IntroductionKiwifruit species have a relatively high rate of root oxygen consumption, making them very vulnerable to low root zone oxygen concentrations resulting from soil waterlogging. Recently, kiwifruit rootstocks have been increasingly used to improve biotic and abiotic stress tolerance and crop performance under adverse conditions. The aim of the present study was to evaluate morpho-physiological changes in kiwifruit rootstocks and grafting combinations under short-term waterlogging stress.MethodsA pot trial was conducted at the ALSIA PhenoLab, part of the Phen-Italy infrastructures, using non-destructive RGB and NIR image-based analysis and physiological measurements to identify waterlogging stress indicators and more tolerant genotypes. Three pot-grown kiwifruit rootstocks (‘Bounty 71,’ Actinidia macrosperma—B; ‘D1,’ Actinidia chinensis var. deliciosa—D; and ‘Hayward,’ A. chinensis var. deliciosa—H) and grafting combinations, with a yellow-fleshed kiwifruit cultivar (‘Zesy 002,’ A. chinensis var. chinensis) grafted on each rootstock (Z/B, Z/D, Z/H), were subjected to a control irrigation treatment (WW), restoring their daily water consumption, and to a 9-day waterlogging stress (WL), based on substrate saturation. Leaf gas exchange, photosynthetic activity, leaf temperature, RGB, and NIR data were collected during waterlogging stress.ResultsStomatal conductance and transpiration reached very low values (less than 0.05 mol m−2 s−1 and 1 mmol m−2 s−1, respectively) in both waterlogged D and H rootstocks and their grafting combinations. In turn, leaf temperature was significantly increased and photosynthesis was reduced (1–6 μmol m−2 s−1) from the first days of waterlogging stress compared to B rootstock and combination.DiscussionThe B rootstock showed prolonged leaf gas exchange and photosynthetic activity, indicating that it can cope with short-term and temporary waterlogging and improve the tolerance of grafted kiwi vines, which showed a decrease in stomatal conductance 5 days after the onset of stress. Morphometric and colorimetric parameters from the image-based analysis confirmed the greater susceptibility of D and H rootstocks and their grafting combinations to waterlogging stress compared to B. The results presented confirm the role of physiological measurements and enhance that of RGB and NIR images in detecting the occurrence of water stress and identifying more tolerant genotypes in kiwifruit.

Colorectal cancer (CRC) is a major public health concern and identifying prognostic molecular biomarkers can help stratify patients based on risk profiles, thus enabling personalized medicine. Epitranscriptomic modifications play a relevant role in controlling gene expression, N6-methyladenosine (m6A) regulators play crucial roles in cancer progression, but their clinical significance in CRC cancer has thus far not been elucidated. Thus, we aimed to examine by immunohistochemical techniques and RT-qPCR, protein levels and RNAs expression of m6A writers (METTL3, WTAP) and eraser (FTO) in a cohort of 10 patients affected by CRC. The patients were followed for 5 years and values of METTL3, WTAP and FTO RNAs in alive vs dead patients were compared. Proteins expression and RNAs expression had a different trend, METTL3, WTAP and FTO proteins’ expression showed an increasing trend from non-cancerous adjacent (N) tissue vs carcinoma (CA) tissue G1 stage, and then a decreasing trend from G1 to G2 and G3 stages. The most marked increase was observed in WTAP that, from a 40% of protein expression positivity in N tissue raised to the 81% of positivity in G1 stage K tissue. RNAs expression of METTL3, WTAP and FTO genes in N tissue vs G1 stage CA tissue was significantly different, the analysis and comparison of RNAs values in patient alive after 5 years (0.58±0.04) vs patients dead after 5 years (1.69±0.29) showed that only WTAP values resulted significantly high in dead patients. The fact that WTAP protein expression levels lower while WTAP RNA expression remains high, lets us hypothesize a sort of inhibition of protein expression, but further studies are needed to clarify the mechanism. Although the results suggest a relationship between biological meaning and prognostic utility of WTAP, this prognostic utility must be confirmed by further studies on a larger sample.

