1. Sublinear-time Sampling of Spanning Trees in the Congested Clique

Abstract

We present the first sublinear round algorithm for approximately sampling uniform spanning trees in the CongestedClique model of distributed computing. In particular, our algorithm requires $\~O(n^{0.658})$ rounds for sampling a spanning tree from a distribution within total variation distance $1/n^c$, for arbitrary constant $c > 0$, from the uniform distribution. More precisely, our algorithm requires $\~O(n^{1/2 + \alpha})$ rounds, where $O(n^\alpha)$ is the running time of matrix multiplication in the CongestedClique model, currently at $\alpha = 1 - 2/\omega = 0.158$, where $\omega$ is the sequential matrix multiplication time exponent. In addition, we show how to take somewhat shorter random walks even more efficiently in the CongestedClique model. Specifically, we show how to construct length-$\tau$ walks, for $\tau = \Omega(n/\log n)$, in $O\left(\frac{\tau}{n} \log \tau \log n\right)$ rounds and for $\tau = O(n/\log n)$ in $O(\log \tau)$ rounds. This implies an $O(\log^3 n)$-round algorithm in the CongestedClique model for sampling spanning trees for Erd\H{o}s-R\'enyi graphs and regular expander graphs due to the $O(n \log n)$ bound on their cover time. This also implies that polylogarithmic-length walks, which are useful for page rank estimation, can be constructed in $O(\log \log n)$ rounds in the CongestedClique model. These results are obtained by adding a load balancing component to the random walk algorithm of Bahmani, Chakrabarti and Xin (SIGMOD 2011) that uses the ``doubling'' technique.

9. Towards singular optimality in the presence of local initial knowledge

Abstract

The Knowledge Till rho CONGEST model is a variant of the classical CONGEST model of distributed computing in which each vertex v has initial knowledge of the radius-rho ball centered at v. The most commonly studied variants of the CONGEST model are KT0 CONGEST in which nodes initially know nothing about their neighbors and KT1 CONGEST in which nodes initially know the IDs of all their neighbors. It has been shown that having access to neighbors' IDs (as in the KT1 CONGEST model) can substantially reduce the message complexity of algorithms for fundamental problems such as BROADCAST and MST. For example, King, Kutten, and Thorup (PODC 2015) show how to construct an MST using just Otilde(n) messages in the KT1 CONGEST model, whereas there is an Omega(m) message lower bound for MST in the KT0 CONGEST model. Building on this result, Gmyr and Pandurangen (DISC 2018) present a family of distributed randomized algorithms for various global problems that exhibit a trade-off between message and round complexity. These algorithms are based on constructing a sparse, spanning subgraph called a danner. Specifically, given a graph G and any delta in [0,1], their algorithm constructs (with high probability) a danner that has diameter Otilde(D + n^{1-delta}) and Otilde(min{m,n^{1+delta}}) edges in Otilde(n^{1-delta}) rounds while using Otilde(min{m,n^{1+\delta}}) messages, where n, m, and D are the number of nodes, edges, and the diameter of G, respectively. In the main result of this paper, we show that if we assume the KT2 CONGEST model, it is possible to substantially improve the time-message trade-off in constructing a danner. Specifically, we show in the KT2 CONGEST model, how to construct a danner that has diameter Otilde(D + n^{1-2delta}) and Otilde(min{m,n^{1+delta}}) edges in Otilde(n^{1-2delta}) rounds while using Otilde(min{m,n^{1+\delta}}) messages for any delta in [0,1/2].

