Injectable and in situ photocurable biomaterials are receiving a lot of attention due to their ease of application via syringe or dedicated applicator and ability to be used in laparoscopic and robotic minimally invasive procedures. The aim of this work was to synthesize photocurable ester-urethane macromonomers using a heterometallic magnesium-titanium catalyst, magnesium-titanium (IV) butoxide for elastomeric polymer networks. The progress of the two-step synthesis of macromonomers was monitored using infrared spectroscopy. The chemical structure and molecular weight of the obtained macromonomers were characterized using nuclear magnetic resonance spectroscopy and gel permeation chromatography. The dynamic viscosity of the obtained macromonomers was evaluated by a rheometer. Next, the photocuring process was studied under both air and argon atmospheres. Both the thermal and dynamic mechanical thermal properties of the photocured soft and elastomeric networks were investigated. Finally, in vitro cytotoxicity screening of polymer networks based on ISO10993-5 revealed high cell viability (over 77%) regardless of curing atmosphere. Overall, our results indicate that this heterometallic magnesium-titanium butoxide catalyst can be an attractive alternative to commonly used homometallic catalysts for the synthesis of injectable and photocurable materials for medical applications.

Bacterial biofilms generally contribute to chronic infections, including wound infections. Due to the antibiotic resistance mechanisms protecting bacteria living in the biofilm, they are a serious problem in the wound healing process. To accelerate the wound healing process and avoid bacterial infection, it is necessary to select the appropriate dressing material. In this study, the promising therapeutic properties of alginate lyase (AlgL) immobilised on BC membranes for protecting wounds from Pseudomonas aeruginosa infection were investigated. The AlgL was immobilised on never dried BC pellicles via physical adsorption. The maximum adsorption capacity of AlgL was 6.0 mg/g of dry BC, and the equilibrium was reached after 2 h. The adsorption kinetics was studied, and it has been proven that the adsorption was consistent with Langmuir isotherm. In addition, the impact of enzyme immobilisation on bacterial biofilm stability and the effect of simultaneous immobilisation of AlgL and gentamicin on the viability of bacterial cells was investigated. The obtained results showed that the AlgL immobilisation significantly reduced the amount of polysaccharides component of the P. aeruginosa biofilm. Moreover, the biofilm disruption by AlgL immobilised on BC membranes exhibited synergism with the gentamicin, resulting in 86.5% more dead P. aeruginosa PAO-1 cells.

The versatility and unique properties of bacterial cellulose (BC) motivate research into enhancing its synthesis. Here a silicone polyether surfactant (SPS) was synthesized and tested as a non-nutritional additive to the cultivation media of Komagataeibacter xylinus. The addition of SPS to the Hestrin-Schramm (HS) medium resulted in a concentration-dependent decrease in surface tension from 59.57 ± 0.37 mN/m to 30.05 ± 0.41 mN/m (for 0.1% addition) that was correlated with an increased yield of BC, up to 37% wet mass for surfactant concentration close to its critical micelle concentration (0.008%). Physicochemical characterization of bacterial cellulose obtained in presence of SPS, showed that surfactant is not incorporated into BC structure and has a moderate effect on its crystallinity, thermal stability. Moreover, the water holding capacity was enhanced by over 40%. Importantly, obtained BC did not affect L929 murine fibroblast cell viability. We conclude that SPS provides an eco-friendly approach to increasing BC yield in static culture, enabling more widespread industrial and biomedical applications.

Due to the global spread of the SARS-CoV-2 virus and the resultant pandemic, there has been a major surge in the demand for surgical masks, respirators, and other air filtration devices. Unfortunately, the fact that these filters are made of petrochemical-derived, non-biodegradable polymers means that the surge in production has also led to a surge in plastic waste. In this work, we present novel, sustainable filters based on bacterial cellulose (BC) functionalized with low-pressure argon plasma (LPP-Ar). The “green” production process involved BC biosynthesis by Komagataeibacter xylinus, followed by simple purification, homogenization, lyophilization, and finally LPP-Ar treatment. The obtained LPP-Ar-functionalized BC-based material (LPP-Ar-BC-bM) showed excellent antimicrobial and antiviral properties, with no cytotoxicity versus murine fibroblasts in vitro. Further, filters consisting of three layers of LPP-Ar-BC-bM had >99% bacterial and viral filtration efficiency, while maintaining sufficiently low airflow resistance (6 mbar at an airflow of 95 L/min). Finally, as a proof-of-concept, we were able to prepare 80 masks with LPP-Ar-BC-bM filter and ~85% of volunteer medical staff assessed them as good or very good in terms of comfort. We conclude that our novel sustainable, biobased, biodegradable filters are suitable for respiratory personal protective equipment (PPE), such as surgical masks and respirators. Further, with scale-up, they may be adapted for indoor air handling filtration in hospitals or schools.Graphical abstract

Minimally invasive surgical approaches require the development of new injectable materials. A particularly attractive strategy involves using photocurable materials that can be delivered as liquids and rapidly cured in situ using UV light. The aim of this work was to synthesize and characterize fatty-acid-derived ester-urethane telechelic (methacrylate) macromonomers, suitable for photopolymerization. The commonly used dibutyltin dilaurate catalyst was replaced with bismuth neodecanoate, bismuth tris(2-ethylhexanoate), and zinc (II) acetyloacetonate as less-toxic alternative catalysts. Additionally, ethyl acetate was used as a “green” solvent. The progress of the two-step synthesis was monitored with infrared spectroscopy. The chemical structure and molecular weight of the obtained viscous materials was characterized with nuclear magnetic resonance spectroscopy and gel permeation chromatography. Photocrosslinking of the macromonomers into elastomeric films was achieved using 150 s per spot of UV light (20 mW/cm2) exposure. Mechanical tensile testing of the films confirmed tensile strength and modulus values in a range of values typical of soft tissue. Finally, in vitro cytotoxicity tests showed high cell viability for the case of materials synthesized using bismuth and zinc catalysts. Overall, our results indicate that bismuth and zinc catalysts are excellent alternatives to organotin compounds in the synthesis of photocurable methacrylate ester-urethanes for biomedical applications, such as minimally invasive surgical protocols.

