41. Enabling Visible Light Sensitization of Yb III , Nd III and Er III in Dimeric Ln III /Ga III Metallacrowns through Functionalization with Ru II Complexes for NIR-II Multiplex Imaging.

Abstract

Multiplex imaging in the second near-infrared window (NIR-II, 1000-1700 nm) provides exciting opportunities for more precise understanding of biological processes and more accurate diagnosis of diseases by enabling real-time acquisition of images with improved contrast and spatial resolution in deeper tissues. Today, the number of imaging agents suitable for this modality remains very scarce. In this work, we have synthesized and fully characterized, including theoretical calculations, a series of dimeric Ln III /Ga III metallacrowns bearing Ru II polypyridyl complexes, LnRu-3 (Ln=Y III , Yb III , Nd III , Er III ). Relaxed structures of YRu-3 in the ground and the excited electronic states have been calculated using dispersion-corrected density functional theory methods. Detailed photophysical studies of LnRu-3 have demonstrated that characteristic emission signals of Yb III , Nd III and Er III in the NIR-II range can be sensitized upon excitation in the visible range through Ru II -centered metal-to-ligand charge transfer (MLCT) states. We have also showed that these NIR-II signals are unambiguously detected in an imaging experiment using capillaries and biological tissue-mimicking phantoms. 42. Investigation of Ln 3+ complexation by a DOTA derivative substituted by an imidazothiadiazole: synthesis, solution structure, luminescence and relaxation properties.

Abstract

We investigated the coordination properties of original macrocyclic Ln 3+ complexes comprising an imidazothiadiazole heterocycle. The thermodynamic stability of the Gd 3+ complex was determined by a combination of potentiometric and photophysical measurements. The kinetic inertness was assessed in highly acidic media. The solution structure of the Ln 3+ complex was unambiguously determined by a set of photophysical measurements and 1 H, 13 C, 89 Y NMR data in combination with DFT calculations, which proved coordination of the heterocycle to Ln 3+ . The ability of the imidazothiadiazole moiety to sensitize Tb 3+ luminescence was investigated. Finally, the relaxation properties were investigated by recording 1 H nuclear magnetic relaxation dispersion (NMRD) profiles and 17 O measurements. The water exchange rate is similar to that of GdDOTA as the less negative charge of the ligand is compensated for by the presence of a bulky heterocycle. Relaxivity is constant over a large range of pH values, demonstrating the favorable properties of the complex for imaging purposes.

43. A Dual-Mode Near-Infrared Optical and Photoacoustic Imaging Agent Based on a Low Energy Absorbing Ytterbium Complex.

Abstract

Near-infrared (NIR) luminescence and photoacoustic (PA) imaging have attracted increasing attention for the real-time monitoring of biological samples due to high sensitivity, resolution, and pronounced signal detection depth, respectively. For improved contrast, both techniques require imaging agents possessing high absorption in the red-NIR range. Herein, we took advantage of a ternary complex formed with the anionic ytterbium(III) tetrakis (2-thenoyltrifluoroacetonate) ([Yb(tta) 4 ] - ) and the cationic NIR-absorbing chromophore, 1,1'-diethyl-2,2'-dicarbocyanine (Cy + ), to evaluate its potential to act as a dual-mode NIR luminescence and PA imaging agent. We demonstrated that, upon excitation with red-NIR light, Cy[Yb(tta) 4 ] encapsulated into polystyrene nanoparticles is able to generate both NIR Yb 3+ emission and a PA signal in an imaging experiment performed in a tissue-mimicking phantom.

44. Tuning white light emission using single-component tetrachroic Dy 3+ metallacrowns: the role of chromophoric building blocks.

Abstract

White light production is of major importance for ambient lighting and technological displays. White light can be obtained by several types of materials and their combinations, but single component emitters remain rare and desirable towards thinner devices that are, therefore, easier to control and that require fewer manufacturing steps. We have designed a series of dysprosium(iii)-based luminescent metallacrowns (MCs) to achieve this goal. The synthesized MCs possess three main structural types LnGa 4 (L') 4 (L'') 4 (type A), Ln 2 Ga 8 (L') 8 (L''') 4 (type B) and LnGa 8 (L') 8 (OH) 4 (type C) (H 3 L', HL'' and H 2 L''' derivatives of salicylhydroxamic, benzoic and isophthalic acids, respectively). The advantage of these MCs is that, within each structural type, the nature of the organic building blocks does not affect the symmetry around Dy 3+ . By detailed studies of the photophysical properties of these Dy 3+ -based MCs, we have demonstrated that CIE coordinates can be tuned from warm to neutral to cold white by (i) defining the symmetry about Dy 3+ , and (ii) choosing appropriate chromophoric building blocks. These organic building blocks, without altering the coordination geometry around Dy 3+ , influence the total emission profile through changing the probability of different energy transfer processes including the 3 T 1 ← Dy 3+ * energy back transfer and/or by generating ligand-centered fluorescence in the blue range. 45. Multi-Technique Characterization of 3.45 Ga Microfossils on Earth: A Key Approach to Detect Possible Traces of Life in Returned Samples from Mars.

