This paper shows how the Open5GENESIS suite can be used for executing 5G experiments at rural and underserved areas, integrating technologies such as edge computing and satellite backhaul. The use case considered is a smart farming application for weed detection, realized by means of Virtualized Network Functions and tailored functions. The 5GENESIS experimentation methodology is summarized, showing its flexibility for orchestrating, managing and retrieving metrics of this particular experiment. Results show that the use of edge computing in the use case considered reduces the end-to-end latency significantly.

Although rodents are arguably the easiest animals to use for studying brain function, relying on them as model species for translational research comes with its own set of limitations. Here, we propose sheep as a practical large animal species to use for in vivo brain function studies performed in naturalistic settings. We conducted proof-of-principle deep brain electrophysiological recording experiments using unrestrained sheep during behavioral testing. Recordings were made from cortex and hippocampus, both while sheep performed goal-directed behaviors (two-choice discrimination tasks) and across states of vigilance, including sleep. Hippocampal and cortical oscillatory rhythms were consistent with those seen in rodents and non-human primates, and included cortical alpha oscillations and hippocampal sharp wave ripple oscillations (∼150 Hz) during immobility and hippocampal theta oscillations (5-6 Hz) during locomotion. Recordings were conducted over a period of many months during which time the animals participated willingly in the experiments. Over 3,000 putative neurons were identified, including examples whose activity was modulated by task, speed of locomotion, spatial position, reward and vigilance states, and one whose firing rate was potentially modulated by the sight of the investigator. Together, these experiments demonstrate that sheep are excellent experimental animals to use for longitudinal studies requiring a large-brained mammal and/or large-scale recordings across distributed neuronal networks. Sheep could be used safely for studying not only neural encoding of decision-making and spatial-mapping in naturalistic environments outside the confines of the traditional laboratory but also the neural basis of both intra- and inter-species social interactions.

Recordings were conducted over a period of many months. Due to the exceptional stability of individual electrodes we were able to record from some neurons repeatedly across the span of a month. Together these experiments demonstrate that sheep are an excellent experimental animal model to use in longitudinal electrophysiological and imaging studies, particularly those requiring a large brained mammal, large scale recordings across distributed neuronal networks, experimentation outside the confounds of the traditional laboratory, or all the above concomitantly.

Pathogenic variants in SYNGAP1 are one of the most common genetic causes of nonsyndromic intellectual disability (ID) and are considered a risk for autism spectrum disorder (ASD). SYNGAP1 encodes a synaptic GTPase activating protein that modulates the intrinsic GTPase activity of several small G-proteins and is implicated in regulating the composition of the postsynaptic density. By targeting the deletion of exons encoding the calcium/lipid binding (C2) and GTPase activating protein (GAP) domains, we generated a novel rat model to study SYNGAP related pathophysiology. We find that rats heterozygous for the C2/GAP domain deletion (Syngap+/Δ-GAP) exhibit reduced exploration and fear extinction, altered social behaviour, and spontaneous seizures, while homozygous mutants die within days after birth. This new rat model reveals that the enzymatic domains of SYNGAP are essential for normal brain function and provide an important new model system in the study of both ID/ASD and epilepsy.

The advent of implantable active dense CMOS neural probes opened a new era for electrophysiology in neuroscience. These single shank electrode arrays, and the emerging tailored analysis tools, provide for the first time to neuroscientists the neurotechnology means to spatiotemporally resolve the activity of hundreds of different single-neurons in multiple vertically aligned brain structures. However, while these unprecedented experimental capabilities to study columnar brain properties are a big leap forward in neuroscience, there is the need to spatially distribute electrodes also horizontally. Closely spacing and consistently placing in well-defined geometrical arrangement multiple isolated single-shank probes is methodologically and economically impractical. Here, we present the first high-density CMOS neural probe with multiple shanks integrating thousand’s of closely spaced and simultaneously recording microelectrodes to map neural activity across 2D lattice. Taking advantage from the high-modularity of our electrode-pixels-based SiNAPS technology, we realized a four shanks active dense probe with 256 electrode-pixels/shank and a pitch of 28 µm, for a total of 1024 simultaneously recording channels. The achieved performances allow for full-band, whole-array read-outs at 25 kHz/channel, show a measured input referred noise in the action potential band (300-7000 Hz) of 6.5 ± 2.1µVRMS, and a power consumption µW/electrode-pixel. Preliminary recordings in awake behaving mice demonstrated the capability of multi-shanks SiNAPS probes to simultaneously record neural activity (both LFPs and spikes) from a brain area >6 mm2, spanning cortical, hippocampal and thalamic regions. High-density 2D array enables combining large population unit recording across distributed networks with precise intra- and interlaminar/nuclear mapping of the oscillatory dynamics. These results pave the way to a new generation of high-density and extremely compact multi-shanks CMOS-probes with tunable layouts for electrophysiological mapping of brain activity at the single-neurons resolution.

