Abstract Objective Bladder cancer(BCa) was a disease that seriously affects patients’ quality of life and prognosis. To address this issue, many researches suggested that the gut microbiota modulated tumor response to treatment; however, this had not been well-characterized in bladder cancer. In this study, our objective was to determine whether the diversity and composition of the gut microbiota or the density of specific bacterial genera influence the prognosis of patients with bladder cancer. Methods We collected fecal samples from a total of 50 bladder cancer patients and 22 matched non-cancer individuals for 16S rDNA sequencing to investigate the distribution of Parabacteroides in these two groups. Further we conducted follow-up with cancer patients to access the impact of different genera of microorganisms on patients survival. We conducted a Fecal Microbiota Transplantation (FMT) and mono-colonization experiment with Parabacteroides distasonis to explore its potential enhancement of the efficacy of anti-PD-1 immunotherapy in MB49 tumor-bearing mice. Immunohistochemistry, transcriptomics and molecular experiment analyses were employed to uncover the underlying mechanisms. Results The 16S rDNA showed that abundance of the genus Parabacteroides was elevated in the non-cancer control group compared to bladder cancer group. The results of tumor growth curves showed that a combination therapy of P. distasonis and ICIs treatment significantly delayed tumor growth and increased the intratumoral densities of both CD4+T and CD8+T cells. The results of transcriptome analysis demonstrated that the pathways associated with antitumoral immune response were remarkably upregulated in the P. distasonis gavage group. Conclusion P. distasonis delivery combined with α-PD-1 mAb could be a new strategy to enhance the effect of anti-PD-1 immunotherapy. This effect might be achieved by activating immune and antitumor related pathways.

Abstract To improve the extraction of coal resources, gob‐side entry driving is gradually being promoted and applied. However, as the thickness of the coal seam increases, the deformation control of the roadway becomes more difficult. Aiming at this problem, a combination of theoretical calculation, numerical simulation, and field test was used to analyze the aspects of the stress environment, surrounding rock properties, and support forms. The phenomenon and mechanization of intensified deformation and failure of roadway with increasing coal seam thickness were revealed. The specific results include: (1) The ratio of principal stress at the excavation position controls the maximum failure depth of roadway, and the direction of principal stress determines the location of the maximum failure depth; (2) As the thickness of the coal seam increases, the properties of the surrounding rock at the excavation position decrease, the principal stress ratio increases, and the deflection angle of the principal stress increases, which lead to an intensification of the deformation and failure of the roadway. Based on the deformation and failure characteristics of the roadway and the shortcomings of the original support form, a control strategy and support scheme based on a new support structure called “Anchor Cable with C‐shaped Tube” is proposed, which has achieved better deformation control effect in on‐site application.

Caries represents one of the most prevalent bacterial infectious diseases in the oral cavity. Over decades, researchers have been actively seeking nanomaterials capable of inhibiting oral pathogenic bacteria. We utilized a one-pot hydrothermal method to synthesize CQDs with multicolor luminescence and CQD-loaded ZnO composite nanomaterials. FE-SEM revealed that the rod-like structure of CQDs/ZnO becomes finer with increased CQDs content. XRD and XPS confirmed the presence of all components within the nanocomposites. We compared the antimicrobial activities of ZnO, 7.7% and 14.3% CQDs/ZnO against Streptococcus mutans, Enterococcus faecalis, Escherichia coli, and Staphylococcus aureus. The 7.7% CQDs/ZnO composite exhibited the most effective antibacterial activity. The 7.7% CQDs/ZnO composite exhibited the most effective antibacterial activity. FE-SEM and ONPG assays demonstrated that 7.7% CQDs/ZnO disrupted the membrane structure and alters membrane permeability. ESR, DPBF and ROS assayed further confirmed that 1O2 produced by 7.7% CQDs/ZnO accumulated at the cell membrane and disrupts its structure.

Abstract Objective Progressive infarction (PI) has a negative effect on functional prognosis. Our study aimed to develop and validate a risk score for predicting PI in patients with anterior circulation single subcortical infarction (ACSSI). Methods Between January 2020 and October 2022, we retrospectively enrolled 638 eligible patients with ACSSI. Two‐thirds of the eligible patients were randomly allocated to the training cohort (n = 425). Another resampling sample was formed through the bootstrap method and was used as the validation group (n = 425). Multivariate logistic regression analysis was used to identify the independent factors associated with PI. Each factor was then point assigned based on β‐coefficient and a risk scoring system was developed. This scoring system was internally validated through 1000‐bootstrap resamplings. The C‐statistic and Hosmer‐Lemeshow test were used to assess model discrimination and calibration. Results PI occurred in 121 patients, accounting for 19.0% of the total patients. A 7‐point NTS score system based on the initial NIHSS score, triglyceride‐glucose index, and the number of infarct slices on axial diffusion‐weighted imaging was developed. The NTS score showed good discrimination and calibration in the training cohort (C‐statistic = 0.686; p value of Hosmer–Lemeshow test = 0.797) and validation cohort (C‐statistic = 0.681; p value of Hosmer–Lemeshow test = 0.451). The three risk levels for predicting PI in the training and validation cohorts based on NTS score were as follows: low (0–2, 9.6% vs. 9.3%), intermediate (3–5, 28.2% vs. 26.7%), and high risk (6–7, 60.2% vs. 57.4%). Interpretation The NTS score is a valid and convenient risk score for predicting PI in ACSSI patients.

