in particular the cheniers close to the coast, whereas these spaces are subject to intense changes - colonization and destruction of the mangrove ecosystem, erosion, modification of the estuaries - which have significant consequences for living conditions and access to natural resources. The interdisciplinary approach brought together historians, geographers, geomorphologists, ecologists and anthropologists. Three contrasted areas were studied: Awala-Yalimapo, a Kali'na Amerindian village located at the Maroni estuary, the savannas region, between Sinnamary and Iracoubo where the Creole peasantry flourished, and finally, the particular case of the city of Kourou, built from the 1960s by the sea to house the engineers and technicians of the Guiana Space Center. This paper aims to propose a reflection on mobility and adaptability of the traditional habitat of the Amerindian and Creole populations, based on the collective appropriation of the land, and what could be described as a light approach to development. On the contrary, Kourou was built with a relationship to the sea largely imported from metropolitan France at a time when private property became the norm in French Guiana. The city is now facing serious erosion problems. As we question how to inhabit the coastline at a time when global change is likely to bring rapid transformations, knowing this history can be a valuable source of reflection for future coastal management strategies.

Abstract: International audience; The microbial communities inhabiting the Atlantic-East Pacific (AEP) mangroves have been poorly studied, and mostly comprise chronically polluted mangroves. In this study, we characterized changes in the structure and diversity of microbial communities of mangroves along the urban-to-rural gradient of the Cayenne estuary (French Guiana, South America) that experience low human impact. The microbial communities were assigned into 50 phyla. Proteobacteria, Chloroflexi, Acidobacteria, Bacteroidetes, and Planctomycetes were the most abundant taxa. The environmental determinants found to significantly correlated to the microbial communities at these mangroves were granulometry, dieldrin concentration, pH, and total carbon (TC) content. Furthermore, a precise analysis of the sediment highlights the existence of three types of anthropogenic pressure among the stations: (i) organic matter (OM) enrichment due to the proximity to the city and its wastewater treatment plant, (ii) dieldrin contamination, and (iii) naphthalene contamination. These forms of weak anthropogenic pressure seemed to impact the bacterial population size and microbial assemblages. A decrease in Bathyarchaeota, “Candidatus Nitrosopumilus”, and Nitrospira genera was observed in mangroves subjected to OM enrichment. Mangroves polluted with organic contaminants were enriched in Desulfobacteraceae, Desulfarculaceae, and Acanthopleuribacteraceae (with dieldrin or polychlorobiphenyl contamination), and Chitinophagaceae and Geobacteraceae (with naphthalene contamination). These findings provide insights into the main environmental factors shaping microbial communities of mangroves in the AEP that experience low human impact and allow for the identification of several potential microbial bioindicators of weak anthropogenic pressure.

Abstract: Particle mixing and irrigation of the seabed by benthic fauna (bioturbation) have major impacts on ecosystem functions such as remineralization of organic matter and sediment-water exchange. As a tribute to Prof. Gaston Desrosiers by the Nereis Park association, eighteen laboratories carried out a collaborative experiment to acquire a global snapshot of particle reworking by the polychaete Hediste diversicolor at 16 sites surrounding the Northern Atlantic. Organisms and soft sediments were collected during May – July at different geographical locations and, using a common laboratory protocol, particulate fluorescent tracers (‘luminophores’) were used to quantify particle transport over a 10-day period. Particle mixing was quantified using the maximum penetration depth of tracers (MPD), particle diffusive coefficients (Db), and non-local transport coefficients (r). Non-local coefficients (reflecting centimeter scale transport steps) ranged from 0.4 to 15 yr−1, and were not correlated across sites with any measured biological (biomass, biovolume) or environmental parameters (temperature, grain size, organic matter). Maximum penetration depths (MPD) averaged ~10.7 cm (6.5–14.5 cm), and were similar to the global average bioturbation depth inferred from short-lived radiochemical tracers. MPD was also not correlated with measures of size (individual biomass), but increased with grain size and decreased with temperature. Biodiffusion (Db) correlated inversely with individual biomass (size) and directly with temperature over the environmental range (Q10 ~ 1.7; 5–21 °C). The transport data were comparable in magnitude to rates reported for localized H. diversicolor populations of similar size, and confirmed some but not all correlations between sediment reworking and biological and environmental variables found in previous studies. The results imply that measures of particle reworking activities of a species from a single location can be generally extrapolated to different populations at similar conditions.

Abstract: International audience; Bioindicators assess the mangroves ecological state according to the types of pressures but they differ with the ecosystem’s specificities. We investigated benthic meiofauna diversity and structure within the low human-impacted mangroves in French Guiana (South America) in response to sediment variables with various distances to the main city. Contaminant’s concentrations differed among the stations, but they remained below toxicity guidelines. Meiofauna structure (Foraminifera, Kinorhyncha, Nematoda) however varied accordingly. Nematode’s identification brought details on the sediment’s quality. The opportunistic genus Paraethmolaimus (Jensen, 1994) strongly correlated to the higher concentrations of Hg, Pb. Anoxic sediments were marked by organic enrichment in pesticides, PCB, and mangrove litter products and dominance of two tolerant genus, Terschellingia (de Man, 1888) and Spirinia (Gerlach, 1963). In each of these two stations, we found many Desmodora individuals (de Man, 1889) with the presence of epibionts highlighting the nematodes decreased fitness and defenses. Oxic sediments without contaminants were distinguished by the sensitive genera Pseudocella (Filipjev, 1927) and a higher diversity of trophic groups. Our results suggested a nematodes sensitivity to low contaminants concentrations. Further investigations at different spatio-temporal scales and levels of deterioration, would be necessary to use of this group as bioindicator of the mangroves’ ecological status; Les bioindicateurs évaluent l'état écologique des mangroves en fonction des types de pressions mais ils diffèrent selon les spécificités de l'écosystème. Nous avons étudié la diversité et la structure de la méiofaune benthique dans des mangroves à faible impact humain en Guyane française (Amérique du Sud) en réponse à des variables sédimentaires et à différentes distances de la ville principale. Les concentrations de contaminants différaient selon les stations, mais elles restaient inférieures aux recommandations de toxicité. La structure de la méiofaune (Foraminifera, Kinorhyncha, Nematoda) a cependant varié en conséquence. L'identification des nématodes a apporté des détails sur la qualité des sédiments. Le genre opportuniste Paraethmolaimus (Jensen, 1994) était fortement corrélé aux concentrations plus élevées de Hg, Pb. Les sédiments anoxiques étaient marqués par un enrichissement organique en pesticides, PCB, et produits de la litière de mangrove et par la dominance de deux genres tolérants, Terschellingia (de Man, 1888) et Spirinia (Gerlach, 1963). Dans chacune de ces deux stations, nous avons trouvé de nombreux individus Desmodora (de Man, 1889) avec la présence d'épibiontes mettant en évidence la diminution de la fitness et des capacités de défense des nématodes. Les sédiments oxiques sans contaminants se distinguaient par la présence de genres sensibles Pseudocella (Filipjev, 1927) et une plus grande diversité de groupes trophiques. Nos résultats suggèrent une sensibilité des nématodes à de faibles concentrations de contaminants. Des études supplémentaires à différentes échelles spatio-temporelles et à différents niveaux de détérioration seraient nécessaires pour utiliser ce groupe comme bioindicateur de l'état écologique des mangroves.

