ABSTRACT: Bracken ferns (Pteridium spp.) have been associated with intoxication in cattle and is the cause of bovine enzootic hematuria (BEH) characterized by intermittent hematuria, anemia, and poor body condition. Acute phase proteins have not previously been investigated in detail in BEH. Thus, this investigation determined serum concentrations of haptoglobin, selected hematological (complete blood count and fibrinogen), biochemical (total serum protein, albumin, urea, creatinine, aspartate aminotransferase, gamma glutamyl transferase, and creatine kinase), and urinary analytes, in 50 Nelore cows from farms with history of natural bracken fern chronic ingestion in the southern region of Brazil. Twenty-nine cows had macrohematuria and 21 microhematuria. From the 29 cows with macrohematuria, 13 had a countless number of erythrocytes per high power macroscopic field and anemia. All cows had dipstick proteinuria. Median haptoglobin concentrations were elevated for 47 BEH cows with a 3.9-fold increase, suggesting a chronic inflammatory response while the number of total leukocytes and segmented neutrophils were above the reference intervals for 14 cows. Besides inflammation, there is a hypothesis that bracken fern could cause hemolysis in the circulation. Therefore, there could be a larger acute phase reaction but much of the haptoglobin was being removed as it was being produced. In our study, increases in haptoglobin, creatine kinase, hyperglobulinemia and anemia were found in cows with BEH, contributing to the understanding of the chronic inflammatory response which could be associated with neoplastic lesions in the urinary bladder caused by long-term and repeated ingestion of Pteridium species.

Introduction The number of robotic-assisted hip replacement procedures has expanded globally with the intended aim of improving outcomes. Intraoperative robotic-arm systems add additional costs to total hip replacement (THR) surgery but may improve surgical precision and could contribute to diminished pain and improved function. Additionally, these systems may reduce the need for expensive revision surgery. Surgery with conventional instruments may be just as successful, quick and affordable. There is timely demand for a robust evaluation of this technology.Methods and analysis The Robotic Arthroplasty Clinical and cost Effectiveness Randomised controlled trial for Hips (RACER-Hip) is a multicentre (minimum of six UK sites), participant–assessor blinded, randomised controlled trial. 378 participants with hip osteoarthritis requiring THR will be randomised (1:1) to receive robotic-assisted THR, or THR using conventional surgical instruments. The primary outcome is the Forgotten Joint Score at 12 months post-randomisation; a patient-reported outcome measure assessing participants’ awareness of their joint when undertaking daily activities. Secondary outcomes will be collected post-operatively (pain, blood loss and opioid usage) and at 3, 6, 12, 24 months, then 5 and 10 years postrandomisation (including function, pain, health-related quality of life, reoperations and satisfaction). Allocation concealment will be accomplished using a computer-based randomisation procedure on the day of surgery. Blinding methods include the use of sham incisions for marker clusters and blinded operation notes. The primary analysis will adhere to the intention-to-treat principle. Results will adhere to Consolidated Standards of Reporting Trials statements.Ethics and dissemination The trial was approved by an ethics committee (Solihull Research Ethics Committee, 30 June 2021, IRAS: 295831). Participants will provide informed consent before agreeing to participate. Results will be disseminated using peer-reviewed journal publications, presentations at international conferences and through the use of social media. We will develop plans to disseminate to patients and public with our patient partners.Trial registration number ISRCTN13374625.

Rural farmers face different challenges from climate change and weather variability, which have threatened crop production and productivity. Small-scale farmers try to cope with the prevailing situations by adjusting to different mechanisms, including adopting irrigation services. This study seeks to determine factors influencing rice farmers' decision to adopt irrigation technology and determine the significant contribution of irrigation to rice yields in the Mbeya Region of Tanzania. Data was collected through structured questionnaires, interviews, and focus discussions. Descriptive statistics, Logistic regression, and Ordinary Least Square regression by Stata software performed data analyses. The descriptive statistics characterized households' socio-demographic and economic characteristics. Logistic regression results affirmed that households' education, labor size, meteorological information, access to financial services, extension services, and previous farm outputs significantly influenced irrigation adoption by farmers. Ordinary Least Square regression results confirmed that irrigation significantly contributed to rice yield at P>0.05 level. The study recommends adopting irrigation technology in Tanzania as a coping strategy for the negative impact of climate change and weather variations.

Dilated cardiomyopathy is one of the important diseases in dogs and humans. The second most common cause of heart failure in dogs is idiopathic dilated cardiomyopathy (iDCM), which results in heart failure or sudden cardiac death due to arrhythmia. This study aimed to determine changes in the plasma metabolome of dogs with iDCM compared to healthy dogs. For that purpose, a multiplatform mass-spectrometry-based approach was used. In this study, we included two groups of dogs: 12 dogs with iDCM and 8 healthy dogs. A total of 272 metabolites were detected in the plasma samples of dogs by combining three approaches but four MS-based platforms (GC-MS, LC-MS (untargeted), LC-MS (targeted), and FIA-MS (targeted) methods). Our findings demonstrated changes in the canine plasma metabolome involved in the development of iDCM, including the different concentrations of amino acids, biogenic amines, acylcarnitines, triglycerides and diglycerides, sphingomyelins, and organic acids. The results of this study will enable the detection and monitoring of pathophysiological mechanisms involved in the development of iDCM in the future.

Background: Sleep deprivation and disruptions are associated with both psychological and physiological disorders. Medical students have an increased risk for sleep deprivation. This study aimed to determine the quality of sleep among clinical medical students of Bayero University Kano (BUK). Materials and Methods: This is a cross-sectional study that was conducted between February and March 2019 among clinical students of BUK. The data were obtained using sociodemographic questionnaire as well as the Pittsburgh Sleep Quality Index (PSQI) and Epworth Sleepiness Scale (ESS) self-administered questionnaires. Two hundred questionnaires were administered to the students in their 4th, 5th, and 6th academic year. SPSS version 20 was used for the statistical analysis of the data collected. Results: A total of 181 questionnaires were completed out of the 200 that were administered, making a response rate of 90.5%. There were 114 males (63%) and 67 females (37%), giving a male-to-female ratio of 1.7:1, which is in keeping with the gender distribution of the students. The mean age (± standard deviation) of the students was 23.8 ± 2.9 years. The overall poor sleep quality was found to be 53.0% based on the PSQI, whereas the overall excessive daytime sleepiness was found to be 38.3% using the ESS. Poor sleep quality was found to be 61.1%, 51.7%, and 42.9% among the students in the 4th, 5th, and 6th academic years, respectively (P = 0.14). Excessive daytime sleepiness was found to be 45.8%, 41.7%, and 22.9% among the students in the 4th, 5th, and 6th academic years, respectively (P = 0.03). Conclusion: The prevalence of poor sleep quality is high among the clinical students of BUK.

