Liver fibrosis, and potentially cirrhosis, induced by copper accumulation are critical factors in the pathogenesis of WD. CircRNAs exhibit high stability and play crucial roles in numerous biological processes.Methods: RNA-seq technology was employed to conduct transcriptome sequencing of the liver from 12 homozygous (TX) mice in the model group (NL group) and 12 wild-type (WT) mice in the control group (N group). Differentially expressed circular RNAs (DE-circRNAs) were identified, and following GO and KEGG analysis, a competitive endogenous RNA (ceRNA) regulatory network was constructed. The identified DE-circRNAs were then randomly validated using RT-qPCR.Results: Utilizing RNA sequencing (RNA-seq), the study identified 54 DE-circRNAs in TX-j mice with WD-induced liver fibrosis model, among which 19 were up-regulated and 35 were down-regulated. GO analysis revealed multiple biological processes, including single-organism process, cellular process, and metabolic process. Further pathway identification using KEGG implicated several pathways, including the HIF-1, PI3K-Akt, AMPK, FoxO, signaling pathway regulating pluripotency of stem cells, phospholipase D, mTOR, Ras, cGMP-PKG, and MAPK signaling pathway, among others. A ceRNA regulatory network was constructed with 20 circRNAs, 7 miRNAs, and 75 mRNAs as crucial core components. Additionally, RT-qPCR validation was performed on randomly selected DE-circRNAs, yielding consistent results (P< 0.05).Conclusion: The findings provide a significant molecular biology foundation for understanding the pathogenesis of liver fibrosis in WD and offer new insights for exploring potential diagnostic and therapeutic targets.Keywords: circRNA, circRNA-miRNA-mRNA, ceRNA, Wilson’s disease, liver fibrosis

Abstract

The upgrade of the CMS experiment for the high luminosity operation of the LHC comprises the replacement of the current endcap calorimeter by a high granularity sampling calorimeter (HGCAL). The electromagnetic section of the HGCAL is based on silicon sensors interspersed between lead and copper (or copper tungsten) absorbers. The hadronic section uses layers of stainless steel as an absorbing medium and silicon sensors as an active medium in the regions of high radiation exposure, and scintillator tiles directly readout by silicon photomultipliers in the remaining regions. As part of the development of the detector and its readout electronic components, a section of a silicon-based HGCAL prototype detector along with a section of the CALICE AHCAL prototype was exposed to muons, electrons and charged pions in beam test experiments at the H2 beamline at the CERN SPS in October 2018. The AHCAL uses the same technology as foreseen for the HGCAL but with much finer longitudinal segmentation. The performance of the calorimeters in terms of energy response and resolution, longitudinal and transverse shower profiles is studied using negatively charged pions, and is compared to GEANT4 predictions. Substantial room for improvement exists. Environmental factors, including urbanization level and unemployment rate, wielded a pronounced influence on the operational efficiency of China’s employee medical insurance. Conversely, the fiscal revenue and expenditure ratio hindered the enhancement of employee medical insurance efficiency.Conclusion: This study held valuable insights for enhancing the operational efficiency of China’s urban employee basic medical insurance. To effectively improve insurance efficiency, the following recommendations are put forth: firstly, relevant departments should escalate resource investments and optimize resource utilization based on rational allocation; secondly, enhance legislation and regulations, fortify fund oversight, and ensure equitable and judicious utilization of the medical insurance fund; finally, leveraging modern high-tech advancements can comprehensively elevate the operational efficiency of the employee medical insurance fund. This dataset contained all hospital’s electronic medical records from January 1, 2015 to December 31, 2018 including both public and private hospitals. The target population was identified based on the principal diagnosis of appendicitis (ICD-10: K35). To examine the impact of various factors on hospitalization costs, we conducted a multivariate linear regression analysis. Furthermore, we employed the Shapley value decomposition method to gain a more comprehensive understanding of the factors that influenced hospitalization costs and their respective levels of importance.Results: Our study comprised 317,200 cases. During the period from 2015 to 2018, the average hospitalization expenses for patients with acute appendicitis were estimated at approximately 7014 RMB (1061 USD), which accounts for a considerable 12% of China’s per capita GDP. The results of this study demonstrate a significant correlation between various factors, such as the patient’s age, gender, marital status, occupation, payment method, number of complications, treatment method, hospital tier, and ownership, and the total hospitalization costs and subcomponents of hospitalization costs. Notably, the treatment method employed had the most substantial impact on hospitalization costs.Conclusion: To the best of knowledge, this is one of the first studies to investigate the hospitalization costs of acute appendicitis incorporating both patient-level and hospital-level covariates, using a large sample size. To reduce the costs associated with acute appendicitis in China, it is recommended to consider suitable treatment options and explore the option of receiving medical care at lower-tier and privately-owned healthcare facilities.Keywords: appendicitis, hospitalization costs, influencing factors, multivariate regression

