Loss of chromosome 9p21 is observed in one-thirds of clear-cell renal cell carcinoma (ccRCC) and is associated with poorer patient survival. Unexpectedly, 9p21 LOH does not lead to decreased expression of the 9p21 tumor suppressor genes, CDKN2A and CDKN2B, suggesting alternative mechanisms of 9p-mediated tumorigenesis. Concordantly, CRISPR-mediated 9p21 deletion promotes growth of immortalized human embryonic kidney epithelial cells independently of the CDKN2A/B pathway inactivation. The 9p21 locus has a highly accessible chromatin structure, suggesting that 9p21 loss might contribute to kidney cancer progression by dysregulating genes distal to the 9p21 locus. We identified several 9p21 regulatory hubs by assessing which of the 9p21-interacting genes are dysregulated in 9p21-deleted kidney cells and ccRCCs. By focusing on the analysis of the homeobox gene 13 (HOXB13) locus, we found that 9p21 loss relieves the HOXB13 locus, decreasing HOXB13 methylation and promoting its expression. Upregulation of HOXB13 facilitates cell growth and is associated with poorer survival of patients with ccRCC.Implications:The results of our study propose a novel tumor suppressive mechanism on the basis of coordinated expression of physically associated genes, providing a better understanding of the role of chromosomal deletions in cancer.

Meningiomas are the most common benign brain tumors. Mutations of the E3 ubiquitin ligase TRAF7 occur in 25% of meningiomas and commonly cooccur with mutations in KLF4, yet the functional link between TRAF7 and KLF4 mutations remains unclear. By generating an in vitro meningioma model derived from primary meningeal cells, we elucidated the cooperative interactions that promote meningioma development. By integrating TRAF7-driven ubiquitinome and proteome alterations in meningeal cells and the TRAF7 interactome, we identified TRAF7 as a proteostatic regulator of RAS-related small GTPases. Meningioma-associated TRAF7 mutations disrupted either its catalytic activity or its interaction with RAS GTPases. TRAF7 loss in meningeal cells altered actin dynamics and promoted anchorage-independent growth by inducing CDC42 and RAS signaling. TRAF deficiency–driven activation of the RAS/MAPK pathway promoted KLF4-dependent transcription that led to upregulation of the tumor-suppressive Semaphorin pathway, a negative regulator of small GTPases. KLF4 loss of function disrupted this negative feedback loop and enhanced mutant TRAF7-mediated cell transformation. Overall, this study provides new mechanistic insights into meningioma development, which could lead to novel treatment strategies.Significance:The intricate molecular cross-talk between the ubiquitin ligase TRAF7 and the transcription factor KLF4 provides a first step toward the identification of new therapies for patients with meningioma.

Endothelial cells (ECs) grant access of disseminated cancer cells to distant organs. However, the molecular players regulating the activation of quiescent ECs at the premetastatic niche (PMN) remain elusive. Here, we find that ECs at the PMN coexpress tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) and its cognate death receptor 5 (DR5). Unexpectedly, endothelial TRAIL interacts intracellularly with DR5 to prevent its signaling and preserve a quiescent vascular phenotype. In absence of endothelial TRAIL, DR5 activation induces EC death and nuclear factor κB/p38-dependent EC stickiness, compromising vascular integrity and promoting myeloid cell infiltration, breast cancer cell adhesion, and metastasis. Consistently, both down-regulation of endothelial TRAIL at the PMN by proangiogenic tumor-secreted factors and the presence of the endogenous TRAIL inhibitors decoy receptor 1 (DcR1) and DcR2 favor metastasis. Long non-coding RNAs (lncRNAs) can exhibit cell-type and cancer-type specific expression profiles, making them highly attractive as therapeutic targets. Pan-cancer RNA sequencing data revealed broad expression of the SAMMSON lncRNA in uveal melanoma (UM), the most common primary intraocular malignancy in adults. Currently, there are no effective treatments for UM patients with metastatic disease, resulting in a median survival time of 6–12 months. We aimed to investigate the therapeutic potential of SAMMSON inhibition in UM. Antisense oligonucleotide (ASO)-mediated SAMMSON inhibition impaired the growth and viability of a genetically diverse panel of uveal melanoma cell lines. These effects were accompanied by an induction of apoptosis and were recapitulated in two uveal melanoma patient derived xenograft (PDX) models through subcutaneous ASO delivery. SAMMSON pulldown revealed several candidate interaction partners, including various proteins involved in mitochondrial translation. Consequently, inhibition of SAMMSON impaired global, mitochondrial and cytosolic protein translation levels and mitochondrial function in uveal melanoma cells. The present study demonstrates that SAMMSON expression is essential for uveal melanoma cell survival. ASO-mediated silencing of SAMMSON may provide an effective treatment strategy to treat primary and metastatic uveal melanoma patients.