IntroductionAtrial Septal Defect/Patent Foramen Ovale (ASD/PFO) occlusion is performed to prevent paradoxical embolism and reduce the risk of recurrent ischemic stroke. Left atrial appendage (LAA) closure is used as an alternative to medical therapy of non-valvular atrial fibrillation for prevention of stroke. Multiple studies have examined performing LAA and ASD/PFO occlusion. However, the feasibility and safety of combined occlusion of the left atrial appendage and ASD/PFO are not clear, furthermore, these studies are limited by their small sample sizes and retrospective analysis. In this study, we aimed to systematically review and meta-analyze the feasibility and safety of combining left atrial appendage and ASD/PFO closure.MethodsPubMed, Web of Science, CNKI, Cochrane Library, Embase, and WanFang database were searched up to April 2022 to identify peer-reviewed human studies on assessing the feasibility, safety, and efficacy of combining left atrial appendage and ASD/PFO closure. The primary outcome was calculated: procedural feasibility outcome and procedural safety outcome.ResultsA total of 10 articles, including 340 patients from multiple countries, were included in the analysis. The principal findings of our study are: compared with single LAA closure, (i) combining PFO/ASD occlusion and LAA closure had similar procedural success proportion (98.43%, 95% CI: 96.67–100.00%), (ii) similar safety event incidences developed (1.67%, 95% CI: 0.24–3.92%), subgroup analyzed safety event incidences in death was 0.00 (95% CI: 0.00–0.33%), cardiac tamponade was 0.87% (95% CI: 0.00–2.77%), device embolization was 0.00 (95% CI: 0.00–0.60%), major bleeding was 0.00 (95% CI: 0.00–0.33%), stroke was 0.00 (95% CI: 0.00–0.02%).ConclusionAlthough this systematic review and meta-analysis demonstrate the technical feasibility and safety of combining closure of PFO/ASD and LAA, further studies of sufficient sample size, long-term follow-up, and rigor endpoint criteria are yet needed to fully evaluate this combination procedure for its role in clinical outcomes.

S100 proteins are a group of calcium-binding proteins which received this name because of their solubility in a 100% saturated solution of ammonium sulphate. They have a similar molecular mass of 10–12 KDa and share 25–65% similarity in their amino acid sequence. They are expressed in many tissues, and to date 25 different types of S100 proteins have been identified. This review aims to provide updated information about S100 proteins and their use as biomarkers in veterinary science, with special emphasis on the family of calgranulins that includes S100A8 (calgranulin A; myeloid-related protein 8, MRP8), S100A9 (calgranulin B; MRP14), and S100A12 (calgranulin C). The proteins SA100A8 and S100A9 can be linked, forming a heterodimer which is known as calprotectin. Calgranulins are related to the activation of inflammation and the immune system and increase in gastrointestinal diseases, inflammation and sepsis, immunomediated diseases, and obesity and endocrine disorders in different animal species. This review reflects the current knowledge about calgranulins in veterinary science, which should increase in the future to clarify their role in different diseases and potential as biomarkers and therapeutic targets, as well as the practical use of their measurement in non-invasive samples such as saliva or feces.

Equine gastric ulcer syndrome (EGUS) is a highly prevalent disease that affects horses worldwide. Within EGUS, two different forms have been described: equine squamous gastric disease (ESGD) and equine glandular gastric disease (EGGD). The associated clinical signs cause detrimental activity performance, reducing the quality of life of animals. Saliva can contain biomarkers for EGUS that could be potentially used as a complementary tool for diagnosis. The objective of this work was to evaluate the measurements of calprotectin (CALP) and aldolase in the saliva of horses as potential biomarkers of EGUS. For this purpose, automated assays for the quantification of these two proteins were analytically validated and applied for detecting EGUS in a total of 131 horses divided into 5 groups: healthy horses, ESGD, EGGD, combined ESGD and EGGD, and horses with other intestinal pathologies. The assays showed good precision and accuracy in analytical validation, and they were able to discriminate between horses with EGUS and healthy horses, especially in the case of CALP, although they did not show significant differences between horses with EGUS and horses with other diseases. In conclusion, salivary CALP and aldolase can be determined in the saliva of horses and further studies are warranted to elucidate the potential of these analytes as biomarkers in EGUS.

Calprotectin (CALP, S100A8/A9), also named myeloid-related protein 8/14, is a dimer complex of S100A8 and S100A9 that belongs to the S-100 protein family. It is involved in inflammation and has a wide range of proinflammatory functions, such as cytokine production and regulation of leukocyte adhesion, migration, and phagocytosis. In humans, CALP traditionally can be measured in faeces, serum, and saliva as a biomarker of inflammation and sepsis. The objective of this study was to validate an automated assay for CALP measurements in the saliva of pigs, having the advantage of the use of a non-invasive sample that is easy to collect. The assay was precise and accurate. CALP in saliva measured by this assay showed significant changes depending on the hour of the day. It also showed significant increases in the saliva of pigs after the administration of lipopolysaccharide (LPS), and showed a rise, although with increases of lower magnitude, after a stressful stimulus. Further studies should be made to gain knowledge about the possible practical applications of the measurements of CALP in the saliva of pigs as a biomarker to evaluate the animals’ health and welfare.

Comprehensive profiling of serum proteome provides valuable clues of health status and pathophysiological processes, making it the main strategy in biomarker discovery. However, the high dynamic range significantly decreases the number of detectable proteins, obstructing the insights into the underlying biological processes. To circumvent various serum enrichment methods, obtain high-quality proteome wide information using the next-generation proteomic, and study host response in canine leishmaniosis, we applied data-independent acquisition mass spectrometry (DIA-MS) for deep proteomic profiling of clinical samples. The non-depleted serum samples of healthy and naturally Leishmania-infected dogs were analyzed using the label-free 60-min gradient sequential window acquisition of all theoretical mass spectra (SWATH-MS) method. As a result, we identified 554 proteins, 140 of which differed significantly in abundance. Those were included in lipid metabolism, hematological abnormalities, immune response, and oxidative stress, providing valuable information about the complex molecular basis of the clinical and pathological landscape in canine leishmaniosis. Our results show that DIA-MS is a method of choice for understanding complex pathophysiological processes in serum and serum biomarker development.

