We combined the neuroanatomists' approach of defining a fascicle as all fibers passing through its compact stem with diffusion-weighted tractography to investigate the cortical terminations of two association tracts, the inferior fronto-occipital fasciculus (IFOF) and the uncinate fasciculus (UF), which have recently been implicated in the ventral language circuitry. The aim was to provide a detailed and quantitative description of their terminations in 60 healthy subjects and to do so to apply an anatomical stem-based virtual dissection, mimicking classical post-mortem dissection, to extract with minimal a priori the IFOF and UF from tractography datasets. In both tracts, we consistently observed more extensive termination territories than their conventional definitions, within the middle and superior frontal, superior parietal and angular gyri for the IFOF and the middle frontal gyrus and superior, middle and inferior temporal gyri beyond the temporal pole for the UF. We revealed new insights regarding the internal organization of these tracts by investigating for the first time the frequency, distribution and hemispheric asymmetry of their terminations. Interestingly, we observed a dissociation between the lateral right-lateralized and medial left-lateralized fronto-occipital branches of the IFOF. In the UF, we observed a rightward lateralization of the orbito-frontal and temporal branches. We revealed a more detailed map of the terminations of these fiber pathways that will enable greater specificity for correlating with diseased populations and other behavioral measures. The limitations of the diffusion tensor model in this study are also discussed. We conclude that anatomical stem-based virtual dissection with diffusion tractography is a fruitful method for studying the structural anatomy of the human white matter pathways.

Hemispheric lateralization for language production and its relationships with manual preference and manual preference strength were studied in a sample of 297 subjects, including 153 left-handers (LH). A hemispheric functional lateralization index (HFLI) for language was derived from fMRI acquired during a covert sentence generation task as compared with a covert word list recitation. The multimodal HFLI distribution was optimally modeled using a mixture of 3 and 4 Gaussian functions in right-handers (RH) and LH, respectively. Gaussian function parameters helped to define 3 types of language hemispheric lateralization, namely "Typical" (left hemisphere dominance with clear positive HFLI values, 88% of RH, 78% of LH), "Ambilateral" (no dominant hemisphere with HFLI values close to 0, 12% of RH, 15% of LH) and "Strongly-atypical" (right-hemisphere dominance with clear negative HFLI values, 7% of LH). Concordance between dominant hemispheres for hand and for language did not exceed chance level, and most of the association between handedness and language lateralization was explained by the fact that all Strongly-atypical individuals were left-handed. Similarly, most of the relationship between language lateralization and manual preference strength was explained by the fact that Strongly-atypical individuals exhibited a strong preference for their left hand. These results indicate that concordance of hemispheric dominance for hand and for language occurs barely above the chance level, except in a group of rare individuals (less than 1% in the general population) who exhibit strong right hemisphere dominance for both language and their preferred hand. They call for a revisit of models hypothesizing common determinants for handedness and for language dominance.

Jean Piaget's theory is a central reference point in the study of logico-mathematical development in children. One of the most famous Piagetian tasks is number conservation. Failures and successes in this task reveal two fundamental stages in children's thinking and judgment, shifting at approximately 7 years of age from visuospatial intuition to number conservation. In the current study, preschool children (nonconservers, 5-6 years of age) and school-age children (conservers, 9-10 years of age) were presented with Piaget's conservation-of-number task and monitored by functional magnetic resonance imaging (fMRI). The cognitive change allowing children to access conservation was shown to be related to the neural contribution of a bilateral parietofrontal network involved in numerical and executive functions. Background: A real-world visual scene consists of local elements (e.g. trees) that are arranged coherently into a global configuration (e.g. a forest). Children show psychological evolution from a preference for local visual information to an adult-like preference for global visual information, with the transition in visual preference occurring around 6 years of age. The brain regions involved in this shift in visual preference have not been described., Methods and Results: We used voxel-based morphometry (VBM) to study children during this developmental window to investigate changes in gray matter that underlie the shift from a bias for local to global visual information. Six-year-old children were assigned to groups according to their judgment on a global/local task. The first group included children who still presented with local visual processing biases, and the second group included children who showed global visual processing biases. VBM results indicated that compared to children with local visual processing biases, children with global visual processing biases had a loss of gray matter in the right occipital and parietal visuospatial areas., Conclusions: These anatomical findings are in agreement with previous findings in children with neurodevelopmental disorders and represent the first structural identification of brain regions that allow healthy children to develop a global perception of the visual world.

