Changes in energy and environmental policies along with changes in the energy markets of New York State over the past two decades, have spurred interest in evaluating their impacts on emissions from various energy generation sectors. This study focused on quantifying these effects on VOC (volatile organic compounds) emissions and their subsequent impacts on air quality within the New York City (NYC) metropolitan area. NYC is an EPA nonattainment region for ozone (O 3 ) and likely is a VOC limited region. NYC has a complex coastal topography and meteorology with low-level jets and sea/bay/land breeze circulation associated with heat waves, leading to summertime O 3 exceedances and formation of secondary organic aerosol (SOA). To date, no comprehensive source apportionment studies have been done to understand the contributions of local and long-range sources of VOCs in this area. This study applied an improved Positive Matrix Factorization (PMF) methodology designed to incorporate atmospheric dispersion and photochemical reaction losses of VOCs to provide improved apportionment results. Hourly measurements of VOCs were obtained from a Photochemical Assessment Monitoring Station located at an urban site in the Bronx from 2000 to 2021. The study further explores the role of VOC sources in O 3 and SOA formation and leverages advanced machine learning tools, XGBoost and SHAP algorithms, to identify synergistic interactions between sources and provided VOC source impacts on ambient O 3 concentrations. Isoprene demonstrated a substantial influence in the source contribution of the biogenic factor, emphasizing its role in O 3 formation. Notable contributions from anthropogenic emissions, such as fuel evaporation and various industrial processes, along with significant traffic-related sources that likely contribute to SOA formation, underscore the combined impact of natural and human-made sources on O 3 pollution. The fate of volatile organic compounds (VOC) vapors in the unsaturated zone is the basis for evaluating the natural attenuation potential and vapor intrusion risk. Microcosm and column experiments were conducted to study the effects chemical speciation and soil types/properties on the fate of petroleum VOCs in unsaturated zone. The biodegradation and total attenuation rates of the seven VOCs obtained by microcosm experiments in black soil and yellow earth were also generally higher than those in floodplain soil, lateritic red earth, and quartz sand. The VOC vapors in floodplain soil, lateritic red earth, and quartz sand showed slow total attenuation rates (<0.3 d -1 ). N-pentane, methylcyclopentane, and methylcyclohexane showed lower biodegradation rates than octane and three monoaromatic hydrocarbons. Volatilization into the atmosphere and biodegradation are two important natural attenuation paths for VOCs in unsaturated soil columns. The volatilization loss fractions of different volatile hydrocarbons in all five unsaturated soils were generally in the order: n-pentane (93.5%-97.8%) > methylcyclopentane (77.2%-85.5%) > methylcyclohexane (53.5%-69.2%) > benzene (17.1%-73.3%) > toluene (0-45.7%) > octane (1.9%-34.2%) > m-xylene (0-5.7%). The fractions by volatilization into the atmosphere of all seven hydrocarbons in quartz sand, lateritic red earth, and floodplain soil were close and higher compared to the yellow earth and black soil. The leaves of many trees emit volatile organic compounds (abbreviated as BVOCs), which protect them from various damages, such as herbivory, pathogens, and heat stress. For example, isoprene is highly volatile and is known to enhance the resistance to heat stress. In this study, we analyze the optimal seasonal schedule for producing isoprene in leaves to mitigate damage. We assume that photosynthetic rate, heat stress, and the stress-suppressing effect of isoprene may vary throughout the season. We seek the seasonal schedule of isoprene production that maximizes the total net photosynthesis using Pontryagin's maximum principle. The isoprene production rate is determined by the changing balance between the cost and benefit of enhanced leaf protection over time. If heat stress peaks in midsummer, isoprene production can reach its highest levels during the summer. However, if a large portion of leaves is lost due to heat stress in a short period, the optimal schedule involves peaking isoprene production after the peak of heat stress. Both high photosynthetic rate and high isoprene volatility in midsummer make the peak of isoprene production in spring. In the breath research community's search for volatile organic compounds that can act as non-invasive biomarkers for various diseases, hundreds of endogenous volatiles have been discovered. Whilst these systemic chemicals result from normal and abnormal metabolic activities or pathological disorders, to date very few are of any use for the development of clinical breath tests that could be used for disease diagnosis or to monitor therapeutic treatments. The reasons for this lack of application are manifold and complex, and these complications either limit or ultimately inhibit the analytical application of endogenous volatiles for use in the medical sciences. One such complication is a lack of knowledge on the biological origins of the endogenous volatiles. A major exception to this is isoprene. Since 1984, i.e. for 40 years, it has been generally accepted that the pathway to the production of human isoprene, and hence the origin of isoprene in exhaled breath, is through cholesterol biosynthesis via the mevalonate (MVA) pathway within the liver. However, various studies between 2001 and 2012 provide compelling evidence that human isoprene is produced in skeletal muscle tissue. A recent multi-omic investigation of genes and metabolites has revealed that this proposal is correct by showing that human isoprene predominantly results from muscular lipolytic cholesterol metabolism. Despite the overwhelming proof for a muscular pathway to isoprene production in the human body, breath research papers still reference the hepatic MVA pathway. The major aim of this perspective is to review the evidence that leads to a correct interpretation for the origins of human isoprene, so that the major pathway to human isoprene production is understood and appropriately disseminated. This is important, because an accurate attribution to the endogenous origins of isoprene is needed if exhaled isoprene levels are to be correctly interpreted and for assessing isoprene as a clinical biomarker., (Creative Commons Attribution license.)

Exhaled volatile organic compounds (VOCs) are of interest due to their minimally invasive sampling procedure. Previous studies have investigated the impact of exercise, with evidence suggesting that breath VOCs reflect exercise-induced metabolic activity. However, these studies have yet to investigate the impact of maximal exercise to exhaustion on breath VOCs, which was the main aim of this study. Two-litre breath samples were collected onto thermal desorption tubes using a portable breath collection unit. Samples were collected pre-exercise, and at 10 and 60 min following a maximal exercise test (VO 2MAX ). Breath VOCs were analysed by thermal desorption-gas chromatography-mass spectrometry using a non-targeted approach. Data showed a tendency for reduced isoprene in samples at 10 min post-exercise, with a return to baseline by 60 min. However, inter-individual variation meant differences between baseline and 10 min could not be confirmed, although the 10 and 60 min timepoints were different ( p = 0.041). In addition, baseline samples showed a tendency for both acetone and isoprene to be reduced in those with higher absolute VO 2MAX scores (mL(O 2 )/min), although with restricted statistical power. Baseline samples could not differentiate between relative VO 2MAX scores (mL(O 2 )/kg/min). In conclusion, these data support that isoprene levels are dynamic in response to exercise.

A baseline correction method is developed for comprehensive two-dimensional (2D) chromatography (GC × GC) with flame-ionization detection (FID) using dynamic pressure gradient modulation (DPGM). The DPGM-GC × GC-FID utilized porous layer open tubular (PLOT) columns in both dimensions to focus on light hydrocarbon separations. Since DPGM is nominally a stop-flow modulation technique, a rhythmic baseline disturbance is observed in the FID signal that cycles with the modulation period (P M ). This baseline disturbance needs to be corrected to optimize trace analysis. The baseline correction method has three steps: collection of a background "blank" chromatogram and multiplying it by an optimized normalization factor, subtraction of the normalization-optimized background chromatogram from a sample chromatogram, and application of Savitzky-Golay smoothing. An alkane standard solution, containing pentane, hexane and heptane was used for method development, producing linear calibration curves (r 2  > 0.991) over a broad concentration range (7.8 ppm - 4000 ppm). Further, the limit-of-detection (LOD) and limit-of-quantification (LOQ) were determined for pentane (LOD = 2.5 ppm, LOQ = 8.2 ppm), hexane (LOD = 0.9 ppm, LOQ = 3.0 ppm), and heptane (LOD = 1.9 ppm, LOQ = 6.4 ppm). A natural gas sample separation illustrated method applicability, whereby the DPGM produced a signal enhancement (SE) of 30 for isopentane, where SE is defined as the height of the tallest 2 D peak in the modulated chromatogram for the analyte divided by the height of the unmodulated 1 D peak. The 30-fold SE resulted in about a 10-fold improvement in the signal-to-noise ratio (S/N) for isopentane. The aim of this study was to analyze the exhaled volatile organic compounds (VOCs) profile, airway microbiome, lung function and exercise performance in congenital diaphragmatic hernia (CDH) patients compared to healthy age and sex-matched controls. A total of nine patients (median age 9 years, range 6-13 years) treated for CDH were included. Exhaled VOCs were measured by GC-MS. Airway microbiome was determined from deep induced sputum by 16S rRNA gene sequencing. Patients underwent conventional spirometry and exhausting bicycle spiroergometry. The exhaled VOC profile showed significantly higher levels of cyclohexane and significantly lower levels of acetone and 2-methylbutane in CDH patients. Microbiome analysis revealed no significant differences for alpha-diversity, beta-diversity and LefSe analysis. CDH patients had significantly lower relative abundances of Pasteurellales and Pasteurellaceae . CDH patients exhibited a significantly reduced Tiffeneau Index. Spiroergometry showed no significant differences. This is the first study to report the VOCs profile and airway microbiome in patients with CDH. Elevations of cyclohexane observed in the CDH group have also been reported in cases of lung cancer and pneumonia. CDH patients had no signs of impaired physical performance capacity, fueling controversial reports in the literature.