Abstract: Petroleum can pollute pristine shorelines as a consequence of accidental spills or chronic leaks. In this study, the fate of petroleum hydrocarbons in soft pristine sediment of Caleta Valdés (Argentina) subject to ex situ simulated oil pollution was assessed. Sedimentary columns were exposed to medium and high concentrations of Escalante Crude Oil (ECO) and incubated in the laboratory during 30 days. Levels of aliphatic hydrocarbons at different depths of the sedimentary column were determined by gas chromatography. Oil penetration was limited to the first three centimetres in both treatments, and under this depth, hydrocarbons were clearly biogenic (terrestrial plants) as in the whole sedimentary column of the control assay. Bioturbation by macrobenthic infauna was strongly impacted by oil pollution which resulted in reduced sediment oxygenation and low burial of petroleum hydrocarbons. This may partly explain the limited hydrocarbon biodegradation observed, as indicated by the relatively high values of the ratios nC17/pristane, nC18/phytane, and total resolved aliphatic hydrocarbons/unresolved complex mixture. Correspondingly, at the end of the experiment the most probable number of hydrocarbon-degrading bacteria reached ~ 103 MPN g−1 dry weight. These values were lower than those found in chronically polluted coastal sediments, reflecting a low activity level of the oil-degrading community. The results highlight the low attenuation capacities of Caleta Valdés pristine sediments to recover its original characteristics in a short time period if an oil spill occurs. Abstract: 2017-410 Mangroves are among the most sensitive marine ecosystems to oil pollution due both to the sensitivity of mangroves species and to the high persistence of hydrocarbons in these environments. Despite their ecological and socio-economic value, the potential effects of an oil spill on French Guiana mangroves remain so far unknown. Yet, there is an increasing transboundary risk of oil spill due to Brazilian offshore oil exploitation (in mid-April 2013, there were 122 offshore drilling rigs in Brazil, including 29 under construction – Brazilian Amapá region located in the French Guiana border area is thought to become an important world oil production area in the coming years). The aim of the PRISME project was to assess the natural degradation of oil in mangrove sediment as well as its impact on benthic communities (micro, meio and macrobenthos): a one-month in situ experiment was conducted in the young French Guianese mangrove (around 3 years old) at the mouth of the Sinnamary estuary. The experimental units consisted in eight plastic cores (Ø : 10 cm ; height: 30 cm) manually introduced within sediments. A thin layer of oiled sediment (2 cm, 20 000 ppm) was applied on four cores while the remaining four cores were considered as control (no oil addition). Three cores were additionally sampled at the beginning of the experiment as initial control sediments. After one month in situ, the eight cores were collected and sliced on site into different sedimentary layers aliquots for later analyses (hydrocarbons, bacterial, meio, macrofauna fauna diversity, bioturbation, biogeochemical parameters). Samples were sent to the different laboratories involved in this multidisciplinary project. Results and knowledge gained from this experimental work were used to develop an approach for assessing coastal vulnerability for oil spills preparedness in mangroves.

Abstract: International audience; An experimental oil spill was carried out in order toassess in situ responses of a macrobenthic community of shallow subtidal sediments historically exposed to petroleum contamination. Both structural and functional (bioturbation activity) parameters of the community, subjected or not to a pulse acute contamination (25,000 ppm), were studied for 18 months. No difference in the community structure was detected between contaminated and control sediments, from 6 to 18 months of experimentation. Vertical distributions of organisms, however, were affected by the presence of oil contamination leading to a deeper burial of some polychaete species. In the same time, changes in sediment-reworking activity and more especially a deeper particle burying in sediments subjected to acute oil contamination were shown.These results highlight the need to complete the analysis of community structure by assessing functional aspects, such as bioturbation activity, a process integrating various aspects of benthic behaviour (e.g. feeding, locomotion, burrow building)in order to estimate real (structural and functional) and longterm effects of oil contamination on benthic communities.

Abstract: The potential for surfactant production by the extreme halophilic archaeon Haloferax sp. MSNC14 in the presence of individual hydrocarbon substrates was studied. This strain was selected for its ability to grow on different types of hydrocarbons at high NaCl concentrations. Linear (n-heptadecane or C17) and isoprenoid (pristane) alkanes, a polyaromatic hydrocarbon (phenanthrene) and ammonium acetate (highly water-soluble control compound) were used as growth substrates. The adherence potential was demonstrated by the ability of the cells to adhere to liquid or solid hydrocarbons. The biosurfactant production was indicated by the reduction of the surface tension (ST) and by the emulsification activity (EA) of cell-free supernatants. Growth on acetate was accompanied by a low EA (lower than 0.1) and a high ST (~70 mN/m), whereas an important EA (up to 0.68 ± 0.08) and a reduction of ST (down to 32 ± 2.3 mN/m) were observed during growth on the different hydrocarbons. Both ST and EA varied with the growth phase. The adhesion to hydrocarbons was higher when cells were grown on C17 (by 60-70 %) and pristane (by 30-50 %) than on phenanthrene (~25 %). The results demonstrated that strain MNSC14 was able to increase the bioavailability of insoluble hydrocarbons, thus facilitating their uptake and their biodegradation even at high salt concentration.