In February 2021, the Wisconsin DNR implemented a wolf season in which > 20% of the population was killed in 63 h. Wisconsin’s Ojibwe tribes had a visceral reaction to this killing. This paper provides a perspective for this reaction by reviewing the Ojibwe relationship with Ma’iingan. This relationship maintains that Ma’iingan and Ojibwe are to be considered relatives whose fates are intertwined. Ma’iingan and Ojibwe have lived parallel histories, suffering from the effects of colonization, the decimation of wolf populations and decline of tribal culture. The Ojibwe tribes ceded vast territories in treaties with the United States while retaining common use rights, including the right to hunt and fish. These rights were reaffirmed just as wolves were reestablishing themselves in Wisconsin. The tribes continue to strengthen their culture, while wolf populations continue to recover. By examining these comparative histories, it becomes apparent that “whatever happens to one happens to the other.” Unfortunately, Ma’iingan were not adjudicated during the Wisconsin treaty case, creating uncertainty over how the relationship between the Ojibwe and Ma’iingan is to be respected by the state. The tribes believe their treaty right includes protection for wolves, so that wolves can fulfill their cultural and ecological purposes. Tribes maintain that Ma’iingan should determine their own population levels, in order to provide ecological and cultural benefits. A respectful and appreciative relationship with Ma’iingan should be maintained so that the future well-being of both Ma’iingan and the Ojibwe will be assured.

Abstract Background Currently, the pathogenesis of congestive heart failure (CHF) in cats is not fully understood. Objective To identify novel biomarkers for CHF in cats caused by primary cardiomyopathy, particularly related to cardiovascular‐renal axis disorder and systemic inflammatory response. Animals Twenty‐five cats in CHF caused by primary cardiomyopathy, 12 cats with preclinical cardiomyopathy, and 20 healthy controls. Methods Case control and observational case series. The following serum biomarkers were compared among the 3 cat groups: a cardiorenal profile that included N‐terminal pro‐brain natriuretic peptide (NT‐proBNP), symmetric dimethylarginine (SDMA), and creatinine and an inflammatory profile that included 7 acute‐phase proteins (APPs). Survival analyses and longitudinal studies were performed in CHF cats. Results All cardiorenal biomarkers were positively correlated and higher in CHF cats, and high NT‐proBNP and SDMA were associated with poor clinical outcome. Cats with CHF had significantly higher leucine‐rich alpha‐2‐glycoprotein 1, serum amyloid A, and ceruloplasmin, and these APPs were positively correlated with NT‐proBNP and left atrial size. In a multivariable survival analysis, alpha‐1‐acid glycoprotein concentration (P = .01), body weight (P = .02) and left atrial‐to‐aortic root ratio (P = .01) were independent prognostic factors for CHF in these cats. Conclusions and Clinical Importance In cats, CHF is an inflammatory disorder and outcome in CHF may be determined by the extent of inflammation and possibly the amount of residual renal function. These novel biomarkers have potential use for the clinical management, prognosis, and future research into CHF and cardiomyopathy in cats.

Satellite altimetry data provide a solution to the lack of in situ tide gauge data, which are essential for comprehending various marine processes worldwide. In the present study, we seek to validate ALES-retrieved sea-level data against tide gauge observations from four ground stations on the coast of Mozambique. The approach consisted of extracting data from selected tracks of the Jason-1, Jason-2 and Jason-3 missions, and processing it to (i) remove outliers, (ii) collocate alongside tide gauge data, (iii) remove the tidal component and detrend, and (iv) perform a set of statistical analyses. Good agreement was found between the altimetry and tide gauge data in three of the four stations (Maputo, r = 0.59; Inhambane, r = 0.87; and Pemba, r = 0.75), with the exception of Beira. The annual and semi-annual cycles in the two datasets revealed that the altimetry signal is smaller in amplitude and ahead (with a few exceptions) of tide gauge by a varying number of days in each location. Both the annual and semi-annual cycles are far more comparable in Pemba, where the amplitude in particular has the same order of magnitude, followed by the Maputo station. The study concluded that the selected altimetry data for Pemba and Maputo stations are valid and can be used for coastal risk analysis and other applications. No altimetry data could be validated for Inhambane and Beira stations due to lack of consistent and sufficiently long tide gauge records. This difficulty urges the need for improved maintenance practices of ground stations located near human settlements that rely on sound information of the sea level and its variability to protect lives, infrastructure and livelihoods.

Objective Tourniquet use in total knee replacement (TKR) is believed to improve the bone-cement interface by reducing bleeding, potentially prolonging implant survival. This study aimed to compare the risk of revision for primary cemented TKR performed with or without a tourniquet.Design We analysed data from the National Joint Registry (NJR) for all primary cemented TKRs performed in England and Wales between April 2003 and December 2003. Kaplan-Meier plots and Cox regression were used to assess the influence of tourniquet use, age at time of surgery, sex and American Society of Anaesthesiologists (ASA) classification on risk of revision for all-causes.Results Data were available for 16 974 cases of primary cemented TKR, of which 16 132 had surgery with a tourniquet and 842 had surgery without a tourniquet. At 10 years, 3.8% had undergone revision (95% CI 2.6% to 5.5%) in the no-tourniquet group and 3.1% in the tourniquet group (95% CI 2.8% to 3.4%). After adjusting for age at primary surgery, gender and primary ASA score, the HR for all-cause revision for cemented TKR without a tourniquet was 0.82 (95% CI 0.57 to 1.18).Conclusions We did not find evidence that using a tourniquet for primary cemented TKR offers a clinically important or statistically significant reduction in the risk of all-cause revision up to 13 years after surgery. Surgeons should consider this evidence when deciding whether to use a tourniquet for cemented TKR.

Abstract High energy X-ray phase contrast tomography is tremendously beneficial to the study of thick and dense materials with poor attenuation contrast. Recently, the X-ray speckle-based imaging technique has attracted widespread interest because multimodal contrast images can now be retrieved simultaneously using an inexpensive wavefront modulator and a less stringent experimental setup. However, it is time-consuming to perform high resolution phase tomography with the conventional step-scan mode because the accumulated time overhead severely limits the speed of data acquisition for each projection. Although phase information can be extracted from a single speckle image, the spatial resolution is deteriorated due to the use of a large correlation window to track the speckle displacement. Here we report a fast data acquisition strategy utilising a fly-scan mode for near field X-ray speckle-based phase tomography. Compared to the existing step-scan scheme, the data acquisition time can be significantly reduced by more than one order of magnitude without compromising spatial resolution. Furthermore, we have extended the proposed speckle-based fly-scan phase tomography into the previously challenging high X-ray energy region (120 keV). This development opens up opportunities for a wide range of applications where exposure time and radiation dose are critical.

