20. Optical Fiber pH Sensors Based on PAni-Coated Microstructured Optical Fibers

Abstract

In this article, we report the analysis of a novel optical fiber pH sensor based in polyaniline (PAni) coating applied on a trenched core-free (only-bridge) silica fiber. The results are compared to another pH sensor based on side-polished silica fiber. The pH-sensitive polymer is synthesized over the fiber sample by means of oxidative polymerization, keeping records of the time where the reaction occurs to improve the sensitivity. The response to pH is enhanced in the case of the trenched core-free bridge fiber due to the higher interaction of the evanescent field with the PAni film and, thus, the surrounding media. The sensitivity in the linear zone of operation is 1.1 mW/pH, and a total transmittance of 55% in the pH range is 4.2–8.1. The repeatability of both sensors was checked, showing a high capability to perform studies in liquid samples due to its temperature independence, good sensitivity, and long-term stability.

Abstract

Udotea is an exclusively marine green macroalgal genus with wide distribution in both tropical and subtropical regions. Due to the difficulty in properly delimiting infrageneric taxa based on morpho-anatomical characteristics by the presence of cryptic species and phenotypic plasticity, the use of techniques of DNA barcode and phylogenetic analysis have been fundamental to assess the Udotea species diversity. Information concerning this genus in Brazil is currently restricted to a few publications in checklists and/or regional floristic surveys along the tropical coast. This study aimed to evaluate the diversity of Udotea in northeastern Brazil through morphological-based taxonomy allied with plastid molecular markers, tufA and rbcL. We generated phylogenies of Udotea sensu lato, and we used different species delimitation methods to assess the diversity among sequences of selected tufA clades within Udotea sensu stricto. None of the eight infrageneric taxa reported for Brazil based only on morphology were confirmed by using molecular data. Udotea dotyi was revealed by tufA and rbcL sequences and represents its first occurrence for the western south Atlantic Ocean. A taxon named here as ‘Udotea cyathiformis’ requires further studies for its definition. This first molecular study on Udotea from the Brazilian coast shows that a broader sampling will be necessary to assess the true diversity of the genus in the western south Atlantic, which may be overestimated due to its high morphological plasticity.

37. Effects of the rainy season and urbanization on the phenology and richness of the community of Dictyotales (Phaeophyceae) on Brazilian tropical reefs

Abstract

The phycological diversity of Dictyotales, macroalgal species with great biotechnological potential, is still unknown in Alagoas, Brazil. Therefore, we carried out a phenological survey of Dictyotales and provided ecological comments. Samples were collected at four stations during the dry season (November and December 2013 and January 2014) and the rainy season (May–July 2014). Eighteen species were identified, five of which were recorded for the first time in Alagoas (Canistrocarpus crispatus, Dictyopteris jamaicensis, D. polypodioides, Dictyota pinnatifida and Padina antillarum). Canistrocarpus cervicornis and Dictyopteris delicatula made the highest biomass contribution in all the stations in both seasons. The stations studied showed differences between them, except for stations with less anthropogenic interference and offer greater local species diversity. This highlights that in Maragogi and Pajuçara Stations aspects of urbanization could negatively affect the Dictyotales community, such as the disappearance of endemic species. Sporophyte phase and fertility showed differences between stations and seasons. In both analyses, the rainy season collected the most of the records. The analysis of richness showed that the rainy season had higher values. Thus, the rainy season is the time when Dictyotales invest in reproduction and we find higher biodiversity. Therefore, phenological studies, biodiversity research and commercial research of Dictyotales should take place during the rainy season.

Abstract

This thesis is composed of four empirical studies which use data from Portugal, the United Kingdom and Germany to examine four topical aspects of employment, careers and productivity in these countries' labour markets. The second chapter studies the impact that a 49.3% change in the legal minimum wage for workers aged 18 and 19 in Portugal had on the wages and employment of this age group of workers. It uses firm-level micro data to compare the employment growth of 18-19 year old workers with employment growth of older workers. It also looks separately at firms more and less likely to be affected by the minimum wage shock. The third chapter studies the impact of foreign direct investment (FDI) on the productivity of domestic firms in the UK. It uses a plant-level panel covering UK manufacturing to find evidence of FDI spillovers. It does so by investigating the correlation between a domestic plant's TFP and the foreign-affiliate share of employment in that plant's industry and, independently, in that plant's region. A number of different specifications are estimated in order to minimise potential endogeneity bias. The fourth chapter estimates returns to job tenure and labour market experience in the United Kingdom and Germany using various methods to correct for heterogeneity and endogeneity biases. It also estimates the returns to tenure and experience by qualification group. Results are interpreted in light of the differences between the two labour market's institutions. The fifth chapter compares returns to tenure and experience in union and non-union jobs in the United Kingdom in the 80s and 90s. It uses longitudinal data and instrumental variables methods to correct for potential individual and job match heterogeneity biases. Returns are also calculated separately for jobs with and without seniority wage scales.

Abstract

Therapies for bladder cancer patients are limited by side effects and failures, highlighting the need for novel targets to improve disease management. Given the emerging evidence highlighting the key role of innate lymphoid cell subsets, especially type 2 innate lymphoid cells (ILC2s), in shaping the tumor microenvironment and immune responses, we investigated the contribution of ILC2s in bladder tumor development. Using the orthotopic murine MB49 bladder tumor model, we found a strong enrichment of ILC2s in the bladder under steady-state conditions, comparable to that in the lung. However, as tumors grew, we observed an increase in ILC1s but no changes in ILC2s. Targeting ILC2s by blocking IL-4/IL-13 signaling pathways, IL-5, or IL-33 receptor, or using IL-33-deficient or ILC2-deficient mice, did not affect mice survival following bladder tumor implantation. Overall, these results suggest that ILC2s do not contribute significantly to bladder tumor development, yet further investigations are required to confirm these results in bladder cancer patients. Abstract

Molecular phylogenetic studies have revealed the polyphyletic nature of Cladophora as traditionally circumscribed, which is separated into four main clades: Cladophoraceae, Siphonocladus, Pithophoraceae and Pseudocladophoraceae. Several taxonomic proposals have recently been made to resolve its polyphyletic nature. The present work focuses on the cryptic diversity of Cladophora coelothrix, within the Siphonocladus clade. Based on new collections from northeastern Brazil and phylogenetic analyses of partial SSU and LSU rDNA sequence alignments, we propose the new genus and species Leliaertia repens to accommodate one of the cryptic species hitherto included in C. coelothrix, occurring in the tropical Atlantic, Indian and Pacific Oceans. Leliaertia repens was recovered in the Siphonocladus clade and is phylogenetic distinct from the genuine Cladophora species in the Cladophoraceae. The new species occurs in marine and estuarine environments and forms conspicuous mats of semi-prostrate filaments. It represents a cryptic species and genus since morphological features, including the diameter and length:diameter ratio of the apical cell, show high levels of overlap with other species, including C. socialis. Our phylogenetic data indicate that L. repens, C. coelothrix and C. socialis represent a complex of cryptic species with at least four lineages, some of which co-occur in tropical seas.
