To overcome the environmental changes occurring now and predicted for the future, it is essential that fruit breeders develop cultivars with better physiological performance. During the last few decades, high-throughput plant phenotyping and phenomics have been developed primarily in cereal breeding programs. In this study, plant reflectance, at the level of the leaf, was used to assess several physiological traits in five Vaccinium spp. cultivars growing under four controlled conditions (no-stress, water deficit, heat stress, and combined stress). Two modeling methodologies [Multiple Linear Regression (MLR) and Partial Least Squares (PLS)] with or without (W/O) prior wavelength selection (multicollinearity, genetic algorithms, or in combination) were considered. PLS generated better estimates than MLR, although prior wavelength selection improved MLR predictions. When data from the environments were combined, PLS W/O gave the best assessment for most of the traits, while in individual environments, the results varied according to the trait and methodology considered. The highest validation predictions were obtained for chlorophyll a/b (R2Val ≤ 0.87), maximum electron transport rate (R2Val ≤ 0.60), and the irradiance at which the electron transport rate is saturated (R2Val ≤ 0.59). The results of this study, the first to model modulated chlorophyll fluorescence by reflectance, confirming the potential for implementing this tool in blueberry breeding programs, at least for the estimation of a number of important physiological traits. Additionally, the differential effects of the environment on the spectral signature of each cultivar shows this tool could be directly used to assess their tolerance to specific environments.

Abstract  Beyond their participation in fruit pigmentation and because of their high antioxidant activity, flavonoids are considered important constituents of fruits and vegetables. We have previously reported that in the ripe receptacles of Fragaria chiloensis only traces of flavonoids can be found, while cinnamic acid derivatives are highly accumulated. In order to characterize the molecular background of this uncommon phenotype we analyzed the transcriptional profile of different biosynthetic genes, with special regard to the gene encoding Cinnamate 4-Hydroxylase (C4H), the enzyme transforming cinnamic acid into the next intermediary of the phenylpropanoid pathway. Northern blot and quantitative RT-PCR showed low transcript abundance for the gene encoding C4H and also for a series of structural genes responsible for flavonoid biosynthesis. Together with this, high transcript levels were found for a repressive transcription factor, suggesting that the pathway would be inhibited at the transcriptional level, thus correlating to our previous findings on the chemical phenotype. Our results contribute to the comprehension of the pigmentation phenotype in strawberries, allowing the utilization of Fragaria chiloensis as a model system for the study of antioxidant pigment biosynthesis. Abstract  In Chile Vasconcellea pubescens is cropped to produce canned fruit, juice, jam and processed sweets. Additionally this species produces latex with a high level of papain, an important and valuable proteolytic enzyme with industrial applications. In this investigation seven ISSR primers were used to study the level and organization of genetic diversity in 333 samples of V. pubescens. Out of the 114 bands recorded, 63 proved to be polymorphic (P = 55.3%). At the species level, the genetic diversity was rather low (h = 0.01 ± 6,80188E-05, Shannon’s Index I = 0.16 ± 0,000148). The major portion of the genetic diversity was found within groups (65%). The genetic differentiation between the different groups was significant, as the AMOVA analysis suggested (Φpt = 0.35). When analysing the Northern area alone, the differentiation increased to Φpt = 0.40. When only the Southern area was analysed, Φpt decreased to 0.18, indicating greater genetic similarity among the samples. The results generated from Structure and Bayesian Analysis of Population Structure distinguished 8 genetically different groups, five of them located in the north and three in the south. Gomortega keule (Molina) Baillon is an endangered, rare species, the only representative of its genus, and endemic to Central Chile. Populations of this tree are now fragmented and few individuals can be found in any of them. Genetic diversity was studied in 33 individuals from three populations in Cauquenes, a coastal mountain area (35°58'S-72°41'W). Fifteen InterSimple Sequence Repeat primers were used to determine the degree of similarity between and within populations. This revealed that 30% of the variation exhibited was between populations while 70% was within; nevertheless individuals were clearly clustered in a pattern which reflected a narrow base of diversity. Three other species from the Laurales order were used in order to provide an external reference as to the degree of diversity. In addition, an external wild population from the native species, Peumus boldus, was used to verify the utility of the markers. We show that the primers are effective in quickly giving an estimate of the degree of diversity of a population, thus giving important topical information relevant to preserving endangered species. Aspects of the conservation and management policy for the species in order to maintain the remaining populations and to preserve the genetic resources are discussed. [ABSTRACT FROM AUTHOR]