Triple-negative breast cancer (TNBC) is the most aggressive breast cancer subtype, lacking effective therapy. Many TNBCs show remarkable response to carboplatin-based chemotherapy, but often develop resistance over time. With increasing use of carboplatin in the clinic, there is a pressing need to identify vulnerabilities of carboplatin-resistant tumors. In this study, we generated carboplatin-resistant TNBC MDA-MB-468 cell line and patient derived TNBC xenograft models. Mass spectrometry-based proteome profiling demonstrated that carboplatin resistance in TNBC is linked to drastic metabolism rewiring and upregulation of anti-oxidative response that supports cell replication by maintaining low levels of DNA damage in the presence of carboplatin. Carboplatin-resistant cells also exhibited dysregulation of the mitotic checkpoint. A kinome shRNA screen revealed that carboplatin-resistant cells are vulnerable to the depletion of the mitotic checkpoint regulators, whereas the checkpoint kinases CHEK1 and WEE1 are indispensable for the survival of carboplatin-resistant cells in the presence of carboplatin. We confirmed that pharmacological inhibition of CHEK1 by prexasertib in the presence of carboplatin is well tolerated by mice and suppresses the growth of carboplatin-resistant TNBC xenografts. Thus, abrogation of the mitotic checkpoint by CHEK1 inhibition re-sensitizes carboplatin-resistant TNBCs to carboplatin and represents a potential strategy for the treatment of carboplatin-resistant TNBCs.

Low molybdenum (Mo) bioavailability in acidic soil obstructs vegetable nitrogen assimilation and thus increases the health risk of vegetable ingestion due to nitrate accumulation. Constantly providing available Mo in acidic soil is a challenge for decreasing nitrate accumulation in vegetables. In this study, three Mo application methods, including biochar-based Mo slow-release fertilizer (Mo-biochar), seed dressing, and basal application, were investigated to enhance Mo bioavailability in acidic soil and nitrogen assimilation in Chinese flowering cabbage (Brassica parachinensis). The results showed that Mo-biochar constantly and sufficiently supplied Mo nutrients throughout the growing period of Brassica parachinensis, as evidenced by the soil available Mo, plant Mo uptake, and Mo values. The improved Mo supply was attributed to the alleviation of acidic soil (pH from 5.10 to 6.99) and the slow release of Mo adsorbed on biochar. Mo-biochar increased the nitrate reductase (NR) activity by 238.6% and glutamate dehydrogenase activity by 27.5%, indicating an enhancement of the rate-limiting steps of nitrogen assimilation, especially for nitrate reduction and amino acid synthesis. The increase in Mo-containing NR could be directly ascribed to the high level of Mo in Brassica parachinensis. Compared with the control, the nitrate content of Brassica parachinensis decreased by 42.9% due to the nitrate reduction induced by increased NR. Dysregulated splicing is a common event in cancer even in the absence of mutations in the core splicing machinery. The aberrant long non-coding transcriptome constitutes an uncharacterized level of regulation of post-transcriptional events in cancer. Here, we found that the stress-induced long non-coding RNA (lncRNA), LINC02657 or LASTR (lncRNA associated with SART3 regulation of splicing), is upregulated in hypoxic breast cancer and is essential for the growth of LASTR-positive triple-negative breast tumors. LASTR is upregulated in several types of epithelial cancers due to the activation of the stress-induced JNK/c-JUN pathway. Using a mass-spectrometry based approach, we identified the RNA-splicing factor SART3 as a LASTR-interacting partner. We found that LASTR promotes splicing efficiency by controlling SART3 association with the U4 and U6 small nuclear ribonucleoproteins (snRNP) during spliceosome recycling. Intron retention induced by LASTR depletion downregulates expression of essential genes, ultimately decreasing the fitness of cancer cells.