ABSTRACTMyotonia congenita (MC) is a rare hereditary muscle disease caused by variants in the CLCN1 gene. Currently, the correlation of phenotype-genotype is still uncertain between dominant-type Thomsen (TMC) and recessive-type Becker (BMC). The clinical data and auxiliary examinations of MC patients in our clinic were retrospectively collected. Electromyography was performed in 11 patients and available family members. Whole exome sequencing was conducted in all patients. The clinical and laboratory data of Chinese MC patients reported from June 2004 to December 2022 were reviewed. A total of 11 MC patients were included in the study, with a mean onset age of 12.64 ± 2.73 years. The main symptom was muscle stiffness of limbs. Warm-up phenomenon and percussion myotonia were found in all patients. Electromyogram revealed significant myotonic charges in all patients and two asymptomatic carriers, while muscle MRI and biopsy showed normal or nonspecific changes. Fourteen genetic variants including 6 novel variants were found in CLCN1. Ninety-eight Chinese patients were re-analyzed and re-summarized in this study. There were no significant differences in the demographic data, clinical characteristics, and laboratory findings between 52 TMC and 46 BMC patients. Among the 145 variants in CLCN1, some variants, including the most common variant c.892 G>A, could cause TMC in some families and BMC in others. This study expanded the clinical and genetic spectrum of Chinese patients with MC. It was difficult to distinguish between TMC and BMC only based on the clinical, laboratory, and genetic characteristics.

As screen sizes are becoming larger and larger, exceeding human physical limitations for direct interaction via touching, remote control is inevitable. However, among the current solutions, inertial gyroscopes are susceptible to positional inaccuracies, and gesture recognition is limited by cameras’ focus depths and viewing angles. Provided that the issue of ghost points can be effectively addressed, grating antenna light-trapping technology is an ideal candidate for multipoint inputs. Therefore, we propose a differential amplitude modulation scheme for grating antenna-based multi-beam optical touch, which can recognize different incidence points. The amplitude of the incident beams was first coded with different pulse widths. Then, following the capture of incident beams by the grating antenna and their conversion into electrical currents by the aligned detector arrays, the incident points of the individual beams were recognized and differentiated. The scheme was successfully verified on an 18-inch screen, where two-point optical touch with a position accuracy error of under 3 mm and a response time of less than 7 ms under a modulation frequency of 10 kHz on both incident beams was achieved. This work demonstrates a practical method to achieve remote multi-point touch, which can make digital mice more accurately represent the users’ pointing directions by obeying the natural three-point one-line aiming rule instantaneously.

Abstract Background The contributors predicting progressive infarction (PI) in patients with anterior circulation single subcortical infarction (ACSSI) and pontine single infarction (PSI) may be unidentical. The role of triglyceride to high‐density lipoprotein cholesterol (TG/HDL‐c) ratio on PI is unclear. The purpose of our study is to evaluate the correlation between TG/HDL‐c ratio and PI in patients with ACSSI or PSI. Methods Between January 2020 and October 2022, we retrospectively enrolled 738 patients including 638 ACSSI patients and 100 PSI patients to analyze. Demographic characteristics, clinical information, and laboratory data were collected within 24 h of admission. Results PI occurred in 143 (19.4%) patients. In univariate analysis, patients with PI had higher initial National Institutes of Health Stroke Scale (NIHSS) scores, higher discharge NIHSS scores, higher levels of fasting glucose, total cholesterol, TG, low‐density lipoprotein cholesterol, and TG/HDL‐c ratio, but lower levels of creatinine compared to patients with non‐PI (p

An important way to reduce urban–rural disparity lies in encouraging migrant workers to return to their hometowns for entrepreneurship. This paper examines the effect of the Integrated Medical Insurance System on the return-to-hometown entrepreneurship among migrant workers. Using microdata from the China Household Finance Survey (CHFS) spanning from 2013 to 2019, we find that the Integrated Medical Insurance System (IMIS) significantly increases the likelihood of migrant workers returning to their hometowns for entrepreneurship by 0.44%. This result remains stable after a series of robustness checks. Heterogeneity results indicate that this “pullback effect” is more pronounced for those who are male and with lower educational levels, higher income, larger social networks, and lower risk preferences. Finally, the interaction between the Mass Entrepreneurship and Innovation policy (MEI) and IMIS can create a more significant combined effect in promoting the return of migrant workers to their hometowns for entrepreneurial activities.

For airborne radar, detecting a low–slow–small (LSS) target is a hot and challenging topic, which results from the rapidly increasing number of non-cooperative flying LSS targets becoming of widespread concern, and the low signal-to-clutter ratio (SCR) of LSS targets results in the targets being particularly easily overwhelmed by the clutter. In this paper, a novel light gradient boosting machine (LightGBM)-based LSS target detection algorithm for airborne radar is proposed. The proposed method, based on the current real-time clutter environment of the range cell to be detected, firstly designs a specific real-time space-time LSS target signal repository with special dimensions and structures. Then, the proposed method creatively designs a new fast-built real-time training feature dataset specifically for the LSS target and the current clutter, together with a series of unique data transformations, sample selection, data restructuring, feature extraction, and feature processing. Finally, the proposed method develops a unique machine learning-based LSS target detection classifier model for the designed training dataset, by fully excavating and utilizing the advantages of the ensemble decision trees-based LightGBM. Consequently, the pre-processed data in the range cell of interest are classified using the proposed algorithm, which achieves LSS target detection by evaluating the output results of the designed classifier. Compared with the traditional classical target detection methods, the proposed algorithm is capable of providing markedly superior performance for LSS target detection. With an appropriate computational time, the proposed algorithm attains the highest probability of detecting LSS targets under the low SCR. The simulation outcomes and detection results with the experimental data are employed to validate the effectiveness and merits of the proposed algorithm.