A novel non-silicon-based micro-preconcentration device, as a pretreatment component in a portable gas chromatography system, was developed for the preconcentration one of the trace volatile organic compounds (VOCs) in the exhaled gases, which is one typical biomarker for the chronic liver disease (CLD). The device was designed as an array of manifold-shaped rectangular metal micro-channels with flat dimensions of 16 mm × 12.6 mm and the internal empty volume is 14.4 μL on the copper substrate. Instead of the non-silicon fabrication process, the traditional laser etching technology (LET) was optimized to etch micro-channels, and vacuum diffusion welding (VDW) was applied to form internal channels. The fabricated chip was filled with Carbopack X adsorbent. In the testing, the metal gas preconcentrator (MGP) was installed in a commercial GC (gas chromatography) and nitrogen was used as carrier gas and desorbed gas. With the MPG, up to 352 of concentration factor can be achieved for 10 ppb isoprene. The developed MGP, which has advantages of high strength, low cost, good thermal conductivities, can potentially be used for non-invasive screening of advanced liver fibrosis by monitoring isoprene concentrations in exhaled breath., (Copyright © 2018 Elsevier B.V. Continuous observation of isoprene, α-pinene and β-pinene was carried out in a typical urban area of Beijing from March 2014 to February 2015, using an AirmoVOC online analyzer. Based on the analysis of the ambient level and variation characteristics of isoprene, α-pinene and β-pinene, the chemical reactivity was studied, and their sources were identified. Results showed that the concentrations of isoprene, α-pinene and β-pinene in the urban area of Beijing were lower than those in richly vegetated areas; the concentrations of isoprene were at a moderate level compared with those of previous studies of Beijing. Concentrations of isoprene, α-pinene and β-pinene showed different seasonal, monthly, daily and diurnal variations, and all of the three species showed higher level at night than those in the daytime as a whole, the variations of isoprene, α-pinene and β-pinene mainly influenced by emission of sources, photochemical reaction, and meteorological parameters. Isoprene was the largest contributor to the total OFP values than α-pinene and β-pinene. α-Pinene was the largest contributor to the total SOAFP values than isoprene and β-pinene in autumn, while isoprene was the largest one in other seasons. Isoprene, α-pinene and β-pinene were derived mainly from biological sources; and α-pinene level were also affected by industrial sources. To reduce the concentrations of isoprene, α-pinene and β-pinene, it is necessary to scientifically select urban green plant species, and more strict control measures should be taken to reduce the emission of α-pinene from industrial sources, such as artificial flavors and resins synthesis processes., (Copyright © 2018. Volatile organic compounds (VOCs) play a key role in air pollution of China. Among various sources, petrochemical industry is one of the important contributors, but its VOC emission estimate still exists a big uncertainty. Therefore, this study developed an inverse-dispersion calculation method (IDM), and applied it in a typical petrochemical plant that manufactures ethylene (170 kt/year), polyethylene (100 kt/year), and polypropylene (61 kt/year), and determined the VOC emission amount for this complex industrial area source. Firstly, this study monitored VOC concentrations around this plant in April of 2017, and found that the VOCs at downwind receptors was obviously higher than the level at background, higher by 20.7 ppb on average. This VOC increment was mainly contributed by ethylene (30.4%), propylene (17.8%), pentanes (16.4%), and butanes (13.4%), which is consistent with the knowledge of VOC components emitted from the manufacturing of ethylene, polyethylene, and polypropylene. Then, by using the inverse-dispersion calculation method (IDM), we determined the relationship coefficient γ between source emission rate and ambient concentration for each receptor of each test based on an assumed source emission rate, combined γ with the actual VOC concentrations measured in monitoring tests, and estimated the average VOCs emission of 666.0 tons/year for this plant, including 18.1 tons for ethane, 21.1 tons for propane, 61.6 tons for isobutane, 44.3 tons for n-butane, 79.3 tons for isopentane, 56.8 tons for n-pentane, 115.4 tons for ethylene, 102.5 tons for propylene, 92.7 tons for benzene, and 74.1 tons for toluene. Our IDM estimate was in the same order with the traditional emission factor method estimate (916.4 tons VOCs per year for this plant), and we believed the IDM can be applied to effectively estimate the VOCs emissions for those complicated industrial sources.

In this study, the measurement of volatile organic compounds (VOCs) was conducted at Beijing Capital International Airport (ZBAA) and a background reference site in four seasons of 2015. Total concentrations of VOCs were 72.6 ± 9.7, 65.5 ± 8.7, 95.8 ± 11.0, and 79.2 ± 10.8 μg/m 3 in winter, spring, summer, and autumn, respectively. The most abundant specie was toluene (10.1%-17.4%), followed by benzene, ethane, isopentane, ethane, acetylene, and n-butane. Seasonal variations of VOCs were analyzed, and it was found that the highest concentration occurring in summer, while the lowest in spring. For the diurnal variation, the concentration of VOCs in the daytime (9:00-15:00) was less than that at night (15:00-21:00) obviously. Ozone Formation Potential (OFP) was calculated by using Maximum Incremental Reactivity (MIR) method. The greatest contribution to OFP from alkenes and aromatics, which accounted for 27.3%-51.2% and 36.6%-58.6% of the total OFP. The WRF-CMAQ model was used to simulate the impact of airport emissions on the surrounding area. The results indicated that the maximum impact of VOCs emissions and all sources emissions at the airport on O 3 was 0.035 and -23.8 μg/m 3 , respectively. Based on the RCP2.6, RCP4.5, RCP6.0 and RCP8.5 climate change scenarios produced by the global climate model NorESM1-M and plant isoprene emissions model, the effects of climate change on the isoprene emission rate from leaves of Pleioblastus amarus in Yixing City of Jiangsu Province, Longmen County of Guangdong Province, Yulong Naxi Autonomous County of Yunnan Province and Wanyuan City of Sichuan Province were simulated. The differences of isoprene emission rate from leaves of P. amarus distributed in four regions were compared under future climate change scenarios. The results showed that mean annual air temperature would increase, annual precipitation and radiation intensity would greatly fluctuate, with the coexistence of increasing and decreasing trends in the four regions. In the baseline scenario, daily mean emission rate of isoprene from leaves of P. amarus was 71-470 μg·g -1 ·d -1 , and annual mean value was 25954-171231 μg·g -1 ·a -1 . The daily and annual emission rates in the four regions decreased with the order of Longmen, Yixing, Wanyuan and Yulong. Compared with the baseline scenario, daily mean emission rate of isoprene from leaves of P. amarus was about 4-45 μg·g -1 ·d -1 higher in future climate change scenario, and which was about 23, 29, 4, and 14 μg·g -1 ·d -1 higher than that in baseline in Yixing, Longmen, Yulong and Wanyuan, respectively. In addition, the emission rate of isoprene from leaves of P. amarus was more than 5% higher in the future climate change scenario than that in the baseline scenario, which was higher in Wanyuan and Yixing (>13%) than and lower in Longmen and Yulong (>5%). All the four regions reached the highest rate under RCP8.5 scenario (increased by about 11%-18%). Compared with the baseline scenario, annual emission rate of isoprene in the future climate change scenario was about 1500-17000 μg·g -1 ·a -1 , and which was about 8560-13208 μg·g -1 ·a -1 higher in Yixing, 10862-16131 μg·g -1 ·a -1 higher in Longmen, 1574-3028 μg·g -1 ·a -1 higher in Yulong, 5288-8532 μg·g -1 ·a -1 higher in Wanyuan. In addition, the increasing rate of annual isoprene emission rates was 6%-14%. The rates in Yixing (8%-12%) and Wanyuan (8%-14%) were higher than that in the other two regions, the rate in Yulong (6%-12% increase) was the lowest, with all four regions increasing substantially (9%-14%) under RCP8.5 scenario. The results suggested that climate change would have different effects on the rate of isoprene emissions from leaves of P. amarus distributed in diffe-rent regions.