Aims In treating patients with melanoma, the order in which therapy is administered, choosing between targeted therapy and immune checkpoint inhibition, has garnered growing interest.Patients and Methods We conducted a retrospective, real-world analysis of patients with advanced melanoma undergoing immunotherapy or targeted therapy as first-line at a single center.Results A total of 88 patients diagnosed with melanoma were identified. At 7 years, in this cohort, 68.4% (95% CI: 55.9%-83.6%) of patients were alive. In all, 47 tumors harbored BRAF mutations; 10 patients who did not receive therapy were excluded from this subgroup. Of the 37 patients with a BRAF mutation, 29 received first-line targeted therapy and 8 received first-line immunotherapy. At 2 years, 28 (76%) patients were alive and 9 (24%) had died. Of the 28 survivors, 22 received first-line targeted therapy and 6 received first-line immunotherapy. In addition, 29 patients were administered a MEK inhibitor in first line. Of these, 66.4% (95% CI: 48.3–91.2) of patients were alive at 7 years.Conclusions There was no significant difference between survival and first-line immunotherapy or first-line targeted therapy. Additional studies are required to establish whether front-line immunotherapy is linked to more effective long-term disease control compared to first-line targeted therapy.

Objective: To compare 12-month post-treatment health-related quality of life (HR-QoL) and symptom severity (SS) changes among patients with symptomatic uterine fibroids (SUF) not seeking fertility and undergo a hysterectomy, abdominal myomectomy (AM), or uterine artery embolization (UAE). Materials and Methods: The Comparing Options for Management: Patient-Centered Results for Uterine Fibroids (COMPARE-UF) Registry is a multi-institutional prospective observational cohort study of patients treated for SUF. A subset of 1465 women 31-45 years of age, who underwent either hysterectomy (n = 741), AM (n = 446), or UAE (n = 155) were included in this analysis. Demographics, fibroid history, and symptoms were obtained by baseline questionnaires and at 1 year post-treatment. Results were stratified by all treatments and propensity score weighting to adjust for differences in baseline characteristics. Results: Women undergoing UAE reported the lowest baseline HR-QoL and highest SS scores (mean = 40.6 [standard deviation (SD) = 23.8]; 62.3 [SD = 24.2]) followed by hysterectomy (44.3 [24.3]; 59.8 [SD = 24.1]). At 12 months, women who underwent a hysterectomy experienced the largest change in both HR-QoL (48.7 [26.2]) and SS (51.9 [25.6]) followed by other uterine-sparing treatments. Propensity score weighting revealed all treatments produced substantial improvement, with hysterectomy patients reporting the highest HR-QoL score (92.0 [17.8]) compared with myomectomy (86.7 [17.2]) and UAE (82.6 [21.5]) (p

The paper focuses on the seasonal oil accumulation in traditional and super-high density (SHD) olive plantations and its modelling employing image-based linear models. For these purposes, at 7-10-day intervals, fruit samples (cultivar Arbequina, Fasola, Frantoio, Koroneiki, Leccino, Maiatica) were pictured and images segmented to extract the Red (R), Green (G), and Blue (B) mean pixel values which were re-arranged in 35 RGB-derived colorimetric indexes (CIs). After imaging, the samples were crushed and oil concentration was determined (NIR). The analysis of the correlation between oil and CIs revealed a differential hysteretic behavior depending on the covariates (CI and cultivar). The hysteresis area (Hyst) was then quantified and used to rank the CIs under the hypothesis that CIs with the maximum or minimum Hyst had the highest correlation coefficient and were the most suitable predictors within a general linear model. The results show that the predictors selected according to Hyst-based criteria had high accuracy as determined using a Global Performance Indicator (GPI) accounting for various performance metrics (R2, RSME, MAE). The use of a general linear model here presented is a new computational option integrating current methods mostly based on artificial neural networks. RGB-based image phenotyping can effectively predict key quality traits in olive fruit supporting the transition of the olive sector towards a digital agriculture domain.