Cognitive impairment is typically reflected in the time and frequency variations of electroencephalography (EEG). Integrating time-domain and frequency-domain analysis methods is essential to better understand and assess cognitive ability. Timely identification of cognitive levels in early Parkinson’s disease (ePD) patients can help mitigate the risk of future dementia. For the investigation of the brain activity and states related to cognitive levels, this study recruited forty ePD patients for EEG microstate analysis, including 13 with mild cognitive impairment (MCI) and 27 without MCI (control group). To determine the specific frequency band on which the microstate analysis relies, a deep learning framework was employed to discern the frequency dependence of the cognitive level in ePD patients. The input to the convolutional neural network consisted of the power spectral density of multi-channel multi-point EEG signals. The visualization technique of gradient-weighted class activation mapping was utilized to extract the optimal frequency band for identifying MCI samples. Within this frequency band, microstate analysis was conducted and correlated with the Montreal Cognitive Assessment (MoCA) Scale. The deep neural network revealed significant differences in the 1-11.5Hz spectrum of the ePD-MCI group compared to the control group. In this characteristic frequency band, ePD-MCI patients exhibited a pattern of global microstate disorder. The coverage rate and occurrence frequency of microstate A and D increased significantly and were both negatively correlated with the MoCA scale. Meanwhile, the coverage, frequency and duration of microstate C decreased significantly and were positively correlated with the MoCA scale. Our work unveils abnormal microstate characteristics in ePD-MCI based on time-frequency fusion, enhancing our understanding of cognitively related brain dynamics and providing electrophysiological markers for ePD-MCI recognition.

In order to achieve the high-value utilization of heavy tar for the production of enhanced-performance graphite foam carbon, the carbon mesophase was ready from the heavy component of low-temperature coal tar, and the coal tar was modified by styrene-butadiene-styrene (SBS), polyethylene (PE) and ethylene-vinyl-acetate (EVA) copolymers. The order degree of the carbonite mesophase was analyzed using a polarizing microscope test, Fourier transform infrared spectroscopy and X-ray diffraction to screen out the most suitable copolymer type and addition amount. Furthermore, the mechanism of modification by this copolymer was analyzed. The results showed that adding SBS, PE and EVA to coal tar would affect the order of carbonaceous mesophase; however, at an addition rate of 10.0 wt.%, the linear-structure SBS copolymer with a styrene/butadiene ratio (S/B) of 30/70 exhibited the optimal degree of ordering in the carbonaceous mesophase. Its foam carbon prepared by polymer modification is the only one that forms a graphitized structure, with d002 of 0.3430 nm, and the maximum values of Lc and La are 3.54 nm and 2.22 nm, respectively. This is because, under elevated pressure and high-temperature conditions, SBS underwent chain scission, releasing a more significant number of methyl and other free radicals that interacted with the coal tar constituents. As a result, it reduced the affinity density of heavy coal tar molecules, enhanced fluidity, promoted the stacking of condensed aromatic hydrocarbons and increased the content of soluble carbonaceous mesophase, ultimately leading to a more favorable alignment of the carbonaceous mesophase.

For the first time, the ultrasonic surface rolling processing (USRP) technology was applied to a real bearing raceway, and a finite element simulation model for USRP on a bearing raceway was established. Practical tests were conducted to evaluate the surface integrity of the bearing raceway before and after USRP treatment. The USRP parameters were optimized using Taguchi-gray correlation analysis. The results show that the optimal process parameters for the 7224 bearing raceway are: an amplitude of 10 μm, a static pressure of 2400 N, a spindle speed of 45 rad/min, and rolling times of 7. In the process of USRP, residual compressive stress results from inhomogeneous elastic and plastic deformation. Adjacent tempered martensitic laths merge and deflect at a certain angle along the rolling direction to form directional grain stacks, resulting in a flocculent plastic deformation layer. The USRP based on the finishing mechanism of “peak-cutting and valley-filling” can improve the surface micro-morphology, reduce roughness value Ra to approximately 0.16 μm, and effectively eliminate microscopic cracks caused by grinding. The micro-hardness of the bearing raceway surface increased from 760 HV to 840 HV under the dual action of surface grain refinement and work hardening effect.

Background/purpose: Recent studies have pointed to the crucial role of microRNAs (miRNAs) in chronic periodontitis (CP). This study investigated the regulation and potential mechanisms of miR-28-5p in CP patients and lipopolysaccharide (LPS)-induced periodontal ligament cells (PDLCs). Materials and methods: 76 CP patients and 71 periodontally healthy subjects were included. RT-qPCR was employed to examine miR-28-5p and sphingosine kinase −1 (SPHK1) in subjects’ gingival sulcus fluid and PDLCs. To comprehend the potential impacts of both natural phenomena and human activities on ecological risk, a thorough examination of the spatial and temporal evolution characteristics of Landscape Ecological Risk (LER) in arid river basins is imperative. This investigation holds paramount importance for the proactive prevention and mitigation of LER, as well as for the preservation of ecological security within these basins. In this scholarly inquiry, the Kriya River Basin (KRB) serves as the focal point of analysis. Leveraging three historical land use and land cover (LULC) images and incorporating a diverse array of drivers, encompassing both natural and anthropogenic factors, the study employs the PLUS model to forecast the characteristics of LULC changes within the basin under three distinct scenarios projected for the year 2030. Concurrently, the research quantitatively assesses the ecological risks of the basin through the adoption of the Landscape Ecological Risk Assessment (LERA) methodology and the Spatial Character Analysis (SCA) methodology. The results showed the following: (1) The study area is primarily composed of grassland and unused land, which collectively account for over 97% of the total land. However, there has been a noticeable rise in cropland and considerable deterioration in grassland between 2000 and 2020. The key observed change in LULC involves the transformation of grassland and unused land into cropland, forest, and construction land. (2) The overall LER indices for 2000, 2010, and 2020 are 0.1721, 0.1714, and 0.16696, respectively, showing strong positive spatial correlations and increasing autocorrelations over time. (3) Over time, human activities have come to exert a greater influence on LER compared to natural factors between 2000 and 2020. (4) In the natural development scenario (NDS), cropland protection scenario (CPS), and ecological priority scenario (EPS), the LER of KRB experienced notable variations in the diverse 2030 scenarios. Notably, the CPS exhibited the highest proportion of low-risk areas, whereas Daryaboyi emerged as the focal point of maximum vulnerability. These findings offer theoretical and scientific support for sustainable development planning in the watershed.