In this work, we verified the possibility of valorizing a major waste product of the potato starch industry, potato tuber juice (PJ). We obtained a cost-effective, ecological-friendly microbiological medium that yielded bacterial cellulose (BC) with properties equivalent to those from conventional commercial Hestrin–Schramm medium. The BC yield from the PJ medium (&gt, 4 g/L) was comparable, despite the lack of any pre-treatment. Likewise, the macro- and microstructure, physicochemical parameters, and chemical composition showed no significant differences between PJ and control BC. Importantly, the BC obtained from PJ was not cytotoxic against fibroblast cell line L929 in vitro and did not contain any hard-to-remove impurities. The PJ-BC soaked with antiseptic exerted a similar antimicrobial effect against Staphylococcus aureus and Pseudomonas aeruginosa as to BC obtained in the conventional medium and supplemented with antiseptic. These are very important aspects from an application standpoint, particularly in biomedicine. Therefore, we conclude that using PJ for BC biosynthesis is a path toward significant valorization of an environmentally problematic waste product of the starch industry, but also toward a significant drop in BC production costs, enabling wider application of this biopolymer in biomedicine.

In this work, we present novel ex situ modification of bacterial cellulose (BC) polymer, that significantly improves its ability to absorb water after drying. The method involves a single inexpensive and easy-to-perform process of BC crosslinking, using citric acid along with catalysts, such as disodium phosphate, sodium bicarbonate, ammonium bicarbonate or their mixtures. In particular, the mixture of disodium phosphate and sodium bicarbonate was the most promising, yielding significantly greater water capacity (over 5 times higher as compared to the unmodified BC) and slower water release (over 6 times as compared to the unmodified BC). Further, our optimized crosslinked BC had over 1.5x higher water capacity than modern commercial dressings dedicated to highly exuding wounds, while exhibiting no cytotoxic effects against fibroblast cell line L929 in vitro. Therefore, our novel BC biomaterial may find application in super-absorbent dressings, designed for chronic wounds with imbalanced moisture level.

Bacterial cellulose (BC) is recognized as a wound dressing material well-suited for chronic wounds; however, it has no intrinsic antimicrobial activity. Further, the formation of biofilms can limit the effectiveness of the pre-saturation of BC with antimicrobial agents. Bioactive films find more and more applications in various industries, including packaging and biomedicine. This work describes the preparation, characterization and physicochemical, antioxidant and antimicrobial properties of alginate films modified with melanin from watermelon (Citrullus lanatus) seeds at concentrations of 0.10%, 0.25% and 0.50% w/w and with silver and zinc oxide nanoparticles (10 mM film casting solutions for both metal nanoparticles). Melanin served as the active ingredient of the film and as a nanoparticle stabilizer. The additives affected the color, antioxidant (~90% ABTS and DPPH radicals scavenging for all melanin modified films) and antimicrobial activity (up to 4 mm grow inhibition zones of E. coli and S. aureus for both zinc oxide and silver nanoparticles), mechanical (silver nanoparticles addition effected two-fold higher tensile strength), thermal and barrier properties for water and UV-vis radiation. The addition of ZnONP resulted in improved UV barrier properties while maintaining good visible light transmittance, whereas AgNP resulted in almost complete UV barrier and reduced visible light transmittance of the obtained films. What is more, the obtained films did not have an adverse effect on cell viability in cytotoxicity screening. These films may have potential applications in food packaging or biomedical applications.

We present an ink platform for a printable polymer–graphene nanocomposite that is intended for the development of modular biosensors. The ink consists of catechol-modified chitosan and reduced graphene oxide decorated with platinum nanoparticles (rGO–Pt). We modified the chitosan with catechol groups, in order to obtain adhesive properties and improve solubility. Dispersions of rGO–Pt in ethylene glycol were admixed with an aqueous solution of modified chitosan to yield an ink that is suitable for non-contact piezoelectric printing using a commercial microplotter (Sonoplot GIX Microplotter Desktop). As a proof of concept, printed patterns were biofunctionalized with DNA oligonucleotide probes for Streptococcus agalactiae (Group B streptococcus) using glutaraldehyde as a linker. Confocal microscopy revealed the successful hybridization of complementary polymerase chain reaction (PCR) products and low non-specific binding. Our results demonstrate that catechol-modified chitosan/rGO–Pt nanocomposites can be used as inks for piezoelectric printing and facilitate the attachment of biorecognition elements for biosensor applications.