Isoprene (2-methyl-1,3-butadiene), the most abundantly produced biogenic volatile organic compound (BVOC) on Earth, is highly reactive and can have diverse and often detrimental atmospheric effects, which impact on climate and health. Most isoprene is produced by terrestrial plants, but (micro)algal production is important in aquatic environments, and the relative bacterial contribution remains unknown. Soils are a sink for isoprene, and bacteria that can use isoprene as a carbon and energy source have been cultivated and also identified using cultivation-independent methods from soils, leaves and coastal/marine environments. Bacteria belonging to the Actinobacteria are most frequently isolated and identified, and Proteobacteria have also been shown to degrade isoprene. In the freshwater-sediment isolate, Rhodococcus strain AD45, initial oxidation of isoprene to 1,2-epoxy-isoprene is catalyzed by a multicomponent isoprene monooxygenase encoded by the genes isoABCDEF. The resultant epoxide is converted to a glutathione conjugate by a glutathione S-transferase encoded by isoI, and further degraded by enzymes encoded by isoGHJ. Genome sequence analysis of actinobacterial isolates belonging to the genera Rhodococcus, Mycobacterium and Gordonia has revealed that isoABCDEF and isoGHIJ are linked in an operon, either on a plasmid or the chromosome. In Rhodococcus strain AD45 both isoprene and epoxy-isoprene induce a high level of transcription of 22 contiguous genes, including isoABCDEF and isoGHIJ. Sequence analysis of the isoA gene, encoding the large subunit of the oxygenase component of isoprene monooxygenase, from isolates has facilitated the development of PCR primers that are proving valuable in investigating the ecology of uncultivated isoprene-degrading bacteria.

The potential role of isoprene oxidative processes, as well as the possible impact of air pollution on isoprene emissions, are more important in tropical cities, surrounded by rainforests. In this study, the contribution of isoprene to ozone formation was determined considering different scenarios, mainly volatile organic compounds/NO x (VOC/NO x ) ratios, and typical atmospheric conditions for the city of Rio de Janeiro, where more than 36% of the urbanized area is covered by vegetation. Ozone isopleths and incremental reactivity coefficients (IR) were evaluated to understand the direct contribution of isoprene to ground-level ozone formation and the negative impact of anthropogenic NO x emissions on the natural atmospheric balance. Although isoprene accounted for only 2.7% of the total VOC mass, excluding the isoprene concentration from the model reduced the maximum ozone value by 14.1%. The calculated IR coefficient (grams of O 3 formed per gram of added isoprene) was 2.2 for a VOC/NO x ratio of 8.86.

Hypoxia-like incidents in-flight have increased over the past decade causing severe safety concerns across the aviation community. As a result, the need to monitor flight crews in real-time for the onset of hypoxic conditions is paramount for continued aeronautical safety. Here, hypoxic events were simulated in the laboratory via a reduced oxygen-breathing device to determine the effect of recovery gas oxygen concentration (21% and 100%) on exhaled breath volatile organic compound composition. Data from samples collected both serially (throughout the exposure), prior to, and following exposures yielded 326 statistically significant features, 203 of which were unique. Of those, 72 features were tentatively identified while 51 were verified with authentic standards. A comparison of samples collected serially between recovery and hypoxia time points shows a statistically significant reduction in exhaled breath isoprene (2-methyl-1,3-butadiene, log 2 FC -0.399, p = 0.005, FDR = 0.034, q = 0.033), however no significant difference in isoprene abundance was observed when comparing recovery gases (21% or 100% O 2 , p = 0.152). Furthermore, examination of pre-/post-exposure 1 l bag breath samples illustrate an overall increase in exhaled isoprene abundance post-exposure (log 2 FC 0.393, p = 0.005, FDR = 0.094, q = 0.033) but again no significant difference between recovery gas (21% and 100%, p = 0.798) was observed. A statistically significant difference in trend was observed between isoprene abundance and recovery gases O 2 concentration when plotted against minimum oxygen saturation (p = 0.0419 100% O 2 , p = 0.7034 21% O 2 ). Collectively, these results suggest exhaled isoprene is dynamic in the laboratory ROBD setup and additional experimentation will be required to fully understand the dynamics of isoprene in response to acute hypoxic stress.

A study has been carried out on the volatile organic compounds (VOCs) in the exhaled breath of patients suffering from inflammatory bowel disease (IBD), comprising 136 with Crohn's disease (CD) and 51 with ulcerative colitis (UC), together with a cohort of 14 healthy persons as controls. Breath samples were collected by requesting the patients to inflate Nalophan bags, which were then quantitatively analysed using selected ion flow tube mass spectrometry (SIFT-MS). Initially, the focus was on n-pentane that had previously been quantified in single exhalations on-line to SIFT-MS for smaller cohorts of IBD patients. It was seen that the median concentration of pentane was elevated in the bag breath samples of the IBD patients compared to those of the healthy controls, in accordance with the previous study. However, the absolute median pentane concentrations in the bag samples were about a factor of two lower than those in the directly analysed single exhalations-a good illustration of the dilution of VOCs in the samples of breath collected into bags. Accounting for this dilution effect, the concentrations of the common breath VOCs, ethanol, propanol, acetone and isoprene, were largely as expected for healthy controls. The concentrations of the much less frequently measured hydrogen sulphide, acetic acid, propanoic acid and butanoic acid were seen to be more widely spread in the exhaled breath of the IBD patients compared to those for the healthy controls. The relative concentrations of pentane and these other VOCs weakly correlate with simple clinical activity indices. It is speculated that, potentially, hydrogen sulphide and these carboxylic acids could be exhaled breath biomarkers of intestinal bacterial overgrowth, which could assist therapeutic intervention and thus alleviate the symptoms of IBD.

The paper presents a portable breath sampling unit (BSU) for human breath analyzers. The developed unit can be used to probe air from the upper airway and alveolar for clinical and science studies. The BSU is able to operate as a patient interface device for most types of breath analyzers. Its main task is to separate and to collect the selected phases of the exhaled air. To monitor the so-called I, II, or III phase and to identify the airflow from the upper and lower parts of the human respiratory system, the unit performs measurements of the exhaled CO 2 (ECO 2 ) in the concentration range of 0%-20% (0-150 mm Hg). It can work in both on-line and off-line modes according to American Thoracic Society/European Respiratory Society standards. A Tedlar bag with a volume of 5 dm 3 is mounted as a BSU sample container. This volume allows us to collect ca. 1-25 selected breath phases. At the user panel, each step of the unit operation is visualized by LED indicators. This helps us to regulate the natural breathing cycle of the patient. There is also an operator's panel to ensure monitoring and configuration setup of the unit parameters. The operation of the breath sampling unit was preliminarily verified using the gas chromatography/mass spectrometry (GC/MS) laboratory setup. At this setup, volatile organic compounds were extracted by solid phase microextraction. The tests were performed by the comparison of GC/MS signals from both exhaled nitric oxide and isoprene analyses for three breath phases. The functionality of the unit was proven because there was an observed increase in the signal level in the case of the III phase (approximately 40%). The described work made it possible to construct a prototype of a very efficient breath sampling unit dedicated to breath sample analyzers.