Abstract: In coastal marine sediment, oxygen fluctuations induced by bioturbating activities are widespread and exert a great influence, not only on the structure and diversity of the microbenthic communities, but also on their activities. Thus, the activity of benthic organisms can have a significant influence on the degradation of hydrocarbons (HC) and can favor the development of hydrocarbonoclastic microorganisms in contaminated marine sediments. Here, we have generated metatranscriptomic data from coastal marine sediments affected by oil addition and/or by the reworking activity of the marine polychaete Hediste diversicolor to gain insights into the active microbial groups involved in the response to oil addition under the oxygen-fluctuating conditions. The preliminary results suggest that the macrofauna promote the diversity of active aerobic hydrocarbonoclastic bacteria in marine sediments, even if its influence cannot be strongly observed at the microbial community expression profiles level.

Abstract: International audience; Abiotic and biotic processes associated with the degradation of a light petroleum in brines close to the salt-saturation (~31 %) and the effect of labile organic matter (LOM) supply (casaminoacids/citrate; 0.2 and 0.1 % w/v, respectively) were followed during an incubation of 30 days. After 4-week incubation at 40 °C under light/dark cycles, a 24 % of abiotic degradation was observed in untreated brines. The stimulation of native brines community with LOM addition allowed an additional 12.8 % oil attenuation due to biodegradation processes. Successional changes in the active microbial community structure due to the oil contamination (16S rRNA DGGE approach) showed the selection of one phylotype affiliated to Salinibacter and the disappearance of Haloquadratum walsbyi in untreated brines. In LOM-amended microcosms, phylotypes related to Salinibacter, Haloarcula, Haloterrigena and Halorhabdus were selected. An effect of hydrocarbon contamination was only observed in the bacterial community with the inhibition of two dominant proteobacterial phylotypes. This study further confirms that short-term and moderate oil biodegradation is possible in LOM-stimulated brines. Biodegradation should be much more reduced under in situ conditions. Self-cleaning capacities of close to saturation hypersaline lakes appears, therefore very limited compared to non-extreme haline environments.

Abstract: The Patagonian coast is characterized by the existence of pristine ecosystems which may be particularly sensitive to oil contamination. In this study, a simulated oil spill at acute and chronic input levels was carried out to assess the effects of contamination on the macrobenthic community structure and the bioturbation activity of sediments sampled in Caleta Valdés creek. Superficial sediments were either noncontaminated or contaminated by Escalante crude oil and incubated in the laboratory for 30 days. Oil contamination induced adverse effects on macrobenthic community at both concentrations with, for the highest concentration, a marked decrease of approximately 40 and 55 % of density and specific richness, respectively. Besides the disappearance of sensitive species, some other species like Oligochaeta sp. 1, Paranebalia sp., and Ostracoda sp. 2 species have a higher resistance to oil contamination. Sediment reworking activity was also affected by oil addition. At the highest level of contamination, nearly no activity was observed due to the high mortality of macroorganisms. The results strongly suggest that an oil spill in this protected marine area with no previous history of contamination would have a deep impact on the non-adapted macrobenthic community. Abstract: Coastal shorelines suffer a variety of pollution injuries from both sides, from the sea, predominantly by crude oil from shipping and offshore mining, and from the land, principally through agricultural practices, urban wastewaters, and industrial activities. As a result, multi-pollution hot spots are found along shorelines, especially in estuaries and harbors. Microbial communities have evolved to adapt their metabolism to the presence of multi-contaminants (Duran et al. 2008; Gillan et al. 2005; Iannelli et al. 2012; Kaci et al. 2014; SabadiniSantos et al. 2014; Wang and Tam 2012). However, hydrocarbon compounds and crude oil-derived products are the most abundant pollutants, the more spectacular source being oil spill as illustrated by the recent catastrophe of Deepwater Horizon in 2010, the largest oil spill so far observed (Atlas and Hazen 2011). Although they are frequently found at unacceptably high concentrations, hydrocarbons are natural compounds, and thus, most of them can be biodegraded by the collective catabolic diversity of microorganisms (Duran and Goni Urriza 2010; Head et al. 2006; Leahy and Colwell 1990; Miralles et al. 2007; Paisse et al. 2011; Paisse et al. 2010), particularly demonstrated in coastal marine ecosystems such as salt marshes with microbial mat structures (Bordenave et al. 2004a; Bordenave et al. 2008; Bordenave et al. 2007; Bordenave et al. 2004b), mangroves (Brito et al. 2009; Brito et al. 2006) and estuaries (Chronopoulou et al. 2013; Coulon et al. 2012). However, coastal marine sediments constitute particular ecosystems submitted to fluctuating oxygenation and redox conditions from the tidal cycles and burrowing activities of the macrofauna that in turn drive microbial degradation processes (Cravo-Laureau and Duran 2014). The biodegradation of hydrocarbon compounds in such oxic/anoxic oscillating environments still remains poorly understood (Cravo-Laureau et al. 2011; Cuny et al. 2011; Vitte et al. 2013; Vitte et al. 2011). The understanding of the particular microbes involved, their ecology, their genetic and enzymatic capacities, their interactions, as well as their functioning in the changing redox conditions, is crucial for the implementation of efficient bioremediation strategies (Goni-Urriza et al. 2013; McGenity 2014). The DECAPAGE project (ANR CESA-2011-006 01; http://ipremeem.univ-pau.fr/live/DECAPAGE), funded by the French National Agency for Research (ANR), was precisely devoted to understand the adaptation mechanisms driving the reorganization of bacterial communities in response to petroleum in coastal sediments. In this special issue, the scientists involved in the DECAPAGE project present their contribution for understanding the microbial ecology in hydrocarbon-polluted mudflat sediments. The special issue also includes articles from scientists involved in other projects and programs in order to enlarge the topic to diverse ecosystems, to different biological organization levels from microbial and macrobenthic communities to bacterial populations, and addressing remediation strategies as well. Thus, the 15 articles compiled in this special issue explore diverse facets of the microbial ecology of hydrocarbon-polluted coastal sediments. The article of Acosta-Gonzalez et al. Abstract: International audience; The present study aimed to examine whether the use of dispersant would be suitable for favoring the hydrocarbon degradation in coastal marine sediments without impacting negatively micro- and macrobenthic organisms. Mudflat sediments, maintained during 286 days in mesocosms designed to simulate natural conditions, were contaminated or not with Ural blend crude oil (REBCO) and treated or not with third-generation dispersant (Finasol OSR52). While the dispersant did not lead to an increase of hydrocarbon biodegradation, its use enables an attenuation of more than 55 % of the sediment concentration of total petroleum hydrocarbons (TPH). Canonical correspondence analysis (CCA) correlating T-RFLP patterns with the hydrocarbon content and bacterial abundance indicated weak differences between the different treatments except for the mesocosm treated with oil and dispersant for which a higher bacterial biomass was observed. The use of the dispersant did not significantly decrease the macrobenthic species richness or macroorganisms' densities in uncontaminated or contaminated conditions. However, even if the structure of the macrobenthic communities was not affected, when used in combination with oil, biological sediment reworking coefficient was negatively impacted. Although the use of the dispersant may be worth considering in order to accelerate the attenuation of hydrocarbon-contaminated mudflat sediments, long-term effects on functional aspects of the benthic system such as bioturbation and bacterial activity should be carefully studied before.

