2. Mitochondria from osteolineage cells regulate myeloid cell-mediated bone resorption

Abstract Interactions between osteolineage cells and myeloid cells play important roles in maintaining skeletal homeostasis. Herein, we find that osteolineage cells transfer mitochondria to myeloid cells. Impairment of the transfer of mitochondria by deleting MIRO1 in osteolineage cells leads to increased myeloid cell commitment toward osteoclastic lineage cells and promotes bone resorption. In detail, impaired mitochondrial transfer from osteolineage cells alters glutathione metabolism and protects osteoclastic lineage cells from ferroptosis, thus promoting osteoclast activities. Furthermore, mitochondrial transfer from osteolineage cells to myeloid cells is involved in the regulation of glucocorticoid-induced osteoporosis, and glutathione depletion alleviates the progression of glucocorticoid-induced osteoporosis. These findings reveal an unappreciated mechanism underlying the interaction between osteolineage cells and myeloid cells to regulate skeletal metabolic homeostasis and provide insights into glucocorticoid-induced osteoporosis progression.

Abstract Mitochondria, with their intricate networks of functions and information processing, are pivotal in both health regulation and disease progression. Particularly, mitochondrial dysfunctions are identified in many common pathologies, including cardiovascular diseases, neurodegeneration, metabolic syndrome, and cancer. However, the multifaceted nature and elusive phenotypic threshold of mitochondrial dysfunction complicate our understanding of their contributions to diseases. Nonetheless, these complexities do not prevent mitochondria from being among the most important therapeutic targets. In recent years, strategies targeting mitochondrial dysfunction have continuously emerged and transitioned to clinical trials. Advanced intervention such as using healthy mitochondria to replenish or replace damaged mitochondria, has shown promise in preclinical trials of various diseases. Mitochondrial components, including mtDNA, mitochondria-located microRNA, and associated proteins can be potential therapeutic agents to augment mitochondrial function in immunometabolic diseases and tissue injuries. Here, we review current knowledge of mitochondrial pathophysiology in concrete examples of common diseases. We also summarize current strategies to treat mitochondrial dysfunction from the perspective of dietary supplements and targeted therapies, as well as the clinical translational situation of related pharmacology agents. Finally, this review discusses the innovations and potential applications of mitochondrial transplantation as an advanced and promising treatment.

Abstract Transcortical vessels (TCVs) provide effective communication between bone marrow vascular system and external circulation. Although osteocytes are in close contact with them, it is not clear whether osteocytes regulate the homeostasis of TCVs. Here, we show that osteocytes maintain the normal network of TCVs by transferring mitochondria to the endothelial cells of TCV. Partial ablation of osteocytes causes TCV regression. Inhibition of mitochondrial transfer by conditional knockout of Rhot1 in osteocytes also leads to regression of the TCV network. By contrast, acquisition of osteocyte mitochondria by endothelial cells efficiently restores endothelial dysfunction. Administration of osteocyte mitochondria resultes in acceleration of the angiogenesis and healing of the cortical bone defect. Our results provide new insights into osteocyte-TCV interactions and inspire the potential application of mitochondrial therapy for bone-related diseases.

Abstract Background While laparoscopic assistance is often entrusted to less experienced individuals, such as residents, medical students, and operating room nurses, it is important to note that they typically receive little to no formal laparoscopic training. This deficiency can lead to poor visibility during minimally invasive surgery, thus increasing the risk of errors. Moreover, operating room nurses and medical students are currently not included as key users in structured laparoscopic training programs. Objectives The aim of this study is to evaluate the laparoscopic skills of OR nurses, clinical medical postgraduate students, and residents before and after undergoing virtual reality training. Additionally, it aimed to compare the differences in the laparoscopic skills among different groups (OR nurses/Students/Residents) both before and after virtual reality training. Methods Operating room nurses, clinical medical postgraduate students and residents from a tertiary Grade A hospital in China in March 2022 were selected as participants. All participants were required to complete a laparoscopic simulation training course in 6 consecutive weeks. One task from each of the four training modules was selected as an evaluation indicator. A before-and-after self-control study was used to compare the basic laparoscopic skills of participants, and laparoscopic skill competency was compared between the groups of operating room nurses, clinical medical postgraduate students, and residents. Results Twenty-seven operating room nurses, 31 clinical medical postgraduate students, and 16 residents were included. 7. Construction of nursing-sensitive quality indicators for the care of patients with prone position ventilation using the Delphi method

Abstract Background Prone position ventilation (PPV) has gradually become an adjuvant treatment to improve oxygenation in patients with acute respiratory distress syndrome. Scientific and comprehensive evaluation of the quality of nursing care for patients with PPV is of great significance to ensure the effectiveness of treatment and patient safety. However, there are no established objective indicators for evaluating the quality of nursing care for patients with PPV. This study intended to identify a set of scientific, systematic and clinically applicable nursing-sensitive quality indicators for the care of patients with PPV. Methods Based on the Donabedian structure-process-result theory model, the quality evaluation indicators of nursing care for patients with PPV were preliminarily constructed based on an evidence-based perspective, and two rounds of Delphi surveys were conducted with the purpose of collecting opinions from a panel of independent experts. Results The questionnaire recovery rates of the two rounds of correspondence were 100.00% and 95.00%, the recovery rates of expert opinions were 80.00% and 26.32%, the expert authority coefficient values were 0.89, and the Kendall coordination coefficient W values were 0.110 and 0.133, respectively. The final nursing-sensitive quality indicators for the care of patients with PPV included 3 first-level indicators, 9 s-level indicators and 29 third-level indicators. Conclusion The constructed nursing-sensitive quality indicators for the care of patients with PPV involve quality supervision during the whole process of PPV from three dimensions: structure, process and results. These indicators have strong operability, reliability, practicability and scientificity and can provide a reference for the quality evaluation and monitoring of nursing care for patients with PPV. Implications for nursing management The quality indicators of nursing care for patients with PPV constructed in this research are scientific and reliable, and the content of the quality indicators can better reflect the technical characteristics of special nursing. Nursing managers are encouraged to use these quality indicators to evaluate the quality of clinical nursing care and improve safety for patients with PPV.

