Two new cytochalasin derivatives, peniotrinins A (1) and B (2), three new citrinin derivatives, peniotrinins C–E (4, 5, 7), and one new tetramic acid derivative, peniotrinin F (12), along with nine structurally related known compounds, were isolated from the solid culture of Peniophora sp. SCSIO41203. Their structures, including the absolute configurations of their stereogenic carbons, were fully elucidated based on spectroscopic analysis, quantum chemical calculations, and the calculated ECD. Interestingly, 1 is the first example of a rare 6/5/5/5/6/13 hexacyclic cytochalasin. We screened the above compounds for their anti-prostate cancer activity and found that compound 3 had a significant anti-prostate cancer cell proliferation effect, while compounds 1 and 2 showed weak activity at 10 μM. We then confirmed that compound 3 exerts its anti-prostate cancer effect by inducing methuosis through transmission electron microscopy and cellular immunostaining, which suggested that compound 3 might be first reported as a potential anti-prostate methuosis inducer. In order to evaluate the blood pressure-lowering peptide Tyr-Pro (YP) derived from casein, we wanted to develop an efficient fermentation method. Therefore, we chose to use a jar fermentor for this purpose. Strains with an excellent antihypertensive peptide-releasing ability from casein were selected from basidiomycete fungi that grow well in milk under shaking conditions accompanied by physical stimulation. Among them, the white-rot fungus Peniophora sp., which is suited for growth only in cow’s milk or low-fat milk under vigorous shaking conditions, was found to release peptides and amino acids from milk. When comparing the growth in cow’s milk and low-fat milk, there was no particular difference in the growth of mycelia between the two, but this fungus tended to preferentially consume lactose under low-fat conditions. The fermented milk exhibited good production of the target peptide YP. The expression of many genes encoding proteolytic enzymes, such as aminopeptidases and carboxypeptidases, was observed during the milk fermentation. Furthermore, this fungus showed good growth in a jar fermentor culture using only cow’s milk or low-fat milk, which enabled the efficient production of YP and ACE-inhibitory activity. At this time, it was more effective to use cow’s milk than low-fat milk. These results suggest that Peniophora sp. could be potentially useful in the production of the functional YP peptide from milk.

Agarwood is one of the non-timber forest products that have high economic value. Agarwood is widely used to make incense, perfume and other products. Sapwood on agarwood is a group of secondary metabolites of agarwood plants that form a lump and have a certain color and aroma. The fragrant aroma of sapwood on agarwood is formed due to pathogenic infection of the agarwood tree. Until now, most studies of fungi forming sapwood are only oriented to virulent pathogenic fungi in nature and have never been reported to form hypovirulent agarwood. This study aimed to evaluate the potential of fungi originating from sapwood on agarwood especially those that are hypovirulent in inducing sesquiterpene compounds. This study included exploration, isolation, identification, and induction of fungi associated with sapwood on agarwood from four districts in the Bangka Belitung Islands Province. Hypovirulence test in cucumber sprouts and sesquiterpene induction test on agarwood plantlets was conducted in the laboratory. Based on the isolation results, 48 fungal isolates associated with sapwood on agarwood were found: fungi of genus Fusarium, Trichoderma, Aspergillus, Penicillium, Curvularia, Peniophora, and six isolates were unidentified. Based on the hypovirulence test on cucumber sprouts, 46 isolates of the fungus 46 isolates were virulent and 2 isolates were hypovirulent (isolates 4A and 17A). The induction of sesquiterpene compounds on the agarwood plant was employed using 5 sample isolates consisting of 4 virulent isolates (2A, 7A, 18A, and 25A) and 1 hypovirulent isolate (Isolate 4A). The results showed that hypovirulent isolates were able to produce sesquiterpenes even in small amounts compared with virulent isolates. Isolates produced many sesquiterpene compounds were isolates 18A (Fusarium sp.). Sesquiterpene compounds formed were pinene, terpineol, patchouli alcohol, trimethyl-naphthalene, beta-caryophyllene, camphor, eugenol, trimethyl- benzene, phenanthrene, citronella, eucalyptol, 4-hydroxy-4-methyl-2-pentanone. In this study also found fungi associated with sapwood on agarwood which had never been reported by previous researchers, Peniophora sp. (isolate 25A).

A novel laccase producing Basidiomycete Peniophora sp. (NFCCI-2131) was isolated from pulp and paper mill effluent. The optimal temperature and initial pH for laccase production by the isolate in submerged culture were found to be 30 and 4.6° C, respectively. Maltose (20 g l) and tryptone (1.0 g l) were the most suitable carbon and nitrogen sources for laccase production. Cu (1.0 mM) and veratryl alcohol induced maximum laccase production giving 6.6 and 6.07 U/ml laccase activity, respectively. Under optimised culture conditions, 7.6 U/ml activity was obtained, which was 2.4 times higher than that was achieved in basal medium. An evaluation of the delignification efficiency of the crude enzyme in the presence of redox mediators [2,2'-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid) and (1-hydroxybenzotriazole)] revealed structural changes in lignin and existence of many active centres for both chemical and biological degradation of lignin following enzymatic treatment. Some 25 fungi, including at least 14 basidiomycetes, one ascomycete, and five anamorphic fungi were evaluated for their cellulose-degrading abilities in Difco potato dextrose broth or Difco malt extract broth cultures with cellulosic substrates (e.g., filter paper) in plastic Petri dishes. Among them, Peniophora sp. 06-13 and Phlebia sp. 99-335 reduced the dry weights of the whole cultures with these substrates more than the dry weights of the respective original substrates after 30 days of culture, showing definite cellulose degradation. In the cultures with more than 10 test fungi including Pycnoporus coccineus 84-117, such weight losses did not occur.