Concentration- and flux-based O 3 dose-responses of isoprene emission from single leaves and whole plants were developed. Two poplar clones differing in O 3 sensitivity were exposed to five O 3 levels in open-top chambers for 97 d: charcoal-filtered ambient air (CF), non-filtered ambient air (NF) and NF plus 20 ppb (NF + 20), 40 ppb (NF + 40) and 60 ppb (NF + 60). At both leaf and plant level, isoprene emission was significantly decreased by NF + 40 and NF + 60 for both clones. Although intra-specific variability was found when the emissions were up-scaled to the whole plant, both leaf- and plant-level emissions decreased linearly with increasing concentration-based (AOT40, cumulative exposure to hourly O 3 concentrations >40 ppb) and flux-based indices (POD Y , cumulative stomatal uptake of O 3  > Y nmol O 3 m -2 PLA s -1 ). AOT40- and POD 7 -based dose-responses performed equally well. The two clones responded differently to AOT40 and similarly to POD Y (with a slightly higher R 2 for POD 7 ) when the emission was expressed as change relative to clean air. Chromatographic studies on breath composition are aimed at finding volatile markers useful for medical diagnostics or in screening investigations. Studies leading to the development of screening breath tests are especially important for the diagnostics of chronic kidney disease (CKD) and type 2 diabetes mellitus (T2DM). The aim of the presented study was to confirm diagnostic usefulness of chosen volatile compounds detected in breath, which are suggested as potential biomarkers of renal dysfunction and diabetes. Breath analysis were carried out in three groups: 10 healthy volunteers, 10 patients with CKD and 10 patients with CKD and T2DM. All exhaled air samples were analyzed using gas chromatograph (Agilent 6890GC) coupled with mass spectrometer (5975MSD). Thermal desorption was applied as the enrichment method. TMA was detected only in CKD patients. Higher breath concentrations of methanethiol (MeSH) were observed in CKD patients with coexisting diabetes than in patients with renal dysfunction only or in the healthy group. There was a tendency of increasing MeSH concentration in breath with increasing total glutathione in plasma (r=0.53, p=0.0026). Also, a trend of increasing dimethylsulfide (DMS) levels detected in breath was noticed with an increase of hydrogen sulfide concentration in plasma (r=0.74; p=0.00001) as well as with aspartate aminotransferase (AST), (r=0.61; p=0.001). The sources of submicrometer particulate matter (PM 1 ) remain poorly characterized in the industrialized city of Houston, TX. A mobile sampling approach was used to characterize PM 1 composition and concentration across Houston based on high-time-resolution measurements of nonrefractory PM 1 and trace gases during the DISCOVER-AQ Texas 2013 campaign. Two pollution zones with marked differences in PM 1 levels, character, and dynamics were established based on cluster analysis of organic aerosol mass loadings sampled at 16 sites. The highest PM 1 mass concentrations (average 11.6 ± 5.7 µg/m 3 ) were observed to the northwest of Houston (zone 1), dominated by secondary organic aerosol (SOA) mass likely driven by nighttime biogenic organonitrate formation. Zone 2, an industrial/urban area south/east of Houston, exhibited lower concentrations of PM 1 (average 4.4 ± 3.3 µg/m 3 ), significant organic aerosol (OA) aging, and evidence of primary sulfate emissions. Diurnal patterns and backward-trajectory analyses enable the classification of airmass clusters characterized by distinct PM sources: biogenic SOA, photochemical aged SOA, and primary sulfate emissions from the Houston Ship Channel. Principal component analysis (PCA) indicates that secondary biogenic organonitrates primarily related with monoterpenes are predominant in zone 1 (accounting for 34% of the variability in the data set). The relevance of photochemical processes and industrial and traffic emission sources in zone 2 also is highlighted by PCA, which identifies three factors related with these processes/sources (~50% of the aerosol/trace gas concentration variability). PCA reveals a relatively minor contribution of isoprene to SOA formation in zone 1 and the absence of isoprene-derived aerosol in zone 2. The relevance of industrial amine emissions and the likely contribution of chloride-displaced sea salt aerosol to the observed variability in pollution levels in zone 2 also are captured by PCA., Implications: This article describes an urban-scale mobile study to characterize spatial variations in submicrometer particulate matter (PM 1 ) in greater Houston. The data set indicates substantial spatial variations in PM 1 sources/chemistry and elucidates the importance of photochemistry and nighttime oxidant chemistry in producing secondary PM 1 . These results emphasize the potential benefits of effective control strategies throughout the region, not only to reduce primary emissions of PM 1 from automobiles and industry but also to reduce the emissions of important secondary PM 1 precursors, including sulfur oxides, nitrogen oxides, ammonia, and volatile organic compounds. Such efforts also could aid in efforts to reduce mixing ratios of ozone.

Spatial distribution and sea-to-air flux of isoprene in the East China Sea and the South Yellow Sea in July 2013 were investigated. This study is the first to report the concentrations of isoprene in the China marginal seas. Isoprene concentrations in the surface seawater during summer ranged from 32.46 to 173.5 pM, with an average of 83.62 ± 29.22 pM. Distribution of isoprene in the study area was influenced by the diluted water from the Yangtze River, which stimulated higher in-situ phytoplankton production of isoprene rather than direct freshwater input. Variations in isoprene concentrations were found to be diurnal, with high values observed during daytime. A significant correlation was observed between isoprene and chlorophyll a in the study area. Relatively higher isoprene concentrations were recorded at stations where the phytoplankton biomass was dominated by Chaetoceros, Skeletonema, Pennate-nitzschia, and Thalassiosira. Positive correlation was observed between isoprene and methyl iodide. In addition, sea-to-air fluxes of isoprene approximately ranged from 22.17 nmol m -2  d -1 -537.2 nmol m -2  d -1 , with an average of 161.5 ± 133.3 nmol m -2  d -1 . Objective: N-butane and n-pentane can both produce general anesthesia. Both compounds potentiate γ-aminobutyric acid type A (GABA A ) receptor function, but only butane inhibits N-methyl-d-aspartate (NMDA) receptors. It was hypothesized that butane and pentane would exhibit anesthetic synergy due to their different actions on ligand-gated ion channels., Study Design: Prospective experimental study., Animals: A total of four Xenopus laevis frogs and 43 Sprague-Dawley rats., Methods: Alkane concentrations for all studies were determined via gas chromatography. Using a Xenopus oocyte expression model, standard two-electrode voltage clamp techniques were used to measure NMDA and GABA A receptor responses in vitro as a function of butane and pentane concentrations relevant to anesthesia. The minimum alveolar concentrations (MAC) of butane and pentane were measured separately in rats, and then pentane MAC was measured during coadministration of 0.25, 0.50 or 0.75 times MAC of butane. An isobole with 95% confidence intervals was constructed using regression analysis. A sum of butane and pentane that was statistically less than the lower-end confidence bound isobole indicated a synergistic interaction., Results: Both butane and pentane dose-dependently potentiated GABA A receptor currents over the study concentration range. Butane dose-dependently inhibited NMDA receptor currents, but pentane did not modulate NMDA receptors. Butane and pentane MAC in rats was 39.4±0.7 and 13.7±0.4 %, respectively. Purpose Human breath analysis is proposed with increasing frequency as a useful tool in clinical application. We performed this study to find the characteristic volatile organic compounds (VOCs) in the exhaled breath of patients with idiopathic pulmonary fibrosis (IPF) for discrimination from healthy subjects. Methods VOCs in the exhaled breath of 40 IPF patients and 55 healthy controls were measured using a multi-capillary column and ion mobility spectrometer. The patients were examined by pulmonary function tests, blood gas analysis, and serum biomarkers of interstitial pneumonia. Results We detected 85 VOC peaks in the exhaled breath of IPF patients and controls. IPF patients showed 5 significant VOC peaks; p-cymene, acetoin, isoprene, ethylbenzene, and an unknown compound. The VOC peak of p-cymene was significantly lower (p < 0.001), while the VOC peaks of acetoin, isoprene, ethylbenzene, and the unknown compound were significantly higher (p < 0.001 for all) compared with the peaks of controls. Comparing VOC peaks with clinical parameters, negative correlations with VC (r =-0.393, p = 0.013), %VC (r =-0.569, p < 0.001), FVC (r = -0.440, p = 0.004), %FVC (r =-0.539, p < 0.001), DLco (r =-0.394, p = 0.018), and %DLco (r =-0.413, p = 0.008) and a positive correlation with KL-6 (r = 0.432, p = 0.005) were found for p-cymene. Conclusion We found characteristic 5 VOCs in the exhaled breath of IPF patients. Among them, the VOC peaks of p-cymene were related to the clinical parameters of IPF. These VOCs may be useful biomarkers of IPF.

