The interaction between Actinobacillus pleuropneumoniae and Influenza A-virus in experimentally infected mice Models for infecting mice with Influenza A-Virus (A/PR 8/34, H0N1) and Actinobacillus pleuropneumoniae (serotype 9) were developed in Han: NMRI-mice. After infecting mice with sublethal doses of one of the infectious agents, or both together as a mixed infection, animals were subsequently exsanguated and the lungs washed by bronchoalveolar lavage. Clinical symptoms were recorded daily, examination of lung lavage fluid and sera as well as histology of the lungs were done. An increase in mortality, weight reduction and total cell yield of lung lavage fluid was observed after mixed infection. Compared to mixed infections total protein content and elastase in sera and lung lavage fluid after singular ones were raised not as much. In lung lavage fluid the total cell yield was increased more marked. These alterations indicate a synergistic effect of viruses and bacteria, developed by mixed infection as well as a bacterial infection on top of a viral one. Histopathologically the lung alterations were found to depend on the infectious agent and the mode of infection. Zusammenfassung Es wurden Infektionsmodelle für Influenza A-Virus (A/PR 8/34, H0N1) und Actinobacillus pleuropneumoniae (Serovar 9) an Han: NMRI-Mäusen entwickelt. Nach Einzelinfektionen und Mischinfektionen mit subletalen Dosen wurden die Tiere zu verschiedenen Zeiten durch Entbluten getötet und ihre Lungen gespült. Untersuchungen erfolgten an der Maus selbst (klinische Symptome), an der Lungenspülflüssigkeit (bronchoalveoläre Lavage), am Serum und histologisch (Lungen). Bei den Mischinfektionen waren die infektionsbedingte Gewichtsabnahme und die Mortalität gegenüber den Einzelinfektionen erhöht. In den Lungenspülflüssigkeiten und Seren stiegen Gesamtprotein- und Elastasewert nach Mischinfektion stärker an als nach Einzelinfektion, die Gesamtzellzahlen in der Lungenspülflüssigkeit waren erhöht. Diese Hinweise auf ein Zusammenwirken von Virus und Bakterium wurden bei simultaner Mischinfektion und bei bakterieller Sekundärinfektion deutlich. Crude dermonecrotic toxins (DNT) were prepared from Pasteurella. multocida (P. m.) type D and type A strains isolated from pigs with atrophic rhinitis. Rabbits were immunized with the DNT of P. m. type D. This serum neutralized the DNT of P. m. type A to the same degree as the homologous one both in vitro (cytopathogenicity for tissue culture cells) and in vivo (mouse lethality and dermonecrotic activity in guinea pig). Zusammenfassung Antigenverwandtschaft der dermonekrotischen Toxine vom Pasteurella multocida Typ D und Typ A Von Pasteurella multocida-Stämmen Typ A und D, die aus Schweinen mit Rhinitis atrophicans isoliert worden waren, konnte ein dermonekrotisches Rohtoxin (DNT) präpariert werden. Mit dem DNT aus P. mult Typ D wurden Kaninchen immunisiert. Zui Uberprüfung der Wirksamkeit der Antikörper dienten in vitro die Neutralisation des cytopathischen Effekts in Gewebszellkulturen und in vivo der Seroneutralisationstest an Mäusen und die Neutralisation der dermonekrotischen Aktivität bei Meerschweinchen. Mit den gewonnenen Antikörpern gegen DNT von P. mult Typ D konnte in vitro wie in vivo die Wirkung des DNT aus P. mult Typ A in gleichem Maß neutralisiert werden wie die des homologen Toxins von P. mult Typ D. A crude dermonecrotic toxin (DNT) of Pasteurella multocida (P. m.) type D was prepared by repeated sonication and freezing. It was sterilized by filtration. A toxoid was then made and pigs were hyperimmunized with it to get an antiserum. A control serum was obtained by hyperimmunization of pigs with a preparation derived from nontoxigenic P. m. type D in the same manner as the toxoid. Three gnotobiotic piglets were injected with the antiserum. This resulted in neutralization