Radiotherapy is one of the most used treatment approaches for head and neck squamous cell carcinoma (HNSCC). Targeted inhibition of DNA repair machinery has the potential to improve treatment response by tailoring treatment to cancer cells lacking specific DNA repair pathways. Human papillomavirus (HPV)‐negative and HPV‐positive HNSCCs respond differently to radiotherapy treatment, suggesting that different approaches of DNA repair inhibition should be employed for these HNSCC groups. Here, we searched for optimal radiosensitization approaches for HPV‐positive and HPV‐negative HNSCCs by performing a targeted CRISPR‐Cas9 screen. We found that inhibition of base excision repair resulted in a better radiotherapy response in HPV‐positive HNSCC, which is correlated with upregulation of genes involved in base excision repair. In contrast, inhibition of nonhomologous end‐joining and mismatch repair showed strong effects in both HNSCC groups. We validated the screen results by combining radiotherapy with targeted inhibition of DNA repair in several preclinical models including primary and recurrent patient‐derived HNSCC xenografts. These findings underline the importance of stratifying HNSCC patients for combination treatments., What's new? Genomic loss of in Neurofibromin 1 (NF1) is a recurrent mechanism driving metastatic progression and resistance to endocrine therapy in ER-positive breast cancer, via activation of the MAPK pathway, induction of cyclin D1 expression and modulation of estrogen receptor signaling. To identify specific mediators of tumor growth in NF1 mutant cancers, we performed a genetic knockout (KO) screen in isogenic WT and NF1 KO breast cancer cells. After employing stringent cutoff criteria, we found 28 genes that are specifically essential for NF1 KO cells but not WT cells. Among these genes, NR2F2, encoding an orphan nuclear receptor, was one of the top candidates. We validated that its loss significantly impaired the growth of multiple models of NF1 null, ER+ breast cancer. Loss of NF1 leads to the hyperactivation of Ras which sequentially activates multiple effector pathways, including the MAPK pathway and the PI3K-AKT pathway. We found that MAPK activation upon NF1 loss led to elevated NR2F2 expression levels. A negative correlation between NF1 and NR2F2 expression was further confirmed in public gene expression databases. These data suggested that induction of NR2F2 expression might be one mechanism that mediates endocrine resistance caused by NF1 loss. Consistently, we found that exogenous expression of NR2F2 could confer endocrine resistance while its knockout hypersensitized ER+ breast cancer cells to endocrine therapies and significantly attenuated NF1 loss induced resistance. RNA-seq analysis revealed that estrogen response gene sets were among the most enriched signatures upon NF1 KO, NR2F2 over-expression (OE) or NR2F2 KO. NF1 KO or NR2F2 OE decreased the expression of early estrogen response genes and impaired their E2-induced expression, most of which could be rescued by NR2F2 KO, suggesting a regulatory effect of NR2F2 on ER transcriptional activity. To further define these mechanisms, ongoing integrative analyses of NR2F2 qPlex-RIME, ER and NR2F2 ChIP-Sequencing and ATAC-Sequencing will be presented. Taken together, our studies identify NR2F2 as a MAPK driven transcript whose induction mediates endocrine resistance, and may serve as a novel therapeutic target to overcome anti-estrogen resistant breast cancer growth. Rationale: Noonan syndrome (NS) is one of the most frequent genetic disorders. Bleeding problems are among the most common, yet poorly defined complications associated with NS. A lack of consensus on the management of bleeding complications in patients with NS indicates an urgent need for new therapeutic approaches. Objective: Bleeding disorders have recently been described in patients with NS harboring mutations of LZTR1 (leucine zipper-like transcription regulator 1), an adaptor for CUL3 (CULLIN3) ubiquitin ligase complex. Here, we assessed the pathobiology of LZTR1-mediated bleeding disorders. Methods and Results: Whole-body and vascular specific knockout of Lztr1 results in perinatal lethality due to cardiovascular dysfunction. Lztr1 deletion in blood vessels of adult mice leads to abnormal vascular leakage. We found that defective adherent and tight junctions in Lztr1 -depleted endothelial cells are caused by dysregulation of vesicular trafficking. LZTR1 affects the dynamics of fusion and fission of recycling endosomes by controlling ubiquitination of the ESCRT-III (endosomal sorting complex required for transport III) component CHMP1B (charged multivesicular protein 1B), whereas NS-associated LZTR1 mutations diminish CHMP1B ubiquitination. LZTR1-mediated dysregulation of CHMP1B ubiquitination triggers endosomal accumulation and subsequent activation of VEGFR2 (vascular endothelial growth factor receptor 2) and decreases blood levels of soluble VEGFR2 in Lztr1 haploinsufficient mice. Inhibition of VEGFR2 activity by cediranib rescues vascular abnormalities observed in Lztr1 knockout mice Conclusions: Lztr1 deletion phenotypically overlaps with bleeding diathesis observed in patients with NS. ELISA screening of soluble VEGFR2 in the blood of LZTR1 -mutated patients with NS may predict both the severity of NS phenotypes and potential responders to anti-VEGF therapy. VEGFR inhibitors could be beneficial for the treatment of bleeding disorders in patients with NS.