Performance losses during scale-up are described since decades, but are still one of the major obstacles for industrial bioprocess development. Consequently, robustness to inhomogeneous cultivation environments is an important quality of industrial production organisms. Especially, Corynebacterium glutamicum was proven to have an outstanding resistance against rapid changes of oxygen and substrate availability as occurring in industrial scale bioreactors. This study focuses on the identification of metabolic key mechanisms for this robustness to get a deeper insight and provide future targets for process orientated strain development. A 1,5-diaminopentane producing C. glutamicum strain was cultivated in a two compartment scale-down device to create short-term environmental changes simulating industrial scale cultivation conditions. Using multi omics based methods, it is shown, that central metabolism is flexibly rearranged under short-term oxygen depletion and carbon source excess to overcome shortage in NAD + recycling. In order to balance the redox state, key enzymes for the non-oxygen dependent fermentative NAD + regeneration were significantly up-regulated while parts of non-essential pathways were down-regulated. The transfer of the cells back into the well aerated zones with low substrate concentration triggers an additional upregulation of genes for the re-assimilation of previously formed side products, showing L-lactate forming and utilizing reactions being active at the same time. Especially L-lactate as reversible and flexible external buffer for carbon and redox equivalents puts C. glutamicum in a robust position to deal with inhomogeneity in large scale processes. Methane emissions in oil sands tailings ponds are sustained by anaerobic biodegradation of unrecovered hydrocarbons. Naphtha (primarily C 6 -C 10 ; n- iso- and cycloalkanes) is commonly used as a solvent during bitumen extraction process and its residue escapes to tailings ponds during tailings deposition. To investigate biodegradability of hydrocarbons in naphtha, mature fine tailings (MFT) collected from Albian and CNRL tailings ponds were amended with CNRL naphtha at ∼0.2 wt% (∼2000 mg L -1 ) and incubated under methanogenic conditions for ∼1600 d. Microbial communities in both MFTs started metabolizing naphtha after a lag phase of ∼100 d. Complete biodegradation/biotransformation of all n-alkanes (except partial biodegradation of n-octane in CNRL MFT) followed by major iso-alkanes (2-methylpentane, 3-methylhexane, 2- and 4-methylheptane, iso-nonanes and 2-methylnonane) and a few cycloalkanes (derivatives of cyclopentane and cyclohexane) was observed during the incubation. 16S rRNA gene pyrosequencing showed dominance of Peptococcaceae and Anaerolineaceae in Albian MFT and Anaerolineaceae and Syntrophaceae in CNRL MFT bacterial communities with co-domination of Methanosaetaceae and "Candidatus Methanoregula" in archaeal populations during active biodegradation of hydrocarbons. Analysis of exhaled VOCs can provide information on physiology, metabolic processes, oxidative stress and lung diseases. In critically ill patients, VOC analysis may be used to gain complimentary information beyond global clinical parameters. This seems especially attractive in mechanically ventilated patients frequently suffering from impairment of gas exchange. This study was intended to assess (a) the effects of recruitment maneuvers onto VOC profiles, (b) the correlations between electrical impedance tomography (EIT) data and VOC profiles and (c) the effects of recruitment onto distribution of ventilation. Eleven mechanically ventilated patients were investigated during lung recruitment after cardiac surgery. Continuous breath gas analysis by means of PTR-ToF-MS, EIT and blood gas analyses were performed simultaneously. More than 300 mass traces could be detected and monitored continuously by means of PTR-ToF-MS in every patient. Exhaled VOC concentrations varied with recruitment induced changes in minute ventilation and cardiac output. Ammonia exhalation depended on blood pH. The improvement in dorsal lung ventilation during recruitment ranged from 9% to 110%. Correlations between exhaled concentrations of acetone, isoprene, benzene sevoflurane and improvement in regional ventilation during recruitment were observed. Extent and quality of these correlations depended on physico-chemical properties of the VOCs. Combination of continuous real-time breath analysis and EIT revealed correlations between exhaled VOC concentrations and distribution of ventilation. This setup enabled immediate recognition of physiological and therapeutic effects in ICU patients. In a perspective, VOC analysis could be used for non-invasive control and optimization of ventilation strategies.

A breath analysis system based on ultraviolet-absorption spectroscopy was developed by using a hollow optical fiber as a gas cell for real-time monitoring of isoprene in breath. The hollow optical fiber functions as an ultra-small-volume gas cell with a long path. The measurement sensitivity of the system was evaluated by using nitric-oxide gas as a gas sample. The evaluation result showed that the developed system, using a laser-driven, high-intensity light source and a 3-m-long, aluminum-coated hollow optical fiber, could successfully measure nitric-oxide gas with a 50 ppb concentration. An absorption spectrum of a breath sample in the wavelength region of around 200-300 nm was measured, and the measured spectrum revealed the main absorbing components in breath as water vapor, isoprene, and ozone converted from oxygen by radiation of ultraviolet light. The concentration of isoprene in breath was estimated by multiple linear regression. The regression analysis results showed that the proposed analysis system enables real-time monitoring of isoprene during the exhaling of breath. Volatile organic compounds (VOCs) play a central role in atmospheric chemistry. In this work, VOCs in the Botanical Garden of Rio de Janeiro were determined using the TO-15 Method. The park occupies 1,370,000 m 2 in the southern area of the city and is next to the Tijuca Forest, which is considered the largest secondary urban forest in the world. The total VOC concentrations ranged from 43.52 to 168.75 µg m -3 , depending on the sampling site and dates. In terms of concentration isoprene represented 4 %-14 % of the total VOC masses. The results suggested that the differences in biomass, distance from the street and activities within the park affected the concentrations of VOCs. The ratios of isoprene/aromatic compounds were higher than those determined in other areas of the city, confirming that the atmosphere of this green area has the contribution of other sources. Kinetic and mechanistic reactivities were also evaluated.