Abstract As key organelles involved in cellular metabolism, mitochondria frequently undergo adaptive changes in morphology, components and functions in response to various environmental stresses and cellular demands. Previous studies of mitochondria research have gradually evolved, from focusing on morphological change analysis to systematic multiomics, thereby revealing the mitochondrial variation between cells or within the mitochondrial population within a single cell. The phenomenon of mitochondrial variation features is defined as mitochondrial heterogeneity. Moreover, mitochondrial heterogeneity has been reported to influence a variety of physiological processes, including tissue homeostasis, tissue repair, immunoregulation, and tumor progression. Here, we comprehensively review the mitochondrial heterogeneity in different tissues under pathological states, involving variant features of mitochondrial DNA, RNA, protein and lipid components. Then, the mechanisms that contribute to mitochondrial heterogeneity are also summarized, such as the mutation of the mitochondrial genome and the import of mitochondrial proteins that result in the heterogeneity of mitochondrial DNA and protein components. Additionally, multiple perspectives are investigated to better comprehend the mysteries of mitochondrial heterogeneity between cells. Finally, we summarize the prospective mitochondrial heterogeneity-targeting therapies in terms of alleviating mitochondrial oxidative damage, reducing mitochondrial carbon stress and enhancing mitochondrial biogenesis to relieve various pathological conditions. The possibility of recent technological advances in targeted mitochondrial gene editing is also discussed.

BackgroundWith population aging, the prevalence of both cancer and atrial fibrillation (AF) have increased. However, there is scarce epidemiological data concerning the comorbid state of cancer and AF in low- and middle-income countries, including China. ObjectiveWe aimed to evaluate the site-, sex-, and age-specific profiles of cancer and AF comorbidities in Chinese populations. MethodsData from the Shanghai Municipal Health Commission database between 2015 and 2020 were screened, covering all medical records of Shanghai residents with medical insurance. Site-specific cancer profiles were evaluated for the population with AF relative to the age- and sex-adjusted population of residents without AF. The sex distribution and peak age of cancer diagnosis were also assessed. ResultsA total of 25,964,447 adult patients were screened. Among them, 22,185 patients presented cancers comorbid with AF (median 77, IQR 67-82 years of age; men: n=13,631, 61.44%), while 839,864 presented cancers without AF (median 67, IQR 57-72 years of age; men: n=419,020, 49.89%), thus yielding a higher cancer prevalence among residents with AF (8.27%) than among those without AF (6.05%; P

10. Mannan oligosaccharides selenium ameliorates intestinal mucosal barrier, and regulate intestinal microbiota to prevent Enterotoxigenic Escherichia coli -induced diarrhea in weaned piglets

Enterotoxigenic Escherichia coli (ETEC) is a common diarrheal pathogen in humans and animals. To prevent and treat ETEC induced diarrhea, we synthesized mannan oligosaccharide selenium (MOSS) and studied its beneficial effect on ETEC-induced diarrhea. A total of 32 healthy weaned piglets (6.69 ± 0.01 kg) were randomly divided into four groups: NC group (Basal diet), MOSS group (0.4 mg/kg MOSS supplemented diet), MOET group (0.4 mg/kg MOSS supplemented diet + ETEC treatment), ETEC group (ETEC treatment). NC and ETEC group fed with basal diet, MOSS and MOET group fed with the MOSS supplemented diet. On the 8th and 15th day of the experiment, MOET and ETEC group were gavaged with ETEC, and NC and MOSS group were gavaged with stroke-physiological saline solution. Our data showed that dietary MOSS supplementation increased average daily gain (ADG) and average daily feed intake (ADFI) and significantly decreased diarrhea index and frequency in ETEC-treated piglets. MOSS did not affect the α diversity and β diversity of ileal microbial community, but it significantly decreased the proportion of lipopolysaccharide biosynthesis in ileal microbial community. MOSS supplementation regulated colonic microbiota community composition, which significantly increased carbohydrate metabolism, and inhibited lipopolysaccharide biosynthesis pathway in colonic microbial community. Moreover, MOSS significantly decreased inflammatory stress, and oxidative stress in ETEC treated piglets. Furthermore, dietary MOSS supplementation significantly decreased intestinal barrier permeability, and alleviated ETEC induced intestinal mucosa barrier irritation. In conclusion, our study showed that dietary MOSS supplementation ameliorated intestinal mucosa barrier, and regulated intestinal microbiota to prevent ETEC induced diarrhea in weaned piglets.