Data herein describe the quantitative changes in the plasma proteome in chickens challenged with lipopolysaccharide (LPS), a bacterial endotoxin known to stimulate the host innate immune system obtained by shotgun quantitative proteomic tandem mass tags approach using high-resolution Orbitrap technology. Statistical and bioinformatic analyses were performed to specify the effect of bacterial endotoxin. Plasma from chicken (N=6) challenged with Escherichia coli (LPS) (2 mg/kg body weight) was collected pre (0 h) and at 12, 24, 48, and 72 h post injection along with plasma from a control group (N=6) challenged with sterile saline. Protein identification and relative quantification were performed using Proteome Discoverer, and data were analysed using R. Gene Ontology terms were analysed by the Cytoscape application ClueGO based on Gallus gallus GO Biological Process database, and refined by REVIGO. Absolute quantification of several acute phase proteins, e.g. alpha-1-acid glycoprotein (AGP), serum amyloid A (SAA) and ovotrensferrin (OVT) was performed by immunoassays to validate the LC-MS results. The data contained within this article are directly related to our research article”Quantitative proteomics using tandem mass tags in relation to the acute phase protein response in chicken challenged with Escherichia coli lipopolysaccharide endotoxin” [1]. The proper use of agricultural extension and technological information service provides essential interventions required for sustainable agriculture in developing countries. This study provides insights into the availability and uses of agricultural information by small scale farmers in the Mbeya Region of Tanzania. The research used structured questionnaires to interview 240 rice farmers in the Mbeya region. The data were coded by Epidata and transferred to Stata statistical software for descriptive statistics analyzes. The findings revealed that farmers accessed agricultural information from various agents including local government extension staff, neighbours and friends, advertisements, electronic media including internet, televisions, radios, and other channels. The identified technological information application methods are by lectures, field demonstrations, exposure visits to various places, printed production, and others. Based on satisfaction levels, 43%, 25%, 20%, 8%, and 4% perceived to have dissatisfied, strongly dissatisfied, strongly satisfied, satisfied, and no opinions by farmers respectively. The majority of farmers claimed that both agricultural information and technological communication are strongly needed for agricultural performance. The study suggests that government and non-government organizations should collaborate together to bridge the existing information-sharing gap between farmers and information providers

Introduction Tourniquets are routinely used during total knee replacement (TKR) surgery. They could increase the risk of thromboembolic events including cerebral emboli, cognitive decline, pain and other adverse events (AEs). A randomised controlled trial to assess whether tourniquet use might safely be avoided is therefore warranted but it is unclear whether such a trial would be feasible.Methods In a single-site feasibility study and pilot randomised controlled trial, adults having a TKR were randomised to surgery with an inflated tourniquet versus a non-inflated tourniquet. Participants underwent brain MRI preoperatively and within 2 days postoperatively. We assessed cognition using the Mini-Mental State Examination (MMSE), Montreal Cognitive Assessment (MoCA) and Oxford Cognitive Screen (OCS) and thigh pain using a Visual Analogue Scale at baseline and days 1 and 2, and 1 week postsurgery. AEs related to surgery were recorded up to 12 months.Results We randomised 53 participants (27 tourniquet inflated and 26 tourniquet not inflated). Fifty-one participants received care per-protocol (96%) and 48 (91%) were followed up at 12 months. One new ischaemic brain lesion was detected. Of the cognitive tests, MoCA was easy to summarise, sensitive to change with lower ceiling effects compared with OCS and MMSE. There was a trend towards more thigh pain (mean 49.6 SD 30.4 vs 36.2 SD 28 at day 1) and more AEs related to surgery (21 vs 9) in participants with an inflated tourniquet compared with those with a tourniquet not inflated.Conclusion A full trial is feasible, but using MRI as a primary outcome is unlikely to be appropriate or feasible. Suitable primary outcomes would be cognition measured using MoCA, pain and AEs, all of which warrant investigation in a large multicentre trial.Trial registration number ISRCTN20873088.

Global habitat deterioration of marine ecosystems has led to a need for active interventions to halt or reverse the loss of ecological function. Restoration has historically been a key tool to reverse habitat loss and restore functions, but the extent to which this will be sufficient under future climates is uncertain. Emerging genetic technologies now provide the ability for restoration to proactively match adaptability of target species to predicted future environmental conditions, which opens up the possibility of boosting resistance to future stress in degraded and threatened habitats. As such, the choice of whether to restore to historical baselines or anticipate the future remains a key decision that will influence restoration success in the face of environmental and climate change. Here, we present an overview of the different motives for restoration – to recover or revive lost or degraded habitats to extant or historical states, or to reinforce or redefine for future conditions. We focus on the genetic and adaptive choices that underpin each option and subsequent consequences for restoration success. These options span a range of possible trajectories, technological advances and societal acceptability, and represent a framework for progressing restoration of marine habitat forming species into the future.

The construction of artificial structures, such as seawalls, is increasing globally, resulting in loss of habitat complexity and native species biodiversity. There is increasing interest in mitigating this biodiversity loss by adding topographic habitat to these structures, and/or seeding them with habitat-forming species. Settlement tile experiments, comparing colonisation of species to more and less complex habitats, have been used to inform eco-engineering interventions prior to their large-scale implementation. Most studies have focused on applying one type of intervention (either adding habitat structure or seeding with native organisms), so it is unclear whether there are greater benefits to biodiversity when multiple interventions are combined. Using a fully orthogonal experiment, we assessed the independent and interactive effects of habitat structure (flat vs. crevice/ridges) and seeding with native oysters (unseeded vs. seeded) on the biodiversity of four different functional groups (sessile and mobile taxa, cryptobenthic and pelagic fishes). Concrete tiles (flat unseeded, flat seeded, complex unseeded and complex seeded) were deployed at two sites in Sydney Harbour and monitored over 12 months, for the survival and colonisation of oysters and the species density and abundances of the four functional groups. The survival of seeded oysters was greater on the complex than flat tiles, at one of the two sites, due to the protective role of crevices. Despite this, after 12 months, the species density of sessile invertebrates and the percentage cover of seeded and colonising oysters did not differ between complex and seeded tiles each of which supported more of these variables than the flat unseeded tiles. In contrast, the species density of mobile invertebrates and cryptobenthic fishes and the MaxN of pelagic fishes, at 1 month, were only positively influenced by seeding with oysters, which provided food as well as habitat. Within the complex seeded and unseeded tiles, there was a greater species density of sessile taxa, survival and percentage cover of oysters in the crevices, which were more humid and darker at month 12, had lower high temperature extremes at months 1 and 12, than on the ridges or flat tiles. Our results suggest that eco-engineering projects which seek to maximise the biodiversity of multiple functional groups on seawalls, should apply a variety of different microhabitats and habitat-forming species, to alter the environmental conditions available to organisms.