Abstract: International audience; The present study aimed to examine whether the physical reworking of sediments by harrowing would be suitable for favouring the hydrocarbon degradation in coastal marine sediments. Mudflat sediments were maintained in mesocosms under conditions as closer as possible to those prevailing in natural environments with tidal cycles. Sediments were contaminated with Ural blend crude oil, and in half of them, harrowing treatment was applied in order to mimic physical reworking of surface sediments. Hydrocarbon distribution within the sediment and its removal was followed during 286 days. The harrowing treatment allowed hydrocarbon compounds to penetrate the first 6 cm of the sediments, and biodegradation indexes (such as n-C18/phytane) indicated that biodegradation started 90 days before that observed in untreated control mesocosms. However, the harrowing treatment had a severe impact on benthic organisms reducing drastically the macrofaunal abundance and diversity. In the harrowing-treated mesocosms, the bacterial abundance, determined by 16S rRNA gene Q-PCR, was slightly increased; and terminal restriction fragment length polymorphism (T-RFLP) analyses of 16S rRNA genes showed distinct and specific bacterial community structure. Co-occurrence network and canonical correspondence analyses (CCA) based on T-RFLP data indicated the main correlations between bacterial operational taxonomic units (OTUs) as well as the associations between OTUs and hydrocarbon compound contents further supported by clustered correlation (ClusCor) analysis. Abstract: International audience; Microbial communities inhabiting coastal marine sediments at intertidal zones are subjected to fluctuating environmental conditions according to tide level and reworking activities of macroorganisms. Such fluctuations are important drivers for the microbial communities’ assemblages and organization that in turn determine the microbial hydrocarbon-degradation processes. Notwithstanding the difficulties to access the entire ecosystem complexity, experimental ecology studies, involving more or less sophisticated experimental systems, have demonstrated to be suitable approaches. Several systems have been developed to tackle the role of microorganisms in hydrocarbon degradation in marine sediments in order to decipher the role of microbial communities in hydrocarbon degradation and understand the microbial behavior in response to an oil spill. We describe here a mesocosm system that maintains sediments under conditions as close as possible to those prevailing in the environment. We explain how to collect mudflat sediments conserving their structure and how to apply crude oil in order to simulate an oil spill. Subsampling procedures are also provided for following the fate of crude oil hydrocarbons, the macrofauna burrowing activity, as well as the behavior and dynamic of microbial communities.

Abstract: Kinetics of phenanthrene degradation, emulsification and surface tension activities of this hydrocarbon upon degradation by a Pseudomonas putida (COB 3-2) isolated from a coastal microbial mat were investigated. After 27 days of incubation at 20°C, 71.4±3.4 % of the initial phenanthrene amount (0.4 g l-1) were biodegraded. Observation of both tension-active and emulsifying activities indicated that biosurfactants, probably rhamnolipids, are generated. The high phenanthrene degrading capacity and biosurfactant production potential of this newly isolated P. putida strain may contribute positively to oil bioremediation processes within the microbial mat.

Abstract: Aims: To isolate and identify alkane-degrading bacteria from deep-sea superficial sediments sampled at a north-western Mediterranean station. Methods and Results: Sediments from the water/sediment interface at a 2400 m depth were sampled with a multicorer at the ANTARES site off the French Mediterranean coast and were promptly enriched with Maya crude oil as the sole source of carbon and energy. Alkane-degrading bacteria belonging to the genera Alcanivorax, Pseudomonas, Marinobacter, Rhodococcus and Clavibacter-like were isolated, indicating that the same groups were potentially involved in hydrocarbon biodegradation in deep sea as in coastal waters. Conclusions: These results confirm that members of Alcanivorax are important obligate alkane degraders in deep-sea environments and coexist with other degrading bacteria inhabiting the deep-subsurface sediment of the Mediterranean. Significance and Impact of the Study: The results suggest that the isolates obtained have potential applications in bioremediation strategies in deep-sea environments and highlight the need to identify specific piezophilic hydrocarbon-degrading bacteria (HCB) from these environments.

Abstract: The biodegradation of phenanthrene by the marine strain Sphingomonas sp. 2MPII (DSMZ 11572) was enhanced by the solubilizating properties of the nonionic surfactant Tween 80. After 197 h of incubation, 85 +/- 4% of the initial amount of phenanthrene (0.4 g l-1) was biodegraded in presence of Tween 80 (0.5 g l-1) as opposed to 52 +/- 5% without this synthetic surfactant. These results confirm that the activity of the strain 2MPII is limited by the bioavailability of the polycyclic aromatic hydrocarbon (PAH) substrate in the aqueous phase. Tween 80 appears to be efficient in increasing the bioavailability of hydrophobic compounds such as PAHs.