Abstract Background In modern animal husbandry, breeders pay increasing attention to improving sow nutrition during pregnancy and lactation to favor the health of neonates. Sow milk is a main food source for piglets during their first three weeks of life, which is not only a rich repository of essential nutrients and a broad range of bioactive compounds, but also an indispensable source of commensal bacteria. Maternal milk microorganisms are important sources of commensal bacteria for the neonatal gut. Bacteria from maternal milk may confer a health benefit on the host. Methods Sow milk bacteria were isolated using culturomics followed by identification using 16S rRNA gene sequencing. To screen isolates for potential probiotic activity, the functional evaluation was conducted to assess their antagonistic activity against pathogens in vitro and evaluate their resistance against oxidative stress in damaged Drosophila induced by paraquat. In a piglet feeding trial, a total of 54 newborn suckling piglets were chosen from nine sows and randomly assigned to three treatments with different concentrations of a candidate strain. Multiple approaches were carried out to verify its antioxidant function including western blotting, enzyme activity analysis, metabolomics and 16S rRNA gene amplicon sequencing. Results The 1240 isolates were screened out from the sow milk microbiota and grouped into 271 bacterial taxa based on a nonredundant set of 16S rRNA gene sequencing. Among 80 Pediococcus isolates, a new Pediococcus pentosaceus strain (SMM914) showed the best performance in inhibition ability against swine pathogens and in a Drosophila model challenged by paraquat. Pretreatment of piglets with SMM914 induced the Nrf2-Keap1 antioxidant signaling pathway and greatly affected the pathways of amino acid metabolism and lipid metabolism in plasma. In the colon, the relative abundance of Lactobacillus was significantly increased in the high dose SMM914 group compared with the control group. Conclusion P. pentosaceus SMM914 is a promising probiotic conferring antioxidant capacity by activating the Nrf2-Keap1 antioxidant signaling pathway in piglets. Purpose This study compares doctor staffing level and the scale of medical education in China with those of other countries and proposes policy recommendations for future adjustments to the scale of China's medical education. Design/Approach/Methods This study employs a literature review and descriptive analysis. Findings China had 1.98 medical doctors per 1,000 people in 2018, ranking 85th out of the 193 member-states of the World Health Organization (WHO). In 2017, China had 1.99 practicing doctors per 1,000 people, only ranking above Turkey (1.88) in Organisation for Economic Co-operation and Development (OECD) countries. China had only 10.28 medical graduates per 100,000 people—placing in the bottom third of OECD countries. China's provision of 1.4 medical schools per 10 million people was also significantly lower than the global average (3.9). However, the average number of students enrolled in medical schools (509) in China was significantly higher than the global average (160). Originality/Value Although the scale of admission in undergraduate medical education must be expanded in China, this needs to be achieved while controlling the average number of medical students per school and reducing enrollment in low-quality medical schools. Furthermore, it is necessary to establish new medical schools while improving the operating level of existing ones.

13. Dynamic Marketing Resource Allocation with Two-Stage Decisions

In the precision marketing of a new product, it is a challenge to allocate limited resources to the target customer groups with different characteristics. We presented a framework using the distance-based algorithm, K-nearest neighbors, and support vector machine to capture customers’ preferences toward promotion channels. Additionally, online learning programming was combined with machine learning strategies to fit a dynamic environment, evaluating its performance through a parsimonious model of minimum regret. A resource optimization model was proposed using classification results as input. In particular, we collected data from an institution that provides financial credit products to capital-constrained small businesses. Our sample contained 525,919 customers who will be introduced to a new product. By simulating different scenarios between resources and demand, we showed an up to 22.42% increase in the number of expected borrowers when KNN was performed with an optimal resource allocation strategy. Our results also show that KNN is the most stable method to perform classification and that the distance-based algorithm has the most efficient adoption with online learning.

This study investigated sediment-bound magnetic properties and selected trace elements level in the karst ditch wetland, Caohai National Nature Reserve, Guizhou Province, China. Sediment-bound magnetic signals were quantified using low-frequency mass magnetic susceptibility (χLF), anhysteretic remanent magnetization susceptibility (χARM), saturation isothermal remanent magnetization (SIRM), and percentage frequency-dependent susceptibility (χfd%). Concentrations of Cd, Cr, Sb and Zn were determined using inductively coupled plasma mass spectrometry. Sediment χLF, χARM, SIRM, and χfd% were higher than those of bedrocks and mainly altered by the pedogenic processes. The estimated χfd% ranged from 6.15 % to 14.62 % and reflected the magnetic grain sizes were largely concentrated in the range of superparamagnetic particles. The elevated concentrations of sediment-bound Cd, Cr, Sb, and Zn supported the significant contribution of the anthropogenic sources in the karst ditch wetlands. Weaning stress induces the depressed digestive and absorptive capacity and insufficient intestinal energy supply. Medium-chain fatty acid glycerides have shown to improve the growth performance and intestinal barrier function of weaned piglets in the previous study. This study was aimed to investigate the regulation of medium-chain fatty acid glyceride on the nutrient absorption and energy utilization of weaned piglets. Nighty healthy weaned piglets were randomly assigned into five treatments: NP (Normal protein, normal-protein diet no antibiotics included); NC (Negative control, low-protein diet no antibiotics included); PC (Positive control, low-protein diet +75 mg/kg quinocetone, 20 mg/kg virginiamycin and 50 mg/kg aureomycin); MCT (tricaprylin + tricaprin group, low-protein diet + tricaprylin + tricaprin); GML (glycerol monolaurate group, low-protein diet + glycerol monolaurate). The results showed that GML treatment increased the ALP activity, concentrations of serine and methionine, MCT treatment increased concentrations of serine and 3-methyl-histidine but decreased TG concentration in serum. MCT and GML supplementations significantly promoted the lipase activity in the jejunum and ileum, as well as the AMP content in the ileal mucosa. GML addition significantly decreased the contents of butyric acid, isobutyric acid and total volatile fatty acid. In addition, medium chain fatty acid glycerides altered gene expressions involved in lipid metabolism, which showing the increases of AMPK2, CD36 and CGI58 and the decreases of MGAT2 and DGAT2 in the liver, as well as the increases of CD36, CGI58, MGAT2 and DGAT2 in the subcutaneous adipose tissue. These findings showed that medium-chain fatty acid glyceride can effectively improve the absorption of nutrients and lipid metabolism of piglets to meet the energy demand of weaned piglets, and then regulate the growth and development of weaned piglets.

BackgroundThe COVID-19 pandemic has led to concerns around its subsequent impact on global health.ObjectiveTo investigate the health-seeking behavior, reflected by ECG utilization patterns, of patients with non-COVID-19 diseases during and after COVID-19 epidemic.MethodsTaking advantage of the remote ECG system covering 278 medical institutions throughout Shanghai, the numbers of medical visits with ECG examinations during the lockdown (between January 23 and April 7, 2020), post-lockdown (between April 8 and December 31, 2020) and post-SARS-CoV-2 (between January 23 and April 7, 2021) periods were analyzed and compared against those during the same periods of the preceding years (2018 and 2019).ResultsCompared with the same period during pre-COVID years, the number of medical visits decreased during the lockdown (a 38% reduction), followed by a rebound post-lockdown (a 17% increase) and a fall to the baseline level in post-SARS-CoV-2 period. The number of new COVID-19 cases announced on a given day significantly correlated negatively with the numbers of medical visits during the following 7 days. Medical visit dynamics differed for various arrhythmias. Whereas medical visits for sinus bradycardia exhibited a typical decrease-rebound-fallback pattern, medical visits for atrial fibrillation did not fall during the lockdown but did exhibit a subsequent increase during the post-lockdown period. By comparison, the volume for ventricular tachycardia remained constant throughout this entire period.ConclusionThe ECG utilization patterns of patients with arrhythmias exhibited a decrease-rebound-fallback pattern following the COVID-19 lockdowns. Medical visits for diseases with more severe symptoms were less influenced by the lockdowns, showing a resilient demand for healthcare.