Pharmaceutical medicine has evolved to be a distinct medical scientific discipline over time. Pharmaceutical medicine has distinctive features related to complex innovative medicines development activities in an often commercially focused competitive environment. This sometimes uneasy mix of professionalism and commercialisation demands of its medical and scientific researchers alike, a focus on strict adherence to ethical standards, guidelines, practices and behaviors in the interest of delivering new, effective, high-quality lifesaving and life-enhancing medicines quickly and reliably to patients in need. To support the speciality, codes of ethical standards and practices have been developed, with several being recently updated. These various codes are outlined in this paper along with relevant historical perspectives and interrelationship with concepts of professionalism. Reflecting the longer history of pharmaceutical medicine as a speciality in the UK and experience of the authors, there is a focus on the UK for the historical perspectives.

Canine babesiosis is an important tick-borne disease worldwide, caused by parasites of the Babesia genus. Although the disease process primarily affects erythrocytes, it may also have multisystemic consequences. The goal of this study was to explore and characterize the serum metabolome, by identifying potential metabolites and metabolic pathways in dogs naturally infected with Babesia canis using liquid and gas chromatography coupled to mass spectrometry. The study included 12 dogs naturally infected with B. canis and 12 healthy dogs. By combining three different analytical platforms using untargeted and targeted approaches, 295 metabolites were detected. The untargeted ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) metabolomics approach identified 64 metabolites, the targeted UHPLC-MS/MS metabolomics approach identified 205 metabolites, and the GC-MS metabolomics approach identified 26 metabolites. Biological functions of differentially abundant metabolites indicate the involvement of various pathways in canine babesiosis including the following: glutathione metabolism; alanine, aspartate, and glutamate metabolism; glyoxylate and dicarboxylate metabolism; cysteine and methionine metabolism; and phenylalanine, tyrosine, and tryptophan biosynthesis. This study confirmed that host–pathogen interactions could be studied by metabolomics to assess chemical changes in the host, such that the differences in serum metabolome between dogs with B. canis infection and healthy dogs can be detected with liquid chromatography-mass spectrometry (LC-MS) and gas chromatography-mass spectrometry (GC-MS) methods. Our study provides novel insight into pathophysiological mechanisms of B. canis infection.

Summary: Certain Gram-negative bacteria use the type VI secretion system (T6SS) to kill and lyse competing bacteria. Here, we show that the T6SS-dependent lysis of prey cells by the naturally competent Acinetobacter baylyi results in the extensive filamentation of a subpopulation of A. baylyi cells. Filamentation is dependent on the release of DNA from the prey and its uptake by the competence system. The analysis of A. baylyi transcriptome and the response of transcriptional reporters suggest that the uptake of DNA results in the upregulation of the SOS response, which often leads to cell-division arrest. Long-term competition between competent and non-competent strains shows that the strain lacking the DNA uptake machinery outcompetes the parental strain only in the presence of the T6SS-dependent lysis of prey cells. Our data suggest that the cost of the induced SOS response may drive the selection of tight regulation or the loss of DNA uptake in bacteria capable of lysing their competitors. : Lin et al. demonstrate that Acinetobacter baylyi uses its type VI secretion system to lyse competing bacteria. Subsequent DNA uptake induces SOS response and division arrest in A. baylyi. This results in slower growth of the competent bacteria during bacterial competition and highlights the importance of the regulation of DNA uptake. Abstract Tendon is a commonly injured soft musculoskeletal tissue, however, poor healing potential and ineffective treatment strategies result in persistent injuries and tissue that is unable to perform its normal physiological function. The identification of a stem cell population within tendon tissue holds therapeutic potential for treatment of tendon injuries. This study aimed, for the first time, to characterise and compare tenocyte and tendon-derived stem cell (TDSC) populations in murine tendon. Tenocytes and TDSCs were isolated from murine tail tendon. The cells were characterised for morphology, clonogenicity, proliferation, stem cell and tenogenic marker expression and multipotency. TDSCs demonstrated a rounded morphology, compared with a more fibroblastic morphology for tenocytes. Tenocytes had greater clonogenic potential and a smaller population doubling time compared with TDSCs. Stem cell and early tenogenic markers were more highly expressed in TDSCs, whereas late tenogenic markers were more highly expressed in tenocytes. Multipotency was increased in TDSCs with the presence of adipogenic differentiation which was absent in tenocytes. The differences in morphology, clonogenicity, stem cell marker expression and multipotency observed between tenocytes and TDSCs indicate that at least two cell populations are present in murine tail tendon. Determination of the most effective cell population for tendon repair is required in future studies, which in turn may aid in tendon repair strategies.

Bacteria use type VI secretion systems (T6SSs) to manipulate host cells during pathogenesis or to kill competing bacteria, which, in some cases, increases horizontal gene transfer. These functions largely depend on T6SS regulation, dynamics, and the set of effectors that the system delivers into the target cells. Here, we show that Acinetobacter baylyi ADP1 assembles a highly dynamic T6SS capable of killing and lysing bacterial cells. T6SS function depends on conserved T6SS components as well as Acinetobacter-specific genes of unknown function. Five different effectors, encoded next to VgrG or PAAR proteins and their cognate immunity proteins, cause distinct changes in the prey cells, resulting in various degrees of their lysis. Prey lysis correlates with the rate of DNA transfer from prey to predator, suggesting that lytic effectors are required for efficient T6SS-dependent horizontal gene transfer in naturally competent bacteria.