Abstract: Rates of photodegradation of chlorophylls in their entirety and of chlorophyll phytyl chains, and time evolution of the Chlorophyll Phytyl side-Chain Photodegradation Index (CPPI) under visible light exposure were determined and compared in laboratory experiments. Killed phytoplanktonic cells of two widespread Bacillariophyceae (Thalassiosira rotula Meun. and Skeletonema costatum (Grev.) Cleve), one widespread Prymnesiophyceae (Emiliania huxleyi (Lohm.) May and Mohler) and one Chlorophyceae (Dunaliella tertiolecta Butch) were used as source of pigments (chlorophyll a, chlorophyll b and phaeophytin a). Chlorophyll and chlorophyll phytyl chain photodegradation and CPPI evolution showed apparent first-order kinetics with respect to light exposure. The in vitro determined degradation rates were approximately 3.5 times higher for the chlorophyll tetrapyrrolic structure than for the phytyl chain. Few variations of the relative photodegradation constant rates were observed between strains: less than 34% for chlorophyll a and 25% for chlorophyll phytyl chain at 20°C. Nonetheless, the photo-oxidation process of the phytyl chain (Q10=2.0) seems more affected by temperature than does chlorophyll a (Q10=1.4). Phytol loss in the dark controls have been attributed to bacterial degradation, and, in the case of S. costatum, chlorophyllase dephytylation seems to have enhanced the potential for bacterial biodegradation. From these results, it clearly appears that in small detrital particles not only chlorophyll a, chlorophyll b and phaeophytin but also their phytyl chains may be rapidly photodegraded in the upper portion of the euphotic zone. The use of the CPPI for monitoring the chlorophyll photo-oxidation process in the aquatic environment is discussed.

Abstract: Bacterial bioluminescence is commonly found in the deep sea and depends on environmental conditions. Photobacterium phosphoreum ANT-2200 has been isolated from the NW Mediterranean Sea at 2200-m depth (in situ temperature of 13 degrees C) close to the ANTARES neutrino telescope. The effects of hydrostatic pressure on its growth and luminescence have been investigated under controlled laboratory conditions, using a specifically developed high-pressure bioluminescence system. The growth rate and the maximum population density of the strain were determined at different temperatures (from 4 to 37 degrees C) and pressures (from 0.1 to 40 MPa), using the logistic model to define these two growth parameters. Indeed, using the growth rate only, no optimal temperature and pressure could be determined. However, when both growth rate and maximum population density were jointly taken into account, a cross coefficient was calculated. By this way, the optimum growth conditions for P. phosphoreum ANT-2200 were found to be 30 degrees C and, 10 MPa defining this strain as mesophile and moderately piezophile. Moreover, the ratio of unsaturated vs. saturated cellular fatty acids was found higher at 22 MPa, in agreement with previously described piezophile strains. P. phosphoreum ANT-2200 also appeared to respond to high pressure by forming cell aggregates. Its maximum population density was 1.2 times higher, with a similar growth rate, than at 0.1 MPa. Strain ANT-2200 grown at 22 MPa produced 3 times more bioluminescence. The proposed approach, mimicking, as close as possible, the in situ conditions, could help studying deep-sea bacterial bioluminescence and validating hypotheses concerning its role into the carbon cycle in the deep ocean.

Abstract: International audience; Oil spills threaten coastlines where biological processes supply essential ecosystem services. Therefore, it is crucial to understand how oil influences the microbial communities in sediments that play key roles in ecosystem functioning. Ecosystems such as sediments are characterized by intensive bioturbation due to burrowing macrofauna that may modify the microbial metabolisms. It is thus essential to consider the bioturbation when determining the impact of oil on microbial communities. In this study, an experimental laboratory device maintaining pristine collected mudflat sediments in microcosms closer to true environmental conditions - with tidal cycles and natural seawater - was used to simulate an oil spill under bioturbation conditions. Different conditions were applied to the microcosms including an addition of: standardized oil (Blend Arabian Light crude oil, 25.6 mg.g21 wet sediment), the common burrowing organism Hediste (Nereis) diversicolor and both the oil and H. diversicolor. The addition of H. diversicolor and its associated bioturbation did not affect the removal of petroleum hydrocarbons. After 270 days, 60% of hydrocarbons had been removed in all microcosms irrespective of the H. diversicolor addition. However, 16S-rRNA gene and 16S-cDNA T-RFLP and RT-PCR-amplicon libraries analysis showed an effect of the condition on the bacterial community structure, composition, and dynamics, supported by PerMANOVA analysis. The 16S-cDNA libraries from microcosms where H. diversicolor was added (oiled and un-oiled) showed a marked dominance of sequences related to Gammaproteobacteria. However, in the oiled-library sequences associated to Deltaproteobacteria and Bacteroidetes were also highly represented. The 16S-cDNA libraries from oiled-microcosms (with and without H. diversicolor addition) revealed two distinct microbial communities characterized by different phylotypes associated to known hydrocarbonoclastic bacteria and dominated by Gammaproteobacteria and Deltaproteobacteria. In the oiled-microcosms, the addition of H. diversicolor reduced the phylotype-richness, sequences associated to Actinobacteria, Firmicutes and Plantomycetes were not detected. These observations highlight the influence of the bioturbation on the bacterial community structure without affecting the biodegradation capacities.

Abstract: Comparison of the stability of intact and photooxidized chlorophyll phytyl chain with depth in a core of recent sediment (where high bacterial activity was observed) allowed us to select potential markers for monitoring past photodegradation of chlorophyll. “Esterified + bound” 3-methylidene-7,11,15-trimethylhexadecan-1,2-diol appeared to be particularly suitable for such an application, but we excluded “esterified + bound” 6,10,14-trimethyl-pentadecan-2-one which is not sufficiently stable nor is it exclusively produced by photooxidation. For example, it can be produced during alkaline hydrolysis of tocopherols. Bacterial processes appeared to be particularly active on “free” isoprenoid compounds.