indices (NI) of 25 in their sera, as tested on mice. Three litter-mated controls were given the control serum. Their NI remained 1. All piglets were challenged intramuscularly 4 times, every third day, with 30 mouse LD50 of the DNT. When euthanized 15 days after the last DNT administration no snout lesions were found in passively immunized piglets, whereas control animals showed severe turbinate atrophy and other changes typical for atrophic rhinitis. The next experiment was identical to the previous one except for the challenge, which was given intranasally (4 times 300 mouse LD50). Also in this case circulating antitoxin protected the piglets from damage of the nasal turbinates caused by the DNT. Typ D isoliert werden. Nach der Sterilfiltration wurde zur Gewinnung von Antiserum ein Toxoid hergestellt, mit de***m Schweine hyperimmunisiert wurden. Das Kontrollserum stammte von Schweinen, die mit einer aus nichttoxinogener P. m. Typ D isolierten Substanz immunisiert worden waren. Die Herstellung erfolgte in gleicher Weise wie bei dem Toxoid. Drei gnotobiotischen Ferkeln wurde das Antiserum gespritzt. In deren Sera wurden im Mäuseschutztest Neutralisations-Indizes (NI) von 25 gemessen. Die NI der drei mit Kontrollserum behandelten Wurfgeschwister blieben dagegen bei 1. Alle Ferkel wurden mit 30 Maus LD50 von DNT 4mal im Abstand von jeweils 3 Tagen intramuskulär belastet. 15 Tage nach der letzten Applikation von DNT erfolgte die Tötung der Ferkel. Es konnten keine Veränderungen im Nasenbereich der passiv immunisierten Ferkel gefunden werden. Dagegen zeigten die Kontrolltiere schwere Nasenmuschelatrophien und andere für Rhinitis atrophicans typische Veränderungen. Das folgende Experiment war mit dem oben beschriebenen identisch bis auf den Challenge der Tiere, der intranasal erfolgte (4mal 300 Maus LD50). Following systemic application of dermonecrotic crude toxin (DNT) from a sonic extract of Pasteurella multocida type D into 20 young adult male rats, its distribution and time dependence as well as the toxin induced organ alterations were studied. In all animals, liver, kidney, spleen, lymph nodes, thymus, cross-sections of the nose, duodenum, gastric wall, lung, and adrenal gland were excised, fixed, dehydrated and embedded in glycol methacrylate. The immunoperoxidase-avidin-biotin-complex-technique was chosen to detect the toxin in various organs. Toxin was found in decreasing amounts in liver, kidney, spleen, lymph nodes and thymus. In the other organs the essay remained negativ. The DNT was responsible for 3 major histopathological findings: a dystrophic alteration of the parenchymic cells of liver, kidney and adrenal gland; a necrosis of the lymphocytes in thymus, spleen and lymph nodes; an accumulation of neutrophilic granulocytes in the altered tissues, mainly, however, in the cortical and medullary sinus of the lymph nodes. Zusammenfassung Der toxische Kulturüberstand von Pasteurella multocida Typ D und seine Verteilung in der Ratte Nach systemischer Gabe des im Meerschweinchenhauttest Dermatonekrose-positiv reagierenden Rohtoxins aus Pasteurella multocida Typ D wurden die räumliche und zeitliche Verteilung sowie die durch das Toxin hervorgerufenen Organveränderungen in 20 jungadulten, männlichen Ratten demonstriert. Es folgte die Untersuchung von Leber, Niere, Milz, Lymphknoten, Thymus, Nasenquerschnitten, Duodenum, Magenwand, Lunge, und Nebenniere, welche fixiert, entwässert und in Glycolmethacrylat eingebettet wurden. Die Toxindarstellung erfolgte mittels der Immunperoxidase-Avidin-Biotin-Komplex-Technik. Hinsichtlich der räumlichen Verteilung konnte das Toxin immun-histochemisch in abnehmender Menge in Leber, Niere, Milz, Lymphknoten und Thymus nachgewiesen werden. In den übrigen untersuchten Organen verlief der Toxinnachweis negativ. In bezug auf die patho-histologischen Veränderungen