Abstract

As hospitals move towards automating and integrating their computing systems, more fine-grained hospital operations data are becoming available. These data include hospital architectural drawings, logs of interactions between patients and healthcare professionals, prescription data, procedures data, and data on patient admission, discharge, and transfers. This has opened up many fascinating avenues for healthcare-related prediction tasks for improving patient care. However, in order to leverage off-the-shelf machine learning software for these tasks, one needs to learn structured representations of entities involved from heterogeneous, dynamic data streams. Here, we propose DECENT, an auto-encoding heterogeneous co-evolving dynamic neural network, for learning heterogeneous dynamic embeddings of patients, doctors, rooms, and medications from diverse data streams. These embeddings capture similarities among doctors, rooms, patients, and medications based on static attributes and dynamic interactions. DECENT enables several applications in healthcare prediction, such as predicting mortality risk and case severity of patients, adverse events (e.g., transfer back into an intensive care unit), and future healthcare-associated infections. The results of using the learned patient embeddings in predictive modeling show that DECENT has a gain of up to 48.1% on the mortality risk prediction task, 12.6% on the case severity prediction task, 6.4% on the medical intensive care unit transfer task, and 3.8% on the Clostridioides difficile (C.diff) Infection (CDI) prediction task over the state-of-the-art baselines. Abstract

Fast distributed algorithms that output a feasible solution for constraint satisfaction problems, such as maximal independent sets, have been heavily studied. There has been much less research on distributed sampling problems, where one wants to sample from a distribution over all feasible solutions (e.g., uniformly sampling a feasible solution). Recent work (Feng, Sun, Yin PODC 2017; Fischer and Ghaffari DISC 2018; Feng, Hayes, and Yin arXiv 2018) has shown that for some constraint satisfaction problems there are distributed Markov chains that mix in $O(\log n)$ rounds in the classical LOCAL model of distributed computation. However, these methods return samples from a distribution close to the desired distribution, but with some small amount of error. In this paper, we focus on the problem of exact distributed sampling. Our main contribution is to show that these distributed Markov chains in tandem with techniques from the sequential setting, namely coupling from the past and bounding chains, can be used to design $O(\log n)$-round LOCAL model exact sampling algorithms for a class of weighted local constraint satisfaction problems. This general result leads to $O(\log n)$-round exact sampling algorithms that use small messages (i.e., run in the CONGEST model) and polynomial-time local computation for some important special cases, such as sampling weighted independent sets (aka the hardcore model) and weighted dominating sets.

Abstract

Delayed hemolytic transfusion reaction (DHTR) poses a significant challenge in patients receiving blood transfusions. This case report highlights the complexities of managing DHTR in a newly diagnosed chronic myelomonocytic leukemia (CMML) patient with clinically significant JKa and little c antibodies during induction chemotherapy. A 46-year-old woman with CMML-2 who presented for induction chemotherapy was found to have hemolytic anemia. Due to presence of JKa and little c antibodies, she required intensive monitoring and supportive care measures. The coexistence of JKa and little c antibodies complicates transfusion management and chemotherapy tolerance in CMML patients.

In myelodysplastic syndrome (MDS) and acute myeloid leukemia (AML) with TP53 aberrations, dissecting the interaction amongst patient, disease and treatment factors are important for therapeutic decisions and prognostication. This retrospective analysis included patients with newly diagnosed MDS (>5% blasts) and AML with TP53 mutation(s) treated at MD Anderson Cancer Center. We factored patient age, TP53 aberration burden, therapy intensity and use of venetoclax in the AML subgroup, and allogeneic hematopoietic stem cell transplantation (HSCT) to interrogate outcomes. Abstract

Introduction Leprosy, a neglected tropical disease, is caused by Mycobacterium leprae or less often by M. lepromatosis. (1) The disease is a chronic, moderately infectious condition affecting mostly skin, peripheral [...], The World Health Organization (WHO) alms to reduce new leprosy cases by 70% by 2030, necessitating advancements In leprosy diagnostics. Here we discuss the development of two WHO's target product profiles for such diagnostics. These profiles define criteria for product use, design, performance, configuration and distribution, with a focus on accessibility and affordability. The first target product profile outlines requirements for tests to confirm diagnosis of leprosy in individuals with clinical signs and symptoms, to guide multidrug treatment initiation. The second target product profile outlines requirements for tests to detect Mycobacterium leprae or M. lepromatosis infection among asymptomatic contacts of leprosy patients, aiding prophylactic interventions and prevention. Statistical modelling was used to assess sensitivity and specificity requirements for these diagnostic tests. The paper highlights challenges in achieving high specificity, given the varying endemicity of M. leprae, and identifying target analytes with robust performance across leprosy phenotypes. We conclude that diagnostics with appropriate product design and performance characteristics are crucial for early detection and preventive intervention, advocating for the transition from leprosy management to prevention. Se utilizaron modelos estadisticos para evaluar los requisitos de sensibilidad y especificidad de estas pruebas diagnosticas. El articulo destaca las dificultades para lograr una alta especificidad, dada la diferente endemicidad de M. leprae, y para identificar analitos diana con un rendimiento solido en todos los fenotipos de lepra. Concluimos que los diagnosticos con un diseno de producto y unas caracteristicas de rendimiento adecuados son fundamentales para la deteccion precoz y la intervencion preventiva, lo que favorece la transicion del manejo de la lepra a la prevencion.