In order to study the characteristics and sources of heavy metal pollution within different grassland soil categories in Altay, 147 soil samples were collected in Fuyun County according to grassland type, and the contents of seven heavy metals (Cu, Pb, Cr, Zn, As, Ni, and Cd) were determined. The heavy metal pollution in the soil was evaluated using the Nemerow pollution and geological accumulation index methods. The absolute principal component score–multiple linear regression model (APCS-MLR) model was used to analyze the sources of heavy metals and the contribution rate of the pollution sources. The results showed that (1) the heavy metal content of 0~10 cm soil in the mining area was generally higher than the remaining two layers. Natural grassland was the most seriously contaminated, and the order of its contamination was Zn > Pb > Ni > Cd > Cu > Cr > As. (2) The evaluation results of the comprehensive pollution index showed that Ni and Cd were the main pollutants in five grassland types, and the evaluation results of the geological accumulation index method showed that natural grasslands were more polluted. (3) Three pollution sources were obtained according to the analysis using the APCS-MLR model, including natural and mining activity sources (32.6%), industrial coal combustion and sewage sources (24.7%), and an unknown source (42.7%). The results of the study can provide a basis for pollution control and ecological protection in the grasslands of Altay.

IntroductionCerebral sparganosis is a rare parasitic infection of the brain tissue. The remission of MRI change and clinical symptom has been used to evaluate the therapeutic effect. However, there is no study to correlate the serum IgG antibody level of sparganum to the prognosis of disease after treatment. Methods87 patients with cerebral sparganosis were collected from three medical centers. Clinical symptoms and MRI changes were evaluated at 12 months after initial treatment, and serum IgG antibody level of sparganum was evaluated at 2, 6, and 12 months after treatment. The positive cut-off value was based on 2.1 times the optical density (OD) of negative control. The index value was defined as the sample OD divided by the cut-off value.ResultsAmong the 87 patients after treatment, 71 patients had good clinical outcomes, and 16 had poor clinical outcomes. The area under the curve (AUC) showed that the index value measured at 12 months after treatment had the best prediction effect, with a value of 2.014. In the good-outcome group, the index values were less than 2.014 in all 71 patients, and only 8 patients had mildly enhanced residual lesions on MRI. In the poor-outcome group, the index values were more than 2.014 in all 16 patients, and all patients still showed significantly enhanced lesions on MRI. Compared with poor-outcome patients, only 2 patients with good outcomes had disease recurrence after treatment.DiscussionThis study provided evidence that the serum IgG antibody level of sparganum was a promising biomarker to evaluate the prognosis of patients with cerebral sparganosis after treatment.

Anchor cables often break in various forms. Tensile failure can be resisted by using high-strength materials, while there is no good solution for shear failure. A new supporting structure called Anchor Cable and C-shaped tube that can bear the transverse shear force was invented and a preliminary introduction to the mechanism of the structure is given. Field investigations and in-situ stress tests were carried out on the panel 3210 in the Nanguan Mine. The failure modes of the surrounding rock and cables and the in-situ stress test results show that the working face is obviously affected by the horizontal stress. Thus, a new full-section anchor cable and C-shaped tube support scheme were proposed. The results of FLAC3D show that the new scheme is excellent in controlling the surface convergence and plastic zone extension. The rationality of the scheme was verified by two-and-a-half months of monitoring. Compared with the original scheme, the amount of surface displacement was at least 20% smaller, and the time for the displacement to reach equilibrium was at least 27.4% shorter. The surface of the roadway is flat without supporting material damaging. The new scheme is worth popularizing in similar working conditions.

Abstract Background Urogenital microbiota may be associated with the recurrence of bladder cancer, but the underlying mechanism remains unclear. The notion that microbiota can upregulate PD-L1 expression in certain epithelial tumors to promote immune escape has been demonstrated. Thus, we hypothesized that the urogenital microbiota may be involved in the recurrence and progression of non-muscle invasive bladder cancer (NMIBC) by upregulating the PD-L1 expression. To test this hypothesis, we investigated the relationship between urogenital microbial community and PD-L1 expression in male patients with NMIBC. Results 16S rRNA gene sequencing was performed to analyse the composition of urogenital microbiota, and the expression of PD-L1 in cancerous tissues was detected by immunohistochemistry. The subjects (aged 43–79 years) were divided into PD-L1-positive group (Group P, n = 9) and PD-L1-negative group (Group N, n = 19) respectively based on their PD-L1 immunohistochemical results. No statistically significant differences were found in the demographic characteristics between group P and N. We observed that group P exhibited higher species richness (based on Observed species and Ace index, both P < 0.05). Furthermore, subgroup analysis showed that the increase in number of PD-L1 positive cells was accompanied by increased richness of urogenital microbiota. Significantly different composition of urogenital microbiota was found between group P and group N (based on weighted Unifrac and unweighted Unifrac distances metric, both P < 0.05). Enrichment of some bacterial genera (e.g., Leptotrichia, Roseomonas, and Propionibacterium) and decrease of some bacterial genera (e.g., Prevotella and Massilia) were observed in group P as compared with group N. These findings indicated that these genera may affect the expression of PD-L1 through some mechanisms to be studied. Conclusion Our study provided for the first time an overview of the association between urogenital microbiota and PD-L1 expression in male patients with NMIBC, indicating that urogenital microbiota was an important determinant of PD-L1 expression in male NMIBC patients.