The aim of present study was to determine the hemocompatibility, cellular response of endothelial cells and bacterial adhesion to a new polyester nanocomposite. The carbon nanoparticle nanocomposite was prepared via in situ polymerization of monomers to obtain material of hardness 55 Sh D similar to polyurethanes used in medical applications, for example, in heart-assisting devices. The carbon nanoparticle-containing polyester exhibits markedly reduced bacterial colonization, as compared to commercially available polyurethanes. Further the nanocomposite possesses markedly improved hemocompatibility, as determined by flow cytometry, and robust endothelialization. Possible explanations for these beneficial properties include surface nanoroughness of carbon nanoparticle-containing nanocomposites and presence of fatty acid sequences within polymer structure.

Despite on-going medical advances, ovarian cancer survival rates have stagnated. In order to improve IP delivery of platinum-based antineoplastics, we aimed to develop a sustained drug delivery system for carboplatin (CPt). Toward this aim, we pursued a double emulsion process for obtaining CPt-loaded microcapsules composed of poly(ethylene terephthalate-ethylene dilinoleate) (PET-DLA) copolymer. We were able to obtain PET-DLA microspheres in the targeted size range of 10&ndash, 25 &micro, m (median: 18.5 &micro, m), to reduce intraperitoneal clearance by phagocytosis and lymphoid transit. Empty microspheres showed the lack of toxicity in vitro. The double emulsion process yielded 2.5% w/w CPt loading and obtained microcapsules exhibited sustained (&gt, 20 day) zero-order release. The encapsulated CPt was confirmed to be bioavailable, as the microcapsules demonstrated efficacy against human ovarian adenocarcinoma (SK-OV-3) cells in vitro. Following intraperitoneal injection in mice, we did not observe adhesions, only mild, clinically-insignificant, local inflammatory response. Tissue platinum levels, monitored over 14 days using atomic absorption spectroscopy, revealed low burst and reduced systemic uptake (plasma, kidney), as compared to neat carboplatin injection. Overall, the results demonstrate the potential of the developed microencapsulation system for long-term intraperitoneal sustained release of carboplatin for the treatment of ovarian cancer.

Electrospinning is one of the most investigated methods used to produce polymeric fiber structures that mimic the morphology of native extracellular matrix. These structures have been extensively studied in the context of scaffolds for tissue regeneration. However, the compactness of materials obtained by traditional electrospinning, collected as two-dimensional non-woven scaffolds, can limit cell infiltration and tissue ingrowth. In addition, for applications in smooth muscle tissue engineering, highly elastic scaffolds capable of withstanding cyclic mechanical strains without suffering significant permanent deformations are preferred. In order to address these challenges, we report the fabrication of microscale 3D helically coiled structures (referred as 3D-HCS) by wet-electrospinning method, a modification of the traditional electrospinning process in which a coagulation bath (non-solvent system for the electrospun material) is used as the collector. The present study, for the first time, successfully demonstrates the feasibility of using this method to produce various architectures of 3D-HCS from segmented copolyester of poly(butylene succinate-co- dilinoleic succinate) (PBS-DLS), a thermoplastic elastomer. A mechanism for the HCS formation is proposed and verified with experimental data. Fabricated 3D-HCS showed high specific surface area, high porosity, and good elasticity. Further, the marked increase in cell proliferation on 3D-HCS confirmed the suitability of these materials as scaffolds for soft tissue engineering.

Proteins adsorbed onto biomaterial surfaces facilitate cell-material interactions, including adhesion and migration. Of particular importance are provisional matrix components, fibrinogen (Fg) and fibronectin (Fn), which play an important role in the wound healing process. Here, to assess the potential of a series of elastomeric poly(butylene succinate) copolymers for soft tissue engineering and regenerative medicine applications, we examined the adsorption of Fg and Fn. We prepared spin-coated thin films of poly(butylene succinate) homopolymer and a series of elastomeric poly(butylene succinate) copolymers with butylene succinate (PBS, hard segment) to succinate-dimer linoleic diol units (DLS, soft segments) weight ratios of 70:30, 60:40, and 50:50. X-ray diffraction was used to assess crystallinity, while the obtained thin films were characterized using quartz crystal microbalance with dissipation monitoring (QCM-D) and atomic force microscopy (AFM). Protein adsorption was assessed using QCM-D, followed by data analysis using viscoelastic modeling. On all three copolymers, we observed robust adsorption of both key provisional matrix proteins. Poly(butylene succinate) (PBS) is a thermoplastic and biodegradable polyester characterized by high rigidity due to its high crystallinity. However, the use of long chain biobased monomers to produce segmented copolymers is an effective strategy to tailor the properties of PBS, such as greater flexibility. In this paper, a series of aliphatic bio-copolyesters of poly(butylene succinate-dilinoleic succinate) (PBS-DLS) were successfully synthesized via a direct two-step polycondensation method using a semi-pilot scale reactor for melt polymerization and titanium dioxide/silicon dioxide coprecipitate catalyst (C-94). In this study, the thermal and mechanical properties were investigated and compared, focusing on the effect of varying the amount of biobased dilinoleic diol in the structure. With increasing amount of long chain diol, a decrease in molecular weight, density, and melt flow index was observed. The semicrystalline nature of the copolymers was confirmed using differential scanning calorimetry (DSC) and dynamic mechanical thermal analysis (DMTA) methods. These copolymers exhibit two main transition temperatures and vary in softness and processing flexibility. Furthermore, in the DSC data a linear trend was observed with increasing wt.% of hard PBS segments, which can be described by the Gordon-Taylor equation. Increasing the soft DLS segment content in the copolymer series resulted in an increase in the elastic behavior of the polymers. The broad range of crystallization temperatures and melt flow index values indicates that a polyester library with customizable properties that spans PBS applications has been successfully obtained.