Abstract

Leukemic masses are a known complication in patients with hematologic malignancies. Here we present a case regarding a patient with recently diagnosed B-acute lymphoblastic leukemia (B-ALL) who presented with multiple sites of extramedullary involvement including an anterior mediastinal mass. This mass persisted despite multiple rounds of multiagent cytotoxic therapy. In this report, we summarize the literature regarding mediastinal masses in the setting of B-ALL and illustrate that such masses in patients with leukemias may have surprising etiology, separate from the primary disease.

Abstract

Blastic plasmacytoid dendritic cell neoplasm (BPDCN) is a rare and clinically aggressive hematologic malignancy with limited treatment options. Currently, standard treatment strategies include clinical trials; chemotherapy regimens such as hyperfractionated cyclophosphamide, vincristine, doxorubicin, and dexamethasone (HCVAD); and tagraxofusp-erzs (TAG, previously SL-401) which is the first-in-class targeted therapy against CD123. TAG received Food and Drug Administration approval for frontline BPDCN treatment in December 2018 and has increasingly become an alternative to chemotherapy, offering potentially more effective and less toxic options. However, despite promising results, there are still patients who may be resistant to TAG monotherapy and/or who respond but eventually relapse. Herein, we discuss an important patient case of BPDCN treated with TAG and review BPDCN treatment strategies.

20. Intrapleural Fibrinolysis with Urokinase versus Alteplase in Complicated Pleural Effusions and Empyema: A Prospective Randomized Controlled Trial

Abstract

Background Intrapleural fibrinolytic therapy (IPFT) has been used as an effective agent since 1949 for managing complicated pleural effusion and empyema. Several agents, such as streptokinase, urokinase (UK), and recombinant tissue plasminogen activator (rt-PA), have been found to be effective with variable effectiveness. However, a head-to-head controlled trial comparing the efficacy of the most frequently used agents, i.e., UK and rt-PA (alteplase) for managing complicated pleural effusion has rarely been reported. Methods A total of 50 patients were randomized in two intervention groups, i.e., UK and rt-PA. The dose of rt-PA was 10 mg, and that of UK was 1.0 lac units. UK was given thrice daily for 2 days, followed by clamping to allow the retainment of drugs in the pleural space for 2 hours. rt-PA was instilled into the pleural space twice daily for 2 days, and intercostal drainage was clamped for 1 hour. Results A total of 50 patients were enrolled into the study, of which 84% (n=42) were males and 16% (n=8) were females. Among them, 30 (60%) patients received UK, and 20 (40%) patients received alteplase as IPFT agents. The percentage of mean± standard deviation changes in pleural opacity was –33.0%±9.9% in the UK group and –41.0%±14.9% in the alteplase group, respectively (p=0.014). Pain was the most common adverse side effect, occurring in 60% (n=18) of the patients in the UK group and in 40% (n=8) of the patients in the alteplase group (p=0.24), while fever was the second most common side effect. Patients who reported early (within 6 weeks of onset of symptoms) showed a greater response than those who reported late for the intervention. Conclusion IPFT is a safe and effective option for managing complicated pleural effusion or empyema, and newer agents, such as alteplase, have greater efficacy and a similar adverse effect profile when compared with conventional agents, such as UK.