Herniation of the intervertebral disc (IVDH) is the most common cause of neurological and intervertebral disc degeneration-related diseases. Since the disc starts to degenerate before it can be observed by currently available diagnostic methods, there is an urgent need for novel diagnostic approaches. To identify molecular networks and pathways which may play important roles in intervertebral disc herniation, as well as to reveal the potential features which could be useful for monitoring disease progression and prognosis, multi-omics profiling, including high-resolution liquid chromatography-mass spectrometry (LC-MS)-based metabolomics and tandem mass tag (TMT)-based proteomics was performed. Cerebrospinal fluid of nine dogs with IVDH and six healthy controls were used for the analyses, and an additional five IVDH samples were used for proteomic data validation. Furthermore, multi-omics data were integrated to decipher a complex interaction between individual omics layers, leading to an improved prediction model. Together with metabolic pathways related to amino acids and lipid metabolism and coagulation cascades, our integromics prediction model identified the key features in IVDH, namely the proteins follistatin Like 1 (FSTL1), secretogranin V (SCG5), nucleobindin 1 (NUCB1), calcitonin re-ceptor-stimulating peptide 2 precursor (CRSP2) and the metabolites N-acetyl-D-glucosamine and adenine, involved in neuropathic pain, myelination, and neurotransmission and inflammatory response, respectively. Their clinical application is to be further investigated. The utilization of a novel integrative interdisciplinary approach may provide new opportunities to apply innovative diagnostic and monitoring methods as well as improve treatment strategies and personalized care for patients with degenerative spinal disorders.

Abstract Background Omega-3 long-chain (≥C20) polyunsaturated fatty acids (ω3 LC-PUFA) confer important attributes to health-conscious meat consumers due to the significant role they play in brain development, prevention of coronary heart disease, obesity and hypertension. In this study, the ω3 LC-PUFA content of raw and cooked Longissimus thoracis et lumborum (LTL) muscle from genetically divergent Australian prime lambs supplemented with dietary degummed crude canola oil (DCCO) was evaluated. Methods Samples of LTL muscle were sourced from 24 first cross ewe and wether lambs sired by Dorset, White Suffolk and Merino rams joined to Merino dams that were assigned to supplemental regimes of degummed crude canola oil (DCCO): a control diet at 0 mL/kg DM of DCCO (DCCOC); 25 mL/kg DM of DCCO (DCCOM) and 50 mL/kg DCCO (DCCOH). Abstract Background Regeneration of neurons in the central nervous system is poor in humans. In other vertebrates neural regeneration does occur efficiently and involves reactivation of developmental processes. Within the neural retina of zebrafish, Müller glia are the main stem cell source and are capable of generating progenitors to replace lost neurons after injury. However, it remains largely unknown to what extent Müller glia and neuron differentiation mirror development. Methods Following neural ablation in the zebrafish retina, dividing cells were tracked using a prolonged labelling technique. We investigated to what extent extrinsic feedback influences fate choices in two injury models, and whether fate specification follows the histogenic order observed in development. Results By comparing two injury paradigms that affect different subpopulations of neurons, we found a dynamic adaptability of fate choices during regeneration. Both injuries followed a similar time course of cell death, and activated Müller glia proliferation. However, these newly generated cells were initially biased towards replacing specifically the ablated cell types, and subsequently generating all cell types as the appropriate neuron proportions became re-established. This dynamic behaviour has implications for shaping regenerative processes and ensuring restoration of appropriate proportions of neuron types regardless of injury or cell type lost. Conclusions Our findings suggest that regenerative fate processes are more flexible than development processes. Compared to development fate specification we observed a disruption in stereotypical birth order of neurons during regeneration Understanding such feedback systems can allow us to direct regenerative fate specification in injury and diseases to regenerate specific neuron types in vivo.

ABSTRACT: Rice farming is characterized by various factors including environmental and non-environmental factors. The current paper analyses the influence of households’ characteristics, and perceptions of weather variability on rice yield. Authors used primary data collected from small-scale rice farmers in the Mbeya region of Tanzania. Garret technique and Stata software were used for data analyses. Results confirmed that farmer’s education, marital status, gender, and land ownership have a positive influence on rice yield. Thus, for every 1% increase in each variable increases rice yield by 14%, 98%, 26%, and 21% respectively. Owing to empirical results on farmers’ perceptions, it is confirmed that if the drought period increased by 1%, would on average reduce rice yield by 20%. The majority of farmers (70%) have perceived that drought period reduced their rice yield. In addition, farmers’ used averagely less quantity of rice seeds which have impaired their rice yields. If farmers increased the seed quantity by 1%, rice yields would increase by 17%. Our findings also revealed that the previous market price has positive association with the next seasons’ rice yield. Therefore, our study concluded that among households’ variables and their perceptions have positive implications on rice yield; however, with the empirical analyses other variables were insignificant due to multicollinearity effects. Authors recommended to the government and other responsible organizations to capitalize on irrigation infrastructures, agricultural extension and technological services to restraint the enduring food insecurity coercions in Tanzania.

Research into the identification and use of protein biomarkers for use in veterinary clinical pathology has produced numerous potential analytes that could become common tests in the future. One problem that has to be overcome in the general acceptance of a novel biomarker is that differing standards for calibration may be developed by individual laboratories or the diagnostic companies that will provide kits for widespread use. This has been apparent in the development of acute phase protein biomarkers such as canine C-reactive protein. In order to overcome this problem an international initiative is required to ensure that assays developed in separate laboratories would have a consistent calibration protocol so that results produced are equivalent. International reference preparations for serum protein analysis for each relevant species should be established for use as primary standard in the calibration of biomarkers for veterinary diagnosis.