This paper uses evolutionary game theory to analyse regional tourism cooperation. Asymmetricevolutionary game model is established to explore the intrinsic motivation of regional tourismcooperation, and we via rigorous mathematical reasoning to prove a series of proposition. At the same time, through numerical simulation on each proposition by the MATLAB software, it vividly reflects the evolution process of the regional tourism cooperation. The results show that, first, it is necessary to cooperate, which is the inevitable trend in the development; Second, we shall specify the necessary condition to maintain cooperation, when the no-cooperative party can obtain additional benefit of freeriding from the cooperation party, and the additional benefit is greater than the benefit from bilateral cooperation, cooperation will ultimately fail; third, we must focus on the sufficient conditions to guarantee cooperation, which mainly reflected on the utilization of shared resources, cooperation cost, the benefit of “free-riding” etc.. In view of the results, we propose some suggestions to promote regional tourism cooperation from the regulations of regional tourism cooperation, government and the utilization of shared resources.

Porous flower-like hematite microstructures assembled with porous nanosheets were prepared by thermal decomposition of a precursor which was obtained using FeCl 3 ·6H 2 O, urea, and polyvinylpyrrolidone (PVP) in the solvent of ethylene glycol (EG) via a simple solution method. Because of the high specific surface area and porous structure, such α-Fe 2 O 3 microflowers showed excellent adsorption performances for the Congo red in wastewater treatment with a maximum removal capacity of 628.9 mg/g. The adsorption rate and adsorption isotherm were also investigated, and were well represented by the Langmuir model. These hematite flowers may hold great potential as environmentally friendly dye adsorbent candidate in decontamination of water.

Density and neutron logs play an important role in the exploration of coalbed methane (CBM) reservoirs. However, the study of these two log responses of coalbeds is deficient. Based on laboratory data in the published literature and field logs of CBM reservoirs from several coal basins in China, this paper focuses on acquiring and analyzing variations in bulk density and thermal neutrons of coal with different ranks. Two new methods are introduced to correct the effect of ash on field logs to obtain the log values of ash-free coal. The corrected coalbed density logs are in accordance with the density of coal samples in the laboratory. Then, hydrogen indices of coals with different ranks are simulated, and the corrected neutron logs are well verified by simulated values. After obtaining the variations of density and neutrons of coalbeds with different ranks, the behaviors of density and neutron logs are analyzed and discussed. Based on the laboratory data and the simulated and corrected field logs, the specific values of density and neutron porosity for ash-free coals with higher ranks are listed. Furthermore, a crossplot of density versus neutron logs is proposed to identify coal ranks, which has been successfully applied to various CBM fields of several basins in China.

A new well test model is developed for the hydraulic fractured well in coalbed by considering the following aspects: methane desorption phenomena, finite conductivity vertical fractures, and asymmetry of the fracture about the well. A new parameter is introduced to describe the storage of the fracture, which is named as a combined fracture storage. Another new concept called the fracture asymmetry coefficient is used to define the asymmetry of the fracture about the well. Finite element method (FEM) is used to solve the new mathematical model. The well test type curves and pressure fields are obtained and analyzed. The effects of the combined fracture storage, desorption factor, fracture conductivity, and fracture asymmetry coefficient on the well test type curves are discussed in detail. In order to verify the new model, a set of field well test data is analyzed.

The novel intumescent flame retardant rigid polyurethane foams ((PEPS-RPUF) were prepared based on a P-N containing intumescent flame retardant, poly ethanediamine spirocyclic pentaerythritol bisphosphonate (PEPS). The flammability, thermal, and mechanical properties of PEPS-RPUF were discussed. Scanning electron microscopy (SEM) and compression strength tests showed PEPS exhibited favorable compatibility with polyurethane matrix and smaller negative influence on the mechanical properties of PEPS-RPUF. Cone calorimeter, vertical burning test (UL-94) and limiting oxygen index (LOI) showed when the content of PEPS was 25 pph, the peak heat release rate (PHRR) and total heat release (THR) decrease, respectively, from 75.2 to 52.2 kW m−2 and from 5.7 to 4.1 MJ m−2. The LOI of PEPS-RPUF could reach 27%, and a UL-94 V-0 rating was achieved. The PEPS-RPUF exhibited an outstanding water resistance that it could still obtain a V-0 rating after water soaking. Thermogravimetric analysis (TGA) showed the charring ability of PEPS-RPUF was improved greatly compared to RPUF. SEM and Fourier transform-infrared spectroscopy (FT-IR) were utilized to characterize the surface morphology and chemical structure of the intumescent chars formed from PEPS-RPUF. The results indicated the chars were compact and smooth, which was a critical factor for protecting the substrate material from burning. POLYM. ENG. In this paper, zinc phytate was prepared and used as a synergist in intumescent flame-retarded polypropylene composites. The results showed that the polypropylene composites with 17 wt% intumescent flame retardant and 2 wt% zinc phytate have a limiting oxygen index 29.2 and achieve the UL-94 V-0 rating. Moreover, the peak heat release rate of the polypropylene composites decreases from 374 to 275 kW/m2. Real-time Fourier-transform infrared spectroscopy showed that the zinc phytate delays the emission of carbon dioxide indicating that zinc phytate slows the degradation of polypropylene which regulates the suitability of intumescent flame-retardant system in polypropylene.