Abstract: 6,10,14-Trimethylpentadecan-2-one, 3-methylidene-7,11,15-trimethylhexadecan-1,2-diol, (Z and E) 3,7,11,15-tetramethyl-2,3-epoxyhexadecanols, (Z and E) 3,7,11,15-tetramethyl-2,3-epoxyhexadecanals and (Z and E) 3,7,11,15-tetramethylhexadec-2-enals were unambiguously identified in attached leaves of senescing Avena sativa collected during late summer. These isoprenoid compounds result from the photooxidation of the chlorophyll phytyl chain during senescence. In vitro dark and light-induced senescence experiments carried out with Petroselinum sativum confirmed these results.

Abstract: Unbound phenolic compounds of leaves of the marine phanerogam Posidonia oceanica (L.) Delile from an area colonised by the introduced chlorophyte Caulerpa taxifolia (Vahl) C. Ag. have been investigated and their concentration assessed using high performance liquid chromatography (HPLC). Leaves of the colonised area exhibited high maximum values of phenolic contents in comparison with a reference area. Nonetheless, because of the high variability of the values, further studies will be needed to confirm that P. oceanica plants are producing unbound phenolic compounds in reaction to growth of C. taxifolia. Ferulic acid, phloridzin, phloroglucinol, p-anisic acid, acetosyringone, sinapic acid, phenol, p-hydroxybenzoic acid, p-coumaric acid and cinnamic acid represent more than 95% of the mean relative concentration of phenolic compounds measured. Several of these compounds had not hitherto been reported in P. oceanica.

Abstract: The heterogeneity of oxygen distribution in a Hediste diversicolor burrow environment was investigated in a laboratory experiment using a 6-mm thick tank equipped with oxygen planar optodes. The two-dimensional oxygen distribution in a complete burrow was monitored every 2 min for 4 h. Oxygen concentrations fluctuated over a scale of minutes in the burrow lumen and wall (up to 2 mm) reflecting the balance between worm ventilation activity and oxygen consumption. The magnitude of the three surrounding micro-horizons (oxic, oscillating and anoxic) induced by the intermittent worm ventilation was spatially and temporally variable within the structure. Oxygen variations appeared to be controlled by distance from the sediment-water interface and the direction of water circulation. Moreover, there was an apparent 'buffer effect', induced by the proximity to the overlying water, which reduced the variations of lumen and wall oxygen in the upper part of the structure. These results highlight the heterogeneous distribution and dynamics of oxygen associated with H. diversicolor burrows and ventilation activity. They also highlight the necessity of integrating this complexity into the current burrow-base models in order to estimate the ecological importance of burrowing species in coastal ecosystems. Abstract: Bioturbation is known to stimulate microbial communities, especially in macrofaunal burrows where the abundance and activities of bacteria are increased. Until now, these microbial communities have been poorly characterized and an important ecological question remains: do burrow walls harbor similar or specific communities compared with anoxic and surface sediments? The bacterial community structure of coastal sediments inhabited by the polychaete worm Hediste diversicolor was investigated. Surface, burrow wall and anoxic sediments were collected at the Carteau beach (Gulf of Fos, Mediterranean Sea). Bacterial diversity was determined by analyzing small subunit ribosomal RNA (16S rRNA) sequences from three clone libraries (168, 179 and 129 sequences for the surface, burrow wall and anoxic sediments, respectively). Libraries revealed 306 different operational taxonomic units (OTUs) belonging to at least 15 bacterial phyla. Bioinformatic analyses and comparisons between the three clone libraries showed that the burrow walls harbored a specific bacterial community structure which differed from the surface and anoxic environments. More similarities were nevertheless found with the surface assemblage. Inside the burrow walls, the bacterial community was characterized by high biodiversity, which probably results from the biogeochemical heterogeneity of the burrow system. (C) 2011 Institut Pasteur. Abstract: ISBN: 978-1-904455-83-7; Sediments can serve as sinks for hydrocarbon contaminants in marine ecosystems. Once settled, hydrocarbons fate will be dominated by several abiotic and biotic processes that will result in either their partial or total degradation or in a selective preservation when buried within the sediment. Biodegradation of hydrocarbons in marine sediments is mainly due to the existence of prokaryotes harbouring specific catabolic genes enabling the degradation of these compounds under oxic, suboxic and anoxic conditions. The interplay of the various factors that govern hydrocarbons biodegradation in marine sediments is highly complex as illustrated by bioturbation processes carried out by macrofaunal organisms. For instance, the redox oscillation regimes generated by macrofaunal organisms, and the efficiency of metabolic coupling between functional groups associated to these specific redox regimes, are probably determinant factors controlling the biodegradation rates of hydrocarbons in marine sediments. From the understanding of how these natural occurring factors may modulate the rates of hydrocarbons biodegradation, innovative bioremediation strategies may emerge.

Abstract: Muddy areas and more or less coastal anoxic zones play a key role in maintaining the integrity of estuarine and coastal ecosystems. By their localization, these areas are frequently exposed to pollutant damage, such as oil spills or accidental events, and accumulate various pollutants released by continental hydrosystems. Due to their composition (fine silt sediments), actual remediation techniques are not adapted. These artificial compounds accumulate in different sites, constituting “pollutant reservoirs” that are threats for the ecosystem as well as for human health. Microorganisms play a key role in hydrocarbon degradation. However, most of our knowledge has been obtained by studies on isolated bacterial strains. In addition, bacterial cooperation within different microbial groups has been demonstrated for the degradation of complex hydrocarbon mixtures. Silt sediments are located at the oxic/anoxic interface where various bacterial metabolisms coexist and follow one another through the function of the tides. Moreover, burrowing organisms can influence the bacterial metabolisms by making the oxygen penetration easier. It thus seems judicious to tackle the problem of hydrocarbon degradation in a global way, trying to understand how bacterial metabolisms interact in the degradation of the pollutants and to estimate the bioturbation effects of these activities. This is the main objective of the project DHYVA (Degradation des HYdrocarbures dans les VAsieres), with a special focus on anaerobic degradation mechanisms, about which we have limited knowledge.