Abstract

The NASA Mars 2020 Perseverance rover is actively exploring Jezero crater to conduct analyses on igneous and sedimentary rock targets from outcrops located on the crater floor (Máaz and Séítah formations) and from the delta deposits, respectively. The rock samples collected during this mission will be recovered during the Mars Sample Return mission, which plans to bring samples back to Earth in the 2030s to conduct in-depth studies using sophisticated laboratory instrumentation. Some of these samples may contain traces of ancient martian life that may be particularly difficult to detect and characterize because of their morphological simplicity and subtle biogeochemical expressions. Using the volcanic sediments of the 3.45 Ga Kitty's Gap Chert (Pilbara, Australia), containing putative early life forms (chemolithotrophs) and considered as astrobiological analogues for potential early Mars organisms, we document the steps required to demonstrate the syngenicity and biogenicity of such biosignatures using multiple complementary analytical techniques to provide information at different scales of observation. These include sedimentological, petrological, mineralogical, and geochemical analyses to demonstrate macro- to microscale habitability. New approaches, some unavailable at the time of the original description of these features, are used to verify the syngenicity and biogenicity of the purported fossil chemolithotrophs. The combination of elemental (proton-induced X-ray emission spectrometry) and molecular (deep-ultraviolet and Fourier transform infrared) analyses of rock slabs, thin sections, and focused ion beam sections reveals that the carbonaceous matter present in the samples is enriched in trace metals ( e.g. , V, Cr, Fe, Co) and is associated with aromatic and aliphatic molecules, which strongly support its biological origin. Transmission electron microscopy observations of the carbonaceous matter documented an amorphous nanostructure interpreted to correspond to the degraded remains of microorganisms and their by-products (extracellular polymeric substances, filaments…). Nevertheless, a small fraction of carbonaceous particles has signatures that are more metamorphosed. They probably represent either reworked detrital biological or abiotic fragments of mantle origin. This study serves as an example of the analytical protocol that would be needed to optimize the detection of fossil traces of life in martian rocks.

Abstract

By combining advantages of two series of lanthanide(III)/zinc(II) metallacrowns (MCs) assembled using pyrazine- (pyzHA 2- ) and quinoxaline- (quinoHA 2- ) hydroximate building blocks ligands, we created here water-soluble mixed-ligand MCs with extended absorption to the visible range. The Yb III analogue demonstrated improved photophysical properties in the near-infrared (NIR) range in cell culture media, facilitating its application for NIR optical imaging in living HeLa cells., (© 2023 The Authors. Abstract

The effect of ligands' energy levels on thermal dependence of lanthanide emission was examined to create new molecular nanothermometers. A series of Ln 2 Ga 8 L 8 ' L 8 ″ metallacrowns (shorthand Ln 2 L 8 ' ), where Ln = Gd 3+ , Tb 3+ , or Sm 3+ (H 3 L' = salicylhydroxamic acid (H 3 shi), 5-methylsalicylhydroxamic acid (H 3 mshi), 5-methoxysalicylhydroxamic acid (H 3 iph), was synthesized and characterized. Within the series, ligand-centered singlet state (S 3 nha)) and H 2 L″ = isophthalic acid (H 2 iph), was synthesized and characterized. Within the series, ligand-centered singlet state (S 1 ) energy levels ranged from 23,300 to 27,800 cm -1 , while triplet (T 1 ) energy levels ranged from 18,150 to 21,980 cm -1 . We demonstrated that the difference between T 1 levels and relevant energies of the excited 4 G 5/2 level of Sm 3+ (17,800 cm -1 ) and 5 ) is the major parameter controlling thermal dependence of the emission intensity via the back energy transfer mechanism. However, when the energy difference between S 4 level of Tb 3+ (20,400 cm -1 ), the S 1 and T 1 intersystem crossing (and its reverse, S -1 ), the S 1 → T 1 intersystem crossing (and its reverse, S 1 ← T 1 ) mechanism contributes to the thermal behavior of metallacrowns. Both mechanisms affect Ln 3+ -centered room-temperature quantum yields with values ranging from 2.07(6)% to 31.2(2)% for Tb 2 L 8 ' and from 0.0267(7)% to 2.27(5)% for Sm 2 L 8 ' . The maximal thermal dependence varies over a wide thermal range (ca. 150-350 K) based on energy gaps between relevant ligand-based and lanthanide-based electronic states. By mixing Tb 2 moshi 8 ' with Sm 2 moshi 8 ' in a 1:1 ratio, an optical thermometer with a relative thermal sensitivity larger than 3%/K at 225 K was created. Other temperature ranges are also accessible with this approach.