This study included 3509 patients (2567 females and 942 males) with normal fasting plasma glucose (FPG) and no MS. Multivariate logistic regression was used to analyze the correlation between FPG and MS ROC was used to analyze the effective cutoff value of FPG for the incidence of MS.Results: After 3-year follow-up, 675 patients had MS (567 females and 108 males). MS incidence in the total population was 19.24%, 11.46% in males, and 22.09% in females, and it increased with FPG. After multivariate logistic regression analysis, the risk of MS corresponding to FPG in females and males was OR=4.607,95% CI (3.477– 6.105) and OR=2.944, 95% CI (1.785– 4.855), respectively. ROC results demonstrated that FPG could predict the onset of MS (AUC: 0.720 in males and 0.666 in females).Conclusion: Increased FPG correlated with the incidence of metabolic syndrome. Subjects with FPG in the normal range still had a high incidence of MS. The population cutoff value for predicting effective FPG for metabolic syndrome was 5.545 mmol/L in men and 5.605 mmol/L in women. Epidemiological investigations are needed to determine whether a lower FPG cutoff value is required to diagnose MS. FPG not only diagnoses diabetes but also serves as a cost-effective and convenient screening method for developing of MS in the general Chinese population.Keywords: metabolic syndrome, incidence, fasting plasma glucose, cutoff value

In total, 2935 subjects in the normal BP range and without MS (795 males, 2140 females) aged 40– 80 years were included in this study. Follow-up subjects were classified by baseline BP and grouped by age. After the 3-year follow-up, the MS incidence and valid BP cutoff value for predicting MS were calculated for subjects within the normal BP range.Results: After the 3-year follow-up study, the crude and standardized MS incidences in the cohort were 19.83% and 16.89% for follow-up subjects, respectively, with incidences of 10.94% and 10.50% for males and 23.13% and 20.66% for females. The incidence of MS in normotensive subjects (15.53%) was lower than that in subjects with high-normal BP (29.08%). After adjusting for age, sex, BMI, smoking, and drinking, the risk of having MS in the high-normal BP group was 1.823-fold [HR 1.823 95% CI (1.538– 2.162)] higher than that in the normotensive group. The ROC curve showed that the BP cutoff values were more than 117/74 mmHg in males and 112/74 mmHg in females.Conclusion: When BP was within the normal range, the incidence of MS in participants was very high. The MS incidence was higher among high-normal BP subjects than among normotensive subjects, as was the risk of having MS. The valid BP cutoff value for predicting MS in the population was 117/74 mmHg in males and 112/74 mmHg in females. Abstract

Molybdenum is used as plasma-facing material in tokamaks and as material for plasma optical diagnostics mirrors. Harsh conditions of neutron irradiation, exposure to hydrogen isotopes and helium ions, and high operating temperatures result in degradation of the molybdenum surface and ultimately limit their lifetime in a fusion power plant. In the current paper, intake and subsequent thermal release of deuterium from self-irradiated by high energy (1 MeV) ions molybdenum as a function of irradiation dose are investigated. Several characteristic temperature regions where deuterium release takes place are identified and attributed to trapping of deuterium in intrinsic and radiation-induced microstructure defects. This attribution is further validated by molecular dynamics modeling, which confirms that increase and saturation of vacancy concentration found in simulations follows increase and saturation of experimentally determined deuterium content. Deuterium inventory and vacancy content saturate at a damage level of around 0.2 dpa (displacement per atom), similar to recent modeling and experimental studies of iron and tungsten. Reflectivity measurements of irradiated molybdenum show that it is only slightly affected by damage up to 1 dpa. [ABSTRACT FROM AUTHOR]