The effect of transistors in abutted rows on charge sharing is investigated by changing the configuration of the transistors in abutted rows in this work. 3D TCAD numerical simulations indicate that the existence of transistors in abutted rows can mitigate the occurring probability of charge sharing, especially charge sharing induced by ion striking at the vicinity of n-well contact. The simulations also indicate that the single event double transient (SEDT) pulse width is reduced obviously by the transistors in abutted rows for ion strike location near n-well contact. A 65 nm test chip was designed in commercial 65 nm twin-well bulk CMOS process, and heavy-ion experiment was conducted. The experiment results agree well with the simulation results, which indicates that the effect of transistors in abutted rows on single event sensitivity and the occurring probability of charge sharing is more than 10%, and then considering the effect of transistors in abutted rows is necessary in nanometer technology.

Chromatin regulators (CRs) are essential upstream regulatory factors of epigenetic modification. The role of CRs in the pathogenesis of renal ischemia-reperfusion injury (IRI) remains unclear. We analyzed a bioinformatic analysis on the differentially expressed chromatin regulator genes in renal IRI patients using data from public domains. The hub CRs identified were used to develop a risk prediction model for renal IRI, and their expressions were also validated using Western blot, qRT-PCR, and immunohistochemistry in a murine renal IRI model. We also examined the relationships between hub CRs and infiltrating immune cells in renal IRI and used network analysis to explore drugs that target hub CRs and their relevant downstream microRNAs. The results of machine learning methods showed that five genes (DUSP1, GADD45A, GADD45B, GADD45G, HSPA1A) were upregulated in renal IRI, with key roles in the cell cycle, p38 MAPK signaling pathway, p53 signaling pathway, FoxO signaling pathway, and NF-κB signaling pathway. Two genes from the network, GADD45A and GADD45B (growth arrest and DNA damage-inducible protein 45 alpha and beta), were chosen for the renal IRI risk prediction model. They all showed good performance in the testing and validation cohorts. Mice with renal IRI showed significantly upregulated GADD45A and GADD45B expression within kidneys compared to sham-operated mice. GADD45A and GADD45B showed correlations with plasmacytoid dendritic cells (pDCs) in infiltrating immune cell analysis and enrichment in the MAPK pathway based on the weighted gene co-expression network analysis (WGCNA) method. Candidate drugs that target GADD45A and GADD45B include beta-escin, sertraline, primaquine, pimozide, and azacyclonol. The dysregulation of GADD45A and GADD45B is related to renal IRI and the infiltration of pDCs, and drugs that target GADD45A and GADD45B may have therapeutic potential for renal IRI.

Here, the authors show that periaqueductal gray neuronal ensembles encode the sequential hunting motor program, which might provide a framework for decoding complex instinctive behaviors.

ObjectiveOveractive bladder (OAB) is a disease that seriously affects patients’ quality of life and mental health. To address this issue, more and more researchers are examining the relationship between OAB treatment and urinary microecology. In this study, we sought to determine whether differences in treatment efficacy were related to microbiome diversity and composition as well as the abundance of specific genera. Machine learning algorithms were used to construct predictive models for urine microbiota-based treatment of OAB.MethodsUrine samples were obtained from 64 adult female OAB patients for 16S rRNA gene sequencing. Patients’ overactive bladder symptom scores (OABSS) were collected before and after mirabegron treatment and patients were divided into effective and ineffective groups. The relationship between the relative abundance of certain genera and OABSS were analyzed. Three machine learning algorithms, including random forest (RF), supporting vector machine (SVM) and eXtreme gradient boosting (XGBoost) were utilized to predict the therapeutic effect of mirabegron based on the relative abundance of certain genera in OAB patients’ urine microbiome.ResultsThe species composition of the two groups differed. For one, the relative abundance of Lactobacillus was significantly higher in the effective group than in the ineffective group. In addition, the relative abundance of Gardnerella and Prevotella in the effective group was significantly lower than in the ineffective group. Alpha-diversity and beta-diversity differed significantly between the two groups. LEfSe analysis revealed that Lactobacillus abundance increased while Prevotella and Gardnerella abundance decreased in the effective group. The Lactobacillus abundance ROC curve had high predictive accuracy. The OABSS after treatment was negatively correlated with the abundance of Lactobacillus, whereas the relationship between OABSS and Prevotella and Gardnerella showed the opposite trend. In addition, RF, SVM and XGBoost models demonstrated high predictive ability to assess the effect of mirabegron in OAB patients in the test cohort.ConclusionsThe results of this study indicate that urinary microbiota might influence the efficacy of mirabegron, and that Lactobacillus might be a potential marker for evaluating the therapeutic efficacy of mirabegron in OAB patients.