Abstract: International audience; Little information exists about the ability of halophilic archaea present in hypersaline environments to degrade hydrocarbons. In order to identify the potential actors of hydrocarbon degradation in these environments, enrichment cultures were prepared using samples collected from a shallow crystallizer pond with no known contamination history in Camargue, France, with n-alkanes provided as source of carbon and energy. Five alkane-degrading halophilic archaeal strains were isolated: one (strain MSNC 2) was closely related to Haloarcula and three (strains MSNC 4, MSNC 14, and MSNC 16) to Haloferax. Biodegradation assays showed that depending on the strain, 32 to 95% (0.5 g/l) of heptadecane was degraded after 30 days of incubation at 40A degrees C in 225 g/l NaCl artificial medium. One of the strains (MSNC 14) was also able to degrade phenanthrene. This work clearly shows for the first time the potential role of halophilic archaea belonging to the genera Haloarcula and Haloferax in the degradation of hydrocarbons in both pristine and hydrocarbon-contaminated hypersaline environment

Abstract: To achieve a better understanding of the ecological significance of Archaea in relation to petroleum contamination, we examined the composition of archaeal communities in anoxic coastal marine infralittoral sediments and their response to simulated petroleum contamination. Sediments were collected at an experimental site, and half were mixed with oil. Control and oiled sediments were then reworked into PVC cores that were inserted into the sediment at their sampling site at 20 m water depth. After 503 d of in situ incubation, molecular and statistical analyses clearly showed that petroleum contamination induced significant shifts in the composition of archaeal communities inhabiting these anoxic sediments. Overall, contamination led to a significant decrease in diversity. Control sediments were dominated by Crenarchaeota, whereas in oiled sediments,Euryarchaeota became dominant, as attested by the particular contribution of Methanococcoides, Methanosarcina, and Methanolobus sequences. Methanogens could be indirectly related to the degradation of the added hydrocarbons by participating in syntrophic consortia with hydrocarbon-degrading acetogenic bacteria. In addition, the abundance of anaerobic methane-oxidizing Archaea sequences in oiled sediments could indicate that anaerobic oxidation of methane (AOM) occurs at the depth selected for the analyses. Further studies undertaken to quantify vertical distribution of AOM, methanogenic activity, and their potential causative Archaea would help to enhance understanding of this complex petroleum-contaminated sedimentary system.

39. Oxygen Distribution Heterogeneity Related to Bioturbation Quantified by Planar Optode Imaging

Abstract: Oxygen plays a key role in benthic microbial ecology. Until recently, oxygen concentration in sediments was measured with oxygen microsensors along a vertical profile (one dimension) from the surface until a few centimeters into the sediment. With this approach, however, it is a tedious job to describe or overcome the heterogeneity of oxygen distribution in environments such as bioturbated environments. Recently, a new technique has been introduced that allows the investigation of two-dimensional oxygen distribution and dynamics at a high resolution in the upper sediment column. This non-destructive technique takes advantage of an oxygen-quenchable fluorophore, which is cast into a thin sheet, the planar optode. The latter may be introduced in sediments and is used in situ or in laboratory experimentation, coupled with an optical system allowing the oxygen quantification. Oxygen optodes were used in experimentation dedicated to the study of oxygen heterogeneity induced by macrofaunal bioturbation. Oxygen images of sediments inhabited by a biodiffusor, the gastropod Cyclope neritea, and two gallery diffusors, the annelids Nereis virens and Nereis diversicolor, were used to compare the impact of these organisms on oxygen distribution in sediments. Diffusive oxygen flux and a heterogeneity index were quantified based on oxygen images. Results showed that all species increased oxygen distribution heterogeneity, and that this heterogeneity increased with increasing total diffusive fluxes.

Abstract: International audience; The diffusive oxygen uptake (DOU) of sediments inhabited by Chironomus riparius and Tubifex tubifex was investigated using a planar oxygen optode device, and complemented by measurements of bioturbation activity. Additional experiments were performed within contaminated sediments to assess the impact of uranium on these processes. After 72 h, the two invertebrate species significantly increased the DOU of sediments (13–14%), and no temporal variation occurred afterwards. Within contaminated sediments, it was already 24% higher before the introduction of the organisms, suggesting that uranium modified the sediment biogeochemistry. Although the two species firstly reacted by avoidance of contaminated sediment, they finally colonized it. Their bioturbation activity was reduced but, for T. tubifex, it remained sufficient to induce a release of uranium to the water column and an increase of the DOU (53%). These results highlight the necessity of further investigations to take into account the interactions between bioturbation, microbial metabolism and pollutants.

Abstract: International audience; The influence of sediment oxygen heterogeneity, due to bioturbation, on diffusive oxygen flux was investigated. Laboratory experiments were carried out with 3 macrobenthic species presenting different bioturbation behaviour patterns:the polychaetes Nereis diversicolor and Nereis virens, both constructing ventilated galleries in the sediment column, and the gastropod Cyclope neritea, a burrowing species which does not build any structure. Oxygen two-dimensional distribution in sediments was quantified by means of the optical planar optode technique. Diffusive oxygen fluxes (mean and integrated) and a variability index were calculated on the captured oxygen images. All species increased sediment oxygen heterogeneity compared to the controls without animals. This was particularly noticeable with the polychaetes because of the construction of more or less complex burrows. Integrated diffusive oxygen flux increased with oxygen heterogeneity due to the production of interface available for solute exchanges between overlying water and sediments. This work shows that sediment heterogeneity is an important feature of the control of oxygen exchanges at the sediment–water interface.