In a multi-robot system, situation assessment evaluates the current situation quantitatively to help decision-makers make the best decision. Conventional situation assessment methods ignore the initiative of each robot, so it often encounters bottlenecks. Collaborative intelligence shows better performance than a single global decision. To address this problem, this work introduces a deep learning-based fuzzy adaptive method (DLFA) to achieve the real-time situation assessment for a multi-robot system. The proposed method employs the shortest path faster algorithm to achieve information sharing between agents. The shortest path faster algorithm ensures that the agent distributes its state information to its teammates in the fastest way. Each agent gets the information from teammates and treats their state as the observation of the scene. Deep neural network maps current observations into a local situation assessment result by combining a large number of nonlinear processing layers. Finally, each local assessment result is regarded as a brick to construct the final situation assessment via a fuzzy ensemble method. Experimental results show that the proposed method outperforms competitors.

21. Organic Phosphorus Mineralization Dominates the Release of Internal Phosphorus in a Macrophyte-Dominated Eutrophication Lake

Macrophyte-dominated eutrophication (MDE) lakes have attracted wide attention due to the high phosphorus (P) loading in sediments that poses a wide spread risk for P release and pollution management. However, because of the superior productivity characteristics, the role of organic P mineralization in sediments in the internal P loading of MDE lake is still under debate. This study investigated the release dynamic of P in the sediments of Lake Caohai, a MDE lake in southwest of China, using a combination of the modified Huffer sequential extraction method, 31P nuclear magnetic resonance spectroscopy (NMR), and composite diffusive gradient in thin films (DGT) technology. Results showed that the apparent P diffusion flux at the sediment-water interface was remarkably high, with a mean value of 0.37 mg m−2 d−1. The phosphate ester organophosphorus components (i.e., Mono-P and Diester-P) continuously deposited and degraded in the sediments maintained the high productivity of the lake, and the mineralization process plays a critical role in the release of internal P. Although the content of inorganic P in sediment is relatively high (accounting for approximately 60% of total P), the reductive mechanism based on P-containing iron oxide/hydroxide has a low contribution to the internal P loading, as was indicated by the low release rate of P-combination iron-manganese (Fe-Mn)/iron-aluminum (Fe-Al) (BD-P and NaOH-P) and the insignificant positive correlations between DGT-labile P and DGT-labile Fe in the sediment cores. Additionally, organic P in sediments could transfer to P-combination Fe-Al/Fe-Mn. However, in severely expropriated environments, the enrichment of P-combination Fe-Al/Fe-Mn in surface sediments inhibited the mineralization of monophosphate to some degree. Taken together, this study emphasized the impact of sediment organic P loading on the release of internal P in lake, highlighting that organic P is also the valuable objects for avoiding eutrophication of MDE lakes.

22. Evaluating the Influence of Different Recommended Dietary Levels of Cu and Zn on Finishing Pigs

This study was conducted to evaluate the effects of dietary supplementation of different recommended levels of Cu and Zn on antioxidant capacity, tissue mineral status, minerals excretion, meat quality, digestive enzyme activity, and metal transporters in finishing pigs. A total of 120 pigs (with an average initial body weight (BW) of 70.0 ± 2.1 kg) were randomly divided into four treatments: (1) basal diet without added Cu or Zn (control), (2) basal diet+35 mg cupreous N-carbamylglutamate chelate (NCG-Cu) +150 mg zinc-methionine chelate (Zn-Met) (AC), (3) basal diet + 3.0 mg of NCG-Cu + 43 mg Zn-Met (CN), and (4) basal diet + 3.5 mg NCG-Cu + 50 mg Zn-Met (NRC100). Pig growth performance was not affected by the level of Cu or Zn. Among the four treatments, the AC treatment had the highest concentration (P < 0.05) of glutathione peroxidase (GSH-Px). Pigs fed the AC diet had the highest (P < 0.05) liver Zn, fecal Cu, and fecal Zn among the four treatments. The protein levels of trypsin and aminopeptidase N (APN) in the intestinal mucosa showed their highest levels (P < 0.05) in the NRC100 and AC treatments. The mRNA levels of trypsinogen and APN were significantly up-regulated (P < 0.05) in the AC, CN, and NRC100 treatments compared with the control. The mRNA levels for the Zn transporter genes SLC30A1 (ZnT1) and SLC30A2 (ZnT2) were significantly up-regulated (P < 0.05) in the AC treatment, and the mRNA levels for SLC39A4 (ZIP4) and metallothionein 1 (MT) in the AC, CN, and NRC100 treatments were significantly up-regulated (P < 0.05) compared with the control. Meat quality were not affected (P > 0.05) by the different recommended levels of Cu and Zn. These results indicated that the supplemental Cu and Zn levels routinely used in AC diets in Chinese commercial feed enterprises should be reduced.

In this study, based on the physical and mechanical parameters of Camellia oleifera, the mechanical model of Camellia oleifera was rebuilt and analysed to reveal the damage mechanism of fruit shell breakage. The results revealed that under the same conditions (e.g., axial loading form), the stress of the fruit uniformly diffused from the extrusion point to the periphery and depth, and the maximum equivalent stress was 9.4104 Mpa. While under radial loading, the stress of the fruit extended axially along the dorsal line of the tea seed, and the maximum equivalent stress was 6.9467 Mpa. The maximum stress under the two loading modes occurred at the joint between the middle column of the shell and the calyx. The increased loading displacement decreased the stress on the fruit, making it easier to break the shell of Camellia oleifera by radial extrusion. The results can serve as a reference for the development of different equipment to break the shell of the Camellia oleifera fruit.