Background: The tumor microenvironment is highly heterogeneous, and it is understood to affect tumor progression and patient outcome. A number of studies have reported the prognostic significance of tumor-infiltrating lymphocytes and tumor budding in colorectal cancer (CRC). However, the significance of the intratumoral heterogeneity present in the spatial distribution of these features within the tumor immune microenvironment (TIME) has not been previously reported. Evaluating this intratumoral heterogeneity may aid the understanding of the TIME's effect on patient prognosis as well as identify novel aggressive phenotypes which can be further investigated as potential targets for new treatment. Methods: In this study, we propose and apply two spatial statistical methodologies for the evaluation of the intratumor heterogeneity present in the distribution of CD3 + and CD8 + lymphocytes and tumor buds (TB) in 232 Stage II CRC cases. Getis-Ord hotspot analysis was applied to quantify the cold and hotspots, defined as regions with a significantly low or high number of each feature of interest, respectively. A novel spatial heatmap methodology for the quantification of the cold and hotspots of each feature of interest, which took into account both the interpatient heterogeneity and the intratumor heterogeneity, was further developed. Results: Resultant data from each analysis, characterizing the spatial intratumor heterogeneity of lymphocytes and TBs were used for the development of two new highly prognostic risk models. Conclusions: Our results highlight the value of applying spatial statistics for the assessment of the intratumor heterogeneity. Both Getis-Ord hotspot and our proposed spatial heatmap analysis are broadly applicable across other tissue types as well as other features of interest. The ratio of omega-6 (ω6) to omega-3 (ω3) polyunsaturated fatty acids (PUFAs) in the diet contributes to animal health and performance modulations because they have mostly opposite physiological functions. Increasing ω3 PUFAs content in the maternal diet can stimulate antioxidative capacity in sow and piglets; however, the optimal ratio of ω6 and ω3 PUFAs in the sow diet is still under discussion. Rich sources of bioactive constituents such as brown seaweed are an excellent supplementation to promote animal health and antioxidant status. However, the knowledge of the effects of this compound, specifically in post-weaning piglets, is still limited. Moreover, the combined effect of a low ω6:ω3 PUFAs ratio in sow diet and seaweed supplementation in post-weaning piglets’ diet has never been studied. This research aims to assess the combined effect of a low ω6:ω3 ratio in sow diets and seaweed supplementation in piglet diets on their growth and oxidative status. We also assessed the impact of a low ω6:ω3 ratio in the maternal diet on reproduction, milk fatty acid (FA) profile, and plasma leptin concentration. Two sow diets (n = 8 each) contained either a control ratio (CR, 13:1 during gestation, starting from day 28 (G28) and 10:1 during lactation) or a low ratio (LR, 4:1 from G28 until the end of lactation (L-End)) of ω6:ω3 FA by adding soybean oil or linseed oil, respectively. Reproductive performance was evaluated. Colostrum and milk at lactation day 7 (L7) and L-End were collected to analyze FA profile. Plasma was collected at G28, G79, G108, L7, L14, and L-End for determination of leptin and oxidative status. At weaning, 20 male piglets were selected per sow group to form 4 diet treatments (n = 10 each), which were supplemented with or without 4 g/kg seaweed. Recording of growth performance and collection of blood were performed at days 0, 7, 15, and 21 of post-weaning for oxidative status. LR diet increased (p < 0.05) the survival rate of piglets at weaning, and individual and litter weight gains. Colostrum and milk at L7 and L-End had lower (p < 0.05) ω6:ω3 ratio in LR sows. Interaction between dietary treatments on sows and piglets was revealed for all examined growth parameters at most time points (p < 0.05). LR diet did not affect plasma leptin levels and oxidative status. These findings suggest that the seaweed supplement during post-weaning could not improve growth rate and oxidative status of piglets born from mothers receiving a low dietary ω6:ω3 ratio (4:1) during gestation and lactation. However, this low ratio was beneficial for weaning survival rate, sucking piglets’ weight gain, and ω3 enrichment in colostrum and milk.

Teachers are simultaneously regarded as the most important influence on student learning and, paradoxically, as untrustworthy agents who can't be relied upon to deliver quality learning outcomes. Globally, this contested view of teaching is reflected in policies that limit teacher agency by prescribing how teaching will be conducted; for example, insisting on so-called evidence-based instructional models. Such policies also affect initial teacher education, which is increasingly policed for compliance with national teaching standards. However, teaching standards that reduce the teacher's role to ensuring fidelity to acceptable pedagogies have been widely criticised, with calls for teacher agency and professional judgement to be more highly valued. We endorse this critique and draw on Bakhtin's dialogic theory to illuminate data generated using collaborative autoethnography that reveals the worthwhile struggle for "good" initial teacher education.

Ligament is prone to injury and degeneration and has poor healing potential and, with currently ineffective treatment strategies, stem cell therapies may provide an exciting new treatment option. Ligament-derived stem cell (LDSC) populations have been isolated from a number of different ligament types with the majority of studies focussing on periodontal ligament. To date, only a few studies have investigated LDSC populations in other types of ligament, for example, intra-articular ligaments; however, this now appears to be a developing field. This literature review aims to summarise the current information on nondental LDSCs including in vitro characteristics of LDSCs and their therapeutic potential. The stem cell niche has been shown to be vital for stem cell survival and function in a number of different physiological systems; therefore, the LDSC niche may have an impact on LDSC phenotype. The role of the LDSC niche on LDSC viability and function will be discussed as well as the therapeutic potential of LDSC niche modulation.

Calcium binding proteins show stereotypical expression patterns within diverse neuron types across the central nervous system. Here, we provide a characterization of developmental and adult secretagogin-immunolabelled neurons in the zebrafish retina with an emphasis on co-expression of multiple calcium binding proteins. Secretagogin is a recently identified and cloned member of the F-hand family of calcium binding proteins, which labels distinct neuron populations in the retinas of mammalian vertebrates. Both the adult distribution of secretagogin labeled retinal neurons as well as the developmental expression indicative of the stage of neurogenesis during which this calcium binding protein is expressed was quantified. Secretagogin expression was confined to an amacrine interneuron population in the inner nuclear layer, with monostratified neurites in the center of the inner plexiform layer and a relatively regular soma distribution (regularity index > 2.5 across central-peripheral areas). However, only a subpopulation (~60%) co-labeled with gamma-aminobutyric acid as their neurotransmitter, suggesting that possibly two amacrine subtypes are secretagogin immunoreactive. Quantitative co-labeling analysis with other known amacrine subtype markers including the three main calcium binding proteins parvalbumin, calbindin and calretinin identifies secretagogin immunoreactive neurons as a distinct neuron population. The highest density of secretagogin cells of ~1800 cells / mm2 remained relatively evenly along the horizontal meridian, whilst the density dropped of to 125 cells / mm2 towards the dorsal and ventral periphery. Thus, secretagogin represents a new amacrine label within the zebrafish retina. The developmental expression suggests a possible role in late stage differentiation. This characterization forms the basis of functional studies assessing how the expression of distinct calcium binding proteins might be regulated to compensate for the loss of one of the others.

Dye-sensitized solar cells (DSSCs) have been intensely researched for more than two decades. Electrolyte formulations are one of the bottlenecks to their successful commercialization, since these result in trade-offs between the photovoltaic performance and long-term performance stability. The corrosive nature of the redox shuttles in the electrolytes is an additional limitation for industrial-scale production of DSSCs, especially with low cost metallic electrodes. Numerous electrolyte formulations have been developed and tested in various DSSC configurations to address the aforementioned challenges. Here, we comprehensively review the progress on the development and application of electrolytes for DSSCs. We particularly focus on the improvements that have been made in different types of electrolytes, which result in enhanced photovoltaic performance and long-term device stability of DSSCs. Several recently introduced electrolyte materials are reviewed, and the role of electrolytes in different DSSC device designs is critically assessed. To sum up, we provide an overview of recent trends in research on electrolytes for DSSCs and highlight the advantages and limitations of recently reported novel electrolyte compositions for producing low-cost and industrially scalable solar cell technology.