This study evaluates the electrical potential and chemical alterations in laboratory-induced colistin-resistant Klebsiella pneumoniae, as compared to the susceptible strain using spectroscopic analyses. The minimal inhibitory concentration (MIC) of colistin, ζ-potential and chemical composition analysis of K. pneumoniae strains are determined. The results obtained for the K. pneumoniaeCol-R with induced high-level colistin resistance (MIC = 16.0 ± 0.0 mg/L) are compared with the K. pneumoniaeCol-S strain susceptible to colistin (MIC = 0.25 ± 0.0 mg/L). Fourier transform infrared (FTIR) and Raman spectroscopic studies revealed differences in bacterial cell wall structures and lipopolysaccharide (LPS) of K. pneumoniaeCol-R and K. pneumoniaeCol-S strains. In the beginning, we assumed that the obtained results could relate to a negative charge of the bacterial surface and different electrostatic interactions with cationic antibiotic molecules, reducing the affinity of colistin and leading to its lower penetration into K. pneumoniaeCol-R cell. However, no significant differences in the ζ-potential between the K. pneumoniaeCol-R and K. pneumoniaeCol-S strains are noticed. In conclusion, this mechanism is most probably associated with recognisable changes in the chemical composition of the K. pneumoniaeCol-R cell wall (especially in LPS) when compared to the susceptible strain.

Further, 1D and 2D infrared spectroscopy analysis indicated systematic reduction of polysaccharide biofilm elements, confirming the specificity of immobilized PelAh. Importantly, BC-PelAh was not cytotoxic towards L929 fibroblast cells. Thus, we conclude that PelAh can be used in BC wound dressings for safe and specific protection against biofilm formation by P. aeruginosa.

Valorization of food industry waste and plant residues represents an attractive path towards obtaining biodegradable materials and achieving &ldquo, zero waste&rdquo, goals. Here, melanin was isolated from watermelon (Citrullus lanatus) seeds and used as a modifier for whey protein concentrate and isolate films (WPC and WPI) at two concentrations (0.1% and 0.5%). The modification with melanin enhanced the ultraviolet (UV) blocking, water vapor barrier, swelling, and mechanical properties of the WPC/WPI films, in addition to affecting the apparent color. The modified WPC/WPI films also exhibited high antioxidant activity, but no cytotoxicity. Overall, the effects were melanin concentration-dependent. Thus, melanin from watermelon seeds can be used as a functional modifier to develop bioactive biopolymer films with good potential to be exploited in food packaging and biomedical applications.

Nanocomposites based on poly(ethylene terephthalate-ethylene dilinoleate) (PET-DLA) copolymers of different hard to soft segment ratios (40:60 and 60:40) and three different carbon nanofillers of different aspect ratios (dimensions), as 0D carbon black, 1D multiwalled carbon nanotubes, and 2D graphene, have been prepared in situ during two-stage polymerization. Fourier transform infrared (FTIR) and nuclear magnetic resonance (NMR) spectroscopy were used to characterize the chemical structures of the obtained nanocomposites. Scanning electron microscopy (SEM) indicated very good dispersions of all carbon nanofillers in both polymer matrices. Differential scanning calorimetry (DSC) results revealed that the addition of nano-sized fillers eliminated cold crystallization of materials containing 40% hard segments in polymer matrix. We found that the high aspect ratio, 1D nano-filler (multiwalled carbon nanotubes) strongly nucleated crystallization of materials containing 60% of hard segments. This nanofiller also yielded the greatest improvement in the Young’s modulus as assessed by tensile tests, both at 24 oC and 37 oC. We did not observe reduced bacterial adhesion to nanocomposites, likely due to increased roughness. Importantly, in vitro cytocompatibility tests with L929 murine fibroblasts demonstrated cell viability and growth on all materials except those containing carbon nanotubes. Finally, both high aspect ratio nanofillers markedly improved the barrier properties of obtained nanocomposites. New materials were successfully used for manufacturing of prototype of heart assist device, with pneumatic membrane made of graphene nanocomposite.

Biodegradable polymers are an active area of investigation, particularly ones that can be produced from sustainable, biobased monomers, such as copolymers of poly(butylene succinate) (PBS). In this study, we examine the enzymatic degradation of poly(butylene succinate-dilinoleic succinate) (PBS-DLS) copolymers obtained by &ldquo, green&rdquo, enzymatic synthesis using lipase B from Candida antarctica (CALB). The copolymers differed in their hard to soft segments ratio, from 70:30 to 50:50 wt %. Enzymatic degradation was carried out on electrospun membranes (scaffolds) and compression-moulded films using lipase from Pseudomomas cepacia. Poly(&epsilon, caprolactone) (PCL) was used as a reference aliphatic polyester. The degradation process was monitored gravimetrically via water uptake and mass loss. After 24 days, approx. 40% mass loss was observed for fibrous materials prepared from the PBS-DLS 70:30 copolymer, as compared to approx. 10% mass loss for PBS-DLS 50:50. Infrared spectroscopy (FTIR) and size exclusion chromatography (SEC) analysis were used to examine changes in chemical structure. Differential scanning calorimetry (DSC) and scanning light microscopy (LSM) revealed changes in degree of crystallinity, and changes in surface morphology, consistent with a surface erosion mechanism. We conclude that the obtained copolymers are suitable for tissue engineering applications thanks to tuneable degradation and lack of acidification during breakdown.