Satellite/terrestrial integration in the context of 5G is a very promising aspect, as it combines the unrivaled performance of 5G with the unprecedented benefits of satellite communications, such as ubiquitous broadband coverage and inherent multicast capabilities. This paper presents the design and implementation of an end-to-end experimental testbed for integrated satellite/terrestrial 5G services, developed in the frame of the EU 5GENESIS project. The testbed encompasses all the components of the 5G network and it is suitable for wide-area field trials over several use cases corresponding to the needs of vertical industries.

Background For reasons of cost and ethical concerns, models of neurodegenerative disorders such as Huntington disease (HD) are currently being developed in farm animals, as an alternative to non-human primates. Developing reliable methods of testing cognitive function is essential to determining the usefulness of such models. Nevertheless, cognitive testing of farm animal species presents a unique set of challenges. The primary aims of this study were to develop and validate a mobile operant system suitable for high throughput cognitive testing of sheep. New method We designed a semi-automated testing system with the capability of presenting stimuli (visual, auditory) and reward at six spatial locations. Fourteen normal sheep were used to validate the system using a two-choice visual discrimination task. Four stages of training devised to acclimatise animals to the system are also presented. Results All sheep progressed rapidly through the training stages, over eight sessions. All sheep learned the 2CVDT and performed at least one reversal stage. The mean number of trials the sheep took to reach criterion in the first acquisition learning was 13.9 ± 1.5 and for the reversal learning was 19.1 ± 1.8. Comparison with existing method(s) This is the first mobile semi-automated operant system developed for testing cognitive function in sheep. Conclusions We have designed and validated an automated operant behavioural testing system suitable for high throughput cognitive testing in sheep and other medium-sized quadrupeds, such as pigs and dogs. Sheep performance in the two-choice visual discrimination task was very similar to that reported for non-human primates and strongly supports the use of farm animals as pre-clinical models for the study of neurodegenerative diseases.

With their large brains and long lives, sheep offer advantages over rodents for studying progressive neurodegenerative disease in timeframes relevant to humans. Perentos et al. demonstrate the validity of an ovine model of Batten disease by revealing in vivo EEG abnormalities that resemble those in affected children., Creating valid mouse models of slowly progressing human neurological diseases is challenging, not least because the short lifespan of rodents confounds realistic modelling of disease time course. With their large brains and long lives, sheep offer significant advantages for translational studies of human disease. Here we used normal and CLN5 Batten disease affected sheep to demonstrate the use of the species for studying neurological function in a model of human disease. We show that electroencephalography can be used in sheep, and that longitudinal recordings spanning many months are possible. This is the first time such an electroencephalography study has been performed in sheep. We characterized sleep in sheep, quantifying characteristic vigilance states and neurophysiological hallmarks such as sleep spindles. Mild sleep abnormalities and abnormal epileptiform waveforms were found in the electroencephalographies of Batten disease affected sheep. These abnormalities resemble the epileptiform activity seen in children with Batten disease and demonstrate the translational relevance of both the technique and the model. Given that both spontaneous and engineered sheep models of human neurodegenerative diseases already exist, sheep constitute a powerful species in which longitudinal in vivo studies can be conducted. This will advance our understanding of normal brain function and improve our capacity for translational research into neurological disorders.