One novel technology that can be useful is topology design. Compared with traditional design, topology design not only has more undetermined parameters, but also its topology variables have a greater impact on the design goals. In this perspective, we propose the concept of Artificial Intelligent Topology Design (AITD) for aerodynamic configuration design based on topology design and artificial intelligence technology and discuss its potential in the application of the new concept of aerodynamic configuration design.

25. A Driving-Adapt Strategy for the Electric Vehicle with Magneto-Rheological Fluid Transmission Considering the Powertrain Characteristics

The additional energy consumption caused by the incompatibility between existing electric vehicle (EV) powertrain characteristics and driving conditions inevitably curbs the promotion and development of EVs. Hence, there is an urgent demand for the driving-adapt strategy, which aims to minimize EV energy consumption due to both powertrain characteristics and driving conditions. In order to fully explore the EV driving-adapt potential, this paper equips the EV with a magneto-rheological fluid transmission (MRFT). First, an EV dynamics analysis of the driving conditions, the powertrain model considering the energy transmission process, and the driving-adapt transmission model considering magneto-rheological fluid (MRF) is conducted to clarify the quantitative relation between the driving conditions and the powertrain. Second, a driving-adapt optimization strategy in the specific driving condition is proposed. Finally, the results and discussions are executed to study (i) the determination of the MRFT fixed speed ratio and variable speed ratio range, (ii) the application potential analysis of the proposed strategy, and (iii) the feasibility analysis of the proposed strategy. The results indicate that (i) the urban driving condition has higher requirements for the MRFT, (ii) EVs equipped with MRFT achieve the expected driving performance at the most states of charge (SOCs) and environmental temperatures, except for the SOC lower than 10%, and (iii) the driving time with efficiency greater than 80% can be increased by the MRFT from 10.1% to 58.7% and from 66.8% to 88.8% in the urban and suburban driving conditions, respectively. Thus, the proposed driving-adapt strategy for the EV equipped with the MRFT has the potential to alleviate or eliminate the traffic problems caused by the incompatibility of the EV powertrain characteristics and the driving conditions.

This study aimed to investigate the beneficial effect of baicalin–zinc complex (BZN) on intestinal microorganisms in deoxynivalenol (DON)-challenged piglets and the association between intestinal microorganisms and host immunity and hormone secretion. Forty weaned piglets were randomly divided into four treatments with 10 piglets in each treatment: (1) control (Con) group (pigs fed basal diet); (2) DON group (pigs fed 4 mg DON/kg basal diet); (3) BZN group (pigs fed 0.5% BZN basal diet); and (4) DBZN group (pigs fed 4 mg DON/kg and 0.5% BZN basal diet). The experiment lasted for 14 days. The BZN supplementation in DON-contaminated diets changed the intestinal microbiota composition and increased intestinal microbial richness and diversity of piglets. The BZN supplementation in DON-contaminated diets also alleviated the inflammatory responses of piglets and modulated the secretion of hormones related to the growth axis. Moreover, microbiota composition was associated with inflammatory and hormone secretion. In conclusion, BZN alleviated inflammatory response and hormone secretion in piglets, which is associated with the intestinal microbiome.

Container seaport congestion is a challenging problem in improving the service level and optimizing evacuating container vessels after congestion. There is a lack of research on container vessel evacuation strategies for continuous terminals. In this article, the weight of the objective function is regarded as the index for the service priority of vessels. The effects of the service priority on the continuous terminal are analyzed by establishing a mixed integer programming model. The model minimizes the total weighted delay departure time of vessels. Two sets of weight values are adopted, including handling volume of each ship and the squared handling volume, then the optimization results are compared with the unweighted scenario. The model is solved using a genetic algorithm. Lianyungang Port is selected as a case study. The results show that the method using the square of handled container volume is more conducive to ensuring the shipping period of large vessels after congestion. Besides, the quay crane number of large vessels affecting the scheduling strategy is discussed. The method proposed in this article provides a new idea for arranging scheduling strategies in other ports under congestion situations, which can better ensure the planned shipping period of large vessels.

28. Synergistic Effects and Ecological Responses of Combined In Situ Passivation and Macrophytes toward the Water Quality of a Macrophytes-Dominated Eutrophic Lake

Combined use of in situ passivation and macrophytes is a valuable technology that exerts remarkable effects on aquatic systems. However, the effectiveness and ecological functions of this combined technology for macrophytes-dominated eutrophic (MDE) lakes with organophosphorus-controlled internal phosphorus (P) loading were poorly understood. In this study, aquatic simulation experiments were performed to study the combination of La-modified materials (LMM; La-modified bentonite (LMB), and La/Al co-modified attapulgite (LAA)) with macrophytes (Myriophyllum verticillatum L. (MVL), Hydrilla verticillata (Linn. f.) royle (HVR), and Ceratophyllum demersum L. (CDL)) for the control of P mobility in the water column, and to investigate the passivator effects on the physiological characteristics of macrophytes. The mineralization of organophosphates (BD−Po, HCl−Po, and Res−Po) is an important factor for maintaining high internal P loadings and overlying water P concentrations in the experiments. Compared with individual treatment groups, the reduction of internal P release flux and porewater SRP concentrations was more obvious in the combined treatments. Moreover, the redox-sensitive P forms transformation is more pronounced in the surface sediments. In the LAA+M group, internal P release flux was reduced by 55% and 55% compared with individual passivators and macrophytes retreatment groups, respectively. In contrast, the LMB+M group decreased by 16% and 46%, respectively. Simultaneously, LMM had less effect on macrophytes traits compared with individual macrophytes group and enhanced the absorption of phosphate by macrophytes. The phosphate content of macrophytes in the LAA+M and LMB+M groups increased by 24% and 11%, respectively, in comparison with the individual macrophytes group. Results concluded that the combination of passivator and macrophytes enhanced the effect of ecological restoration and exerts a synergistic effect on internal P pollution with macrophytes.