Abstract: The effects of spilled oil on sedimentary bacterial communities were examined in situ at 20 m water depth in a Mediterranean coastal area. Sediment collected at an experimental site chronically subjected to hydrocarbon inputs was reworked into PVC cores with or without a massive addition of crude Arabian light oil ( approximately 20 g kg(-1) dry weight). Cores were reinserted into the sediment and incubated in situ at the sampling site (20 m water depth) for 135 and 503 days. The massive oil contamination induced significant shifts in the structure of the indigenous bacterial communities as shown by ribosomal intergenic spacer analysis (RISA). The vertical heterogeneity of the bacterial communities within the sediment was more pronounced in the oiled sediments particularly after 503 days of incubation. Response to oil of the deeper depth communities (8-10 cm) was slower than that of superficial depth communities (0-1 and 2-4 cm). Analysis of the oil composition by gas chromatography revealed a typical microbial alteration of n-alkanes during the experiment. Predominant RISA bands in oiled sediments were affiliated to hydrocarbonoclastic bacteria sequences. In particular, a 395-bp RISA band, which was the dominant band in all the oiled sediments for both incubation times, was closely related to hydrocarbonoclastic sulfate-reducing bacteria (SRB). These bacteria may have contributed to the main fingerprint changes and to the observed biodegradation of n-alkanes. This study provides useful information on bacterial dynamics in anoxic contaminated infralittoral sediments and highlights the need to assess more precisely the contribution of SRB to bioremediation in oil anoxic contaminated areas.

Abstract: International audience; The fate of ingested eukaryotic photoautotrophic fatty acids during gut transit in the lugwormArenicolamarina (L.) and the influence of A. marina's faeces on the evolution of fatty acid distribution and bacterial community structure in superficial sediments were studied under laboratory conditions. Dead phytoplanktonic cells (food portions) were fed to individual A. marina and subsequently incubated, or allowed to directly incubate in the presence of fresh egesta or non-ingested sediment. Changes in fatty acid composition and genetic structure of bacterial communities during gut transit and/or incubation were monitored using gas-chromatography/mass-spectrometry and a DNA fingerprint approach (RISA), respectively. Results, supported by principal component analyses, suggest that A. marina's feeding activity can directly and indirectly affect the lipid biomarker composition and the bacterial community structure of inhabited sediments. Faecal casts produced fromfood portions appeared qualitatively enriched in saturated fatty acids relative to (poly)unsaturated ones due, partly, to an increase of some bacterial fatty acids and to the preferential removal of some polyunsaturated fatty acids (PUFAs). The incubation of food portions in the presence of fresh A. marina's egesta (designed to study the indirect impact of feeding by A. marina) induced a significant increase in the concentrations of C20 and C22 polyunsaturated fatty acids (PUFAs), whereas these compounds almost disappeared following direct feeding and subsequent incubation, indicating that some dietary fatty acidsmay be more accessible to biodegradation following passage through the gut of A. marina. The aforementioned increase in PUFAs was attributed to a bacterial production during incubation, suggesting the presence of PUFA-producing bacteria in the fresh egesta of A. marina. Those bacteria were either enteric bacteria thatwere releasedwith the egesta or ingested bacteria that have survived gut passage, as suggested by the variations of the bacterial community structure (i.e. RISA profiles) during incubation. The results suggest that aged faeces from A. marina might be, in some circumstances, of relatively high nutritional value to trophic levels which are unable to synthesize essential PUFAs de novo. The presence of PUFA-producing bacteria in guts of marine lugworms deserves further attention.

Abstract: Particulate samples from the water column were collected monthly from depths of 5-150 m, between May 1996 and March 1997, in the northwestern Mediterranean Sea (Ligurian Sea) as part of the DYFAMED project within the French JGOFS program. These samples were analyzed by gas chromatography-electron impact mass spectrometry for their phytol and 3-methylidene-3,7,11-trimethylhexadecan-1,2-diol (phytyldiol) content. The corresponding Chlorophyll Phytyl side chain Photodegradation Index, molar ratio of phytyldiol to phytol, was calculated and the mean amount of chlorophyll photodegraded within the euphotic zone estimated. Seasonal differences in the chlorophyll photodegradation process appear in the one-year study. The chlorophyll appeared more photodegraded in the surface water (generally more than 40% photodegraded at 5-10m) than at the deep chlorophyll maximum (DCM) (40-50m) observed in the summer stratified waters (about 20% photodegraded). This difference was attributed to the healthy state of the phytoplankton community (coincidence with the highest primary production levels) and to the lower intensity of irradiance at the DCM level. On the other hand, the bulk of the detrital chlorophyll (chlorophyll associated with phytodetritus, phaeopigments) undergoes photodegradation before it sinks out of the photic zone. However, in January (winter mixed water) the pigments exported towards the sea floor were less photodegraded. This is thought to result from a shorter period of residence of the pigments in the photic zone due to vertical convection and grazing activity of macrozooplankton (salps), which are producers of rapid sinking fecal pellets. Abstract: International audience; European Water Framework Directive is enforced in five tropical French Oversea Territories where mangroves are present. Developing bioindication tools to support the ecosystem-based management approach of the Directive is needed. A series of expert workshops was organized and led to the proposal of a strategy and of an applied research program to develop bioindication tools. The proceedings of the workshops are presented as a case study, as this is the first time such an integrative ecosystem-based approach is proposed in mangroves, combining structural and functional aspects, from forest structure to benthic community functioning.

Abstract: International audience; Mangrove forests are formed by mangrove trees and shrubs that grow in the intertidal zone at the sea-continent interface. They constitute major ecosystems of tropical to subtropical muddy coasts that perform several ecological functions, including: mitigation of coastal erosion and flooding hazards associated with storm waves, extreme tides and tsunami, providing nurseries for some estuarine and coastal species (e.g., shrimps, fishes), production and recycling of organic matter, carbon storage, functioning as long-term sinks for several contaminants. World mangroves face a number of threats with increasing habitat destruction caused by direct and indirect anthropogenic pressures coupled with global climate change. They are known to be extremely vulnerable to oil spills. Even if the fate and impact of oil spills in such ecosystems have been partially monitored and experimentally studied (e.g., 30-year TROPICS field experiment in Panama islands, replicated field trials conducted in central Queensland, Australia) significant gaps in knowledge remain. The oil dynamic in such ecosystem is complex and depends on the abiotic-biotic processes interactions. Understanding the fate and impact of the oil spill thus requires an integrated approach of the functioning of the whole mangroves system facing the pollution. The case of the French Guiana mangroves, subjected to intense hydromorpho-sedimentary dynamics under the direct influence of the massive discharge of suspended sediments from the Amazon River, will serve as conceptual model to highlight the importance of the need for a specific Ecosystem-based Management response in case of oil spill.