Abstract

Ion beam damage combined with nanoscale focused ion beam direct milling was used to create manufacturable SNS type Josephson junctions in 100 nm thick MgB$_{2}$ with T$_{C}$ of 38 K. The junctions show non-hysteretic current - voltage characteristics between 36 and 4.2 K. Experimental evidence for the dc and ac Josephson effects in MgB$_{2}$ metal masked ion damage junctions are presented. Abstract

Wintertime deepwater renewal, which is important for heat–oxygen–nutrient exchange in lakes, is traditionally considered to be mainly driven by 1D vertical convective cooling. However, differential cooling between shallow and deep waters can produce density currents that flow into deep layers. In order to determine the role that these two cooling processes play in deepwater renewal, field measurements and 3D numerical modeling were combined to investigate heat content dynamics in Lake Geneva's large basin, the Grand Lac (maximum depth 309 m), during an exceptionally cold air spell in early 2012 where complete overturning had been reported. In a novel approach, the heat budget of the lake was decomposed, which allowed the identification and quantification of the heat budget components. The heat budget decomposition revealed that vertical convective cooling only penetrated to 200 m and that lateral advection was not only caused by density currents being discharged from the shallow littoral zone of the Grand Lac, but also from the Lake's shallow Petit Lac basin (maximum depth 75 m); the latter was found to be the main driver of heat content decrease in the deep layers of the Grand Lac below ∼200‐m depth. These findings provide unique insight into heat exchange processes that cannot be obtained from field data or numerical simulations alone. Heat budget decomposition proved to be a powerful, universally applicable tool for quantifying the contribution of alternative deepwater renewal processes. This is important, since deepwater renewal by convective cooling is weakening due to persistent global warming. Plain Language Summary: Winter cooling is an important process for heat–oxygen–nutrient exchange in lakes. Traditionally considered to be mainly driven by 1D vertical convective cooling, complete overturning occurs when temperatures become homogeneous over the full water depth. In most deep lakes, this only occurs occasionally. Complete overturning in 309‐m deep Lake Geneva was reported for winter 2012. Field measurements taken at the deepest part of the lake, however, showed that the lake's temperature profile during that winter was not uniform. Instead, it had a cooler, deep bottom layer. To understand this temperature profile, we used 3D numerical modeling to decompose the lake's heat budget. This novel approach made it possible to investigate and quantify the contribution of: (a) vertical top‐down convective cooling, and (b) lateral advection of cold water from shallower regions to the lake's deep layers. The results revealed that convective cooling did not reach below ∼200‐m depth. Instead, lateral cold‐water advection from the lake's shallow side basin and nearshore regions were responsible for the cooling and deepwater renewal below that depth. As climate change induced‐warming continues to weaken convective cooling, it will become increasingly important to understand and consider lateral advection as an alternative deepwater renewal process in large, deep lakes. The correlation between MRPL13 expression and clinicopathological parameters was analyzed. Cox regression multivariate analysis was used to explore the factors affecting the prognosis of BC patients. The UALCAN database was used to analyze the expression level of MRPL13 in BC and its relationship with clinical pathological factors. The GSEA method was used to predict the possible regulatory pathways of MRPL13. Immune responses of MRPL13 expression were analyzed using TISIDB and CIBERSORT. Additionally, GEPIA, K-M survival analysis and data from the HPA were used to validate the outcomes.Results: The expression of MRPL13 in BC tissues was significantly higher than normal counterparts, patients with low MRPL13 expression had a better survival prognosis, also indicated an independent prognostic factor. GSEA analysis showed that the regulation of cell migration, positive regulation of endothelial cell migration, and Notch signaling pathway were enriched in tissues with low expression of MRPL13. Additionally, depleting MRPL13 expression inhibited invasion in MCF-10A and MCF-7 cells. Furthermore, PCR showed that MRPL13 affected VEGFA and MMP gene expression. CIBERSORT analysis revealed that the amount of NK cells decreased when MRPL13 expression was high.Conclusion: The expression of MRPL13 mRNA is upregulated in BC tissues, and the expression level of MRPL13 is significantly related to the clinicopathological factors of patients. High MRPL13 expression is a poor prognostic factor for BC, and it can be used as a molecular marker for prognosis judgment and as a potential therapeutic target.Keywords: cancer genome map, breast cancer, MRPL13, clinical significance