Ni-rich layered transition-metal oxides with high specific capacity and energy density are regarded as one of the most promising cathode materials for next generation lithium-ion batteries. However, the notorious surface impurities and high air sensitivity of Ni-rich layered oxides remain great challenges for its large-scale application. In this respect, surface impurities are mainly derived from excessive Li addition to reduce the Li/Ni mixing degree and to compensate for the Li volatilization during sintering. Owing to the high sensitivity to moisture and CO2 in ambient air, the Ni-rich layered oxides are prone to form residual lithium compounds (e.g. LiOH and Li2CO3) on the surface, subsequently engendering the detrimental subsurface phase transformation. Consequently, Ni-rich layered oxides often have inferior storage and processing performance. More seriously, the residual lithium compounds increase the cell polarization, as well as aggravate battery swelling during long-term cycling. This review focuses on the origin and evolution of residual lithium compounds. Moreover, the negative effects of residual lithium compounds on storage performance, processing performance and electrochemical performance are discussed in detail. Finally, the feasible solutions and future prospects on how to reduce or even eliminate residual lithium compounds are proposed.

Multiple organ dysfunction syndrome can follow severe infection or injury, but its relationship to exercise is not well understood. Previous studies have observed that prolonged strenuous exercise can lead to transiently increased level and/or activity of markers for systemic inflammatory response and multiple organ damage. However, few studies have analyzed the pathogenesis of the inflammatory response and subsequent multi-organ injury in exhaustive exercise conditions. In this study, we established a rat model of repetitive bouts of exhaustive running (RBER) and investigated its effects on multiple organ damage. Rats were subjected to RBER in either uphill or downhill running modes daily for a period of 7 days. Morphologically, RBER causes tissue structural destruction and infiltration of inflammatory cells in the skeletal muscles and many visceral organs. RBER also causes sustained quantitative changes in leukocytes, erythrocytes, and platelets, and changes in the concentration of blood inflammatory factors. These inflammatory alterations are accompanied by increases in serum enzyme levels/activities which serve as functional markers of organ damage. In general, RBER in the downhill mode seemed to cause more damage evaluated by the above-mentioned measures than that produced in the uphill mode. A period of rest could recover some degree of damage, especially for organs such as the heart and kidneys with strong compensatory capacities. Together, our data suggest that, as a result of multi-organ interactions, RBER could cause a sustained inflammatory response for at least 24 h, resulting in tissue lesion and ultimately multiple organ dysfunction.

According to the engineering phenomenon of the galloping of ice-coated transmission lines at certain wind speeds, this paper proposes a novel type of energy harvester based on the galloping of a flexible structure. It uses the tension generated by the galloping structure to cause periodic strain on the piezoelectric cantilever beam, which is highly efficient for converting wind energy into electricity. On this basis, a physical model of fluid–structure interaction is established, and the Reynolds-averaged Navier–Stokes equation and SST K -ω turbulent model based on ANSYS Fluent are used to carry out a two-dimensional steady computational fluid dynamics (CFD) numerical simulation. First, the CFD technology under different grid densities and time steps is verified. CFD numerical simulation technology is used to simulate the physical model of the energy harvester, and the effect of wind speed on the lateral displacement and aerodynamic force of the flexible structure is analyzed. In addition, this paper also carries out a parameterized study on the influence of the harvester’s behavior, through the wind tunnel test, focusing on the voltage and electric power output efficiency. The harvester has a maximum output power of 119.7 μW/mm3 at the optimal resistance value of 200 KΩ at a wind speed of 10 m/s. The research results provide certain guidance for the design of a high-efficiency harvester with a square aerodynamic shape and a flexible bluff body.

Head and neck squamous cell carcinomas (HNSCCs) are closely linked with immunosuppression, accompanied by complex immune cell functional activities. The abnormal competition between costimulatory and coinhibitory signal molecules plays an important role in the malignant progression of HNSCC. This review will summarize the features of costimulatory molecules (including CD137, OX40 as well as CD40) and coinhibitory molecules (including CTLA-4, PD-1, LAG3, and TIM3), analyze the underlying mechanism behind these molecules' regulation of the progression of HNSCC, and introduce the clinic application. Vaccines, such as those targeting STING while working synergistically with monoclonal antibodies, are also discussed. A deep understanding of the tumor immune landscape will help find new and improved tumor immunotherapy for HNSCC.

In this paper a novel hybrid piezoelectric and electromagnetic energy harvester for civil engineering low-frequency sloshing environment is reported. The architecture, fabrication and characterization of the harvester are discussed. The hybrid energy harvester is composed of a permanent magnet, copper coil, and PVDF(polyvinylidene difluoride) piezoelectric film, and the upper U-tube device containing a cylindrical fluid barrier is connected to the foundation support plate by a hinge and spring. The two primary means of energy collection were through the vortex street, which alternately impacted the PVDF piezoelectric film through fluid shedding, and the electromotive force (EMF) induced by changes in the magnetic field position in the conducting coil. Experimentally, the maximum output power of the piezoelectric transformer of the hybrid energy harvester was 2.47 μW (circuit load 270 kΩ; liquid level height 80 mm); and the maximum output power of the electromagnetic generator was 2.72 μW (circuit load 470 kΩ; liquid level height 60 mm). The low-frequency sloshing energy collected by this energy harvester can drive microsensors for civil engineering monitoring.