Grapevine is among the most economically important crops suffering environmental constraints, including drought and salt stress. Although imaging is increasingly used to detect abiotic stress in agriculture, image-based phenotyping in grapevine still needs optimisation. This study presents the RGB-(red, green, blue)-based phenotyping of the early stage of salt stress response in potted grapevine (Aleatico/SO4) irrigated with saline water (100 mM NaCl) for 9 days in contrast with vines irrigated with fresh water. The response was measured using stomatal conductance (gs), net photosynthetic rate (A), transpiration (E), maximum potential photosynthetic efficiency (Fv/Fm), stem water potential (SWP) concurrently with RGB imaging via a robotised platform. The image-based phenotyping of salt-stressed vines employed two sets of measurements: (i) the pixel fraction of specific colour bands (Yellow, Green, Brown and Dark Green) and (ii) the mean pixel value of R, G and B and other RGB-based colorimetric indexes. Results show that the responses of gs, A, E, Fv/Fm were closely related to increasing soil electrical conductivity (EC) and that imaging could detect the EC threshold of approx. 4 dS m-1 causing a ~60 % decrease in these physiological traits compared to the pre-stress level. The SWP declined to about –0.7 MPa at the end of the experiment. The change of the relative pixel fraction of Dark Green to increasing EC has been analysed within a dose-response context, showing that a decrease of 1 % of the Dark Green colour band corresponded to the 4 dS m-1 EC threshold. This study also examined the use of the mean pixel value of the R, G and B channels as proxies of EC along with new RGB-based indexes resulting from the rearrangement of original R, G and B mean pixel values. Results show the suitability of the mean pixel value of R and Coloration Index [(R-B)/R] to serve as predictors of EC (R2 >= 0.80).

Aiding optimal plant–environment interaction would favor plant resilience against environmental constrains including salt stress. We test the hypothesis that 6-Benzylaminopurine (BAP) primes grapevine’s salt tolerance in vines (Vitis vinifera) received salt water (NaCl 100 mM) through the modulation of gene expression of BAP (AHK4, AHP1) and salt-stress (CAT, APX) inducible genes and morpho-physiological traits. A subgroup of vines had previously (48 h) been primed with BAP (80 mg/L) before salt stress. The gene expressions were 30% (CAT) and 56% (APX) lower in primed salt-stressed vines than that in un-primed. Salt treatment did not increase leaf Na+ but it lowered stomatal conductance (gs), photosynthesis (A), stem water potential (less negative) and photosystem-II efficiency (Fv/Fm). Chlorophyll-a concentrations were 30% higher in BAP-primed compared to un-primed. Adverse effects of salt were significantly reduced, maintaining high A/gs, Fv/Fm and growth. After the relief of the stress, the BAP primed vines had a fast recovery.

Abstract X‐ray direct detectors based on hybrid lead‐halide perovskite have seen a dramatic increase of interest in the last years. A rush for the achievement of high performing devices drives the scientific community. In this context, several photoconductor sensors employ functional layers to increase the gain effect, but the full comprehension of the mechanism is still lacking. Here, X‐ray nanoanalysis is used, performed by simultaneous acquisition of X‐ray Fluorescence and X‐ray Beam Induced Current maps, to investigate at the nanoscale level the role of [6,6]‐phenyl‐C61‐butyric acid methyl ester fullerene (PCBM) molecules when interacting with MAPbI3 polycrystalline thin films acting as photo‐conductors in X‐ray detectors. At the device‐scale level it shows that the addition of PCBM enhances the X‐ray sensitivity by four times. At the nanoscale level how the perovskite grain boundaries act as high photocurrent generation centers is demonstrated. The addition of the PCBM increases the photocurrent generation, as the macroscopic performance does, and the charge collection becomes uniform over the full crystallite volume. The results clarify the role of grain boundaries and charge selecting layers and establish the X‐ray nanoanalysis techniques as a powerful tool to investigate charge transport and collection in perovskite films.