ObjectiveThe diagnosis of neuronal intranuclear inclusion disease (NIID) is currently based on CGG repeat expansion in the 5′UTR of the NOTCH2NLC gene, or p62-positive intranuclear inclusions in skin biopsy. The purpose of this study is to explore the value of non-invasive pathological findings in urine sediment cells from NIID patients.Materials and methodsTen patients with clinically suspected NIID were enrolled for skin biopsy and gene screening. Morning urine (500 ml) was collected from each patient, and cell sediment was obtained by centrifugation. Urine cytology, including Giemsa staining, p62 immunostaining, and electron microscopic examination, were conducted on cell sediment.ResultsThe main clinical symptoms of 10 patients included episodic disturbance of consciousness, cognitive impairment, tremor, limb weakness, and so on. Cerebral MRI showed that 9 patients had linear DWI high signal in the corticomedullary junction. Genetic testing found that the number of CGG repeat ranged from 96 to 158 in the NOTCH2NLC gene. Skin biopsy revealed that all patients showed p62-positive intranuclear inclusions in 18.5 ± 6.3% of the duct epithelial cells of sweat gland. In contrast, urine sediment smears revealed that only 3 patients had p62 positive intranuclear inclusions in 3.5 ± 1.2% of the sedimentary cells. Ultrastructural examinations showed that intranuclear inclusions were also identified in the cell sediment of the 3 patients.ConclusionUrine cytology may be a new and non-invasive pathological diagnosis technique for some NIID patients, although the positive rate is not as high as that of skin biopsy, which is a sensitive and reliable pathological method for NIID.

Background: Primary serine deficiency disorders have a broad range of the phenotypic spectrum. As an inborn error of metabolism, individuals with severe phenotype may be easily recognized with Neu-Laxova syndrome. However, late-onset mild phenotypes may be underdiagnosed and will lead to disastrous consequences due to treatment delays.Materials and Methods: Clinical features of patients with serine deficiency disorders were summarized in two unrelated patients. Skin and sural nerve biopsies were conducted on the patients. Whole exome sequencing (WES) was performed in the index patients. Sanger sequencing was used to analyze family cosegregation.Results: Patient 1 was a 19-year-old male presenting with infancy-onset ichthyosis and juvenile-onset neuropathy. Patient 2 was a 17-year-old male manifesting childhood-onset ichthyosis and juvenile-onset neuropathy. Except for nystagmus, no other developmental or neurodegenerative disorders were found in the patients. Electrophysiological studies indicated a severe sensorimotor axonal neuropathy with a possible demyelinating component. High-dose oral L-serine and glycine completely alleviated skin lesions and only slightly improved neuropathy symptoms. Skin biopsies showed typical features consistent with ichthyosis and severe loss of unmyelinated axons. Sural biopsies revealed a severe loss of axons and a few thinly myelinated fibers. WES found the same homozygous variant c.43G > C (p.A15P) in the PSAT1 gene, which was cosegregated in the two families.Conclusions: The skin and nervous system may be the main affected targets in serine deficiency disorders. Our patients show a more simple and mild phenotype of PSAT1-related serine deficiency disorder. The pathological changes and regenerative ability of skin and peripheral nerves determine their response to serine supplements.

Background: CircPUM1 acts as an oncogene in a variety of tumors, and there is no related research on oral squamous cell carcinoma. This study aimed to evaluate the clinical significance of CircPUM1 in oral squamous cell carcinoma radiotherapy.Methods: Radio-resistant cell lines were established by increasing the X-ray dose. Analysis of CircPUM1 expression in oral squamous cell carcinoma was carried out using bioinformatics tools. Cell proliferation was analyzed with CCK-8 and colony formation. Protein and gene expressions were detected by Western blotting and qPCR. RNA interference inhibits endogenous gene expression. A luciferase reporter system and immunoprecipitation were used to validate the target of CircPUM1.Result: CircPUM1 was highly expressed in OSCC. The higher the expression level of CircPUM1 in OSCC, the worse the clinical features and prognosis. Knockdown of CircPUM1 enhances the sensitivity of OSCC cells to X-rays, and expression of exogenous CircPUM1 makes OSCC cells acquire radiation resistance. The absence of CircPUM1 blocked the cells in the G0/G1 phase and triggered apoptosis. The prediction of mir-580-binding site, luciferase reporter system, and immunoprecipitation confirmed that mir-580 is the binding site of CircPUM1. In addition, STAT3 was predicted and confirmed as the binding site of mir-580. Overexpression of STAT3 partially attenuated the radiosensitivity of OSCC cells to knockdown of CircPUM1.Conclusion: CircPUM1 has the oncogene expression profile in oral squamous cell carcinoma; patients with high expression of CircPUM1 have less benefit from radiotherapy and need more frequent follow-up. In addition, CircPUM1 may be a potential therapeutic target for oral squamous cell carcinoma. The CircPUM1/mir-580/STAT3 axis has a certain effect on the radiosensitivity of OSCC. These results suggest that patients with low expression of CircPUM1 may gain more benefits.

The frequent occurrence of meteorological disasters in China has caused huge losses to agriculture. Risk assessment serves as a bridge from disaster crisis management to disaster risk management. Therefore, it is necessary to carry out a refined comprehensive risk assessment of meteorological disasters in typical areas. However, several limitations remain in the disaster loss risk research, such as too coarse resolution and too single risk indicator. Additionally, less research has examined geographical information on risk clustering and barycenter migration, as well as temporal information on the sustainability of trends. Consequently, it is significant to unearth the geographical and temporal information on disaster loss and identify the refined spatial and temporal evolution pattern of crop risk. For this reason, we evaluated the risk of corn production in Gansu Province. First, based on maize yield data, a risk evaluation index system was constructed using the characteristics of variation trends, fluctuations, and extreme values of disaster losses. Then, the spatial distribution patterns and temporal evolution characteristics of maize production risks on a county scale in Gansu Province were determined using spatial analysis and climate diagnosis technology. The results show that there is a large interdecadal fluctuation in risk. In the 1980s, 1990s, 2000s, and 2010s, the average yield reduction rates of maize in Gansu Province were −11.8%, −12.6%, −8.7%, and −8.5%, and the proportions of counties with severe yield reduction were 34.8%, 44.4%, 20.8%, and 9.7%, respectively. Second, most counties belong to medium-low or low-risk areas for maize production. High-risk counties are primarily located in eastern and southern Gansu, whereas low-risk counties are mostly found along the Hexi Corridor. Third, most risk indicators exhibit some geographical aggregation. The Jiuquan region falls within the low-low-risk aggregation zone. In contrast, the Qingyang region is a high-high aggregation zone with a gradual expansion trend. Four, each risk indicator’s geographical barycenter migrates over a complicated path, but the direction and distance vary considerably. The comprehensive risk migrates along the south-northwest-southeast trajectory, albeit at a shorter distance. Five, the proportion of counties with a medium, medium-severe, severe, and total yield reduction tended to decline. In addition, the annual precipitation is significantly or very significantly correlated with most risk indicators and the comprehensive risk level. The results can guide agricultural production processes at all levels, as well as government disaster prevention.