The cytosol-localised mevalonic acid (MVA) pathway delivers the basic isoprene unit isopentenyl diphosphate (IPP). In higher plants, this central metabolic intermediate is also synthesised by the plastid-localised methylerythritol phosphate (MEP) pathway. Both MVA and MEP pathways conspire through exchange of intermediates and regulatory interactions. Products downstream of IPP such as phytosterols, carotenoids, vitamin E, artemisinin, tanshinone and paclitaxel demonstrate antioxidant, cholesterol-reducing, anti-ageing, anticancer, antimalarial, anti-inflammatory and antibacterial activities. Other isoprenoid precursors including isoprene, isoprenol, geraniol, farnesene and farnesol are economically valuable. An update on the MVA pathway and its interaction with the MEP pathway is presented, including the improvement in the production of phytosterols and other isoprenoid derivatives. Such attempts are for instance based on the bioengineering of microbes such as Escherichia coli and Saccharomyces cerevisiae, as well as plants. The role of the aerial mycelium of the fungus Fusarium solani in the biodegradation of n-pentane was evaluated in a continuous fungal bioreactor (FB) to determine the contribution of the aerial (hyphae) and non-aerial (monolayer) fungal biomass. The experimental results showed that although the aerial biomass fraction represented only 25.9(±3)% on a dry weight basis, it was responsible for 71.6(±4)% of n-pentane removal. The FB attained a maximum elimination capacity (ECmax) of 680(±30) g m(-3) h(-1) in the presence of fungal hyphae (which supported an interfacial area of 5.5(±1.5) × 10(6) m(2) m(-3)). In addition, a mathematical model capable of describing n-pentane biodegradation by the filamentous fungus was also developed and validated against the experimental data. This model successfully predicted the influence of the aerial biomass fraction and its partition coefficient on the n-pentane removal, with EC decreasing from 680(±30) g m(-3) h(-1) to values of 200(±14) g m(-3) h(-1) when the dimensionless partition coefficient increased from 0.21(±0.09) with aerial biomass to 0.88(±0.06) without aerial biomass., (Copyright © 2016 Elsevier Ltd. Breath volatile organic compound (VOC) analysis can open a non-invasive window onto pathological and metabolic processes in the body. Decades of clinical breath-gas analysis have revealed that changes in exhaled VOC concentrations are important rather than disease specific biomarkers. As physiological parameters, such as respiratory rate or cardiac output, have profound effects on exhaled VOCs, here we investigated VOC exhalation under respiratory manoeuvres. Breath VOCs were monitored by means of real-time mass-spectrometry during conventional FEV manoeuvres in 50 healthy humans. Simultaneously, we measured respiratory and hemodynamic parameters noninvasively. Tidal volume and minute ventilation increased by 292 and 171% during the manoeuvre. FEV manoeuvre induced substance specific changes in VOC concentrations. pET-CO2 and alveolar isoprene increased by 6 and 21% during maximum exhalation. Then they decreased by 18 and 37% at forced expiration mirroring cardiac output. Acetone concentrations rose by 4.5% despite increasing minute ventilation. Blood-borne furan and dimethyl-sulphide mimicked isoprene profile. Exogenous acetonitrile, sulphides, and most aliphatic and aromatic VOCs changed minimally. Reliable breath tests must avoid forced breathing. As isoprene exhalations mirrored FEV performances, endogenous VOCs might assure quality of lung function tests. Analysis of exhaled VOC concentrations can provide additional information on physiology of respiration and gas exchange.

In addition to oxygen, hydrocarbons are the most reactive chemical compounds produced by plants into the atmosphere. These compounds are part of the family of volatile organic compounds (VOCs) and are discharged in a great variety of forms. Among the VOCs produced by natural sources such as vegetation, the most studied until today are the isoprene and monoterpene. These substances can play an important role in the chemical balance of the atmosphere of a region. In this project, we develop a methodology to estimate the natural (vegetation) emission of isoprene and monoterpenes and applied it to the Monterrey Metropolitan Area, Mexico and its surrounding areas. Landsat-TM data was used to identify the dominant vegetation communities and field work to determine the foliage biomass density of key species. The studied communities were submontane scrub, oak, and pine forests and a combination of both. We carried out the estimation of emissions for isoprene and monoterpenes compounds in the different plant communities, with two different criteria: (1) taking into account the average foliage biomass density obtained from the various sample point in each vegetation community, and (2) using the foliage biomass density obtained for each transect, associated to an individual spectral class within a particular vegetation type. With this information, we obtained emission maps for each case. The results show that the main producers of isoprene are the communities that include species of the genus Quercus, located mainly on the Sierra Madre Oriental and Sierra de Picachos, with average isoprene emissions of 314.6 ton/day and 207.3 ton/day for the two methods utilized. The higher estimates of monoterpenes were found in the submontane scrub areas distributed along the valley of the metropolitan zone, with an estimated average emissions of 47.1 ton/day and 181.4 tons for the two methods respectively.

Emissions of biogenic volatile organic compounds (BVOCs) from dry ecosystems at high latitudes respond strongly to small increases in temperature, and warm canopy surface temperatures drive emissions to higher levels than expected. However, it is not known whether emissions from wetlands, cooled by through-flowing water and higher evapotranspiration show similar response to warming as in drier ecosystems. Climate change will cause parts of the Arctic to experience increased snow fall, which delays the start of the growing season, insulates soil from low temperatures in winter, and increases soil moisture and possibly nutrient availability. Currently the effects of increasing snow depth on BVOC emissions are unknown. BVOC emissions were measured in situ across the growing season in a climate experiment, which used open top chambers to increase temperature and snow fences to increase winter snow depth. The treatments were arranged in a full factorial design. Measurements took place during two growing seasons in a fen ecosystem in west Greenland. BVOC samples collected by an enclosure technique in adsorbent cartridges were analysed using gas chromatography-mass spectrometry. Gross ecosystem production (GEP) was measured with a closed chamber technique, to reveal any immediate effect of treatments on photosynthesis, which could further influence BVOC emissions. Isoprene made up 84-92% of the emitted BVOCs. Isoprene emission increased 240 and 340% due to an increase in temperature of 1.3 and 1.6°C in 2014 and 2015, respectively. Isoprene emissions were 25 times higher in 2015 than in 2014 most likely due to a 2.4°C higher canopy air temperature during sampling in 2015. Snow addition had no significant effect on isoprene emissions even though GEP was increased by 24%. Human beings continuously emit chemicals into the air by breath and through the skin. In order to determine whether these emissions vary predictably in response to audiovisual stimuli, we have continuously monitored carbon dioxide and over one hundred volatile organic compounds in a cinema. It was found that many airborne chemicals in cinema air varied distinctively and reproducibly with time for a particular film, even in different screenings to different audiences. Application of scene labels and advanced data mining methods revealed that specific film events, namely "suspense" or "comedy" caused audiences to change their emission of specific chemicals. These event-type synchronous, broadcasted human chemosignals open the possibility for objective and non-invasive assessment of a human group response to stimuli by continuous measurement of chemicals in air. Such methods can be applied to research fields such as psychology and biology, and be valuable to industries such as film making and advertising.

Saccharides are important constituents of atmospheric particulate matter (PM). In order to better understand the sources and seasonal variations of saccharides in aerosols in Beijing, China, saccharide composition was measured in ambient PM samples collected at an urban site in Beijing. The highest concentrations of total saccharides in Beijing were observed in autumn, while an episode with abnormal high total saccharide levels was observed from 15 to 23 June, 2011, due to extensive agricultural residue burning in northern China during the wheat harvest season. Compared to the other two categories of saccharides, sugars and sugar alcohols, anhydrosugars were the predominant saccharide group, indicating that biomass burning contributions to Beijing urban aerosol were significant. Ambient sugar and sugar alcohol levels in summer and autumn were higher than those in spring and winter, while they were more abundant in PM2.5 during winter time. Levoglucosan was the most abundant saccharide compound in both PM2.5 and PM10, the annual contributions of which to total measured saccharides in PM2.5 and PM10 were 61.5% and 54.1%, respectively. To further investigate the sources of the saccharides in ambient aerosols in Beijing, the PM10 datasets were subjected to positive matrix factorization (PMF) analysis. Isoprene is the predominant non-methane volatile organic compound emitted to the atmosphere and shapes tropospheric composition and biogeochemistry through its effects on ozone, other oxidants, aerosols, and the nitrogen cycle. Isoprene is emitted naturally by vegetation during daytime, when its photo-oxidation is rapid, and in the presence of nitrogen oxides (NOx) produces ozone and degrades air quality in polluted regions. Here, we show for a city downwind of an isoprene-emitting forest (St. Louis, MO) that isoprene actually peaks at night; ambient levels then endure, owing to low nighttime OH radical concentrations. Nocturnal chemistry controls the fate of that isoprene and the likelihood of a high-ozone episode the following day. When nitrate (NO3) radicals are suppressed, high isoprene persists through the night, providing photochemical fuel upon daybreak and leading to a dramatic late-morning ozone peak. On nights with significant NO3, isoprene is removed before dawn; days with low morning isoprene then have lower ozone with a more typical afternoon peak. This biogenic-anthropogenic coupling expands the daily high-ozone window and likely has an opposite O3-NOx response to what would otherwise be expected, with implications for exposure and air-quality management in cities that, like St. Louis, are downwind of major isoprene-emitting forests.