Presented our outcomes graphically by incorporating the parameters. The elasticity of the Maxwell fluid is directly proportional to temperature, resulting in a reduction in its velocity. The factors that affect fluid flow include viscous dissipation, Lewis number, Brownian motion, and the reversibility of temperature and velocity. Increasing the parameters of Brownian motion leads to significant movement of the nanofluid particles, which in turn increases their kinetic energy and enhances heat generation in the boundary layer. The Lorentz force, which hinders the movement of fluid, leads to a decrease in velocity profiles. This is determined by the magnetic parameter. The heat transfer rate, the velocity decay rate, and the occurrence of viscous dissipation are all occurring in close proximity to the sheet. Also, the model tabular validation presented and current results align well with previously published studies. Cancer treatment and the cooling process in industries, polymer processing, biotechnology and medicine, food industry, cosmetics, oil and gas, textiles, aerospace and automotive, and construction are just a few of the technical and biological uses for it. Industries may enhance material performance, process efficiency, and product quality in a variety of applications by using Maxwell fluid models.

In this article, we illustrate how the Coulomb field of a highly relativistic electron beam can be shaped into a broadband pulse suitable for driving ultrafast and strong-field physics. In contrast to a solid-state laser, the Coulomb field creates a pulse which can be intrinsically synchronized with an x-ray free electron laser (XFEL), can have a cutoff frequency which is broadly tunable from THz to EUV, and which acts on target systems as a "half-cycle" impulse. Abstract

The constrained-search formulation of Levy and Lieb provides a concrete mapping from N-representable densities to the space of N-particle wavefunctions and explicitly defines the universal functional of density functional theory. We numerically implement the Levy-Lieb procedure for a paradigmatic lattice system, the Hubbard dimer, using a modified variational quantum eigensolver approach. We demonstrate density variational minimization using the resulting hybrid quantum-classical scheme featuring real-time computation of the Levy-Lieb functional along the search trajectory. We further illustrate a fidelity based quantum kernel associated with the density to pure-state embedding implied by the Levy-Lieb procedure and employ the kernel for learning observable functionals of the density. Abstract

In this paper we present a deterministic $O(\log\log n)$-round algorithm for the 2-ruling set problem in the Massively Parallel Computation model with $\tilde{O}(n)$ memory; this algorithm also runs in $O(\log\log n)$ rounds in the Congested Clique model. This is exponentially faster than the fastest known deterministic 2-ruling set algorithm for these models, which is simply the $O(\log \Delta)$-round deterministic Maximal Independent Set algorithm due to Czumaj, Davies, and Parter (SPAA 2020). Our result is obtained by derandomizing the 2-ruling set algorithm of Kothapalli and Pemmaraju (FSTTCS 2012).

Abstract

Strong-field ionization of nanoscale clusters provides excellent opportunities to study the complex correlated electronic and nuclear dynamics of near-solid density plasmas. Yet, monitoring ultrafast, nanoscopic dynamics in real-time is challenging, which often complicates a direct comparison between theory and experiment. Here, near-infrared laser-induced plasma dynamics in ∼600  nm diameter helium droplets are studied by femtosecond time-resolved x-ray coherent diffractive imaging. An anisotropic, ∼20  nm wide surface region, defined as the range where the density lies between 10% and 90% of the core value, is established within ∼100  fs, in qualitative agreement with theoretical predictions. At longer timescales, however, the width of this region remains largely constant while the radius of the dense plasma core shrinks at average rates of ≈71  nm/ps along and ≈33  nm/ps perpendicular to the laser polarization. These dynamics are not captured by previous plasma expansion models. The observations are phenomenologically described within a numerical simulation; details of the underlying physics, however, remain to be explored.