Biodegradable polymers are an active area of investigation, particularly ones that can be produced from sustainable, biobased monomers, such as copolymers of poly(butylene succinate) (PBS). In this study, we examine the enzymatic degradation of poly(butylene succinate-dilinoleic succinate) (PBS-DLS) copolymers obtained by “green” enzymatic synthesis using lipase B from Candida antarctica (CALB). Electrospinning is one of the most investigated methods used to produce polymeric fiber structures that mimic the fiber morphology of native extracellular matrix. These structures have been extensively studied in the context of scaffolds for tissue regeneration. However, the compactness of materials obtained by traditional electrospinning, collected as two-dimensional non-woven membranes, can limit cell infiltration and tissue ingrowth. In addition, for applications in smooth muscle tissue engineering, highly elastic scaffolds capable of withstanding cyclic mechanical strains without suffering significant permanent deformations are preferred. In order to address these challenges, we report the fabrication of microscale 3D helically coiled structures (referred as 3D-HCS) by wet-electrospinning method, a modification of the traditional electrospinning process in which a coagulation bath (non-solvent system for the electrospun material) is used as the collector. The present study successfully demonstrates the feasibility of using this method to process segmented copolyester poly(butylene succinate-co-dilinoleic succinate) (PBS-DLS), a thermoplastic elastomer, into 3D helically coiled structures (HCS). A mechanism for the HCS formation is proposed and verified with experimental data. Fabricated 3D-HCS showed high specific surface area, high porosity, and good elasticity. Further, the marked increase in cell proliferation on 3D-HCS confirmed the suitability of these materials as scaffolds for soft tissue engineering.

In this contribution the results on the development of a novel measurement system for electrochemical biosensing are presented. It covers the most important work on innovative graphene-based nanocomposites used in preparation of biosensing electrodes, design of different electrode types and a specialized flow chamber, specialized hardware and software for two versions (mobile and laboratory) of the electronic control and read-out system. The results of lab tests performed with the prototype system GruntMaster 6001 are shown in comparison to the measurements obtained by means of commercially available professional platforms used as a reference. The prototype is currently in the process of commercialization.

Cardiac catheterization results in interactions between the catheter and surfaces and the artery lumen, which is lined by the endothelium. These interactions can range from minor rubbing to severe mechanical injury. Further, in the case of radial access, even atraumatic interactions have consequences ranging from clinical complications, such as radial spasm and radial occlusion, to lasting endothelial cell dysfunction. These consequences may be underappreciated; however, endothelial cells play a central role in maintaining vascular homeostasis via nitric oxide production. Existing treatment paradigms do not address endothelial dysfunction or damage and, thus, novel therapeutic approaches are needed. Nanomedicine, in particular, offers great potential in the form of targeted drug delivery, via functionalized coatings or nanocarriers, aimed at increased nitric oxide bioavailability or reduced inflammation.

Traditionally, fibronectin has been used as a physisorbed surface coating (physFN) in cell culture experiments due to its critical role in cell adhesion. However, because the resulting layer is thick, unstable, and of unpredictable uniformity, this method of fibronectin deposition is unsuitable for some types of research, including quartz crystal microbalance (QCM) experiments involving cells. Here, we present a new method for chemical immobilization of fibronectin onto silicon oxide surfaces, including QCM crystals pre-coated with silicon oxide. We characterize these chemically coated fibronectin surfaces (chemFN) as well as physFN ones using spectroscopic ellipsometry (SE), Fourier transform infrared spectroscopy (FTIR), atomic force microscopy (AFM), and contact angle measurements. A cell culture model demonstrates that cells on chemFN and physFN surfaces exhibit similar viability, structure, adhesion and metabolism. Finally, we perform QCM experiments using cells on both surfaces which demonstrate the superior suitability of chemFN coatings for QCM research, and provide real-time QCM-D data from cells subjected to an actin depolymerizing agent. Overall, our method of chemical immobilization of fibronectin yields great potential for furthering cellular experiments in which thin, stable and uniform coatings are desirable. As QCM research with cells has been rather limited in success thus far, we anticipate that this new technique will particularly benefit this experimental system by availing it to the much broader field of cell mechanics.

The aim of this work was to assess whether synthesized random copolyester, poly(butylene terephthalate-r-butylene dilinoleate) (PBT&ndash, DLA), containing bio-based components, can effectively compatibilize polypropylene/poly(butylene terephthalate) (PP/PBT) blends. For comparison, a commercial petrochemical triblock copolymer, poly(styrene-b-ethylene/butylene-b-styrene) (SEBS) was used. The chemical structure and block distribution of PBT&ndash, DLA was determined using nuclear magnetic resonance spectroscopy and gel permeation chromatography. PP/PBT blends with different mass ratios were prepared via twin-screw extrusion with 5 wt% of each compatibilizer. Thermogravimetric analysis, differential scanning calorimetry, and dynamic mechanical analysis were used to assess changes in phase structure of PP/PBT blends. Static tensile testing demonstrated marked improvement in elongation at break, to ~18% and ~21% for PBT&ndash, DLA and SEBS, respectively. Importantly, the morphology of PP/PBT blends compatibilized with PBT&ndash, DLA copolymer showed that it is able to act as interphase modifier, being preferentially located at the interface. Therefore, we conclude that by using polycondensation and monomers from renewable resources, it is possible to obtain copolymers that efficiently modify blend miscibility, offering an alternative to widely used, rubber-like petrochemical styrene compatibilizers.