traten folgende Hauptbefunde auf: eine dystrophische Veränderung der Parenchymzellen von Leber, Niere und Nebennierenmark, eine Nekrose der Lymphozyten in Thymus, Milz und Lymphknoten und eine Ansammlung von neutrophilen Granulozyten im Bereich der o. g. The adhesive properties of 5 toxigenic Pasteurella multocida (P. m.) strains isolated from pigs with atrophic rhinitis and 7 non-toxigenic strains derived from nasal cavities of normal pigs were studied in vitro. The bacteria were grown either in brain heart infusion broth or on blood agar. For adherence testing, swine nasal epithelial cells, nasopharyngeal and tracheobronchial epithelia, bladder epithelia, human buccal epithelia and tissue culture cells (Buffalo green monkey and HeLa cells) were used. The time of incubation of bacteria and cell mixtures ranged between 0.5 and 2 hours. All P. m. strains showed very poor adherence to swine nasal epithelial cells and other cell types used, regardless of the conditions of their cultivation or modifications of adherence testing. The average adherence was 0.07 ± 0.3 bacteria per cell, whereas a Bordetella bronchiseptica strain tested as a control adhered at a mean level of 15.3 bacteria per cell. Die Anzüchtung der Bakterien erfolgte entweder in 'Brain Heart Infusion Bouillon' oder auf Blutagar. Für den Anheftungstest wurden nasale Epithelzellen vom Schwein, nasopharyngeales und tracheobronchiales Epithel, Blasenepithel, humanes Mundschleimhautepithel und Gewebekulturzellen (Buffalo green monkey und HeLa-Zellen) verwendet. Die Inkubationszeit der Bakterien und der Zellkulturen lag zwischen 0,5 und 2 Stunden. Alle P. m.-Stämme hefteten sich nur sehr schwach an die Nasenepithelzellen vom Schwein und an die anderen untersuchten Zellarten an, worauf auch die Wachstumsbedingungen oder Veränderungen des Anheftungstestes keinen Einfluß hatten. Die durchschnittlichen Anheftungswerte lagen bei 0,07 ± 0,3 Bakterien pro Zelle, während ein Bordetella bronchiseptica-Stamm, welcher als Kontrolle diente, einen mittleren Anheftungswert von 15,3 Bakterien pro Zelle aufwies. Some methods for the isolation of sIgA and to production of anti-sIgA are suggested for establishment of an sIgA determination test. Three methods for sIgA purification from sow milk and three methods for testing its purity are compared. Additionally, anti-sIgA was produced in rabbits. The methods used are described. To investigate the antibody response after local application of lung-pathogenic bacteria, pigs were immunized with viable or inactivated Actinohacillus pleuropneumoniae by the oral and aerogenous route. After 3 weeks class-specific immunoglobulins against purified A. pleuropneumoniae capsular polysaccharides (CP) were determined in scrum and BALF by ELISA. A significant increase of IgA antibodies was found in BALF but not in sera of all immunized pigs. Oral immunization with viable A. pleuropneumoniae and aerosol immunization with either viable or inactivated bacteria resulted in a significant increase of IgG antibodies to the CP antigen in BALF, whereas only aerosol exposure to viable bacteria resulted in a significant increase in IgG antibodies in serum. A significant increase in anti-CP IgM in BALF was observed after aerosol exposure but not after oral immunization. IgM antibodies towards CP increased significantly by both routes of immunization with viable bacteria. The anti-CP activity of all three isolypesin sera and BALF was low in all groups compared with the positive controls, although inoculation of viable A. pleuropneumoniae led to higher levels of antibody concentration than inactivated bacteria. Our results indicate a traffic of primed lymphocytes from the gut into the bronchoalveolar airways and further support the hypothesis that polysaccharide-specific B cells may functionally mature at the mucosal surfaces.