Legal philosophical discourse tends to be animated by some conception of self and the parameters of community. Reflecting a vast heritage of humanist philosophy and theology, western legal concepts reflect anthropocentric values. Theories of law and legal concepts promote human beings as separate to the environment and define frameworks for the exploitation of nature. Against this paradigm, environmental philosophers have sought to redefine human beings as integral members of a greater Earth community – nature is a community of subjects, not a collection of objects. This alternative conception of self carries important consequences for legal philosophy. This paper explores these consequences first by analysing the ecological conception of self and community articulated by ‘geologian’ Thomas Berry (1914-2009). Second, this paper uses Berry’s analysis to develop an ecological theory of jurisprudence. This theory connects human law with ecological integrity and holds that human law attains legal quality (in part) when enacted for the common good of the comprehensive Earth community. Throughout this analysis, the paper also highlights the limitations Berry’s philosophical and legal writing and seeks dialogue with leftist political theory. El discurso filosófico jurídico tiende a estimularse por una concepción del individuo y por parámetros de comunidad. Como reflejo de una vasta herencia de la filosofía humanista y la teología, los conceptos jurídicos occidentales reflejan valores antropocéntricos. Teorías del derecho y conceptos legales promueven seres humanos separados del medio ambiente y definen los marcos para la explotación de la naturaleza. Frente a este paradigma, los filósofos ambientales han tratado de redefinir los seres humanos como miembros de una comunidad de la Tierra más amplia – la naturaleza es una comunidad de sujetos, no una colección de objetos. Esta concepción alternativa del individuo conlleva importantes consecuencias para la filosofía del derecho. Este artículo explora estas consecuencias, primero mediante el análisis de la concepción ecológica del individuo y de la comunidad articulada por el geólogo Thomas Berry (1914-2009). En segundo lugar, este trabajo utiliza el análisis de Berry para desarrollar una teoría ecológica de la jurisprudencia. Esta teoría conecta el derecho humano con la integridad ecológica y sostiene que el derecho humano alcanza (en parte) la calidad jurídica cuando se promulga por el bien común de la comunidad global de la Tierra. Mesenchymal stem cells (MSC) are capable of multipotent differentiation into connective tissues and as such are an attractive source for autologous cell-based regenerative medicine and tissue engineering. Epigenetic mechanisms, like DNA methylation, contribute to the changes in gene expression in ageing. However there was a lack of sufficient knowledge of the role that differential methylation plays during chondrogenic, osteogenic and tenogenic differentiation from ageing MSCs. This study undertook genome level determination of the effects of DNA methylation on expression in engineered tissues from chronologically aged MSCs. We compiled unique DNA methylation signatures from chondrogenic, osteogenic, and tenogenic engineered tissues derived from young; n = 4 (21.8 years ± 2.4 SD) and old; n = 4 (65.5 years±8.3SD) human MSCs donors using the Illumina HumanMethylation 450 Beadchip arrays and compared these to gene expression by RNA sequencing. Unique and common signatures of global DNA methylation were identified. There were 201, 67 and 32 chondrogenic, osteogenic and tenogenic age-related DE protein-coding genes respectively. Findings inferred the nature of the transcript networks was predominantly for 'cell death and survival', 'cell morphology', and 'cell growth and proliferation'. Further studies are required to validate if this gene expression effect translates to cell events. Alternative splicing (AS) was dysregulated in ageing with 119, 21 and 9 differential splicing events identified in chondrogenic, osteogenic and tenogenic respectively, and enrichment in genes associated principally with metabolic processes. Gene ontology analysis of differentially methylated loci indicated age-related enrichment for all engineered tissue types in 'skeletal system morphogenesis', 'regulation of cell proliferation' and 'regulation of transcription' suggesting that dynamic epigenetic modifications may occur in genes associated with shared and distinct pathways dependent upon engineered tissue type. An altered phenotype in engineered tissues was observed with ageing at numerous levels. These changes represent novel insights into the ageing process, with implications for stem cell therapies in older patients. In addition we have identified a number of tissue-dependant pathways, which warrant further studies.

BACKGROUND:There has been considerable debate as to whether different modalities of simulated hypoxia induce similar cardiac responses. MATERIALS AND METHODS:This was a prospective observational study of 14 healthy subjects aged 22-35 years. Echocardiography was performed at rest and at 15 and 120 minutes following two hours exercise under normobaric normoxia (NN) and under similar PiO2 following genuine high altitude (GHA) at 3,375 m, normobaric hypoxia (NH) and hypobaric hypoxia (HH) to simulate the equivalent hypoxic stimulus to GHA. RESULTS:All 14 subjects completed the experiment at GHA, 11 at NN, 12 under NH, and 6 under HH. The four groups were similar in age, sex and baseline demographics. Sheep and cattle farmers need pastoral systems that are more productive and environmentally sustainable. The role that high feeding value herb and legume forage species can play in the farms of the future is highlighted. It is shown that species such as chicory (Cichorium intybus), plantain (Plantago lanceolata), red clover (Trifolium pratense) and white clover (T.repens) can provide live weight gains in lambs that are 70% greater than those from perennial ryegrass (Lolium perenne) based pastures. Macroalgal surfaces support abundant and diverse microorganisms within biofilms, which are often involved in fundamental functions relating to the health and defence of their seaweed hosts, including algal development, facilitation of spore release and chemical antifouling. Given these intimate and important interactions, environmental changes have the potential to negatively impact macroalgae by disrupting seaweed-microbe interactions. We used the disappearance of the dominant canopy-forming fucoid Phyllospora comosa from the metropolitan coast of Sydney, Australia as a model system to study these interactions. We transplanted Phyllospora individuals from nearby, extant populations back onto reefs in Sydney to test whether bacterial assemblages associated with seaweed surfaces would be influenced by (i) the host itself, independently of where it occurs, (ii) the type of habitat where the host occurs or (iii) site-specific differences. Analyses of bacterial DNA fingerprints (TRFLPs) indicated that assemblages of bacteria on Phyllospora were not habitat-specific. Rather, they were primarily influenced by local, site-specific conditions with some evidence for host-specificity in some cases. This could suggest a lottery model of host-surface colonisation, by which hosts are colonised by ‘suitable’ bacteria available in the local species pool, resulting in high variability in assemblage structure across sites, but where some species in the community are specific to the host and possibly influenced by differences in host traits.