Biosensing is an important and rapidly developing field, with numerous potential applications in health care, food processing, and environmental control. Polymer-graphene nanocomposites aim to leverage the unique, attractive properties of graphene by combining them with those of a polymer matrix. Molecular imprinted polymers, in particular, offer the promise of artificial biorecognition elements. A variety of polymers, including intrinsically conducting polymers (polyaniline, polypyrrole), bio-based polymers (chitosan, polycatechols), and polycationic polymers (poly(diallyldimethylammonium chloride), polyethyleneimine), have been utilized as matrices for graphene-based nanofillers, yielding sensitive biosensors for various biomolecules, such as proteins, nucleic acids, and small molecules.

Hemoglobin vesicles (HbVs) are artificial oxygen carriers encapsulating purified and concentrated Hb solution in phospholipid vesicles (liposomes). We examined in-vitro reaction profiles of a formulation of HbV with NO and CO in anaerobic and aerobic conditions using stopped-flow spectrophotometry and a NO electrode. Reaction rate constants of NO to deoxygenated and oxygenated HbV were considerably smaller than those of cell-free Hb because of the intracellular NO-diffusion barrier. The reaction of CO with deoxygenated HbV was slightly slower than that of cell-free Hb solely because of the co-encapsulated allosteric effector, pyridoxal 5'-phosphate. The NO depletion in an aerobic condition in the presence of empty vesicles was monitored using a NO electrode, showing that the hydrophobic bilayer membrane of HbV, which might have higher gas solubility, does not markedly facilitate the O(2) and NO reaction, and that the intracellular Hb is the major component of NO depletion. In conclusion, HbV shows retarded gas reactions, providing some useful information to explain the absence of vasoconstriction and hypertension when they are intravenously injected.

Background— Local drug delivery from polymer-coated stents has demonstrated efficacy for preventing in-stent restenosis; however, both the inflammatory effects of polymer coatings and concerns about late outcomes of drug-eluting stent use indicate the need to investigate innovative approaches, such as combining localized gene therapy with stent angioplasty. Thus, we investigated the hypothesis that adenoviral vectors (Ad) could be delivered from the bare-metal surfaces of stents with a synthetic complex for reversible vector binding. Methods and Results— We synthesized the 3 components of a gene vector binding complex: (1) A polyallylamine bisphosphonate with latent thiol groups (PABT), (2) a polyethyleneimine (PEI) with pyridyldithio groups for amplification of attachment sites [PEI(PDT)], and (3) a bifunctional (amine- and thiol-reactive) cross-linker with a labile ester bond (HL). HL-modified Ad attached to PABT/PEI(PDT)-treated steel surfaces demonstrated both sustained release in vitro over 30 days and localized green fluorescent protein expression in rat arterial smooth muscle cell cultures, which were not sensitive to either inhibition by neutralizing anti-Ad antibodies or inactivation after storage at 37°C. In rat carotid studies, deployment of steel stents configured with PABT/PEI(PDT)/HL-tethered adenoviral vectors demonstrated both site-specific arterial Ad GFP expression and adenovirus-luciferase transgene activity per optical imaging. Rat carotid stent delivery of adenovirus encoding inducible nitric oxide synthase resulted in significant inhibition of restenosis. Conclusions— Reversible immobilization of adenovirus vectors on the bare-metal surfaces of endovascular stents via a synthetic complex represents an efficient, tunable method for sustained release of gene vectors to the vasculature.

Rational development of adjunct or anti-disease therapy for severe Plasmodium falciparum malaria requires cellular and molecular definition of malarial pathogenesis. Nitric oxide (NO) is a potential target for such therapy but its role during malaria is controversial. It has been proposed that NO is produced at high levels to kill Plasmodium parasites, although the unfortunate consequence of elevated NO levels might be impaired neuronal signaling, oxidant damage and red blood cell damage that leads to anemia. In this case, inhibitors of NO production or NO scavengers might be an effective adjunct therapy. However, increasing amounts of evidence support the alternate hypothesis that NO production is limited during malaria. Furthermore, the well-documented NO scavenging by cell-free plasma hemoglobin and superoxide, the levels of which are elevated during malaria, has not been considered. Low NO bioavailability in the vasculature during malaria might contribute to pathologic activation of the immune system, the endothelium and the coagulation system: factors required for malarial pathogenesis. Therefore, restoring NO bioavailability might represent an effective anti-disease therapy.