Oxidative stress represents a hallmark for many degenerative pathologies of the Central Nervous System. Throughout life, the constant pressure of noxious stimuli and/or episodes of traumatic events may expose the brain to a microenvironment where the non-balanced reactive oxygen species inevitably lead to neuronal loss and cognitive decline. HO-1, a 32 kDa heat-shock protein catalyzing the degradation of heme into carbon monoxide (CO), iron and biliverdin/bilirubin is considered one of the main antioxidant defense mechanisms playing pivotal roles in neuroprotection. Restoring the redox homeostasis is the goal of many natural or synthetic antioxidant molecules pursuing beneficial effects on brain functions. Here, we investigated the antioxidant capacity of four selected benzofuran-2-one derivatives in a cellular model of neurodegeneration represented by differentiated SH-SY5Y cells exposed to catechol-induced oxidative stress. Our main results highlight how all the molecules have antioxidant properties, especially compound 9, showing great abilities in reducing intracellular ROS levels and protecting differentiated SH-SY5Y cells from catechol-induced death. This compound above all seems to boost HO-1 mRNA and perinuclear HO-1 protein isoform expression when cells are exposed to the oxidative insult. Our findings open the way to consider benzofuran-2-ones as a novel and promising adjuvant antioxidant strategy for many neurodegenerative disorders.

Formamidinium lead iodide (FAPbI3) is the benchmark material for the most efficient near‐infrared perovskite light‐emitting diodes (LEDs) and a promising gain medium for perovskite‐based coherent light sources. Thus, it is crucial to understand and control its defect chemistry to harness the full potential of its exceptional radiative recombination properties. Here, this topic is addressed by tailoring the I− to Br− ratio in the perovskite composition. It is found that introducing small Br− quantities improves the yield of radiative recombination with a beneficial impact on both spontaneous and amplified spontaneous emission (ASE) and improves the semiconductor photostability leading to reduced luminescence efficiency roll‐off and enhanced radiance in LEDs. By employing photoemission electron microscopy (PEEM), this improvement in optoelectronic performance can be directly correlated to a reduced hole‐trapping activity achieved by replacing iodide with bromide, thus, providing a convenient yet powerful synthetic approach to control the defect chemistry of the material and fostering its implementation in advanced photonic platforms. To predict oil and phenol concentrations in olive fruit, the combination of back propagation neural networks (BPNNs) and contact-less plant phenotyping techniques was employed to retrieve RGB image-based digital proxies of oil and phenol concentrations. Fruits of cultivars (×3) differing in ripening time were sampled (~10-day interval, ×2 years), pictured and analyzed for phenol and oil concentrations. Prior to this, fruit samples were pictured and images were segmented to extract the red (R), green (G), and blue (B) mean pixel values that were rearranged in 35 RGB-based colorimetric indexes. Three BPNNs were designed using as input variables (a) the original 35 RGB indexes, (b) the scores of principal components after a principal component analysis (PCA) pre-processing of those indexes, and (c) a reduced number (28) of the RGB indexes achieved after a sparse PCA. The results show that the predictions reached the highest mean R2 values ranging from 0.87 to 0.95 (oil) and from 0.81 to 0.90 (phenols) across the BPNNs. In addition to the R2, other performance metrics were calculated (root mean squared error and mean absolute error) and combined into a general performance indicator (GPI). The resulting rank of the GPI suggests that a BPNN with a specific topology might be designed for cultivars grouped according to their ripening period. The present study documented that an RGB-based image phenotyping can effectively predict key quality traits in olive fruit supporting the developing olive sector within a digital agriculture domain.

Basal cell carcinoma (BCC) is a skin cancer with low local aggressiveness and a low tendency to metastasize. Basosquamous Carcinoma (BSC) represents an aggressive histological subtype of BCC with intermediate features between Squamous Cell Carcinoma (SCC) and BCC. Cemiplimab is currently approved as first-line therapy in SCC and second-line therapy in BCC patients who have progressed on or are intolerant of a Hedgehog pathway Inhibitor (HHI). Our study describes the case of a 59-year-old man with BSC who was successfully treated with 5 cycles of Cemiplimab as first-line therapy and Sonidegib as second-line therapy. Currently, the efficacy of Cemiplimab against BSC and other histopathological subtypes of BCC has not been fully elucidated, as has the role of sequential or combination therapy with Cemiplimab and HHI in the management of BSC. The aim of this case report is to highlight the need to outline the use of checkpoint inhibitors in BCCs and focus attention on the synergistic role of Cemiplimab and HHIs in such a controversial entity as BSC.
