Contains fulltext : 232881.pdf (Publisher’s version ) (Open Access), Clear cell meningioma represents an uncommon variant of meningioma that typically affects children and young adults. Although an enrichment of loss-of-function mutations in the SMARCE1 gene has been reported for this subtype, comprehensive molecular investigations are lacking. Here we describe a molecularly distinct subset of tumors (n = 31), initially identified through genome-wide DNA methylation screening among a cohort of 3093 meningiomas, of which most were diagnosed histologically as clear cell meningioma. This cohort was further supplemented by an additional 11 histologically diagnosed clear cell meningiomas for analysis (n = 42). Targeted DNA sequencing revealed SMARCE1 mutations in 33/34 analyzed samples, accompanied by a nuclear loss of expression determined via immunohistochemistry and a decreased SMARCE1 transcript expression in the tumor cells. Analysis of time to progression or recurrence of patients within the clear cell meningioma group (n = 14) in comparison to those with meningioma WHO grade 2 (n = 220) revealed a similar outcome and support the assignment of WHO grade 2 to these tumors. Our findings indicate the existence of a highly distinct epigenetic signature of clear cell meningiomas, separate from all other variants of meningiomas, with recurrent mutations in the SMARCE1 gene. This suggests that these tumors may arise from a different precursor cell population than the broad spectrum of the other meningioma subtypes.

Contains fulltext : 219243.pdf (Publisher’s version ) (Closed access), Desmoplastic small round cell tumors (DSRCTs) are highly aggressive sarcomas that most commonly occur intra-abdominally, and are defined by EWSR1-WT1 gene fusion. Intracranial DSRCTs are exceptionally rare with only seven previously reported fusion-positive cases. Herein, we evaluate the clinical, morphologic, immunohistochemical and molecular features of five additional examples. All patients were male (age range 6-25 years; median 11 years), with four tumors located supratentorially and one within the posterior fossa. The histologic features were highly variable including small cell, embryonal, clear cell, rhabdoid, anaplastic and glioma-like appearances. A prominent desmoplastic stroma was seen in only two cases. The mitotic index ranged from <1 to 12/10 HPF (median 5). While all tumors showed strong desmin positivity, epithelial markers such as EMA, CAM 5.2 and other keratins were strongly positive in only one, focally positive in two and negative in two cases. EWSR1-WT1 gene fusion was present in all cases, with accompanying mutations in the TERT promoter or STAG2 gene in individual cases. Given the significant histologic diversity, in the absence of genetic evaluation these cases could easily be misinterpreted as other entities. Desmin immunostaining is a useful initial screening method for consideration of a DSRCT diagnosis, prompting confirmatory molecular testing. Demonstrating the presence of an EWSR1-WT1 fusion provides a definitive diagnosis of DSRCT. Genome-wide methylation profiles of intracranial DSRCTs matched those of extracranial DSRCTs. Thus, despite the occasionally unusual histologic features and immunoprofile, intracranial DSRCTs likely represent a similar, if not the same, entity as their soft tissue counterpart based on the shared fusion and methylation profiles.

Abstract

Anaplastic meningiomas that resemble sarcomas often reveal clues to their meningothelial differentiation or develop in a plausible setting that confirms their meningothelial origin. Malignant mesenchymal neoplasms without obvious evidence of meningothelial differentiation or origin are more likely to be true primary or metastatic sarcomas. Because of their clinical and biological differences, it is important to distinguish anaplastic meningioma from a sarcoma. We present a 67-year-old woman with multiple meningiomas, who developed a high-grade spindle cell tumor 6 months after the resection of a World Health Organization grade I meningioma. It was not clear whether this tumor represented a malignant transformation of meningioma or a primary sarcoma. Malignant transformation of a meningioma is exceptional within this short period and a coexisting sarcoma and meningioma are equally uncommon. Even though these malignant neoplasms are rare in general, they appear to be more prevalent in patients with multiple meningiomas including those with neurofibromatosis type 2.

Abstract

Bu çalışmada, örtüaltında topraksız kültürle çilek (Fragaria×ananassa Duch.) yetiştiriciliğinde, değişik fide tipleri ile yetiştirme ortamlarının, bitkilerin fenolojik özellikleri ile erkencilik ve verim üzerine etkileri incelenmiştir. Denemede çeşit olarak ‘Camarosa’ çeşidi; fide tipi olarak, tüplü fide ile frigo fide; yetiştirme ortamı olarak ise torf, perlit, kokopit, volkanik tüf ve bunların karışımları denenmiştir. Araştırma sonucunda, çiçeklenme ve erkencilik bakımından tüplü fidenin; bitki başına düşen toplam verim bakımından ise frigo fidenin avantajlı olduğu belirlenmiştir. Nitekim frigo fidelerde ilk çiçeklenmeler ocak ayı başında, ilk derimler ise şubat ayında başlarken; tüplü fidelerde ilk çiçeklenmeler kasım ayı başında ve ilk derimler ise aralık ayında gerçekleşmiştir. Bitki başına düşen toplam verim değeri, deneme yıllarına göre değişmekle birlikte, frigo fidede 1057,61 g bitki-1 ile 1151,90 g bitki-1 ; tüplü fidede 745,42 g bitki-1 ile 739,11 g bitki-1 arasında değişim göstermiştir. Yetiştirme ortamları bakımından ise özellikle kokopit ve kokopit+volkanik tüf ortamları, gerek bitki gelişimi, gerek erkencilik ve gerekse verim bakımından denenen diğer ortamlardan daha olumlu sonuçlar vermiştir. Volkanik tüf ve perlit ortamlarında yetiştirilen bitkilerde ise erkencilik önemli oranda gecikirken, bitki başına düşen verim değeri de azalmıştır.