Abstract

Abstract Blastic plasmacytoid dendritic cell neoplasm (BPDCN) predominantly occurs in adults ≥60 years old; 10–20% of cases are pediatric or adolescent/young adult (AYA) patients. Tagraxofusp (TAG, Elzonris®) is the only approved treatment for BPDCN; in the United States it is approved for patients aged ≥2 years. Data on treating pediatric and AYA BPDCN patients are limited. We present a case series of pediatric and AYA patients with BPDCN treated with TAG. Eight patients (five newly diagnosed; three relapsed/refractory [R/R]), aged 2–21 years, received 12 mcg/kg TAG. Seven patients were female; most had skin (n = 6) and/or bone marrow (n = 4) involvement. No new safety signals were identified. Grade 3 adverse events were headache (n = 1) and transaminitis (n = 2). Three patients with newly diagnosed BPDCN achieved complete response, one achieved partial response, and one had stable disease (SD). One patient with R/R BPDCN achieved a minor response; one had SD. Seven patients (88%) were bridged to stem cell transplant: 80% of newly diagnosed patients and 100% of R/R patients. Five patients remained alive at last follow‐up. These cases highlight the efficacy and safety of TAG in pediatric and AYA patients for whom there is no other approved BPDCN therapy.

Abstract

Aim: Lattice is a three-dimensional technical development of the two-dimensional grid technology, which forms high-dose “islands” amid a “sea” of lower dose. When compared to grid, lattice offers enhanced dose distribution and organs-at-risk (OAR) sparing, which may be useful for big or deep-seated cancers. In this study, we used a Halcyon 2.0 (6MV) linear accelerator to treat the palliative cases with lattice radiation using volumetric modulated arc treatment (VMAT). With new machines and techniques, it's becoming more difficult to adapt and reinvent traditional grid radiation. In our study, we used VMAT to deliver high-dose radiation to the tumor while sparing the surrounding areas and normal tissues. New methodologies and the same thing we showed in our study are bringing development to lattice planning. Materials and Methods: Four patients were treated with grid technique. All the structures of respective patients were manually contoured on Eclipse SomaVision for lattice planning, important parameters include the vertex volume diameter and center-to-center spacing. Spheres were contoured in the axial slice inside the gross tumor volume. VMAT was planned with a single isocenter and multiple arcs (full arcs 360°). Optimization was set giving high priorities to the target volume, ensuring that the tumor received the required dose and that the OARs were met. Results: VMAT lattice therapy as a strategy to improve initial responsiveness while reducing OAR toxicity. All the target spheres achieved >95% of the prescription dose and dose fall-off outside the target sphere was achieved due to the ring shape sphere constraints. Conclusions: Bulky tumors were treated with lattice radiation with a high dose per fraction in this study, with the vertices receiving the specified dose and the region between the vertices being spared. As it was a task for physicists to design a lattice radiation considering the vertices and periphery regions and to achieve the OARs according to the dose constraint protocol, the VMAT plan was well realized using optimization and computation.

Abstract

Blastic plasmacytoid dendritic cell neoplasm (BPDCN) is a rare hematologic malignancy with poor outcomes with conventional therapy. Nearly 100% of BPDCNs overexpress interleukin 3 receptor subunit alpha (CD123). Given that CD123 is differentially expressed on the surface of BPDCN cells, it has emerged as an attractive therapeutic target. UCART123 is an investigational product consisting of allogeneic T cells expressing an anti-CD123 chimeric antigen receptor (CAR), edited with TALEN® nucleases. In this study, we examine the antitumor activity of UCART123 in preclinical models of BPDCN. We report that UCART123 have selective antitumor activity against CD123-positive primary BPDCN samples (while sparing normal hematopoietic progenitor cells) in the in vitro cytotoxicity and T cell degranulation assays; supported by the increased secretion of IFNγ by UCART123 cells when cultured in the presence of BPDCN cells. UCART123 eradicate BPDCN and result in long-term disease-free survival in a subset of primary patient-derived BPDCN xenograft mouse models. One potential challenge of CD123 targeting therapies is the loss of CD123 antigen through diverse genetic mechanisms, an event observed in one of three BPDCN PDX studied. In summary, these results provide a preclinical proof-of-principle that allogeneic UCART123 cells have potent anti-BPDCN activity.

