Abstract

Parasites generally have a number of different strategies for avoiding destruction by the immune system of their host: the production of immune regulators, special structural characteristics, and the mimicry of host molecular structures. Unfortunately, the host sometimes contributes to this avoidance by developing ineffective responses. In the case of the parasite Schistosoma mansoni, an infected individual develops an immune response, which is only partially effective. Working with laboratory mice, investigators have now shown that the anti-idiotype network may regulate antibodies to the disadvantage of a host infected with the agent for schistosomiasis mansoni. Idiotypes are the lowest common denominator of antibody molecules; a particular antibody molecule has unique regions which distinguish it from all other antibody molecules, save those which descend from the same original B cell. These unique idiotypes can act as antigens in their own right, and may produce their own immune response. Antibodies directed against the idiotypic region of another antibody molecule are called anti-idiotypes, and they are widely thought to play a role in a complicated network of antibody control. If an antibody has reached the appropriate level in the blood, the production of anti-idiotypes may prevent overproduction. Researchers can experimentally produce anti-idiotypes by injecting stimulated lymphoblasts from another mouse of the same genetic background. When such a mouse, now possessing what researchers have dubbed 'auto-anti-idiotypes', is injected with killed Schistosoma organisms, it fails to mount an adequate immune response. Not only does it fail to produce a normal antibody response, but the response of the cellular immune system to Schistosoma antigens is inadequate as well. These findings suggest that it is not possible to design a vaccine against such parasites without consideration of the anti-idiotypic network of regulation. However, the results also suggest that it may well be possible, by fine-tuning the regulatory network, to produce successful vaccines against organisms which have heretofore been elusive. 30. The Regulation of Resistance to Schistosoma mansoniby a Soluble α/β TCR Analog, Produced by a Cloned T Cell Hybridoma

Abstract

Immunity to Schistosoma mansoniis regulated by an auto-anti-idiotypic network. The T cells in this network interact through soluble regulatory factors. These studies describe a soluble suppressor-inducer factor (TsiF) derived from TCR+suppressor-inducer clone Q37, Q37 is the fusion product of CD4+T cells derived from S. mansoni-infected mice and TCR-, BW1100 thymoma cells. [35S]Methionine biosynthetically labeled TsiF was immunoprecipitated with mAb H57-597 (anti-pan α/β TCR) and adsorbed with protein G-agarose. The eluted 72-kDa TCR analog was reduced with dithiothreitol. Components of 32 and 44 kDa were found on SDSPAGE. These components were analogous to the α and β chains of the TCR, binding antigen, or expressing Vβ determinants, respectively. TsiF suppressed delayed-type hypersensitivity to S. mansoniantigens in a genetically and antigenically restricted manner. TsiF totally abrogated protective immune response against S. mansoni. These soluble TCR analogs may idiotypically regulate the resistance to S mansoni.Q37 is an appropriate source of a suppressor TCR analog for the molecular analyses of this idiotypic regulation.

33. The cinnamic acid pathway and hispidin biosynthesis in cultures of Polyporus hispidus Fries

Abstract

The biosynthesis of hispidin, 6-(3,4-dihydroxystyryl)-4-hydroxy-2-pyrone, was examined in cultures of Polyporus hispidus Fr. Cultural studies were undertaken to determine the most suitable medium for investigating the biosynthesis of this pigment. These studies showed that light was necessary for hispidin formation and that the development of basidiocarps with viable spores could be achieved on agar media. On the liquid medium employed for biochemical studies, the maximum rate of hispidin production was observed to lag the maximum rate of growth by about five days. Trimethylhispidin, 4-methylhispidin and yangonin were synthesized for comparative purposes and for dilution in tracer experiments. These and numerous other phenolic and aromatic compounds were employed as references in examinations of nonradioactive culture extracts. P-coumaric, caffeic, p-hydroxybenzoic, protocatechuic and o- and p-hydroxyphenyl-acetic acids were detected in extracts of the culture medium. Bis-noryangonin (6-(4-hydroxystyryl)-4-hydroxy-2-pyrone) and other possible styrylpyrones were detected in extracts of the mycelium. Tracer experiments established that phenylalanine was metabolized to cinnamic, benzoic, p-hydroxybenzoic and protocatechuic acids. The incorporation of radioactivity into phenyllactic, phenylpyruvic, phenylacetic and p-hydroxy-phenylacetic acids also was observed. Furthermore, p-coum-aric acid, caffeic acid and hispidin incorporated radioactivity from phenylalanine. Hispidin also was shown to incorporate radioactivity from tyrosine, cinnamic acid, p-coum-aric acid, caffeic acid, malonic acid and sodium acetate. Degradation of the labelled hispidin obtained from these precursors confirms the hypothesis that this molecule is bio-synthesized from a phenylpropanoid moiety with the addition of two equivalents of acetate. Crude and partially purified preparations of several enzymes related to aromatic metabolism were obtained. Phenylalanine and tyrosine ammonia-lyase activity were demonstrated in cell-free preparations. Maximum phenylalanine ammonia-lyase activity was obtained from cultures during the logarithmic phase of growth. Enzymes capable of hydroxylating cinnamic acid, benzoic acid and bis-noryangonin also were obtained in vitro.