Abstract

A set of (Ln[14-MC Zn(II)N(quinHA) -5]) 2 Ln 2 Zn 2 (quinHA) 2 (ph) 2 (Hph) 4 (OH) 8 (H 2 O) 4 metallacrowns ( Ln-1 , Ln = Tb, Gd, or Yb; H 2 quinHA = quinaldic hydroxamic acid, H 2 ph = phthalic acid) have been synthesized via solution-state self-assembly. The metallacrowns possess an uncommon topology within the metallacrown family where two rarely seen 14-metallacrown-5 moieties are fused by a Yb 2 Zn 2 (quinHA) 2 bridge. Moreover, Yb-1 analyzed in the solid state exhibits a characteristic near-infrared luminescence signal arising from Yb 3+ 2 F 5/2 → 2 F 7/2 transition despite the proximity of high energy O-H oscillators.

Abstract

The luminescence properties of two types of heterotrimetallic aluminum-lanthanide-sodium 12-metallacrown-4 compounds are presented here, LnNa(ben) 4 [12-MC Al(III)N(shi) -4] (LnAl4Na) and {LnNa[12-MC Al(III)N(shi) -4]} 2 (iph) 4 (Ln2Al8Na2), where Ln = Gd III , Tb III , Er III , and Yb III , MC is metallacrown, ben - is benzoate, shi 3- is salicylhydroximate, and iph 2- is isophthalate. The aluminum-lanthanide-sodium metallacrowns formed with benzoate are discrete monomers while, upon replacement of the benzoate with the dicarboxylate isophthalate, two individual metallacrowns can be joined to form a dimer. In the solid state, the terbium version of each structure type displays emission in the visible region, and the erbium and ytterbium complexes emit in the near-infrared. The luminescence lifetimes ( τ obs ) and quantum yields have been collected under ligand excitation ( Q LLn) for both LnAl4Na monomers and Ln2Al8Na2 dimers. Several of these values tend to be shorter (luminescence lifetimes) and smaller (quantum yields) than the corresponding values recorded for the structurally similar gallium-lanthanide monomer and dimer 12-MC-4 molecules. However, the quantum yield value recorded for the visible emitting Tb2Al8Na2 dimer, 43.9%, is the highest value observed in the solid state to date for a Tb III based metallacrown.

Abstract

Today, there is a very strong demand for versatile near-infrared (NIR) imaging agents suitable for non-invasive optical imaging in living organisms ( in vivo imaging). Here, we created a family of NIR-emitting macromolecules that take advantage of the unique structure of dendrimers. In contrast to existing fluorescent dendrimers bearing fluorophores at their periphery or in their cavities, a NIR fluorescent structure is incorporated into the core of the dendrimer. Using the poly(amidoamine) dendrimer structure, we want to promote the biocompatibility of the NIR-emissive system and to have functional groups available at the periphery to obtain specific biological functionalities such as the ability to deliver drugs or for targeting a biological location. We report here the divergent synthesis and characterization by NMR and mass spectrometries of poly(amidoamine) dendrimers derived from the fluorescent NIR-emitting anthraquinone core ( AQ-PAMAF ). AQ-PAMAFs ranging from the generation -0.5 up to 3 were synthesized with a good level of control resulting in homogeneous and complete dendrimers. Absorption, excitation, and emission spectra, as well as quantum yields, of AQ-PAMAFs have been determined in aqueous solutions and compared with the corresponding properties of the AQ-core . It has been demonstrated that the absorption bands of AQ-PAMAFs range from UV to 750 nm while emission is observed in the range of 650-950 nm. Fluorescence macroscopy experiments confirmed that the NIR signal of AQ-PAMAFs can be detected with a satisfactory signal-to-noise ratio in aqueous solution, in blood, and through 1 mm thick tissue-mimicking phantom. The results show that our approach is highly promising for the design of an unprecedented generation of versatile NIR-emitting agents.