The impact of having left-handers (LHs) among one's close relatives, called familial sinistrality (FS), on neuroanatomical markers of left-hemisphere language specialization was studied in 274 normal adults, including 199 men and 75 women, among whom 77 men and 27 women were positive for FS. Measurements of the surface of a phonological cortical area, the "planum temporale" (PT), and gray and white matter hemispheric volumes and asymmetries were made using brain magnetic resonance images. The size of the left PT of subjects with left-handed close relatives (FS+) was reduced by 10%, decreasing with the number of left-handed relatives, and lowest when the subject's mother was left-handed. Such findings had no counterparts in the right hemisphere, and the subject's handedness and sex were found to have no significant effect or interaction with FS on the left PT size. The FS+ subjects also exhibited increased gray matter volume, reduced hemispheric gray matter leftward asymmetry, and, in LHs, reduced strength of hand preference. These results add to the increasing body of evidence suggesting multiple and somewhat independent mechanisms for the inheritance of hand and language lateralization.

Internal senses of the position of the eye in the orbit may influence the cognitive processes that take into account gaze and limb positioning for movement or guiding actions. Neuroimaging studies have revealed eye position-dependent activity in the extrastriate visual, parietal, and frontal areas, but, at the earliest vision stage, the role of the primary visual area (V1) in these processes remains unclear. Functional MRI (fMRI) was used to investigate the effect of eye position on V1 activity evoked by a quarter-field stimulation using a visual checkerboard. We showed that the amplitude of V1 activity was modulated by the position of the eye, the activity being maximal when both the eye and head positions were aligned. Previous studies gave impetus to the emerging view that V1 activity is a cortical area in which contextual influences take place. Using voxel-based morphometry, we measured the cerebral anatomical asymmetries in a sample of 56 young right-handed males and then compared voxelwise asymmetry indices of these subjects to those of 56 young left-handed males. In the right-handed, the clusters of grey matter asymmetry corresponding to the leftward occipital petalia and planum temporale asymmetries were retrieved. Strong rightward temporo-parietal asymmetries were also observed, but the rightward grey matter asymmetry in the frontal lobe was less massive than previously described. Group comparisons of left- and right-handed subjects' asymmetry maps, performed at a statistical threshold not corrected for multiple comparisons, revealed significant effects of handedness on this pattern of anatomical asymmetry in frontal regions, notably in the lower central and precentral sulci, and also in the planum temporale, with right-handed subjects being more leftward asymmetric. Concerning white matter, although almost no focal differences between left- and right-handed subjects were detected, volumetric analyses at the hemispheric level revealed a leftward asymmetry, which happened to be significantly less marked in the left-handed. This latter result, together with the pattern of leftward white matter asymmetries, suggested that anatomical correlates of the left hemispheric specialization for language would exist in white matter. In the population we studied, differences in anatomical asymmetry between left- and right-handed subjects provided structural arguments for a greater functional ambilaterality in left-handed subjects.

The morphology of the central sulcus (CS), at the level of the hand primary motor cortex, has been shown to be related to hand preference and skill. Differences in the cerebral functional organisation of left and right-handers have been described, notably with respect to hemispheric specialisation, which might cause the neural substrate of hand dominance or skill to differ between the two groups. Here, we further explored the relationship between the anatomical variability of the central sulcus and hand skill in two groups of young male subjects differing by handedness (n = 56 right-handers and n = 55 left-handers). Grey matter volume (GMV) in the upper region of the central sulcus was estimated with Voxel Based Morphometry, using a probabilistic region of interest approach, while hand motor skill was measured with the finger tapping test. No significant anatomical differences could be evidenced between the two hand preference groups, a rightward hemispheric asymmetry being observed in both samples. However, multiple regression analyses showed that, in the right-handed group, the maximum tapping rate of the right hand correlated positively with the left central sulcus GMV, but negatively with the right. Similar analyses showed that, in left-handers, the maximum tapping rate of the non-dominant right hand was strongly correlated with the GMV of the ipsilateral CS but not significantly with that of the contralateral CS. These results may be due to differences in the organisation of motor systems between these two groups, possibly concerning a left hemispheric specialisation for fast repetitive movements in right-handers, which would be different in left-handers.

A new method of instantaneous EEG analysis based on amplitude modulation (AM-EEG) was applied to analyze the AM-EEG changes in the alpha frequency band (8.20-12.89 Hz) for successive 5 ms epochs. Repeated auditory tone-burst stimuli (of 220 ms duration) were presented at fixed 2.56 second intervals to 12 attending right-handed young female volunteers, who were EEG-recorded over 19 EEG channels at 200 Hz sampling frequency. The time-course of functional activation was characterized in terms of percent decrease in instantaneous amplitude modulation, as compared to baseline, in analogy with the classical event-related desynchronization paradigm. A dynamic sequence of the successive 122 AM-EEG maps obtained for the 610 ms from beginning of tone-burst was stored and later animated on PC microcomputer. Topological changes among successive maps were extracted into 40 specific AM-EEG maps. Early left temporal and centro-temporal activation was observed, followed first by strong bilateral frontal, and then by left temporal activation. These changes induced by a warning tone-burst are discussed in terms of functional neurophysiology. The present method therefore allows an improved time-resolution for functional brain activation paradigms.