Abstract

Solid systems with strong correlations and interactions under light illumination have the potential for exhibiting interesting bulk photovoltaic behavior in the nonperturbative regime, which has remained largely unexplored in past theoretical studies. We investigate the bulk photovoltaic response of a perovskite manganite with strongly coupled electron-spin-lattice dynamics using real-time simulations performed with a tight-binding model. The transient changes in the band structure and the photoinduced phase transitions, emerging from spin and phonon dynamics, result in a nonlinear current versus intensity behavior beyond the perturbative limit. The current rises sharply across a photoinduced magnetic phase transition, which later saturates at higher light intensities due to excited phonon and spin modes. The predicted peak photoresponsivity is an order of magnitude higher than another known transition metal ferroelectric oxide BiFeO3. We disentangle phonon-and spin-assisted components to the ballistic photocurrent, showing that they are comparable in magnitude. Our results illustrate a promising alternative way for controlling and optimizing the bulk photovoltaic response through the photoinduced phase transitions in strongly correlated systems.

Abstract

Optical constants characterize the interaction of materials with light and are important properties in material design. Here we present a Python-based Corvus workflow for simulations of full spectrum optical constants from the UV-VIS to hard x-ray wavelengths based on the real-space Green's function code FEFF10 and structural data from the Materials Project (MP). The Corvus workflow manager and its associated tools provide an interface to FEFF10 and the MP database. The workflow parallelizes the FEFF computations of optical constants over all absorption edges for each material in the MP database specified by a unique MP-ID. The workflow tools determine the distribution of computational resources needed for that case. Similarly, the optical constants for selected sets of materials can be computed in a single-shot. To illustrate the approach, we present results for nearly all elemental solids in the periodic table, as well as a sample compound, and compared with experimental results. As in x-ray absorption spectra, these results are interpreted in terms of an atomic-like background and fine-structure contributions.

Abstract

The real-space Green's function code FEFF has been extensively developed and used for calculations of x-ray and related spectra, including x-ray absorption (XAS), x-ray emission (XES), inelastic x-ray scattering, and electron energy loss spectra (EELS). The code is particularly useful for the analysis and interpretation of the XAS fine-structure (EXAFS) and the near-edge structure (XANES) in materials throughout the periodic table. Nevertheless, many applications, such as non-equilibrium systems, and the analysis of ultra-fast pump-probe experiments, require extensions of the code including finite-temperature and auxiliary calculations of structure and vibrational properties. To enable these extensions, we have developed in tandem, a new version FEFF10, and new FEFF based workflows for the Corvus workflow manager, which allow users to easily augment the capabilities of FEFF10 via auxiliary codes. This coupling facilitates simplified input and automated calculations of spectra based on advanced theoretical techniques. The approach is illustrated with examples of high temperature behavior, vibrational properties, many-body excitations in XAS, super-heavy materials, and fits of calculated spectra to experiment., Comment: 9 pages, 9 figures. Abstract

Interlayer van der Waals (vdW) coupling is generic in two-dimensional materials such as graphene and transition metal dichalcogenides, which can induce very low-energy phonon modes. Using high-resolution inelastic hard x-ray scattering, we uncover the ultra-low energy phonon mode along the Cu-O bond direction in the high-$T_c$ cuprate (Bi,Pb)$_2$(Sr,La)$_2$CuO$_{6+\delta}$ (Bi2201). This mode is independent of temperature, while its intensity decreases with doping in accordance with an increasing c-axis lattice parameter. We compare the experimental results to first-principles density functional theory simulations and identify the observed mode as a van der Waals phonon, which arises from the shear motion of the adjacent Bi-O layers. This shows that Bi-based cuprate has similar vibrational properties as graphene and transition metal dichalcogenides, which can be exploited to engineer novel heterostructures.

Abstract

We present INQ, a new implementation of density functional theory (DFT) and time-dependent DFT (TDDFT) written from scratch to work on graphical processing units (GPUs). Besides GPU support, INQ makes use of modern code design features and takes advantage of newly available hardware. By designing the code around algorithms, rather than against specific implementations and numerical libraries, we aim to provide a concise and modular code. The result is a fairly complete DFT/TDDFT implementation in roughly 12,000 lines of open-source C++ code representing a modular platform for community-driven application development on emerging high-performance computing architectures for the simulation of materials., Comment: 25 pages, 10 figures