The versatility of NG makes them ideal candidates for drug delivery and bioimaging [1]. NG, often characterized as soft nanoparticles, are hydrophilic 3D networks that can absorb large amounts of water or biological fluids while maintaining their structure. In the swollen state, the low surface tension between the fluid and the NG minimizes nonspecific interactions, such as protein and cell adhesion, enhancing biocompatibility. In addition, the sub-micrometer size of the nanogels ensures that most nanogels are able to be taken up by cells, while their large surface area allows for tailoring to various in vitro/vivo applications and rapid responses to environment stimuli [2]. Research on drug delivery applications has mostly focused on core-shell NG. Generally, a hydrophilic shell stabilizes the NG in water/biological fluid and provides the NG with a “stealth” property, allowing them to avoid mononuclear phagocytic system (MPS) recognition as well as opsonization. The hydrophobic core allows for loading of cargo/drug via hydrophobic and electrostatic interactions. Polymers frequently used to prepare NG include polyethylene glycol-poly(ethylene imine) [3], poly(4-vinylpyridine)-poly(N-isopropylacrylamide) [4], and natural polymers. The use of polysaccharides, such as dextran, for the hydrophilic shell is particularly appealing for biomedical applications due to well-documented biocompatibility and biodegradability, multiple reactive sites for attachment of specific ligands, and steric protection against protein adsorption [5–13]. Common synthesis methods to prepare core-shell NG include (inverse) emulsion polymerization [14], precipitation polymerization [15], and photopolymerization [16]. Alternatively, core-shell NG can be prepared via two simple steps, Maillard reaction and heat gelation. The Maillard reaction is a natural nontoxic reaction that links a protein and a polysaccharide. Subsequent heat gelation ensures partial denaturation of the protein and stable formation of the NG. This technique has several notable advantages: it is low-cost, completely “green” (i.e., it takes place in an aqueous solution) and initiator-free. Following this methodology, Li et al. synthesized NG with a LDNG that could be tailored from 80 nm to 1 μm in diameter [7]. In particular, LDNG (200 nm diameter) were spherical in shape, stable across a broad range of pH values and ionic strength, and capable of loading ibuprofen. Similar methodology has been used to prepare other protein-polysaccharide core-shell NG, such as bovine serum albumin (BSA)-dextran [9], BSA-dextran-chitosan [17], and doxorubicin-BSA-dextran [18] core-shell nanoparticles. The latter NG was found to prolong the life of mice having hepatoma H22 tumors [18]. In this work, we prepare rhodamine-labeled LDNG (LRDNG) and demonstrate their potential for use as carriers for drug delivery. This was accomplished by loading a fluorescent molecular probe (HAF) into the core of the LRDNG to model a drug. HUVEC were used as a cell culture model to verify the uptake of the LRDNG and drug release, as well as to assess cytotoxicity. Finally, the “stealth” characteristics of the LRDNG were confirmed using differentiated macrophages (PMA-stimulated THP-1 cells), as a model for the MPS.

Vascular air embolism resulting from too rapid decompression is a well-known risk in deep-sea diving, aviation and space travel. It is also a common complication during surgery or other medical procedures when air or other endogenously administered gas is entrained in the circulation. Preventive and post-event treatment options are extremely limited for this dangerous condition, and none of them address the poorly understood pathophysiology of endothelial response to intravascular bubble presence. Using a novel apparatus allowing precise manipulation of microbubbles in real time fluorescence microscopy studies, we directly measure human umbilical vein endothelial cell responses to bubble contact. Strong intracellular calcium transients requiring extracellular calcium are observed upon cell-bubble interaction. The transient is eliminated both by the presence of the stretch activated channel inhibitor, gadolinium, and the transient receptor potential vanilliod family inhibitor, ruthenium red. No bubble induced calcium upsurge occurs if the cells are pretreated with an inhibitor of actin polymerization, cytochalasin-D. This study explores the biomechanical mechanisms at play in bubble interfacial interactions with endothelial surface layer (ESL) macromolecules, reassessing cell response after selective digestion of glycocalyx glycosoaminoglycans, hyaluran (HA) and heparin sulfate (HS). HA digestion causes reduction of cell-bubble adherence and a more rapid induction of calcium influx after contact. HS depletion significantly decreases calcium transient amplitudes, as does pharmacologically induced sydencan ectodomain shedding. The surfactant perfluorocarbon Oxycyte abolishes any bubble induced calcium transient, presumably through direct competition with ESL macromolecules for interfacial occupancy, thus attenuating the interactions that trigger potentially deleterious biochemical pathways.

Gas embolism is a serious complication of decompression events and clinical procedures, but the mechanism of resulting injury remains unclear. Previous work has demonstrated that contact between air microbubbles and endothelial cells causes a rapid intracellular calcium transient and can lead to cell death. Here we examined the mechanism responsible for the calcium rise. Single air microbubbles (50–150 μm), trapped at the tip of a micropipette, were micromanipulated into contact with individual human umbilical vein endothelial cells (HUVECs) loaded with Fluo-4 (a fluorescent calcium indicator). Changes in intracellular calcium were then recorded via epifluorescence microscopy. First, we confirmed that HUVECs rapidly respond to air bubble contact with a calcium transient. Next, we examined the involvement of extracellular calcium influx by conducting experiments in low calcium buffer, which markedly attenuated the response, or by pretreating cells with stretch-activated channel blockers (gadolinium chloride or ruthenium red), which abolished the response. Finally, we tested the role of intracellular calcium release by pretreating cells with an inositol 1,4,5-trisphosphate (IP3) receptor blocker (xestospongin C) or phospholipase C inhibitor (neomycin sulfate), which eliminated the response in 64% and 67% of cases, respectively. Collectively, our results lead us to conclude that air bubble contact with endothelial cells causes an influx of calcium through a stretch-activated channel, such as a transient receptor potential vanilloid family member, triggering the release of calcium from intracellular stores via the IP3 pathway.