The photochemical reaction of Fe2(S2)(CO)6 and Ph2CS affords the perthiolate Fe2(S3CPh2)(CO)6 (1) in good yield. As confirmed crystallographically, 1 contains a previously elusive perthiolate ligand. The related reaction of Fe2(S2)(CO)5-(PPh3) and Ph2CS gave Fe2(S3CPh2)(CO)5(PPh3). Although Fe3S2(CO)9 and Ph2CS failed to efficiently give Fe2(S2CPh2)(CO)6, this compound could be prepared by desulfurization of 1 using PPh3.

The mathematical model of dynamic loads was developed based on an analysis of the polished rod load of beam pumps, and the variation of the dynamic loads and the computation of the minimum and maximum limits during a complete pumping cycle were given out by solving the model. Field examples verify that it is necessary to take into account the inertial and vibration loads while calculating polished rod loads. During the prophase of the pumping production, the dynamic to polished rod load ratio is relatively large. Then the ratio decreases rapidly and becomes small after entering stable production. Moreover, the total deformation of rod and tubing in CBM wells is much smaller than that in oil fields, and the deformation caused by the dynamic loads is also relatively small. The result of this work is the calculation of the dynamic loads. The application of this calculation for the sucker rod pumping system in CBM wells can give the desired accuracy of polished rod load and the dynamometer cards, which provides a reasonable basis for the design and selection of beam pumps.

Hirschsprung-associated enterocolitis (HAEC) is a life-threatening complication of Hirschsprung’s disease (HD). Although the pathological mechanisms are still unclear, studies have shown that HAEC has a close relationship with the disturbance of intestinal microbiota. This study aimed to investigate the characteristics of the intestinal microbiome of HD patients with or without enterocolitis. During routine or emergency surgery, we collected 35 intestinal content samples from five patients with HAEC and eight HD patients, including three HD patients with a history of enterocolitis who were in a HAEC remission (HAEC-R) phase. Using Illumina-MiSeq high-throughput sequencing, we sequenced the V4 region of bacterial 16S rRNA, and operational taxonomic units (OTUs) were defined by 97% sequence similarity. Principal coordinate analysis (PCoA) of weighted UniFrac distances was performed to evaluate the diversity of each intestinal microbiome sample. The microbiota differed significantly between the HD patients (characterized by the prevalence of Bacteroidetes) and HAEC patients (characterized by the prevalence of Proteobacteria), while the microbiota of the HAEC-R patients was more similar to that of the HAEC patients. We also observed that the specimens from different intestinal sites of each HD patient differed significantly, while the specimens from different intestinal sites of each HAEC and HAEC-R patient were more similar. In conclusion, the microbiome pattern of the HAEC-R patients was more similar to that of the HAEC patients than to that of the HD patients. The HD patients had a relatively distinct, more stable community than the HAEC and HAEC-R patients, suggesting that enterocolitis may either be caused by or result in a disruption of the patient’s uniquely adapted intestinal flora. The intestinal microbiota associated with enterocolitis may persist following symptom resolution and can be implicated in the symptom recurrence.

In the process of codebook design of vector quantization, traditional LBG algorithm owns the advantage of fast convergence, but it is prone to local optimum and is influenced greatly by initial codebook. Given that the Genetic Algorithm has the capability to produce global optimal results,this paper proposes a new clustering algorithm GA-L based on GA and LBG to improve the quality of codebook. This paper applies GA-L algorithm to Mandarin Continuous Digit Speech Recognition,the experiments results show GA-L algorithm is more effective than traditional LBG algorithm.