Abstract Background The current study was carried out to provide a reference for monitory of aflatoxin B1 (AFB1), zearalenone (ZEN) and deoxynivalenol (DON) contamination in feed ingredients and complete feeds were collected from different Province in China from 2013 to 2015. Methods A total of 443 feed ingredients, including 220 corn, 24 wheat, 24 domestic distillers dried grains with soluble (DDGS), 55 bran, 20 wheat shorts and red dog, 37 imported DDGS, 34 corn germ meal and 29 soybean meal as well as 127 complete feeds including 25 pig complete feed (powder), 90 pig complete feed (pellet), six duck complete feed and six cattle complete feed were randomly collected from different Province in China, respectively, by high-performance chromatography in combined with UV or fluorescence analysis. Results The incidence rates of AFB1, ZEN and DON contamination of feed ingredients and complete feeds were 80.8, 92.3 and 93.9 %, respectively. The percentage of positive samples for DON ranged from 66.7 to 100 %. Domestic DDGS and imported DDGS presented the most serious contamination AFB1, ZEN and DON contamination levels of feeds ranged from 61.5 to 100 %, indicated that serious contamination over the studied 3-year period. Conclusion The current data provide clear evidence that AFB1, ZEN and DON contamination of feed ingredients and complete feeds in different Province in China is serious and differs over past 3-year. The use of corn, domestic DDGS, imported DDGS and corn germ meal, which may be contaminated with these three mycotoxins, as animal feed may triggered a health risk for animal. Feeds are most contaminated with DON followed by ZEN and AFB1. Mycotoxins contamination in feed ingredients and complete feeds should be monitored routinely in China.

Medium-chain fatty acid glycerides have been shown to provide energy for rapid oxidation in the body. The study was conducted to investigate the effects of dietary supplementation with medium-chain fatty acid glyceride on the growth performance and intestinal health of weaned piglets fed with a low-protein diet. Nighty healthy weaned piglets were randomly divided into five treatments: NP (Normal protein treatment, normal-protein diet no antibiotics included); NC (Negative control, low-protein diet no antibiotics included); PC (Positive control, low-protein diet +75 mg/kg quinocetone, 20 mg/kg virginiamycin and 50 mg/kg aureomycin); MCT (tricaprylin + tricaprin treatment, low-protein diet + tricaprylin + tricaprin); GML (glycerol monolaurate treatment, low-protein diet + glycerol monolaurate). The results showed that the average daily feed intake (ADFI) of the MCT treatment was significantly higher than that of the NP, NC treatments (p < 0.05). In the jejunum, the villus height of the GML treatment was significantly lower than that of the PC treatment (p < 0.05), and the number of goblet cells in the GML treatment was higher than that in the NC treatment (p < 0.05). Compared with the NC treatment, the MCT treatment significantly increased the level of claudin-1, Zonula occludens-1(ZO-1), while the GML treatment significantly increased the level of claudin-1, occludin, ZO-1 (p < 0.05). In the ileum, the level of ZO-1 in the GML treatment was significantly higher than that in the NP, NC, PC treatments (p < 0.05). Compared with the NC treatment, the GML treatment significantly increased the level of Secretory immunoglobulin A (SIgA) in the ileum and serum, while the MCT treatment significantly increased the level of SIgA and decreased the level of interleukin-6 (IL-6) in the ileum (p < 0.05). These results showed that the addition of medium-chain fatty acid glycerides to a low-protein diet could improve the growth performance and intestinal functional barrier of weaned piglets and also improve the immune function of weaned piglets.

The present experiment assessed the inflammatory responses, hormone secretion, and gut microbiota of weanling piglets administered baicalin-copper complex (BCU) or deoxynivalenol (DON) supplementation diets. Twenty-eight piglets were randomly assigned to four groups: control diet (Con group), a 4 mg DON/kg diet (DON group), a 5 g BCU/kg diet (BCU group), a 5 g BCU + 4 mg DON/kg diet (DBCU group). After 14 days, the results showed that dietary BCU supplementation remarkably increased the relative abundance of Clostrium bornimense and decreased the relative abundance of Lactobacillus in the DBCU group (p < 0.05). BCU decreased the serum concentration of IgG, IL-2, IFN-γ, and IgA in DON treated piglets (p < 0.05), and promoted the serum concentration of IL-1β, IgG, IL-2, IFN-γ, IgA, IL-6, IgM, and TNFα in normal piglets (p < 0.05). BCU increased the concentrations of serum IGF1, insulin, NPY, GLP-1, and GH, and decreased the concentrations of serum somatostatin in no DON treated piglets (p < 0.05). Dietary BCU supplementation significantly promoted the secretion of somatostatin, and inhibited the secretion of leptin in piglets challenged with DON (p < 0.05). BCU regulated the expression of food intake-related genes in the hypothalamus and pituitary of piglets. Collectively, dietary BCU supplementation alleviated inflammatory responses and regulated the secretion of appetite-regulating hormones and growth-axis hormones in DON challenged piglets, which was closely linked to changes of intestinal microbes.

Trillions of bacteria reside in the human gut and they metabolize dietary substances to obtain nutrients and energy while producing metabolites. Therefore, different dietary components could affect human health in various ways through microbial metabolism. Many such metabolites have been shown to affect human physiological activities, including short-chain fatty acids metabolized from carbohydrates; indole, kynurenic acid and para-cresol, metabolized from amino acids; conjugated linoleic acid and linoleic acid, metabolized from lipids. Here, we review the features of these metabolites and summarize the possible molecular mechanisms of their metabolisms by gut microbiota. We discuss the potential roles of these metabolites in health and diseases, and the interactions between host metabolism and the gut microbiota. We also show some of the major dietary patterns around the world and hope this review can provide insights into our eating habits and improve consumers’ health conditions.

Ribosomal proteins (RPs) play important roles in modulating the MDM2-p53 pathway. However, less is known about the upstream regulators of the RPs. Here, we identify SPIN1 (Spindlin 1) as a novel binding partner of human RPL5/uL18 that is important for this pathway. SPIN1 ablation activates p53, suppresses cell growth, reduces clonogenic ability, and induces apoptosis of human cancer cells. Mechanistically, SPIN1 sequesters uL18 in the nucleolus, preventing it from interacting with MDM2, and thereby alleviating uL18-mediated inhibition of MDM2 ubiquitin ligase activity toward p53. SPIN1 deficiency increases ribosome-free uL18 and uL5 (human RPL11), which are required for SPIN1 depletion-induced p53 activation. Analysis of cancer genomic databases suggests that SPIN1 is highly expressed in several human cancers, and its overexpression is positively correlated with poor prognosis in cancer patients. Altogether, our findings reveal that the oncogenic property of SPIN1 may be attributed to its negative regulation of uL18, leading to p53 inactivation.