Leaves of fast-growing, woody bioenergy crops often emit volatile organic compounds (VOC). Some reactive VOC (especially isoprene) play a key role in climate forcing and may negatively affect local air quality. We monitored the seasonal exchange of VOC using the eddy covariance technique in a 'coppiced' poplar plantation. The complex interactions of VOC fluxes with climatic and physiological variables were also explored by using an artificial neural network (Self Organizing Map). Isoprene and methanol were the most abundant VOC emitted by the plantation. Rapid development of the canopy (and thus of the leaf area index, LAI) was associated with high methanol emissions and high rates of gross primary production (GPP) since the beginning of the growing season, while the onset of isoprene emission was delayed. The highest emissions of isoprene, and of isoprene photo-oxidation products (Methyl Vinyl Ketone and Methacrolein, iox ), occurred on the hottest and sunniest days, when GPP and evapotranspiration were highest, and formaldehyde was significantly deposited. Canopy senescence enhanced the exchange of oxygenated VOC. Isoprene is a substantial contributor to global secondary organic aerosol (SOA). The formation of isoprene SOA (SOAI) is highly influenced by anthropogenic emissions. Currently, there is rare information regarding SOAI in polluted regions. In this study, one-year concurrent observation of SOAI tracers was undertaken at 12 sites across China for the first time. The tracers formed from the HO2-channel exhibited higher concentrations at rural sites, while the tracer formed from the NO/NO2-channel showed higher levels at urban sites. 3-Methyltetrahydrofuran-3,4-diols exhibited linear correlations with their ring-opening products, C5-alkenetriols. And the slopes were steeper in the southern China than the northern China, indicating stronger ring-opening reactions there. The correlation analysis of SOAI tracers with the factor determining biogenic emission and the tracer of biomass burning (levoglucosan) implied that the high level of SOAI during summer was controlled by biogenic emission, while the unexpected increase of SOAI during winter was largely due to the elevated biomass burning emission. The estimated secondary organic carbon from isoprene (SOCI) exhibited the highest levels in Southwest China. The significant correlations of SOCI between paired sites implied the regional impact of SOAI in China. Our findings implicate that isoprene origins and SOAI formation are distinctive in polluted regions.

Unlabelled: Ambient air observations of hazardous air pollutant (HAPs), also known as air toxics, derived from routine monitoring networks operated by states, local agencies, and tribes (SLTs), are analyzed to characterize national concentrations and risk across the nation for a representative subset of the 187 designated HAPs. Observations from the National Air Toxics Trend Sites (NATTS) network of 27 stations located in most major urban areas of the contiguous United States have provided a consistent record of HAPs that have been identified as posing the greatest risk since 2003 and have also captured similar concentration patterns of nearly 300 sites operated by SLTs. Relatively high concentration volatile organic compounds (VOCs) such as benzene, formaldehyde, and toluene exhibit the highest annual average concentration levels, typically ranging from 1 to 5 µg/m(3). Halogenated (except for methylene chloride) and semivolatile organic compounds (SVOCs) and metals exhibit concentrations typically 2-3 orders of magnitude lower. Formaldehyde is the highest national risk driver based on estimated cancer risk and, nationally, has not exhibited significant changes in concentration, likely associated with the large pool of natural isoprene and formaldehyde emissions. Benzene, toluene, ethylbenzene, and 1,3-butadiene are ubiquitous VOC HAPs with large mobile source contributions that continue to exhibit declining concentrations over the last decade. Common chlorinated organic compounds such as ethylene dichloride and methylene chloride exhibit increasing concentrations. The variety of physical and chemical attributes and measurement technologies across 187 HAPs result in a broad range of method detection limits (MDLs) and cancer risk thresholds that challenge confidence in risk results for low concentration HAPs with MDLs near or greater than risk thresholds. From a national monitoring network perspective, the ability of the HAPs observational database to characterize the multiple pollutant and spatial scale patterns influencing exposure is severely limited and positioned to benefit by leveraging a variety of emerging measurement technologies., Implications: Ambient air toxics observation networks have limited ability to characterize the broad suite of hazardous air pollutants (HAPs) that affect exposures across multiple spatial scales. While our networks are best suited to capture major urban-scale signals of ubiquitous volatile organic compound HAPs, incorporation of sensing technologies that address regional and local-scale exposures should be pursued to address major gaps in spatial resolution. Caution should be exercised in interpreting HAPs observations based on data proximity to minimum detection limit and risk thresholds.

Background: To deal with the increasingly severe energy crisis and environmental consequences, biofuels and biochemicals generated from renewable resources could serve as a promising alternative for replacing petroleum as a source of fuel and chemicals, among which isoprene (2-methyl-1,3-butadiene) in particular is of great significance in that it is an important platform chemical, which has been used in industrial production of synthetic rubber for tires and coatings or aviation fuel., Results: We firstly introduced fatty acid decarboxylase (OleTJE) from Jeotgalicoccus species into E. coli to directly convert MVA(mevalonate) into 3-methy-3-buten-1-ol. And then to transform 3-methy-3-buten-1-ol to isoprene, oleate hydratase (OhyAEM) from Elizabethkingia meningoseptica was overexpressed in E. coli. A novel biosynthetic pathway of isoprene in E. coli was established by co-expressing the heterologous mvaE gene encoding acetyl-CoA acetyltransferase/HMG-CoA reductase and mvaS gene encoding HMG-CoA synthase from Enterococcus faecalis, fatty acid decarboxylase (OleTJE) and oleate hydratase (OhyAEM). Furthermore, to enhance isoprene production, a further optimization of expression level of OleTJE, OhyAEM was carried out by using different promoters and copy numbers of plasmids. Thereafter, the fermentation process was also optimized to improve the production of isoprene. The final engineered strain, YJM33, bearing the innovative biosynthetic pathway of isoprene, was found to produce isoprene up to 2.2 mg/L and 620 mg/L under flask and fed-batch fermentation conditions, respectively., Conclusions: In this study, by using metabolic engineering techniques, the novel MVA-mediated biosynthetic pathway of isoprene was successfully assembled in E. coli BL21(DE3) with the heterologous MVA upper pathway, OleTJE from Jeotgalicoccus species and OhyAEM from Elizabethkingia meningoseptica. Compared with traditional MVA pathway, the novel pathway is shortened by 3 steps. In addition, this is the first report on the reaction of converting MVA into 3-methy-3-buten-1-ol by fatty acid decarboxylase (OleTJE) from Jeotgalicoccus species. In brief, this study provided an alternative method for isoprene biosynthesis, which is largely different from the well-developed MEP pathway or MVA pathway.

Evaluating the environmental benefits and consequences of urban trees supports their sustainable management in cities. Models such as i-Tree Eco enable decision-making by quantifying effects associated with particular tree species. Of specific concern are emissions of biogenic volatile organic compounds, particularly isoprene, that contribute to the formation of photochemical smog and ground level ozone. Few studies have quantified these potential disservices of urban trees, and current models predominantly use emissions data from trees that differ from those in our target region of subtropical Australia. The present study aimed (i) to quantify isoprene emission rates of three tree species that together represent 16% of the inventoried street trees in the target region; (ii) to evaluate outputs of the i-Tree Eco model using species-specific versus currently used, generic isoprene emission rates; and (iii) to evaluate the findings in the context of regional air quality. Isoprene emission rates of (Myrtaceae) and (Proteaceae) were 2.61 and 2.06 µg g dry leaf weight h, respectively, whereas (Sapindaceae) was a nonisoprene emitter. We substituted the generic isoprene emission rates with these three empirical values in i-Tree Eco, resulting in a 182 kg yr (97%) reduction in isoprene emissions, totaling 6284 kg yr when extrapolated to the target region. From these results we conclude that care has to be taken when using generic isoprene emission factors for urban tree models. Pilots have reported experiencing in-flight hypoxic-like symptoms since the inception of high-altitude aviation. As a result, the need to monitor pilots, in-flight, for the onset of hypoxic conditions is of great interest to the aviation community. We propose that exhaled breath is an appropriate non-invasive medium for monitoring pilot hypoxic risk through volatile organic compound (VOC) analysis. To identify changes in the exhaled breath VOCs produced during periods of reduced O2 levels, volunteers were exposed to simulated flight profiles, i.e. sea level for 5 min, O2 levels found at elevated altitudes for 5 min or placebo and 5 min at 100% O2 recovery gas, using a modified flight mask interfaced with a reduced O2 breathing device. During the course of these test events, time series breath samples from the flight mask and pre/post bag samples were collected and analyzed by gas chromatography/mass spectrometry (GC/MS). Seven compounds (pentanal, 4-butyrolactone, 2-pentanone, 2-hexanone, 2-cyclopenten-1-one, 3-methylheptane and 2-heptanone) were found to significantly change in response to hypoxic conditions. Additionally, the isoprene, 2-methyl-1,3-butadiene, was found to increase following the overall exposure profile. This study establishes an experimental means for monitoring changes in VOCs in response to hypoxic conditions, a computational workflow for compound analysis via the Metabolite Differentiation and Discovery Lab and MatLab(©) software and identifies potential volatile organic compound biomarkers of hypoxia exposure.