The Gangdese magmatic rocks of the southern Lhasa terrane, are generally thought to be an important window to witness the formation and evolution of the Neo-Tethys oceanic opening, subduction, and closure, and India-Eurasian continental collision. We investigated a new occurrence of granodiorite in the Nuocang district of western Gangdese, southern Lhasa terrane, and conducted a series of analyses on their petrology, chronology, and geochemistry. The Nuocang granodiorites have the zircon U-Pb ages of 151–154 Ma, which suggest that Late Jurassic granitoids are present in the western Gangdese of southern Lhasa terrane. They are relatively high in SiO2, Al2O3, low K2O, Na2O, and Sr/Y ratios, enrichments of LILE and LREE, and depletion of HFSE, with the positive correlation between Rb and Th, and negative correlations between SiO2 and P2O5, Rb, and Y, showing the features of I-type granites. The relatively high (87Sr/86Sr)i values from 0.712231 to 0.712619, low εNd(t) values from −9.56 to −8.99, together with the negative εHf(t) values from −10.8 to −5.0 (mean value −8.9) suggested that the Nuocang granodiorites probably sourced from the partial melting of the ancient Lhasa terrane, with parts of mantle materials involving in. Combined with the previous geochronology and geochemical data of Mesozoic magmas in the Gangdese belt, as well as the Late Jurassic granodiorite, in this paper, we propose that the Nuocang granodiorites formed in a continental margin arc environment triggered by the northward subduction of Neo−Tethys oceanic crust.

The free section of prestressed anchor cable is a weak part of support. A new supporting structure named Anchor Cable and C-Shaped Tube, which can bear transverse shear force, solves the problem that rock bolt and anchor cable are prone to shear failure in the free section and also solves the contradiction between high preload and low shear bearing capacity of support materials. Double-shear tests of Anchor Cable and C-Shaped Tube with smooth joint planes were carried out. Double-shear tests were carried out on the anchor cables with the diameter of 21.6 mm and the same type of Anchor Cable and C-Shaped Tube under different preload conditions. The influence of the preload on the shear performance of supporting materials and the enhancement effect of Anchor Cable and C-Shaped Tube supporting structure on the shear performance of anchor cables were analyzed. The test results confirm that Anchor Cable and C-Shaped Tube can improve the transverse shear resistance of the supporting material and increase the axial ultimate bearing capacity of the anchor cable during the shearing process. It is found that, during the double-shear test, the fracture form of the supporting materials is tensile fracture, and when it is sheared, Anchor Cable and C-Shaped Tube can reduce the stress concentration of the interaction between the joint surface and the surrounding rock and reduce the damage to the surrounding rock.

Deep roadway deformation due to soft rock, rock dip, and horizontal tectonic stress is uneven and asymmetrical primarily in large loose zones. Traditional anchor support is influenced by the yield strength and shear strength of the anchors and has a limited prestress capacity or shear resistance. When the roadway roof is laminated rock or when the roadway passes through layered rock or rock interfaces, interlayer sliding commonly occurs, which can easily lead to anchor cables being sheared off. The tape tunnel in the Zhengling Mine passes through several rock strata and requires anchors to achieve a high shear resistance and prestress. To solve these problems, an anchor cable and C-shaped tube that can bear lateral shear forces were developed, and a full-section anchor cable and C-shaped tube support system were created based on extruded arch theory. Numerical results from FLAC3D show that the new scheme effectively controls surface convergence and plastic zone extension. Field tests have demonstrated that the amount of surface displacement was at least 42% smaller in the new support scheme. The extruded arch formed by the highly prestressed anchor cable and concrete spray layer can effectively control the bulking load within the loose zone, and the ACC effectively resists interlayer shear.

Template dictionary construction is an important issue in sparse representation (SP)-based tracking algorithms. In this article, a drift-free visual tracking algorithm is proposed via the construction of an effective template dictionary. The constructed dictionary is composed of three categories of atoms (templates): nonpolluted atoms, variational atoms, and noise atoms. Moreover, the linear combinations of nonpolluted atoms are also added to the dictionary for the diversity of atoms. All the atoms are selectively updated to capture appearance changes and alleviate the model drifting problem. A bidirectional tracking process is used and each process is optimized by two-step SP, which greatly reduces the computational burden. Compared with other related works, the constructed dictionary and tracking algorithm are both robust and efficient.

In this paper, the optimization of the extraction/purification process of multiple components was performed by the entropy weight method (EWM) combined with Plackett–Burman design (PBD) and central composite design (CCD). We took the macroporous resin purification of Astragalus saponins as an example to discuss the practicability of this method. Firstly, the weight of each component was given by EWM and the sum of the product between the componential content and its weight was defined as the comprehensive score, which was taken as the evaluation index. Then, the single factor method was adopted for determining the value range of each factor. PBD was applied for screening the significant factors. Important variables were further optimized by CCD to determine the optimal process parameters. After the combination of EWM, PBD and CCD, the resulting optimal purification conditions were as follows: pH value of 6.0, the extraction solvent concentration of 0.15 g/mL, and the ethanol volume fraction of 75%. Under the optimal conditions, the practical comprehensive score of recoveries of saponins was close to the predicted value (n = 3). Therefore, the present study provided a convenient and efficient method for extraction and purification optimization technology of multiple components from natural products.