Luminescent dinoflagellates respond to flow by the production of light. The primary mechanotransduction event is unknown, although downstream events include a calcium flux in the cytoplasm, a self-propagating action potential across the vacuole membrane, and a proton flux into the cytoplasm that activates the luminescent chemistry. Given the role of GTP-binding (G) proteins in the mechanotransduction of flow by nonmarine cells and the presence of G-proteins in dinoflagellates, it was hypothesized that flow-stimulated dinoflagellate bioluminescence involves mechanotransduction by G-proteins. In the present study, osmotic swelling of cells of the dinoflagellate Lingulodinium polyedrum was used as a drug delivery system to introduce GDPβS, an inhibitor of G-protein activation. Osmotically swollen cells produced higher levels of flow-stimulated bioluminescence at a lower threshold of shear stress, indicating they were more flow sensitive. GDPβS inhibited flow-stimulated bioluminescence in osmotically swollen cells and in cells that were restored to the isosmotic condition following hypoosmotic treatment with GDPβS. These results provide evidence that G-proteins are involved in the mechanotransduction of flow in dinoflagellates and suggest that G-protein involvement in mechanotransduction may be a fundamental evolutionary adaptation.

The role of nitric oxide (NO) in the genesis of cerebral malaria is controversial. Most investigators propose that the unfortunate consequence of the high concentrations of NO produced to kill the parasite is the development of cerebral malaria. Here we have tested this high NO bioavailability hypothesis in the setting of experimental cerebral malaria (ECM), but find instead that low NO bioavailability contributes to the genesis of ECM. Specifically, mice deficient in vascular NO synthase showed parasitemia and mortality similar to that observed in control mice. Exogenous NO did not affect parasitemia but provided marked protection against ECM; in fact, mice treated with exogenous NO were clinically indistinguishable from uninfected mice at a stage when control infected mice were moribund. Administration of exogenous NO restored NO-mediated signaling in the brain, decreased proinflammatory biomarkers in the blood, and markedly reduced vascular leak and petechial hemorrhage into the brain. Low NO bioavailability in the vasculature during ECM was caused in part by an increase in NO-scavenging free hemoglobin in the blood, by hypoargininemia, and by low blood and erythrocyte nitrite concentrations. Exogenous NO inactivated NO-scavenging free hemoglobin in the plasma and restored nitrite to concentrations observed in uninfected mice. We therefore conclude that low rather than high NO bioavailability contributes to the genesis of ECM.

Reactive oxygen species (ROS) are widely believed to kill malarial parasites. C57BL/6 mice injected withP. bergheiinocula incubated with supraphysiological doses of NO (≤150 μM) or with peroxynitrite (220 μM), however, exhibited parasitemia similar to that seen with those given control inocula, and there was no difference in disease development. Only treatment of inocula with NO doses nearing saturation (≥1.2 mM) resulted in no detectable parasitemia in the recipients; flow cytometric analysis with a vital dye (hydroethidine) indicated that 1.5 mM NO lysed the erythrocytes rather than killing the parasites. The hemoglobin level in the inocula was about 8 μM; the hemoglobin was mainly oxyhemoglobin (oxyHb) (96%), which was converted to methemoglobin (>95%) after treatment with 150 μM NO. The concentrations of 150 μM of NO and 220 μM of peroxynitrite were far in excess of the hemoglobin concentration (∼8 μM), and yet no parasite killing was detected. We therefore conclude that hemoglobin protectsPlasmodiumparasites from ROS, but the parasite likely possesses intrinsic defense mechanisms against ROS.

Our understanding of how the host immune response kills Plasmodium, the causative agent of malaria, is limited and controversial. One widely held belief is that reactive oxygen species are crucial for controlling parasite replication. One of the hallmarks of blood-stage malaria is the cyclic rupture of erythrocytes by the parasite, which releases free hemoglobin into the circulation. We propose that this free hemoglobin, as well as the hemoglobin within the erythrocyte and surrounding the parasite, effectively shields Plasmodium from reactive oxygen species well in excess of those achievable in vivo.

Objective Gas microembolism remains a serious risk associated with surgical procedures and decompression. Despite this, the signaling consequences of air bubbles in the vasculature are poorly understood and there is a lack of pharmacological therapies available. Here, we investigate the mitochondrial consequences of air bubble contact with endothelial cells. Methods and results Human umbilical vein endothelial cells were loaded with an intracellular calcium indicator (Fluo-4) and either a mitochondrial calcium indicator (X-Rhod-1) or mitochondrial membrane potential indicator (TMRM). Contact with 50-150 µm air bubbles induced concurrent rises in intracellular and mitochondrial calcium, followed by a loss of mitochondrial membrane potential. Pre-treating cells with 1 µmol/L ruthenium red, a TRPV family calcium channel blocker, did not protect cells from the mitochondrial depolarization, despite blocking the intracellular calcium response. Mitigating the interactions between the air-liquid interface and the endothelial surface layer with 5% BSA or 0.1% Pluronic F-127 prevented the loss of mitochondrial membrane potential. Finally, inhibiting protein kinase C-α (PKCα), with 5 µmol/L Go6976, protected cells from mitochondrial depolarization, but did not affect the intracellular calcium response. Conclusions Our results indicate that air bubble contact with endothelial cells activates a novel, calcium-independent, PKCα-dependent signaling pathway, which results in mitochondrial depolarization. As a result, mitochondrial dysfunction is likely to be a key contributor to the pathophysiology of gas embolism injury. Further, this connection between the endothelial surface layer and endothelial mitochondria may also play an important role in vascular homeostasis and disease.