Abstract

In this study, Marsh Seedless grapefruit variety has been investigated; due to its recent increasing market demands. The followings have been chosen as rootstocks: Sour orange, which is used widespread in our region, and Carrizo and Troyer citrange which were turned out as hopeful in recent studies. By investigating the growing of trees on the rootstock varieties, yield, fruit quality, tree growing, the best rootstock X scion combination has been determined. The trees, which are eighteen years old in Batı Akdeniz Agricultural Research Institute, have been used as experiment materials. As a result of our findings, on the Carrizo citrange rootstocks have been determined as the best result with regard to yield and quality. As a result of our study, when we consider all of the factors in our regional conditions, for Marsh Seedless grapefruit the Carrizo citrange rootstock have given the best results., Bu araştırmada, önemli bir altıntop çeşidi olan Marsh Seedless çeşidi, anaç olarak da bölgemizde yaygın olarak kullanılan turunç anacı ve son yıllarda yapılan çalışmalarda ümitvar olarak görülen Troyer ve Carrizo sitranjı anaçları seçilmiştir. Bu anaçlar üzerindeki çeşitlere ait ağaçların gelişme durumları, verim ve meyve kalitesi incelenerek, en iyi anaç X kalem kombinasyonları saptanmıştır. Deneme materyali olarak, Batı Akdeniz Tarımsal Araştırmalar Enstitüsündeki 18 yaşlı Marsh Seedless ağaçları kullanılmıştır. Bulgularımız sonucunda, verim ve kalite açısından en iyi sonuçlar Carrizo sitranjı anacı üzerindeki ağaçlarda olup, bölgemiz koşullarındaki Marsh Seedless altıntopu için Carrizo sitranjı anacının, en iyi sonucu verdiği saptanmıştır

Tiplerin seçiminde; ağaç formu, gövde formu, gövde rengi, dal rengi, gövde çevresi ve yaprak sayısı, meyvelerde ise; meyve ağırlığı, boyu, eni, kalınlığı, kabuk rengi, meyve eti ağırlığı, meyve eti randımanı, tohum sayısı ve ağırlığı, tohum randımanı, tohum rengi ve suda çözünebilir kuru madde miktarları dikkate alınmıştır. 25. Pitfalls in the use of whole slide imaging for the diagnosis of central nervous system tumors: a pilot study in surgical neuropathology

Abstract

Background: Whole slide imaging (WSI) finds increasingly higher value in everyday surgical pathology in addition to its well-established use for educational and research purposes. However, its diagnostic utility, especially in subspecialty settings such as neuropathology, is not fully validated. Neuropathology practice is unique with smaller overall tissue size and frequent need for high-power evaluation. In addition, tumor grade is an integral part of the initial diagnosis. The purpose of this study is to assess the feasibility of primary pathology diagnosis of surgical neuropathology specimens using WSI. Materials and Methods: We reviewed consecutive surgical neuropathology cases diagnosed in our institution during a 2-month period and identified a single diagnostic slide, which was scanned at 40× magnification. Two neuropathologists who were blinded to the original diagnoses reviewed the whole slide image and rendered a diagnosis including tumor grade when applicable. They reviewed the single diagnostic slide after a wash-out period. Intra- and inter-observer discrepancies, as well as reasons for discrepancies, were evaluated. Results: The concordance rates were 94.9% and 88% for two neuropathologists. Two critical issues leading to discrepancies were identified: (1) identification of mitoses and (2) recognition of nuclear details. Conclusions: Given the current study is exclusively for surgical neuropathology cases, an all-encompassing conclusion about the utility of WSI for diagnostic purposes may not be available. Nevertheless, pathologists should be aware of the potential pitfalls due to identification of mitotic figures and nuclear details. Abstract

In this study, it was intended to identify the most suitable rootstock X scion combination, taking into account the carbohydrate content of grapefruit leaves of Marsh Seedless, which is amongst important citrus varieties, in terms of our country and our region, hanging on Troyer and Carrizo citrange rootstocks and considered an alternative to sour orange. The trees, which were 18 years old at West Mediterranean Agricultural Research Institute, have been used as experiment material. The highest carbohydrates contents were found at trees on citrange rootstocks. When we look for seasonal changes, we determined that total carbohydrates, starch and carbohydrate/nitrogen (C/N) rate increases from spring to summer and decreases to fall; total sugar, reduced sugar and sucrose content decreases from spring to fall and increases to winter. As a result of our study, the highest total carbohydrates contents were found at trees on Carrizo citrange rootstocks. On the other hand, higher starch, total sugar, reduced sugar and sucrose were determined on citrange rootstocks.
