15. LAP1 Interactome Profiling Provides New Insights into LAP1's Physiological Functions.

Abstract

The nuclear envelope (NE), a protective membrane bordering the nucleus, is composed of highly specialized proteins that are indispensable for normal cellular activity. Lamina-associated polypeptide 1 (LAP1) is a NE protein whose functions are just beginning to be unveiled. The fact that mutations causing LAP1 deficiency are extremely rare and pathogenic is indicative of its paramount importance to preserving human health, anticipating that LAP1 might have a multifaceted role in the cell. Mapping the LAP1 protein interactome is, thus, imperative to achieve an integrated view of its potential biological properties. To this end, we employed in silico- and mass spectrometry-based approaches to identify candidate LAP1-interacting proteins, whose functional attributes were subsequently characterized using bioinformatics tools. Our results reveal the complex and multifunctional network of protein–protein interactions associated to LAP1, evidencing a strong interconnection between LAP1 and cellular processes as diverse as chromatin and cytoskeleton organization, DNA repair, RNA processing and translation, as well as protein biogenesis and turnover, among others. Novel interactions between LAP1 and DNA repair proteins were additionally validated, strengthening the previously proposed involvement of LAP1 in the maintenance of genomic stability. Overall, this study reaffirms the biological relevance of LAP1 and the need to deepen our knowledge about this NE protein, providing new insights about its potential functional partners that will help guiding future research towards a mechanistic understanding of LAP1's functioning. Abstract

Cymoxanil (CYM) is a widely used synthetic acetamide fungicide, but its biochemical mode of action remains elusive. Since CYM inhibits cell growth, biomass production, and respiration in Saccharomyces cerevisiae, we used this model to characterize the effect of CYM on mitochondria. We found it inhibits oxygen consumption in both whole cells and isolated mitochondria, specifically inhibiting cytochrome c oxidase (CcO) activity during oxidative phosphorylation. Based on molecular docking, we propose that CYM blocks the interaction of cytochrome c with CcO, hampering electron transfer and inhibiting CcO catalytic activity. Although other targets cannot be excluded, our data offer valuable insights into the mode of action of CYM that will be instrumental in driving informed management of the use of this fungicide. Composting units which handle lignocellulosic residues are suitable sources of novel and promising lignocellulose-degrading enzymes such as cellulases, xylanases and amylases. These enzymes have practical application in many industries where lignocellulose is converted into several added-value bioproducts. 36. Questioning the cultural identity at the folkloric group “os pastores de São Romão”

Abstract

It was in 1978 that the Rancho Folclórico "Os Pastores de São Romão" began its activity, being an institution to collect, preserve and disseminate the material and immaterial heritage of the western slope of Serra da Estrela. It is not only a folk group, but an institution strongly linked to the local population, supporting cultural activities such as music, dance, and studies about the past of the region and its inhabitants, having, inclusively, the Ethnographic Museum situated in São Romão. This article intends to analyse, understand and characterise notions of tradition through the customs and various characteristic aspects of this folkloric group, such as the costumes, instruments, songs, and the performance itself. Through interviews and analysis of the cultural identity transmitted by the group, we have as objectives to question and identify the veracity of this set of customs, values, and practices, assessing the authenticity of the traditions.

Abstract

Food webs represent trophic interactions within ecosystems. Matching traits of consumers and resources helps infer trophic interactions and food-web properties. Environmental (e)DNA, commonly used for detecting species occurrences, is rarely used in trait-matching studies because abundance estimates and descriptions of relevant traits are generally missing. We synthesized recent literature on inferences of trophic interactions with eDNA and trait matching to identify challenges and opportunities for coupled eDNA–trait recording schemes. Our case study shows how coupling eDNA and trait data collection improves the ability to characterize greater numbers of food webs across multiple scales ranging from spatiotemporal to trait variation. Future-proofing eDNA data sets requires the collection of new traits or the compilation of existing trait data at spatiotemporal scales that are relevant to detect current and future changes in food webs and ecosystems.

Abstract

Sebastião Martins Mestre foi um militar e político algarvio que participou em dois acontecimentos decisivamente marcantes para a sociedade portuguesa do século XIX: as Invasões Francesas e as Guerras Liberais. Durante a juventude, Martins Mestre gloriou-se graças à intervenção nas invasões napoleónicas, mas quando decidiu enveredar pelo caminho da política, a sua imagem ficou para sempre ensombrada pela conduta. A presente dissertação tem como principal objetivo apresentar novos dados biográficos (relacionados com a família, atividade militar, atividade política, morte) que permitam compreender melhor a figura histórica de Sebastião Martins Mestre. O estudo inicia-se com o capítulo dedicado à família e funções militares, seguido do período referente às Invasões Francesas e, por último, será feita a análise à vida do biografado no âmbito das Guerras Liberais, onde centramos a nossa atenção no período em que foi governador de Vila Real de Santo António. Sebastião Martins Mestre was an algarvian military and politician who participated in two main events for Portuguese society in the 19th century: the French Invasions and the Liberal Wars. During his youth, Martins Mestre gloried due to his intervention in the Napoleonic invasions, but when he decided to take the path of politics, his image was forever overshadowed by his conduct. The main goal of this dissertation is to present new biographical data (family-related, military activity, political activity, death...) that will allow a better understading of the historical figure of Sebastião Martins Mestre. The study begins with a chapter dedicated to the family and military positions, followed by another alluding to the period of the French Invasions and a final chapter analyzes the life of the biographer in the context of the Liberal Wars, where we focus our attention on the period in which he was governor of Vila Real de Santo António.