Pipelines are convenient, economical and widely used mode of transportation of crude oil. However, the inevitable or otherwise accidents during such transport of crude oil lead to large scale oil spills, which consequently result in both soil and air pollution. When such pollution occurs, crude oil VOC concentrations in air, soil pollution evaluation and VOC propagation in soil provide important evidence for airborne detection of oils spills. Therefore, several issues, including determination method for VOC, isotherm parameters of VOC sorption on soil surfaces, and VOC diffusion flux simulation, are significant. In our previous study, n-butane and n-pentane were proved to be the maximum VOCs in studied crude oils. Therefore, a predictive method using n-pentane as a representative component is proposed in this paper. Firstly, a headspace solid phase microextraction (SPME) method was developed for determination of n-pentane in non-equilibrium mass transfer conditions. Secondly, Brunauer-Emmett-Teller (BET) analysis with liquid nitrogen was carried out to predict isotherm parameters for n-pentane. Finally, two models were used to predict the emission process. Probably influenced by gas vapor density below and above the soil layer, the experimental data amounted to 74% of the deduced value from the simplified analytical model. Gas-phase low volatility organic compounds (LVOC), produced from oxidation of isoprene 4-hydroxy-3-hydroperoxide (4,3-ISOPOOH) under low-NO conditions, were observed during the FIXCIT chamber study. Decreases in LVOC directly correspond to appearance and growth in secondary organic aerosol (SOA) of consistent elemental composition, indicating that LVOC condense (at OA below 1 μg m(-3)). This represents the first simultaneous measurement of condensing low volatility species from isoprene oxidation in both the gas and particle phases. The SOA formation in this study is separate from previously described isoprene epoxydiol (IEPOX) uptake. Assigning all condensing LVOC signals to 4,3-ISOPOOH oxidation in the chamber study implies a wall-loss corrected non-IEPOX SOA mass yield of ∼4%. By contrast to monoterpene oxidation, in which extremely low volatility VOC (ELVOC) constitute the organic aerosol, in the isoprene system LVOC with saturation concentrations from 10(-2) to 10 μg m(-3) are the main constituents. These LVOC may be important for the growth of nanoparticles in environments with low OA concentrations. LVOC observed in the chamber were also observed in the atmosphere during SOAS-2013 in the Southeastern United States, with the expected diurnal cycle. This previously uncharacterized aerosol formation pathway could account for ∼5.0 Tg yr(-1) of SOA production, or 3.3% of global SOA.

Isoprene emissions from poplar (Populus spp.) plantations can influence atmospheric chemistry and regional climate. These emissions respond strongly to temperature, [CO2], and drought, but the superimposed effect of these three climate change factors are, for the most part, unknown. Performing predicted climate change scenario simulations (periodic and chronic heat and drought spells [HDSs] applied under elevated [CO2]), we analyzed volatile organic compound emissions, photosynthetic performance, leaf growth, and overall carbon (C) gain of poplar genotypes emitting (IE) and nonemitting (NE) isoprene. We aimed (1) to evaluate the proposed beneficial effect of isoprene emission on plant stress mitigation and recovery capacity and (2) to estimate the cumulative net C gain under the projected future climate. During HDSs, the chloroplastidic electron transport rate of NE plants became impaired, while IE plants maintained high values similar to unstressed controls. During recovery from HDS episodes, IE plants reached higher daily net CO2 assimilation rates compared with NE genotypes. Irrespective of the genotype, plants undergoing chronic HDSs showed the lowest cumulative C gain. Under control conditions simulating ambient [CO2], the C gain was lower in the IE plants than in the NE plants. In summary, the data on the overall C gain and plant growth suggest that the beneficial function of isoprene emission in poplar might be of minor importance to mitigate predicted short-term climate extremes under elevated [CO2]. Moreover, we demonstrate that an analysis of the canopy-scale dynamics of isoprene emission and photosynthetic performance under multiple stresses is essential to understand the overall performance under proposed future conditions., (© 2015 American Society of Plant Biologists. Isoprene and other volatile organic compounds emitted from vegetation play a key role in governing the formation of ground-level ozone. Emission rates of such compounds depend critically on the plant species. The cultivation of biofuel feedstocks will contribute to future land use change, altering the distribution of plant species and hence the magnitude and distribution of emissions. Here we use relationships between biomass yield and isoprene emissions derived from experimental data for 29 commercially available poplar hybrids to assess the impact that the large-scale cultivation of poplar for use as a biofuel feedstock will have on air quality, specifically ground-level ozone concentrations, in Europe. We show that the increases in ground-level ozone across Europe will increase the number of premature deaths attributable to ozone pollution each year by up to 6%. Substantial crop losses (up to ∼9 Mt y(-1) of wheat and maize) are also projected. We further demonstrate that these impacts are strongly dependent on the location of the poplar plantations, due to the prevailing meteorology, the population density, and the dominant crop type of the region. Our findings indicate the need for a concerted and centralized decision-making process that considers all aspects of future land use change in Europe, and not just the effect on greenhouse gas emissions.

A modified SAPRC-11 (S11) photochemical mechanism with more detailed treatment of isoprene oxidation chemistry and additional secondary organic aerosol (SOA) formation through surface-controlled reactive uptake of dicarbonyls, isoprene epoxydiol and methacrylic acid epoxide was incorporated in the Community Multiscale Air Quality Model (CMAQ) to quantitatively determine contributions of isoprene to summertime ambient SOA concentrations in the eastern United States. The modified model utilizes a precursor-origin resolved approach to determine secondary glyoxal and methylglyoxal produced by oxidation of isoprene and other major volatile organic compounds (VOCs). Predicted OC concentrations show good agreement with field measurements without significant bias (MFB ∼ 0.07 and MFE ∼ 0.50), and predicted SOA reproduces observed day-to-day and diurnal variation of Oxygenated Organic Aerosol (OOA) determined by an aerosol mass spectrometer (AMS) at two locations in Houston, Texas. On average, isoprene SOA accounts for 55.5% of total predicted near-surface SOA in the eastern U.S., followed by aromatic compounds (13.2%), sesquiterpenes (13.0%) and monoterpenes (10.9%). Aerosol surface uptake of isoprene-generated glyoxal, methylglyoxal and epoxydiol accounts for approximately 83% of total isoprene SOA or more than 45% of total SOA. A domain wide reduction of NOx emissions by 40% leads to a slight decrease of domain average SOA by 3.6% and isoprene SOA by approximately 2.6%. Although most of the isoprene SOA component concentrations are decreased, SOA from isoprene epoxydiol is increased by ∼16%.

Volatile organic compounds were collected and analyzed from a variety of indoor and outdoor air samples to test whether human-derived compounds can be readily detected in the air and if they can be associated with human occupancy or presence. Compounds were captured with thermal desorption tubes and then analyzed by gas chromatography with mass spectrometry. Isoprene, a major volatile organic compound in exhaled breath, was shown to be the best indicator of human presence. Acetone, another major breath-borne compound, was higher in unoccupied or minimally occupied areas than in human-occupied areas, indicating that its majority may be derived from exogenous sources. The association of endogenous skin-derived compounds with human occupancy was not significant. In contrast, numerous compounds that are found in foods and consumer products were detected at elevated levels in the occupied areas. Our results revealed that isoprene and many exogenous volatile organic compounds consumed by humans are emitted at levels sufficient for detection in the air, which may be indicative of human presence., (© 2015 WILEY-VCH Verlag GmbH & Co.