IR-drop is a fundamental constraint by almost all integrated circuits (ICs) physical designs, and many iterations of timing engineer change order (ECO), IR-drop ECO, or other ECO are needed before design signoff. However, IR-drop analysis usually takes a long time and wastes so many resources. In this work, we develop a fast dynamic IR-drop predictor based on a machine learning technique, XGBoost, and the prediction method can be applied to vector-based and vectorless IR-drop analysis simultaneously. Correlation coefficient is often used to characterize the symmetry of prediction data and golden data, and our experiments show that the prediction correlation coefficient is more than 0.96 and the average error is no more than 1.3 mV for two industry designs, which are of 2.4 million and 3.7 million instances, respectively, and that the analysis is speeded up over 4.3 times compared with the IR-drop analysis by commercial tool, Redhawk.

The influence of aerosols on climate varies greatly within different spatial zones. China has a very prominent summer monsoon climate and summer monsoon activity basically determines the climate distribution pattern. Consequently, we need to understand the aerosol optical properties and spatial distribution under the background of summer monsoon activity in China, which is the basis for further research on the impact of aerosols on the climate system. Based on Moderate Resolution Imaging Spectroradiometer (MODIS) and Modern-Era Retrospective Analysis for Research and Applications (MERRA-2) data, the spatial response of the high aerosol optical depth (AOD) region in China to the advance and retreat of summer monsoon was analyzed. The main types of aerosol and the contribution of each type of aerosol particles to the total AOD were discussed. The results showed that before the landing of summer monsoon, the high value areas of AOD were distributed in the eastern Sichuan Basin, Changsha, Wuhan and Pearl River Delta regions. With the northward advance of the monsoon, the high value areas moved to the transition region affected by the summer monsoon and the AOD in this region was highly sensitive to the summer monsoon. The main aerosol types were dust and sulfate in this region and the contribution to total AOD was 27% and 57%, respectively; before the monsoon onset, the contribution of dust to total AOD was 16%, and that of sulfate was 18%; after the monsoon onset, the contribution of dust decreased by half to 8%, while the contribution of sulfate aerosol increased to 20%.

The construction of a virtual debris flow disaster environment is of great significance in debris flow disaster prevention, risk assessment, accurate simulation, and disaster emergency response. However, existing research on virtual disaster environments mainly focus on the specific visualization task requirements of single-type users, and the multilevel visualization task requirements of multitype users are generally not met. In this paper, an adaptive construction method for virtual debris flow disaster environments driven by multilevel visualization task is proposed based on the characteristics of users with different professional knowledge backgrounds and requirements in disaster emergency response scenarios. The on-demand construction of virtual debris flow disaster environments and the corresponding diverse organization and dynamic scheduling technologies are discussed in detail. Finally, the Qipan Gully debris flow disaster is selected for experimental analysis, and a prototype system is developed. The experimental results show that the proposed method can adaptively construct virtual debris flow disaster environments according to the multilevel visualization task requirements of multitype users in debris flow disaster emergency response scenarios. This approach can provide efficient rendering of disaster scenes and appropriate disaster information to multitype users who are involved in debris flow disaster emergency response scenarios.

Scientific and appropriate visualizations increase the effectiveness and readability of disaster information. However, existing fusion visualization methods for disaster scenes have some deficiencies, such as the low efficiency of scene visualization and difficulties with disaster information recognition and sharing. In this paper, a fusion visualization method for disaster information, based on self-explanatory symbols and photorealistic scene cooperation, was proposed. The self-explanatory symbol and photorealistic scene cooperation method, the construction of spatial semantic rules, and fusion visualization with spatial semantic constraints were discussed in detail. Finally, a debris flow disaster was selected for experimental analysis. The experimental results show that the proposed method can effectively realize the fusion visualization of disaster information, effectively express disaster information, maintain high-efficiency visualization, and provide decision-making information support to users involved in the disaster process.

In a triple-well NMOSFET, a deep n+ well (DNW) is buried in the substrate to isolate the substrate noise. The presence of this deep n+ well leads to changes in single-event transient effects compared to bulk NMOSFET. In space, a single cosmic particle can deposit enough charge in the sensitive volume of a semiconductor device to cause a potential change in the transient state, that is, a single-event transient (SET). In this study, a quantitative characterization of the effect of a DNW on a SET in a 65 nm triple-well NMOSFET was performed using heavy ion experiments. Compared with a bulk NMOSFET, the experimental data show that the percentages of average increase of a SET pulse width are 22% (at linear energy transfer (LET) = 37.4 MeV·cm2/mg) and 23% (at LET = 22.2 MeV·cm2/mg) in a triple-well NMOSFET. This study indicates that a triple-well NMOSFET is more sensitive to a SET, which means that it may not be appropriate for radiation hardened integrated circuit design compared with a bulk NMOSFET.

Tetrabutylammonium hydroxide with methanol as an additive was found to be a highly active catalyst for the cleavage of 4-aryl-2-methyl-3-butyn-2-ols. The reaction was performed at 55–75 °C and gave terminal arylacetylenes in good to excellent yields within several minutes. Compared with the usual reaction conditions (normally >110 °C, several hours), this novel catalyst system can dramatically decrease the reaction time under much milder conditions.