This study was conducted to determine the positive effects of dietary supplementation with l-arginine (Arg) on piglets fed a deoxynivalenol (DON)-contaminated diet. A total of eighteen, 28-day-old healthy weanling pigs were randomly assigned into one of three groups: uncontaminated basal diet (control group), 6 mg/kg DON-contaminated diet (DON group) and 6 mg/kg DON + 1% l-arginine (DON + ARG group). After 21 days of Arg supplementation, piglets in the DON and DON + ARG groups were challenged by feeding 6 mg/kg DON-contaminated diet for seven days. The results showed that DON resulted in damage to piglets. However, clinical parameters, including jejunal morphology, amino acid concentrations in the serum, jejunum and ileum, were improved by Arg (p < 0.05). Furthermore, the mRNA levels for sodium-glucose transporter-1 (SGLT-1), glucose transporter type-2 (GLUT-2) and y+l-type amino acid transporter-1 (y+LAT-1) were downregulated in the DON group, but the values were increased in the DON + ARG group (p < 0.05). Collectively, these results indicate that dietary supplementation with Arg exerts a protective role in pigs fed DON-contaminated diets.

This paper builds the equivalent circuit model for a single cell of zinc nickel single flow battery (ZNB) with 300 Ah. According to the experimental data of the single cell under 100 A pulse discharge conditions, the model parameters can be obtained by parameter identification, and the analytical expressions for each model parameter can be obtained by using the method of high degree polynomial fitting and exponential function fitting, then the mathematical model of the stack voltage can be built. The relative error of the simulation results for stack voltage is controlled within 3.2% by experimental comparison, which verifies the accuracy of the model and model parameters. The parameter formulas obtained by fitting method can effectively solve calculation problem of the battery parameters. And under 100 A constant-current discharge condition, the stack voltage of the battery is dropping relatively flat over about 110 minutes after loading current, and dropping dramatically within about 50 minutes at the end of discharge due to the increasing polarization.

SCP1 as a nuclear transcriptional regulator acts globally to silence neuronal genes and to affect the dephosphorylation of RNA Pol ll. However, we report the first finding and description of SCP1 as a plasma membrane-localized protein in various cancer cells using EGFP- or other epitope-fused SCP1. Membrane-located SCP1 dephosphorylates AKT at serine 473, leading to the abolishment of serine 473 phosphorylation that results in suppressed angiogenesis and a decreased risk of tumorigenesis. Consistently, we observed increased AKT phosphorylation and angiogenesis followed by enhanced tumorigenesis in Ctdsp1 (which encodes SCP1) gene - knockout mice. Importantly, we discovered that the membrane localization of SCP1 is crucial for impeding angiogenesis and tumor growth, and this localization depends on palmitoylation of a conserved cysteine motif within its NH2 terminus. Thus, our study discovers a novel mechanism underlying SCP1 shuttling between the plasma membrane and nucleus, which constitutes a unique pathway in transducing AKT signaling that is closely linked to angiogenesis and tumorigenesis.

Based on the working principle of the zinc-nickel single flow batteries (ZNBs), this paper builds the electrochemical model and mechanical model, analyzes the effect of electrolyte flux on the battery performance and obtains a single cell with a 216 Ah charge-discharge capacity as an example, and thereafter conducts a simulation to obtain several results under the condition of constant current charge and discharge. The simulation results are well matched in comparison with the experimental results. An optimal flux exists during the charge and discharge, which indicates that the model can well simulate the charge and discharge characteristics of the ZNBs under the condition of constant current.

Increasing evidence highlights the important roles of microRNAs in mediating p53’s tumor suppression functions. Here, we report miR-139-5p as another new p53 microRNA target. p53 induced the transcription of miR-139-5p, which in turn suppressed the protein levels of phosphodiesterase 4D (PDE4D), an oncogenic protein involved in multiple tumor promoting processes. Knockdown of p53 reversed these effects. Also, overexpression of miR-139-5p decreased PDE4D levels and increased cellular cAMP levels, leading to BIM-mediated cell growth arrest. Furthermore, our analysis of human colorectal tumor specimens revealed significant inverse correlation between the expression of miR-139-5p and that of PDE4D. Finally, overexpression of miR-139-5p suppressed the growth of xenograft tumors, accompanied by decrease in PDE4D and increase in BIM. These results demonstrate that p53 inactivates oncogenic PDE4D by inducing the expression of miR-139-5p.

Cancer develops and progresses often by inactivating p53. Here, we unveil nerve growth factor receptor (NGFR, p75NTR or CD271) as a novel p53 inactivator. p53 activates NGFR transcription, whereas NGFR inactivates p53 by promoting its MDM2-mediated ubiquitin-dependent proteolysis and by directly binding to its central DNA binding domain and preventing its DNA-binding activity. Inversely, NGFR ablation activates p53, consequently inducing apoptosis, attenuating survival, and reducing clonogenic capability of cancer cells, as well as sensitizing human cancer cells to chemotherapeutic agents that induce p53 and suppressing mouse xenograft tumor growth. NGFR is highly expressed in human glioblastomas, and its gene is often amplified in breast cancers with wild type p53. Altogether, our results demonstrate that cancers hijack NGFR as an oncogenic inhibitor of p53.

In this study of zinc nickel single-flow batteries (ZNB), the ion concentration of the convection area and the electrode surface of the battery runner were investigated first. Then, the relationships between the electrode over-potential (or equilibrium potential) and the charge time were studied. This was based on the electrochemical reaction rate equation and the equilibrium potential equation, from which a mathematical model of the stack voltage (as affected by the internal parameters of the battery) was obtained.