UNLABELLED: Sheep have large brains with human-like anatomy, making them a useful species for studying brain function. Sleep homeostasis has not been studied in sheep. Here, we establish correlates of sleep homeostasis in sheep through a sleep deprivation experiment. We then use these correlates to elucidate the nature of sleep deficits in a naturally occurring ovine model of neuronal ceroid lipofuscinosis (NCL, Batten disease) caused by a mutation in CLN5 In humans, mutations in this gene lead to cortical atrophy and blindness, as well as sleep abnormalities. We recorded electroencephalograms (EEGs) from unaffected and early stage CLN5(-/-) (homozygous, affected) sheep over 3 consecutive days, the second day being the sleep deprivation day. In unaffected sheep, sleep deprivation led to increased EEG delta (0.5-4 Hz) power during non-rapid eye movement (NREM) sleep, increased time spent in the NREM sleep state, and increased NREM sleep bout length. CLN5(-/-) sheep showed comparable increases in time spent in NREM sleep and NREM sleep bout duration, verifying the presence of increased sleep pressure in both groups. Importantly, CLN5(-/-) sheep did not show the increase in NREM sleep delta power seen in unaffected sheep. This divergent delta power response is consistent with the known cortical degeneration in CLN5(-/-) sheep. We conclude that, whereas sleep homeostasis is present in CLN5(-/-) sheep, underlying CLN5(-/-) disease processes prevent its full expression, even at early stages. Such deficits may contribute to early abnormalities seen in sheep and patients and warrant further study. SIGNIFICANCE STATEMENT: Sleep abnormalities pervade most neurological diseases, including the neuronal ceroid lipofuscinoses (NCLs). Here, we show that, in an ovine model of a variant late-infantile NCL, there is abnormal expression of sleep homeostasis. Whereas some sleep pressure correlates respond to sleep deprivation, the strongest electroencephalogram (EEG) correlate of sleep pressure, non-REM delta power, failed to increase. This highlights the relevance of sleep deficits in this disease, in which the drive for sleep exists but the underlying disease prevents its full expression. Sleep abnormalities could contribute to early disease symptoms such as behavioral disorder and cognitive decline. Our study also shows sleep homeostatic EEG correlates in sheep, opening up new opportunities for studying sleep in a large social mammal with complex human-like brain neuroanatomy.

A dual-loop optoelectronic oscillator incorporating an optoelectronic recirculating delay line in order to circumvent the limitations of optical coherence associated with all-optical loops is demonstrated. The optoelectronic oscillator produces a very stable signal at 5.4 GHz (suitable for WLAN 802.11n and 4G-WiMAX systems) with a sub-Hz 3-dB bandwidth and a Q-factor in excess of 1010. A phase noise of −115 dBc/Hz is recorded at 10 kHz offset, owing to the reduction in phase induced intensity noise, whilst side modes are significantly suppressed for offset values in the range 100 kHz – 1 MHz.

We experimentally demonstrate a 60 km long radio-over-fiber link for transmission at 30 GHz with error-free transmission up to 1 Gb/s.

A dual-loop OEO using a SOA-based RDL is presented. The active loop creates a high Q-factor whilst the dual-loop suppresses side-modes. The OEO operates in the incoherent regime and in a proof-of-principle experiment produces a very stable 5.38 GHz oscillation with a Q-factor of 1010.

A WDM optoelectronic oscillator (OEO) based on a cascaded optical multi-loop configuration and multiple photodiodes is proposed and demonstrated experimentally. By employing up to three lasers widely separated in wavelength along with two cascaded multi-loop fiber sections and two photodiodes, we demonstrate OEO topologies that scale up to six effective loops revealing an ultra-high quality factor in excess of 1010 and a phase noise performance down to −119 dBc/Hz at 10 kHz offset

Rumination is a precisely timed process that occupies a large part of a sheep's day. The complex motor coordination required to chew and swallow means that quantification of rumination may provide a surrogate marker for effective motor function. Here, data from 24h in vivo electrophysiological recordings, collected as part of an earlier study, were reanalysed for chewing- and swallowing-related activity. The electroencephalographic (EEG) and electromyographic (EMG) data were collected from sheep with surgically-implanted electrodes. An algorithm was designed to detect coordinated, rhythmic muscle activity. This could distinguish episodes of eating from those of rumination. Normal sheep spent ~29% of their time ruminating. Rumination comprised ~40s bouts of regular (~1.7s(-1)) chewing interspersed by ~6.5s intervals during which time no chewing took place. Eating was significantly less regular than rumination, with quicker chewing (~2.7s(-1)). Biomarkers for measuring progression of disease would be invaluable for studying neurodegenerative disease such as Huntington's disease (HD). To test the feasibility of using rumination as such a biomarker, we also made recordings from two neurologically impaired sheep. These showed deviations from the pattern of rumination and eating seen in normal sheep. This validates not only our use of rumination as a measure of normal motor function, but also as a surrogate biomarker for measuring motor dysfunction in impaired sheep.

In this paper, we experimentally demonstrate the benefit of polarization insensitive dual-band optical phase conjugation for up to ten 400 Gb/s optical super-channels using a Raman amplified transmission link with a realistic span length of 75 km. We demonstrate that the resultant increase in transmission distance may be predicted analytically if the detrimental impacts of power asymmetry and polarization mode dispersion are taken into account.

An optoelectronic oscillator topology based on a class AB analogue optical link is proposed. The motivation for this approach is based on the unique property of class AB links for mitigating both shot noise and relative intensity noise contributions. The class AB optoelectronic oscillator is compared with a conventional single loop optoelectronic oscillator.
